The human middle-temporal region MT+ is highly specialized in processing visual motion. However, recent studies have shown that this region is modulated by extraretinal signals, suggesting a possible involvement in processing motion information also from non-visual modalities. Here, we used functional MRI data to investigate the influence of retinal and extraretinal signals on MT+ in a large sample of subjects. Moreover, we used resting-state functional MRI to assess how the subdivisions of MT+ (i.e., MST, FST, MT, and V4t) are functionally connected. We first compared responses in MST, FST, MT, and V4t to coherent vs. random visual motion. We found that only MST and FST were positively activated by coherent motion. Furthermore, regional analyses revealed that MST and FST were positively activated by leg, but not arm, movements, while MT and V4t were deactivated by arm, but not leg, movements. Taken together, regional analyses revealed a visuomotor role for the anterior areas MST and FST and a pure visual role for the anterior areas MT and V4t. These findings were mirrored by the pattern of functional connections between these areas and the rest of the brain. Visual and visuomotor regions showed distinct patterns of functional connectivity, with the latter preferentially connected with the somatosensory and motor areas representing leg and foot. Overall, these findings reveal a functional sensitivity for coherent visual motion and lower-limb movements in MST and FST, suggesting their possible involvement in integrating sensory and motor information to perform locomotion.

Despite extensive research, the functional architecture of the subregions of the dorsal posterior parietal cortex (PPC) involved in sensorimotor processing is far from clear. Here, we draw a thorough picture of the large-scale functional organization of the PPC to disentangle the fronto-parietal networks mediating visuomotor functions. To this aim, we reanalyzed available human functional magnetic resonance imaging data collected during the execution of saccades, hand, and foot pointing, and we combined individual surface-based activation, resting-state functional connectivity, and effective connectivity analyses. We described a functional distinction between a more lateral region in the posterior intraparietal sulcus (lpIPS), preferring saccades over pointing and coupled with the frontal eye fields (FEF) at rest, and a more medial portion (mpIPS) intrinsically correlated to the dorsal premotor cortex (PMd). Dynamic causal modeling revealed feedforward-feedback loops linking lpIPS with FEF during saccades and mpIPS with PMd during pointing, with substantial differences between hand and foot. Despite an intrinsic specialization of the action-specific fronto-parietal networks, our study reveals that their functioning is finely regulated according to the effector to be used, being the dynamic interactions within those networks differently modulated when carrying out a similar movement (i.e. pointing) but with distinct effectors (i.e. hand and foot).

Neuroimaging studies have revealed two separate classes of category-selective regions specialized in optic flow (egomotion-compatible) processing and in scene/place perception. Despite the importance of both optic flow and scene/place recognition to estimate changes in position and orientation within the environment during self-motion, the possible functional link between egomotion- and scene-selective regions has not yet been established. Here we reanalyzed functional magnetic resonance images from a large sample of participants performing two well-known “localizer” fMRI experiments, consisting in passive viewing of navigationally relevant stimuli such as buildings and places (scene/place stimulus) and coherently moving fields of dots simulating the visual stimulation during self-motion (flow fields). After interrogating the egomotion-selective areas with respect to the scene/place stimulus and the scene-selective areas with respect to flow fields, we found that the egomotion-selective areas V6+ and pIPS/V3A responded bilaterally more to scenes/places compared to faces, and all the scene-selective areas (parahippocampal place area or PPA, retrosplenial complex or RSC, and occipital place area or OPA) responded more to egomotion-compatible optic flow compared to random motion. The conjunction analysis between scene/place and flow field stimuli revealed that the most important focus of common activation was found in the dorsolateral parieto-occipital cortex, spanning the scene-selective OPA and the egomotion-selective pIPS/V3A. Individual inspection of the relative locations of these two regions revealed a partial overlap and a similar response profile to an independent low-level visual motion stimulus, suggesting that OPA and pIPS/V3A may be part of a unique motion-selective complex specialized in encoding both egomotion- and scene-relevant information, likely for the control of navigation in a structured environment. Visual cues coming from the lower visual field (VF) play an important role in the visual guidance of upper and lower limb movements. A recently described region situated in the dorsomedial parietal cortex, area hPEc (Pitzalis et al. in NeuroImage 202:116092, 2019), might have a role in integrating visually derived information with somatomotor signals to guide limb interaction with the environment. In macaque, it has been demonstrated that PEc receives visual information mostly from the lower visual field but, to date, there has been no systematic investigation of VF preference in the newly defined human homologue of macaque area PEc (hPEc). Here we examined the VF preferences of hPEc while participants performed a visuomotor task implying spatially directed delayed eye-, hand- and foot-movements towards different spatial locations within the VF. By analyzing data as a function of the different target locations towards which upcoming movements were planned (and then executed), we observed the presence of asymmetry in the vertical dimension of VF in area hPEc, being this area more strongly activated by limb movements directed towards visual targets located in the lower compared to the upper VF. This result confirms the view, first advanced in macaque monkey, that PEc is involved in processing visual information to guide body interaction with the external environment, including locomotion. We also observed a contralateral dominance for the lower VF preference in the foot selective somatomotor cortex anterior to hPEc. This result might reflect the role of this cortex (which includes areas PE and S-I) in providing highly topographically organized signals, likely useful to achieve an appropriate foot posture during locomotion. The existence of neural correlates of spatial attention is not limited to the reactive stage of stimulus processing: neural activities subtending spatial attention are deployed well ahead of stimulus onset. ERP evidence supporting this proactive (top-down) attentional control is based on trial-by-trial S1-S2 paradigms, where the onset of a directional cue (S1) indicates on which side attention must be directed to respond to an upcoming target stimulus (S2). Crucially, S1 onset trigger both attention and motor preparation, therefore, these paradigms are not ideal to demonstrate the effect of attention at preparatory stage of processing. To isolate top-down anticipatory attention, the present study used a sustained attention paradigm based on a steady cue that indicates the attended side constantly throughout an entire block of trials, without any onset of an attentional cue. The main result consists in the description of the attention effect on the visual negativity (vN) component, a growing neural activity starting before stimulus presentation in extrastriate visual areas. The vN was consistently lateralized in the hemisphere contralateral to the attended side, regardless of the hand to be used. At the opposite, the lateralized motor activity emerged long after, confirming that the hand-selection process followed the spatial attention orientation process. The present study confirms the anticipatory nature of the vN component and corroborate its role in terms of preparatory visuospatial attention.

Receiving feedback on action correctness is a relevant factor in learning, but only a few recent studies have investigated the neural bases involved in feedback processing and its consequences on performance. Several event-related potentials (ERP) studies investigated the feedback-related negativity, which is an ERP occurring after the presentation of a feedback stimulus. In contrast, the present study investigates the effect of providing feedback on brain activities before and after the presentation of an imperative stimulus with the aim to show how this could have an impact on cognitive functions related to anticipatory and post-stimulus task processing. Participants performed a standard visuomotor task and a modified version of the same task in which feedback sounds were emitted when participants committed performance errors. Overall, results showed that in the feedback task subjects have better cognitive control than in the standard task. All behavioral measures were improved in the feedback task. At the brain level, all the studied components were modulated by the presence of the feedback cue. Results pointed to a possible increase of anticipatory activity in the prefrontal cortex, a reduction of perceptual awareness in areas previously associated with the anterior insular cortex, and an increase of activity associated with selective attention in sensory cortices.

Integration of proprioceptive signals from the various effectors with visual feedback of self-motion from the retina is necessary for whole-body movement and locomotion. Here, we tested whether the human visual motion areas involved in processing optic flow signals simulating self-motion are also activated by goal-directed movements (as saccades or pointing) performed with different effectors (eye, hand, and foot), suggesting a role in visually guiding movements through the external environment. To achieve this aim, we used a combined approach of task-evoked activity and effective connectivity (PsychoPhysiological Interaction, PPI) by fMRI. We localized a set of six egomotion-responsive visual areas through the flow field stimulus and distinguished them into visual (pIPS/V3A, V6+ , IPSmot/VIP) and visuomotor (pCi, CSv, PIC) areas according to recent literature. We tested their response to a visuomotor task implying spatially directed delayed eye, hand, and foot movements. We observed a posterior-to-anterior gradient of preference for eye-to-foot movements, with posterior (visual) regions showing a preference for saccades, and anterior (visuomotor) regions showing a preference for foot pointing. No region showed a clear preference for hand pointing. Effective connectivity analysis showed that visual areas were more connected to each other with respect to the visuomotor areas, particularly during saccades. We suggest that visual and visuomotor egomotion regions can play different roles within a network that integrates sensory-motor signals with the aim of guiding movements in the external environment.

Application of a passive and fully articulated exoskeleton, called Human Body Posturizer (HBP), has been demonstrated to improve mobility, response accuracy and ambulation in multiple sclerosis (MS) patients. By using functional magnetic imaging (fMRI) during a visuomotor discrimination task, we performed a pilot study to evaluate the effect of HBP over the neural correlates of motor and cognitive functions which are typically impaired in MS patients. Specifically, we tested the effect of a 6-week multidisciplinary rehabilitation intervention on two groups of MS patients: a control group who followed a standard physiotherapeutic rehabilitation protocol, and an experimental group who used the HBP during physical exercises in addition to the standard protocol. We found that, after treatment, the experimental group exhibited a significant lower activity (as compared to the control group) in the inferior frontal gyrus. This post-treatment activity reduction can be explained as a retour to a normal range, being the amount of iFg activity observed in the experimental patients very similar to that observed in healthy subjects. These findings indicate that the use of HBP during rehabilitation intervention normalizes the prefrontal activity, mitigating the cortical hyperactivity associated to MS.

During locomotion, leg movements define the direction of walking (forward or backward) and the path one is taking (straight or curved). These aspects of locomotion produce characteristic visual motion patterns during movement. Here, we tested whether cortical regions responding to either egomotion-compatible visual motion, or leg movements, or both, are sensitive to these locomotion-relevant aspects of visual motion. We compared a curved path (typically the visual feedback of a changing direction of movement in the environment) to a linear path for simulated forward and backward motion in an event-related fMRI experiment. We used an individual surface-based approach and two functional localizers to define (1) six egomotion-related areas (V6+, V3A, intraparietal motion area [IPSmot], cingulate sulcus visual area [CSv], posterior cingulate area [pCi], posterior insular cortex [PIC]) using the flow field stimulus and (2) three leg-related cortical regions (human PEc [hPEc], human PE [hPE] and primary somatosensory cortex [S-I]) using a somatomotor task. Then, we extracted the response from all these regions with respect to the main event-related fMRI experiment, consisting of passive viewing of an optic flow stimulus, simulating a forward or backward direction of self-motion in either linear or curved path. Results showed that some regions have a significant preference for the curved path motion (hPEc, hPE, S-I, IPSmot) or a preference for the forward motion (V3A), while other regions have both a significant preference for the curved path motion and for the forward compared to backward motion (V6+, CSv, pCi). We did not find any significant effects of the present stimuli in PIC. Since controlling locomotion mainly means controlling changes of walking direction in the environment during forward self-motion, such a differential functional profile among these cortical regions suggests that they play a differentiated role in the visual guidance of locomotion.

International audience; In humans, several neuroimaging studies have demonstrated that passive viewing of optic flow stimuli activates higher-level motion areas, like V6 and the cingulate sulcus visual area (CSv). In macaque, there are few studies on the sensitivity of V6 and CSv to egomotion compatible optic flow. The only fMRI study on this issue revealed selectivity to egomotion compatible optic flow in macaque CSv but not in V6 (Cotterau et al. Cereb Cortex 27(1):330-343, 2017, but see Fan et al. J Neurosci. 35:16303-16314, 2015). Yet, it is unknown whether monkey visual motion areas MT + and V6 display any distinctive fMRI functional profile relative to the optic flow stimulation, as it is the case for the homologous human areas (Pitzalis et al., Cereb Cortex 20(2):411-424, 2010). Here, we described the sensitivity of the monkey brain to two motion stimuli (radial rings and flow fields) originally used in humans to functionally map the motion middle temporal area MT + (Tootell et al. J Neurosci 15: 3215-3230, 1995a; Nature 375:139-141, 1995b) and the motion medial parietal area V6 (Pitzalis et al. 2010), respectively. In both animals, we found regions responding only to optic flow or radial rings stimulation, and regions responding to both stimuli. A region in the parieto-occipital sulcus (likely including V6) was one of the most highly selective area for coherently moving fields of dots, further demonstrating the power of this type of stimulation to activate V6 in both humans and monkeys. We did not find any evidence that putative macaque CSv responds to Flow Fields.

Key points: Monovision is an optical correction for presbyopes that consists of correcting one eye for far distance and the other for near distance, creating a superimposition of an in-focus with a blurred image. Brain adaptation to monovision was studied in unexperienced observers by measuring visual evoked potentials from 64-channels. The first clear effect of monovision on visual evoked potentials was the C1 amplitude reduction, indicating that the unilateral blurring induced by monovision reduces feed-forward activity in primary visual area. Monovision led also to an increased amplitude of the P1 and pP1 components, with the latter originating in prefrontal regions. This effect probably works as an attentional compensatory activity used to compensate for the degraded V1 signal. A common and often successful option to correct presbyopia with contact lenses is monovision. This is an unbalanced correction across the two eyes where one eye is corrected for far vision and the other eye is corrected for near vision. Monovision is therefore a form of acquired anisometropia that causes a superimposition of an in-focus image with a blurred image. In spite of this visual anisometropia, monovision has been successfully used for many decadesl however the brain mechanism supporting monovision is not well understood. The present study aimed to measure the visual evoked potentials with a high-density electrode array (64-channel) in a group of presbyopes and to provide a detailed spatiotemporal analysis of the cortical activity after a short period of adaptation to monovision with contact lenses. When compared with a balanced eye near correction, monovision produced both a clear reduction of the earliest visual evoked potential components, the C1 and the N1, and an amplitude increase of the P1 and pP1. These results indicate that the unilateral blurring induced by wearing monovision contact lenses reduces feed-forward activity in the primary visual area and feedback activity in extrastriate areas (C1 and N1 reduction). Interestingly, other brain activities in both extrastriate visual areas (the P1 component) and in the anterior insula (the pP1 component) appear to compensate for this dysfunction, increasing their activity during monovision. These changes confirm the presence of fluid brain adaptation in visual and non-visual areas during monocular interferences.

Research on preparatory brain processes taking place before acting shows unexpected connections with cognitive processing. From 50 years, we know that motor-related brain activity can be measured by electrocortical recordings 1-3s before voluntary actions. This readiness potential has been associated with increasing excitably of premotor and motor areas and directly linked to the kinematic of the upcoming action. Now we know that the mere motor preparation is only one function of a more complex preparatory activity. Recent research shows that before any action many cognitive processes may occur depending on various aspects of the action, such as complexity, meaning, emotional valence, fatigue and consequences of the action itself. In addition to studies on self-paced action, the review considers also studies on externally-triggered paradigms showing differences in preparation processes related to age, physical exercise, and task instructions. Evidences from electrophysiological and neuroimaging recording indicate that in addition to the motor areas, the prefrontal, parietal and sensory cortices may be active during action preparation to anticipate future events and calibrate responses.

Object prehension typically includes a transport phase (reaching) and a grip phase (grasping). Within the posterior parietal cortex (PPC), grasping movements have been traditionally associated to a lateral activation, although recent monkey evidence suggests also a medial involvement. Here, we wanted to determine whether grasping-related activities are present in the human dorsomedial parietal cortex, by focusing on two cortical regions specialized in the monkey in controlling limb movements, i.e., V6A (composed by its ventral and dorsal sectors, V6Av and V6Ad, respectively) and PEc, both recently defined also in humans. We acquired functional magnetic resonance images while participants performed both real (pantomimed) and imagined grasping of visually-presented objects. We found that the human areas V6Ad (hV6Ad) and PEc (hPEc) were both activated by real grasping, whereas hV6Ad only was activated by the imagery of grasping movements. hV6Av was not involved in either types of grasping. These results speak against the traditional notion of a medial-to-lateral segregation of reaching versus grasping information within the PPC and strengthen the idea that the human dorsomedial parietal cortex implements the whole complex pattern of visuomotor transformations required for object-oriented actions. Our findings suggest that hV6Ad is particularly involved in implementing all the visuomotor transformations needed to create an abstract representation of the object-directed action, while hPEc is involved in implementing the sensorimotor transformations needed to actually perform that action.

Prediction about event timing plays a leading role in organizing and optimizing behavior. We recorded anticipatory brain activities and evaluated whether temporal orienting processes are reflected by the novel prefrontal negative (pN) component, as already shown for the contingent negative variation (CNV). Fourteen young healthy participants underwent EEG and fMRI recordings in separate sessions; they were asked to perform a Go/No-Go task in which temporal orienting was manipulated: the external condition (a visual display indicating the time of stimulus onset) and the internal condition (time information not provided). In both conditions, the source of the pN was localized in the pars opercularis of the iFg; the source of the CNV was localized in the supplementary motor area and cingulate motor area, as expected. Anticipatory activity was also found in the occipital-parietal cortex. Time on task EEG analysis showed a marked learning effect in the internal condition, while the effect was minor in the external condition. In fMRI, the two conditions had a similar pattern; similarities and differences of results obtained with the two techniques are discussed. Overall, data are consistent with the view that the pN reflects a proactive cognitive control, including temporal orienting.

To plan movements toward objects our brain must recognize whether retinal displacement is due to self‐motion and/or to object‐motion. Here, we aimed to test whether motion areas are able to segregate these types of motion. We combined an event‐related functional magnetic resonance imaging experiment, brain mapping techniques, and wide‐field stimulation to study the responsivity of motion‐sensitive areas to pure and combined self‐ and object‐motion conditions during virtual movies of a train running within a realistic landscape. We observed a selective response in MT to the pure object‐motion condition, and in medial (PEc, pCi, CSv, and CMA) and lateral (PIC and LOR) areas to the pure self‐motion condition. Some other regions (like V6) responded more to complex visual stimulation where both object‐ and self‐motion were present. Among all, we found that some motion regions (V3A, LOR, MT, V6, and IPSmot) could extract object‐motion information from the overall motion, recognizing the real movement of the train even when the images remain still (on the screen), or moved, because of self‐movements. We propose that these motion areas might be good candidates for the “flow parsing mechanism,” that is the capability to extract object‐motion information from retinal motion signals by subtracting out the optic flow components.

Monkey neurophysiology and human neuroimaging studies have demonstrated that passive viewing of optic flow stimuli activates a cortical network of temporal, parietal, insular, and cingulate visual motion regions. Here, we tested whether the human visual motion areas involved in processing optic flow signals simulating self‐motion are also activated by active lower limb movements, and hence are likely involved in guiding human locomotion. To this aim, we used a combined approach of task‐evoked activity and resting‐state functional connectivity by fMRI. We localized a set of six egomotion‐responsive visual areas (V6+, V3A, intraparietal motion/ventral intraparietal [IPSmot/VIP], cingulate sulcus visual area [CSv], posterior cingulate sulcus area [pCi], posterior insular cortex [PIC]) by using optic flow. We tested their response to a motor task implying long‐range active leg movements. Results revealed that, among these visually defined areas, CSv, pCi, and PIC responded to leg movements (visuomotor areas), while V6+, V3A, and IPSmot/VIP did not (visual areas). Functional connectivity analysis showed that visuomotor areas are connected to the cingulate motor areas, the supplementary motor area, and notably to the medial portion of the somatosensory cortex, which represents legs and feet. We suggest that CSv, pCi, and PIC perform the visual analysis of egomotion‐like signals to provide sensory information to the motor system with the aim of guiding locomotion.

Background: Motor and inhibitory control rely on frontal cortex activity, which is known to reach full maturation only in late adolescence. The development of inhibitory control has been studied using event-related potentials (ERP), focusing on reactive processing (i.e. the N2 and the P3 components). Scarce information exists concerning pre-stimulus activity as that represented by the Bereinshafstpotential (BP) and by the prefrontal negativity (pN). Further, no literature exists concerning the post-stimulus components originating within the anterior insula (pN1, pP1, pP2). This study aims at associating children performance with these motor-cognitive processing in frontal brain areas. Methods: High-resolution EEG recordings were employed to measure ERPs from 18 children (12 years old) and 18 adults (28 years old) during a visuo-motor discriminative response task. Response time (RT), commission (CE) and omission errors, and RT variability were compared between groups. At brain level, two pre-stimulus (BP and pN) and seven post-stimulus (P1; pN1; N1; pP1; N2; pP2; P3) ERP components were compared between groups. Results: Children showed slower and more variable RTs and poorer inhibition (higher CEs) than adults. At electrophysiological level, children presented smaller BP and pN. After stimulus onset, children showed lower amplitude of N1, pP1, P3, and pP2 components. The P1, pP1, N2 and P3 were delayed compared to adults. Conclusions: Our results demonstrate that children are characterized by less intense task-related proactive activities in frontal cortex, which may account for subsequent poor and delayed reactive processing and, thus, for inaccurate and slow performance.

Purpose: To evaluate, in an amateur sports-playing population, the prevalence of refractive error, the type of vision correction used during sport and attitudes toward different kinds of vision correction used in various types of sports. Method: A questionnaire was used for people engaging in sport and data was collected from sport centres, gyms and universities that focused on the motor sciences. Results: One thousand, five hundred and seventy-three questionnaires were collected (mean age 26.5 ± 12.9 years; 63.5 per cent male). Nearly all (93.8 per cent) subjects stated that their vision had been checked at least once. Fifty-three subjects (3.4 per cent) had undergone refractive surgery. Of the remainder who did not have refractive surgery (n = 1,519), 580 (38.2 per cent) reported a defect of vision, 474 (31.2 per cent) were myopic, 63 (4.1 per cent) hyperopic and 241 (15.9 per cent) astigmatic. Logistic regression analysis showed that the best predictors for myopia prevalence were gender (p < 0.001) and location of sport practice (p < 0.001). Sports that present higher prevalence of outdoor activity have lower prevalence of myopia. Contact lens penetration over the study sample was 18.7 per cent. Contact lenses were the favourite system of correction among people interviewed compared to spectacles and refractive surgery (p < 0.001). Conclusions: This study showed that sport was not associated with different levels of myopia prevalence in the adult population. However, subjects engaging in outdoor sports had lower rates of myopia prevalence. Penetration of contact lens use in sport was four times higher than the overall adult population. Contact lenses were the preferred system of correction in sports compared to spectacles or refractive surgery, but this preference was affected by the type of sport practised and by the age and level of sports activity for which the preference was required.

According to recent models of visuo-spatial processing, the medial parieto-occipital cortex is a crucial node of the dorsal visual stream. Evidence from neurophysiological studies in monkeys has indicated that the parieto-occipital sulcus (POS) contains three functionally and cytoarchitectonically distinct areas: the visual area V6 in the fundus of the POS, and the visuo-motor areas V6Av and V6Ad in a progressively dorsal and anterior location with respect to V6. Besides different topographical organization, cytoarchitectonics, and functional properties, these three monkey areas can also be distinguished based on their patterns of cortico-cortical connections. Thanks to wide-field retinotopic mapping, areas V6 and V6Av have been also mapped in the human brain. Here, using a combined approach of resting-state functional connectivity and task-evoked activity by fMRI, we identified a new region in the anterior POS showing a pattern of functional properties and cortical connections that suggests a homology with the monkey area V6Ad. In addition, we observed distinct patterns of cortical connections associated with the human V6 and V6Av which are remarkably consistent with those showed by the anatomical tracing studies in the corresponding monkey areas. Consistent with recent models on visuo-spatial processing, our findings demonstrate a gradient of functional specialization and cortical connections within the human POS, with more posterior regions primarily dedicated to the analysis of visual attributes useful for spatial navigation and more anterior regions primarily dedicated to analyses of spatial information relevant for goal-directed action.

Temporal and spatial filtering of fMRI data is often used to improve statistical power. However, conventional methods, such as smoothing with fixed-width Gaussian filters, remove fine-scale structure in the data, necessitating a tradeoff between sensitivity and specificity. Specifically, smoothing may increase sensitivity (reduce noise and increase statistical power) but at the cost loss of specificity in that fine-scale structure in neural activity patterns is lost. Here, we propose an alternative smoothing method based on Gaussian processes (GP) regression for single subjects fMRI experiments. This method adapts the level of smoothing on a voxel by voxel basis according to the characteristics of the local neural activity patterns. GP-based fMRI analysis has been heretofore impractical owing to computational demands. Here, we demonstrate a new implementation of GP that makes it possible to handle the massive data dimensionality of the typical fMRI experiment. We demonstrate how GP can be used as a drop-in replacement to conventional preprocessing steps for temporal and spatial smoothing in a standard fMRI pipeline. We present simulated and experimental results that show the increased sensitivity and specificity compared to conventional smoothing strategies. Flexible and adaptive behavior requires the ability to contextually stop inappropriate actions and select the right one as quickly as possible. Recently, it has been proposed that three brain regions, i.e., the inferior frontal gyrus (iFg), the anterior insula (aIns), and the anterior intraparietal sulcus (aIPs), play an important role in several processing phases of perceptual decision tasks, especially in the preparation, perception and action phases, respectively. However, little is known about hemispheric differences in the activation of these three areas during the transition from perception to action. Many studies have examined how people prepare to stop upcoming responses through both proactive and reactive inhibitory control. Although inhibitory control has been associated with activity in the right prefrontal cortex (PFC), we have previously reported that, during a discriminative response task performed with the right hand, we observed: 1) a bilateral activity in the iFg during the preparation phase, and 2) a left dominant activity in the aIns and aIPs during the transition from perception to action, i.e., the so-called stimulus-response mapping. To clarify the hemispheric dominance of these processes, we combined the high temporal resolution of event-related potentials (ERPs) with the high spatial resolution of event-related functional magnetic resonance imaging (fMRI) while participants performed a discriminative response task (DRT) and a simple response task (SRT) using their non-dominant left hand. We confirmed that proactive inhibitory control originates in the iFg: its activity started one second before the stimulus onset and was released concomitantly to the stimulus appearance. Most importantly, we confirmed the presence of a bilateral iFg activity that seems to reflect a bilateral proactive control rather than a right-hemisphere dominance or a stronger control of the hemisphere contralateral to the responding hand. Further, we observed a stronger activation of the left aIns and a right-lateralized activation of the aIPs reflecting left-hemisphere dominance for stimulus-response mapping finalized to response execution and a contralateral-hand parietal premotor activity, respectively.

We investigated the movement related cortical potentials (MRCPs) associated with self-paced horizontal voluntary saccades and evaluated their cortical sources by applying dipole model. A fixation point and two targets (6° of eccentricity in the left and right fields) were continuously displayed on a screen. A first group (15 subjects) performed a saccade toward one of the lateral targets immediately followed by a re-centering saccade. A second group (15 subjects) performed a saccade followed by a long fixation (few seconds) before a re-centering. Results showed a sequence of activities in contralateral Intra-Parietal Sulcus (IPS), Supplementary Eye Fields and Frontal Eye Fields. In the case of long-fixation-saccades, an additional source in ipsilateral IPS was detected in the planning phase. The amplitude and timing of the planning phase, associated with BP components, were influenced by task demands. Similarity and difference between MRCPs for eyes and limbs movements are also presented.

We studied pre-movement cortical activity related to praxic actions performed at self-paced rate and having ecological meanings and functions. Motor-related cortical potentials were recorded using 64-channels EEG in two experiments. Experiment 1 included 15 subjects performing in separate blocks two object-oriented actions: grasping a tea-cup and impossible grasping of a tea-cup (same goal but the grasp was mechanically hindered). Experiment 2 included a subset of 7 subjects from Exp. 1 and the action was reaching a tea-cup; this control condition had a different goal but was kinematically similar to impossible grasping. Different activity patterns in terms of onset, amplitude, duration and, at least in part, sources were recorded in the preparation phase (BP component) according to the specific action and to the possibility of accomplishing it. The main result is that parietal areas were involved in grasping preparation (called "posterior" BP) and not in reaching and impossible grasping preparation. The anterior frontal-central activity (called "anterior" BP) during preparation for grasping started earlier than the other two conditions. The cortical activity during preparation for reaching was similar to that for impossible grasping, except for a frontal activity only detected in the latter condition. It is concluded that the action preparation, even in its early phase, is affected by action meaning and by the awareness of being able to perform the requested action.

In the monkey posterior parietal cortex (PPC), there is clear evidence of anatomically segregated neuronal populations specialized for planning saccades and arm-reaching movements. However, functional neuroimaging studies in humans have yielded controversial results. Here we show that the human PPC contains distinct subregions responsive to salient visual cues, some of which combine spatial and action-related signals into ‘intentional’ signals. Participants underwent event-related functional magnetic resonance imaging while performing delayed saccades and long-range arm reaches instructed by visual cues. We focused on activity in the time period following the cue and preceding the actual movement. The use of individual cortical surface reconstructions with detailed sulcal labeling allowed the definition of six responsive regions with distinctive anatomical locations in the PPC. Each region exhibited a distinctive combination of transient and sustained signals during the delay, modulated by either the cue spatial location (contralateral vs. ipsilateral), the instructed action (saccades vs. reaching) or both. Importantly, a lateral and a medial dorsal parietal region showed sustained responses during the delay preferentially for contralateral saccadic and reaching trials, respectively. In the lateral region, preference for saccades was evident only as a more sustained response during saccadic vs. reaching delays, whereas the medial region also showed a higher transient response to cues signaling reaching vs. saccadic actions. These response profiles closely match the behavior of neurons in the macaque lateral and medial intraparietal area, respectively, and suggest that these corresponding human regions are encoding spatially directed action plans or ‘intentions’.

Infection with high-risk HPV genotypes is considered an essential step in cervical carcinogenesis. Recently, prophylactic anti-HPV vaccines have shown to provide effective protection in clinical trials. However, protection appears to be type-restricted, thus, its real extent will depend on HPV types prevalence in the target population. Here we report HPV prevalence in 555 Italian women with various stages of HPV-related cervical disease (343 CIN1/L-SIL, 156 CIN2-3/HIGH-SIL, 43 invasive squamous cervical carcinoma and 13 adenocarcinoma), as well as in 315 women with smears negative for intraepithelial lesions or malignancy (NIL). HPV was found in 50.7% of CIN1/L-SIL, 87.2% of CIN2-3/HIGH-SIL, 97.7% of squamous carcinomas, 69.2% of adenocarcinomas and in 29.5% of NIL. HPV 16 was present in 69.0% of squamous carcinomas, 58.8% of CIN2-3/HIGH-SIL, in 22.9 and 16.1% of CIN1/L-SIL and NIL, respectively. HPV 18 was found in 7.3% and 4.8%, respectively, while in 35.3% and 26.2% of CIN2-3/HIGH-SIL and invasive carcinomas, respectively, were present high-risk HPV types other than type 16 and 18. Based on these data, it is expected that current HPV prophylactic vaccines could effectively prevent up to 70% of invasive cervical cancers and a slightly smaller proportion of high-grade lesions, in our population.

Human awareness of left space may be disrupted by cerebral lesions to the right hemisphere (hemispatial neglect). Current knowledge on the anatomical bases of this complex syndrome is based on the results of group studies that investigated primarily the best known aspect of the syndrome, which is visual neglect for near extrapersonal (or peripersonal) space. However, another component-neglect for personal space-is more often associated with, than double-dissociated from, extrapersonal neglect, especially, in chronic patients. The present investigation aimed at exploring the anatomical substrate of both extrapersonal and personal neglect by using different advanced methodological approaches to lesion-function correlation. Fifty-two right ischaemic patients were submitted to neuropsychological assessment and in-depth MRI evaluation. The borders of each patient's lesion were delimited onto its own high-resolution anatomical image and then submitted to an automated spatial normalization algorithm. Besides conventional lesion density plots and subtraction analysis, region-based statistical analyses were performed on percentage values of the lesioned tissue also using a new parcellation of the white matter (WM). Data were finally submitted to voxelwise statistical analysis using a recently proposed method (voxel-based lesion-symptom mapping). Results converged in showing that awareness of extrapersonal space is based on the integrity of a circuit of right frontal (ventral premotor cortex and middle frontal gyrus) and superior temporal regions, whereas awareness of personal space is rooted in right inferior parietal regions (supramarginal gyrus, post-central gyrus and especially the WM medial to them). Common but less crucial regions for both neglect sub-types were located in the temporo-peri-Sylvian cortex. We suggest that extrapersonal space awareness critically involves a ventral circuit recently described for the exogenous allocation and reorienting of attention in space. Disruption of personal space awareness, instead, seems to be due to a functional disconnection between regions important for coding proprioceptive and somatosensory inputs, and regions coding more abstract egocentric representations of the body in space. In conclusion, present data strongly support a segregation of personal and extrapersonal spatial awareness in humans, both from a functional and an anatomical point of view.

Summary Cancer increases with age and often arises from the selective clonal growth of altered cells. Thus, any environment favoring clonal growth per se poses a higher risk for cancer development. Using a genetically tagged animal model, we investigated whether aging is associated with increased clonogenic potential. Groups of 4-, 12-, 18-, and 24-month-old Fischer 344 rats were infused (via the portal vein) with 2 × 106 hepatocytes isolated from a normal syngenic 2-month-old donor. Animals deficient in dipeptidyl-peptidase type IV (DPP-IV–) enzyme were used as recipients, allowing for the histochemical detection of injected DPP-IV+ cells. Groups of animals were sacrificed at various times thereafter. No growth of DPP-IV+ transplanted hepatocytes was present after either 2 or 6 months in the liver of rats transplanted at young age, as expected. In striking contrast, significant expansion of donor-derived cells was seen in animals transplanted at the age of 18 months: clusters comprising 7–10 DPP-IV+ hepatocytes/cross-section were present after 2 months and were markedly enlarged after 6 months (mean of 88 ± 35 cells/cluster/cross-section). These results indicate that the microenvironment of the aged liver supports the clonal expansion of transplanted normal hepatocytes. Such clonogenic environments can foster the selective growth of pre-existing altered cells, thereby increasing the overall risk for cancer development associated with aging.

Background/Aims We reported massive liver repopulation by transplanted hepatocytes in rats given retrorsine (RS), a pyrrolizidine alkaloid which blocks proliferation of resident cells. In these studies, molecular alterations induced by RS on hepatocyte cell cycle were investigated. Methods Animals were treated according to the protocol for liver repopulation, i.e. two injections of RS (30mg/kg) followed by two-thirds partial hepatectomy (PH) and were sacrificed at various time points thereafter. Livers were analyzed for the expression of cell cycle-related genes. Results Prior to PH, increased cyclin D1 mRNA and protein levels were found in livers of RS-treated rats. Expression of PCNA was also increased; however, DNA synthesis was not significantly changed. Other cyclins, including cyclin B and cyclin E, were not induced. Cyclin D1 expression increased in controls post-PH and then declined by 48h, as expected. By contrast, no such modulation of cyclin D1 levels was seen in RS group receiving PH and expression remained high at 48h, without mitotic division. Conclusions Exposure to RS is able to block cell cycle progression after cyclin D1 and PCNA induction, but prior to S phase. Such persistent block outside the resting phase may contribute to the selective replacement of resident cells during liver repopulation.

One to four vertical bars were tachistoscopically presented at various eccentricities along the horizontal meridian to patients with right brain damage and neglect (n=7) and to a control group of right brain damaged patients without neglect (n=4). Luminance contrast stimuli and isoluminant chromatic contrast stimuli were used. The patients' task was to report the number of bars. With luminance stimuli the neglect patients' discrimination in the left visual field was reduced, particularly at the most leftward position tested (ca 20 degrees ) where about 20% of the bars were omitted. The loss of information was limited to luminance contrast stimuli. When chromatic contrast stimuli were used, accuracy was comparable in the two hemifields and between groups of patients. The differential pattern of results for chromatic and luminance contrast stimuli is consistent with the hypothesis of a selective deficit of the magno-cellular pathway in neglect.

This study tested the hypothesis that elementary visuo-motor functions involved in visual scanning, as measured by fixation and saccadic tasks, are better in a group of high-level clay target shooters (N=7) than in a control group (N=8). In the fixation task, subject were told to keep fixation as still as possible on a target for 1 min, both in the presence and absence of distracters. For shooters, time did not have an effect on fixation stability, and they had more stable fixation than controls in the distracters condition. Results indicate a difference between groups on both the temporal span of attention and selective attention. In the saccadic task, subjects were asked to saccade, as fast as possible, towards a peripherally displayed target. Two conditions were used: simple reaction to target onset and discrimination between targets and distracters. Shooters had faster saccadic latency to targets than controls in both conditions. Finally, to evaluate the effect of exercise on saccadic latency, we trained one control subject to saccade to a target displayed at a constant spatial position. At the end of the training, saccadic latency reached a value comparable to that recorded in shooters. Learning was largely retinotopic, not showing transfer to untrained spatial positions.

The internal organization of a higher level visual area in the human parietal cortex was mapped. Functional magnetic resonance images were acquired while the polar angle of a peripheral target for a delayed saccade was gradually changed. A region in the superior parietal cortex showed robust retinotopic mapping of the remembered target angle. The map reversed when the direction of rotation of the remembered targets was reversed and persisted unchanged when study participants detected rare target reappearances while maintaining fixation, or when the eccentricity of successive remembered targets was unpredictable. This region may correspond to the lateral intraparietal area in macaque monkeys.
