International audience; Spinal cord injuries remain a real public health issue. However, despite more than half a century of intensive research and numerous clinical trials, the different strategies for repairing the spinal cord remain still inconclusive, even if the diagnosis and the patient care have improved considerably. When Japan and China are embarking on cell therapy trials for chronic lesions, there is currently no treatment that would allow spinal cord injured people to preserve damaged tissue avoiding the secondary expansion of the lesion and recovering of their sensorimotor functions. We thought the stromal vascular fraction (SVF) from fat could be neuroprotective in the acute phase of spinal cord injuries. SVF is extractable in just 2 h and contains mesenchymal and hematopoietic stem cells known for their regenerative and anti-inflammatory properties. Our strategy based on an autologous injection of the SVF within 4 hours after spinal cord injuries. To check our hypothesis, we conducted a preclinical study in adult male rats. Contusions performed at thoracic level using an impactor. From a fat sample of 11.7 +/- 3.5 cc it is possible to extract 8.9 +/- 2.1 million cells of the SVF with 92.2 +/- 1.6% of viability and 77.7 +/- 0.04% of yield. A million cells then injected around injuries. The following 3 months devoted to kinematic analyses of movement, evaluation of sensorimotor recoveries using different behavioral tests (BBB test, Ladder rung walking test, CatWalk…), electrophysiology (evoked potentials, sensorimotor reflexes…), biochemistry (Elisa…), immunohistology (neurofilament labeling…). We present for the first time the results.; Les lésions de la moelle épinière restent un véritable problème de santé publique. Cependant, malgré plus d'un demi-siècle de recherches intensives et de nombreux essais cliniques, les différentes stratégies de réparation de la moelle épinière restent encore peu concluantes, même si le diagnostic et la prise en charge des patients se sont considérablement améliorés. Alors que le Japon et la Chine se lancent dans des essais de thérapie cellulaire pour les lésions chroniques, il n'existe actuellement aucun traitement qui permettrait aux blessés médullaires de préserver les tissus endommagés en évitant l'expansion secondaire de la lésion et en récupérant leurs fonctions sensorimotrices. Nous avons pensé que la fraction vasculaire stromale (FVS) de la graisse pourrait avoir un effet neuroprotecteur dans la phase aiguë des lésions de la moelle épinière. Notre stratégie repose sur une injection autologue de SVF dans les 4 heures suivant les lésions de la moelle épinière. Individuals born after intrauterine growth restriction (IUGR) are at risk of developing cardiovascular diseases (CVDs). Endothelial dysfunction plays a role in the pathogenesis of CVDs; and endothelial colony-forming cells (ECFCs) have been identified as key factors in endothelial repair. In a rat model of IUGR induced by a maternal low-protein diet, we observed an altered functionality of ECFCs in 6-month-old males, which was associated with arterial hypertension related to oxidative stress and stress-induced premature senescence (SIPS). Resveratrol (R), a polyphenol compound, was found to improve cardiovascular function. In this study, we investigated whether resveratrol could reverse ECFC dysfunctions in the IUGR group. ECFCs were isolated from IUGR and control (CTRL) males and were treated with R (1 μM) or dimethylsulfoxide (DMSO) for 48 h. In the IUGR-ECFCs, R increased proliferation (5'-bromo-2'-deoxyuridine (BrdU) incorporation, p < 0.001) and improved capillary-like outgrowth sprout formation (in Matrigel), nitric oxide (NO) production (fluorescent dye, p < 0.01), and endothelial nitric oxide synthase (eNOS) expression (immunofluorescence, p < 0.001). In addition, R decreased oxidative stress with reduced superoxide anion production (fluorescent dye, p < 0.001); increased Cu/Zn superoxide dismutase expression (Western blot, p < 0.05); and reversed SIPS with decreased beta-galactosidase activity (p < 0.001), and decreased p16 ink4a (p < 0.05) and increased Sirtuin-1 (p < 0.05) expressions (Western blot). No effects of R were observed in the CTRL-ECFCs. These results suggest that R reverses long-term ECFC dysfunctions related to IUGR.
