SUMMARY An improved method for reinforcing freeze-fracture replicas, which is especially suitable for tough tissues, is described. For this purpose, polystyrene dissolved in chloroform is used to produce a solid protecting layer of plastic on top of the replica, enabling it to withstand the stresses associated with the thawing and digestion of the tissue with strong acids. The method results in production of large clean replicas from tissues such as skin or peripheral nerve which are difficult to process. The method can also be used profitably for reinforcing other softer and more homogeneous tissues. [ABSTRACT FROM AUTHOR]

Chlorobenzene is a volatile organic compound (VOC) that is widely used as a solvent, degreasing agent and chemical intermediate in many industrial settings. Occupational studies have shown that acute and chronic exposure to chlorobenzene can cause irritation of the mucosa of the upper respiratory tract and eyes. Using in vitro assays, we have shown in a previous study that human bronchial epithelial cells release inflammatory mediators such as the cytokine monocyte chemoattractant protein-1 (MCP-1) in response to chlorobenzene. This response is mediated through the NF-kappaB signaling pathway. Here, we investigated the effects of monochlorobenzene on human lung cells, with emphasis on potential alterations of the redox equilibrium to clarify whether the chlorobenzene-induced inflammatory response in lung epithelial cells is caused via an oxidative stress-dependent mechanism. We found that expression of cellular markers for oxidative stress, such as heme oxygenase 1 (HO-1), glutathione S-transferase pi1 (GSTP1), superoxide dismutase 1 (SOD1), prostaglandin-endoperoxide synthase 2 (PTGS2) and dual specificity phosphatase 1 (DUSP1), were elevated in the presence of monochlorobenzene. Likewise, intracellular reactive oxygen species (ROS) were increased in response to exposure. However, in the presence of the antioxidants N-(2-mercaptopropionyl)-glycine (MPG) or bucillamine, chlorobenzene-induced upregulation of marker proteins and release of the inflammatory mediator MCP-1 are suppressed. These results complement our previous findings and point to an oxidative stress-mediated inflammatory response following chlorobenzene exposure.

Styrene is a volatile organic compound that is widely used as an intermediate in many industrial settings. There are known adverse health effects at environmentally significant concentrations, but little is known about the molecular effect of exposure to styrene at sub-acute toxic concentrations. We exposed human lung epithelial cells, at a wide range of concentrations (1 mg/m(3)-10 g/m(3)), to styrene and analyzed the effects on the proteome level by 2-DE, where 1380 proteins spots were detected and 266 were identified unambiguously by MS. A set of 16 protein spots were found to be significantly altered due to exposure to styrene at environmentally significant concentrations of 1-10 mg/m(3) (0.2-2.3 ppm). Among these, superoxide dismutase as well as biliverdin reductase A could be correlated with the molecular pathway of oxidative stress, while eukaryotic translation initiation factor 5A-1, ezrin, lamin B2 and voltage-dependent anion channel 2 have been reported to be involved in apoptosis. Treatment with styrene also caused the formation of styrene oxide-protein adducts, specifically for thioredoxin reductase 1. These results underline the relevance of oxidative stress as a primary molecular response mechanism of lung epithelial cells to styrene exposure at indoor-relevant concentrations.

Chronic exposure to styrene can result in irritation of the mucosa of the upper respiratory tract. Contact of styrene with epithelial cells stimulates the expression of a variety of inflammatory mediators, including the chemotactic cytokine monocyte chemoattractant protein-1 (MCP-1). To characterise the underlying mechanisms of the induction of inflammatory signals by styrene, we investigated the influence of this compound on the induction of oxidative stress and the activation of the nuclear factor-kappa B (NF-kappaB) signalling pathway in human lung epithelial cells (A549). The results demonstrate that styrene-induced MCP-1 expression, as well as the expression of the oxidative stress marker glutathione S-transferase (GST), is associated with a concentration dependent pattern of NF-kappaB activity. An inhibitor of NF-kappaB, IKK-NBD, and the anti-inflammatory antioxidant N-acetylcysteine (NAC) were both effective in suppressing styrene-induced MCP-1 secretion. In addition, NAC was capable of inhibiting the upregulation of GST expression. Our findings suggest that the activation of the NF-kappaB signalling pathway by styrene is mediated via a redox-sensitive mechanism.

Chlorobenzene is a volatile organic compound that is used as a solvent in many industrial settings and has been shown to be related with irritations of the respiratory tract. Exposure to chlorobenzene induces the release of monocyte chemoattractant protein 1 (MCP-1) by lung epithelial cells, a chemokine involved in inflammatory reactions. To characterize the underlying mechanisms we investigated the influence of chlorobenzene on the activation of two intracellular signalling pathways: the nuclear factor-kappa B (NF-kappa B) and the p38 mitogen-activated protein kinase (MAPK) pathways. Human lung epithelial cells (A549) were stimulated with tumor necrosis factor (TNF)-alpha in the presence or absence of specific inhibitors of NF-kappaB or the p38 MAP kinase and exposed to chlorobenzene using an air-liquid cell culture system. Exposure of lung epithelial cells to chlorobenzene resulted in an activation of NF-kappa B and p38 MAP kinase and a release of the chemokine MCP-1. In the presence of IKK-NBD, a specific NF-kappa B inhibitor, or the inhibitors of the p38 MAP kinase SB 203580 and SB 202190, the chlorobenzene-related MCP-1 release was suppressed, suggesting an involvement of both pathways in the chlorobenzene induced expression of MCP-1. Our data show that the release of MCP-1 following chlorobenzene exposure is dependent on the NF-kappa B and MAPK pathways.

An in vitro cell culture system based on an air/liquid culture technique was developed which allows a direct exposure of cells to volatile chemicals without medium coverage. For the establishment of the experimental system, chlorobenzene was used as a model compound. Chlorobenzene is a volatile organic compound which is mainly used as a solvent. Beside other adverse health effects, chlorobenzene exposure has been shown to be associated with respiratory tract irritations, Th2 differentiation, and allergic sensitizations. Human peripheral blood mononuclear cells (PBMC) and lung epithelial cells (A549) were exposed to chlorobenzene via gas phase for 20 h. Additionally, PBMC were incubated with culture supernatants from exposed lung epithelial cells. High chlorobenzene concentrations (100 g/m(3)) induced IL-8 production in A549 cells, whereby lower concentrations (10 microg/m(3)-1 g/m(3)) stimulated the secretion of the monocyte chemoattractant protein-1 (MCP-1). A direct effect of chlorobenzene on the cytokine secretion of PBMC was not found. However, if PBMC were incubated with culture supernatants of exposed lung cells, an enhanced production of the Th2 cytokine IL-13 was observed. This induction was prevented in the presence of an anti-MCP-1 antibody. Our data suggest that chlorobenzene induces the production of inflammatory mediators in lung cells. The primary chlorobenzene caused release of MCP-1 in lung epithelial cells may secondarily result in a Th2 differentiation in T lymphocytes. These findings may contribute to the understanding of how chlorobenzene mediates the development of inflammatory reactions in the airways and contributes to the development of an allergic reactivity.

Fixed and unfixed human tendons originating from cadavers and postoperation specimens were examined using inclined parallel beams of light in a reflecting mode. Along the tendon, numerous planes, constantly inclined to the axis, were observed edge-on at the surface and within the interior. Their angle of inclination, with respect to the distal end was very nearly +/- 50 degrees. The planes consisted of individual segments arranged in steps which were on average 190 x 50 microns. Similar configurations were also observed with the scanning electron microscope. Using this technique, the segments were identified with collagen bundles turning at a sharp angle with respect to the axis of the tendon at the level of the inclined plane. Crimped planes were found to be irregularly distributed along the tendons. On longer flatter tendons the average distance between planes was in the range of 1-12 mm. On stretching, the inclined pattern disappeared and was rapidly reestablished in the previously observed position when the strain was released. It is suggested that the observed structure forms a mechanism which is responsible for the appearance of the first part of 'foot' region of the tendon's stress-strain diagram.

A regular pattern of parallel reflections with a periodicity of approximately 39 microns has been detected on peripheral nerve fascicles. The reflections have been found to originate from a layer of wavy epineurial collagen fibrils arranged in register around the entire circumference of the fascicle. The waves were observed to be disposed in parallel with the plane of the fascicular sheath and along its axis. The pattern was observed in cut or relaxed fascicles in situ as well as in isolated and split layers of the epineurium. The pattern was not observed on nerve fascicles under tension. An additional structural feature consisting of longitudinally disposed elastic fibres was also detected among the epineurial collagen fibrils. The perineurial collagen associated with sheets of cells was found to be disposed in flattened waves as in the inner layer of the epineurium. The period of the waves, however, was much shorter, in the range of 6-9 microns. From the nature of the wavy structure it can be surmised that on stretching or contraction the sheath length may change, thus accommodating displacement and movement of nerve fibres.

Peripheral nerves in rats fed a diet containing 40% galactose develop endoneurial oedema related to the accumulation of interstitial galactitol and sodium. Biochemical studies of the perineurium in galactose neuropathy indicate an increase in membrane bound Na+/K(+)-ATPase activity. The present study has analysed the intramembraneous particles (IMPs) on the P membrane fracture faces of the perineurium in galactose-fed rats and has demonstrated a significant increase in the density of IMPs. This finding may reflect changes in the perineurial cell membranes in galactose neuropathy, such as increased membrane bound Na+/K(+)-ATPase activity.

Twenty two mid-trimester human fetal tracheae were processed for light microscopy and scanning, transmission and freeze-fracture replication electron microscopy. The tracheal pseudostratified columnar epithelium was found to be composed of four cell types: ciliated in various stages of development, non-ciliated, basal and degenerating cells. The ratio of ciliated to non-ciliated cells was approximately one-to-one. The non-ciliated cells were of two morphological sub-types. Of these, the vast majority contained characteristic secretory granules, approximately 0.5 microns diameter, found in abundance within the apical region of the cell cytoplasm. The second sub-type of the non-ciliated cell, the well differentiated goblet cell, was observed very infrequently. The relatively large number of non-ciliated cells in the fetal tracheal epithelium suggests a transition stage to the adult where the ratio of ciliated to non-ciliated cells is near 5-to-1. The nature of the first type of the non-ciliated cells, which have not been studied in depth previously, remains obscure. Their probable secretory function is discussed and comparisons are made with previously reported cell types showing similar features, in fetal or adult material from the distal airways of animal and human tissues. Approximately 15 to 20% of the epithelial cells show signs of degeneration. The probable causes of this degenerative process are discussed. Other cell types such as brush cells, endocrine and wandering lymphocytes were not observed. Well-differentiated muco-serous glands were observed within the tela submucosa.
