Abstract

Despite decades of rigorous research and numerous clinical trials, Alzheimer's disease (AD) stands as a notable healthcare challenge of this century, with effective therapeutic solutions remaining elusive. Recently, the endocannabinoid system (ECS) has emerged as an essential therapeutic target due to its regulatory role in different physiological processes, such as neuroprotection, modulation of inflammation, and synaptic plasticity. This aligns with previous research showing that cannabinoid receptor ligands have the potential to trigger the functional structure of neuronal and brain networks, potentially impacting memory processing. Therefore, our study aims to assess the effects of prolonged, intermittent exposure (over 90 days) to JWH-133 (0.2 mg/kg) and an EU-GMP certified Cannabis sativa L. (Cannabixir® Medium Flos, 2.5 mg/kg) on recognition memory, as well as their influence on brain metabolism and modulation of the expanded endocannabinoid system in APP/PS1 mice. Chronic therapy with cannabinoid receptor ligands resulted in reduced anxiety-like behavior and partially reversed the cognitive deficits. Additionally, a reduction was observed in both the number and size of Aβ plaque deposits, along with decreased cerebral glucose metabolism, as well as a decline in the expression of mTOR and CB2 receptors. Furthermore, the study revealed enlarged astrocytes and enhanced expression of M1 mAChR in mice subjected to cannabinoid treatment. Our findings highlight the pivotal involvement of the extended endocannabinoid system in cognitive decline and pathological aspects associated with AD, presenting essential preclinical evidence to support the continued exploration and assessment of cannabinoid receptor ligands for AD treatment. Abstract

Recently, numerous side effects of synthetic drugs have lead to using medicinal plants as a reliable source of new therapy. Pain is a global public health problem with a high impact on life quality and a huge economic implication, becoming one of the most important enemies in modern medicine. The medicinal use of plants as analgesic or antinociceptive drugs in traditional therapy is estimated to be about 80% of the world population. The Lamiaceae family, one of the most important herbal families, incorporates a wide variety of plants with biological and medical applications. In this study, the analgesic activity, possible active compounds of Lamiaceae genus, and also the possible mechanism of actions of these plants are presented. The data highlighted in this review paper provide valuable scientific information for the specific implications of Lamiaceae plants in pain modulation that might be used for isolation of potentially active compounds from some of these medicinal plants in future and formulation of commercial therapeutic agents.

Abstract

Alzheimer's disease, a major and increasing global health challenge, is an irreversible, progressive form of dementia, associated with an ongoing decline of brain functioning. The etiology of this disease is not completely understood, and no safe and e ective anti-Alzheimer's disease drug to prevent, stop, or reverse its evolution is currently available. Current pharmacotherapy concentrated on drugs that aimed to improve the cerebral acetylcholine levels by facilitating cholinergic neurotransmission through inhibiting cholinesterase. These compounds, recognized as cholinesterase inhibitors, o er a viable target across key sign domains of Alzheimer's disease, but have a modest influence on improving the progression of this condition. In this paper, we sought to highlight the current understanding of the cholinergic system involvement in Alzheimer's disease progression in relation to the recent status of the available cholinesterase inhibitors as e ective therapeutics. Abstract

Aim: The results presented here are a part of an exploratory positron emission tomography (PET)/magnetic resonance imaging (MRI) study in a rat model of cardiorenal syndrome (CRS), where we have used parametric quantification in target organs (heart, kidney, vessels), based on image analysis. As an incremental approach to the final goal of the experiment, we aimed at obtaining a valid technique for approximating the glomerular filtration rate (GFR) using F-18-FDG tracer dynamics on control lot versus CRS induced by heart failure (HF). Method: We studied a number of 12 male Wistar rats, divided into 2 lots: control (L1) and heart failure (L2). Heart failure was induced by intraperitoneal doxorubicin administration for a period of 6 weeks. F-18- FDG PET/MRI scans (nanoScan PET/MRI, Mediso) were acquired at week 0 for L1 and at week 6 for L2. Each rat underwent a succession of MRI examination (T1 weighted), injection of F-18-FDG and PET data acquisition, in the same session. Manually defined regions of interest were drawn on MRI images for kidneys and left ventricular cavity, which were used for volumetric assessment and delineation of VOIs on PET frames. Weight based standardized uptake values (SUV) for F-18-FDG were calculated for each frame of the dynamic PET study, with the 3 to 20 minute interval being selected for data analysis. PET derived GFR estimation was calculated based on the following formula: GFR: Kiright[min-1]*Vright kidney[ml]+ Kileft[min-1]*Vleft kidney[mL], where Ki is approximated as the slope of the linear regression of arterial tracer concentration by taking the renal activity as independent variable. Results: Results are reported as mean ± standard error of the mean. The variation in arterial concentration explained by renal tracer dynamics of the groups was R2 L1= 0.97, R2 L2= 0.99. Estimated GFR for the control group, compared to literature data (1.38±0.1 mL/min), was within 1.26±0.23 mL/ min. L2 group showed a departure from the control group with values of 0.88±0.14 mL/min. Conclusion: Our results are in agreement with the reported literature for healthy Wistar rats and it provides us with an intra-experiment assessment method for kidney dysfunction by F-18-FDG PET/MRI.
