Abstract

Background Persistent sensorimotor impairments after stroke can negatively impact quality of life. The hippocampus is vulnerable to poststroke secondary degeneration and is involved in sensorimotor behavior but has not been widely studied within the context of poststroke upper-limb sensorimotor impairment. We investigated associations between non-lesioned hippocampal volume and upper limb sensorimotor impairment in people with chronic stroke, hypothesizing that smaller ipsilesional hippocampal volumes would be associated with greater sensorimotor impairment. Methods and Results Cross-sectional T1-weighted magnetic resonance images of the brain were pooled from 357 participants with chronic stroke from 18 research cohorts of the ENIGMA (Enhancing NeuoImaging Genetics through Meta-Analysis) Stroke Recovery Working Group. Abstract

Lesion load of the corticospinal tract (CST-LL), a measure of overlap between a stroke lesion and the CST, is one of the strongest predictors of motor outcomes following stroke. CST-LL is typically calculated by using a probabilistic map of the CST originating from the primary motor cortex (M1). However, higher order motor areas also have projections that contribute to the CST and motor control. In this retrospective study, we examined whether evaluating CST-LL from additional motor origins is more strongly associated with post-stroke motor severity than using CST-LL originating from M1 only. We found that lesion load to both the ventral premotor (PMv) cortex and M1 were more strongly related to stroke motor severity indexed by Fugl-Meyer Assessment cut-off scores than CST-LL of M1 alone, suggesting that higher order motor regions add clinical relevance to motor impairment.

Abstract

Accurate lesion segmentation is critical in stroke rehabilitation research for the quantification of lesion burden and accurate image processing. Current automated lesion segmentation methods for T1-weighted (T1w) MRIs, commonly used in stroke research, lack accuracy and reliability. Manual segmentation remains the gold standard, but it is time-consuming, subjective, and requires neuroanatomical expertise. We previously released an open-source dataset of stroke T1w MRIs and manually-segmented lesion masks (ATLAS v1.2, N = 304) to encourage the development of better algorithms. However, many methods developed with ATLAS v1.2 report low accuracy, are not publicly accessible or are improperly validated, limiting their utility to the field. Here we present ATLAS v2.0 (N = 1271), a larger dataset of T1w MRIs and manually segmented lesion masks that includes training (n = 655), test (hidden masks, n = 300), and generalizability (hidden MRIs and masks, n = 316) datasets. Algorithm development using this larger sample should lead to more robust solutions; the hidden datasets allow for unbiased performance evaluation via segmentation challenges. We anticipate that ATLAS v2.0 will lead to improved algorithms, facilitating large-scale stroke research.

Abstract

The goal of the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Stroke Recovery working group is to understand brain and behavior relationships using well-powered meta- and mega-analytic approaches. ENIGMA Stroke Recovery has data from over 2,100 stroke patients collected across 39 research studies and 10 countries around the world, comprising the largest multisite retrospective stroke data collaboration to date. This article outlines the efforts taken by the ENIGMA Stroke Recovery working group to develop neuroinformatics protocols and methods to manage multisite stroke brain magnetic resonance imaging, behavioral and demographics data. Specifically, the processes for scalable data intake and preprocessing, multisite data harmonization, and large-scale stroke lesion analysis are described, and challenges unique to this type of big data collaboration in stroke research are discussed. Finally, future directions and limitations, as well as recommendations for improved data harmonization through prospective data collection and data management, are provided.

Up to two-thirds of stroke survivors experience persistent sensorimotor impairments. Recovery relies on the integrity of spared brain areas to compensate for damaged tissue. Deep grey matter structures play a critical role in the control and regulation of sensorimotor circuits. 17. Working memory enhancement using real-time phase-tuned transcranial alternating current stimulation.

Abstract

Prior work has shown that transcranial alternating current stimulation (tACS) of parietooccipital alpha oscillations (8–14 Hz) can modulate working memory (WM) performance as a function of the phase lag to endogenous oscillations. However, leveraging this effect using real-time phase-tuned tACS has not been feasible so far due to stimulation artifacts preventing continuous phase tracking. We aimed to develop a system that tracks and adapts the phase lag between tACS and ongoing parietooccipital alpha oscillations in real-time. We hypothesized that such real-time phase-tuned tACS enhances working memory performance, depending on the phase lag. We developed real-time phase-tuned closed-loop amplitude-modulated tACS (CLAM-tACS) targeting parietooccipital alpha oscillations. CLAM-tACS was applied at six different phase lags relative to ongoing alpha oscillations while participants (N = 21) performed a working memory task. To exclude that behavioral effects of CLAM-tACS were mediated by other factors such as sensory co-stimulation, a second group of participants (N = 25) received equivalent stimulation of the forehead. WM accuracy improved in a phase lag dependent manner (p = 0.0350) in the group receiving parietooccipital stimulation, with the strongest enhancement observed at 330° phase lag between tACS and ongoing alpha oscillations (p = 0.00273, d = 0.976). Moreover, across participants, modulation of frontoparietal alpha oscillations correlated both in amplitude (p = 0.0248) and phase (p = 0.0270) with the modulation of WM accuracy. No such effects were observed in the control group receiving frontal stimulation. Our results demonstrate the feasibility and efficacy of real-time phase-tuned CLAM-tACS in modulating both brain activity and behavior, thereby paving the way for further investigation into brain-behavior relationships and the exploration of innovative therapeutic applications. • TACS is phase-tuned to alpha oscillations in real-time during stimulation. • TACS enhances working memory performance. • Working memory effects are associated with frontoparietal alpha synchrony. • No phase-dependent modulation in active control condition. 32. How can an educational chatbot's feedback influence human attention?

Abstract

Educational chatbots have been shown to be useful assistants in computer-supported learning settings. However, how does feedback of an educational chatbot impact on the learner's attention? Thus, this paper proposes a study to measure changes in human attention when learning by using an educational chatbot Liza that is intended to improve human reasoning ability. In total, 18 participants participated in the study and had a conversation with Liza. During the interaction with Liza, the attention of the study participants was measured using a mobile electroencephalogram (EEG) device. Three findings have been determined based on statistics methods. First, it was found that there was a significant attention effect occurring in 54% of the times, after the educational chatbot showed feedback and the attention measurement took place over the length of a task. Second, when differentiating the type of feedback, positive feedback had a significant effect in 71 of these 137 cases (51.82%) and negative feedback had a significant effect in 66 of the 137 cases (48.18%). Third, statistical results showed that there was no significant difference in attention at the significant level of 0.05 during the 10 seconds before, and 10 seconds after positive feedback is received. Die Alkoholsucht ist ein global verbreitetes Phänomen und kennzeichnet sich durch eine Transition von kontrolliertem zu zwanghaftem Alkoholkonsum.Die Tendenz, einem neutralen Reiz eine Anreizwirkung zuzuschreiben, ist individuell unterschiedlich und stellt einen Risikofaktor für die Entwicklung einer Abhängigkeit dar.Zur Entwicklung spezifischer Präventionsstrategien ist ein besseres Verständnis der Zusammenhänge zwischen der Anreizsalienz und Alkoholabhängigkeit erforderlich.Der Übergang von mäßigem zu zwanghaftem Alkoholkonsum wurde durch das Modell des Alkoholentzugseffekts simuliert, das den menschlichen Alkoholrückfall nachahmt.Die Ratten erhielten freiwilligen Zugang zu verschieden Alkohollösungen mit wiederholten Deprivations- und Alkoholphasen.Die Ratten durchliefen zusätzlich den Pavlovian Conditioned Approach getestet, welcher die individuellen Tendenzen auf einen bedingten Reiz/Belohnung quantifiziert.Während des letzten ADE-Zyklus wurde mit Geschmacksverfälschung zwanghaftes Trinken ermittelt.Nach der Identifizierung zuverlässiger Prädiktoren für Suchtverhalten wurde präventive Neurostimulation durchgeführt, um die Tendenz der Tiere alkoholbezogenen Reizen eine motivationale Bedeutung beizumessen, zu beeinflussen, und die Manifestation eines Abhängigkeitsverhaltens zu verhindern. Weibchen tranken mehr Alkohol als Männchen und zeigten ST Verhalten im PavCA, während Männchen GT aufwiesen.Die Anwendung von transkranieller Gleichstromstimulation während PavCA führte zu mehr GT-Verhalten bei stimulierten Ratten.Frühe tDCS während des Trinkens hatte keinen Einfluss auf das akute und das Langzeit-Trinken. Die Ergebnisse zeigen eine komplexere Beziehung zwischen Anreizsalienz und Alkoholsucht und unterstreichen,individuelle Unterschiede und beide Geschlechter in der präklinischen Forschung zu berücksichtigen., The consequences of alcohol dependence cause the global deaths of million people yearly.The ability of the environment can trigger dependent behavior and promote drinking.The tendency to attribute incentive salience to cues differs between subjects.By forming a cue-alcohol association, neutral cues receive motivational value,thereby predicting the likelihood of alcohol reward occurrence,known as Pavlovian learning.Understanding the relationship between incentive salience and alcohol addiction help inform treatment strategies.We study the relationship between incentive salience and alcohol addiction.The transition from moderate to compulsive alcohol intake can be captured by the alcohol deprivation effect rat model (mimics alcohol relapse in humans).Rats were given voluntary access to alcohol solutions with repeated abstinence/reintroduction phases.Further,rats were tested in the PavCA,which quantifies individual tendency toward a conditional cue and the reward, thus allowing to trace the process of attributing incentive salience to rewardcues.During the final ADE cycle,rats underwent a bitter taste adulteration test to assess for compulsive-like behavior.After identifying reliable predictors of addictive behavior,preventive tDCS was performed to influence the tendency of animals to attach motivational importance to alcohol-related stimuli,and to prevent the manifestation of alcohol addictive behavior.Females drank more alcohol than males and exhibited more ST behavior in the PavCA, whereas males showed GT behavior.PavCA phenotypes emerged early and remained stable.The application of tDCS during PavCA results in high GT numbers in stimulated rats.Early tDCS on drinking does not affect acute or long-term drinking.Our findings indicate a complex relationship between incentive salience and alcohol addiction and emphasize the importance of considering individual differences and both sexes in preclinical research.
