Each year, hundreds of millions of people are infected with arboviruses such as dengue, yellow fever, chikungunya, and Zika, which are all primarily spread by the notorious mosquito Aedes aegypti. Traditional control measures have proven insufficient, necessitating innovations. In response, here we generate a next-generation CRISPR-based precision-guided sterile insect technique (pgSIT) for Ae. aegypti that disrupts genes essential for sex determination and fertility, producing predominantly sterile males that can be deployed at any life stage. Using mathematical models and empirical testing, we demonstrate that released pgSIT males can effectively compete with, suppress, and eliminate caged mosquito populations. This versatile species-specific platform has the potential for field deployment to effectively control wild populations of disease vectors.

Background Matings between male Aedes aegypti mosquitoes infected with w AlbB strain of Wolbachia and wildtype females yield non-viable eggs. We evaluated the efficacy of releasing w AlbB-infected Ae. aegypti male mosquitoes to suppress dengue. Methods We specified the protocol of a two-arm cluster-randomized test-negative controlled trial (cRCT) and emulated it using a nationally representative test-negative/positive database of individuals reporting for febrile illness to any public hospital, general practitioner or polyclinic. We retrospectively built a cohort of individuals who reside in Wolbachia locations vs a comparator control group who do not reside in Wolbachia locations, using a nationally representative database of all individuals whom report for febrile illness and were tested for dengue at the Environmental Health Institute/hospital laboratories/commercial diagnostic laboratories, through general practitioner clinic, polyclinic or public/private hospital from epidemiological week (EW) 1 2019 to EW26 2022. We emulated a constrained randomization protocol used in cRCTs to balance dengue risk between intervention and control arms in the pre-intervention period. We used the inverse probability weighting approach to further balance the intervention and control groups using a battery of algorithmically selected sociodemographic, environmental and anthropogenic variables. Intention-to-treat analyses were conducted to estimate the risk reduction of dengue given Wolbachia exposure. Results Intention-to-treat analyses revealed that, compared with controls, Wolbachia releases for 3, 6 and ≥12 months was associated to 47% (95% confidence interval: 25–69%), 44% (33–77%) and 61% (38–78%) protective efficacy against dengue, respectively. When exposed to ≥12 months of Wolbachia releases, protective efficacies ranged from 49% (13–72%) to 77% (60–94%) across years. The proportion of virologically confirmed dengue cases was lower overall in the intervention arm. Protective efficacies were found across all years, age and sex subgroups, with higher durations of Wolbachia exposure associated to greater risk reductions of dengue. Sterile insect technique (SIT) is a useful strategy for preventing and mitigative establishment of invasive insect species. SIT of the pest tephritid Mediterranean fruit fly, Ceratitis capitata (Wiedemann, 1824) WiedemannWiedemann , has been effective in preventing population establishment in vulnerable agricultural areas of the United States. However, irradiation-induced sterilization can have detrimental impacts resulting in reduced performance metrics. Mediterranean fruit fly males reared for SIT have been shown to have differences in their microbiomes relative to other population sources, which has been postulated to be a factor in how well flies compete with wild conspecifics. To identify baseline performance metrics on the effects of irradiation on the gut microbiome of mass-reared flies in Hawai'i, a study was performed to assess performance metrics and microbiome (bacterial 16S rRNA) variation across multiple timepoints. Mediterranean fruit fly pupae were selected from mass-reared trays intended for release, and paired samples were either irradiated or remained as controls and transported to the laboratory for evaluation. Irradiated flies exhibited fewer successful fliers, more rapid mortality rates, and were less active relative to control nonirradiated flies. Contrary to initial expectations, irradiation did not exert substantial impacts on the composition or diversity of bacterial reads. Samples were primarily comprised of sequences classified as Klebsiella and there were low levels of both read and taxonomic diversity relative to other 16S surveys of medfly. BACKGROUND: An innovative version of the sterile insect technique (SIT) for pest control, called boosted SIT, relies on the use of sterile males coated with a biocide to control a target wild pest population of the same species. The objective of the present study was to assess the relevance of such technology to control the fruit fly Bactrocera dorsalis and fruit losses in mango orchards using. An agent‐based simulation model named BOOSTIT was used to explore the reduction of fruit losses thank to sterile male fruit flies control and economic benefits according to different strategies of sterile male release. The simulation considered a landscape of 30.25 ha made up of four mango orchards. RESULTS: The SIT and the boosted SIT reduced fruit losses when releases were made before the mango fruiting period. According to model simulations, releases should be performed at least seven times at 2‐week intervals and with a sterile/wild male ratio of at least 10:1. Considering the benefit/cost ratio (BCR), few releases should be done with a late start date. The BCR showed economic gains from the two control methods, the number of saved fruits and BCR being higher for SIT. CONCLUSION: Our simulations showed that SIT would have better results than the boosted SIT to contribute to an effective control of Bactrocera dorsalis at the scale of a small landscape. We highlight the need for laboratory studies of other types of pathogen to find a suitable one with higher incubation time and lower cost. © 2024 The Author(s). Simple Summary: For the sterile insect technique to be successful, sterile male mosquitoes need to outcompete fertile males and mate with wild females. Some female insects, however, have been shown to be able to select for "better sperm" to secure high-quality offspring. This study used stable isotopes to mark the sperm of sterile and fertile males to find any evidence of female tiger mosquitoes selecting the fertile sperm over the sterile one when mated by both types of males. Although this study does not prove that a complex mechanism of sperm selection does not occur, we were not able to find any evidence that double-mated Aedes albopictus females favor fertile sperm for egg fertilization. The key to success in the application of the sterile insect technique (SIT) relies on the ability of released, sterile males to outcompete their fertile wild male counterparts to mate with wild females. However, many insect species exhibit multiple-mating behavior, which can be a way for females to select paternity for their progeny. This study aims to recognize the consequences of potential double-matings during an SIT program and to detect any evidence of sperm selection favoring sperm from fertile mates. This report provides a descriptive analysis of the storage and use of sperm by female Aedes albopictus. Stable isotopes were used to mark the sperm of fertile and sterile males. Mated females were allowed to oviposit before dissecting the spermathecae to link the presence of each type of sperm to the sterility of the eggs laid. It was found that sperm in females inseminated by both males was distributed in the three spermathecae with no obvious pattern, mostly mixed but also separately, and no evidence of any mechanism for sperm selection, sperm precedence, or sperm competition in Ae. albopictus females could be found. The fact that only a few double-mated females were double-inseminated and could also produce semi-sterile eggs, together with the finding that the sperm of sterile males appeared to be no less viable than that of fertile males, is an encouraging outcome for SIT approaches. Background: Bovine tuberculosis (bTB) is a chronic disease that results from infection with any member of the Mycobacterium tuberculosis complex. Infected animals are typically diagnosed with tuberculin-based intradermal skin tests according to World Organization of Animal Health which are presently in use. However, tuberculin is not suitable for use in BCG-vaccinated animals due to a high rate of false-positive reactions. Peptide-based defined skin test (DST) antigens have been identified using antigens (ESAT-6, CFP-10 and Rv3615c) which are absent from BCG, but their performance in buffaloes remains unknown. To assess the comparative performance of DST with the tuberculin-based single intradermal test (SIT) and the single intradermal comparative cervical test (SICCT), we screened 543 female buffaloes from 49 organized dairy farms in two districts of Haryana state in India. Results: We found that 37 (7%), 4 (1%) and 18 (3%) buffaloes were reactors with the SIT, SICCT and DST tests, respectively. Of the 37 SIT reactors, four were positive with SICCT and 12 were positive with the DST. The results show that none of the animals tested positive with all three tests, and 6 DST positive animals were SIT negative. Together, a total of 43 animals were reactors with SIT, DST, or both, and the two assays showed moderate agreement (Cohen's Kappa 0.41; 95% Confidence Interval (CI): 0.23, 0.59). In contrast, only slight agreement (Cohen's Kappa 0.18; 95% CI: 0.02, 0.34) was observed between SIT and SICCT. Using a Bayesian latent class model, we estimated test specificities of 96.5% (95% CI, 92–99%), 99.7% (95% CI: 98–100%) and 99.0% (95% CI: 97–100%) for SIT, SICCT and DST, respectively, but considerably lower sensitivities of 58% (95% CI: 35–87%), 9% (95% CI: 3–21%), and 34% (95% CI: 18–55%) albeit with broad and overlapping credible intervals. Conclusion: Taken together, our investigation suggests that DST has a test specificity comparable with SICCT, and sensitivity intermediate between SIT and SICCT for the identification of buffaloes suspected of tuberculosis. Our study highlights an urgent need for future well-powered trials with detailed necropsy, with immunological and microbiological profiling of reactor and non-reactor animals to better define the underlying factors for the large observed discrepancies in assay performance, particularly between SIT and SICCT. The sterile insect technique (SIT) is used for the management of tephritid fruit fly pests. The South American fruit fly, Anastrepha fraterculus (Wiedemann) (Diptera: Tephritidae), is one potential pest to be targeted by means of SIT. The success of SIT depends, to a large extent, on the sexual performance of sterile males. Various approaches have been evaluated with the aim of improving their sexual performance. These include the exposure to plant‐derived compounds and/or the provision of protein sources in the adults' diet capable of stimulating male mating success. The present study aimed to determine whether exposure to volatiles of Citrus limon (L.) Burm. F. (Rutaceae) essential oil and limonene confers a mating advantage to A. fraterculus laboratory males fed two distinct dietary regimes when competing with wild males for wild females under field cage conditions. Dietary regimes were, one, with non‐hydrolysed brewer's yeast and sugar (1:3 ratio), and the other with brewer's yeast hydrolysate enzymatic and sugar (1:12 ratio). The effect was evaluated in four variables associated with mating success: number of copulas obtained, latency to mate, copula duration, and copula location. Exposure to volatiles did not affect the number of matings achieved, irrespective of the diet given to the males. When laboratory males were fed with brewer's yeast hydrolysate, the effect of volatile exposure was shown in latency to mate, copula duration, and copula location. When the laboratory males were fed with non‐hydrolysed brewer's yeast, the effect of volatile exposure was shown only in copula duration. Laboratory males fed brewer's yeast hydrolysate achieved the same number of matings as wild males, whereas laboratory males fed non‐hydrolysed brewer's yeast had lower performance. Simple Summary: The brown marmorated stink bug, Halyomorpha halys, native to Asia, is now an insect pest in several countries. The sterile insect technique (SIT), involving the sterilization of insects by ionizing radiation and field release, is one method to control insect pest. This study examines the ultrastructure of H. halys spermiogenesis in both irradiated and wild males. Typical hemipteran sperm features were observed, such as mitochondrial derivative–axoneme bridges, the absence of accessory bodies, crystalline inclusions in mitochondrial derivatives, an acrosome with packed tubules, and a plaque-shaped microtubular organizing center (MTOC) in the centriolar region. Occasionally, two centrioles were observed in spermatids, with one disappearing later—a novel feature for insect spermiogenesis. In irradiated adults, cysts exhibited inconsistent radiation effects, including disorganized sperm arrangement, incomplete sperm cell divisions, multiple organelle copies within cells, failure to reabsorb cytoplasm, and a lack of axonemes. Additionally, rod-shaped viruses or virus-like particles were found in vasa deferentia, regardless of irradiation. Halyomorpha halys (Heteroptera: Pentatomidae) is an insect pest native to Asia that has spread over the last two decades to most of the North America, parts of South America, Europe and North Africa. Its impact is significant as it can feed on more than 300 host plants, rendering affected fruits and vegetable crops unsellable or of lower quality. Various chemical and biological methods have been used to control this pest, with varying degrees of success. The sterile insect technique (SIT) is a pest control method involving the sterilization of insects via ionizing radiation and their subsequent mass release into the field. In the present contribution, the spermiogenesis of H. halys was studied from an ultrastructural point of view in both irradiated and non-irradiated adult males. In both cases, we observed ultrastructural characteristics typical of hemipteran sperm cells: bridges connecting the mitochondrial derivatives and the axonemal microtubules, the absence of accessory bodies, and the presence of two or three crystalline inclusions within the mitochondrial derivatives, an acrosome composed of tightly packed tubules, and an atypical, plaque-shaped microtubular organizing center (MTOC) in the centriolar region. Moreover, in the same region, we seldom observed the presence of two centrioles in the spermatids, one of which disappeared at a later stage of maturation. This feature is a novelty for insect spermiogenesis. The cysts of irradiated adults were not all uniformly affected by the radiation. However, irradiated cysts sometimes exhibited a general disorganization of sperm arrangement, incomplete divisions of sperm cells resulting in multiple copies of the same organelle within the same cell, failure to reabsorb the cytoplasm, and the lack of axonemes. In this study, we employ a Random Forest approach to estimate sea ice thickness (SIT) using Fengyun-3E (FY-3E) and Soil Moisture Ocean Salinity (SMOS) data. This method relies on four input parameters: incidence angle (θ), reflectivity (Γ), sea ice salinity (S), and sea ice temperature (T). In addition, FY-3E can receive both Global Positioning System (GPS) and Beidou Navigation Satellite System (BDS) reflected signals. Evaluation for the Arctic region based on data spanning from October 2022 to April 2023 reveals that the proposed models trained on GPS and BDS signals from FY-3E achieve high consistency and low error. Take GPS signals as an example, coefficients of determination are 0.97 and 0.91 and mean absolute errors are 0.019 m and 0.032 m for the training and test sets, respectively. In general, SIT inversion based on GPS signals slightly exhibits a higher accuracy than that based on BDS signals, but both approaches display high performances. The areas with the highest accuracy of SIT estimation based on GPS and BDS signals are the Shelikhov Bay and the Okhotsk Sea, followed by the Bering Sea and the Bering Strait. Drosophila suzukii (D. suzukii), commonly known as the spotted wing drosophila, is a highly invasive crop pest that is difficult to control using chemical insecticides. To address the urgent need for alternative and more sustainable control strategies, the sterile insect technique (SIT) is improved, which involves the release of sterilized male insects to mate with fertile conspecifics, thereby reducing the size of the pest population in the subsequent generation. The three critical aspects that influence the success of SIT programs in D. suzukii are addressed. First, an accurate and nondestructive method is established to determine the sex of individual insects based on the differential weight of male and female pupae. Second, conditions for X‐ray sterilization are systematically tested and an optimal dose (90 kV/40 Gy) is identified that ensures the efficient production of sterile D. suzukii for release. Finally, the inherent thermosensitivity of D. suzukii males is exploited to develop a temperature‐based sterilization technique, offering an alternative or additional SIT method for this pest. These advances will contribute to the development of a comprehensive and effective strategy for the management of D. suzukii populations, reducing their impact on agriculture and helping to safeguard crop yields. Background & objectives: Biological approaches for vector mosquito control such as sterile insect technique (SIT) requires sterilization of male mosquitoes through specific radiation doses for sterility induction in males. Under the SIT program, the males used must be compatible with the wild males in vigor, flight and selection of mate. Much of this potential is determined by the diet provided in the larval stages and optimizing the irradiation doses that cause complete sterility but have very minimal effect on the lives of irradiated males. The current study is designed to evaluate gamma radiation doses for inducing sterility in male Aedes aegypti mosquitoes with a genomic background from Pakistan (Swat district) and to assess the effects of radiation exposure on the competency of irradiated males and other life traits of irradiated mosquitoes. Methods: Sterilization of 17-hour-old male pupae (groups of 50 pupa/cup in three replicates) of the Ae. aegypti Swat strain was conducted using radiation doses of 30, 60, 70, 90, and 105 gray (Gy) from (Co60) at the Nuclear Institute for Food and Agriculture (NIFA) in Peshawar, Pakistan. Post-irradiated pupae were released in adult cages. Virgin females of the same cohort were released (1:1) to mate with the irradiated males. Mortality, longevity, mating competitiveness of males, and female fecundity were recorded. Results: An average of 71 eggs per female was recorded in control with 86% hatch rate. Individual females mated with a 60 Gy treated males produced 60 ± 0.6 eggs per female with 17% hatch rate, and those mated with 70 Gy males produced 42 ± 0.01 eggs with a nil hatch rate, whereas females mated with males treated with ≥70 Gy doses did not reproduce to next generation. Females in groups of 50 mated with 60, 70 Gy treated males (equal pairs), produced 369±1.3 and 98±0.01 eggs with 15% and zero hatch rate. Significant dose dependent reduction in longevity was observed for >30 Gy doses. The matting competence of irradiated males was about half that of un-irradiated males. Interpretation & conclusion: Aedes aegypti with a Pakistani genomic background treated with a ≥70 Gy dose of gamma radiation induced complete sterility in males and provided the first-step foundation for SIT application in Pakistan. Further extensive studies are required to optimize the SIT techniques so that fully sterile males with very minor quality changes can be produced on large scales for field trials.

Simple Summary: Aedes, Culex, and Anopheles mosquitoes are the most prolific arthropod vectors of viral and parasitic agents of debilitating and lethal diseases in humans and animals. Despite some success in integrated pest management programs to control vectors, mosquito-borne diseases, such as dengue and dengue hemorrhagic fever, yellow fever, chikungunya, West Nile, and Zika, and parasitic diseases, such as malaria, lymphatic filariasis, and river blindness, continue to threaten the health and well-being of half the world's population, many of whom live in economically and medically challenged societies. The perpetual problem inflicted by vector-borne diseases is compounded by the selection for resistance to synthetic pesticides, globalization, and climate change. The latter appears to be the most significant factor implicated in the geographic expansion of mosquitoes. Here, we present a review of these challenges and highlight traditional vector control strategies that employ synthetic pesticides, and "green" eco-friendly technologies that include SIT, IIT, RIDL, CRISPR/Cas9/Cas13 gene drive systems, and biological control, with an emphasis on Lysinibacillus sphaericus and Bacillus thuringiensis subsp. israelensis (Bti). Mosquitoes are the most notorious arthropod vectors of viral and parasitic diseases for which approximately half the world's population, ~4,000,000,000, is at risk. Integrated pest management programs (IPMPs) have achieved some success in mitigating the regional transmission and persistence of these diseases. However, as many vector-borne diseases remain pervasive, it is obvious that IPMP successes have not been absolute in eradicating the threat imposed by mosquitoes. Moreover, the expanding mosquito geographic ranges caused by factors related to climate change and globalization (travel, trade, and migration), and the evolution of resistance to synthetic pesticides, present ongoing challenges to reducing or eliminating the local and global burden of these diseases, especially in economically and medically disadvantaged societies. Abatement strategies include the control of vector populations with synthetic pesticides and eco-friendly technologies. These "green" technologies include SIT, IIT, RIDL, CRISPR/Cas9 gene drive, and biological control that specifically targets the aquatic larval stages of mosquitoes. Regarding the latter, the most effective continues to be the widespread use of Lysinibacillus sphaericus (Ls) and Bacillus thuringiensis subsp. israelensis (Bti). Here, we present a review of the health issues elicited by vector mosquitoes, control strategies, and lastly, focus on the biology of Ls and Bti, with an emphasis on the latter, to which no resistance has been observed in the field. Tuta absoluta (Meyrick) is a destructive pest of tomato crops. The cryptic nature of larvae often leads to low control efficacy. Sterile insect technique (SIT) has the potential to be an alternative management option for T. absoluta. In this study, the optimal dose for T. absoluta sterility was determined by estimating the effect of gamma radiation on parental generation (F0) and their descendants (first and second generation, F1 and F2). Adult emergence was not affected when T. absoluta pupae were exposed to gamma radiation at doses of 100~400 Gy. However, increasing doses of radiation increased deformed individuals and shortened their longevity. Furthermore, as the dose increased, F0 females showed decreased fecundity and fertility, and they were completely sterile when exposed to 300 Gy. The survival and sex ratio of females declined significantly in F1 of irradiated males with increasing doses. Moreover, we found that 300 Gy of gamma radiation did not affect the mating competitiveness of F0 males. Interestingly, this treatment caused a considerable reduction in the number of descendants and caused inherited sterility in F1 by lowering their fecundity and fertility. Therefore, the optimal radiation dose was 300 Gy, and the release of sterile adults may suppress T. absoluta population growth in the field. BACKGROUND: Improving the mating competitiveness and survival of sterile males are direct means to increase the effectiveness of the sterile insect technique (SIT). Some insecticide growth regulators, such as the juvenile hormone analogue (JHA) methoprene, have been used to improve the mating competitiveness of male tephritid flies by reducing their sexual maturation period. However, the application of methoprene reduces fly resistance to stress and decreases survival. Here, we compared the effects of methoprene and pyriproxyfen (PPF), another JHA, in Anastrepha ludens males. PPF is an insect growth regulator that exhibits higher negative effects on the larval molting process than methoprene or natural juvenile hormone. Both compounds were administered at two doses (0.05% and 0.10%) via the male diet immediately after emergence. RESULTS: Our results show that both PPF and methoprene reduced male sexual maturation. However, PPF‐treated males exhibited a shorter maturation period and obtained more matings at a given age than methoprene‐treated males. No significant differences were observed between the two PPF doses tested (0.05% and 0.10%). Male survival was equally reduced by the two compounds. ObjectiveThe aim of this study was to evaluate the effect of sprint interval training (SIT) and [high intensive interval training (HIIT)] carried out during the cool-down period of the physical education classes on body composition, blood pressure variables (BP) and pulse rate (PR), and cardiorespiratory fitness of adolescents who are overweight and obese, and to compare the differences in enjoyment in response to SIT vs. HIIT.MethodsFor this randomized controlled trial, forty-five adolescents were recruited from a high school and were randomly placed into three groups. SIT and HIIT trained for 8 weeks, twice a week, for 12 min/session. Experimental group (EG) 3 was the control, and they maintained their regular physical education class schedule. The SIT group performed 6 sets of 60 s of work (90-95%HRmax) / 60 s of rest (50-55%HRmax), and the HIIT group performed 3 sets of 2 min of work (80-85%HRmax) / 2 min of rest (50-55%HRmax).ResultsBoth experimental groups showed a significant improvement in fat mass (FM) (%) and trunk FM (kg). In addition, EG2 reported a significance improvement in lean mass (kg), blood pressure BP (mmHG), systolic blood pressure (SBP) (mmHg), diastolic blood pressure (DBP) (mmHg), PR (bpm), and VO2max (ml/kg/min).ConclusionThe present study found that a HIIT protocol performed during the cool-down period of the physical education classes generated adaptations such as improvement in body composition, BP variables and PR, and cardiorespiratory fitness, in overweight and obese adolescents. In contrast, the group of overweight and obese adolescents who performed SIT showed limited benefits, with changes in fat mass only.

The key to success in the application of the sterile insect technique (SIT) relies on the ability of released, sterile males to outcompete their fertile wild male counterparts to mate with wild females. However, many insect species exhibit multiple-mating behavior, which can be a way for females to select paternity for their progeny. This study aims to recognize the consequences of potential double-matings during an SIT program and to detect any evidence of sperm selection favoring sperm from fertile mates. This report provides a descriptive analysis of the storage and use of sperm by female Aedes albopictus. Stable isotopes were used to mark the sperm of fertile and sterile males. Mated females were allowed to oviposit before dissecting the spermathecae to link the presence of each type of sperm to the sterility of the eggs laid. It was found that sperm in females inseminated by both males was distributed in the three spermathecae with no obvious pattern, mostly mixed but also separately, and no evidence of any mechanism for sperm selection, sperm precedence, or sperm competition in Ae. albopictus females could be found. The fact that only a few double-mated females were double-inseminated and could also produce semi-sterile eggs, together with the finding that the sperm of sterile males appeared to be no less viable than that of fertile males, is an encouraging outcome for SIT approaches.

Halyomorpha halys (Heteroptera: Pentatomidae) is an insect pest native to Asia that has spread over the last two decades to most of the North America, parts of South America, Europe and North Africa. Its impact is significant as it can feed on more than 300 host plants, rendering affected fruits and vegetable crops unsellable or of lower quality. Various chemical and biological methods have been used to control this pest, with varying degrees of success. The sterile insect technique (SIT) is a pest control method involving the sterilization of insects via ionizing radiation and their subsequent mass release into the field. In the present contribution, the spermiogenesis of H. halys was studied from an ultrastructural point of view in both irradiated and non-irradiated adult males. In both cases, we observed ultrastructural characteristics typical of hemipteran sperm cells: bridges connecting the mitochondrial derivatives and the axonemal microtubules, the absence of accessory bodies, and the presence of two or three crystalline inclusions within the mitochondrial derivatives, an acrosome composed of tightly packed tubules, and an atypical, plaque-shaped microtubular organizing center (MTOC) in the centriolar region. Moreover, in the same region, we seldom observed the presence of two centrioles in the spermatids, one of which disappeared at a later stage of maturation. This feature is a novelty for insect spermiogenesis. The cysts of irradiated adults were not all uniformly affected by the radiation. However, irradiated cysts sometimes exhibited a general disorganization of sperm arrangement, incomplete divisions of sperm cells resulting in multiple copies of the same organelle within the same cell, failure to reabsorb the cytoplasm, and the lack of axonemes. Finally, rod-shaped viruses or virus-like particles were observed in vasa deferentia independently of irradiation.

Each year, hundreds of millions of people are infected with arboviruses such as dengue, yellow fever, chikungunya, and Zika, which are all primarily spread by the notorious mosquito Aedes aegypti. Traditional control measures have proven insufficient, necessitating innovations. In response, here we generate a next-generation CRISPR-based precision-guided sterile insect technique (pgSIT) for Ae. aegypti that disrupts genes essential for sex determination and fertility, producing predominantly sterile males that can be deployed at any life stage. Using mathematical models and empirical testing, we demonstrate that released pgSIT males can effectively compete with, suppress, and eliminate caged mosquito populations. This versatile species-specific platform has the potential for field deployment to effectively control wild populations of disease vectors.

In this study, four doses 50, 100, 150 and 200 Gy of gamma radiation were tested against 3 days old pupae of Spodoptera frugiperda to investigate some biological aspects and morphological changes. The results showed that the percentage mortality and malformed of pupal increased significantly with the increasing doses. In addition, all treatment cussed significantly prolonged in pupal duration compared with unirradiated, except the dose 50 Gy. When females crossed with male emergence from pupae treated with gamma radiation, fecundity decreased by increasing radiation dose. The decrease of egg hatchability percent and increase of sterility induced by gamma radiation were found to be positively correlated with the dose level. The parentage moth mortality resulted from pupal irradiated increased significantly with the increase of used doses. Females affected with irradiation more than males at the doses of 100, 150 and 200 Gy, with prolonged longevity. In general, the results obtained indicated that the biological action of gamma irradiation against S. frugiperda pupae was more remarkable effects on the appearance of male and female pupae after radiation treatment; some damage in the ventral surface of abdomen at 150 Gy and enlargement in the dorsal surface of thorax and the inability of adult to leave its puparium at 200 Gy as compared with normal pupae. Also, features of malformation in some adults resulted from irradiated pupae were noticed in the wings. Gamma radiation affected the reproductive potential of S. frugiperda and therefore thought to be one of the factors responsible for the reduction of the fecundity and development. Larval diet is one of the key factors in establishing a mass-rearing/production system for Wolbachia-based approaches that promotes high-quality and high-performance adult mosquitoes at a low/reasonable cost. To identify a suitable larval diet for Aedes aegypti infected with the Wolbachia line (wMID) and wild-type lab-established line (MID), four diets with different protein sources (ranging from 42 to 75%) were compared: fish food (TIL), bovine liver powder (COW), porcine powder meal (PIG), and a mix standard laboratory diet (MFOOD). The COW diet for wMID and MID (without affecting survival to the pupal stage) showed a shorter time to pupation, and the average time was 6 to 7 days, respectively. No significant differences were observed on pupation for wMID and MID, which ranged between 92 and 95% and 96–98%, respectively. A larger pupae size was observed among the TIL, COW, and MFOOD diets for wMID; no differences were recorded for MID. With the COW diet, wMID (7.90 ± 0.06) and MID (7.76 ± 0.10) males had shorter development times from LI to emergence. The stability of the Wolbachia infection was not affected by the diets evaluated. Independently of the Wolbachia infection, all diets are suitable for mass-production and maintenance of Ae. aegypti. Overall, no negative impact was observed on the life history traits evaluated. Considering these results, along with the relative costs of the four diets, PIG and TIL are viable choices given their affordability, accessibility of ingredients in the area. Oogenesis in insects is a carefully orchestrated process, facilitating the formation of female gametes, which is regulated by multiple extrinsic and intrinsic factors, including ovary serine protease (Osp). As a member of the serine protease family, Osp is a homolog of Nudel, a maternally required protease defining embryonic dorsoventral polarity in Drosophila. In this study, we used CRISPR/Cas9-mediated mutagenesis to functionally characterize Osp in the Asian corn borer, Ostrinia furnacalis, a devastating maize pest throughout Asia and Australia. Building on previous knowledge, we hypothesized that knockout of Osp would disrupt embryonic development in O. furnacalis females. To examine this overarching hypothesis, we (1) cloned and characterized Osp from O. furnacalis, (2) designed target sites on exons 1 and 4 to construct a CRISPR/Cas9 mutagenesis system, and (3) documented phenotypic impacts among O. furnacalis Osp mutants. As a result, we (1) examined the temporal-spatial expression profiles of OfOsp, which has an open reading frame of 5648 bp in length and encodes a protein of 1873 amino acids; (2) established O. furnacalis Osp mutants; and (3) documented recessive, female-specific sterility among OfOspF mutants, including absent or deformed oviducts and reduced fertility in female but not male mutants. Overall, the combined results support our initial hypothesis that Osp is required for embryonic development, specifically ovarian maturation, in O. furnacalis females. Given its substantial impacts on female sterility, Osp provides a potential target for the Sterile Insect Technique (SIT) to manage Lepidoptera pests in general and the species complex Ostrinia in particular. Plant‐derived volatiles can have positive or negative effects on insects. Exposure to volatiles of guava (Psidium guajava L.) (Myrtaceae) enhances male mating success in the South American fruit fly, Anastrepha fraterculus (Wiedemann) (Diptera: Tephritidae), an important pest of various fruits. These volatiles can therefore be used as a post‐teneral, pre‐release supplement in sterile insect technique (SIT) programs. In SIT, mass‐reared sterile males compete for and mate with wild females, in order to reduce populations of insect pests. However, the success of SIT will be diminished if males are not able to inhibit females from remating with other (potentially wild) males. Here, we investigated the remating propensity of A. fraterculus females mated with males exposed to guava volatiles or non‐exposed (control) in choice and no‐choice tests. In addition, we tested whether exposure to guava volatiles affected male reproductive organ size (testes, accessory glands, and seminal vesicles) and cuticular hydrocarbon (CHC) profile. Females mated with guava‐exposed males were more likely to remate than females mated with control males, but there was no difference in the length of the refractory period (i.e., the time between the first and second mating event). Reproductive organ size did not differ between exposed and non‐exposed males, but exposed males had a higher quantity of waxes than non‐exposed males, and CHC profiles differed. The peach fruit fly, Bactrocera zonata (Saunders) (Diptera: Tephritidae), is a pest of economic importance, endemic to South and South‐East Asia. It is highly invasive and due to its quarantine pest status, it interferes with horticultural trade. Males of many Bactrocera species are strongly attracted to methyl eugenol (ME) [1,2‐dimethoxy‐4‐(2‐propenyl) benzene], a phenylpropanoid compound naturally occurring in many plant species. Feeding on ME is also reported to enhance male mating success in several Bactrocera species. Such an effect can increase the effectiveness of the sterile insect technique (SIT). The current study was designed to identify the sexual maturity age of males of B. zonata, the relation between male age and attraction to ME, and the effect of ME on male mating success. The results showed that males initiated their mating at 8 days of age and attained the highest mating success at 16 days of age. The percentage of immature males responding to ME increased with age and the maximum response was observed during the age of sexual maturity. Methyl eugenol treatment at sexual maturity significantly enhanced male mating success over untreated males. The males treated with ME at a sexually immature age (5 days old) achieved significantly higher mating success after reaching sexual maturity. These findings suggested that B. zonata males can be treated with ME at the age of 5 days in sterile‐male 'holding and release' facilities and thereafter released in the field, where they are expected to achieve higher mating success after attaining sexual maturity. Introduction: Maximal oxygen uptake (V̇O2max) is a pivotal factor for aerobic endurance performance. Recently, aerobic high‐intensity interval training (HIIT) was documented to be superior to sprint interval training (SIT) in improving V̇O2max in well‐trained males. However, as mounting evidence suggests that physiological responses to training are sex‐dependent, examining the effects of HIIT versus SIT on V̇O2max, anaerobic capacity, and endurance performance in females is warranted. Methods: We randomized 81 aerobically well‐trained females (22 ± 2 years, 51.8 ± 3.6 mL∙kg−1∙min−1 V̇O2max), training three times weekly for 8 weeks, to well‐established protocols: (1) HIIT 4 × 4 min at ~95% of maximal aerobic speed (MAS), with 3 min active recovery (2) SIT 8 × 20 s at ~150% of MAS, with 10 s passive recovery (3) SIT 10 × 30 s at ~175% of MAS, with 3.5 min active recovery. Results: Only HIIT 4 × 4 min increased V̇O2max (7.3 ± 3.1%), different from both SIT groups (all p < 0.001). Anaerobic capacity (maximal accumulated oxygen deficit) increased following SIT 8 × 20 s (6.5 ± 10.5%, p < 0.05), SIT 10 × 30 s (14.4 ± 13.7%, p < 0.05; different from HIIT 4 × 4 min, p < 0.05). SIT 10 × 30 s resulted in eight training‐induced injuries, different from no injuries following HIIT 4 × 4 min and SIT 8 × 20 s (p < 0.001). All groups improved long‐distance (3000‐meter) and sprint (300‐meter) running performance (all p < 0.001). SIT protocols improved sprint performance more than HIIT 4 × 4 min (p < 0.05). Compared to previous male results, no increase in V̇O2max following SIT 8 × 20 s (p < 0.01), and a higher injury rate for SIT 10 × 30 s (p < 0.001), were evident. Background: Recovery is a key factor to promote adaptations and enhance performance. Sprint Interval Training (SIT) is known to be an effective approach to improve overall physical function and health. Although a 2-day rest period is given between SIT sessions, the time-course of recovery after SIT is unknown. Purpose: The aim of this study was to determine whether the neuromuscular and autonomic nervous systems would be impaired 24 and 48 h after an SIT session. Methods: Twenty-five healthy subjects performed an 8 × 15 s all-out session on a braked cycle ergometer with 2 min of rest between repetitions. Isometric maximal voluntary contraction (iMVC) and evoked forces to electrical nerve stimulation during iMVC and at rest were used to assess muscle contractile properties and voluntary activation before (Pre), 1 (Post24h), and 2 (Post48h) days after the session. Two maximal 7 s sprints with two different loads were performed at those same time-points to evaluate the maximal theoretical force (F0), velocity (V0) and maximal power (Pmax) production during a dynamic exercise. Additionally, nocturnal heart rate variability (HRV) was assessed the previous and the three subsequent nights to the exercise bout. Results: No significant impairments were observed for the iMVC or for the force evoked by electrical stimulation 1 day after the session. Similarly, F0, V0, and Pmax were unchanged at Post24h and Post48h. Furthermore, HRV did not reveal any temporal or frequential significant difference the nights following SIT compared to Pre. Aedes aegypti spread devastating viruses such as dengue, which causes disease among 100 to 400 million people annually. A potential approach to control mosquito disease vectors is the sterile insect technique (SIT). The strategy involves repeated release of large numbers of sterile males, which reduces insect populations because the sterile males mate and thereby suppress the fertility of females that would otherwise mate with fertile males. While SIT has been successful in suppressing certain agricultural pests, it has been less effective in depressing populations of Ae. aegypti This limitation is in part because of the fitness effects resulting from mutagenizing the mosquitoes nonspecifically. Here, we introduced and characterized the impact on female fertility of an Ae. aegypti mutation that disrupts a gene that is specifically expressed in testes. We used CRISPR/Cas9 to generate a null mutation in the Ae. aegypti β2-tubulin (B2t) gene, which eliminates male fertility. When we allowed wild-type females to first mate with B2t mutant males, most of the females did not produce progeny even after being subsequently exposed to wild-type males. We also introduced B2t mutant and wild-type males simultaneously with wild-type females and found that a larger number of B2t mutant males relative to the wild-type males was effective in significantly suppressing female fertility. These results raise the possibility of employing B2t sterile males to improve the efficacy of SIT in suppressing populations of Ae. aegypti through repeated releases and thereby reduce the transmission of viruses by these invasive mosquitoes.

The solubility and hydrolysis of Be(II) was investigated from undersaturation conditions in alkaline, dilute to concentrated CaCl2 solutions (0.05–3.5 M). Experiments were performed with α-Be(OH)2(cr) under Ar atmosphere at T = (22 ± 2)°C. Aqueous Be speciation was further investigated by means of molecular dynamics (MD) calculations. For the most diluted CaCl2 systems (0.05 and 0.25 M), a solubility minimum is observed at pHm ≈ 9.5 {with [Be(II)] ≈ 10−7 M}, consistent with solubility data previously reported in NaCl and KCl solutions. Above this pHm, and at higher CaCl2 concentrations, a steep increase in the solubility with a slope of ∼ +2 is observed, hinting towards the predominance of the moiety [Be(OH)42–] in the aqueous phase. In NaCl and KCl systems, this hydrolysis species prevails only above pHm ∼ 13, thus supporting the formation of ternary complex/es Ca–Be(II)–OH(aq) in CaCl2 solutions. The analysis of solubility data in combination with MD calculations underpin the key role of the complex Ca2[Be(OH)4]2+ in alkaline to hyperalkaline systems containing Ca. In combination with our previous work in NaCl–NaOH and KCl–KOH systems, complete chemical, thermodynamic and (SIT) activity models are derived for the first time for the system Be2+–Ca2+–Na+–K+–H+–Cl––OH––H2O(l). This model provides an accurate and robust tool for the evaluation of Be(II) solubility and speciation in a diversity of geochemical conditions, including source term calculations of beryllium in the context of repositories for nuclear waste disposal with a high cement inventory.

Whether multiple representations in visual working memory (VWM) can simultaneously guide attention is controversial. The multiple-item-template (MIT) hypothesis holds that multiple VWM representations can be active to guide visual attention simultaneously, whereas the single-item-template (SIT) hypothesis argues that only one VWM representation can interact with attention at a time. Recent study form (Fan et al., Acta Psychologica 192:126–137, 2019) provided strong evidence on MIT through the method of comparing the patterns of guidance effects in different experimental conditions. In the present study, we aimed to reexamine SIT and MIT by sharing a new condition wherein two search distractors match the single memory representation, using the original Fan et al. tilted-line search task. The guidance effect when two colored-shapes were held in VWM and both were present as distractors in the search array were observed and equal to that when the single colored-shape in VWM matched two of the distractors, which is consistent with the hypothesis of MIT. Moreover, the comparison on the degree of linear fitting between the actual data and two hypothesized models according to SIT and MIT showed that the Model of MIT fits the data best with a higher R2 of 0.125. Aedes aegypti (L.) is an invasive mosquito responsible for vectoring diseases such as dengue, Zika and Chikungunya. Dengue affects a large proportion of the global population, with the World Health Organization estimating that half the global population is at risk, with 390 million infections occurring each year. Control of mosquito vector populations over large geographical scales can be improved and made economically viable by computer models with potential to aid decision support.We introduce a method for estimating mosquito abundance (population size) over time in biocontrol programmes that involve the release of sterilised insects. We employ Bayesian state‐space modelling to provide insight into population trajectories using data from an application of incompatible insect technique (IIT) biological control, in Far North Queensland, Australia. The general approach could be adapted to other insect species.We demonstrate how the modelling approach can estimate trajectories of abundance over time as an unmarked–release–recapture analysis. Additionally, it provides a means for quantifying population suppression in IIT programmes (a statistic that can be challenging to estimate in practice) using counterfactuals.Modelling results show that estimated population trajectories exhibit similar temporal patterns to raw trapping rate data collected in the field, for example, the presence of peaks (and troughs) associated with the timing of rainfall events. Additional confidence in our model was demonstrated through a cross‐validation study where we left out each of the six landscapes from our dataset, fit the model using the remaining five regions and assessed its predictive skill. Modelled counterfactuals allowed us to estimate that population suppression in treated landscapes was 95%–99%.Synthesis and applications. Our model can provide valuable insights that can shape decision support systems in sterile insect technique and incompatible insect technique programmes operating over large geographical scales. The model helps determine how many sterile/incompatible insects should be released over time and how population control is progressing (via use of counterfactual scenarios). These outcomes are achieved because the model provides estimates of wild‐type populations over time, even when there has been no differentiation between sterile/incompatible and wild‐type insects caught in traps. Improvements are needed in mosquito mass-rearing to effectively implement the sterile insect technique (SIT). However, managing this technique is challenging and resource intensive. SIT relies on mass rearing, sterilization, and release of adult males to reduce field populations. Maintaining an acceptable level of female presence, who can transmit viruses through biting, is crucial. Females are also essential for facility sustainability. Sex sorting plays a vital role in the production process, and our current mechanical sorting approach aims to obtain a high number of adult males with minimal female contamination within 24 h of pupation. Utilizing protandry helps control female contamination. While the 24-h sorting period achieves desired contamination levels, it may not yield enough females to sustain breeding lines, leading to increased labor costs that impact project sustainability. By delaying the sorting procedure to 48 h, we obtained sufficient females to sustain breeding lines, achieving a balance between male production and female contamination using the automatic version of the Fay–Morlan device as the sorting tool. Background This study examined the effects of regulated and controlled supramaximal high-intensity interval training (HIT) adapted for older adults, compared to moderate-intensity training (MIT), on cardiorespiratory fitness; cognitive, cardiovascular, and muscular function; and quality of life. Methods Sixty-eight nonexercising older adults (66–79 years, 44% males) were randomized to 3 months of twice-weekly HIT (20-minute session including 10 × 6-second intervals) or MIT (40-minute session including 3 × 8-minute intervals) on stationary bicycles in an ordinary gym setting. Individualized target intensity was watt controlled with a standardized pedaling cadence and individual adjustment of the resistance load. Primary outcomes were cardiorespiratory fitness (V̇ o2 peak) and global cognitive function (unit-weighted composite). Results V̇ o2 peak increased significantly (mean 1.38 mL/kg/min, 95% CI [0.77, 1.98]), with no between-group difference (mean difference 0.05 [−1.17, 1.25]). Global cognition did not improve (0.02 [−0.05, 0.09]), nor differed between groups (0.11 [−0.03, 0.24]). Significant between-group differences in change were observed for working memory (0.32 [0.01, 0.64]), and maximal isometric knee extensor muscle strength (0.07 N·m/kg [0.003, 0.137]), both in favor of HIT. Irrespective of the group, there was a negative change in episodic memory (−0.15 [−0.28, −0.02]), a positive change in visuospatial ability (0.26 [0.08, 0.44]), and a decrease in systolic (−2.09 mmHg [−3.54, −0.64]) and diastolic (−1.27 mmHg [−2.31, −0.25]) blood pressure. Conclusions In nonexercising older adults, 3 months of watt-controlled supramaximal HIT improved cardiorespiratory fitness and cardiovascular function to a similar extent as MIT, despite half the training time. Simple Summary: The tiger mosquito, Aedes albopictus (Skuse, 1894), is a competent vector of arboviruses such as dengue, Zika, and chikungunya among others. The sterile insect technique (SIT) is presented as an innovative and environmentally friendly control method to be implemented as a main component of integrated vector control management (IVM). In the Valencian region (Spain), an IVM pilot project led and funded by the Department of Agriculture, based on the use of SIT to control Ae. albopictus, has been carried out from 2017 to 2020. It has proven to be effective in reducing tiger mosquito populations. This manuscript analyzes the impact of the migration of wild individuals from peri-urban areas into non-isolated urban centers on the control strategy. In recent years, Aedes albopictus (Skuse, 1984) has expanded its distribution globally due to its high ecological plasticity. This expansion has increased the population's susceptibility to contracting diseases such as dengue, Zika, and chikungunya, among others, which are transmitted by this mosquito species. In the absence of effective control methods, the application of the sterile insect technique (SIT) is proposed as part of an integrated vector management (IVM) program. From 2007 to 2020, this strategy has been tested in a non-isolated mosquito population urban area of 45 ha, representative of the municipalities of the Valencian region (Spain). The population levels of adult females and eggs collected in the traps have been reduced by 70–80% compared to the control area, demonstrating its efficacy in reducing mosquito populations. Simple Summary: Halyomorpha halys is an alien stink bug species native to south-eastern Asia, and is now widely distributed and very invasive worldwide. Management of this pest by chemical insecticides is not very effective because of the high mobility of the pest species and its innate ability to develop resistance to most of the synthetic insecticides applied. For this reason, classical biological control has been considered, with attention focused on co-evolved egg parasitoids, which oviposit and complete their larval development within a single egg of the host. The detection and collection of the egg parasitoid species is often conducted by exposing newly oviposited egg-masses of the stink bug pest in the field for a few days as sentinels, which can then be parasitized by the egg parasitoids. In newly invaded areas, limiting factors in the use of sentinel eggs include the short amount of time that they are a suitable substrate for the oviposition of the egg parasitoid and the risk of the unintentional release of additional pests from unparasitized eggs. A valid potential alternative is to use sterile sentinel eggs of the pest species. This study focused on the evaluation of the performance of three types of sterile sentinel eggs for the oviposition and the larval development of the egg parasitoid Trissolcus japonicus. The brown marmorated stink bug (BMSB), Halyomorpha halys, is a phytophagous invasive pest native to south-eastern Asia, and it is now distributed worldwide. This species is considered to be one of the most damaging insect pests in North America and in Europe. In agriculture, the predominant approach to managing BMSB is based on the use of insecticides, specifically pyrethroids and neonicotinoids. Unfortunately, the biology of the species and its facility to develop mechanisms of resistance to available pesticides has induced farmers and scientists to develop different, least-toxic, and more effective strategies of control. In a territorial area-wide approach, the use of a classical biological control program in combination with other least-toxic strategies has been given prominent consideration. Following exploratory surveys in the native range, attention has focused on Trissolcus japonicus, a small scelionid egg parasitoid wasp that is able to oviposit and complete its larval development in a single egg of H. halys. A common method for detecting egg parasitoids in the native range involves the placement of so-called 'sentinel' egg masses of the pest in the environment for a short period, which are then returned to the laboratory to determine if any of them are parasitized. Outside of the area of origin, the use of fertile sentinel eggs of the alien species may lead to the further release of the pest species; an alternative is to use sterile sentinel eggs to record the presence of new indigenous egg parasitoids or to detect the dispersal of alien species (in this case, T. japonicus) released in a new environment to control the target insect pest species. This study evaluated the performance of three types of sterile sentinel eggs as a suitable substrate for the oviposition and larval development of the egg parasitoid T. japonicus in a context of combining classical biological control with a Sterile Insect Technique (SIT) approach. Citrus leafminer (CLM), Phyllocnistis citrella Stainton (Lepidoptera: Gracillariidae), and European corn borer (ECB), Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae), are two key pests of young citrus and maize plants in Iran, respectively. In this study, the effects of sub‐sterilizing doses of gamma radiation (100–250 Gy for CLM, 90–180 Gy for ECB) on the biological and reproductive parameters of F1 progeny from irradiated males were determined. The results indicated that males irradiated at doses of ≥200 and ≥150 Gy produced highly sterile F1 descents for CLM and ECB, respectively. No adverse effects of radiation doses applied to males were observed on the F1 pupal emergence for both species, nor on the adult longevity of male and female ECB moths. However, the life span of CLM F1 male and female adults decreased with increasing doses. The sex ratio of emerged adults of F1 progeny from irradiated males was skewed towards males with increasing doses; the proportion of males to all emerged adults reached 0.52 and 0.54 at doses of 250 and 150 Gy for CLM and ECB, compared to 0.44 and 0.49 in the controls, respectively. The competitiveness index of irradiated and F1 males of both species fluctuated in acceptable ranges and in general F1 males were stronger competitors than their irradiated male parents. The results of the field cage experiments showed that the larval infestation in all cages including the released irradiated CLM and ECB moths decreased compared to the control treatment – the percentage of damaged leaves and stems in the cages with ratios of 20:1 and 7:1 irradiated‐to‐normal insects (highest ratios assayed) reached half and one‐third of the control for CLM and ECB, respectively. Methoprene, a juvenile hormone analog, is used to accelerate sexual maturation in males of species of economic importance in support to the sterile insect technique (SIT). In the SIT, mass-reared sterile males are released into the field and need to survive until they reach sexual maturation, find a wild female, mate with her and then induce female sexual refractoriness, so she will not remate with a wild counterpart. The use of methoprene shortens the time between release and copulation. However, in South American fruit flies, Anastrepha fraterculus , the ability of methoprene-treated males to inhibit female remating has been shown to be lower than wild males, when methoprene was applied by pupal immersion or topical application. Here we evaluated the possibility of incorporating methoprene into the male diet at different doses and the ability of those males to inhibit female remating, as well as the effect of methoprene on male reproductive organ size, due to the possible correlation between male accessory gland size and their content, and the role of male accessory gland proteins in female inhibition. We found that A. fraterculus males fed with methoprene in the adult protein diet at doses as high as 1% were less likely to inhibit female remating, however, at all other lower doses males had the same ability as untreated males to inhibit female remating. Males fed with methoprene had bigger male accessory glands and testes compared to methoprene-deprived males. We demonstrate that the incorporation of methoprene in adult male diets is possible in this species and potentially useful as a post-teneral, pre-release supplement at doses as low as 0.01%. Even at higher doses, the percentage of females remating after 48 h from the first copulation is sufficiently low in this species so as not compromise the efficiency of the SIT. BACKGROUND: Zeugodacus cucurbitae is an agricultural pest species with robust reproductive capabilities capable of causing extensive damage. The advent of novel male fertility‐related pest control strategies has been an area of active entomological research focused on the sterile insect technique (SIT) strategy. RESULTS: RNA‐sequencing analyses were conducted using 16 tissue samples from adult male Z. cucurbitae, leading to the identification of 5338 genes that were differentially expressed between the testes and three other analyzed tissue types. Of these genes, 808 exhibited high levels of testis expression. A quantitative polymerase chain reaction (qPCR) approach was used to validate the expression of ten of these genes selected at random, including ZcTSSK1 and ZcTSSK3, which are testis‐specific serine/threonine protein kinase (TSSK) genes. Evaluation via a loss‐of‐function‐based knockdown assay showed that the down‐regulation of either of these two genes in males was associated with significantly decreased egg hatching rates. In situ hybridization analyses revealed the expression of both of these transcripts in the transformation zone, and significant decreases in Z. cucurbitae sperm numbers were observed following double‐stranded RNA treatment. Together, these results suggested that inhibiting ZcTSSK1 and ZcTSSK3 expression was sufficient to alter male fertility in Z. cucurbitae. Conclusion: These transcriptional sequencing results provide a foundation for further efforts to clarify the regulators of Z. cucurbitae male fertility. Simple Summary: The red palm weevil Rhynchophorus ferrugineus is an invasive pest insect that feeds on more than 20 species of palms, many of which are widely grown for food and ornamental use. Due to its vast spread and the huge economic damage this pest can cause, it is very important to find sustainable and effective eradication strategies. Sterile insect techniques are biological control approaches based on the release of mass-reared sterilized males that could improve the likelihood of successful pest control. However, for the successful implementation of these approaches, it is necessary to develop a deep understanding of the insect's mating system. For this purpose, we built a paternity assignment strategy based on previously developed microsatellite loci to help the future study of the reproductive mechanisms of this species in laboratory-controlled mating experiments. We evaluated the reliability of our genetic tool using a simulation approach that aimed to explore its resolution power in paternity tests, both in the laboratory and in nature. The main goal of this research was to assess the performance of a previously developed microsatellite panel in terms of the paternity assignment of progeny from laboratory mating experiments. Using a simulation approach, we evaluated the reliability of the microsatellite markers in the paternity tests both in complex laboratory experiment scenarios and on the progeny of wild-caught gravid females to help future studies on the RPW mating system. As a case study of the simulation results, we performed two double-mating experiments, genotyped the progeny and estimated the P2 values to compare to the expected progeny genotypes according to the crossing scheme of each experiment. The results of our simulations on laboratory experiments showed that it was possible to carry out paternity assignments for all progeny with reliable statistical confidence using our 13 microsatellites set. On the contrary the low genetic variability measured in red palm weevil populations in invaded areas made the resolution power of our loci too low to carry out paternity analyses on natural populations. Abstract Background The global incidence of dry eye disease (DED) is very high. DED seriously affects the quality of life of patients; however, the current curative effect of medicine for moderate to severe DED is poor. This randomized clinical trial was planned to investigate the effect of acupuncture compared with artificial tears (AT) on moderate to severe DED. Methods A randomized clinical trial was performed at 2 hospitals in China. 120 DED patients were randomly equally divided into an acupuncture and an artificial tear group. Either acupuncture or artificial tears was performed for an 8-week period, and a 24-week follow-up was performed. The primary outcome measure was the Schirmer-I test (SIT) change from baseline. The secondary outcome measures included the numerical rating scale (NRS) change from baseline for improvement in ocular symptoms, the ocular surface disease index (OSDI), the tear-film break-up time (TBUT), corneal fluorescein staining (CFS), and acupuncture acceptability. Adverse events also were monitored and documented. Results For the primary outcome, the mean changes from baseline in the SIT values were significantly different between the acupuncture (5.75 [2.53–9.75]) and AT (0.52 [− 1.18–2.46]) groups at week 8 with a between difference of 5.23 (P < 0.05). Between-group differences of 8.49 in OSDI score change from baseline differed significantly at week 8 (P < 0.05). However, between-group differences of the changes in the average symptom NRS score, TBUT, and CFS did not differ significantly at week 8. Five cases experienced acupuncture-related adverse events. Conclusions This randomized clinical trial found that acupuncture at BL1 significantly promoted tear secretion. Acupuncture showed greater benefits than AT for moderate to severe DED. Introduction: The cognition and motivation of family business incumbents and the successors will directly influence succession behavior, yet they face identity challenges during succession due to the intersection of family and firm context, and their ability to overcome identity challenges will determine the success of the succession. However, as studies on their identity are fragmented and lack systematicity, there is a need to assess the relevant literature. Methods: Drawing from social identity theory (SIT) and role identity theory (RIT), this article adopts a systematic literature review approach to analyze 99 SSCI-indexed articles to explore family business succession from an identity perspective. Results: The article finds that the focus on the self-concept of the incumbent and the successor shifts from group identification to role identity perception and multi-roles, and succession behaviors are based on identity perception. Discussion: This article summarizes a knowledge framework of the antecedents, connotations, and behavioral consequences of identity perception, revealing that family business succession froman identity perspective exhibits psychological and multidisciplinary characteristics, highlighting iterative and mutual features. Based on identity theories and succession research, this article proposes future directions from the research topics, research methods, and theoretical perspectives within the existing knowledge framework, such as cross-cultural and diachronic analysis, as well as from the theoretical perspectives of family, personality development, and pedagogy. The sterile insect technique (SIT) is used to control fruit fly pests, such as the Mediterranean fruit fly (medfly), Ceratitis capitata (Wiedemann) (Diptera: Tephritidae). Enhancing sexual competitiveness of mass‐reared sterile males can contribute to making this technique more effective. It has been shown that exposure to volatiles from essential oils (EOs), such as ginger root oil (GRO) and those from host fruits, increases male mating success. We evaluated the effect of EOs from non‐host species native to Argentina, Schinus polygama (Cav.) Cabrera (Anacardiaceae) and Baccharis spartioides (Hook. & Arn.) Remy (Asteraceae), on the sexual competitiveness of sterile C. capitata males. In field cage experiments, sterile males exposed to S. polygama EO increased their mating success. In addition, sterile males exposed to this EO achieved more matings on trees than non‐exposed males, thus suggesting the former are more prone to locate and defend the pheromone‐calling territory. Deprivation of water and/or food significantly reduced males' life span, but exposure to S. polygama EO and GRO did not affect their survival. Schinus polygama EO is composed of mono‐ and sesquiterpenes with behavioral and electroantennographic responses in medflies, indicating that semiochemicals that boost medfly sexual competitiveness combine in this EO. Maximal oxygen uptake (V̇O2max) may be the single most important factor for long‐distance running performance. Interval training, enabling high intensity, is forwarded as the format that yields the largest increase in V̇O2max. However, it is uncertain if an optimal outcome on V̇O2max, anaerobic capacity, and running performance is provided by training with a high aerobic intensity or high overall intensity. Thus, we randomized 48 aerobically well‐trained men (23 ± 3 years) to three commonly applied interval protocols, one with high aerobic intensity (HIIT) and two with high absolute intensity (sprint interval training; SIT), 3× week for 8 weeks: (1) HIIT: 4 × 4 min at ~95% maximal aerobic speed (MAS) with 3 min active breaks. (2) SIT: 8 × 20 s at ~150% MAS with 10 s passive breaks. (3) SIT: 10 × 30 s at ~175% MAS with 3.5 min active breaks. V̇O2max increased more (p < 0.001) following HIIT, 4 × 4 min (6.5 ± 2.4%, p < 0.001) than SIT, 8 × 20 s (3.3 ± 2.4%, p < 0.001) and SIT, 10 × 30 s (n.s.). This was accompanied by a larger (p < 0.05) increase in stroke volume (O2‐pulse) following HIIT, 4 × 4 min (8.1 ± 4.1%, p < 0.001) compared with SIT, 8 × 20 s (3.8 ± 4.2%, p < 0.01) and SIT, 10 × 30 (n.s.). Anaerobic capacity (maximal accumulated oxygen deficit) increased following SIT, 8 × 20 s (p < 0.05), but not after HIIT, 4 × 4 min, nor SIT, 10 × 30 s. Long‐distance (3000‐m) endurance performance increased (p < 0.05–p < 0.001) in all groups (HIIT, 4 × 4 min: 5.9 ± 3.2%; SIT, 8 × 20 s: 4.1 ± 3.7%; SIT, 10 × 30 s: 2.2 ± 2.2%), with HIIT increasing more than SIT, 10 × 30 s (p < 0.05). Sprint (300‐m) performance exhibited within‐group increases in SIT, 8 × 20 s (4.4 ± 2.0%) and SIT, 10 × 30 s (3.3 ± 2.8%). In conclusion, HIIT improves V̇O2max more than SIT. This novel system for long-distance mass transport of mosquitoes may therefore be used to ship sterile males worldwide for journeys of two to four days. Pilot programs of the sterile insect technique (SIT) against Aedes aegypti may rely on importing significant and consistent numbers of high-quality sterile males from a distant mass rearing factory. As such, long-distance mass transport of sterile males may contribute to meet this requirement if their survival and quality are not compromised. This study therefore aimed to develop and assess a novel method for long-distance shipments of sterile male mosquitoes from the laboratory to the field. Different types of mosquito compaction boxes in addition to a simulation of the transport of marked and unmarked sterile males were assessed in terms of survival rates/recovery rates, flight ability and morphological damage to the mosquitoes. The novel mass transport protocol allowed long-distance shipments of sterile male mosquitoes for up to four days with a nonsignificant impact on survival (>90% for 48 h of transport and between 50 and 70% for 96 h depending on the type of mosquito compaction box), flight ability, and damage. In addition, a one-day recovery period for transported mosquitoes post-transport increased the escaping ability of sterile males by more than 20%. This novel system for the long-distance mass transport of mosquitoes may therefore be used to ship sterile males worldwide for journeys of two to four days. This study demonstrated that the protocol can be used for the standard mass transport of marked or unmarked chilled Aedes mosquitoes required for the SIT or other related genetic control programs. Simple Summary: The sterile insect technique (SIT) is an environment-friendly, species-specific pest control method by which target insects are mass-produced in a factory and are made infertile by irradiation—usually with gamma rays. However, gamma sources are becoming more difficult and expensive to purchase, and the regulations surrounding these types of irradiators are becoming stricter. Therefore, there is now increasing interest in alternatives, such as X-ray irradiators. Following a recent technical evaluation of a blood X-ray unit, the aim of this research was to assess the biological responses of a selection of major SIT target insect species to irradiation in the X-ray unit as compared to gamma ray irradiation. It was found that all the insects responded similarly to X-rays as to gamma rays and that the X-ray unit is suitable for small- to medium-sized SIT programs. The sterile insect technique (SIT) is based on the inundatory field release of a target pest following their reproductive sterilization via exposure to radiation. Until recently, gamma irradiation from isotopic sources has been the most widely used in SIT programs. As isotopic sources are becoming increasingly expensive, especially for small programs, and regulations surrounding their procurement and shipment increasingly strict, irradiation capacity is one of the limiting factors in smaller or newly developing SIT projects. For this reason, the possibility of using X-ray irradiators has been evaluated in the recent decade. The availability of "off-the-shelf" blood X-ray irradiators that meet the technical requirements for insect irradiation can provide irradiation capacity for those SIT projects in which the acquisition of gamma ray irradiators is not feasible. Following the recent technical characterization of a Raycell MK2 X-ray blood irradiator, it was found in this study, that MK2 instruments were suitable for the sterilization of fruit flies, tsetse flies and mosquitoes, inducing comparable, even slightly higher, sterility levels compared to those achieved by gamma ray irradiation.
