Using a mixed-methods approach, we examine the complex link between user happiness and technology adoption in the context of smart homes. Our tests show that user happiness and adoption are highly influenced by the versions of smart home technologies, with Version A producing better user satisfaction (7.2) and adoption rates (68%) than Version B (6.8, 62%). Furthermore, consumers engaging with Features A and C reported the greatest adoption rates (80%) and satisfaction (8.1), indicating that certain features, particularly when paired, have a significant influence on user pleasure. Extended training times resulted in higher user satisfaction and adoption rates of the technology; the 6-hour training group had the greatest adoption rate (84%), and the highest satisfaction (8.3%). Furthermore, user age demographics have a substantial impact on adoption and happiness; young adults have the greatest adoption rate (70%) and contentment (7.6). These results highlight the necessity of developing smart home technologies that are appropriate for various age groups, as well as the significance of feature customization, thorough training, and user-centric design in improving user satisfaction and encouraging technology adoption. Vibration monitoring and control have been the main concern of machining industries. Traditional monitoring processes have several drawbacks such as; higher computational time, manual feature detection and requirement of supervision. To overcome these difficulties, this study proposes a hybrid approach based on Self-Organizing Maps (SOM) and multi-layer perceptron – back propagation neural network (MLP-BPNN) using sound signals for monitoring self-induced tool vibration (chatter) and metal removal rate (MRR) in milling operation. Initially, acquired vibration signals are decomposed for extracting desired machining data. Further, SOM has been applied on the reconstructed signals for data mapping and automatic feature selection. Selected features have been used as an input in MLP-BPNN training for the development of prediction models of machining quality and MRR. Finally, it has been observed that the proposed data-driven methodology can be well adapted for the automatic feature selection and can predict machining quality and MRR with nearly 98% accuracy. Chatter is a type of self-induced vibration that reflects fluctuations in both frequency and energy dispersion during the milling process, inevitably resulting in substandard part quality and diminished Material Removal Rates (MRR). It is essential to employ a robust chatter detection method to anticipate its emergence in the early stages. This study introduces an efficient Product Function (PF) based multi-mode signal processing technique, specifically the Spline-Based Local Mean Decomposition (SBLMD). This method is applied to decompose sound signals acquired through experimentation into a series of effective PF's. Subsequently, selected PFs are employed to reconstruct a new chatter signal that is information-rich. Additionally, prediction models based on Artificial Neural Networks (ANN) are established to predict Chatter Indicator (CI) and MRR using three different activation algorithms: Tan Sigmoid (TANSIG), Log Sigmoid (LOGSIG), and Purely Linear (PURELIN). Statistical comparisons have been conducted in order to obtain the optimal activation algorithm and found out that data set trained with LOGSIG gives minimal error. Moreover, an optimal range of input parameters has been selected pertaining to minimum chatter and maximum MRR. The objective of the study was to find out the effect of EI and gender on job satisfaction of primary school teachers. A total of 300 (150 male and 150 female) primary school teachers were selected randomly for the study. Emotional Intelligence Scale (EIS) and Teachers' Job Satisfaction Scale (TJSS) were used to collect the data. The study found a significant positive relationship between emotional intelligence and job satisfaction. Regression analysis showed that emotional management and emotional maturity significantly predicted the job satisfaction. The results of the two-way ANOVA showed that the interaction effect of gender and EI was not significant. Results also showed that level of EI significantly affected the job satisfaction of primary school teachers. However, there is no significant difference between male and female teachers regarding the level of job satisfaction. There are other factors which affect job satisfaction but the role of EI cannot be neglected. So the level of EI must be taken into consideration in order to select the best teachers.

Critical functions of intra-axonally synthesized proteins are thought to depend on regulated recruitment of mRNA from storage depots in axons. Here we show that axotomy of mammalian neurons induces translation of stored axonal mRNAs via regulation of the stress granule protein G3BP1, to support regeneration of peripheral nerves. G3BP1 aggregates within peripheral nerve axons in stress granule-like structures that decrease during regeneration, with a commensurate increase in phosphorylated G3BP1. Colocalization of G3BP1 with axonal mRNAs is also correlated with the growth state of the neuron. Disrupting G3BP functions by overexpressing a dominant-negative protein activates intra-axonal mRNA translation, increases axon growth in cultured neurons, disassembles axonal stress granule-like structures, and accelerates rat nerve regeneration in vivo.

Peripheral nerve regeneration after injury requires a broad program of transcriptional changes. We investigated the basis for the enhanced nerve regenerative capacity of the CAST/Ei mouse strain relative to C57BL/6 mice. RNA sequencing of dorsal root ganglia (DRG) showed a CAST/Ei-specific upregulation of Ascl1 after injury. Ascl1 overexpression in DRG neurons of C57BL/6 mice enhanced their neurite outgrowth. Ascl1 is regulated by miR-7048-3p, which is downregulated in CAST/Ei mice. Inhibition of miR-7048-3p enhances neurite outgrowth. Following injury, CAST/Ei neurons largely retained their mature neuronal profile as determined by single-cell RNA- seq, whereas the C57BL/6 neurons acquired an immature profile. These findings suggest that one facet of the enhanced regenerative phenotype is preservation of neuronal identity in response to injury.

Chemotherapy-induced peripheral neuropathy (CIPN) is a disabling side effect of cancer chemotherapy that can often limit treatment options for cancer patients or have life-long neurodegenerative consequences that reduce the patient's quality of life. CIPN is caused by the detrimental actions of various chemotherapeutic agents on peripheral axons. Currently, there are no approved preventative measures or treatment options for CIPN, highlighting the need for the discovery of novel therapeutics and improving our understanding of disease mechanisms. In this study, we utilized human-induced pluripotent stem cell (hiPSC)-derived motor neurons as a platform to mimic axonal damage after treatment with vincristine, a chemotherapeutic used for the treatment of breast cancers, osteosarcomas, and leukemia. We screened a total of 1902 small molecules for neuroprotective properties in rescuing vincristine-induced axon growth deficits. From our primary screen, we identified 38 hit compounds that were subjected to secondary dose response screens. Six compounds showed favorable pharmacological profiles – AZD7762, A-674563, Blebbistatin, Glesatinib, KW-2449, and Pelitinib, all novel neuroprotectants against vincristine toxicity to neurons. In addition, four of these six compounds also showed efficacy against vincristine-induced growth arrest in human iPSC-derived sensory neurons. In this study, we utilized high-throughput screening of a large library of compounds in a therapeutically relevant assay. We identified several novel compounds that are efficacious in protecting different neuronal subtypes from the toxicity induced by a common chemotherapeutic agent, vincristine which could have therapeutic potential in the clinic.
