International audience; Background: The possible coexistence of different strains in natural prion isolates is the subject of enduring discussions and might have implications for prion strains surveillance. Nonetheless, this possibility has not yet been proved. In 2007, a case of ovine scrapie with features compatible with a mixed infection of classical scrapie and Nor98 was reported in Italy. Objectives: To characterize by bioassay the prion agent(s) involved in this unusual scrapie case. Methods: Previous studies showed that voles are susceptible to the prevalent Italian scrapie agent and resistant to Nor98, while tg338 mice show the contrary. Taking advantage of this differential susceptibility, we inoculated voles and tg338 with the brain stem (BS) and the cerebral cortex (CC) of the unusual case. Discriminatory WB showed a predominance of classical scrapie PrPres in BS, while CC mainly contained Nor98 PrPres. A classical scrapie isolate (ClS) from the same flock of the unusual case, as well as Nor98 isolates from Italy and Norway were used as controls. Survival times and attack rates were calculated in voles and tg338. Brains were analysed by discriminative WB for PrPSc type, by IHC and PET-blot for PrPSc deposition and by E&E for lesion profile. Results: Both CC and BS induced disease in tg338, with survival times similar to Nor98 controls. As expected, Tg338 were resistant to ClS control. In contrast, voles developed disease following inoculation of ClS control and BS, but not of CC and Nor98 controls. In voles, the neuropathological and molecular phenotypes upon infection with the unusual case were the same of those observed with ClS, while in Tg338 were identical to Nor98 controls. Discussion: Our results demonstrate that two independent and separable prion agents coexist in the isolate investigated. This implies that: - mixed prion infections can occur - classical scrapie and Nor98 are distinct entities - the isolation of prion strains from a mixture is affordable.

Biometric and genetic variation were analysed among populations of D. nitedula from Italy, Balkan Peninsula, Asia Minor and Israel. The biometric analysis was carried out on 84 specimens using 13 linear skull measurements. Variations in metrical characters among samples were studied by standard and multivariate analyses. The genetic analysis was carried out on 29 specimens by electrophoresis of 40 gene loci. In both analyses the Israeli sample was the most distinct. Skulls of Israeli specimens are characterized by longer mandibular tooth row, higher rhamus mandibulae, longer bullae, and narrower braincase breadth, rostra1 breadth and interorbital constriction. Electrophoretically the Israeli sample can be discriminated from European populations by four loci (Ldh-1, G6pdh, Pep-1, and Lap-2) fixed for new alleles. The mean value of Nei's genetic distance between Israeli and European populations was D = 0.186, ranging from 0.153 to 0.227. The Israeli population, commonly attributed to the taxon phrygius, was different from this by biometric analysis. D. n. phrygius from the topotype locality in Asia Minor clustered with the European population from Turkish Thrace. In order to clarify the phylogenetic relationships of the Israeli forest dormouse, a comparison with other populations from the Caucasus and Iran and with Dryomys laniger from Turkey is suggested. Riassunto Variabilità genetica e biometrica in Dryomys nitedula (Pallas, 1778) - Variabilità biometrica e genetica sono state analizzate tra popolazioni di D. nitetula provenienti da Italia, Penisola Balcanica, Asia Minore ed Israele. L'analisi biometrica è stata condotta su 84 esemplari mediante 13 misurazioni craniche lineari. La variazione nei caratteri metrici tra i campioni è stata studiata mediante analisi standard e multivariata. L'analisi genetica è stata condotta su 29 esemplari mediante elettroforesi di 40 loci genici. In entrambe le analisi il campione israeliano è risultato il più distinto. I1 cranio degli esemplari israeliani è caratterizzato da fila molare mandibolare più lunga, rhamus mandibolae più alto, bullae più lunghe e da una riduzione nella larghezza della costrizione interorbitale, della scatola cranica e del rostro. Elettroforeticamente il campione israeliano può essere distinto dalle popolazioni europee sulla base di quattro loci fissati per nuovi alleli (Ldh-1, G6pdh, Pep-1 e Lap-2). Il valore medio della distanza genetica di Nei tra le popolazioni europee e quella israeliana è D = 0.186, con range compreso tra 0.153 e 0.227. La popolazione israeliana, generalmente attribuita al taxon phrygius, si differenzia da questo su base morfologica. D. n. phrygius della località topotipica in Asia Minore risulta affine alla popolazione europea della Tracia turca. Per chiarire i rapporti filogenetici del driomio israeliano si suggerisce il confronto con altre popolazioni del Caucaso e dell'Iran e con Dryomys laniger della Turchia.

We examined the glans penis of 34 adult specimens belonging to five genera and six species of Myoxidae. Three measurements of phalli and 11 measurements of bacula were taken and nine ratios were calculated. The baculum of Myoxidae is simple and in all the species studied is fully expressed at the tip of the phallus and, excluding Muscardinus, lies beneath cavernous tissue. In Myoxus, the baculum is triangular, wide and flat and evident interpopulational variability is existing: North Italian specimens display shape differences in comparison with South Italian and Thracian populations. The baculum of Dryomys is relatively thick, wide on the base and narrow at the shaft. There are differences in size between Thracian and Israeli populations of Dryomys nitedula. The baculum of Muscardinus is short and thin, with a small base; the phallus is tubelike and shorter than the baculum. The baculum of Myomimus is very short and robust, with a relatively wide base; the phallus, in contrast to other species, has no pad-like structure. The baculum of Eliomys differs from that of Dryomys and resembles more that of Myoxus, being more curved, more triangular and flat in shape, with widening in the middle. There are marked differences between bacula of E. quercinus and E. melanurus. A cluster analysis showed high affinities between Myoxus and Eliomys, as well as Myomimus and Dryomys, reflecting possible phylogenetic relationships. Riassunto Falli e bacula di Mioxidi europei; descrizione e comparazione - È stato esaminato il glans penis di 34 esemplari adulti appartenenti a cinque generi e sei specie di Myoxidae. Sono state rilevate tre misurazioni sul fallo, undici sul baculum e nove rapporti. Il baculum dei Myoxidae è semplice e in tutte le specie studiate è situato nella parte terminale del fallo e, tranne che nel moscardino, giace sotto il tessuto cavernoso. Il baculum di Myoxus è triangolare, ampio ed appiattito; è presente una evidente variabilità tra popolazioni. Esemplari dell'Italia settentrionale mostrano differenze nella forma del baculum rispetto a popolazioni dell'Italia meridionale e della Tracia. I1 baculum di Dryomys è relativamente spesso, ampio alla base e sottile nella parte terminale. Esistono differenze nelle dimensioni tra popolazioni di Dryomys nitedula della Tracia e di Israele. II baculum di Muscardinus è corto, sottile, con una piccola base; il fallo è tubulare e più corto del baculum. Il baculum di Myomimus è molto corto, robusto, con una base relativamente ampia; il fallo, contrariamente alle altre specie, è privo di protuberanze. Il baculum di Eliomys differisce da quello di Dryomys e ricorda più quello di Myoxus, avendo una forma più curvata, triangolare ed appiattita, con un allargamento nella parte mediana. Esistono marcate differenze tra il baculum di E. quercinus e quello di E. melanurus. Elevata affinità, che potrebbe indicare possibili rapporti filogenetici, tra Myoxus e Eliomys e tra Myomimus e Dryomys è stata osservata mediante cluster analysis.
