Abstract

Although several studies have investigated the factors affecting imputation accuracy, most of these studies involved a large number of genotyped animals. Thus, results from these studies cannot be directly applied to small populations, since the population structure affects imputation accuracy. In addition, factors affecting imputation accuracy may also be intensified in small populations. Therefore, we aimed to compare different imputation strate-gies for the Portuguese Holstein cattle population considering several commercially available single nucleotide poly-morphism (SNP) panels in a relatively small number of genotyped animals. Data from 1359 genotyped animals were used to evaluate imputation in 7 different scenarios. In the S1 to S6 scenarios, imputations were performed from LDv1, 50Kv1, 57K, 77K, HDv3 and Ax58K panels to 50Kv2 panel. In these scenarios, the bulls in 50Kv2 were divided into reference (352) and validation (101) populations based on the year of birth. In the S7 scenario, the validation population consisted of 566 cows genotyped with the Ax58K panel with theirgenotypes masked to LDv1. In general, all sample imputation accuracies were high with correlations ranging from 0.94 to 0.99 and concordance rate rang-ing from 92.59 to 98.18%. SNP-specific accuracy was consistent with that of sample imputation. S4 (40.32% of SNPs imputed) had higher accuracy than S2 and S3, both with less than 7.59% of SNPs imputed. Most probably, this was due to the high number of imputed SNPs with minor allele frequency (MAF) < 0.05 in S2 and S3 (by 18.43% and 16.06% higher than in S4, respectively). Therefore, for these two scenarios, MAF was more relevant than the panel density. These results suggest that genotype imputation using several commercially available SNP panels is feasible for the Portuguese national genomic evaluation. Abstract

1. The aim of this study was to investigate the associations between several carcass, performance and meat quality traits in broilers through factor analysis and use the latent variables (i.e. factors) as pseudo-phenotypes in genetic evaluations. 2. Factors were extracted using the principal components method and varimax rotation algorithm. Genetic parameters were estimated via Bayesian inference under a multiple-trait animal model. 3. All factors taken together explained 71% of the original variance of the data. The first factor, denominated as 'weight', was associated with carcass and body weight traits; and the second factor, defined as 'tenderness', represented traits related to water-holding capacity and shear force. The third factor, 'colour', was associated with traits related to meat colour, whereas the fourth, referenced as 'viscera', was related to heart, liver and abdominal fat. 4. The four biological factors presented moderate to high heritability (ranging from 0.35 to 0.75), which may confer genetic gains in this population. 5. In conclusion, it seems possible to reduce the number of traits in the genetic evaluation of broilers using latent variables derived from factor analysis. [ABSTRACT FROM AUTHOR]

Abstract

Summary: We aimed to estimate transgenerational epigenetic variance for body weight using genealogical and phenotypic information in meat quails. Animals were individually weighted from 1 week after hatching, with weight records at 7, 14, 21, 28, 35 and 42 days of age (BW7, BW14, BW21, BW28, BW35 and BW42, respectively). Single‐trait genetic analyses were performed using mixed models with random epigenetic effects. Variance components were estimated by the restricted maximum likelihood method. A grid search for values of autorecursive parameter (λ) ranging from 0 to 0.5 was used in the variance component estimation. This parameter is directly related to the reset coefficient (ν) and the epigenetic coefficient of transmissibility (1‐ν). The epigenetic effect was only significant for BW7. Direct heritability estimates for body weight ranged in magnitude (from 0.15 to 0.26), with the highest estimate for BW7. Epigenetic heritability was 0.10 for BW7, and close to zero for the other body weights. The inclusion of the epigenetic effect in the model helped to explain the residual and non‐Mendelian variability of initial body weight in meat quails. [ABSTRACT FROM AUTHOR]

Abstract

Cento e vinte e duas amostras de feijão (Phaseolus vulgaris L.) foram coletadas em 14 municípios das regiões serrana e sul do Espírito Santo, para serem caracterizadas e utilizadas em programas de melhoramento genético. As caracterizações pertinentes ao germoplasma coletado envolveram estudos relacionados ao grupo comercial, ao tamanho e à intensidade do brilho das sementes, bem como ao grau de mistura presente. Do total de amostras, 49 pertencem ao grupo comercial preto, 19 ao grupo vermelho, 19 ao grupo pardo e 35 a outros grupos comerciais. No tocante às sementes, 61,4% das amostras são de tamanho pequeno, com peso inferior a 25 g, e, na maioria, com brilho fosco ou opaco. Common bean [Phaseolus vulgaris] cv. BRS Horizonte was developed from the cross EMP 250/4/A769///A429/XAN 252//Pinto VI 114 by Embrapa Rice and Beans and the International Central for Tropical Agriculture -CIAT in Colombia, and was released for the southern and central western regions of Brazil for its erect growth habit, earliness, good lodging resistance, and resistance to 5 pathotypes of Colletotrichum lindemuthianum and to bean common mosaic virus.. This cultivar originated from a multiple cross performance at Embrapa Rice and Beans. After undergoing the bulk method in F2 and F3 and modified SSD in F4 and F5 line LM 93203304 was selected for its remarkable grain yield and erect plant type. Abstract

This study is aimed to develop an electroanalytical methodology using a boron-doped diamond electrode (BDD) associated with experimental design in order to determine simultaneously and selectively carbendazin (CBZ) and fenamiphos (FNP) pesticides. In previous studies oxidation peaks were observed at 1.10 V (CBZ) and 1.20 V (FNP), respectively, with characteristics of irreversible processes controlled by diffusion of species (in pH 2.0 (CBZ) and pH 3.5 (FNP)) using a BR buffer 0.1 mol L−1 as support electrolyte. The differences between the potentials for both pesticides, (about 100 mV) indicate the possibility of selective determination of FNP and CBZ. However, employing an equimolar mixture of analytes, the peaks overlap to form a single oxidation peak. Thus, we used a 34 full factorial design with four parameters to be analyzed in three levels, in order to obtain the optimized parameters for the separation of the peaks. The best separation conditions were pH 5.0, square wave frequency of 300 s−1, pulse amplitude of 10 mV and scan increment of 2 mV. These parameters were used to obtain the calibration curves of CBZ and FNP. For CBZ the analytical curve was obtained in the concentration range of 4.95×10−6 to 6.90×10−5 mol L−1 with good sensitivity and linearity (0.175 A/mol L−1 and 0.999, respectively). The limits of detection (LOD) and quantification (LOQ) were 1.6×10−6 mol L−1 and 5.5×10−6 mol L−1, respectively. For FNP the linear concentration interval was 4.95×10−6 to 3.67×10−5 mol L−1, with a sensitivity of 0,207 A/mol L−1 and linearity of 0.996. The LOD and LOQ were 4.1×10−6 mol L−1 and 13.7×10−6 mol L−1, respectively. Using these experimental conditions it was possible to separate the oxidation peaks of CBZ (Ep=1.08 V) and FNP (Ep=1.23 V). The electroanlytical method was applied in lemon juice samples. The recovery values were 110.0 % and 92.5 % for CBZ and FNP, respectively.
