Abstract Gaining the molecular understanding for myelination development and regeneration has been a long-standing goal in neurological research. Mutations in the transcription cofactor Mediator Med23 subunit are often associated with intellectual disability and white matter defects, although the precise functions and mechanisms of Mediator in myelination remain unclear. In this study, we generated a mouse model carrying an Med23Q649R mutation that has been identified in a patient with hypomyelination features. The MED23Q649R mouse model develops white matter thinning and cognitive decline, mimicking common clinical phenotypes. Further, oligodendrocyte-lineage specific Med23 knockout mice verified the important function of MED23 in regulating central nervous system myelination and postinjury remyelination. Utilizing the in vitro cellular differentiation assay, we found that the oligodendrocyte progenitor cells, either carrying the Q649R mutation or lacking Med23, exhibit significant deficits in their capacity to differentiate into mature oligodendrocytes. Gene profiling combined with reporter assays demonstrated that Mediator Med23 controls Sp1-directed gene programs related to oligodendrocyte differentiation and cholesterol metabolism. Integrative analysis demonstrated that Med23 modulates the P300 binding to Sp1-targeted genes, thus orchestrating the H3K27 acetylation and enhancer activation for the oligodendrocyte lineage progression. Collectively, our findings identified the critical role for the Mediator Med23 in oligodendrocyte fate determination and provide mechanistic insights into the myelination pathogenesis associated with MED23 mutations.

Abstract In this work, we investigate short-packet communications in power beacon (PB)-assisted Internet-of-Things (IoT) networks, where an energy-constrained actuator first harvests energy from a dedicated PB, and then transmits confidential signals to a desired controller in the presence of an eavesdropper. We derive a closed-form lower bound approximation expression for the average achievable effective secrecy rate (AESR) over Nakagami-m fading channels. To gain more insights, we also present the asymptotic average AESR in the high signal-to-noise ratio (SNR) regime. Specifically, analytical results indicate that an average AESR floor appears with the increase of SNR. Moreover, a low complexity one-dimensional search method is employed to maximize the average AESR by optimizing the energy harvesting length. Monte-Carlo simulations are provided to corroborate our analysis.

Abstract Glioma is the most prevalent malignant brain tumor in adults. The development of engineered nanomaterials (ENMs) has led to the emergence of innovative therapeutic strategies for gliomas. Therefore, our aim is to investigate the therapeutic effect of CuO nanoparticles (NPs) on glioma and provide data support for future research. The therapeutic effect of CuO NPs on glioma rats was explored through the detection of inflammatory factors, oxidase, pathological sections, immunofluorescence, neurotransmitter, glioma biomarker proteins and genes, and rat behavioral tests. Additionally, the application prospect of CuO NPs was evaluated by treating U87MG human glioma cell line. In this study, it was found that CuO NPs can alleviate the inflammatory reaction in the hippocampus tissue of glioma rats, promote the production of ·OH and lead to the up-regulation of catalase (CAT) and superoxide dismutase (SOD) enzyme activities. Treatment with CuO NPs also inhibited the expression of matrix metalloproteinase-9 (MMP-9) biomarkers in model rats and glioma cells. Moreover, it enhanced the release of neurotransmitters, which subsequently improved spatial recognition and memory ability of glioma rats. In conclusion, CuO NPs is a potential glioma treatment for ENMs, but still needs modification and modification strategies to improve its biocompatibility and targeted delivery.

Invasive tumors are difficult to be completely resected in clinical surgery due to the lack of clear resection margins, which greatly increases the risk of postoperative recurrence. However, chemotherapy and radiotherapy as the traditional means of postoperative adjuvant therapy, are limited in postoperative applications, such as multi-drug resistance and low sensitivity, etc. Therefore, an engineered magnesium alloy rod is designed as a postoperative implant to completely remove postoperative residual tumor tissue and inhibit tumor recurrence by gas and mild magnetic hyperthermia therapy (MMHT). As a reactive metal, magnesium alloy responds to the acidic tumor microenvironment by continuously generating hydrogen. The in-situ generation of hydrogen not only protects the surrounding normal tissue, but also enables the magnesium alloy to achieve MMHT under low-intensity alternating magnetic field (AMF). Furthermore, the numerous reactive oxygen species (ROS) produced by heat stress will combine with nitric oxide (NO) generated in situ, to produce more toxic reactive nitrogen species (RNS) storm. In summary, engineered magnesium alloy can completely remove residual tumor tissue and inhibit tumor recurrence by MMHT and RNS storm under low-intensity AMF, and the biodegradability of magnesium alloy makes great potential for clinical application.

To solve the problem of rapid following and advancing speed of support in the fully mechanized mining face, a distributed liquid supply system based on accumulators was proposed and tested in the S1204 working face in Ningtiaota Coal Mine. The speed of advancing support movement in the middle of working face was determined based on the coal mining process and capacity requirements, and a simulation model was established for the existing liquid supply system to simulate the advancing process of single, double, and triple supports, and the factors affecting the advancing time of supports were analyzed. The results showed that the following speed should reach 12 m/min to meet the capacity requirements which is 10 million tons per year of the working face. The double or triple support moving in groups was necessary to meet the requirements of advancing time. To solve the problem of large pressure fluctuations and easy support loss in this working condition, the distributed energy storage and pressure stabilization promoting scheme was proposed to balance the contradiction between insufficient instantaneous flow supply and excessive average flow supply capacity in the hydraulic system of the working face by installing “accumulator+check valve” on the hydraulic support. The design method of accumulators based on the flow compensation was also provided. The simulation results showed that after installing the accumulator, the advancing time for double and triple support movement in groups was reduced by 6.2% and 11.5% respectively. Especially after continuous interference, the advancing time for double support movement in groups could be reduce by 15%. The underground application results showed that after installing the distributed accumulators in the liquid supply system, the advancing speed of the supports was significantly improved, and the phenomenon of loss in group pulling was significantly reduced. It played an important supporting role in increasing the average daily footage of the working face from 16.7 to 19.8 cuts, with a normalized daily footage of 22 cuts, achieving an annual production capacity of 10 million tons. This study provides an effective solution method to achieve rapid movement of support in groups.

The petrographical, mineralogical, whole-rock geochemical, and zircon U-Pb geochronological and Lu-Hf isotopic studies were carried out for investigating the provenance and genetic mechanism of the high-grade coal-bearing kaolin within the coal seams of the Permo-Carboniferous Taiyuan Formation in the Xiaoyu mine of the Datong coalfield, Shanxi Province. The coal-bearing kaolin is composed mainly of well-ordered kaolinite with no detrital quartz, feldspars and muscovite. Some samples contain boehmite that were formed through the desilication of kaolinite. The zircons in the coal-bearing kaolin are typically igneous origin with only one age population of ca. 300 Ma, same with the depositional age of the coal, confirming that the coal-bearing kaolin was altered from volcanic ash. Besides, the coal-bearing kaolin commonly contains tabular/columnar and vermicular kaolinite that are also diagnostic feature of alteration from volcanic ash. These coarse-grained kaolinites are thought to be pseudomorphic kaolinite altered from crystal fragments in volcanic ash, such as plagioclase and biotite. The zircon εHf(t) is dominant by a wide range of negative values, indicative of their formation from magma generated by substantial remelting of ancient continental crust, coinciding with that of contemporaneous magmatic rocks formed from continental arc volcanism in the Inner Mongolia Paleo-Uplift at the northern margin of North China. This suggests their cognate relationship. The geochemical fingerprints of trace element of zircons also indicate that they were crystalized from continental arc magma. The northern margin of the North China Craton (NCC) is an Andean-type continental margin in the late Paleozoic. During the coal-accumulating period of the Late Carboniferous to Early Permian in North China, the Paleo-Asian Ocean (PAO) plate was continuously southward subducted beneath the NCC, resulted in a frequent continental arc volcanism. The emitted volcanic ash fell into peat swamps developed in a paralic environment in North China, which were subjected to an intensive leaching and altered to the coal-bearing kaolin.

To comprehensively assess the current state-of-art in asphalt foaming technology, the following four key aspects have been reviewed systematically: foaming principles, test methods, evaluation indicators, and influencing factors. Key findings reveal that asphalt foaming was primarily driven by the vaporization of water, with deterioration processes including bubble collapse and liquid film drainage. However, the current understanding of asphalt foaming principles remains limited, primarily due to difficulties in capturing and precisely measuring its microscopic behaviors during asphalt foaming process. Volume changes provided an intuitive means to evaluate the expansion capacity of asphalt and its foaming stability. Bubble evolution characteristics of foamed asphalt offered promising insights into its foaming performance. Traditional ruler and stopwatch-based assessments were being superseded by automated techniques like laser and ultrasonic ranging. Nevertheless, the current measuring equipment still lacks the capability to comprehensively evaluate the foaming effect of asphalt across various dimensions. Asphalt temperature and foaming water consumption significantly affected asphalt foaming performance, and the inclusion of foaming agents typically led to a notable increase in the half life of foamed asphalt. However, the interaction between foaming agents and asphalt, as well as the underlying mechanisms affecting the foaming effect, are still unclear and require further exploration. Future research should primarily focus on the correlation between asphalt foaming effect and mixture performance, aiming to guide the practical engineering application of foamed asphalt mixtures and enlarge the advantages of such low-emission and sustainable mixtures.

Abstract Background The plasma concentrations of acyl coenzyme A binding protein (ACBP, also known as diazepam-binding inhibitor, DBI, or ‘endozepine’) increase with age and obesity, two parameters that are also amongst the most important risk factors for cancer. Methods We measured ACBP/DBI in the plasma from cancer-free individuals, high-risk patients like the carriers of TP53 or BRCA1/2 mutations, and non-syndromic healthy subjects who later developed cancer. In mice, the neutralization of ACBP/DBI was used in models of non-small cell lung cancer (NSCLC) and breast cancer development and as a combination treatment with chemoimmunotherapy (chemotherapy + PD-1 blockade) in the context of NSCLC and sarcomas. The anticancer T cell response upon ACBP/DBI neutralization was characterized by flow cytometry and single-cell RNA sequencing. Results Circulating levels of ACBP/DBI were higher in patients with genetic cancer predisposition (BRCA1/2 or TP53 germline mutations) than in matched controls. In non-syndromic cases, high ACBP/DBI levels were predictive of future cancer development, and especially elevated in patients who later developed lung cancer. In preclinical models, ACBP/DBI neutralization slowed down breast cancer and NSCLC development and enhanced the efficacy of chemoimmunotherapy in NSCLC and sarcoma models. When combined with chemoimmunotherapy, the neutralizing monoclonal antibody against ACBP/DBI reduced the frequency of regulatory T cells in the tumor bed, modulated the immune checkpoint profile, and increased activation markers. Conclusion These findings suggest that ACBP/DBI acts as an endogenous immune suppressor. We conclude that elevation of ACBP/DBI constitutes a risk factor for the development of cancer and that ACBP/DBI is an actionable target for improving cancer immunosurveillance.

CO2 conversion is gradually seen as a better way for society to effectively use carbon sources and avoid climate crisis associated with fossil CO2 emissions. And the decision to deploy CO2 technology scale should be relied on its environmental impact. In this work, life cycle assessment model evaluates the environmental performance of CO2 conversion by photocatalytic reaction process with two different catalysts (NiAl-LDH and Co-ZIF-9). Six impact categories considered in this analysis, including climate change, acidification potential, depletion of abiotic resources, eutrophication potential, ozone layer depletion potential, and photochemical oxidation potential. Results indicated that CO2 conversion with Co-ZIF-9 photocatalyst has a better environmental impact than the NiAl-LDH photocatalyst route. Moreover, the Co-ZIF-9 catalyst scenario also has a lower total environmental burden than the conventional CO production route. Sensitivity analysis shows that recycle performance of the catalyst is highly sensitive to the production process in two scenarios. This study could provide a framework for robust decisions in CO2 conversion by photocatalytic reaction, which is useful for policymakers to decide the feasibility of industrialization.

Aims: Precise implant positioning, tailored to individual spinopelvic biomechanics and phenotype, is paramount for stability in total hip arthroplasty (THA). Despite a few studies on instability prediction, there is a notable gap in research utilizing artificial intelligence (AI). The objective of our pilot study was to evaluate the feasibility of developing an AI algorithm tailored to individual spinopelvic mechanics and patient phenotype for predicting impingement. Methods: This international, multicentre prospective cohort study across two centres encompassed 157 adults undergoing primary robotic arm-assisted THA. Impingement during specific flexion and extension stances was identified using the virtual range of motion (ROM) tool of the robotic software. The primary AI model, the Light Gradient-Boosting Machine (LGBM), used tabular data to predict impingement presence, direction (flexion or extension), and type. A secondary model integrating tabular data with plain anteroposterior pelvis radiographs was evaluated to assess for any potential enhancement in prediction accuracy. Results: We identified nine predictors from an analysis of baseline spinopelvic characteristics and surgical planning parameters. Using fivefold cross-validation, the LGBM achieved 70.2% impingement prediction accuracy. With impingement data, the LGBM estimated direction with 85% accuracy, while the support vector machine (SVM) determined impingement type with 72.9% accuracy. After integrating imaging data with a multilayer perceptron (tabular) and a convolutional neural network (radiograph), the LGBM’s prediction was 68.1%. Both combined and LGBM-only had similar impingement direction prediction rates (around 84.5%). Conclusion: This study is a pioneering effort in leveraging AI for impingement prediction in THA, utilizing a comprehensive, real-world clinical dataset. Our machine-learning algorithm demonstrated promising accuracy in predicting impingement, its type, and direction. While the addition of imaging data to our deep-learning algorithm did not boost accuracy, the potential for refined annotations, such as landmark markings, offers avenues for future enhancement. Abstract Non-small cell lung cancer (NSCLC), being the most prevalent and lethal malignancy affecting the lungs, poses a significant threat to human health. This research aims at illustrating the precise role and related mechanisms of silent information regulator type-1 (SIRT1) in NSCLC progression. The expression pattern of SIRT1 in NSCLC cell lines was examined using quantitative real-time polymerase chain reaction and western blotting. Functional assays in NSCLC cell lines validated the biological capabilities of SIRT1 on malignant phenotypes, and its impact on tumorigenicity was further evaluated in vivo. In addition, the FOXO1 inhibitor AS1842856 was applied to verify the role of SIRT1 on FOXO pathway in vitro. SIRT1 expression was prominently elevated in NSCLC cell lines. The depletion of SIRT1 retarded the capabilities of proliferation, migration and invasion, while enhancing apoptosis in NSCLC cells. Furthermore, SIRT1 silencing restricted the tumorigenesis of NSCLC in vivo. Additionally, AS1842856 treatment ameliorated the inhibitory effect of SIRT1 deficiency on malignant phenotypes in NSCLC cells. SIRT1 deletion exerted an anti-oncogenic role in NSCLC via activation of FOXO1.

Abstract The search of the new physics (NP) beyond the Standard Model is one of the most important topics in current high energy physics. With the increasing luminosities at the colliders, the search for NP signals requires the analysis of more and more data, and the efficiency in data processing becomes particularly important. As a machine learning algorithm, support vector machine (SVM) is expected to to be useful in the search of NP. Meanwhile, the quantum computing has the potential to offer huge advantages when dealing with large amounts of data, which suggests that quantum SVM (QSVM) is a potential tool in future phenomenological studies of the NP. How to use SVM and QSVM to optimize event selection strategies to search for NP signals are studied in this paper. Taking the tri-photon process at a muon collider as an example, it can be shown that the event selection strategies optimized by the SVM and QSVM are effective in the search of the dimension-8 operators contributing to the anomalous quartic gauge couplings.

Abstract Background Immunosurveillance is pivotal in the effectiveness of anticancer therapies and tumor control. The ineffectiveness of cisplatin in activating the immunosurveillance is attributed to its lack of adjuvanticity resulting from its inability to stimulate endoplasmic reticulum stress. Dihydroartemisinin demonstrates the anti-tumor effects through various mechanisms, including the activation of the endoplasmic reticulum stress. This study aimed to develop a novel strategy to enhance the immunogenicity of dying tumor cells by combining cisplatin with dihydroartemisinin, thereby triggering effective anti-tumor immunosurveillance and improving the efficacy of cisplatin in clinical practice. Methods Lewis lung carcinoma (LLC) and CT26 colon cancer cell lines and subcutaneous tumor models were used in this study. The importance of immunosurveillance was validated in both immunocompetent and immunodeficient mouse models. The ability of dihydroartemisinin and cisplatin therapy to induce immunogenic cell death and tumor growth control in vivo was validated by prophylactic tumor vaccination and therapeutic tumor models. The underlying mechanism was elucidated through the pharmaceutical or genetic intervention of the PERK/eIF2α pathway in vitro and in vivo. Results Dihydroartemisinin enhanced the generation of reactive oxygen species in cisplatin-treated LLC and CT26 cancer cells. The combination treatment of dihydroartemisinin with cisplatin promoted cell death and ensured an optimal release of damage-associated molecular patterns from dying cancer cells, promoting the phagocytosis of dendritic cells. In the tumor vaccination model, we confirmed that dihydroartemisinin plus cisplatin treatment induced immunogenic cell death. Utilizing immunocompetent and immunodeficient mouse models, we further demonstrated that the combination treatment suppressed the tumor growth of CT26 colon cancer and LLC lung cancer, leading to an improved prognosis through the restoration of cytotoxic T lymphocyte responses and reinstatement of anti-cancer immunosurveillance in vivo. Mechanistically, dihydroartemisinin restored the immunogenicity of cisplatin by activating the adjuvanticity of damage-associated molecular patterns, such as calreticulin exposure, through the PERK/eIF2α pathway. Additionally, the inhibition of eIF2α phosphorylation attenuated the anti-tumor efficiency of C + D in vivo. Conclusions We highlighted that dihydroartemisinin acts as an immunogenic cell death rescuer for cisplatin, activating anticancer immunosurveillance in a PERK/eIF2α-dependent manner and offering a strategy to enhance the anti-tumor efficacy of cisplatin in clinical practice.

Abstract Purpose To explore the impact of mobile social media on the psychological well-being of Chinese international students and analyze the mechanisms of influence to enhance their overall psychological health and social interactions in a foreign environment. Methods Convenience sampling was employed, using questionnaires on Mobile Social Media, Psychological Resilience, Body Image, Health Goal Setting, Physical Activity Level, and Mental Health Status as measurement tools. Data were gathered from 378 Chinese international students across 33 universities in South Korea, including Kangwon National University, Myongji University, Kunsan National University, Seoul National University, and Chonbuk National University. Confirmatory factor analysis, correlation analysis, common method bias testing, and chain mediation effect analysis were conducted using SPSS and AMOS 23.0. Results Mobile social media has significant indirect effects on the mental health of international students through various factors: psychological resilience and physical activity level (effect ‘adg’ = 0.080, 95% CI [0.029, 0.144]), body image and physical activity level (effect ‘beg’ = 0.122, 95% CI [0.044, 0.247]), and health goal setting and physical activity level (effect ‘cfg’ = 0.255, 95% CI [0.123, 0.428]). Conclusion The study shows that mobile social media benefits the mental health of Chinese international students by enhancing psychological resilience, physical activity, body image perception, and health goal setting. Collaboration between educational institutions and social media platforms is recommended to promote physical activity among international students. This collaboration can involve sharing encouraging messages, joining health communities, setting goals, and providing accessible exercise resources. Utilizing mobile apps or social media for tracking progress and goal-setting can also improve self-management skills.

Abstract Objective To investigate the accuracy and consistency of MDCT and TEE in the preoperative assessment of aortic root surgery. Methods From January 2021 to September 2022, 118 patients who underwent aortic root surgery were included in this study. All patients underwent high-quality preoperative MDCT and TEE examinations, and the examination results were independently measured and assessed by two senior radiologists or ultrasound specialists. Bland–Altman analysis and Pearson correlation testing were employed to assess the correlation and consistency between MDCT and TEE. These analyses were then compared with actual intraoperative measurement data. Results Among all the patients, 73 (61.86%) had tricuspid aortic valve (TAV), and 45 (38.14%) had bicuspid aortic valve (BAV). A comparison between the TEE and MDCT measurements showed that for the annulus diameter, the area-derived diameter had the best correlation and agreement. For the sinus of Valsalva diameter, the circumference-derived diameter was optimal. However, for the STJ diameter, the minimum cross-sectional diameter showed the best agreement with TEE. In contrast, measurements of geometric height showed a weaker correlation and agreement. Conclusion Contrast-enhanced MDCT can be a valuable tool for perioperative evaluation in aortic root surgery, with good correlation, consistency, and feasibility when compared to TEE. The choice of MDCT measurement methodology, specifically area-derived and circumference-derived diameter, proved to be more accurate than other methods. Further research is required to enhance the understanding of aortic valve repair and associated imaging techniques.

Abstract As a new grinding and maintenance technology, rail belt grinding shows significant advantages in many applications. The dynamic characteristics of the rail belt grinding vehicle largely determines its grinding performance and service life. In order to explore the vibration control method of the rail grinding vehicle with abrasive belt, the vibration response changes in structural optimization and lightweight design are respectively analyzed through transient response and random vibration simulations in this paper. Firstly, the transient response simulation analysis of the rail grinding vehicle with abrasive belt is carried out under operating conditions and non-operating conditions. Secondly, the vibration control of the grinding vehicle is implemented by setting vibration isolation elements, optimizing the structure, and increasing damping. Thirdly, in order to further explore the dynamic characteristics of the rail grinding vehicle, the random vibration simulation analysis of the grinding vehicle is carried out under the condition of the horizontal irregularity of the American AAR6 track. Finally, by replacing the Q235 steel frame material with 7075 aluminum alloy and LA43M magnesium alloy, both vibration control and lightweight design can be achieved simultaneously. The results of transient dynamic response analysis show that the acceleration of most positions in the two working conditions exceeds the standard value in GB/T 17426-1998 standard. By optimizing the structure of the grinding vehicle in three ways, the average vibration acceleration of the whole car is reduced by about 55.1% from 15.6 m/s2 to 7.0 m/s2. The results of random vibration analysis show that the grinding vehicle with Q235 steel frame does not meet the safety conditions of $$3\sigma$$ 3 σ . By changing frame material, the maximum vibration stress of the vehicle can be reduced from 240.7 MPa to 160.0 MPa and the weight of the grinding vehicle is reduced by about 21.7% from 1500 kg to 1175 kg. The modal analysis results indicate that the vibration control of the grinding vehicle can be realized by optimizing the structure and replacing the materials with lower stiffness under the premise of ensuring the overall strength. The study provides the basis for the development of lightweight, diversified and efficient rail grinding equipment.

Abstract Background ICU nurses provide critical care and meticulously document electronic medical records (EMRs), tracking vital signs, interventions, and medication hourly. Despite China’s ICUs effectively integrating real-time monitor and ventilator data into EMRs, challenges persist. Patient movements can introduce inaccuracies, and the demands of critical care may lead nurses to miss assessments like pain and nutrition. Traditional manual EMR verification is inefficient and error-prone, highlighting the urgent need for standardized, technology-aided EMR practices in ICU nursing. Objective This study aimed to describe the development and evaluation of an electronic medical records quality control system implemented in a Chinese tertiary care ICU setting, where current practices impact the accuracy of electronic medical records. Methods A prospective controlled trial was conducted with 600 ICU patients in Zhejiang Province from January to December 2023. An automated EMR quality control system was implemented in July 2023, facilitating real-time data collection and quality control for vital signs, medication management, and nursing evaluations. Results After implementing the ICU nursing electronic medical record quality control system, the prevalence of false data on vital signs decreased from 9 to 1.33%. Additionally, the incidence of incomplete medication administration dropped from 3.33 to 1.67%, and the rate of missing evaluations of assessment items in EMRs was reduced from 8 to 1.33%. Besides, the average time spent on quality control of the electronic medical records was 62 (48,76) seconds per record, which was significantly lower than the 264 (195.5,337.5) seconds using the traditional method. The nurses’ satisfaction with the nursing electronic medical record quality control was (105.73 ± 9.31). Conclusions The ICU nursing electronic medical record quality control system has led to substantial improvements in the quality and reliability of EMRs. The reduction in false data on vital signs, instances of incomplete medication administration, and missing evaluations of assessment items demonstrates the system’s positive impact on nursing documentation practices. These improvements not only enhance the accuracy of patient records but also contribute to better patient care and safety within the ICU setting.

To achieve the rapid and real-time detection of triethylamine (TEA) gas, this study synthesized a gas sensor based on heterostructures of Fe2O3/MoO3 using a hydrothermal method. Fe2O3/MoO3 composites with a narrow bandgap (1.1 eV) were successfully synthesized by constructing heterostructures. The rapid and efficient detection of triethylamine was achieved at 220 °C. The response and response/recovery times of the Fe2O3/MoO3 sensor with 50 × 10−6 triethylamine were 132 s and 5 s/10 s, respectively. The Fe2O3/MoO3 sensor maintained a good response to triethylamine gas, even at 80% relative humidity. The sensing mechanism of the Fe2O3/MoO3 sensor can be described in terms of adsorption energy and electronic behavior of the sensing layer using density functional theory (DFT). The results are consistent with the excellent selectivity and rapid response/recovery of the Fe2O3/MoO3 gas sensor for triethylamine. Therefore, the construction of heterostructures to facilitate electron transmission is an effective strategy to achieve rapid detection of triethylamine and is worthy of further exploration and investigation.

Metal Nanoclusters (MNCs) have shown great potential applications in the field of food safety due to their unique physical and chemical properties. Through the template synthesis method, the specific size, shape and dispersion of MNCs can be regulated, thereby improving their functional characteristics. Based on the recent studies on MNCs, the mechanism of template-assisted synthesis of MNCs and their applications in the detection of food contaminants, food packaging materials, antimicrobial agents and food additives are reviewed. It provides a scientific basis for template synthesis of MNCs, and to provide insights and directions for MNCs in food safety monitoring.

Abstract To explore the multiparameter precursor characteristics of pre- and post-coal burst. Based on a coal burst of LW 1305 in the Zhaolou Coal Mine, an early warning method combining stress‒strain curve and microseismic multiparameter is proposed. The research results show that coal burst was induced by the intrinsic static high-stress concentration and the strong external impact loading generated by fracturing of the key stratum. The precursors mainly characterize the enhancement trend of the S value, the sudden and sharp increase in the A(t) value, the continuous and abnormal decrease in the b value, the increasing absolute value of Z sharply and larger than 2, the continuous and abnormal decrease in the Qt value, and the dominant frequency moving to the low-frequency band. Essentially, many micro-fissures inside the key stratum initiated, converged and connected to form macro-fractures, which was verified by the attenuation rate of the K value. Considering the time-varying effect of the overlying stratum movement, the curves of the six parameters agree well with those of stress vs. strain, which indicates that it is reasonable to take the observed zone as a whole system to investigate the variation in the multiple parameters and fracturing of the key stratum. The research results can be applied to the monitoring, early warning and control of coal burst so that effective safety measures can be taken in real time.

Objective Percutaneous repair is an alternative to open surgical repair of the Achilles tendon with comparable, functional results and low re‐rupture and infection rates; however, sural nerve injury is a known complication. The purpose of this study is to design a new surgical procedure, the minimally invasive peritendinous submembrane access technique (MIS‐PSAT). It offers optimal results, with excellent functional outcomes, and with minimal soft tissue complications and sural nerve injury. Methods This retrospective study included 249 patients with acute closed Achilles tendon ruptures treated at our institution between 2009 and 2019. All patients underwent MIS‐PSAT at our institution and were followed up for 8–48 months. Functional evaluation was based on the Achilles tendon total rupture score (ATRS) and the American Orthopedic Foot and Ankle Society Ankle‐Hindfoot Scale (AOFAS‐AHS), associated with local complications and isokinetic tests. Results None of the patients had infection, necrosis, or sural nerve injury. Re‐rupture occurred in two cases. The average times to return to work and sports was 10.4 and 31.6 weeks, respectively. The average ATRS and AOFAS‐AHS scores were 90.2 and 95.7, respectively, with an excellent rate of 99.5%. Isokinetic tests showed that ankle function on the affected side was comparable with that on the healthy side (p > 0.05). Conclusion The MIS‐PSAT for acute Achilles tendon rupture is easy to perform with few complications. Importantly, the surgical technique reduces the risk of sural nerve injuries. Patients have high postoperative satisfaction, low re‐rupture rates, and muscle strength, and endurance can be restored to levels similar to those on the healthy side.

It is of great scientific and economic value to recycle waste poly (ethylene terephthalate) (PET) into high-value PET-based metal organic frameworks (MOFs) and further convert it into porous carbon for green energy storage applications. In the present study, a facile and cost-effective hydrothermal process was developed to direct recycle waste PET bottles into MIL-53(Al) with a 100% conversation, then the MOF-derived porous carbon was assembled into electrodes for high-performance supercapacitors. The results indicated that the as-synthesized carbon exhibited high SSA of 1712 m2 g−1 and unique accordion-like structure with hierarchical porosity. Benefit to these advantageous characters, the assembled three-electrode supercapacitor displayed high specific capacitances of 391 F g−1 at the current density of 0.5 A g−1 and good rate capability of 73.6% capacitance retention at 20 A g−1 in 6 mol L−1 KOH electrolyte. Furthermore, the assembled zinc ion capacitor still revealed outstanding capacitance of 335 F g−1 at 0.1 A g−1, excellent cycling stability of 92.2% capacitance retention after 10 000 cycles and ultra-high energy density of 150.3 Wh kg−1 at power density of 90 W kg−1 in 3 mol L−1 ZnSO4 electrolyte. It is believed that the current work provides a facile and effective strategy to recycle PET waste into high-valuable MOF, and further expands the applications of MOF-derived carbons for high-performance energy storage devices, so it is conducive to both pollution alleviation and sustainable economic development.

Abstract Background Ankylosing spondylitis (AS) with radiographic damage is more prevalent in men than in women. IL-17, which is mainly secreted from peripheral blood mononuclear cells (PBMCs), plays an important role in the development of AS. Its expression is different between male and female. However, it is still unclear whether sex dimorphism of IL-17 contribute to sex differences in AS. Methods GSE221786, GSE73754, GSE25101, GSE181364 and GSE205812 datasets were collected from the Gene Expression Omnibus (GEO) database. Differential expressed genes (DEGs) were analyzed with the Gene Set Enrichment Analysis (GSEA), Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) methods. CIBERSORTx and EcoTyper algorithms were used for immune infiltration analyses. Machine learning based on the XGBoost algorithm model was used to identify the impact of DEGs. The Connectivity Map (CMAP) database was used as a drug discovery tool for exploring potential drugs based on the DEGs. Results According to immune infiltration analyses, T cells accounted for the largest proportion of IL-17-secreting PBMCs, and KEGG analyses suggested an enhanced activation of mast cells among male AS patients, whereas the expression of TNF was higher in female AS patients. Other signaling pathways, including those involving metastasis-associated 1 family member 3 (MAT3) or proteasome, were found to be more activated in male AS patients. Regarding metabolic patterns, oxidative phosphorylation pathways and lipid oxidation were significantly upregulated in male AS patients. In XGBoost algorithm model, DEGs including METRN and TMC4 played important roles in the disease process. we integrated the CMAP database for systematic analyses of polypharmacology and drug repurposing, which indicated that atorvastatin, famciclocir, ATN-161 and taselisib may be applicable to the treatment of AS. Conclusions We analyzed the sex dimorphism of IL-17-secreting PBMCs in AS. The results showed that mast cell activation was stronger in males, while the expression of TNF was higher in females. In addition, through machine learning and the CMAP database, we found that genes such as METRN and TMC4 may promote the development of AS, and drugs such as atorvastatin potentially could be used for AS treatment.

Objective: This study aims to explore the relationship between different endometrial preparations and pregnancy outcomes among patients with regular ovulatory cycles in order to find the best endometrial preparation methods in the freeze–thaw embryo transfer (FET) cycle. Materials and Methods: This is a retrospective study to investigate FET pregnancy outcomes in women who had a regular menstrual cycle, were younger than 35 years old, and underwent a modified natural cycle (mNC), ovulation induction (OI), or a hormone replacement treatment (HRT) cycle. A total of 1071 frozen cycles were included for analysis. Results: The implantation rate and live birth rate (LBR) in the OI group show a significant difference when compared to the mNC and HRT groups (P < 0.01). After adjusting for confounding factors, the logistic regression analysis revealed that the number of embryos transferred, the embryo stage, and quality were significantly associated with clinical pregnancy rate and LBR. The LBR was additionally affected by the mode of the endometrial preparation; the OI cycle could increase LBR. Conclusions: Endometrial preparation methods affect the LBR in women with a regular menstrual cycle. The OI cycle had an advantage in the LBR of FET.

Abstract Currently, the obstacle avoidance control of patrol robots based on intelligent vision lacks professional controller module assistance. Therefore, this paper proposes a design method of intelligent controller for obstacle avoidance and navigation of electrical inspection mobile robot based on PLC control. The controller designs a laser range finder to determine the required position of electrical patrol inspection. Use PLC as the core controller, and combine sensors, actuators, communication module and PLC selection module in the process of hardware design to achieve autonomous navigation and obstacle avoidance functions of the robot. Then design the software including the PLC compiler system and the virtual machine module. Based on the above steps, design the control module of obstacle avoidance navigation, which realizes the key link of robot autonomous navigation. The test results show that the controller can successfully avoid obstacles, improve the efficiency and quality of inspection, and achieve accurate and fast obstacle avoidance navigation for the electrical inspection mobile robot.

Abstract Kagome vanadates AV3Sb5 display unusual low-temperature electronic properties including charge density waves (CDW), whose microscopic origin remains unsettled. Recently, CDW order has been discovered in a new material ScV6Sn6, providing an opportunity to explore whether the onset of CDW leads to unusual electronic properties. Here, we study this question using angle-resolved photoemission spectroscopy (ARPES) and scanning tunneling microscopy (STM). The ARPES measurements show minimal changes to the electronic structure after the onset of CDW. However, STM quasiparticle interference (QPI) measurements show strong dispersing features related to the CDW ordering vectors. A plausible explanation is the presence of a strong momentum-dependent scattering potential peaked at the CDW wavevector, associated with the existence of competing CDW instabilities. Our STM results further indicate that the bands most affected by the CDW are near vHS, analogous to the case of AV3Sb5 despite very different CDW wavevectors.

Abstract Background Chronic thromboembolic pulmonary hypertension (CTEPH) is a progressive pulmonary vascular disorder with substantial morbidity and mortality, also a disease underdiagnosed and undertreated. It is potentially curable by pulmonary endarterectomy (PEA) in patients with surgically accessible thrombi. Balloon pulmonary angioplasty (BPA) and targeted medical therapy are options for patients with distal lesions or persistent/recurrent pulmonary hypertension after PEA. There is an urgent need to increase the awareness of CTEPH. Qualified CTEPH centers are still quite limited. Baseline characteristics, management pattern and clinical outcome of CTEPH in China needs to be reported. Methods and design The CHinese reAl-world study to iNvestigate the manaGEment pattern and outcomes of chronic thromboembolic pulmonary hypertension (CHANGE) study is designed to provide the multimodality treatment pattern and clinical outcomes of CTEPH in China. Consecutive patients who are ≥ 14 year-old and diagnosed with CTEPH are enrolled. The diagnosis of CTEPH is confirmed in right heart catheterization and imaging examinations. The multimodality therapeutic strategy, which consists of PEA, BPA and targeted medical therapy, is made by a multidisciplinary team. The blood sample and tissue from PEA are stored in the central biobank for further research. The patients receive regular follow-up every 3 or 6 months for at least 3 years. The primary outcomes include all-cause mortality and changes in functional and hemodynamic parameters from baseline. The secondary outcomes include the proportion of patients experiencing lung transplantation, the proportion of patients experiencing heart and lung transplantation, and changes in health-related quality of life. Up to 31 December 2023, the study has enrolled 1500 eligible patients from 18 expert centers. Conclusions As a real-world study, the CHANGE study is expected to increase our understanding of CTEPH, and to fill the gap between guidelines and the clinical practice in the diagnosis, assessment and treatment of patients with CTEPH. Abstract Colon cancer (CC) is a highly malignant tumor with a high incidence and poor prognosis. This study aimed to explore the function and molecular mechanisms of activating transcription factor 4 (ATF4) in CC. The expression levels of ATF4, GCN2, and ASNS in CC tissues were measured using immunohistochemistry (IHC) and reverse transcription quantitative PCR (RT-qPCR). Cell counting kit-8 (CCK-8), clone formation, transwell, and flow cytometry assays were conducted to assess cell viability, clonogenicity, migration, invasion, cell cycle, and apoptosis, respectively, in the ATF4 knockdown and overexpression SW480 cell lines. The effect of ATF4 on the expression of GCN2 and ASNS was detected using RT-qPCR, Chip-qPCR, and western blotting. ATF4, GCN2, and ASNS were expressed at low levels in CC tissues, and all had a significant negative correlation with tumor diameter. ATF4 knockdown promoted cell proliferation, invasion, and S-phase cell cycle and inhibited apoptosis in SW480 cells. In contrast, ATF4 overexpression had the opposite effect. Furthermore, ATF4 overexpression enhanced ATF4 binding to the ASNS promoter region. ATF4 knockdown significantly inhibited the expression of p-GCN2 and ASNS, whereas ATF4 overexpression significantly upregulated their expression. ATF4 inhibited CC cell viability, clone formation ability, migration, and invasion and promoted apoptosis, possibly by regulating the expression of p-GCN2 and ASNS. Our study provides a novel potential therapeutic target for the treatment of CC.

Supergravity is a noncontact volumetric force; when there is a density difference between the two phases, supergravity can strengthen their separation. The removal of nonmetallic inclusions in steel by supergravity technology not only does not cause molten steel to stir back to the mixing but also promotes the rapid floating of inclusions and shortens their floating time in the molten steel. Small inclusions that are difficult to remove can be removed using a supergravity field. The shape of the inclusions affects their floating behavior in molten steel. To study this effect, this study uses the fluid–structure interaction method to track the computational fluid–solid interface state, constructs three inclusions with different aspect ratios in a two-dimensional longitudinal section, and performs simulation studies of the floating behavior of the inclusions in a supergravity field. The effects of the different initial angles of the inclusions on their floating behavior were also compared. The simulation results show that the floating velocity of the inclusions is related to their shape and floating angle, and the faster the floating velocity, the closer the aspect ratio is to 1, or the floating angle is too vertical. For a given length, inclusions with a larger equivalent diameter float faster. In the supergravity field of G = 1000, the inclusions with a length of 1 μm did not undergo rotation; the inclusions with lengths of 10 and 20 μm rotated from the initial angle (45°, 90°) to the horizontal and then floated steadily. The floating velocity of inclusions is related to the real-time angle of inclusions. The rotational state of the inclusions gradually decreases as the gravity coefficient decreases. When G = 50, the inclusions with a length of 20 μm (initial angle of 90°) fail to rotate completely, which also proves that the inclusions are more likely to rotate when initially inclined instead of vertically. In addition, it was concluded that supergravity does not cause anisotropy in steel properties and that the method of removing inclusions from steel by supergravity does not have a substantial negative effect on steel properties. Finally, this study noted that the prediction of the supergravity treatment time using this model should be based on the floating velocity of the inclusions in the horizontal state, and an approximate time for the floating removal of the inclusions is provided based on this conclusion.

Cold mixed epoxy asphalt mixture (CMEAM) has excellent anti-deformation and fatigue performance which shows remarkable superiority on bridge pavements. Previous studies have done comprehensive researches on its properties through macroscopic experiments. However, few studies revealed the impact of gradation design on the road property of CMEAM through microscopic method. Thus, in this paper, X-Ray computed tomography (X-ray CT), nanoindentation (NI), field emission scanning electron microscopy (FESEM) and mercury intrusion porosimetry (MIP) are introduced to investigate the coarse aggregates contact degree, the thickness and elastic modulus of interfacial transition zone (ITZ) and the pore diameter distribution of CMEAM. Results showed that the contact points of coarse aggregates in EA-05 mixture are around 40 while those in EA-10 mixture are around 20. (40°, 50°) is the key inclination angle interval for EA-10 mixture type while (70°, 80°) and (10°, 20°) are of significance for EA-05 mixture type. The contact points and inclination angles are positively and inversely proportional to Marshall stability, respectively. In addition, the ITZ thickness of EA-05 and EA-10 mixture is below 10 μm and above 40 μm, respectively. The ITZ elastic modulus of EA-10 mixture is approximately 1.5 times than that of EA-05. The thickness and elastic modulus of ITZ are inversely proportional to low-temperature indirect tensile strain (εT). Meanwhile, EA-05 mixture has higher proportion in gel micropores and middle capillary pores while EA-10 mixture has higher proportion in mesopores and large capillary pores. Mesopores has the highest correlation coefficients with tensile strength ratio (TSR), followed by large capillary pores. The suggested proportion for above two type pores is around 21.5% and 51.0%, respectively.

Previous studies have delved into slope break zones in faulted basins, yet foreland basins, with their gentle slopes toward the forebulge, underdeveloped large-scale structures, and small terrain undulations, have received limited research on slope-fault belts. During the deposition periods of the 4th and 5th members of the Late Triassic Xujiahe Formation, a foreland basin developed in the Sichuan Basin. Scholars have noted the control exerted by the gentle slope break zone on the sedimentary lithofacies in the central and western Sichuan, particularly in the west of the Luzhou-Kaijiang Paleo-uplift, but related studies remain limited. By interpreting the tectonic slope break zones in seismic reflection profiles of the Sichuan Basin and incorporating existing regional geological data, this study employed particle flow numerical simulation to clarify the types of slope break zones in the central and western Sichuan regions, as well as their relationship with gravity sliding structures. It also analyzed the characte-ristics of new types of oil and gas traps associated with gravity-sliding structures. The findings are as follows: (1) The study area developed a fault slope break zone, with each side bounded by a "front squeezing and rear extension" tectonic combinations formed by gravity sliding. The tectonic deformation is weaker than those of gravity-sliding structures at passive continental margins. (2) The edge fault of the tectonic slope break zone controls the sedimentary facies and sand body types. (3) In the gentle slope zone, the stretching area of gravity-sliding structures lead to the uplift of the footwalls of normal faults, forming syndepositional anticlines with developed sandstones. These sandstones, juxtaposed with organic-rich mudstones within the contemporaneous fault depression, form "side-generated and lateral storage" hydrocarbon reservoirs. (4) In the steep slope zone, gravity sliding forms syndepositional anticlines in the 5th member of the Xujiahe Formation, which are bounded by flank mudstones and post-sliding deposited mudstones, forming in-source sand body reservoirs. (5) Syndepositional anticline hydrocarbon reservoirs related to gravity-sliding structures represent a new type of in-source hydrocarbon accumulation. The 5th member of the Xujiahe Formation in the study area likely contains such reservoirs, distributed in a NE-trending pattern parallel to the slope break zone.

Abstract Background Left ventricular global longitudinal strain (GLS) holds greater diagnostic and prognostic value than left ventricular ejection fraction (LVEF) in the heart failure (HF) patients. The triglyceride-glucose (TyG) index serves as a reliable surrogate for insulin resistance (IR) and is strongly associated with several adverse cardiovascular events. However, there remains a research gap concerning the correlation between the TyG index and GLS among patients with chronic heart failure (CHF). Method 427 CHF patients were included in the final analysis. Patient demographic information, along with laboratory tests such as blood glucose, lipids profiles, and echocardiographic data were collected. The TyG index was calculated as Ln [fasting triglyceride (TG) (mg/dL) × fasting plasma glucose (FPG) (mg/dL)/2]. Results Among CHF patients, GLS was notably lower in the higher TyG index group compared to the lower TyG index group. Following adjustment for confounding factors, GLS demonstrated gradual decrease with increasing TyG index, regardless of the LVEF level and CHF classification. Conclusion Elevated TyG index may be independently associated with more severe clinical left ventricular dysfunction in patients with CHF.

Abstract Micropeptides hidden in long non-coding RNAs (lncRNAs) have been uncovered to program various cell-biological changes associated with malignant transformation-glioblastoma (GBM) cascade. Here, we identified and characterized a novel hidden micropeptide implicated in GBM. We screened potential candidate lncRNAs by establishing a workflow involving ribosome-bound lncRNAs, publicly available MS/MS data, and prognosis-related lncRNAs. Micropeptide expression was detected by western blot (WB), immunofluorescence (IF), and immunohistochemistry (IHC). Cell proliferation rate was assessed by calcein/PI staining and EdU assay. Proteins interacted with the micropeptide were analyzed by proteomics after co-immunoprecipitation (Co-IP). We discovered that lncRNA AF127577.4 indeed encoded an endogenous micropeptide, named AF127577.4-ORF. AF127577.4-ORF was associated with GBM clinical grade. In vitro, AF127577.4-ORF could suppress GBM cell proliferation. Moreover, AF127577.4-ORF reduced m6A methylation level of GBM cells. Mechanistically, AF127577.4-ORF diminished ERK2 interaction with m6A reader methyltransferase like 3 (METTL3) and downregulated phosphorylated ERK (p-ERK) level. The ERK inhibitor reduced p-ERK level and downregulated METTL3 protein expression. AF127577.4-ORF weakened the stability of METTL3 protein by ERK. Also, AF127577.4-ORF suppressed GBM cell proliferation via METTL3. Our study identifies a novel micropeptide AF127577.4-ORF hidden in a lncRNA, with a potent anti-proliferating function in GBM by diminishing METTL3 protein stability by reducing the ERK2/METTL3 interaction. This micropeptide may be beneficial for development of therapeutic strategies against GBM.

Abstract Anchor and fissure grouting are used to repair earthen sites. However, the common method to obtain the compressive strength of grouting slurry would cause material, labor, and time losses. In addition the material properties, environmental and economic benefits have gained increasing attention. This study proposes a design framework for multi-objective proportioning optimization based on machine learning and metaheuristics. The results indicated that the eXtreme Gradient Boosting (XGBoost) model, whose hyper-parameters were optimized by a genetic algorithm, can accurately predicted the compressive strength of the slurries. The impact of the variables on development of compressive strength can explain the internal reaction mechanisms. The analytical framework based on meta-heuristic and technique for order of preference by similarity to an ideal solution (TOPSIS) provided Pareto-optimal solutions in design scenario of each sub-dataset. The framework proposed in this study can efficiently achieve mechanical, environmental, and economic design objectives of anchor grouting and fissure grouting slurries for repairing earthen sites.

Abstract Background To explore a method for screening and diagnosing neonatal congenital heart disease (CHD) applicable to grassroots level, evaluate the prevalence of CHD, and establish a hierarchical management system for CHD screening and treatment at the grassroots level. Methods A total of 24,253 newborns born in Tang County between January 2016 and December 2020 were consecutively enrolled and screened by trained primary physicians via the “twelve-section ultrasonic screening and diagnosis method” (referred to as the “twelve-section method”). Specialized staff from the CHD Screening and Diagnosis Center of Hebei Children’s Hospital regularly visited the local area for definite diagnosis of CHD in newborns who screened positive. Newborns with CHD were managed according to the hierarchical management system. Results The centre confirmed that, except for 2 newborns with patent ductus arteriosus missed in the diagnosis of ventricular septal defect combined with severe pulmonary hypertension, newborns with other isolated or concomitant simple CHDs were identified at the grassroots level. The sensitivity, specificity and diagnostic coincidence rate of the twelve-section method for screening complex CHD were 92%, 99.6% and 84%, respectively. A total of 301 children with CHD were identified. The overall CHD prevalence was 12.4‰. According to the hierarchical management system, 113 patients with simple CHD recovered spontaneously during local follow-up, 48 patients continued local follow-up, 106 patients were referred to the centre for surgery (including 17 patients with severe CHD and 89 patients with progressive CHD), 1 patient died without surgery, and 8 patients were lost to follow-up. Eighteen patients with complex CHD were directly referred to the centre for surgery, 3 patients died without surgery, and 4 patients were lost to follow-up. Most patients who received early intervention achieved satisfactory results. The mortality rate of CHD was approximately 28.86 per 100,000 children. Conclusions The “twelve-section method” is suitable for screening neonatal CHD at the grassroots level. The establishment of a hierarchical management system for CHD screening and treatment is conducive to the scientific management of CHD, which has important clinical and social significance for early detection, early intervention, reduction in mortality and improvement of the prognosis of complex and severe CHDs.

IntroductionNon-small cell lung cancer (NSCLC) is a prominent lung cancer disease worldwide. Currently, commonly used methods, such as surgery and radiotherapy, have significant side effects. Traditional Chinese medicine (TCM) has become a research hotspot because of its safe and effective characteristics. The branches and leaves of Taxus media are abundant in antitumor active compounds, and there has been no research conducted as yet regarding its anti–lung cancer molecular mechanism.ObjectiveThe aim of this study is to investigate the antitumor activity of two samples before and after fermentation of T. media, and to research the molecular mechanism of its inhibitory effect on NSCLC.MethodsThe chemical composition of pre-fermentation T. media (TM) and post-fermentation T. media qu (TMQ) were investigated using UHPLC-Q-Qrbitrap HRMS and high-performance liquid chromatography (HPLC). The anti-lung cancer activities of TM and TMQ were compared using an A549-induced tumor mouse model. An enzyme-linked immunosorbent assay (ELISA), hematoxylin and eosin (H&E) staining, immunohistochemistry, and immunofluorescence were used to determine the of TMQ mechanism of action.ResultsThe results indicated that TM and TMQ contained 83 compounds, consisting primarily of flavonoids, organic acids, and taxanes. Both taxanes and flavonoids in TMQ were higher than that in TM. Both TM and TMQ effectively inhibited the tumor growth in non-small cell lung cancer (NSCLC), and the inhibition rate was greater in TMQ (57.24%) than in TM (49.62%). TMQ administration downregulated the tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and the glutathione (GSH) level and upregulated interferon-γ (IFN-γ), reactive oxygen species (ROS), and malondialdehyde (MDA) levels in the serum of tumor mice. TMQ treatment also increased the protein expression of Bax, Caspase-3, and Beclin-1 in tumor tissues. In contrast, the bcl-2, PI3K, Ki67, ULK1, and mTOR protein levels were suppressed by TMQ. Protein assay analyses reemphasized the superior antitumor effect of TMQ over TM. These cumulative findings demonstrated that the mechanism of action of TMQ was closely related to the activation of transcriptional misregulation in the cancer pathway that inhibited the cholinergic synaptic, AMPK, and PI3K/Akt/mTOR signaling pathways.ConclusionThis study demonstrated that fermentation increased the active ingredient contents and antitumor effects of T. media. In addition, post-fermentation TMQ was superior to TM as a herbal medicine for NSCLC treatment.

Abstract To enhance the anesthetic efficacy and reduce toxic side effects, a strategy is proposed involving the utilization of general anesthetics of Propofol (Pro) and Etomidate (Eto) to synergistic inhibition GABA receptors simultaneously. Four‐in‐one molecular aggregates were prepared to implement this strategy, which comprised of Pro and Eto with the bridging molecule monoglyceride monooleate (GMO) and surfactant F127 through intermolecular forces. The blood‐brain barrier (BBB) targeted lactoferrin (LF) is affixed to their surface, obtaining the final molecular aggregates. By employing lactoferrin enrich aggregates to the BBB, followed by ultrasound combine microbubbles to open the BBB, a remarkable 4.5‐fold enhancement in brain drug delivery was achieved. The molecular aggregates group maintained stable parameters of heart rate, diastolic blood pressure, and systolic blood pressure. A notable increase of more than twice therapeutic index (TI) value was observed, implying their higher anesthesia efficiency and reduced toxicity. Electroencephalogram (EEG) experiments demonstrate a significant elevation in the proportion of δ waves from 28% to 80% for aggregates, accompanied by a nearly fivefold reduction in the proportion of θ waves, meaning a significant improvement in synergistic anesthesia effectiveness (interaction index 0.289) with lower drug dosage. Furthermore, mouse immunofluorescence brain slice experiments suggest Pro and Eto enter the GABA receptor simultaneously, resulting in synergistic inhibition of GABA receptors.

Abstract Proteins play a vital role in different biological processes by forming complexes through precise folding with exclusive inter‐ and intra‐molecular interactions. Understanding the structural and regulatory mechanisms underlying protein complex formation provides insights into biophysical processes. Furthermore, the principle of protein assembly gives guidelines for new biomimetic materials with potential applications in medicine, energy, and nanotechnology. Atomic force microscopy (AFM) is a powerful tool for investigating protein assembly and interactions across spatial scales (single molecules to cells) and temporal scales (milliseconds to days). It has significantly contributed to understanding nanoscale architectures, inter‐ and intra‐molecular interactions, and regulatory elements that determine protein structures, assemblies, and functions. This review describes recent advancements in elucidating protein assemblies with in situ AFM. We discuss the structures, diffusions, interactions, and assembly dynamics of proteins captured by conventional and high‐speed AFM in near‐native environments and recent AFM developments in the multimodal high‐resolution imaging, bimodal imaging, live cell imaging, and machine‐learning‐enhanced data analysis. These approaches show the significance of broadening the horizons of AFM and enable unprecedented explorations of protein assembly for biomaterial design and biomedical research.

IntroductionRadioresistance remains a significant challenge in the treatment of glioblastoma multiforme (GBM), the most prevalent and lethal brain cancer in adults. Metabolic alterations are known to contribute to radioresistance by activating antioxidant responses and promoting DNA repair. However, the role of circular RNAs in this process, particularly circFOXO3, is not well understood.MethodsIn this study, we investigated the expression of circFOXO3 in glioma cells exposed to radiation and in recurrent GBM tissues. We performed knockdown and overexpression experiments in vitro and in vivo to assess the effects of circFOXO3 on radiosensitivity. Metabolomic profiling was conducted to explore the metabolic changes associated with circFOXO3 overexpression following irradiation.ResultsOur results showed significant upregulation of circFOXO3 in glioma cells upon radiation exposure and in recurrent GBM tissues. Knockdown of circFOXO3 increased radiosensitivity both in vitro and in vivo, whereas overexpression of circFOXO3 attenuated radiosensitivity. Metabolomic analysis revealed substantial alterations in lipid and organic compound profiles between circFOXO3-overexpressing and control groups. Additionally, circFOXO3 suppression increased proapoptotic protein levels (Caspase 7 and Bax) and decreased anti-apoptotic protein Bcl-2 levels following radiotherapy.DiscussionThese findings demonstrate the pivotal role of circFOXO3 in promoting tumor radioresistance through metabolic modulation, suggesting that circFOXO3 could serve as a potential diagnostic and therapeutic target for GBM.

Carotid cavernous fistula is a rare but clinically important vascular abnormality that is challenging to diagnose and treat. The clinical data of a patient with bilateral carotid cavernous fistula diagnosed by CT images were retrospectively analyzed. Through the analysis of CT images, the patient was accurately located and the diagnosis was confirmed. CT images can provide detailed anatomical information and accurately show the location, morphology and hemodynamic characteristics of carotid cavernous fistula. Through CT image examination, we successfully diagnosed bilateral carotid cavernous fistula patients, and can provide an important reference for surgical treatment. Therefore, CT image examination can provide accurate diagnosis and surgical planning information, and provide support for the formulation of individual treatment plans for patients. The application of this method is helpful to improve the early diagnosis rate and treatment effect of carotid cavernous fistula.

BackgroundA substantial amount of research has explored the intricate relationship between exercise and sleep quality, consistently confirming that exercise can effectively enhance sleep quality. Nevertheless, previous studies have yet to conclusively determine which specific exercise program is most efficacious in improving sleep quality. To address this gap, the present study systematically evaluated the differential effects of various types of exercise, as well as exercise dosages (including duration, intervention period, frequency, and intensity), on sleep outcomes using a network meta-analysis approach. This endeavor aims to provide evidence-based support for the development of scientifically effective exercise programs tailored to improve sleep quality.MethodsThrough the Web of Science, PubMed, Cochrane Library, Embase, and Scopus databases, we conducted a search for randomized controlled trials investigating the effects of exercise interventions on sleep, with a search cutoff date of April 30, 2024. We rigorously selected the literature according to the PICOS principle, and two independent researchers extracted the data. We would like to change this passage to: Bias risk assessment was conducted using the RevMan 5.4 software, and traditional meta-analysis and network meta-analysis were performed using Stata 17.0 software to generate forest plots, network evidence plots, and funnel plots. Furthermore, we adopted the surface under the cumulative ranking curve (SUCRA) to evaluate and rank the intervention effects of different exercise types and dosages on sleep quality. To verify the robustness of our study results, we performed a sensitivity analysis using the leave-one-out method.ResultsThe study strictly adhered to the PRISMA guidelines and included 58 RCT papers with a total of 5,008 participants. The network meta-analysis revealed significant variations in the impact of exercise frequency on sleep outcomes when compared to the control group. Interventions of 1–2 times per week [SMD = −0.85, 95% CI (−1.43, −0.26)], 3 times per week [SMD = −0.45, 95% CI (−0.80, −0.11)], and 4 times per week [SMD = −1.09, 95% CI (−1.92, −0.26)] demonstrated the most notable effects. Interventions lasting ≤30 min and 60–65 min were significantly more effective than the control group, with ≤30 min proving significantly more effective than 40–55 min [SMD = 0.75, 95% CI (0.01, 1.49)]. Interventions lasting 9–10 weeks [SMD = −1.40, 95% CI (−2.37, −0.44)], 12–16 weeks [SMD = −0.55, 95% CI (−0.90, −0.20)], and ≥ 24 weeks [SMD = −0.71, 95% CI (−1.31, −0.10)] were all significantly more effective than the control group. Additionally, the 9–10 weeks intervention period was found to be significantly more effective than the 6–8 weeks period [SMD = −1.21, 95% CI (−2.37, −0.04)]. Furthermore, interventions of moderate intensity [SMD = −1.06, 95% CI (−1.52, −0.61)] and high intensity [SMD = −1.48, 95% CI (−2.55, −0.40)] exercise interventions yielded significantly greater benefits compared to the control group. Specifically, high intensity interventions [SMD = −1.97, 95% CI (−3.37, −0.56)] and moderate intensity [SMD = −1.55, 95% CI (−2.57, −0.54)] exercise interventions were found to be significantly more effective than moderate-high intensity interventions. In terms of exercise types, aerobic exercise [SMD = −0.56, 95% CI (−0.86, −0.27)], traditional Chinese exercises [SMD = −0.57, 95% CI (−0.97, −0.18)], and combined exercise [SMD = −0.99, 95% CI (−1.66, −0.32)] interventions all produced significant improvements compared to the control group. The study determined that the most effective combination of exercise prescription elements for enhancing sleep quality includes a frequency of 4 times per week (SUCRA = 84.7), a duration of ≤30 min (SUCRA = 92.2), a period of 9–10 weeks (SUCRA = 89.9), and high-intensity (SUCRA = 92.9) combined exercise (SUCRA = 82.7).ConclusionThe current evidence indicates that combined exercise with a frequency of 4 times per week, a duration of ≤30 min, a period of 9–10 weeks, and high intensity is most effective for improving sleep quality. Induction cladding is a promising surface technology that combines the advantages of surface coatings and induction heating. It is an energy-efficient, environment-friendly, and cost-effective method that facilitates the fabrication of coatings with controllable thicknesses and ensures metallurgical bonding between the coating and the substrate. Owing to the high power-conversion efficiency of helical coil, induction cladding is particularly adaptable for the application of coatings on long shafts and rod parts, which find widespread use in mining and energy machinery. This paper provides a comprehensive overview of the state-of-the-art methods in induction cladding. Herein we focus on its mechanisms, cladding process and parameters, commonly used materials, simulations, innovative induction cladding technologies, industrial applications, problems, and future developments in this field.

Corn gluten meal-corn husk mixes (CCM) are an inexpensive and readily available agricultural by-product. This study explores a novel technique by converting CCM into high-value livestock feed protein sources through fermentation with Aspergillus niger AAX and Lactobacillus fermentum LLS, aiming to sustainably meet future global protein needs. The process of fermentation significantly altered the structural composition of high molecular weight proteins, zein, and dietary fibers. This transformation resulted in a marked elevation in the concentrations of peptides, free amino acids, and polyphenols. The acidic environment produced during fermentation prevented lipid oxidation in CCM, thereby extending its storability. After fermentation, the content of anti-nutritional factors decreased, while its antioxidant capacity increased. In vitro simulated digestion suggested that fermentation improved the digestibility of CCM protein. In vivo animal experiments showed that fermented CCM (FCCM) promoted growth and gut health in chicks. This study provides new insights into the utilization of CCM.

ABSTRACT: This investigation sought to reveal the effects of heat stress on the meat quality of geese. Wuzong geese were subjected to heat stress at 35°C for 25 d or 4 h to examine different heat stress time on meat quality. Short-time heat stress reduced muscle drip loss and meat color L* value while increasing pH value and meat color a* and b* values. Long-time heat stress decreased body weight and increased leg muscle pH value and meat color b* value. Amino acid profile of geese breast muscle revealed that both LHS and SHS can induce L-Cystine but reduced L-Cystathionine, which were positive correlated with cooking loss and meat color lightness, respectively. Lipidome analysis indicated that heat stress would alter the synthesis of unsaturated fatty acids, and the difference between LHS and SHS on lipids mainly focused on Hex1Cer and TG. Non-target metabolome analysis indicated effects of heat stress on Glycerolipid metabolism, Arachidonic acid metabolism, and Pyrimidine metabolism. Proteome analysis showed that heat stress mainly affects cellular respiration metabolism and immune response. These findings highlight the diverse effects of heat stress on meat quality, amino acid composition, lipidome, metabolome, and proteome in geese.

IntroductionImmune checkpoint inhibitors (ICIs) have revolutionized the treatment of a number of patients with advanced cancer, and while this has resulted in increased survival times, it has also led to the emergence of novel immune-related adverse events (irAEs). In individuals with advanced cancer, sarcopenia is a significant symptom of cachexia and is linked to poor nutritional status and increased mortality. The present study aimed to evaluate sarcopenia and other risk variables that can affect the emergence of irAEs in patients with lung cancer.MethodsA single-center retrospective analysis of 129 patients with advanced lung cancer treated with programmed cell death protein-1 (PD-1)/programmed cell death ligand-1 (PD-L1) checkpoint inhibitors was conducted from August 2020 to August 2022. Data on baseline characteristics and adverse events of participants were collected. Computed tomography was used to determine the skeletal muscle index at the third lumbar vertebra (L3-SMI) and whether sarcopenia is present.ResultsThe median age of all participants was 60 years old (range, 52-66 years), with men accounting for 68.9% of the total patient cohort. The present study showed that 44 (34%) participants presented with any degree of irAEs, and 79 (61.2%) patients presented with sarcopenia. There were no statistically significant differences in baseline characteristics, such as age and sex, between patients who presented with irAEs and those without irAEs. Using logistic regression analysis, individuals with sarcopenia were 2.635-times more likely to experience any grade of irAEs than those without sarcopenia.DiscussionirAEs are prevalent side effects of PD-1/PD-L1 inhibitor therapy for patients with cancer. By diagnosing and treating sarcopenia early, it is possible to lower the potential risk of irAEs in patients with advanced cancer. Furthermore, sarcopenia can be utilized as a predictor of irAEs.

Infection remains the leading cause of mortality in severe aplastic anemia (SAA) patients, with invasive fungal infections being the great threat. Aspergillus fumigatus accounts for most of the reported fungal infection cases. Here, we present a case of A. flavus infection in a patient with acute very severe aplastic anemia (VSAA) despite persistently negative clinical fungal tests. The patient was admitted to the hospital due to pancytopenia presisting for over a month and intermittent fever for 10 days. Elevated inflammatory indicators and abnormal lung imaging suggested infection, prompting consideration of fungal involvement. Despite negative results from multiple blood, sputum fungal cultures and the serum (1,3)-β-D-glucan/galactomannan tests. Metagenomic next-generation sequencing (mNGS) on multiple blood samples, alongside clinical symptoms, confirmed A. flavus infection. Targeted antifungal treatment with liposomal amphotericin B and voriconazole significantly ameliorated pulmonary symptoms. Additionally, this study reviewed and compared the symptoms, diagnostic approaches, and treatments from prior Aspergillus infections in AA patients. It emphasizes critical role of early mNGS utilization in diagnosing and managing infectious diseases, offering insights for diagnosing and treating fungal infections in VSAA.

Background Coronavirus disease 2019 (COVID-19) has raised concerns about its potential effects on human fertility, particularly among individuals undergoing assisted reproductive therapy (ART). However, the impact of COVID-19 on female reproductive and assisted reproductive outcomes is unclear. In this study, we aimed to evaluate the effects of COVID-19 on pregnancy outcomes during frozen-thawed embryo transfer (FET) cycles. Methods This retrospective cohort study included 327 enrolled patients who underwent FET cycles at a single reproductive centre. The study group consisted of patients treated between 1 January 2023 and 31 March 2023 who recently recovered from COVID-19. The embryos for transfer were generated prior to COVID-19 infection. The control group consisted of patients treated between 1 January 2021 and 31 March 2021 who were not infected and did not receive a severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccine. Demographic and cycle characteristics and outcomes were compared. Results A total of 160 recovered women and 167 controls were included. The primary outcome—the live birth rate—was similar between the two groups (43.8% vs. 43.1%, P > 0.05). IntroductionPorcine Parvovirus (PPV) is a significant pathogen in the pig industry, with eight genotypes, including PPV7, identified since its emergence in 2016. Co-infections with viruses such as Porcine Circovirus 2 (PCV2) and Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) pose serious risks to swine health. Thus, there is an urgent need for rapid, sensitive, and specific detection methods suitable for use in field settings or laboratories with limited resources.MethodsWe developed a CRISPR/Cas12a-based assay combined with recombinase polymerase amplification (RPA) for the rapid detection of PPV7. Specific RPA primers and five CRISPR RNAs (crRNAs) were designed to target a highly conserved region within the NS1 gene of PPV7. Optimization of crRNA and single-stranded DNA (ssDNA) concentrations was performed to enhance the assay’s performance.ResultsCrRNA optimization identified crRNA-05 as the optimal candidate for Cas12a-based detection of PPV7, as all synthesized crRNAs demonstrated similar performance. The optimal crRNA concentration was determined to be 200 nM, yielding consistent results across tested concentrations. For ssDNA optimization, the strongest fluorescence signal was achieved with 500 nM of the FAM-BHQ ssDNA receptor. The assay showed a minimal detection limit of 100copies/μl for PPV7, confirmed through fluorescence and lateral flow detection methods. Specificity testing indicated that only PPV7 DNA samples returned positive results, confirming the assay’s accuracy. In tests of 50 lung tissue samples from diseased pigs, the RPA-Cas12a assay identified 29 positive samples (58%), surpassing the 22 positive samples (44%) detected by conventional PCR. This highlights the RPA-Cas12a method’s enhanced detection capability and its potential utility in clinical surveillance and management of PPV7 in swine populations.DiscussionThe RPA-Cas12a assay effectively detects PPV7 in clinical samples, enhancing disease surveillance and control in pigs. Its adaptability to resource-limited settings significantly improves PPV7 management and prevention strategies, thereby supporting the overall health and development of the pig industry.
