Asian rice gall midge is one of the important pests of rice, which attacks the crop from nursery to the end of the tillering stage. Managing this pest through plant resistance is the most viable and economical. Two hundred‐two rice genotypes were phenotyped against Asian rice gall midge, Orseolia oryzae (Wood‐Mason) and the mechanism of resistance in terms of antixenosis and antibiosis was studied. Antixenosis mechanism for adult settlement and egg laying indicated that the susceptible genotypes were preferred more than the resistant genotypes in a free choice test. First instar maggots were found and did not continue their growth on resistant genotypes further. However, in susceptible genotypes, they molted successfully and emerged as adults. No choice tests revealed that the emergence of adults in susceptible genotypes took less time than in resistant genotypes. Higher adult sex ratio was found in susceptible genotypes. Estimation of biochemical components in rice shoot apices of selected genotypes revealed that higher levels of total phenols, wax content, total flavonoids and total free amino acids were present in the resistant genotypes. Still, the number of total sugars, reducing sugars and total protein contents were significantly higher in the susceptible genotypes. Hence, the resistance mechanism in rice gall midge was displayed as a combination of antixenosis and antibiosis mechanism. Phosphorus nutrition has been known for a long time to influence floral transition in plants, but the underlying mechanism is unclear. Arabidopsis phosphate transporter PHOSPHATE1 (PHO1) plays a critical role in phosphate translocation from roots to shoots, but whether and how it regulates floral transition is unknown. Here, we show that knockout mutation of PHO1 delays flowering under both long- and short-day conditions. The late flowering of pho1 mutants can be partially rescued by Pi supplementation in rosettes or shoot apices. Grafting assay indicates that the late flowering of pho1 mutants is a result of impaired phosphate translocation from roots to shoots. Knockout mutation of SPX1 and SPX2 , two negative regulators of the phosphate starvation response, partially rescues the late flowering of pho1 mutants. PHO1 is epistatic to PHO2, a negative regulator of PHO1, in flowering time regulation. Loss of PHO1 represses the expression of some floral activators, including FT encoding florigen, and induces the expression of some floral repressors in shoots. Genetic analyses indicate that at least jasmonic acid signaling is partially responsible for the late flowering of pho1 mutants. In addition, we find that rice PHO1;2 , the homolog of PHO1 , plays a similar role in floral transition. These results suggest that PHO1 integrates phosphorus nutrition and flowering time, and could be used as a potential target in modulating phosphorus nutrition-mediated flowering time in plants. Formin proteins, characterized by the FH2 domain, are critical in regulating actin-driven cellular processes and cytoskeletal dynamics during abiotic stress. However, no genome-wide analysis of the formin gene family has yet to be conducted in the economically significant plant potato (Solanum tuberosum L.). In this study, 26 formin genes were identified and characterized in the potato genome (named as StFH), each containing the typical FH2 domain and distributed across the ten chromosomes. The StFH was categorized into seven subgroups (A-G) and the gene structure and motif analysis demonstrated higher structural similarities within the subgroups. Besides, the StFH exhibited ancestry and functional similarities with Arabidopsis. The Ka/Ks ratio indicated that StFH gene pairs were evolving through purifying selection, with five gene pairs exhibiting segmental duplications and two pairs exhibiting tandem duplications. Subcellular localization analysis suggested that most of the StFH genes were located in the chloroplast and plasma membrane. Moreover, 54 cis-acting regulatory elements (CAREs) were identified in the promoter regions, some of which were associated with stress responses. According to gene ontology analysis, the majority of the StFH genes were involved in biological processes, with 63 out of 74 GO terms affecting actin polymerization. Six major transcription factor families, including bZIP, C2H2, ERF, GATA, LBD, NAC, and HSF, were identified that were involved in the regulation of StFH genes in various abiotic stresses, including drought. Further, the 60 unique microRNAs targeted 24 StFH by regulating gene expression in response to drought stress were identified. The expression of StFH genes in 14 different tissues, particularly in drought-responsive tissues such as root, stem, shoot apex, and leaf, underscores their significance in managing drought stress. RNA-seq analysis of the drought-resistant Qingshu No. 9 variety revealed the potential role of up-regulated genes, including StFH2, StFH10, StFH19, and StFH25, in alleviating drought stress. Overall, these findings provide crucial insights into the response to drought stress in potatoes and can be utilized in breeding programs to develop potato cultivars with enhanced drought-tolerant traits. SUMMARY The intracellular localization of the florigen FLOWERING LOCUS T (FT) is important for its long‐distance transport toward the shoot apical meristem. However, the mechanisms regulating the FT localization remain poorly understood. Here, we discovered that in Arabidopsis thaliana, the chloroplast‐localized protein THYLAKOID FORMATION 1 (THF1) physically interacts with FT, sequestering FT in the outer chloroplast envelope. Loss of THF1 function led to temperature‐insensitive flowering, resulting in early flowering, especially under low ambient temperatures. THF1 mainly acts in the leaf vasculature and shoot apex to prevent flowering. Mutation of CONSTANS or FT completely suppressed the early flowering of thf1‐1 mutants. FT and THF1 interact via their anion binding pocket and coiled‐coil domain (CCD), respectively. Deletion of the CCD in THF1 by gene editing caused temperature‐insensitive early flowering similar to that observed in the thf1‐1 mutant. FT levels in the outer chloroplast envelope decreased in the thf1‐1 mutant, suggesting that THF1 is important for sequestering FT. Furthermore, THF1 protein levels decreased in seedlings grown at high ambient temperature, suggesting an explanation for its role in plant responses to ambient temperature. A thf1‐1 phosphatidylglycerolphosphate synthase 1 (pgp1) double mutant exhibited additive acceleration of flowering at 23 and 16°C, compared to the single mutants, indicating that THF1 and phosphatidylglycerol (PG) act as independent but synergistic regulators of temperature‐responsive flowering. Collectively, our results provide an understanding of the genetic pathway involving THF1 and its role in temperature‐responsive flowering and reveal a previously unappreciated additive interplay between THF1 and PG in temperature‐responsive flowering. Plants flower in response to environmental signals. These signals change the shape and developmental identity of the shoot apical meristem (SAM), causing it to form flowers and inflorescences. We show that the increases in SAM width and height during floral transition correlate with changes in size of the central zone (CZ), defined by CLAVATA3 expression, and involve a transient increase in the height of the organizing center (OC), defined by WUSCHEL expression. The APETALA2 (AP2) transcription factor is required for the rapid increases in SAM height and width, by maintaining the width of the OC and increasing the height and width of the CZ. AP2 expression is repressed in the SAM at the end of floral transition, and extending the duration of its expression increases SAM width. Transcriptional repression by SUPPRESSOR OF OVEREXPRESSION OF CONSTANS1 (SOC1) represents one of the mechanisms reducing AP2 expression during floral transition. Moreover, AP2 represses SOC1 transcription, and we find that reciprocal repression of SOC1 and AP2 contributes to synchronizing precise changes in meristem shape with floral transition. In multicellular organisms, tissue outgrowth creates a new water sink, modifying local hydraulic patterns. Although water fluxes are often considered passive by-products of development, their contribution to morphogenesis remains largely unexplored. Here, we mapped cell volumetric growth across the shoot apex in Arabidopsis thaliana. We found that, as organs grow, a subpopulation of cells at the organ-meristem boundary shrinks. Growth simulations using a model that integrates hydraulics and mechanics revealed water fluxes and predicted a water deficit for boundary cells. In planta, a water-soluble dye preferentially allocated to fast-growing tissues and failed to enter the boundary domain. Cell shrinkage next to fast-growing domains was also robust to different growth conditions and different topographies. Finally, a molecular signature of water deficit at the boundary confirmed our conclusion. Taken together, we propose that the differential sink strength of emerging organs prescribes the hydraulic patterns that define boundary domains at the shoot apex. Combining cell volumetric analysis, growth simulation, and in planta water flow tracing, the authors reveal the correlation between water flux distribution and cell growth rate in Arabidopsis shoot apex, suggesting the potential contribution of hydraulic patterns to morphogenesis. Chinese artichoke (Stachys sieboldii [Miq.]) is a popular healthcare food owing to its high contents of stachyose. Viral infections have caused the severe degeneration of germplasm in this plant species. An efficient virus elimination and micropropagation system is required to produce Chinese artichoke plants that are free of viruses. A protocol for virus elimination in Chinese artichoke was established by comparing the potential for growth of the terminal buds using shoot apices as starting materials, in media with or without plant growth regulators (PGRs) and various concentrations of sucrose. Murashige and Skoog (MS) medium without any PGR performed better than that supplemented with PGRs as indicated by a higher rate of survival of shoot apices, higher plant height, more root numbers and adventitious buds, and increased biomass (P < 0.05). An experiment to further optimize the sucrose concentration demonstrated that MS medium with 5.0% sucrose (w/v) efficiently induced the growth of both roots and shoots, thus, achieving the efficient micropropagation of Chinese artichoke within 4 wk. Compared with the mother plants in which viruses had not been eliminated, the virus-free plants had significantly higher numbers of tubers, increased yield, and a higher content of stachyose (P < 0.05). Higher contents of endogenous hormones were detected in Chinese artichoke, which may explain the efficient micropropagation without the use of exogenous PGRs. Flowering, the transition from the vegetative to the reproductive stage, is vital for reproductive success, affecting forage quality, the yield of aboveground biomass, and seed production in alfalfa. To explore the transcriptomic profile of alfalfa flowering transition, we compared gene expression between shoot apices (SAs) at the vegetative stage and flower buds (FBs) at the reproductive stage by mRNA sequencing. A total of 3,409 DEGs were identified, and based on gene ontology (GO), 42.53% of the most enriched 15 processes were associated with plant reproduction, including growth phase transition and floral organ development. For the former category, 79.1% of DEGs showed higher expression levels in SA than FB, suggesting they were sequentially turned on and off at the two test stages. For the DEGs encoding the components of circadian rhythm, sugar metabolism, phytohormone signaling, and floral organ identity genes, 60.71% showed higher abundance in FB than SA. Among them, MsAP1, an APETALA1 (AP1) homolog of Arabidopsis thaliana, showed high expression in flower buds and co-expressed with genes related to flower organ development. Moreover, ectopic expression of MsAP1 in Arabidopsis resulted in dwarfism and early flowering under long-day conditions. The MsAP1-overexpression plant displayed morphological abnormalities including fused whorls, enlarged pistils, determinate inflorescence, and small pods. In addition, MsAP1 is localized in the nucleus and exhibits significant transcriptional activity. These findings revealed a transcriptional regulation network of alfalfa transition from juvenile phase to flowering and provided genetic evidence of the dual role of MsAP1 in flowering and floral organ development. Sucrose has a significant role in promoting microtuberization in potato. It acts as a crucial modifier, influencing the growth and development of microtuber in potato. Understanding the role of sucrose in microtuberization is vital for improving potato cultivation practices and enhancing crop productivity. In this experiment, different concentrations of sucrose (3, 6, 9, 12, and 15%) were investigated to determine their effects on the microtuberization of a potato cultivar 'Sante'. The findings revealed that high concentrations of sucrose (12 and 15%) inhibited microtuberization percentage, microtuber count, and shoot growth. Tuberization percentage reached its maximum value (100%) when exposed to a sucrose concentration of 9%. The microtuber count was 2 per explant and 4.6 per vessel. However, using a 12% sucrose concentration resulted in the highest microtuber diameter (5 mm), microtuber fresh weight (120 mg) and dry weight (26 mg), and microtuber yield (FW: 752 mg; DW: 170 mg). The highest explant fresh weight (44 mg) and dry weight (9.5 mg) occurred in response to 15% sucrose concentration. Biomass allocation was influenced by sucrose concentration, with higher concentrations leading to a greater biomass allocation to the microtuber and explant, rather than the shoot. Accordingly, it can be concluded that a sucrose concentration of 12% was an optimal treatment for 'Sante' potato microtuber production. [ABSTRACT FROM AUTHOR]

Plants are dependent on divisions of stem cells to establish cell lineages required for growth. During embryogenesis, early division products are considered to be stem cells, whereas during post-embryonic development, stem cells are present in meristems at the root and shoot apex. PLETHORA/AINTEGUMENTA-LIKE (PLT/AIL) transcription factors are regulators of post-embryonic meristem function and are required to maintain stem cell pools. Despite the parallels between embryonic and post-embryonic stem cells, the role of PLTs during early embryogenesis has not been thoroughly investigated. Here, we demonstrate that the PLT regulome in the zygote, and apical and basal cells is in strong congruence with that of post-embryonic meristematic cells. We reveal that out of all six PLTs, only PLT2 and PLT4/BABY BOOM(BBM) are expressed in the zygote, and that these two factors are essential for progression of embryogenesis beyond the zygote stage and first divisions. Finally, we show that other PLTs can rescue plt2 bbm defectswhen expressed fromthe PLT2 andBBMpromoters, establishing upstream regulation as a key factor in early embryogenesis. Stem cell homeostasis in the shoot apical meristem involves a core regulatory feedback loop between the signalling peptide CLAVATA3 (CLV3), produced in stem cells, and the transcription factor WUSCHEL, expressed in the underlying organising centre. clv3 mutant meristems display massive overgrowth, which is thought to be caused by stem cell over proliferation, although it is unknown how uncontrolled stem cell divisions lead to this altered morphology. Here, we reveal local buckling defects in mutant meristems, and use analytical models to show how mechanical properties and growth rates may contribute to the phenotype. Indeed, clv3 mutant meristems are mechanically more heterogeneous than the wild type, and also display regional growth heterogeneities. Furthermore, stereotypical wild-type meristem organisation, in which cells simultaneously express distinct fate markers, is lost in mutants. Finally, cells in mutant meristems are auxin responsive, suggesting that they are functionally distinguishable from wild-type stem cells. Thus, all benchmarks show that clv3 mutant meristem cells are different from wild-type stem cells, suggesting that overgrowth is caused by the disruption of a more complex regulatory framework that maintains distinct genetic and functional domains in the meristem. Herein, an in vitro multiplication protocol for Prunus campanulata was evaluated. The effects of 15.0, 30.0, and 45.0 g L−1 sucrose and three flask sealing methods (PVC film, rigid polypropylene lid, and rigid lid with membrane) were analyzed at the multiplication, rooting, and acclimatization stages. For in vitro multiplication, shoot tips of approximately 1 cm in length containing two pairs of leaves from seedlings germinated in vitro in Woody Plant Medium (WPM) were used. Evaluations considered the multiplication of shoot apexes in three subcultures (at 40, 80, and 120 d) for survival, number of shoots, shoot length, number of leaves, and multiplication rate. In the rooting phase, the rooting percentage, number of roots, and root length were assessed. In the acclimatization phase under shaded and full sun conditions, seedling survival and biometric characteristics were evaluated, including stem diameter, height, and number of leaves. The addition of sucrose to the medium, at a concentration of 45.0 g L−1, associated with sealing using PVC plastic film, showed inadequate results for in vitro multiplication of P. campanulata. The addition of sucrose to the culture medium in concentrations ranging from 15.0 to 30.0 g L−1 favored in vitro rooting. The use of a permeable membrane for sealing enabled the growth of more vigorous shoots in terms of percentage of rooting and root length, and these seedlings stood out in the acclimatization phases with greater rates of survival and improved biometric characteristics. Gynostemma pentaphyllum (Thunb.) Makino is an important producer of dammarene-type triterpenoid saponins. These saponins (gypenosides) exhibit diverse pharmacological benefits such as anticancer, antidiabetic, and immunomodulatory effects, and have major potential in the pharmaceutical and health care industries. Here, we employed single-cell RNA sequencing (scRNA-seq) to profile the transcriptomes of more than 50,000 cells derived from G. pentaphyllum shoot apexes and leaves. Following cell clustering and annotation, we identified five major cell types in shoot apexes and four in leaves. Each cell type displayed substantial transcriptomic heterogeneity both within and between tissues. Examining gene expression patterns across various cell types revealed that gypenoside biosynthesis predominantly occurred in mesophyll cells, with heightened activity observed in shoot apexes compared to leaves. Furthermore, we explored the impact of transposable elements (TEs) on G. pentaphyllum transcriptomic landscapes. Our findings the highlighted the unbalanced expression of certain TE families across different cell types in shoot apexes and leaves, marking the first investigation of TE expression at the single-cell level in plants. Additionally, we observed dynamic expression of genes involved in gypenoside biosynthesis and specific TE families during epidermal and vascular cell development. The involvement of TE expression in regulating cell differentiation and gypenoside biosynthesis warrant further exploration. Overall, this study not only provides new insights into the spatiotemporal organization of gypenoside biosynthesis and TE activity in G. pentaphyllum shoot apexes and leaves but also offers valuable cellular and genetic resources for a deeper understanding of developmental and physiological processes at singlecell resolution in this species. Flowering is a major developmental transition in plants, but asynchronous flowering hinders the utilization of wild cotton relatives in breeding programs. We performed comparative transcriptomic profiling of early‐ and late‐flowering Gossypium hirsutum genotypes to elucidate genetic factors influencing reproductive timing. Shoot apices were sampled from the photoperiod‐sensitive landrace G. hirsutum purpurascens (GhP) and early‐maturing variety ZhongMianSuo (ZMS) at five time points following the emergence of sympodial nodes. RNA‐sequencing revealed extensive transcriptional differences during floral transition. Numerous flowering‐associated genes exhibited genotype‐specific expression, including FLOWERING LOCUS T (FT) homologs upregulated in ZMS. FT‐interacting factors like SOC1 and CO‐like also showed higher expression in ZMS, implicating florigen pathways in early flowering. Additionally, circadian clock and light signalling components were misregulated between varieties, suggesting altered photoperiod responses in GhP. Weighted co‐expression network analysis specifically linked a module enriched for circadian‐related genes to GhP's late flowering. Through an integrated transcriptome analysis, we defined a regulatory landscape of reproductive phase change in cotton. Differentially expressed genes related to photoperiod, circadian clock, and light signalling likely contribute to delayed flowering in wild cottons. Characterization of upstream flowering regulators will enable modifying photoperiod sensitivity and expand germplasm use for cotton improvement. Nepenthes are carnivorous plants that colonize habitats poor in soil nutrients. To survive, Nepenthes develop pitchers capable of capturing and digesting attracted prey. Prey‐derived nutrients are then absorbed to support plant growth and reproduction. So far, pitcher formation in Nepenthes is a poorly understood biological process. To shed light on the formation of Nepenthes pitchers, we grew dissected shoot apices of 3‐month‐old N. khasiana seedlings in Murashige and Skoog (MS) medium of varying strengths viz. full‐strength MS (1 MS), quarter‐strength MS (1/4 MS), and one‐eighth strength MS (1/8 MS), including those lacking nitrogen (N), phosphorus (P), and potassium (K) and in the presence of N‐1‐naphthylphthalamic acid (NPA). We sequenced the transcriptome and performed gas chromatography–mass spectrometry to determine changes in gene expression patterns and primary metabolite accumulations in response to the varying nutrient conditions. Shoots grown in 1 MS or NPA‐containing 1/4 MS and 1/8 MS failed to develop pitchers. Remarkably, pitcher formation is restored when N was removed from 1 MS. Transcriptomic response to nutrient‐sufficient and nutrient‐deficient conditions are associated with the enrichment of several defence‐related genes, including two JA‐mediated defence response genes, WRKY51 and WRKY11, respectively. Further, metabolomic response to the varying nutrient conditions identifies glutamic acid as a key metabolite, accumulating at lower and higher levels in shoots with and without pitchers, respectively. Together, our findings suggest that failure to form pitchers may be associated with the suppression of the JA‐signalling pathway, whereas the induction of the JA‐mediated defence response is linked to pitcher formation in N. khasiana. Source‐sink balance in plants determines carbon distribution, and altering it can impact carbon fixation, transport, and allocation. We aimed to investigate the effect of altered source‐sink ratios on carbon fixation, transport, and distribution in 'Valencia' sweet orange (Citrus x sinensis) by various defoliation treatments (0%, 33%, 66%, and 83% leaf removal). Gas exchange parameters were measured on 0 and 10 days after defoliation using A/Ci response curves, and leaf export was measured two days after defoliation using radioisotope tracer techniques. Greater defoliation increased the maximum rate of carboxylation (Vcmax), electron transport rate (J1200), and triose‐phosphate utilization rate (TPU). Leaf export was unaffected by defoliation but increased in leaves closer to the shoot apex. Basipetal translocation velocity in the trunk remained unaltered, indicating that more photosynthates remained in the shoot rather than being transported directly to the root sink. Defoliated plants initiated more new flush shoots but accumulated less shoot biomass per plant after 8 weeks. Carbon allocation to fine roots was smaller in defoliated plants, suggesting defoliation led to retention of carbohydrates in aboveground organs such as the trunk and other shoots from previous growing cycles. In conclusion, the low source‐sink ratio increased carbon fixation without impacting individual leaf export in citrus. The results suggest that intermediate sinks such as the aboveground perennial organs play a role in mediating the translocation velocity. In seed plants, growth of shoots and roots is indeterminate, while leaves are typically determinate organs that cease to grow after a certain developmental stage. This is due to the characteristics of the leaf meristem, where cell proliferation activity is retained only for a limited period. However, several plants exhibit indeterminacy in their leaves, exemplified by the pinnate compound leaves of Guarea and Chisocheton genera in the Meliaceae family. In these plants, the leaf meristem at the tip of the leaf retains meristematic activity and produces leaflets over years, resulting in a single leaf that resembles a twig. The molecular mechanism underlying the indeterminate leaf meristem of these plants has not been examined. In this research, we used Guarea glabra as a model to investigate the development of indeterminate pinnate leaves. Transcriptome analyses revealed that the gene expression profile in leaf apex tissue differed from that in the shoot apex. However, a class 1 KNOTTED-LIKE HOMEOBOX (KNOX1) gene which is lost in Brassicaceae was highly expressed in both tissues. We established an in situ hybridisation system for this species using Technovit 9100 to analyse the spatial expression patterns of genes. We revealed that the leaf meristematic region of G. glabra expresses KNOX1, LEAFY and ANGUSTIFORIA3 simultaneously, suggesting the involvement of these genes in the indeterminacy of the leaf meristem. The breeding of low phytic acid (LPA) crops is widely considered an effective strategy to improve crop nutrition, but the LPA crops usually have inferior seed germination performance. To clarify the reason for the suboptimal seed performance of LPA rice, this study investigated the impact of reduced seed phytic acid (InsP6) content in rice ins(3)P synthase1 (EC 5.5.1.4, RINO1), one of the key targets for engineering LPA rice, knockouton cellular differentiation in seed embryos and its relation to myoinositol metabolism and auxin signalling during embryogenesis. The results indicated that the homozygotes of RINO1 knockout could initiate differentiation at the early stage of embryogenesis but failed to form normal differentiation of plumule and radicle primordia. The loss of RINO1 function disrupted vesicle trafficking and auxin signalling due to the significantly lowered phosphatidylinositides (PIs) concentration in seed embryos, thereby leading to the defects of seed embryos without the recognizable differentiation of shoot apex meristem (SAM) and radicle apex meristem (RAM) for the homozygotes of RINO1 knockout. The abnormal embryo phenotype of RINO1 homozygotes was partially rescued by exogenous spraying of inositol and indole-3-acetic acid (IAA) in rice panicle. Thus, RINO1 is crucial for both seed InsP6 biosynthesis and embryonic development. The complex structures of multicellular organisms originate from a unicellular zygote. In most angiosperms, including Arabidopsis thaliana, the zygote is distinctly polar and divides asymmetrically to produce an apical cell, which generates the aboveground part of the plant body, and a basal cell, which generates the root tip and extraembryonic suspensor. Thus, zygote polarity is pivotal for establishing the apical-basal axis running from the shoot apex to the root tip of the plant body. The molecular mechanisms and spatiotemporal dynamics behind zygote polarization remain elusive. Poplar trees use photoperiod as a precise seasonal indicator, synchronizing plant phenology with the environment. Daylength cue determines FLOWERING LOCUS T 2 (FT2) daily expression, crucial for shoot apex development and establishment of the annual growing period. However, limited evidence exists for the molecular factors controlling FT2 transcription and the conservation with the photoperiodic control of Arabidopsis flowering. We demonstrate that FT2 expression mediates growth cessation response quantitatively, and we provide a minimal data-driven model linking core clock genes to FT2 daily levels. GIGANTEA (GI) emerges as a critical inducer of the FT2 activation window, time-bound by TIMING OF CAB EXPRESSION (TOC1) and LATE ELONGATED HYPOCOTYL (LHY2) repressions. CRISPR/Cas9 loss-of-function lines validate these roles, identifying TOC1 as a long-sought FT2 repressor. Additionally, model simulations predict that FT2 downregulation upon daylength shortening results from a progressive narrowing of this activation window, driven by the phase shift observed in the preceding clock genes. This circadian-mediated mechanism enables poplar to exploit FT2 levels as an accurate daylength-meter. Summary: Perennial trees in boreal and temperate regions undergo growth cessation and bud set under short photoperiods, which are regulated by phytochrome B (phyB) photoreceptors and PHYTOCHROME INTERACTING FACTOR 8 (PIF8) proteins. However, the direct signaling components downstream of the phyB‐PIF8 module remain unclear.We found that short photoperiods suppressed the expression of miR156, while upregulated the expression of miR156‐targeted SQUAMOSA‐PROMOTER BINDING PROTEIN‐LIKE 16 (SPL16) and SPL23 in leaves and shoot apices of Populus trees. Accordingly, either overexpression of MIR156a/c or mutagenesis of SPL16/23 resulted in the attenuation of growth cessation and bud set under short days (SD), whereas overexpression of SPL16 and SPL23 conferred early growth cessation.We further showed that SPL16 and SPL23 directly suppressed FLOWERING LOCUS T2 (FT2) expression while promoted BRANCHED1 (BRC1.1 and BRC1.2) expression. Moreover, we revealed that PIF8.1/8.2, positive regulators of growth cessation, directly bound to promoters of MIR156a and MIR156c and inhibited their expression to modulate downstream pathways.Our results reveal a connection between the phyB‐PIF8 module‐mediated photoperiod perception and the miR156‐SPL16/23‐FT2/BRC1 regulatory cascades in SD‐induced growth cessation. Brassica rapa, a major oilseed crop in high-altitude areas, is well known for its indeterminate inflorescences. However, this experiment revealed an intriguing anomaly within the plot: a variant displaying a determinate growth habit (520). Determinate inflorescences have been recognized for their role in the genetic enhancement of crops. In this study, a genetic analysis in a determinate genotype (520) and an indeterminate genotype (515) revealed that two independently inherited recessive genes (Brdt1 and Brdt2) are responsible for the determinate trait. BSA-seq and SSR markers were employed to successfully locate the Brdt1 gene, which is localized within an approximate region 72.7 kb between 15,712.9 kb and 15,785.6 kb on A10. A BLAST analysis of these candidate intervals revealed that Bra009508 (BraA10.TFL1) shares homology with the A. thaliana TFL1 gene. Then, BraA10.TFL1 (gene from the indeterminate phenotype) and BraA10.tfl1 (gene from the determinate phenotype) were cloned and sequenced, and the results indicated that the open reading frame of the alleles comprises 537 bp. Using qRT-PCR, it was determined that BraA10.TFL1 expression levels in shoot apexes were significantly higher in NIL-520 compared to 520. To verify the function of BraA10.TFL1, the gene was introduced into the determinate A. thaliana tfl1 mutant, resulting in the restoration of indeterminate traits. These findings demonstrate that BraA10.tfl1 is a gene that controls the determinate inflorescence trait. Vernalization plays a crucial role in the flowering and yield of Chinese cabbage, a process intricately influenced by long non-coding RNAs (lncRNAs). Our research focused on lncFLC1, lncFLC2a, and lncFLC2b, which emerged as key players in this process. These lncRNAs exhibited an inverse expression pattern to the flowering repressor genes FLOWERING LOCUS C 1 (BrFLC1) and FLOWERING LOCUS C 2 (BrFLC2) during vernalization, suggesting a complex regulatory mechanism. Notably, their expression in the shoot apex and leaves was confirmed through in fluorescent in situ hybridization (FISH). Furthermore, when these lncRNAs were overexpressed in Arabidopsis, a noticeable acceleration in flowering was observed, unveiling functional similarities to Arabidopsis's COLD ASSISTED INTRONIC NONCODING RNA (COOLAIR). This resemblance suggests a potentially conserved regulatory mechanism across species. Summary: Plants can record external stimuli in mobile mRNAs and systemically deliver them to distal tissues to adjust development. Despite the identification of thousands of mobile mRNAs, the functional relevance of mobile mRNAs remains limited.Many mobile mRNAs are synthesized in the source cells that perceive environmental stimuli, but specifically exert their functions upon transportation to the recipient cells. However, the translation of mobile mRNA‐encoded protein in the source cells could locally activate downstream target genes. How plants avoid ectopic functions of mobile mRNAs in the source cells to achieve tissue specificity remains to be elucidated.Here, we show that Arabidopsis AGAMOUS‐LIKE 24 (AGL24) is a mobile mRNA whose movement is necessary and sufficient to specify floral organ identity. Although AGL24 mRNA is expressed in vegetative tissues, AGL24 protein exclusively accumulates in the shoot apex. In leaves, AGL24 proteins are degraded to avoid ectopically activating its downstream target genes.Our results reveal how selective protein degradation in source cells provides a strategy to limit the local effects associated with proteins encoded by mobile mRNAs, which ensures that mobile mRNAs specifically trigger systemic responses only in recipient tissues. Since the first report of the tobamovirus tomato brown rugose fruit virus (ToBRFV) in 2014, it has become globally distributed. Its rapid spread has been primarily attributed to seed-borne transmission. Here, the seed-borne nature of ToBRFV transmission was investigated in different cultivars of tomato, bell pepper, and eggplant. In situ hybridization to localize the virus in reproductive organs of ToBRFV-infected tomato plants revealed that the virus was not present in shoot apices, flower buds, or in ovules during flower opening, indicating the virus may be restricted to the outer integument and transported in the vascular bundles during seed development. However, during early fruit development, the virus was present in the integuments in the ovule. Seeds of tomato cultivars with or without tobamovirus resistance gene Tm-22 transmitted the virus to the progeny seedlings at rates that reflected the ineffectiveness of the gene against ToBRFV. Seeds of bell peppers transmitted ToBRFV at higher rates than tomato seeds, but a bell pepper cultivar that has resistance gene L3 was not systemically infected, and its seeds did not harbor the virus. Three eggplant cultivars were systemically infected with ToBRFV but without showing any obvious symptoms, and even though ToBRFV was present in their seeds, the seedlings were not infected. ToBRFV was detected in the seed coats of contaminated tomato and bell pepper seeds, but not in eggplant seed coats. SUMMARY: Understanding how nutrient stress impacts plant growth is fundamentally important to the development of approaches to improve crop production under nutrient limitation. Here we applied single‐cell RNA sequencing to shoot apices of Pisum sativum grown under boron (B) deficiency. We identified up to 15 cell clusters based on the clustering of gene expression profiles and verified cell identity with cell‐type‐specific marker gene expression. Different cell types responded differently to B deficiency. Specifically, the expression of photosynthetic genes in mesophyll cells (MCs) was down‐regulated by B deficiency, consistent with impaired photosynthetic rate. Furthermore, the down‐regulation of stomatal development genes in guard cells, including homologs of MUTE and TOO MANY MOUTHS, correlated with a decrease in stomatal density under B deficiency. We also constructed the developmental trajectory of the shoot apical meristem (SAM) cells and a transcription factor interaction network. The developmental progression of SAM to MC was characterized by up‐regulation of genes encoding histones and chromatin assembly and remodeling proteins including homologs of FASCIATA1 (FAS1) and SWITCH DEFECTIVE/SUCROSE NON‐FERMENTABLE (SWI/SNF) complex. However, B deficiency suppressed their expression, which helps to explain impaired SAM development under B deficiency. These results represent a major advance over bulk‐tissue RNA‐seq analysis in which cell‐type‐specific responses are lost and hence important physiological responses to B deficiency are missed. The reported findings reveal strategies by which plants adapt to B deficiency thus offering breeders a set of specific targets for genetic improvement. The reported approach and resources have potential applications well beyond P. sativum species and could be applied to various legumes to improve their adaptability to multiple nutrient or abiotic stresses. Turions are vegetative, dormant, and storage overwintering organs formed in perennial aquatic plants in response to unfavorable ecological conditions and originate by extreme condensation of apical shoot segments. The contents of cytokinins, auxins, and abscisic acid were estimated in shoot apices of summer growing, rootless aquatic carnivorous plants, Aldrovanda vesiculosa and Utricularia australis, and in developing turions at three stages and full maturity to reveal hormonal patterns responsible for turion development. The hormones were analyzed in miniature turion samples using ultraperformance liquid chromatography coupled with triple quadrupole mass spectrometry. Photosynthetic measurements in young leaves also confirmed relatively high photosynthetic rates at later turion stages. The content of active cytokinin forms was almost stable in A. vesiculosa during turion development but markedly decreased in U. australis. In both species, auxin content culminated in the middle of turion development and then decreased again. The content of abscisic acid as the main inhibitory hormone was very low in growing plants in both species but rose greatly at first developmental stages and stayed very high in mature turions. The hormonal data indicate a great strength of developing turions within sink–source relationships and confirm the central role of abscisic acid in regulating the turion development. In the monoecious Cucurbita pepo, the transition to female flowering is the time at which the plant starts the production of female flowers after an initial male phase of development. Ethylene plays an essential role in this process since some ethylene deficient and ethylene-insensitive mutants are androecious and only produce male flowers. To gain insight into the molecular mechanisms regulating the specification and early development of female flowers, we have compared the transcriptomic changes occurring in the shoot apices of WT and androecious ethylene-insensitive etr1b mutant plants upon female flowering transition. There were 1160 female flowering-specific DEGs identified in WT plants upon female flowering, and 284 of them were found to be modulated by the ethylene-insensitive etr1b mutation. The function of these DEGs indicated that female flower specification depends on the adoption of a transcriptional program that includes previously identified sex-determining genes in the ethylene pathway, but also genes controlling the biosynthesis and signaling pathways of other phytohormones, and those encoding for many different transcription factors. The transcriptomic changes suggested that gibberellins play a negative role in female flowering, while ethylene, auxins, ABA and cytokinins are positive regulators. Transcription factors from 34 families, including NAC, ERF, bHLH, bZIP, MYB and C2H2/CH3, were found to be regulating female flowering in an ethylene-dependent or -independent manner. Exogenous ethylene is commonly utilized to initiate flower induction in pineapple (Ananas comosus (L.) Merr.). However, the molecular mechanisms and metabolic changes involved are not well understood. In this study, we explored the genetic network and metabolic shifts in the 'Comte de Paris' pineapple variety during ethylene-induced flowering. This was achieved through an integrative analysis of metabolome and transcriptome profiles at vegetative shoot apexes (0 d after ethephon treatment named BL_0d), the stage of bract primordia (8 d after ethephon treatment named BL_8d), stage of flower primordia (18 d after ethephon treatment named BL_18d), and the stage of stopped floret differentiation (34 d after ethephon treatment named BL_34d). We isolated and identified 804 metabolites in the pineapple shoot apex and inflorescence, categorized into 24 classes. Notably, 29, 31, and 46 metabolites showed significant changes from BL_0d to BL_8d, BL_8d to BL_18d, and BL_18d to BL_34d, respectively. A marked decrease in indole was observed, suggesting its role as a characteristic metabolite during flower induction. Transcriptomic analysis revealed 956, 1768, and 4483 differentially expressed genes (DEGs) for BL_0d vs. BL_8d, BL_8d vs. BL_18d, and BL_18d vs. BL_34d, respectively. These DEGs were significantly enriched in carbohydrate metabolism and hormone signaling pathways, indicating their potential involvement in flower induction. Integrating metabolomic and transcriptomic data, we identified several candidate genes, such as Agamous-Like9 (AGL9), Ethylene Insensitive 3-like (ETIL3), Apetala2 (AP2), AP2-like ethylene-responsive transcription factor ANT (ANT), and Sucrose synthase 2 (SS2), that play potentially crucial roles in ethylene-induced flower induction in pineapple. We also established a regulatory network for pineapple flower induction, correlating metabolites and DEGs, based on the Arabidopsis thaliana pathway as a reference. The shoot apical meristem (SAM) of higher plants comprises distinct functional zones. The central zone (CZ) is located at the meristem summit and harbors pluripotent stem cells. Stem cells undergo cell division within the CZ and give rise to descendants, which enter the peripheral zone (PZ) and become recruited into lateral organs. Stem cell daughters that are pushed underneath the CZ form rib meristem (RM). To unravel the mechanism of meristem development, it is essential to know how stem cells adopt distinct cell fates in the SAM. Here, we show that meristem patterning and floral organ primordia formation, besides auxin transport, are regulated by auxin biosynthesis mediated by two closely related genes of the TRYPTOPHAN AMINOTRANSFERASE family. In Arabidopsis SAM, TAA1 and TAR2 played a role in maintaining auxin responses and the identity of PZ cell types. In the absence of auxin biosynthesis and transport, the expression pattern of the marker genes linked to the patterning of the SAM is perturbed. Phytoplasma, plant pathogenic Mollicutes that have a trans-kingdom life cycle, are insect transmitted and have been found to be associated with yield affecting traits in a large number of taxa. Many peculiar symptoms observed in the host taxa are often results of dramatic alterations in the normal development program that are generally controlled at the meristematic regions located in the shoot apices. Phytoplasma being vasculature limited are thus able to bring about genetic reprogramming in the regions of host plant that are far removed from their natural niche namely sieve elements of the phloem. Several proteins secreted by phytoplasma in the host plants have been identified and termed as effector molecules namely SAP54, SAP11, TENGU, SAP21 etc. that enable the colonisation, survival, spread of phytoplasma and also bring about dramatic alterations in the host plant. Nevertheless, the mechanisms underlying these peculiar phenomena are far from understood and remain a challenging area for the phytoplasma biologists. A thorough understanding of the processes involved is needed to provide platforms for developing control measures for phytoplasma associated diseases that will also enhance basic understanding on the plant developmental programs affected. An efficient multiple shoot induction, plant regeneration, and subsequent genetic transformation system for two locally adapted recalcitrant indica rice cultivars ASD16 and ADT43 using shoot apex explants were established. Three different plant growth regulators (PGRs) were tested for their efficacy in forming multiple shoots from shoot apices. Thidiazuron (TDZ) (5.0 mg/L) induced the highest frequencies of multiple shoot induction compared to Benzyl Amino Purine (BAP) and Kinetin (Kin). Shoot apices cultured on MS basal medium devoid of plant growth regulators formed single shoots. All tested PGRs produced multiple shoots at concentrations higher than 3.0 mg/L. Indole-3-Acetic Acid (IAA) and Indole-3-Butyric Acid (IBA) were supplemented to media containing TDZ (5.0 mg/L) for assessing the root induction efficiencies and it was found that media devoid of growth hormones were sufficient to produce better-rooting frequencies comparatively. Among the two tested cultivars, ASD16 responded with better shooting frequencies than ADT43. Further, genetic transformation experiments were carried out with the shoot apex explants of both cultivars. The putative transgenic plants were screened and confirmed by transient GUS expression, PCR, and Southern hybridization in both T0 and T1 generations, respectively. This rapid and efficient regeneration system amenable to Agrobacterium-mediated genetic transformation from shoot apices may be used for genetic manipulation of recalcitrant indica varieties for varietal crop improvement strategies. In temperate and boreal regions, perennial plants adapt their annual growth cycle to the change of seasons. In natural forests, juvenile seedlings usually display longer growth seasons compared to adult trees to ensure their establishment and survival under canopy shade. However, how trees adjust their annual growth according to their age is not known. In this study, we show that age-dependent seasonal growth cessation is genetically controlled and found that the miR156-SPL3/5 module, a key regulon of vegetative phase change (VPC), also triggers age-dependent growth cessation in Populus trees. We show that miR156 promotes shoot elongation during vegetative growth, and its targets SPL3/5s function in the same pathway but as repressors. We find that the miR156-SPL3/5s regulon controls growth cessation in both leaves and shoot apices and through multiple pathways, but with a different mechanism compared to how the miR156-SPL regulon controls VPC in annual plants. Taken together, our results reveal an age-dependent genetic network in mediating seasonal growth cessation, a key phenological process in the climate adaptation of perennial trees. The high-affinity K+ transporter (HAK) family, the most prominent potassium transporter family in plants, which involves K+ transport, plays crucial roles in plant responses to abiotic stresses. However, the HAK gene family remains to be characterized in quinoa (Chenopodium quinoa Willd.). We explored HAKs in quinoa, identifying 30 members (CqHAK1–CqHAK30) in four clusters phylogenetically. Uneven distribution was observed across 18 chromosomes. Furthermore, we investigated the proteins' evolutionary relationships, physicochemical properties, conserved domains and motifs, gene structure, and cis-regulatory elements of the CqHAKs family members. Transcription data analysis showed that CqHAKs have diverse expression patterns among different tissues and in response to abiotic stresses, including drought, heat, low phosphorus, and salt. The expressional changes of CqHAKs in roots were more sensitive in response to abiotic stress than that in shoot apices. Quantitative RT-PCR analysis revealed that under high saline condition, CqHAK1, CqHAK13, CqHAK19, and CqHAK20 were dramatically induced in leaves; under alkaline condition, CqHAK1, CqHAK13, CqHAK19, and CqHAK20 were dramatically induced in leaves, and CqHAK6, CqHAK9, CqHAK13, CqHAK23, and CqHAK29 were significantly induced in roots. Our results establish a foundation for further investigation of the functions of HAKs in quinoa. It is the first study to identify the HAK gene family in quinoa, which provides potential targets for further functional study and contributes to improving the salt and alkali tolerance in quinoa. Dichanthium annulatum is a popular halophyte C4 perennial forage crop of semi-arid tropics that reproduces apomictically. In vitro plant regeneration in apomictic species helps in producing new germplasm that can supplement existing breeding program. Field-collected seeds and in vitro shoot apices were tested for response to 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP) for initiation of somatic embryogenesis (SE) and direct shoot organogenesis (DSO). Explants produced callus on Murashige and Skoog (MS) medium with varying level of 2,4-D and 0.5 mgL–1 BAP. After 4th sub-culture, yellowish brown turns into white nodular callus tissue with a friable appearance, which exhibited the highest percentage of embryogenic callus induction on MS medium with 3.0 mgL–1 2,4-D and 0.5 mgL–1 BAP, was transferred to MS with BAP or kinetin and 0.125 mgL–1 2,4-D to promote somatic embryo germination into plantlet. The developmental stages of SE were identified using scanning electron microscopy and histology. Multiple shoots were observed with the combinations of BAP or kinetin with 2,4-D. The highest number of shoots (27) occurred on MS medium comprising 4.0 mgL–1 BAP and 0.125 mgL–1 2,4-D. In vitro plantlet was rooted on MS medium with 0.4% charcoal. Southern tomato virus (STV) is a dsRNA virus, which belongs to the newly formed Amalgavirus genus of the Amalgaviridae family. Currently there is no report regarding the presence of STV in tomato tissues. In this study, we performed in situ hybridization to examine the distribution of STV in host tissues. STV was found in the leaves, stems, seeds, shoot apexes and root tips of tomato and localized in the cortex tissue, vascular tissues, pith, seed coat, endosperm, cotyledon (including inner cotyledons and outer cotyledons), hypocotyls and radicles of infected tomato tissues. In potato, high levels of nitrogen (N) can lead to excessive vegetative growth at the expense of tuber development, resulting in lower yield and poor-quality tubers. We found that Solanum tuberosum CLE4 (StCLE4) is expressed most strongly in the roots grown in N-rich media, and it positively regulates potato root growth under N-deficient conditions. We noted that StCLE4 functions as a negative regulator of normal shoot apex development similar to CLV3 in Arabidopsis. Transcriptomic analysis revealed that overexpression of StCLE4 resulted in the repression of the StIT1 gene, a regulator of potato tuber initiation. StCLE4-overexpressing stolons were converted into branches, that were similar to a mild phenotype of the it1 (identity of tuber 1) mutant. We also found that NIN-like proteins, key regulators of nitrate signaling bind to the regulatory sequence of StIT1 in a yeast one-hybrid assay. Iochroma orozcoae A.Orejuela & S.D.Sm., sp. nov. (Solanaceae) is described from the Andean forests of Cundinamarca in the eastern cordillera of Colombia. Iochroma orozcoae was first collected by the eminent Spanish priest and botanist José Celestino Mutis in the late part of the 18th century, but the specimens have lain unrecognised in herbaria for over 200 years. The species shares many features with its closest relative, Iochroma baumii S.D.Sm. & S.Leiva, but it differs from it in having small flowers with five corolla lobes and few inflorescences per branch, located near the shoot apex with 1 to 4 (-8) flowers, fruits that are greenish-yellow when ripe and its restricted geographic distribution. A description of I. orozcoae is provided, along with a detailed illustration, photographs of live plants, a comparison with closely-related species and a key to all Colombian species of Iochroma Benth. Light and photoperiod are environmental signals that regulate flowering transition. In plants like Arabidopsis thaliana, this regulation relies on CONSTANS, a transcription factor that is negatively posttranslational regulated by phytochrome B during the morning, while it is stabilized by PHYA and cryptochromes 1/2 at the end of daylight hours. CO induces the expression of FT, whose protein travels from the leaves to the apical meristem, where it binds to FD to regulate some flowering genes. Although PHYB delays flowering, we show that light and PHYB positively regulate XAANTAL1 and other flowering genes in the shoot apices. Also, the genetic data indicate that XAL1 and FD participate in the same signaling pathway in flowering promotion when plants are grown under a long-day photoperiod at 22 °C. By contrast, XAL1 functions independently of FD or PIF4 to induce flowering at higher temperatures (27 °C), even under long days. Furthermore, XAL1 directly binds to FD, SOC1, LFY, and AP1 promoters. Our findings lead us to propose that light and temperature influence the floral network at the meristem level in a partially independent way of the signaling generated from the leaves. The application of organic amendments, biochar, and wood distillate (WD), as well as the exposure to UV-B radiation, are two sustainable ways to enhance soil fertility and increase plant nutraceutical quality, respectively. However, they have always been studied separately, without testing the eventual synergistic or antagonistic effect when applied together. The present study investigated the effects of biochar (2% w/w), WD (1:100), and their combination (BWD) on some biometric and biochemical parameters of basil plants (Ocimum basilicum L.) exposed to different doses of UV-B radiation (0, 1, 2 h d−1; UV-B irradiance of 1.36 W m−2) in controlled conditions. Root and stem length and weight were not affected by soil amendments, while 1 h d−1 UV-B increased the length (+28%) and weight (+62%) of the aerial part. When combining the above- and below-ground factors, a decrease in root length was observed in the 2 h d−1 UV-B-treated plants in both WD (−36%) and BWD (−39%) treatments. The co-application of below- and above-ground treatments generally decreased phenolic and flavonoid concentration in both fully expanded leaves and vegetative shoot apices. This preliminary study highlights an antagonistic action of the combination of the investigated factors, at these doses, on the plant growth and metabolism that should be considered. Cannabis sativa ("cannabis" hereafter) is a valuable recent addition to Canada's economy with the legalization for recreational use in 2018. The vast majority of indoor cannabis cultivators use a 12-h light/12-h dark photoperiod to promote flowering. To test the hypothesis that robust flowering initiation responses can be promoted in indoor-grown cannabis cultivars under longer photoperiods, clones of ten drug-type cannabis cultivars were grown under six photoperiod treatments. All treatments were based on a standard 24-h day and included 12 h, 12.5 h, 13 h, 13.5 h, 14 h, and 15 h of light. The plants were grown in a growth chamber for 3 to 4 weeks, receiving an approximate light intensity of 360 µmol·m−2·s−1 from white LEDs. Flowering initiation, defined as the appearance of ≥3 pairs of stigmas at the apex of the primary shoot, occurred in all cultivars under all photoperiod treatments up to 14 h. Delays in flowering initiation time under 14 h vs. 12 h ranged from no delay to approximately 4 days, depending on the cultivar. Some cultivars also initiated flowering under 15 h, but floral tissues did not further develop beyond the initiation phase. Harvest metrics of some cultivars responded quadratically with increasing photoperiod, with ideal levels of key flowering parameters varying between 12 h and 13 h. These results suggest there is potential to increase yield in some indoor-grown cannabis cultivars by using longer than 12-h photoperiods during the flowering stage of production. This is attributed to the inherently higher daily light integrals. Breeding rapeseed varieties with more main inflorescence siliques is an idea for developing rapeseed varieties that are suitable for light and simplified cultivation. The Brassica napus exhibited cluster bud of the main inflorescence (Bnclib) gene. At the fruiting stage, the main inflorescence had more siliques, higher density, and more main inflorescences. Moreover, the top of the main inflorescence bifurcated. Genetic analysis showed that the separation ratio between Bnclib and the wild type in the F2 generation was 3:1, which indicated that the trait was a single-gene-dominant inheritance. Among the 24 candidate genes, only one gene, BnaA03g53930D, showed differential expression between the groups (False discovery rate, FDR ≤ 0.05, |log2FC|≤ 1). qPCR verification of the BnaA03g53930D gene between Huyou 17 and its Bnclib near-isogenic line showed that BnaA03g53930D was significantly differentially expressed in the stem tissue of Huyou 17 and its Bnclib near-isogenic line (Bnclib NIL). The determination of gibberellin (GA), brassinolide (BR), cytokinin (CTK), jasmonic acid (JA), growth hormone (IAA), and strigolactone (SL) content in the shoot apex of Huyou 17 by Bnclib NIL and wild type showed that all six hormones significantly differed between the Bnclib NIL and Huyou 17. Summary: Alternative splicing of pre‐mRNAs is crucial for plant growth and development. Serine/arginine‐rich (SR) proteins are a conserved family of RNA‐binding proteins that are critical for both constitutive and alternative splicing. However, how phosphorylation of SR proteins regulates gene transcription and alternative splicing during plant development is poorly understood.We found that the Arabidopsis thaliana L. SR protein‐specific kinase II family proteins (SRPKIIs) play an important role in plant development, including flowering. SRPKIIs regulate the phosphorylation status of a subset of specific SR proteins, including SR45 and SC35, which subsequently mediates their subcellular localization.A phospho‐dead SR45 mutant inhibits the assembly of the apoptosis‐and splicing‐associated protein complex and thereby upregulates the expression of FLOWERING LOCUS C (FLC) via epigenetic modification. The splicing efficiency of FLC introns was significantly increased in the shoot apex of the srpkii mutant. Transcriptomic analysis revealed that SRPKIIs regulate the alternative splicing of c. 400 genes, which largely overlap with those regulated by SR45 and SC35‐SCL family proteins.In summary, we found that Arabidopsis SRPKIIs specifically affect the phosphorylation status of a subset SR proteins and regulate the expression and alternative splicing of FLC to control flowering time. Anthocyanins are important natural plant pigments and play diverse roles in plant growth and adaptation. Anthocyanins function as screens to protect photosynthetic tissues from photoinhibition. However, the regulatory mechanisms underlying the biosynthesis and spatial accumulation pattern of anthocyanins remain some unresolved issues. Here, we demonstrate that the GARP‐type transcription factor GOLDEN2‐LIKE 1 (GLK1) functions as a positive factor in anthocyanin accumulation. GLK1 enhances the transcriptional activation activities of MYB75, MYB90, and MYB113 via direct protein‐protein interactions to increase the expression of anthocyanin‐specific biosynthetic genes. Anthocyanins accumulate in an acropetal manner in Arabidopsis. We also found that the expression pattern of GLK1 overall mimicked the accumulation pattern of anthocyanin from the base of the main stem to the shoot apex. Based on these findings, we established a working model for the role of GLK1 in anthocyanin accumulation and propose that GLK1 mediates the spatial distribution pattern of anthocyanins by affecting the transcriptional activation activities of MYB75, MYB90, and MYB113. The aim of the study was to develop an efficient micropropagation protocol of Origanum scabrum, which will allow its commercial exploitation in the pharmaceutical and horticultural industries. First, the effect of the date of the explant collection (20 April, 20 May, 20 June, 20 July, 20 August) and the position of the explant on the plant stem (shoot apex, 1st node, 3rd node, 5th node) on the establishment of in vitro cultures was studied (1st experiment: Stage I). Next, the effect of temperature (15 °C, 25 °C) and the node position (microshoot apex, 1st node, 5th node) on the microplant production and ex vitro survival of plantlets was studied (2nd experiment: Stage II). The optimum season to collect explants from wild plants was shown to be during the vegetative growth of the plants (April to May), while the shoot apex and the 1st node were the most suitable explants. For the proliferation and production of rooted microplants, the best results were obtained from single-node explants excised from microshoots produced from 1st node-explants collected on 20th of May. Temperature did not affect microshoot number, leaf number and the percentage of rooted microplants, while microshoot length was higher at 25 °C. Moreover, microshoot length and the percentage of rooted microplants were higher in those derived from apex explants, while the survival of plantlets was not affected by treatments and ranged between 67% and 100%. Gene duplication played a fundamental role in eukaryote evolution and different copies of a given gene can be present in extant species, often with expressions and functions differentiated during evolution. We assume that, when such differentiation occurs in a gene copy, this may be indicated by its maintenance in all the derived species. To verify this hypothesis, we compared the histological expression domains of the three β-glucuronidase genes (AtGUS) present in Arabidopsis thaliana with the GUS evolutionary tree in angiosperms. We found that AtGUS gene expression overlaps in the shoot apex, the floral bud and the root hairs. In the root apex, AtGUS3 expression differs completely from AtGUS1 and AtGUS2, whose transcripts are present in the root cap meristem and columella, in the staminal cell niche, in the epidermis and in the proximal cortex. Conversely, AtGUS3 transcripts are limited to the old border-like cells of calyptra and those found along the protodermal cell line. The GUS evolutionary tree reveals that the two main clusters (named GUS1 and GUS3) originate from a duplication event predating angiosperm radiation. AtGUS3 belongs to the GUS3 cluster, while AtGUS1 and AtGUS2, which originate from a duplication event that occurred in an ancestor of the Brassicaceae family, are found together in the GUS1 cluster. There is another, previously undescribed cluster, called GUS4, originating from a very ancient duplication event. In domesticated apple (Malus x domestica Borkh.) and other woody perennials, floral initiation can be repressed by gibberellins (GAs). The associated mechanism is a major unanswered question in plant physiology, and understanding organismal aspects of GA signaling in apple has important commercial applications. In plants, the major mechanism for elimination of GAs and resetting of GA signaling is through catabolism by GA2-oxidases (GA2ox). We found that the GA2ox gene family in apple comprises 16 genes representing eight, clearly defined homeologous pairs, which were named as MdGA2ox1A/1B to MdGA2ox8A/8B. Expression of the genes was analyzed in the various structures of the spur, where flowers are initiated, as well as in various structures of seedlings over one diurnal cycle and in response to water-deficit and salt stress. Among the results, we found that MdGA2ox2A/2B dominated expression in the shoot apex and were strongly upregulated in the apex after treatment with exogenous GA3, suggesting potential involvement in repression of flowering. Several MdGA2ox genes also showed preferential expression in the leaf petiole, fruit pedicel, and the seed coat of developing seeds, potentially representing mechanisms to limit diffusion of GAs across these structures. In all contexts studied, we documented both concerted and distinct expression of individual homeologs. This work introduces an accessible woody plant model for studies of GA signaling, GA2ox gene regulation, and conservation/divergence of expression of homeologous genes, and should find application in development of new cultivars of apple and other tree fruits. X‐ray diffraction and small‐angle neutron scattering were used to identify the naturally occurring hormone auxin in Madeira vine. Two kinds of auxins belonging to indole‐3‐acetic acid (IAA, C10H9NO2) and 4‐chloroindole‐3‐acetic acid (4‐Cl‐IAA, C10H8ClNO2) have been found. The appearance of well‐defined x‐ray diffraction peaks from the IAA as well as from the 4‐Cl‐IAA structures shows that the IAA and 4‐Cl‐IAA molecules that are produced at the shoot apex crystallize into periodic arrangements, rather than stay in molecular forms, upon transport downward through in stem. Small angle neutron scattering spectra reveal a progressive increase in the length of crystallized IAA and 4‐Cl‐IAA on transported downward from the shoot apex to the stem, reaching 59 nm in length in the area 8–9 cm below the shoot apex. A portion of IAA loses their carboxyl group COOH becoming inactive in stimulating cell growth, which slows down the growth in an aged stem. [ABSTRACT FROM AUTHOR]

We investigated the transcriptomic changes in the shoot apices during floral transition in Arabidopsis mutants of two closely related splicing factors: AtU2AF65a (atu2af65a) and AtU2AF65b (atu2af65b). The atu2af65a mutants exhibited delayed flowering, while the atu2af65b mutants showed accelerated flowering. The underlying gene regulatory mechanism of these phenotypes was unclear. We performed RNA-seq analysis using shoot apices instead of whole seedlings and found that the atu2af65a mutants had more differentially expressed genes than the atu2af65b mutants when they were compared to wild type. The only flowering time gene that was significantly up- or down-regulated by more than two-fold in the mutants were FLOWERING LOCUS C (FLC), a major floral repressor. We also examined the expression and alternative splicing (AS) patterns of several FLC upstream regulators, such as COOLAIR, EDM2, FRIGIDA, and PP2A-b'ɤ, and found that those of COOLAIR, EDM2, and PP2A-b'ɤ were altered in the mutants. Furthermore, we demonstrated that AtU2AF65a and AtU2AF65b genes partially influenced FLC expression by analyzing these mutants in the flc-3 mutant background. Our findings indicate that AtU2AF65a and AtU2AF65b splicing factors modulate FLC expression by affecting the expression or AS patterns of a subset of FLC upstream regulators in the shoot apex, leading to different flowering phenotypes.
