Abstract Excessive accumulation of reactive oxygen and nitrogen species (RONS) and dysbiosis of intestinal microbiota are pivotal symptoms for inflammatory bowel disease (IBD) and its associated complications, such as intestinal fibrosis. This research introduces a probiotic inulin hydrogel loaded with polypyrrole (PPy) nanozymes and antifibrotic drug pirfenidone (PFD) (PPy/PFD@Inulin gel) designed for the concurrent amelioration of IBD and its fibrotic complication. Upon oral administration, the inulin gel matrix could extend the gastrointestinal residence time of PPy nanozymes and PFD, facilitating the efficient reduction of pro-inflammatory cytokine levels and enhancement of the intestinal epithelial barrier repair as well as the suppression of intestinal fibrosis through sustained RONS scavenging, modulation of gut microbiota and attenuation of the TGF-β/Smad signaling pathway to inhibit fibroblast proliferation. Notably, the PPy/PFD@Inulin gel demonstrated significant prophylactic and therapeutic efficacy in acute and chronic colitis as well as intestinal fibrosis induced by dextran sodium sulfate (DSS) in mouse models. Thus, the engineered ternary PPy/PFD@Inulin gel offered a pioneered paradigm for simultaneous reversal of IBD and its associated complications, such as intestinal fibrosis, in a single therapeutic regimen.

Objective: To investigate the role of RPRD1B in the progression of diffuse large B-cell lymphoma (DLBCL) and its potential as a therapeutic target. Methods: This study analyzed RPRD1B expression in DLBCL and normal tissues using public databases and assessed its prognostic impact through survival analysis. In vitro and in vivo experiments were conducted to explore the mechanisms by which RPRD1B influences tumor growth and apoptosis. Results: RPRD1B expression was significantly elevated in DLBCL compared to normal tissues and was associated with poor prognosis. In vitro and in vivo experiments demonstrated that RPRD1B promoted lymphoma cell proliferation and inhibited apoptosis through the NF-κB signaling pathway. Conclusions: RPRD1B plays a critical role in the progression of DLBCL by modulating apoptosis and cellular proliferation. Targeting RPRD1B may offer a novel therapeutic strategy for DLBCL, suggesting its potential as a prognostic marker and therapeutic target in hematological malignancies.

Abstract This registration study assessed clinical outcomes of TQ-B3525, the dual phosphatidylinositol-3-kinase (PI3K) α/δ inhibitor, in relapsed and/or refractory follicular lymphoma (R/R FL). This phase II study (ClinicalTrials.gov NCT04324879. Registered March 27, 2020) comprised run-in stage and stage 2. R/R FL patients after ≥2 lines therapies received oral 20 mg TQ-B3525 once daily in a 28-day cycle until intolerable toxicity or disease progression. Primary endpoint was independent review committee (IRC)-assessed objective response rate (ORR). Based on results (ORR, 88.0%; duration of response [DOR], 11.8 months; progression-free survival [PFS], 12.0 months) in 25 patients at run-in stage, second stage study was initiated and included 82 patients for efficacy/safety analysis. Patients received prior-line (median, 3) therapies, with 56.1% refractory to previous last therapies; 73.2% experienced POD24 at baseline. At stage 2, ORR was 86.6% (71/82; 95% CI, 77.3–93.1%), with 28 (34.2%) complete responses. Disease control rate was 95.1% due to 7 (8.5%) stable diseases. Median time to response was 1.8 months. Among 71 responders, median DOR was not reached; 18-month DOR rate was 51.6%. with median follow-up of 13.3 months, median PFS was 18.5 (95% CI, 10.2-not estimable) months. Median overall survival (OS) was not reached by cutoff date; 24-month OS rate was estimated as 86.1%. Response rates and survival data were consistent across all subgroups. Grade 3 or higher treatment-related adverse events were observed in 63 (76.8%) cases, with neutropenia (22.0%), hyperglycemia (19.5%), and diarrhea (13.4%) being common. TQ-B3525 showed favorable efficacy and safety for R/R FL patients after ≥2 lines prior therapies.

Over the last 20 years, breakthroughs in accessible therapies for the treatment of multiple myeloma (MM) have been made. Nevertheless, patients with MM resistant to immunomodulatory drugs, proteasome inhibitors, and anti-CD38 monoclonal antibodies have a very poor outcome. Therefore, it is necessary to explore new drugs for the treatment of MM. This review summarizes the mechanism of action of selinexor, relevant primary clinical trials, and recent developments in both patients with relapsed/refractory myeloma and patients with newly diagnosed myeloma. Selinexor may be useful for the treatment of refractory MM.

Objective To evaluate the impacts of equal ratio ventilation (ERV) mode on lung morphology, gastric inflation and laryngeal mask airway ventilation during laparoscopic radical prostatectomy by lung ultrasound（LUS） and gastric ultrasound. Methods A prospective study was performed in 60 patients with prostate cancer undergoing laparoscopy from June 2021 to May 2022 in Nanjing First Hospital. The patients were randomly divided into ERP group (group E) and conventional ventilation group (group C,n=30, each). After induction of routine intravenous anesthesia and mechanical ventilation with laryngeal mask airway, and the ventilation mode was set as follows: the inspiratory-to-expiratory ratio in group E was 1∶1, while that in group C was 1∶2. After anesthesia induction (T0), 30 minutes after surgery begin (T1), 60 minutes after surgery begin (T2) and 10 minutes after surgery (T3), the peak airway pressure (Ppeak), airway plateau pressure(Pplat), mean airway pressure (Pmean),end-expiratory partial pressure of carbon dioxide (PETCO2) and minute ventilation (MV) were observed and compared between two groups. The LUS score, the cross-sectional area of gastric antrum and fundus and the incidence of airway complications were recorded in two groups. Results At T1 and T2, Ppeak in group E was significantly lower than that in group C(P＜0.05), and Pmean was significantly higher than that in group C(P＜0.05). There was no significant difference in PETCO2 and MV between two groups at different time points (P＞0.05). The LUS and the incidence of atelectasis in group E were statistically lower than those in group C (P＜0.05), and there was no significant difference in the cross-sectional area of gastric antrum and fundus（P＞0.05） and the incidence of airway complications（30.0% vs 32.1%，χ2=0.031，P=0.860) between two groups. Conclusion ERV mode during laparoscopic radical prostatectomy can reduce Ppeak, leakage rate of laryngeal mask, atelectasis and LUS scores without increasing the incidence of gastric inflation and laryngeal mask-related adverse complications.

The creep of face-slab concrete in a rockfill dam is critical for determining the restrained stress and cracking resistance of the concrete at an early age. In this article, the mix proportion of the face-slab concrete for a rockfill dam under construction without any cracking resistance additive was taken as the reference concrete (called JC) mix proportion. The carbon nanotubes (CNTs) and magnesium oxide expansion agent (MEA) were incorporated into JC to prepare face-slab concretes called NC and PC, respectively. The temperature–stress tests under temperature matching curing (TMC) and constant temperature curing (CTC) modes were conducted on these three kinds of concretes to investigate the effects of CNTs and MEA on the early-age creep properties of the face-slab concrete under variable stress conditions. The results showed that the creep performance of NC concrete under CTC mode was lower than that under TMC mode. Combined with mercury intrusion porosimetry test results, the mechanism of the effect of CNTs and MEA on creep was analyzed. The results showed that the temperature change may lead to the CNTs debonding from the cementitious matrix or matrix cracking for the NC concrete. The incorporation of CNTs can increase the early-age creep and improve the cracking resistance of concrete.

BackgroundFailure to CD19-targeted chimeric antigen receptor T-cell (CAR-T) therapy for patients with relapsed/refractory (R/R) diffuse large B-cell lymphoma (DLBCL), is an emerging clinical problem. There is no consensus on the treatment for these patients and treatment remains empirical.Case ReportWe reported a case of an elderly R/R DLBCL patient who had TP53 mutation and relapsed 12 months after initial response to CAR T-cell therapy. The patient did not respond to salvage chemotherapy with the GDP regimen and could not tolerate any aggressive chemotherapy. Thereafter, the patient was given chidamide and zanubrutinib. After two months of treatment, the patient achieved sustained complete remission. At the last follow-up, the patient remains in radiographic CR 22 months after CAR-T infusion and 10 months after the initiation of the combination treatment.ConclusionWe report the first successful case of dual inhibition of HDAC and BTK for the treatment of R/R DLBCL after failure to CAR-T cell therapy, which opens a new therapeutic possibility for the future.

Abstract Sb‐based oxides electrodes have received tremendous attention owing to their high theoretical capacities and electrochemical activities. However, the huge volume change during ion insertion/extraction process and poor electronic conductivity limit their wide applications in the electrochemical energy storage. In this study, SbO2/Sb2O3 nanoparticles embedded in N‐doped carbon fibers are successfully synthesized. The composite can display a high reversible specific capacity of 622 mAh g−1 for lithium‐ion batteries and a desirable reversible capacity 307 mAh g−1 for sodium‐ion batteries at current density of 100 mA g−1. Such superior lithium and sodium storage performance can be ascribed to that carbon fibers can effectively restrict the large volume change and enhance the electrochemical stability.

[Introduction]Road design for heliostat field is the important part in the design of solar power tower plant. To reduce power plant investment and improve the efficiency of heliostat field, road design shall be scientific, effective and reasonable. [Method] In this paper, the roads of heliostat field were divided into four categories: main road, radiation road, peripheral ring road and inter-heliostat road. The factors to be considered in the design were analyzed in detail, and the road layout, structural design and ancillary facilities were studied respectively. [Result] The principles and requirements of the road layout are summarized. It is proposed that the pavement structure design should be based on the functional requirements, economic rationality, construction, local material and maintenance conditions. The pavement structure of inter-heliostat road and the road sign system in heliostat field is suggested. [Conclusion] The paper is used for reference in the road design for heliostat field in the future.

[Introduction] In order to analyze the reasonbility of the fan configuration of a 350 MW supercritical heating unit. [Method] In this paper, the single-row configuration and double-row fan configuration was compared, it took the reliability of fans, the requirements of engineering heating and minimum annual fee method into congsideration. [Result] The results demonstrate that the unstopped reliability rate of single-row fan configuration is slightly lower than double-row fan configuration. However, under the condition of high load operation, the heating supply reliability of single-row fan is higher.Single-row fan configuration econmic advantage is obvious.Therefore, it is more reasonable to adopt single-row fan for heating units which operats under high load operation for a long time. [Conclusion] The conclusion of this study is not only applicable to this project, but also can provide guidance for other heating unit.

Most randomized trials for acute promyelocytic leukemia (APL) have investigated highly selected patients under idealized conditions, and the findings need to be validated in the real world. We conducted a population-based study of all APL patients in Zhejiang Province, China, with a total population of 82 million people, to assess the generalization of all-trans retinoic acid (ATRA) and arsenic as front-line treatment. The outcomes of APL patients were also analyzed. Between January 2015 and December 2019, 1,233 eligible patients were included in the final analysis. The rate of ATRA and arsenic as front-line treatment increased steadily from 66.2% in 2015 to 83.3% in 2019, with no difference among the size of the center (≥5 or 10 × 109/L) were independent risk factors for ED and OS in the multivariate analysis. This population-based study showed that ATRA and arsenic as front-line treatment are widely used under real-world conditions and yield a low ED rate and a high survival rate, which mimic the results from clinical trials, thereby supporting the wider application of APL guidelines in the future.

Dysregulation of protein posttranslational modification (PTM) can lead to a variety of pathological processes, such as abnormal sperm development, malignant tumorigenesis, depression, and aging process. SIRT7 is a NAD+-dependent protein deacetylase. Besides known deacetylation, SIRT7 may also have the capacity to remove other acylation. However, the roles of SIRT7-induced other deacylation in aging are still largely unknown. Here, we found that the expression of SIRT7 was significantly increased in senescent fibroblasts and aged tissues. Knockdown or overexpression of SIRT7 can inhibit or promote fibroblast senescence. Knockdown of SIRT7 led to increased pan-lysine crotonylation (Kcr) levels in senescent fibroblasts. Using modern mass spectrometry (MS) technology, we identified 5,149 Kcr sites across 1,541 proteins in senescent fibroblasts, and providing the largest crotonylome dataset to date in senescent cells. Specifically, among the identified proteins, we found SIRT7 decrotonylated PHF5A, an alternative splicing (AS) factor, at K25. Decrotonylation of PHF5A K25 contributed to decreased CDK2 expression by retained intron (RI)-induced abnormal AS, thereby accelerating fibroblast senescence, and supporting a key role of PHF5A K25 decrotonylation in aging. Collectively, our data revealed the molecular mechanism of SIRT7-induced k25 decrotonylation of PHF5A regulating aging and provide new ideas and molecular targets for drug intervention in cellular aging and the treatment of aging-related diseases, and indicating that protein crotonylation has important implications in the regulation of aging progress.

In view of problems of wasting too much fuel, serious pollution of coal-fired reheating furnace caused by difficult temperature control, a heating system model of coal-fired reheating furnace with multi-temperature zones was established. A feedforward decoupling control method based on PID control algorithm was proposed. The method uses feedforward decoupling control to realize decoupling for coupling parts of each heating channel, which can reduce the time required for the heating system to reach steady state, and uses PID control algorithm to optimize control parameters of the heating system, which can make temperature of the heating system rapidly rise and remain stability in a short time. The simulation and experimental results show that the method can greatly improve temperature control speed, and can achieve stable control accuracy in a shorter response time, which avoids fuel wasting and improves heating rate.

Abstract Research on lithium compounds has attracted much attention nowadays. However, to elucidate the precise structure of lithium compounds is a challenge, especially when considering the small ions that may be transferred between the interstitial voids. Here, the discovery of reduced local symmetry (symmetry breaking) in small domains of Li2SrSiO4 is reported by employing Eu3+ as a spectroscopic probe, for which X‐ray, neutron, and electron diffraction have confirmed the average long‐range structure with the space group P3121. However, luminescence shows a lower local symmetry, as confirmed by the extended X‐ray absorption fine structure. By considering the reduced symmetry of the local structure, this work opens the door to a new class of understanding of the properties of materials.

In this paper, a production simulation method of regional integrated energy system considering the randomness of the scene is proposed to solve the production decision problem of integrated energy system including wind power, photovoltaic and other intermittent renewable energy sources. This method firstly establishes the typical scenario set of the integrated energy system operation with the corresponding random probability through cluster analysis. Secondly, taking the regional integrated energy system as the object, a production decision model and algorithm with the goal of minimizing the expected energy consumption cost are proposed. Finally, the effectiveness of the model algorithm is verified by a case study.

Conventional software frame synchronization methods are inefficient in processing huge continuous data without synchronization words. To improve the processing speed, a real-time synchronization algorithm is proposed based on reverse searching. Satellite data are grouped and searched in the reverse direction to avoid searching for synchronization words in huge continuous invalid data; thus, the frame synchronization speed is improved enormously. The fastest processing speed is up to 15445.9 Mbps when HJ-1C data are tested. This method is presently applied to the HJ-1C quick-look system in remote sensing satellite ground stations.

With a large amount of intermittent distributed generation(DG) integrating into active distribution network(ADN), the flexibility requirements during the operation process should be paid more attention. Therefore, a dynamic optimal operation model is built. In this model, the maximization of economic benefits during the production cycle is set as the optimization objective, and the constrains of security and flexibility are considered. Then based on this model, the security, economy and flexibility requirements of power system are satisfied by coordination and interaction of different flexible sources. Finally, a case study is given to illustrate the validity and effectiveness of the models.

Synthetic Aperture Radar (SAR) image processing requires huge computation amount. Traditionally, this task runs on the workstation or server based on Central Processing Unit (CPU) and is rather time-consuming, hence real-time processing of SAR data is impossible. Based on Compute Unified Device Architecture (CUDA) technology, a new plan of SAR imaging algorithm operated on NVIDIA Graphic Processing Unit (GPU) is proposed. The new proposal makes it possible that the data processing procedure and CPU/GPU data exchanging execute concurrently, especially when SAR data size exceeds total GPU global memory size. Multi-GPU is suitably supported by the new proposal and all of computational resources are fully exploited. It is shown by experiment on NVIDIA K20C and INTEL E5645 that the proposed solution accelerates SAR data processing by tens of times. Consequently, the GPU based SAR processing system with the proposed solution embedded is much more power saving and portable, which makes it qualified to be a real-time SAR data processing system. Experiment shows that SAR data of 36 Mega points can be processed in real-time per second by K20C with the new solution equipped.
