Human immunodeficiency virus-1 (HIV-1) infects the central nervous system (CNS) and causes HIV-associated neurocognitive disorders (HAND) in about half of the population living with the virus despite combination anti-retroviral therapy (cART). HIV-1 activates the innate immune system, including the production of type 1 interferons (IFNs) α and β. Transgenic mice expressing HIV-1 envelope glycoprotein gp120 (HIVgp120tg) in the CNS develop memory impairment and share key neuropathological features and differential CNS gene expression with HIV patients, including the induction of IFN-stimulated genes (ISG). Here we show that knocking out IFNβ (IFNβKO) in HIVgp120tg and non-tg control mice impairs recognition and spatial memory, but does not affect anxiety-like behavior, locomotion, or vision. The neuropathology of HIVgp120tg mice is only moderately affected by the KO of IFNβ but in a sex-dependent fashion. Notably, in cerebral cortex of IFNβKO animals presynaptic terminals are reduced in males while neuronal dendrites are reduced in females. The IFNβKO results in the hippocampal CA1 region of both male and female HIVgp120tg mice in an ameliorated loss of neuronal presynaptic terminals but no protection of neuronal dendrites. Only female IFNβ-deficient HIVgp120tg mice display diminished microglial activation in cortex and hippocampus and increased astrocytosis in hippocampus compared to their IFNβ-expressing counterparts. RNA expression for some immune genes and ISGs is also affected in a sex-dependent way. The IFNβKO abrogates or diminishes the induction of MX1, DDX58, IRF7 and IRF9 in HIVgp120tg brains of both sexes. Expression analysis of neurotransmission related genes reveals an influence of IFNβ on multiple components with more pronounced changes in IFNβKO females. In contrast, the effects of IFNβKO on MAPK activities are independent of sex with pronounced reduction of active ERK1/2 but also of active p38 in the HIVgp120tg brain. In summary, our findings show that the absence of IFNβ impairs memory dependent behavior and modulates neuropathology in HIVgp120tg brains, indicating that its absence may facilitate development of HAND. Moreover, our data suggests that endogenous IFNβ plays a vital role in maintaining neuronal homeostasis and memory function.

Adolescent idiopathic scoliosis (AIS), a sideways curvature of the spine, is sexually dimorphic, with increased incidence in females. A genome-wide association study identified a female-specific AIS susceptibility locus near the PAX1 gene. Here, we use mouse enhancer assays, three mouse enhancer knockouts, and subsequent phenotypic analyses to characterize this region. Using mouse enhancer assays, we characterize a sequence, PEC7, which overlaps the AIS-associated variant, and find it to be active in the tail tip and intervertebral disc. Removal of PEC7 or Xe1, a known sclerotome enhancer nearby, or deletion of both sequences lead to a kinky tail phenotype only in the Xe1 and combined (Xe1+PEC7) knockouts, with only the latter showing a female sex dimorphic phenotype. Extensive phenotypic characterization of these mouse lines implicates several differentially expressed genes and estrogen signaling in the sex dimorphic bias. In summary, our work functionally characterizes an AIS-associated locus and dissects the mechanism for its sexual dimorphism.

A putative male advantage in wayfinding ability is the most widely documented sex difference in human cognition and has also been observed in other animals. The common interpretation, the sex-specific adaptation hypothesis, posits that this male advantage evolved as an adaptive response to sex differences in home range size. A previous study a decade ago tested this hypothesis by comparing sex differences in home range size and spatial ability among 11 species and found no relationship. However, the study was limited by the small sample size, the lack of species with a larger female home range and the lack of non-Western human data. The present study represents an update that addresses all of these limitations, including data from 10 more species and from human subsistence cultures. Consistent with the previous result, we found little evidence that sex differences in spatial navigation and home range size are related. We conclude that sex differences in spatial ability are more likely due to experiential factors and/or unselected biological side effects, rather than functional outcomes of natural selection.

Metabolic dysfunction–associated steatotic liver disease (MASLD, previously termed NAFLD, nonalcoholic fatty liver disease) is a complex multifactorial disease showing generally higher prevalence and severity in men than in women. With respect to women, men are also more prone to develop metabolic dysfunction–associated steatohepatitis, fibrosis and liver‐related complications. Several genetic, hormonal, environmental and lifestyle factors may contribute to sex differences in MASLD development, progression and outcomes. However, after menopause, the sex‐specific prevalence of MASLD shows an opposite trend between men and women, pointing to the relevance of oestrogen signalling in the sexual dimorphism of MASLD. The patatin‐like phospholipase domain‐containing protein 3 (PNPLA3) gene, that encodes a triacylglycerol lipase that plays a crucial role in lipid metabolism, has emerged as a key player in the pathogenesis of MASLD, with the I148M variant being strongly associated with increased liver fat content and disease severity. Recent advances indicate that carrying the PNPLA3 I148M variant can be a risk factor for MASLD especially for women. To elucidate the molecular mechanisms underlying the sex‐specific role of PNPLA3 I148M in the development of MASLD, several in vitro, ex vivo and in vivo models have been developed. Background: Blotched snakehead (Channa maculata) displays significant sexual dimorphism, with males exhibiting faster growth rates and larger body sizes compared to females. The cultivation of the all-male population of snakeheads holds substantial economic and ecological value. Nonetheless, the intricate processes governing the development of bipotential gonads into either testis or ovary in C. maculata remain inadequately elucidated. Therefore, it is necessary to determine the critical time window of sex differentiation in C. maculata, providing a theoretical basis for sex control in production practices. Methods: The body length and weight of male and female C. maculata were measured at different developmental stages to reveal when sexual dimorphism in growth initially appears. Histological observations and spatiotemporal comparative transcriptome analyses were performed on ovaries and testes across various developmental stages to determine the crucial time windows for sex differentiation in each sex and the sex-related genes. Additionally, qPCR and MG2C were utilized to validate and locate sex-related genes, and levels of E2 and T were quantified to understand sex steroid synthesis. Results: Sexual dimorphism in growth became evident starting from 90 dpf. Histological observations revealed that morphological sex differentiation in females and males occurred between 20 and 25 dpf or earlier and 30–35 dpf or earlier, respectively, corresponding to the appearance of the ovarian cavity or efferent duct anlage. Transcriptome analyses revealed divergent gene expression patterns in testes and ovaries after 30 dpf. The periods of 40–60 dpf and 60–90 dpf marked the initiation of molecular sex differentiation in females and males, respectively. Male-biased genes (Sox11a, Dmrt1, Amh, Amhr2, Gsdf, Ar, Cyp17a2) likely play crucial roles in male sex differentiation and spermatogenesis, while female-biased genes (Foxl2, Cyp19a1a, Bmp15, Figla, Er) could be pivotal in ovarian differentiation and development. Numerous biological pathways linked to sex differentiation and gametogenesis were also identified. Additionally, E2 and T exhibited sexual dimorphism during sex differentiation and gonadal development. Based on these results, it is hypothesized that in C. maculata, the potential male sex differentiation pathway, Sox11a–Dmrt1–Sox9b, activates downstream sex-related genes (Amh, Amhr2, Gsdf, Ar, Cyp17a2) for testicular development, while the antagonistic pathway, Foxl2/Cyp19a1a, activates downstream sex-related genes (Bmp15, Figla, Er) for ovarian development. Facultative parthenogenesis is a form of reproduction in which females can either lay unfertilised eggs that typically develop into female offspring only or mate and lay fertilised eggs that develop into male and female offspring. Intriguingly, facultative parthenogens often occur in mixed‐sex populations where reproduction is mostly sexual and all‐female populations where reproduction is asexual. How all‐female populations avoid invasion by males remains unknown. Here, we explored the use of pheromones in male–female communication in a facultatively parthenogenetic stick insect, the peppermint stick insect (Megacrania batesii), and compared chemical signals between females descended from sexually versus parthenogenetically reproducing populations. If parthenogenetic females release less attractive pheromones, this could help explain the persistence of all‐female populations. We found that M. batesii exhibits slight sexual dimorphism in antenna morphology, and behavioural assays provided little evidence that males could locate females solely by volatile pheromones. However, CHC profiles differed substantially between different types of females. Analysis of CHC components indicated a clear genetic difference between females descended from all‐female versus mixed‐sex populations, as well as a maternal effect of female parthenogenetic versus sexual development. Together, our results suggest that males might rely more on close‐range chemical cues to differentiate females, and chemical communication could play a role in the persistence of all‐female populations. Objective Animals Studied Procedures Results Conclusions To establish baseline ophthalmic parameters for an endangered, semi‐wild population of healthy whooping cranes (Grus americana) (WHCR) and Mississippi sandhill cranes (Grus canadensis pulla) (SACR).Eighteen WHCR and 16 SACR.Ophthalmic examination was performed by a single observer, followed by conjunctival swab collection for aerobic bacterial culture and measurement of tear production (phenol red thread test, PRTT) and corneal diameter (CD) as tolerated. Measurement of the axial globe (AG) length, anterior chamber (AC) depth, lens thickness, vitreous chamber (VC) depth, and pecten length was performed via ocular ultrasound (OUS) as tolerated.Eyelid cicatrization (n = 1 WHCR), keratitis (n = 2 WHCR), incipient cataracts (n = 1 WHCR, n = 4 SACR), and uveal cysts (n = 1 SACR) were identified. Twenty‐one bacterial species were cultured from SACR, while 18 bacterial species were cultured from WHCR. SACR under 6 months old had increased PRTT values compared to older SACR (p = .0432). AG length and VC depth of male WHCR were greater than in female WHCR (p = .0045 and p = .0008, respectively). WHCR less than 6 months old had greater AC depth and lens thickness than WHCR over 6 months (p < .001 and p = .0013, respectively). SACR less than 6 months old had greater AC depth and lens thickness than WHCR over 6 months (p < .0001 and p < .0001, respectively).WHCR and SACR are amenable to complete ophthalmic examination. Research on the relationship between macrophages and neuropathic pain has flourished in the past two decades. It has long been believed that macrophages are strong immune effector cells that play well-established roles in tissue homeostasis and lesions, such as promoting the initiation and progression of tissue injury and improving wound healing and tissue remodeling in a variety of pathogenesis-related diseases. They are also heterogeneous and versatile cells that can switch phenotypically/functionally in response to the micro-environment signals. Apart from microglia (resident macrophages of both the spinal cord and brain), which are required for the neuropathic pain processing of the CNS, neuropathic pain signals in PNS are influenced by the interaction of tissue-resident macrophages and BM infiltrating macrophages with primary afferent neurons. And the current review looks at new evidence that suggests sexual dimorphism in neuropathic pain are caused by variations in the immune system, notably macrophages, rather than the neurological system. PLAIN LANGUAGE SUMMARY: Neuropathic pain is defined by the International Association for the Study of Pain as pain triggered or caused by primary damage to or dysfunction of the nervous system. Following intensive research into the mechanisms of neuropathic pain, macrophages have been revealed to play an important role in pathologic pain following nerve injury. Macrophages dynamically monitor the microenvironment to maintain tissue homeostasis. Once a macrophage is exposed to a pathologic stimulus, it in turn alters its functional phenotype and interacts with nociceptors, leading to neuropathic pain. Sexual dimorphism in vocal signals is found across many bird species. By evaluating correlations among sex, behaviors, and vocalizations, the meaning and utility of sound production may be inferred. Anna's hummingbirds Calypte anna exhibit pronounced sexual dimorphism, with males being larger and having more vibrant coloration than females, but vocal dimorphism in the species is less studied. A common vocalization of Anna's hummingbirds is the chip note, which is produced by both sexes in a wide array of contexts. Here, we correlated temporal parameters of recorded chip notes with individual sex and behavioral contexts gathered from field observations. The production rate of chip notes differed significantly between male and female hummingbirds but did not vary much with behavioral context. Although Anna's hummingbirds produce chips across a broad spectrum of behaviors, dimorphic chip production may be especially important in territorial behavior. [ABSTRACT FROM AUTHOR]

Brain iron increases in several neurodegenerative diseases are associated with disease progression. However, the causes of increased brain iron remain unclear. This study investigates relationships between subcortical iron, systemic iron and inflammatory status. Brain magnetic resonance imaging (MRI) scans and blood plasma samples were collected from cognitively healthy females (n = 176, mean age = 61.4 ± 4.5 years, age range = 28–72 years) and males (n = 152, mean age = 62.0 ± 5.1 years, age range = 32–74 years). Regional brain iron was quantified using quantitative susceptibility mapping. To assess systemic iron, haematocrit, ferritin and soluble transferrin receptor were measured, and total body iron index was calculated. To assess systemic inflammation, C‐reactive protein (CRP), neutrophil:lymphocyte ratio (NLR), macrophage colony‐stimulating factor 1 (MCSF), interleukin 6 (IL6) and interleukin 1β (IL1β) were measured. We demonstrated that iron levels in the right hippocampus were higher in males compared with females, while iron in the right caudate was higher in females compared with males. There were no significant associations observed between subcortical iron levels and blood markers of iron and inflammatory status indicating that such blood measures are not markers of brain iron. These results suggest that brain iron may be regulated independently of blood iron and so directly targeting global iron change in the treatment of neurodegenerative disease may have differential impacts on blood and brain iron. The strength and direction of sexual selection can vary among populations. However, spatial variability is rarely explored at the level of the social group. Here we investigate sexual selection and sex roles in the paternally mouthbrooding, socially monogamous, and site-attached pajama cardinalfish, Sphaeramia nematoptera. Females were larger and more aggressive and had a longer dorsal fin filament, indicating reversed sex roles. At the scale of social groups, we show that the Bateman gradient and reproductive variance depend on the sex ratio and size of groups. In small and medium-sized groups with balanced or male-biased sex ratios, Bateman gradients were steeper for females, whereas gradients were equally steep for both sexes in large groups or when the sex ratio was female biased. For both sexes, reproductive variance increased with group size and with a higher male-to-female sex ratio. In S. nematoptera , mating opportunities outside the socially monogamous pair appear to impact sexual selection. We conclude that strength and direction of sexual selection can be masked by social dynamics in group-living species when considering only population and large-scale demographic processes. Intraspecific variation in camouflage is common in animals. Sexual dimorphism in camouflage is less common and, where observed, attributed to trade-offs between natural selection for predator avoidance and sexual selection for conspicuous mating signals. Here we report on variation in putatively cryptic ventral hindwing patterns in the American snout butterfly, Libytheana carinenta. We use field surveys and crowdsourced data to characterize three morphs. One is found in both sexes, one is male specific, and one is female specific. The sex-specific morphs constitute a sexually dimorphic set whose frequencies change together in time. Field surveys indicate that butterflies in southern Arizona transition from midsummer dominance of the sexually monomorphic pattern to early-fall dominance of the sexually dimorphic set. Crowdsourced data indicate that the sexually dimorphic set dominates in early spring, transitioning later into a mixture of morphs dominated by the monomorphic pattern, with the dimorphic set rising in frequency again in late fall. Aim: Wallacea is a global biodiversity hotspot and Sulawesi is the largest island in this region, notable for a high proportion of endemic species. The Wakatobi archipelago, off the southeastern peninsular arm of Sulawesi, is home to several endemic bird species. Although islands are known to influence the morphology of their resident species, competitive interactions also exert strong influences on morphology. Here, we consider the contributions of both islands and competitors on two morphological traits of two bird species in a small passerine guild found on the Wakatobi islands. Location: Wakatobi archipelago, Sulawesi Tenggara, Indonesia. Taxon: Wakatobi White‐eye (Zosterops flavissimus) and Wakatobi Sunbird (Cinnyris infrenatus). Methods: Working with morphological measures (from netting studies), population density metrics (from transect surveys) and fundamental data from islands (area, distance from mainland and elevation), we investigated (by means of multiple linear regression) which terms best explained variation in the body size and bill size of two small passerine birds endemic to the Wakatobi islands. Results: Both island metrics and competitor presence/density were useful in describing the variation in body size of both bird species. However, only competitor presence/density was useful in describing variation in bill size of the birds. The best models describing variation in these traits included terms representing both interspecific and intraspecific competition. Main Conclusions: Our findings indicate that simple island metrics may be useful in predicting some of the variation in some functional traits of island species. However, in this example, simple island metrics were insufficient to explain the full variation in body size of endemic birds across the Wakatobi archipelago and were of no use in predicting the variation in their bill size. As bill morphology frequently dictates diet and feeding niche, it is a trait which may determine population divergence and speciation. Cardiovascular diseases (CVDs) remain the leading cause of morbidity and mortality, affecting people of all races, ages, and sexes. Substantial sex dimorphism exists in the prevalence, manifestation, and outcomes of CVDs. Understanding the role of sex hormones as well as sex-hormone-independent epigenetic mechanisms could play a crucial role in developing effective and sex-specific cardiovascular therapeutics. Existing research highlights significant disparities in sex hormones, epigenetic regulators, and gene expression related to cardiac health, emphasizing the need for a nuanced understanding of these variations between men and women. Despite these differences, current treatment approaches for CVDs often lack sex-specific considerations. A pivotal shift toward personalized medicine, informed by comprehensive insights into sex-specific DNA methylation, histone modifications, and non-coding RNA dynamics, holds the potential to revolutionize CVD management. By understanding sex-specific epigenetic complexities, independent of sex hormone influence, future cardiovascular research can be tailored to achieve effective diagnostic and therapeutic interventions for both men and women. Objectives: This study aims to conduct a thorough characterization of hand morphology. Employing a 2D geometric morphometric approach, we scrutinize individual fingers and the palm, delineating the ontogenetic trajectories for each biological sex and investigating the alterations that take place at various stages of human development. Materials and methods: A set of thirty‐two 2D anatomical landmarks were assessed in a sex‐balanced sample of human hands (F = 275, M = 250 males), spanning all stages of human development. Following Procrustes registration, the data on size and shape for individual fingers and the palm were examined for each biological sex and age group. Regression analysis was utilized to quantify ontogenetic trajectories for each biological sex. Results: The findings suggest a gradual escalation in sexual dimorphism throughout human development, with statistically noteworthy distinctions becoming apparent in size starting at the age of 3, and in shape from the age of 7 onwards. Additionally, our analyses uncover a distinctive sigmoid pattern between sexes, indicating that biological male hands exhibit a sturdier build compared to biological female hands from early childhood onward. Conclusions: In conclusion, this study enriches our insights into sexual dimorphism in human hands, stressing the importance of considering both size and shape across different ontogenetic stages. Sexual dimorphism and male combat are observed in many species. Often, the outcome of male combat affects the outcome of subsequent combats, mating success, number of sperm, and fitness of the male's offspring. Also, the quantity and quality of sperm may be regulated by winning or losing, depending on species ecology and mating system. However, very few studies have experimentally examined the influence of fight outcomes on male offspring fitness. We studied male combat in the giant mealworm (Zophobas atratus) in which males bite each other's hind legs. We hypothesized that subsequent fitness could differ between winners and losers in the escalated male combat of this species. We measured several fitness traits including the number of eggs laid by mated females, and the number of hatches sired by uncontested males, winners, and losers in escalated and non-escalated combat, and compared the fitness of each winner and loser to that of an uncontested male. We also measured mating duration. The numbers of eggs and the percentages of hatched eggs of losers in the escalated combat were significantly reduced compared to that of the uncontested males. This reduction may be due to injuries from escalated leg- biting fights and a result of the sperm amount of the uncontested males being greater than that of the loser males. [ABSTRACT FROM AUTHOR]

Chile had a violent military coup (1973–1990) that resulted in 3,000 victims declared detained, missing or killed; many are still missing and unidentified. Currently, the Human Rights Unit of the Forensic Medical Service in Chile applies globally recognised forensic anthropological approaches, but many of these methods have not been validated in a Chilean sample. As current research has demonstrated population-specificity with extant methods, the present study aims to validate sex estimation methods in a Chilean population and thereafter establish population-specific equations. A sample of 265 os coxae of known age and sex of adult Chileans from the Santiago Subactual Osteology Collection were analysed. Visual assessment and scoring of the pelvic traits were performed in accordance with the Phenice (1969) and Klales et al. (2012) methods. The accuracy of Phenice (1969) in the Chilean sample was 96.98%, with a sex bias of 7.68%. Klales et al. (2012) achieved 87.17% accuracy with a sex bias of -15.39%. Although both methods showed acceptable classification accuracy, the associated sex bias values are unacceptable in forensic practice. Therefore, six univariate and eight multivariate predictive models were formulated for the Chilean population. The most accurate univariate model was the ventral arc at 96.6%, with a sex bias of 5.2%. Classification accuracy using all traits was 97.0%, with a sex bias of 7.7%. This study provides Chilean practitioners a population-specific morphoscopic standard with associated classification probabilities acceptable to accomplish legal admissibility requirements in human rights and criminal cases specific to the second half of the 20th century. Anhedonia, a transdiagnostic symptom present in many neuropsychiatric disorders, differs in males and females. Parietal EEG alpha asymmetry is associated with reduced arousal and low positive emotionality, and is, therefore, a promising neurophysiologic biomarker of anhedonia. To date, however, no prior studies have determined whether this measure captures sex differences in anhedonic expression. This preliminary study (N = 36) investigated whether anhedonia severity is associated with EEG resting-state parietal alpha asymmetry in adults and whether sex moderates this relationship. Results showed that there was a significant moderating effect of sex such that, only for females, higher levels of anhedonia were associated with increased parietal alpha asymmetry. These findings suggest that parietal alpha asymmetry is a promising biomarker of anhedonia severity in female adults and reinforces the need to account for sex differences in future research. An important, but generally overlooked source in intraspecific variability of characters used in tardigrades taxonomy is sexual dimorphism. Dioecious tardigrades species with unisexual or bisexual populations are common, but external sexual dimorphism is rarely observed. The discovery and integrative analysis of a Diploechiniscus species population with high sexual dimorphism from Calvert Island (Canada) has allowed its description as a new species. Diploechiniscus dimorphus n. sp. is characterized by a different chaetotaxy in males and females, and by enlarged cephalic sensory appendages in the males. The discovery of this new species shows that it is important to consider sexual dimorphism as a source of intraspecific variability in tardigrade taxonomy due to its implications on the use of traits commonly used to delineate species such as chaetotaxy. Lastly, the finding of individuals of Diploechiniscus horningi (Schuster and Grigarick, 1971), a species previously synonymized with Diploechiniscus oihonnae (Richters, 1903), allows us to prove that it is genetically a separate species and to reinstate it. Preferences for sexual dimorphism in male and female faces can vary based on the viewers' gender, sexual orientation, and intensity of sexual desire. Asexual individuals, who experience little to no sexual attraction, may display different preferences for sexual dimorphism than has been observed in allosexual samples. Researchers have hypothesised that gender conformity may likewise impact preferences for sexual dimorphism. Participants (N = 182) recruited from an online asexual community provided demographic information and responded to measures of romantic attraction, sexual attraction, and conformity with male and female gender norms before they indicated their preference for male and female faces that were digitally altered to be more or less sexually dimorphic. Regardless of gender, asexual participants displayed a high preference for femininity in male and female faces. Preferences for sexual dimorphism displayed complex, and inconsistent associations with reported romantic and sexual attraction, and with conformity to feminine gender norms. This is the first report of preferences for facial sexual dimorphism and assessment of the role of gender conformity in an asexual population. Heterochely denotes the presence of dissimilarly sized chelipeds on opposite sides of the body, a prevalent occurrence in diverse crustaceans. Conversely, heterometry pertains to the quantifiable disparities in size between these chelipeds. Both chelipeds hold pivotal roles in activities such as foraging, mating, and defense. Consequently, individuals of both genders in heterochelic species exhibit this morphological pattern. Previous studies have identified sexual dimorphism in cheliped size, with males displaying larger major chelipeds compared to females, albeit solely relying on propodus length as a size proxy and focusing solely on the major cheliped. In our study, we meticulously examined 190 specimens of the common European hermit crab Pagurus bernhardus from two collections. We sought to elucidate allometric relationships and assess whether heterometry exhibited sex‐based differences when adjusting for body size by using ratios. Our findings revealed that male chelipeds displayed hyperallometric growth relative to females, and all three calculated heterometry indices exhibited significant disparities between the sexes. Consequently, male specimens exhibited larger major and minor chelipeds, even when theoretically matched for body size with females. This phenomenon may be attributed, among other factors, to male–male contests. Blister beetles (Coleoptera: Meloidae) produce a natural defensive toxin cantharidin (CTD), which has been used for various cancer treatments and other diseases. Currently, the lack of chromosome‐level reference genomes in Meloidae limits further understanding of the mechanism of CTD biosynthesis and environmental adaptation. In this study, the chromosome‐level genome assembly of Mylabris phalerata was generated based on PacBio and Hi‐C sequencing. This reference genome was about 136.68 Mb in size with contig N50 of 9.17 Mb and composed of 12 chromosomes. In comparison to six other Coleoptera insects, M. phalerata exhibited multiple expanded gene families enriched in juvenile hormone (JH) biosynthetic process pathway, farnesol dehydrogenase activity, and cytochrome P450, which may be related to CTD biosynthesis. Consistently, the transcriptomic analysis suggested the "terpenoid backbone biosynthesis" pathway and "the juvenile hormone" as putative core pathways of CTD biosynthesis and presented eight up‐regulated differential expression genes in male adults as candidate genes. It is possible that the restricted feeding niche and lifestyle of M. phalerata were the cause of the gene family's contraction of odorant binding proteins. The ABC transporters (ABCs) related to exporting bound toxins out of the cell and the resistance to the self‐secreted toxins (e.g. CTD) were also contracted, possibly due to other self‐protection strategies in M. phalerata. A wide variety of strategies have emerged in different animal species for attracting mates. These strategies, such as chemical signals and calling songs, fuel competition for mates and can lead to the evolution of distinct courtship behaviours within a species. These behaviours often involve males and females taking on different roles in attracting mates. For example, limitations in signal production or perception can make one of the sexes sedentary. Regarding Poecilimon bushcrickets specifically, in 2012 McCartney et al. proposed a theory to explain sex role differentiation within the mate-searching system in certain species. According to this model, sex roles in mate-searching systems propose that in species where females search for males, females should receive a larger spermatophore gift. This study tests this theory for two Poecilimon species with differing strategies in terms of females' response to male calling songs regarding spermatophore investment (P. sureyanus-female searching and P. inflatus-male searching). Given that reproductive effort and physiological activities are both costly for the organism, there is a trade-off due to limited energy resources. To investigate the specific trade-off in allocating resources between reproductive functions and immune defence in these species, the presents study compared two immune parameters (phenoloxidase and lytic activities). In nature, immune activities are much more plastic than the differentiation of reproductive traits between the sexes. The results showed that spermatophore investment may critically depend on differences in mate-searching strategies, particularly in terms of females' response to male calling songs within the species. Renewing prescriptions is important for the continuity of pharmacotherapy. However, renewing without the prescriber meeting the patient might lead to insufficient pharmacotherapy monitoring. This study investigated the prevalence of renewed prescriptions, prescriptions renewed without the prescriber meeting the patient and the factors associated with renewals made without meeting the patient. This register‐based study employed data on electronic prescriptions and health care contacts from Finnish registers. Prescriptions were classified as renewed if there was a renewal request or a previous prescription for the same ATC code. Prescriptions were considered as being renewed without meeting the patient if there was no patient contact on the renewal date. Descriptive and logistic generalized estimating equation analyses were conducted. The random sample of prescriptions (10%) from the year 2019 amounted to 2 804 048. Of these, 41.9% were original, 35.4% were renewals without meeting the patient and 22.7% were renewed with meeting the patient. Characteristics such as male sex, age 35–54 years, prescription for cardiovascular system preparations and the prescription being renewed during the summer (June–August) were associated with renewals made without meeting the patient. Further research is needed on the implementation of pharmacotherapy monitoring in the case of renewals without the prescriber meeting the patient. PLAIN LANGUAGE SUMMARY: This study analyses the frequency and conditions under which prescriptions are renewed without direct patient–prescriber interactions in Finland. Using a random sample of over 2.8 million prescriptions from 2019, it found that while 41.9% were initial issues, 35.4% were renewed without a meeting, and 22.7% with one. Factors like being male, aged 35–54, prescribed cardiovascular medications or renewals during summer were linked to renewals without patient contact. Metabolic processes are intricately linked to the resolution of innate inflammation and tissue repair, two critical steps for treating post-traumatic osteoarthritis (PTOA). Based on lipolytic and immunoregulatory actions of norepinephrine, we hypothesized that intra-articular β-adrenergic receptor (βAR) stimulation would suppress PTOA-associated inflammation in the infrapatellar fat pad (IFP) and synovium. We used the βAR agonist isoproterenol to perturb intra-articular metabolism 3.5 weeks after applying a non-invasive single-load compression injury to knees of 12-week-old male and female mice. We examined the acute effects of intra-articular isoproterenol treatment relative to saline on IFP histology, multiplex gene expression of synovium-IFP tissue, synovial fluid metabolomics, and mechanical allodynia. Injured knees developed PTOA pathology characterized by heterotopic ossification, articular cartilage loss, and IFP atrophy and fibrosis. Isoproterenol suppressed the upregulation of pro-fibrotic genes and downregulated the expression of adipose genes and pro-inflammatory genes (Adam17 , Cd14 , Icam1 , Csf1r , and Casp1) in injured joints of female (but not male) mice. Analysis of published single-cell RNA-seq data identified elevated catecholamine-associated gene expression in resident-like synovial-IFP macrophages after injury. Injury substantially altered synovial fluid metabolites by increasing amino acids, peptides, sphingolipids, phospholipids, bile acids, and dicarboxylic acids, but these changes were not appreciably altered by isoproterenol. Intra-articular injection of either isoproterenol or saline increased mechanical allodynia in female mice, whereas neither substance affected male mice. Acute βAR activation altered synovial-IFP transcription in a sex and injury-dependent manner, suggesting that women with PTOA may be more sensitive than men to treatments targeting sympathetic neural signaling pathways. Of twenty species of beetles across six families and fourteen genera found living within the hives of the western honey bee Apis mellifera, only the small hive beetle (SHB), Aethina tumida, displays attributes characteristic of an integrated parasitic lifestyle. This species is a pest that can cause severe damage to weakened hives. SHB adults tend to disperse over long distances after they eclose and appear to rely on food odors to locate beehives to infest. Thus, we hypothesized that the floral sources of nectars processed into honey stored in hives may influence SHB orientation toward hives and dietary preferences within hives. We carried out preference assays of individual SHB in an arena with several protein patty substitute diets, each of which was prepared with one of three monofloral honeys (white tupelo Nyssa ogeche, black locust Robinia pseudoacacia, and buckwheat Fagopyrum esculentum) or a phytochemical-free control. Male and female beetles displayed different responses to food choices. Female SHB preferred monofloral honey diets (74%) over the phytochemical-free control, and, among the three monofloral honeys, exhibited a strong preference for the white tupelo honey diet over control diets. In contrast, 41% of male SHB in individual assays preferred the control diet over diets containing monofloral honeys. These findings suggest that for males the ability to detect sugar sources is the primary determinant of preference, whereas, based presumably on nutritional requirements in support of oviposition, female SHB may be responsive to a broader range of food-related cues to increase the likelihood of identifying well-provisioned hives. Background Methods Results Conclusions High‐risk medication use is associated with an increased risk of adverse events, but little is known about its chronic utilization by key demographic groups. We aimed to study the associations between age, sex, and race/ethnicity with new chronic use of high‐risk medications in older adults.In this retrospective cohort study, we analyzed data from older adults aged ≥65 years enrolled in a national health insurer who started a high‐risk medication between 2017 and 2022 across 16 high‐risk medication classes. We used generalized estimating equations to estimate the associations between sociodemographic classifications and the onset of chronic high‐risk medication use after initiation (≥90 days' supply across ≥2 fills within 180 days). We adjusted the analyses for sociodemographic and clinical patient characteristics and added three‐way interaction terms for race/ethnicity, sex, and age to explore whether the outcome varied across different subgroups of race/ethnicity, age, and sex.Across 2,751,069 patients (mean age: 74 years [SD = 7], 72% White, 60% Female), 406,075 (15%) became new chronic users of ≥1 high‐risk medication. Compared to White older adults, Asian (RR = 0.81, 95% CI: 0.79–0.84), Black (RR = 0.92, 95% CI: 0.90–0.94), and Hispanic (RR = 0.85, 95% CI: 0.83–0.86) older adults had a lower risk of becoming new chronic users. Men had a higher risk compared to women (RR = 1.09, 95% CI: 1.08–1.10). Age was not significantly associated with new chronic high‐risk medication use (≥75 years: RR = 1.00, 95% CI: 1.00–1.01). We observed differences across some medication classes, like benzodiazepines, first‐generation antihistamines, and antimuscarinics for which non‐White older adults were at a higher risk. The joint presence of specific age, sex, and race/ethnicity characteristics decreased the risk of becoming a new chronic user (e.g., Hispanic/Female/65–74 years: RR = 0.96, 95% CI: 0.94–0.99).New chronic high‐risk medication use varied across older adults by sociodemographic characteristics, suggesting the need to individualize medication optimization approaches and better understand how systematic barriers in access to health care may influence differences in high‐risk medication use in older adults. The sex-specific differences in the growth and population characteristics of the high-commercial-value sand crab Ovalipes punctatus were investigated in Korea. The estimated allometric growth between the sexes showed significant differences in all morphometric measurements. In the classification of growth types, carapace width-chela length exhibited positive and negative allometric growth in males and females, respectively. Carapace width-abdominal width showed positive relative growth in both sexes, and orbital spine width exhibited negative relative growth in both sexes. Consequently, sexual dimorphism was evident in all measured traits. Growth parameters estimated using the ELEFAN function of the FiSAT II program indicated higher values in males compared to females. Asymptotic length (CW∞) for males was estimated at 139.2 mm, whereas for females it was 116.6 mm. Additionally, the growth coefficient (K) was higher in males (0.65) than in females (0.54), suggesting faster growth in males. The winter point (WP) was 1 for males and 0.7 for females, indicating slower growth in males during the colder December and slower growth in females during the spawning period in August. The modified von Bertalanffy growth curves indicated asymptotic growth in all sexes, and the growth performance index (φ') showed higher values in males (4.10) compared to females (3.87), reflecting differences in growth curves. The steady increase in recruitment rates from July to September was associated with the appearance of larvae and their subsequent growth into juveniles, leading to their recruitment into the population during this period. Introduction: Thyroid function during pregnancy fluctuates with gestational weeks, seasons and other factors. However, it is currently unknown whether there is a fetal sex-specific thyroid function in pregnant women. The purpose of this study was to investigate the fetal sex differences of maternal thyroid-stimulating hormone (TSH) and free thyroxine (FT4) in pregnant women. Methods: This single-center retrospective real-world study was performed by reviewing the medical records of pregnant women who received regular antenatal health care and delivered liveborn infants in Shanghai First Maternity and Infant Hospital (Pudong branch), from Aug. 18, 2013 to Jul. 18, 2020. Quantile regression was used to evaluate the relationship between various variables and TSH and FT4 concentrations. The quantile regression also evaluated the sex impact of different gestational weeks on the median of TSH and FT4. Results: A total of 69,243 pregnant women with a mean age of 30.36 years were included. 36197 (52.28%) deliveries were boys. In the three different trimesters, the median levels (interquartile range) of TSH were 1.18 (0.66, 1.82) mIU/L and 1.39 (0.85, 2.05) mIU/L, 1.70 (1.19, 2.40) mIU/L; and the median levels (interquartile range) of FT4 were 16.63 (15.16, 18.31) pmol/L, 14.09 (12.30, 16.20) pmol/L and 13.40 (11.52, 14.71) pmol/L, respectively. The maternal TSH upper limit of reference ranges was decreased more in mothers with female fetuses during gestational weeks 7 to 12, while their FT4 upper limit of the reference ranges was increased more than those with male fetuses. After model adjustment, the median TSH level was 0.11 mIU/L lower (P <0.001), and FT4 level was 0.14 pmol/L higher (P <0.001) for mothers with female fetuses than those with male fetuses during gestational weeks 9 to 12. Discussion: We identified sexual dimorphism in maternal thyroid function parameters, especially during 9-12 weeks of pregnancy. Based on previous research, we speculated that it may be related to the higher HCG levels of mothers who were pregnant with girls during this period. Twisted-wing insects (Strepsiptera) are an enigmatic order of parasites with unusual life histories and striking sexual dimorphism. Males emerge from hosts as free-living winged adults, while females from most species remain as endoparasites that retain larval traits. Due to scarce genomic data and phylogenetic controversies, Strepsiptera was only recently placed as the closest living relative to beetles (Coleoptera). Here, we report the first PacBio HiFi genome assembly of the strepsipteran Xenos peckii (Xenidae). This de novo assembly size is 72.1 Mb, with a BUSCO score of 87.4%, N50 of 7.3 Mb, 23.4% GC content, and 38.41% repeat content. We identified 8 contigs that contain >75% of the assembly and reflect the haploid chromosome number reported from karyotypic data, and 3 contigs that exhibit sex chromosome coverage patterns. Additionally, the mitochondrial genome is 16,111 bp long and has 37 genes. This long-read assembly for Strepsiptera reveals a miniature genome and provides a unique tool to understand complex genome evolution associated with a parasitic lifestyle and extreme sexual dimorphism. Background: Intersex describes a diversity of individuals with variations in sex characteristics (VSC), reflecting underlying differences in reproductive anatomy, hormones, and/or genes and chromosomes. With a shift towards socially-conscious clinical practices, genetic counsellors (GCs) are increasingly needing to provide comprehensive care to individuals with VSC and their families. However, the current quality of training provided to genetic counsellors on intersex health is unclear. Methods: Qualitative interviews were conducted between Jan-Feb 2021 with 20 current and graduated students of Canadian GC training programs to assess the quality of GC education on intersex health topics. An agency-based model of VSC health as proposed by Crocetti et al. was used to guide the inductive thematic data analysis. Results: Results revealed three key themes: limited discussions on psychosocial considerations when caring for intersex individuals, enthusiasm for integrating more intersex training into the curriculum, and personal initiative in ensuring equity and justice in the care of individuals with VSC. Conclusions: These findings demonstrate existing knowledge gaps in the GC curriculum, with a need to increase the profession's overall awareness on intersex issues. Objective Methods Results Conclusion The cortical bone trajectory (CBT) technology is an effective substitute for traditional pedicle screw (PS) technology. However, there is still controversy about the CBT screw technology placement strategy. The objective of this study was to simulate cortical screw placement with the help of three‐dimensional (3D) software, to discuss the differences in screws between genders and vertebral segments, and to explore a safer and more efficient strategy for cortical screw placement.Mimics Medical software was used to construct a 3D model of the lumbar spine, and the placement of CBT screws was simulated. The volume of each vertebral body from L1 to L5, the pedicle isthmus height (IH), the pedicle isthmus width (IW), and the sagittal vertebral distance (SAVD) were measured. The transverse distance (TD) and the longitudinal distance (LD) between the ideal starting point (SP) and the clinical SP (the intersection Q of the midline of the superior articular process and the horizontal line 1 mm below the transverse process) were measured. The cephalad angle (CA), lateral angle (LA), maximum screw diameter (MSD), maximum screw length (MSL) of each trajectory of the L1 to L5 vertebral bodies, and the percentage of the screw insertion depth (PSID) into the vertebral body were measured. Data were statistically analyzed using Student's t‐test, one‐way analysis of variance (ANOVA), and Tukey's test.Vertebral anatomical parameters and CBT screw parameters differed between males and females. Female patients had lower IH, IW, SAVD, CA, LA, MSD, and MSL than males. IH was greatest in L1 (male, 17.81 mm; female, 16.12 mm) and the smallest in L5 (male, 14.11 mm; female, 13.05 mm). IW was smallest in L1 (male, 8.89 mm; female, 7.37 mm) and greatest in L5 (male, 16.59 mm; female, 15.43 mm). The MSD of males was smallest in L1 (6.05 mm) and greatest in L3 (7.06 mm); the MSD of females was smallest in L1 (5.13 mm) and greatest in L4 (6.64 mm). MSL was greatest at L3 (male, 33.63 mm; female, 32.28 mm) and smallest at L5 (male, 31.25 mm; female, 29.97 mm). CA was smallest in L1 (male, 22.80°; female, 21.92°) and greatest in L3 (male, 25.29°; female, 24.33°). LA was smallest in L1 (male 12.37°, female 11.27°) and greatest in L5 (male 13.56°, female 12.96°). Among the males, TD was smallest at L1 (−0.51 mm) and greatest at L5 (1.37 mm), while LD was greatest at L2 (3.46 mm) and smallest at L5 (2.40 mm). In females, TD was greatest at L1 (0.12 mm) and smallest at L3 (−0.51 mm), while LD was greatest at L1 (3.69 mm) and smallest at L5 (2.08 mm). Background and Objectives Materials and Methods Results Conclusion Iron deficiency (ID) poses a prevalent concern among blood donors, especially impacting young donors, premenopausal females and frequent donors. In alignment with recommendations to address ID, routine ferritin testing was implemented in a hospital‐based donor centre.Data set, encompassing 26 164 ferritin values from 16 464 blood donors over 33 months, were analysed retrospectively. Ferritin levels were assessed concerning donor characteristics such as sex, age, ethnicity and donation frequency.Ferritin testing revealed age, sex and ethnicity variations, emphasising the heightened risk of ID in young females meeting all donation criteria under 23 year of age who demonstrated the lowest mean baseline ferritin (41% [CI: 34%–48%] < 26 ng/mL; 20% [CI: 14%–25%] < 15 ng/mL). Postmenopausal females exhibited ferritin levels similar to similarly aged males. Irrespective of sex, donors showcased mean ferritin recovery within 6 months. Analysis of ferritin recovery post‐donation showed a five‐fold increase in risk (compared with first visit) of ID when donors return at a 2‐month interval. ‘Regular’ donors (≥10 visits) approach a median steady ferritin level (~30–35 ng/mL) by the sixth visit.As reliance on regular blood donors increases, donation policies must strike a balance between blood centre resources and the risks posed to both regular and at‐risk donors. Frequent blood donation led to donors attaining a mean steady state ferritin level above the threshold for ID. At‐risk groups, particularly premenopausal females, were several times more likely to experience ID after donation but demonstrated recovery rates similar to their group's baseline levels. The black soldier fly (BSF) Hermetia illucens (Diptera: Stratiomyidae) plays a significant role at the larval stage in the circular economy due to its ability to convert organic waste into valuable products for energy, food, feed, and agricultural applications. Many data are available on larval development and biomass generation, but basic research on this species is lacking and little is known about adult biology, in particular about the cues involved in sexual recognition. In the present study, using various instruments (stereomicroscope, scanning and transmission electron microscope, hyperspectral camera and spectrophotometer), wing ultrastructure of both sexes was analysed, reflectance and transmission spectra of the wings were measured and behavioural bioassays were carried out to measure male response to specific visual stimuli. The collected data showed the existence of sexual dimorphism in the wings of H. illucens due to iridescent structural colouration generated by a multilayer of melanin located in the dorsal lamina of the central part of the wing. Wing sexual dimorphism is particularly evident regarding the strong emission of blue light of female wings. Blue colour induces in males a strong motivation to mate. Scapula shape is highly variable across humans and appears to be sexually dimorphic—differing significantly between biological males and females. However, previous investigations of sexual dimorphism in scapula shape have not considered the effects of allometry (the relationship between size and shape). Disentangling allometry from sexual dimorphism is necessary because apparent sex‐based differences in shape could be due to inherent differences in body size. This study aimed to investigate sexual dimorphism in scapula shape and examine the role of allometry in sex‐based variation. We used three‐dimensional geometric morphometrics with Procrustes ANOVA to quantify scapula shape variation associated with sex and size in 125 scapulae. Scapula shape significantly differed between males and females, and males tended to have larger scapulae than females for the same body height. We found that males and females exhibited distinct allometric relationships, and sexually dimorphic shape changes did not align with male‐ or female‐specific allometry. A secondary test revealed that sexual dimorphism in scapula shape persisted between males and females of similar body heights. Overall, our findings indicate that there are sex‐based differences in scapula shape that cannot be attributed to size‐shape relationships. Our results shed light on the potential role of sexual selection in human shoulder evolution, present new hypotheses for biomechanical differences in shoulder function between sexes, and identify relevant traits for improving sex classification accuracy in forensic analyses. Our study investigated the size at the onset of morphological sexual maturity, sexual size dimorphism, and relative growth in the green porcelain crab Petrolisthes armatus from a population in Northeastern Brazil. Specimens were collected monthly between February 2011 and January 2012 from the Baía do Pontal, Ilhéus, Bahia. We analysed 576 specimens, with females reaching the size at the onset of morphological sexual maturity at a carapace width of 4.7 mm and males at a carapace width of 5.83 mm. Sexual size dimorphism was confirmed, with females exhibiting a broader pleon and males a wider carapace and larger chelipeds. Positive allometry was observed in cheliped growth, with males exhibiting variation among juveniles and adults. Females showed ontogenetic distinctions in pleon width and cheliped dimensions. Our findings reveal that females and males of P. armatus exhibit different energetic investments in the development of secondary sexual characteristics, especially in cheliped growth for males and pleon enlargement for females. Macrobrachium jelskii is an endemic shrimp species of South America, with a wide geographical distribution. Although there have been some studies on this organism, information regarding the relative growth of body structures has never been fully explored. In this regard, we aim to analyse the relative growth of this species, in order to investigate the developmental pattern between juveniles and adults, as well as to estimate the morphological sexual maturity (MSM). For morphometric analysis and MSM estimation, the following structures were measured: carapace length (CL); abdomen length (AL); propodus (CPL) and dactyl length (DL) of the second pair of pereopods; second pleonal pleuron width (SPW); and appendix masculina length (AML). We observed developmental differences between juveniles and adults, especially in males. The MSM size for males was estimated at 7.47 mm CL, while for females it was 8.77 mm CL. Females exhibit significantly larger sizes compared to males. Our study reveals divergent energy allocation between juvenile and adult M. jelskii, with males and females reaching MSM at different sizes. The development of these structures with varying allometric constants throughout life stages suggests a strong connection to reproductive behaviour, meeting the specific needs of each life stage’s behavioural repertoire. Introduction: In colorectal cancer, men exhibit a higher incidence than women, and there is a disturbance in the levels of sex steroids in serum in patients with this disease. Consistently, in animals, males have greater tumor growth than females in diverse models. Nevertheless, the role of sex steroids is not well established. For that, we analyzed the effect of the principal gonadal sex steroids in both sexes. We determined sex as a statistically risk factor for colorectal cancer with data obtained from GLOBOCAN database. Methods: To induce colorectal tumors, we used the gold standard chemical method of azoxymethane and dextran sulphate of sodium. To evaluate the role of sex steroids, we gonadectomized independent males and female animals, reconstituting and substituting them with 17b estradiol and dihydrotestosterone. Finally, we determined, in vitro, the proliferation of a human cell line exposed to 17b estradiol, testosterone, or dihydrotestosterone. Sex, as a risk factor for colorectal cancer, showed a statistically significant susceptibility of men over 50 years old. Results: In vivo, males develop a greater number of tumors and with a larger size than females. In males, orchiectomy prevents tumor growth, whereas in females, ovariectomy promotes the development of neoplasms. DHT acts as a protumoral agent in both sexes. 17b estradiol reduces tumor growth in females but enhances it in males, showing a dimorphic effect. In vitro studies reveal that estradiol decreases the proliferation of the HCT-116 colon cancer cell line, while testosterone boosts proliferation in these cells. Background: Fetal sex and placental development impact pregnancy outcomes and fetal–maternal health, but the critical timepoint of placenta establishment in first trimester is understudied in human pregnancies. Methods: Pregnant subjects were recruited in late first trimester (weeks 10–14) at time of chorionic villus sampling, a prenatal diagnostic test. Leftover placenta tissue was collected and stored until birth outcomes were known, then DNA and RNA were isolated from singleton, normal karyotype pregnancies resulting in live births. DNA methylation was measured with the Illumina Infinium MethylationEPIC BeadChip array (n = 56). Differential methylation analysis compared 25 females versus 31 males using a generalized linear model on 743,461 autosomal probes. Gene expression sex differences were analyzed with RNA-sequencing (n = 74). An integrated analysis was performed using linear regression to correlate gene expression and DNA methylation in 51 overlapping placentas. Results: Methylation analysis identified 151 differentially methylated probes (DMPs) significant at false discovery rate < 0.05, including 89 (59%) hypermethylated in females. Probe cg17612569 (GABPA, ATP5J) was the most significant CpG site, hypermethylated in males. There were 11 differentially methylated regions affected by fetal sex, with transcription factors ZNF300 and ZNF311 most significantly hypermethylated in males and females, respectively. RNA-sequencing identified 152 genes significantly sexually dimorphic at false discovery rate < 0.05. The 151 DMPs were associated with 18 genes with gene downregulation (P < 0.05) in the direction of hypermethylation, including 2 genes significant at false discovery rate < 0.05 (ZNF300 and CUB and Sushi multiple domains 1, CSMD1). Both genes, as well as Family With Sequence Similarity 228 Member A (FAM228A), showed significant correlation between DNA methylation and sexually dimorphic gene expression, though FAM228A DNA methylation was less sexually dimorphic. Comparison with other sex differences studies found that cg17612569 is male-hypermethylated across gestation in placenta and in human blood up to adulthood. Conclusions: Overall, sex dimorphic differential methylation with associated differential gene expression in the first trimester placenta is small, but there remain significant genes that may be regulated through methylation leading to differences in the first trimester placenta. Epigenetics, the "overlay" of regulatory signals on DNA which affects how DNA is read, is not well understood in early pregnancy when critical placenta developments are happening that affect the rest of pregnancy. Here, we use leftover placenta biopsy samples (n = 56) donated by Cedars-Sinai patients with informed consent to learn about first trimester human placenta DNA methylation differences due to fetal sex. Out of the total 743,461 sites analyzed, we identified 151 sites significantly affected by fetal sex after correcting p-values to reduce false positives (false discovery rate < 0.05). We also performed an analysis to look at multiple sites and identified 11 regions across the genome with significant DNA methylation changes due to fetal sex. Furthermore, because DNA methylation is a regulatory mark on DNA which typically dampens gene expression, we also compared the DNA methylation sex differences to placental RNA-sequencing gene expression analysis using the same tissue from a mostly overlapping patient group (n = 74 total sequenced, n = 51 overlap). We identify 18 genes which show both significant DNA methylation differences and gene expression changes. The most significant gene was transcription factor ZNF300 with higher DNA methylation in males and reduced gene expression in males (and thus higher gene expression in females). This study identifies some sex differences that continue until later pregnancy and others that are unique to first trimester. Highlights: We identify sex differences in first trimester human placenta DNA methylation (n = 56) and total RNA-sequencing (n = 74), with a sample overlap of n = 51. Metabolic dysfunction-associated steatotic liver disease (MASLD) comprises a spectrum of liver diseases that span simple steatosis, metabolic dysfunctionassociated steatohepatitis (MASH) and fibrosis and may progress to cirrhosis and cancer. The pathogenesis of MASLD is multifactorial and is driven by environmental, genetic, metabolic and immune factors. This review will focus on the role of the type 3 cytokines IL-17 and IL-22 in MASLD pathogenesis and progression. IL-17 and IL-22 are produced by similar adaptive and innate immune cells such as Th17 and innate lymphoid cells, respectively. IL-17-related signaling is upregulated during MASLD resulting in increased chemokines and proinflammatory cytokines in the liver microenvironment, enhanced recruitment of myeloid cells and T cells leading to exacerbation of inflammation and liver disease progression. IL-17 may also act directly by activating hepatic stellate cells resulting in increased fibrosis. In contrast, IL-22 is a pleiotropic cytokine with a dominantly protective signature in MASLD and is currently being tested as a therapeutic strategy. IL-22 also exhibits beneficial metabolic effects and abrogates MASH-related inflammation and fibrosis development via inducing the production of anti-oxidants and anti-apoptotic factors. A sex-dependent effect has been attributed to both cytokines, most importantly to IL-22 in MASLD or related conditions. Altogether, IL-17 and IL-22 are key effectors in MASLD pathogenesis and progression. We will review the role of these two cytokines and cells that produce them in the development of MASLD, their interaction with host factors driving MASLD including sexual dimorphism, and their potential therapeutic benefits. Males and females often differ in ecology, behaviour and lifestyle, and these differences are expected to lead to sex differences in parasite susceptibility. However, neither the sex differences in parasite prevalence, nor their ecological and evolutionary drivers have been investigated across a broad range of taxa using phylogenetically corrected analyses. Using the most extensive dataset yet that includes 755 prevalence estimates from 151 wild bird species in a meta-analytic framework, here we compare sex differences in blood and gastrointestinal parasites. We show that despite sex differences in parasite infection being frequently reported in the literature, only Haemoproteus infections were more prevalent in females than in males. Notably, only seasonality was strongly associated with the sex-specific parasite prevalence of both Leucocytozoon and Haemoproteus, where birds showed greater female bias in prevalence during breeding periods compared to the non-breeding period. No other ecological or sexual selection variables were associated with sex-specific prevalence of parasite prevalence. We suggest that much of the variation in sex-biased prevalence could be idiosyncratic, and driven by local ecology and behavioural differences of the parasite and the host. Quasipaa spinosa, commonly known as the spiny frog, is an economically valued amphibian in China prized for its tender meat and nutritional value. This species exhibits marked sexual dimorphism, most notably the prominent spiny structures on males that are pivotal for mating success and species identification. The spines of Q. spinosa exhibit strong seasonal variation, changing significantly with the reproductive cycle, which typically spans from April to October. Sexually mature males develop densely packed, irregularly arranged round papillae with black spines on their chests during the breeding season, which may then reduce or disappear afterward, while females have smooth chest skin. Despite their ecological importance, the developmental mechanisms and biological functions of these spines have been inadequately explored. This study integrates morphological, transcriptomic, and metabolomic analyses to elucidate the mechanisms underlying the seasonal variation in spine characteristics of Q. spinosa. Our results demonstrate that spine density inversely correlates with body size and that spine development is accompanied by significant changes in epidermal thickness and keratinization during the breeding season. Comparative transcriptomic analysis across different breeding stages revealed significant gene expression alterations in pathways related to extracellular matrix interactions, tyrosine metabolism, Wnt signaling, and melanogenesis. Metabolomic analysis further identified significant seasonal shifts in metabolites essential for energy metabolism and melanin synthesis, including notable increases in citric acid and β-alanine. These molecular changes are consistent with the observed morphological adaptations, suggesting a complex regulatory mechanism supporting spine development and functionality. This study provides novel insights into the molecular basis of spine morphogenesis and its seasonal dynamics in Q. spinosa, contributing valuable information for the species' conservation and aquaculture. Sperm whales (Physeter macrocephalus) have been studied for decades, but the development of their clicks during the animal growth is not yet well known. The click they emit during socialization and echolocation contains information about the length of their acoustic organs and, therefore the length of the body through the interpulse interval (IPI). This paper provides the first IPI/age relationship for juvenile male and female sperm whales (Physeter macrocephalus) based on field recordings of individuals whose age is largely known. Across 9 years, audiovisual recordings of a Mauritian sperm whale social unit were carried out. Adult female and juvenile sperm whales were identified and aged. The dataset made from those recordings is publicly available. The interpulse interval was measured for individuals whose ages ranged from 7 days to around 38 years. The growth of the acoustic organ of juveniles showed an early inter-individual variability as well as sexual dimorphism. Usual growth models were also fitted, predicting a mean I P I ∞ of 3.5 ms for adults and a physical maturity reached at around 30 years old. The use of passive acoustic monitoring (PAM) is one of the main tools used to study sperm whales. Despite developing prior to the appearance of secondary sexual characteristics of the skeleton, the permanent dentition exhibits sexual dimorphism. Therefore, teeth can serve as a means to estimate sex assigned at birth even in young individuals. This project takes a large global sample of maximum dimensions of the crown as well as measurements of the crown at the cervix to explore sexual dimorphism. Dimorphism is noted in teeth throughout the dental arcade, particularly in the canines. We provide sectioning points as well as the probability of correct classification (ranging from 50.9% to 81.3%) for each measurement to aid the practitioner in sex estimation from the dentition. This research provides a method to estimate sex without arbitrary population specifications. We argue for a global approach that incorporates more population variation to remove the need to estimate “ancestry,” (which in actuality is translated to a social race category) and therefore does not force sexual dimorphism‐related variation into these mutable and ambiguous categories. Further, this paper demonstrates the utility of the dentition as an additional indicator to aid with the estimation of sex assigned at birth in forensic anthropology. Sucrose intake is a potential risk factor for non-alcoholic fatty liver disease (NAFLD). Individual characteristics such as sex, play arole in the biological variation of the disease, potentially related to genetic regulation. This research evaluated sex differences in biochemical, histopathological, and gene expression responses associated with NAFLD in C57bl/6N mice on a high sucrose diet. Female and male mice were assigned to control or high sucrose diets (50% sucrose solution) for 20 weeks. After sacrifice, blood and hepatic tissue were collected for analysis. Female mice revealed moderate-to-high NAFLD, whereas male mice showed mild-to-moderate NAFLD. Sex-specific variations were observed in Cd36 gene expression, an upregulation in females compared with the male group, and Adipor1 gene expression showed significant downregulation in the female group in response to high sucrose diet compared with the control group. AbstractSex plays a crucial role in shaping the immune response of vertebrates, leading to differing susceptibilities to diseases between females and males. This review explores the observed sexually dimorphic patterns in different pathogenic infections, immune responses, and molecular events. First, a compilation of historical guidelines for incorporating gender/sex as a variable in research studies to enhance scientific rigor, improve research outcomes, and promote better health for both sexes has been carried out. Following this, the study focuses on examining sex-based differences in immune responses across different taxa, with an in-depth review of available studies on fish, particularly regarding the prevalence of sex-related differences in parasites, viruses, and bacteria. Furthermore, the benefits of considering sexual dimorphism when evaluating the prevalence of diseases in fish management in aquaculture and ecological contexts have been discussed. Integrating sex in understanding immune responses will positively influence fish welfare, economic impacts, and policymaking, leading to more tailored and effective treatments. Introduction: To explore the distribution of Isthmin-1 (ISM1) level and its association with isolated post-challenge hyperglycemia (IPH). Methods: A total of 522 participants without a history of diabetes were invited to attend a standard 75g 2-h oral glucose tolerance test (OGTT), and 71 subjects were further invited for a 3-h oral minimal model test. Insulin sensitivity and b-cell function were evaluated using both HOMA and estimated from OGTT. Circulating ISM1 levels were determined by a commercially available ELISA kit. Results: A total of 76 (14.6%) participants were diagnosed as IPH, accounting for 61.3% of the newly diagnosed diabetes. ISM1 levels were significantly higher in men than in women (1.74 ng/mL versus 0.88 ng/mL). The inverse correlation between ISM1 and b-cell function and IPH was only significant in men. After multivariate adjustment, per unit increment in ISM1 was associated with 0.68-fold (95% CI: 0.49-0.90) reduced odds ratio (OR) of IPH in men. Compared to men with the lowest ISM1 levels, the adjusted OR of IPH with the highest ISM1 levels decreased by 73% (95% CI: 0.11-0.61). Moreover, incorporation of ISM1 into the New Chinese Diabetes Risk Score (NCDRS) model yielded a substantial improvement in net reclassification improvement of 58% (95% CI: 27%-89%) and integrated discrimination improvement of 6.4% (95% CI: 2.7%-10.2%) for IPH. Conclusions: ISM1 was significantly and independently associated with IPH, and serves as a feasible biomarker for the early identification of men with high risk of IPH. [ABSTRACT FROM AUTHOR]

Ontogenetic sexual dimorphism is observed in different primate species, with ecological and evolutionary relationships explaining this pattern. Understanding the growth of the southern brown howler monkey elucidates not only the ecology and evolution but also contributes to conservation projects for this species. Throughout 20 years of the Centro de Pesquisas Biológicas de Indaial—Projeto Bugio, Brazil, we collected morphological data on 105 howlers of the Alouatta guariba species to identify the growth differences between ontogenetic categories and sexes and generate a growth curve to estimate the age of rescued individuals. Linear measurements were employed to obtain body length as well as the dimensions of the head and limbs. All individuals were also weighed to obtain body mass. We assessed growth rate and duration using allometric analysis based on the individuals' ages. We compared growth rate and duration among infant, juvenile, and adult howlers and between sexes. We provide growth curves for body size for both sexes using the Von Bertalanffy model. Infants have accelerated growth rate in comparison to the juveniles and adults, with no differences between sexes in establishing body length at this ontogenetic stage. Males have a prolonged development duration from the juvenile stage, reaching adulthood later than females, which explains the body length differences found in this species. Variables of head and limbs analyzed also showed differences in growth rate and duration, but not so consistently among ontogenetic stages. Mass was not a good variable to understand the growth differences of the animals, since many arrived feeble in the project and may have lost mass due to different circumstances in old age. Therefore, growth curves were obtained only for body length, allowing the estimation of the age of these animals when rescued from the wild to more effectively provide needed care in captivity. Bumblebees are important pollinators for many natural and agricultural systems in temperate regions. Interspecific and intraspecific variation in floral resource preferences have been proposed to influence bumblebee community structure. In particular, sexual dimorphism is a major source of intraspecific niche variation. Although interspecific resource partitioning is well studied, few studies have explored the intraspecific dynamics between workers and males. Here, we report a study on a total of 11 528 workers and 2220 males of 14 bumblebee species recorded over 5 years in the Hengduan Mountains of Southwest China. We first compared the potential for interspecific and intraspecific competition between workers and males using visitation records and resource partitioning indices (overlap index). We then evaluated the influence of nectar traits on flower preference, including nectar volume and the levels of hexose, sucrose and 10 essential amino acids (EAAs). We found that the niche overlap between intraspecific workers and males was higher than that between different species, and temporal overlap alone did not strongly determine diet overlap. Males of most species preferred flowers with high levels of EAAs and hexose, whereas workers of some species preferred flowers with high nectar volume and sucrose levels. This study suggests that there is floral resource partitioning among bumblebee species, and between workers and males, which may play a key role in alleviating interspecific and intraspecific competition.
