[ES] El proceso de emulsificación de aceites ricos en compuestos bioactivos permite su mejor aplicación y conservación durante el tiempo de almacenamiento. Entre los diversos mecanismos de emulsificación, se destaca el método de Pickering ya que utiliza nanopartículas sólidas naturales en sustitución de los tensioactivos artificiales. Debido a sus propiedades antioxidantes, no toxicidad y disponibilidad, en este trabajo se estudiaron distintas modificaciones del quitosano para su potencial aplicación como partícula de Pickering. Las modificaciones estudiadas fueron la autoagregación, también denominada desprotonación, y el entrecruzamiento con tripolifosfato de sodio. Se evaluó el comportamiento de estas partículas emulsionando aceite de café tostado, un subproducto de la industria de café con un alto contenido de compuestos bioactivos y compuestos volátiles de interés. Posteriormente, se analizaron las propiedades físico-químicas y la estabilidad de las microcápsulas producidas tras el secado de las emulsiones mediante técnicas de secado por atomización y liofilización. Todas las emulsiones se caracterizaron por tener un comportamiento reológico pseudoplástico, sufriendo desintegración a lo largo del proceso de digestión. Las emulsiones formuladas con nanopartículas de quitosano desprotonadas y menor concentración de aceite mostraron una mejor estabilización y, en consecuencia, una mayor bioaccesibilidad de los compuestos fenólicos totales. Las diferentes nanopartículas de quitosano se caracterizaron estudiando su carga superficial, distribución del tamaño de partícula, microestructura y afinidad agua/aceite. A medida que se aumentó la concentración de estas partículas, se afectó positivamente la viscosidad de las emulsiones debido a la formación de una red tridimensional en la fase continua. Las nanopartículas obtenidas al entrecruzar quitosano con tripolifosfato de sodio fueron más pequeñas, dando como resultado emulsiones con gotas más pequeñas. Las emulsiones de Pickering que contenían un 10% de aceite de café tostado se secaron por atomización y se liofilizaron utilizando las diferentes nanopartículas de quitosano estudiadas y maltodextrina como agentes portadores. Las microcápsulas obtenidas tuvieron la humedad, actividad del agua y solubilidad adecuada para su manipulación y almacenamiento. La presencia de nanopartículas de quitosano permitió una mayor retención de aceite en las microcápsulas y mayor eficiencia de encapsulación. Los compuestos bioactivos y las propiedades antioxidantes se conservaron mejor durante la liofilización. Por otro lado, las microcápsulas obtenidas por atomización presentaron mayor protección de estos compuestos durante la digestión. Las microcápsulas formuladas con nanopartículas desprotonadas fueron sometidas a almacenamiento durante 30 días a 25 ºC. Durante el almacenamiento, se evaluó la protección contra la oxidación de lípidos y la liberación de volátiles. Las isotermas de sorción de agua de estas muestras se determinaron también previamente en las condiciones de almacenamiento. Ambas muestras presentaron isotermas del tipo II, lo que permitió un buen ajuste del modelo de GAB a los datos experimentales. La determinación del índice de peróxido y del contenido de dienos conjugados dio lugar a valores adecuados durante el almacenamiento, aunque las muestras liofilizadas presentaron una ligera tendencia a la oxidación debido a la mayor cantidad de aceite superficial. Aunque hubo ligeras diferencias entre las muestras secas, todas mostraron menos pérdida de aromas totales (~28%) en comparación con el aceite no encapsulado (~51%) al final del almacenamiento. Les emulsions formulades amb nanopartícules de quitosà desprotonades i menor concentració d'oli van mostrar una millor estabilització i, en conseqüència, una major bioaccesibilitat als compostos fenòlics totals. Les diferents nanopartícules de quitosà es van caracteritzar estudiant la seua càrrega superficial, distribució del tamany de partícula, microestructura i afinitat aigua/oli. A mesura que es va augmentar la concentració d'aquestes partícules, es va afectar positivament la viscositat de les emulsions a causa de la formació d'una xarxa tridimensional en la fase contínua. Les nanopartícules obtingudes a l'entrecreuar quitosà amb tripolifosfat de sodi van ser més menudes, donant com a resultat emulsions amb gotes més menudes també. Les emulsions de Pickering que contenien un 10% d'oli de cafè torrat es van assecar per atomització i es liofilitzaren utilitzant les diferents nanopartícules de quitosà estudiades i maltodextrina com a agents portadors. Les microcàpsules obtingudes van obtenir una humitat, activitat de l'aigua i solubilitat adequada per a la seua manipulació i emmagatzematge. La presència de nanopartícules de quitosà va permetre major retenció d'oli en les microcàpsules i major eficiència d'encapsulació. Els compostos bioactius i les propietats antioxidants es van conservar millor durant la liofilització. D'altra banda, les microcàpsules obtingudes per atomització presentaren major protecció d'aquests compostos durant la digestió. Les microcàpsules formulades amb nanopartícules desprotonades també van ser sotmeses a la prova d'emmagatzematge durant 30 dies a 25°C. Durant l'emmagatzematge, es va avaluar la seua protecció contra l'oxidació de lípids i l'alliberament de volàtils. Per això, les isotermes de sorció d'aigua d'aquestes mostres es van determinar prèviament en les condicions d'emmagatzematge. Les dues mostres van presentar isotermes de tipus II, el que va permetre un bon ajust del model de GAB a les dades experimentals. L'índex de peròxids i el contingut de diens conjugats van resultar en valors adequats durant l'emmagatzematge, encara que les mostres liofilitzades van presentar una lleugera tendència a l'oxidació a causa de la major quantitat d'oli superficial. Encara que va haver lleugeres diferències entre les mostres seques, totes van mostrar menys pèrdua d'aromes totals (~28%) en comparació amb l'oli no encapsulat (~51%) a la fi de l'emmagatzematge. Així, es va concloure que les nanopartícules de quitosà estudiades van ser eficients per encapsular l'oli de cafè torrat i preservar les seues característiques enfront de l'acció d'agents externs., [EN] The emulsification process of bioactive-rich oils makes possible their better application and preservation over the storage time. Among the many emulsification mechanisms, the Pickering method has been highlighted as it uses natural solid nanoparticles in replacement of artificial surfactants. Due to the antioxidant properties, non-toxicity and availability, this work aimed at studying chitosan modifications to produce potential Pickering particles. The studied modifications comprised self-aggregation, also called deprotonation, and crosslinking with sodium tripolyphosphate. The performance of these particles was evaluated in the emulsification of roasted coffee oil, a by-product of the coffee industry with a high content of bioactive and volatile compounds of interest. Subsequently, the physicochemical properties and stability of the microcapsules produced after drying the emulsions using spray-drying and lyophilization techniques were analyzed. All emulsions were characterized as shear-thinning, being them destabilized over the digestion process. Emulsions formulated with deprotonated chitosan nanoparticles and lower oil concentrations showed better stabilization and, consequently, greater bioaccessibility of total phenolic compounds. The different chitosan nanoparticles were characterized regarding surface charge, particle size distribution, microstructure and oil/water affinity. Deprotonated chitosan nanoparticles had a larger particle size, which resulted in emulsions with larger oil droplets. As the concentration of these particles increased, the viscosity of the emulsions was positively affected by the formation of a three-dimensional network in the continuous phase. The nanoparticles obtained by crosslinking with sodium tripolyphosphate were smaller, resulting in emulsions with smaller droplets. The viscosity of these emulsions was lower and little affected by the concentration of particles. Pickering emulsions containing 10% roasted coffee oil were spray-dried and freeze-dried, using the different studied chitosan nanoparticles and maltodextrin as carrier agents. The resulting microcapsules showed adequate moisture content, water activity and solubility for subsequent handling and storage. The presence of chitosan nanoparticles resulted in greater oil retention in the microcapsules and higher encapsulation efficiency. Microcapsules obtained by spray-drying had a more regular spherical shape, while the microparticles obtained by freeze-drying were larger with irregular morphology. Bioactive compounds and antioxidant properties were more preserved during freeze-drying. On the other hand, spray drying allowed greater protection of these compounds during the in vitro digestion. The spray- and freeze-dried microcapsules formulated with deprotonated nanoparticles were subjected to the storage test for 30 days at 25 ºC. During storage, their protection against lipid oxidation and volatile release were evaluated. The water sorption isotherms of these samples were previously determined under the storage conditions. Both samples presented type II isotherms, which resulted in a good fitting accuracy of the GAB model to the experimental data. The peroxide index and the conjugated dienes content resulted in adequate values during storage, although the freeze-dried samples showed a slightly higher tendency to oxidation due to the higher amount of surface oil. Although slight differences were observed between the dried samples, both of them showed less loss of total volatile compounds (~28%) when compared to the non-encapsulated oil (~51%) at the end of storage. Se estudió el proceso de secado spray como alternativa de menor costo para obtener bacterias probióticas deshidratadas. Se estudió el efecto del proceso sobre la viabilidad y funcionalidad de las mismas. A commercial drinkable yogurt with and without 4% of added trehalose (as cell protectant) was spray-dried obtaining a powder with low water activity (aw). Total bacterial count in the powder was between 8.48–8.90 log cfu/g. The dried yogurt was stored: (i) at 38 °C and aw = 0.33; (ii) at 38 °C in hermetically sealed flasks (aw = 0.21/0.22); (iii) in a cyclic temperature chamber (10–20 °C) in hermetically sealed flasks (aw = 0.21/0.22). Whole milk was then fermented by adding an inoculum of spray-dried yogurt after storage under these different conditions. The kinetics of acidification showed the presence of a lag time which was strongly dependent on storage conditions. The data was fitted with a logistic type equation from which the lag time was calculated. To evaluate structural differences among samples, Fourier Transform Infrared spectra (FTIR) were recorded. Partial Least Squares (PLS) models enabled a good correlation between lag time of fermentation and FTIR spectra. The lag time for yogurt powder stored at aw about 0.21/0.22 and cyclic temperature 10–20 °C remained approximately constant over the 12 weeks of storage, while all the other conditions resulted in a dramatic increase. The addition of trehalose had a small influence on lag time and, therefore, as a protectant of lactobacilli. Fil: Bater, Caterina. El desarrollo de probióticos lácticos deshidratados implica un desafío científico-tecnológico y reviste gran interés como herramienta de innovación para ampliar el mercado de alimentos y/o suplementos dietarios con propiedades funcionales. El proceso de secado por pulverización genera estrés térmico, osmótico y oxidativo para las células, con pérdida de viabilidad y/o de su actividad metabólica. El inmunobiótico Lactobacillus (L.) rhamnosus CRL1505 es objeto de continuo estudio por sus propiedades beneficiosas en salud. El objetivo de este trabajo de tesis doctoral fue conocer su comportamiento frente a situaciones de estrés derivadas del proceso de deshidratación y sentar las bases tecnológicas para el desarrollo de complementos dietarios probióticos. Se formuló un medio de cultivo económico (MCM) de base no láctea para la producción de biomasa, que contiene una elevada concentración de sales de fosfato (9,2 g/L). En este medio se evidenció la presencia de gránulos de polifosfato (polyP) en el citoplasma del inmunobiótico CRL1505, mediante la estandarización de diferentes técnicas (microscopía óptica y electrónica de transmisión, fluorescencia, RT-PCR, PAGE, espectrofotometría). Se relacionó la acumulación intracelular de polyP y la concentración de fosfato del medio de cultivo, estableciéndose dos condiciones para obtener células con alto y bajo contenido de polyP: MCM y MCM(-), respectivamente, según la concentración de fosfato del medio de cultivo. Posteriormente, se evaluó la tolerancia intrínseca de los cultivos a estrés térmico, oxidativo y osmótico, confirmándose que dicha tolerancia requiere 9,2g/L de sales de fosfato inorgánico para obtener la tolerancia a estrés observada. Se evaluó el efecto de pH y la fase de crecimiento sobre la acumulación de polyP y tolerancia a shock térmico usando bioreactores (1,5 L). Se estableció la condición óptima de cultivo para obtener células tolerantes a estrés: crecimiento a pH libre y cosecha en fase estacionaria. La respuesta a estrés del inmunobiótico CRL1505 se evaluó en diferentes medios de shock térmico, obteniéndose los mejores resultados con un pool de sales inorgánicas (MCM3) conteniendo Na2HPO4, KH2PO4, MgSO4 y MnSO4. Establecidas las condiciones óptimas de producción de biomasa con máxima tolerancia a estrés, se procedió al secado por pulverización (escala laboratorio) en dos matrices: maltodextrina (polvo A) y maltodextrina-MCM3 (polvo B). La sobrevida celular fue mayor en el polvo B (72,5 %) respecto al A (42,8 %), poniéndose en evidencia el efecto termoprotector de MCM3. Finalmente, se evaluó el efecto biológico de extractos intracelulares de L. rhamnosus CRL1505 conteniendo polyP y de un polyP sintético (control) mediante ensayos in vivo utilizando un modelo murino de inflamación pulmonar aguda. Se demostró que los polyP evitan el desarrollo de una respuesta inflamatoria local en las vías respiratorias.En esta Tesis Doctoral se establecieron diferentes estrategias para mejorar la sobrevida del probiótico CRL1505 a estrés relacionado con el proceso de secado: medio de cultivo de bajo costo con alto contenido de fosfato inorgánico; aumento de polyP intracelular; agregado de MCM3 (pool de sales inorgánicas) a la matriz de secado. El uso de polyP de L. rhamnosus CRL1505 como un nuevo biocompuesto con propiedades funcionales es una de las proyecciones de este trabajo. The dehydration of lactic acid probiotics implies a scientific-technological challenge, and it is of great interest to expand the market of food and dietary supplements with functional properties. The spray drying process generates thermal, osmotic and oxidative stress for the cells with loss of viability and/or metabolic activity. The immunobiotic Lactobacillus (L.) rhamnosus CRL1505, used in this study, is under continuous study because of its beneficial health properties. The objective of this phD study was to know the behavior of the strain under oxidative and thermic stress which take place during dehydration process, as the first step for probiotic dietary supplements development. An economic non-dairy based growth medium (MCM) was formulated for biomass production, which contains high concentrations of phosphate salts (9.2 g/L/64 Mm). In this medium, the immunobiotic CRL1505 accumulates polyphosphate granules (polyP) in the cytoplasm, which was evinced by setting up different techniques, e.g., optical and transmission electronic microscopy; fluorescence; RT-PCR; PAGE; spectrophotometry. The relationship existing between the intracellular accumulation of polyP and the phosphate concentration in the culture medium was established. From these results, two growth conditions MCM and MCM(-) for obtaining cells with high and low polyP content, respectively, were set up. Afterwards, the intrinsic tolerance of the cultures to thermal, oxidative and osmotic stress was evaluated, thus confirming that 9.2 g/L of phosphate salts in the culture medium are required for the stress tolerance observed. The effect of pH and growth phase on both the accumulation of polyP and the tolerance of the cells to heat shock was evaluated in bioreactors (1.5 L). The optimal culture conditions were free pH and stationary phase for obtaining stress tolerant cells. The stress response of the immunobiotic CRL1505 in different heat shock media was evaluated, best results being obtained with MCM3 (pool of inorganic salts) containing Na2HPO4, KH2PO4, MgSO4 and MnSO4. Once the optimum conditions for biomass production were established, cultures were spray dryed at lab scale using two dry matrix: maltodextrin (powder A) and maltodextrin-MCM3 (powder B). Cell survival was higher in powder B (72.5 %) than in powder A (42.8 %), thus confirming the thermoprotective effect of MCM3. Finally, the biological effect of intracellular extracts of L. rhamnosus CRL1505 containing polyP and a synthetic polyP (control) was evaluated in vivo using a murine model of acute lung inflammation. It was shown that polyPs avoid development of a local inflammatory response in the respiratory tract. In this Doctoral Thesis, different strategies to improve the survival rate of L. rhamnosus CRL1505 to stress conditions involved in the drying process were established: a low-cost culture medium with high inorganic phosphate content; the increase in intracellular polyP; the addition of MCM3 (pool of inorganic salts) in the drying matrix. La microencapsulación del citral se realizó mediante secado por atomización y se optimizó empleando metodología superficie respuesta (MSR), maximizando el rendimiento y eficiencia de encapsulación (EE) respecto a la relación citral: dextrina (Ct:Dx) entre 1:5 y 1:20 y temperatura del aire de entrada (TEA) al secador entre 120 y 200 °C. Se evaluaron las características físico-químicas y actividad antimicrobiana de las micropartículas de citral (MC) óptimas. La predicción del contenido superficial de citral (CSct) en las micropartículas se realizó utilizando espectroscopía NIR y quimiometría. Las biopelículas activas se elaboraron mediante método de casting y se optimizó con MSR, minimizando el valor de transparencia, opacidad y módulo elástico respecto a la relación caseinato de sodio: sorbitol (CS:Sb) entre 1: 0.5 y 1:1.5 y relación CS:MC entre 1:0.5 y 1:1.5. Se caracterizaron las propiedades de color, mecánicas y micro-estructurales de las biopelículas activas óptimas. Los quesos se envasaron en biopelículas con MC, biopelículas con citral libre y bolsas de polietileno; se evaluó su perfil físico, sensorial y microbiológico durante 14 días a 6°C. Las condiciones óptimas de microencapsulación fueron 1:5 de relación Ct:Dx y 187°C de TEA, mostrando características físico-químicas aceptables y actividad antimicrobiana de amplio espectro. Los modelos quimiométricos empleados en la predicción de CSct presentaron R2 entre 0.80 y 0.95, siendo las redes neuronales artificiales las que presentaron mejores resultados. Las condiciones óptimas de biopelículas activas se alcanzaron a relación CS:Sb de 1:0.91 y relación CS:MC de 1:0.95; obteniendo propiedades de color, mecánicas y microestructurales adecuadas para envasado de alimentos. Los quesos en biopelículas con MC presentaron menores recuentos microbianos y mejor perfil físico y sensorial, prolongando lavida útil del producto hasta 14 días en condiciones aceptables. The objective of the work was to develop active biofilms with microencapsulated citral essential oil and its effect on the microbial load of fresh cheese. Microencapsulation of citral was performed by spray drying and was optimized using surface response methodology (MSR), maximizing yield and encapsulation efficiency (EE) with respect to the citral: dextrin (Ct:Dx) ratio between 1: 5 and 1: 20 and inlet air temperature (TEA) to the dryer between 120 and 200 °C. The physicochemical characteristics and antimicrobial activity to the optimal citral microparticles (MC) were evaluated. The prediction of the surface content of citral (CSct) in microparticles was performed using NIR spectroscopy and chemometric. The active biofilms were elaborated by casting method and optimized with MSR, minimizing the transparency value, opacity and elastic modulus with respect to the sodium caseinate: sorbitol (CS:Sb) ratio between 1: 0.5 and 1: 1.5 and CS:MC ratio between 1: 0.5 and 1: 1.5. Color, mechanical and microstructural properties of the optimal active biofilms were characterized. The cheeses were packed in biofilms with MC, biofilms with free citral and polyethylene bags; their physical, sensory and microbiological profile was evaluated during 14 days at 6 °C. The optimal microencapsulation conditions were 1: 5 to Ct:Dx and 187 °C to TEA, showing acceptable physicochemical characteristics and broad-spectrum antimicrobial activity. The chemometric models used in the CSct prediction presented R2 between 0.80 and 0.95; artificial neural networks showed better results. The optimal conditions of active biofilms were reached at CS:Sb ratio of 1: 0.91 and CS:MC ratio of 1: 0.95; obtaining color, mechanical and microstructural properties adequate for food packaging. The cheeses in biofilms with MC presented lower microbial counts and better physical and sensory profile, prolonging the useful life of the product up to 14 days in acceptable conditions. Tesis

Ancellotta red wine (Vitis vinifera L.) was encapsulated by spray-drying (inlet and outlet temperatures were 145 °C and 70 °C, respectively) to obtain a wine powder with a low water activity (aw) using maltodextrin DE10 as an encapsulating agent. The retention of total monomeric anthocyanins (TMA) in the wine powder was found to be greater than 80%. Anthocyanin profiles of Ancellotta liquid wine and wine powder were characterized by using HPLC–DAD and 33 compounds were identified. The wine powder was stored under two different water activity values (aw 0.25 and aw 0.33). Furthermore, the TMA (pH-differential method), total anthocyanins and malvidin-3-glucoside were determined by HPLC–DAD for up to 90 days’ storage at 38 °C. Total anthocyanins and malvidin-3-glucoside decreased very slowly during storage. The stability of anthocyanins and color differences (ΔE*ab) in 1% wine powder solution at different pHs and temperatures were evaluated. These results indicated that Ancellotta wine powder has the potential to be used as a food colorant in low pH and low water activity foods.

Facultad de Ingeniería y Ciencias Agrarias; Argentina Abstract: Epidemiological evidence indicates that moderate consumption of red wine reduces the incidence of coronary disease, atherosclerosis, and platelet aggregation. Wine is very rich in antioxidant compounds because of their phenolic components. However, many people for ethnic, social or religious reasons do not consume wine. Drying/encapsulation of red wine in the presence of adequate carbohydrates leads to water and more than 99% of alcohol removal; a glassy amorphous microstructure is obtained in which the wine’s phenolic compounds are entrapped. The resulting product is a free flowing powder which could be used for the polyphenol enrichment of healthy foods and/or drink powders, as well as in the pharmaceutical industry. The wine industry may take advantage of the dried/encapsulated red wine using as a raw material red wines which have little commercial value for different reasons; i.e. poor quality due to raw material, unfavourable climatic conditions, or wines that suffered some alteration during the wine making process. Dry encapsulated wine may be a new alternative to red wines that cannot be sold as such for different reasons, and open new opportunities to diversify wine products.

RESUMEN: La estabilidad de las propiedades del polvo de aguacate adicionado con vitaminas y minerales (PA+CA) empleando secado por atomización, bajo diferentes temperaturas de almacenamiento (15, 25 y 35°C) y tipo de atmósfera de empaque (vacío y nitrógeno) durante 180 días, fueron evaluadas. Los resultados obtenidos indican mayores cambios negativos en la calidad del PA+CA y una disminución en sus propiedades nutricionales, asociado a los bajos porcentajes de retención de vitamina A (36,44 a 36,48%) y vitamina D (50,01 a 52,53%), con el incremento en la temperatura y el tiempo de almacenamiento. Desde el punto de vista de la fortificación mineral, el PA+CA se presenta como una excelente opción para la industria farmacéutica y de alimentos, con recuperación total del Ca y el Fe adicionados. Context: A functional extract from Lannea microcarpa (Lm), possess interesting antioxidant and anti-inflammatory properties. However, the unprocessed dried extract occurs as sticky and low-water-soluble material showing critical properties for industrial applications. The unprocessed dried extract is not always enough stable to preserve its functional properties, also giving practical difficulties for the manufacturing. Aims: This research aimed to produce Lm extract microparticles with enhanced functional stability and technological characteristics by spray-drying. Methods: Lm extract was microencapsulated by spray-drying using a sodium-carboxymethylcellulose (NaCMC) based matrix. Physicochemical and technological characteristics (determined by UV, HPLC, LLS, SEM, DSC, and in vitro dissolution tests), as well as antioxidant properties (DPPH-test) of the resulting powder (LmC) were examined. Results: The produced spray dried microparticles showed satisfying encapsulation efficiency, good functional stability and enhanced technological properties. The selected carrier and process conditions led to a stable and handling microencapsulated powder form with improved water dissolution rate. Moreover, the matrix was also able to preserve the antioxidant activity of the phenolic compounds-rich extract. Conclusions: The made-up powder resulted in a functional component that can be used with great potential in cosmetics, foods or nutraceutical products.

Instituto de Investigaciones Científicas y Técnicas de las Fuerzas Armadas; Argentina Abstract: Spray dried orange flavour encapsulated in different amorphous matrices comprised of maltodextrin (MD) and different combinations with; sucrose, trehalose, lactose, modified starch and gum arabic (ga); were evaluated by sensory analysis and electronic nose (e-nose). With both techniques the flavours encapsulated in MDsucrose and MD -lactose-sucrose were perceived as similar. However, the e-nose did not detect any differences among the other matrices (MD-trehalose, MD, MD-sucrose at a different concentration and MD-ga). On the contrary, sensory analysis was able to group MD-trehalose and MD describing them by: woody, marmalade, syrup, citrus terpens, and Vitamin C; MD -sucrose at 40% and 10% concentration and MD -lactosesucrose were grouped and represented by powder juice, tangerine and pungency, while MD-ga was differentiated from the rest by the attributes peely, plastic, solvent and green. In this way, it was shown that matrix composition determines the aromatic profile of spray dried encapsulated orange flavours.
