Injury is one of the most common health problems people face in daily life. Finding the causes that prevent healing in wounds and eliminating these causes helps to accelerate the healing process. However, the increase in the elderly population, the inability of people living far from health institutions to access health services, increases in health expenditures and poorly managed wounds in complex patients can lead to the development of chronic wounds after a while. The aim of this study is to investigate the in vitro effects of red LED light, infrared LED light and hypochlorous acid on two different cell types. In the study, a low-cost and portable optical device was prepared for irradiation. Red light, infrared light, hypochlorous acid and the combination of these lights with hypochlorous acid were applied to the keratinocyte cell line (HaCaT) and fibroblast cell line (BJ). At the end of the experiment, keratinocyte and fibroblast cells reacted differently to the treatments. An increase was observed in keratinocyte after 48 and 72 hours, while a decrease in the fibroblast cells. The ability of photobiomodulation to promote wound healing is due to increased proliferation of cells. Yaralarda iyileşmeyi engelleyen sebeplerin bulunması ve bu sebeplerin ortadan kaldırılması iyileşme sürecinin hızlandırılmasına yardımcı olmaktadır. Ancak yaşlı nüfusun artması, sağlık kuruluşlarına uzak yaşayan insanların sağlık hizmetine ulaşamaması, sağlık harcamalarındaki artışların olması ve kompleks hastalardaki iyi yönetilemeyen yaralar, bir süre sonra kronik yaranın gelişmesine yol açabilmektedir. Bu çalışmanın amacı kırmızı LED ışık, infrared LED ışık ve hipokloröz asidin iki farklı hücre tipi üzerindeki in vitro etkilerini araştırmaktır. Çalışmada uygulama için düşük maliyetli ve taşınabilir optik düzenek hazırlanmıştır. Keratinosit hücre hattına (HaCaT) ve fibroblast hücre hattına (BJ) kırmızı ışık, infrared ışık, hipokloröz asit ve bu ışıkların hipokloröz asit ile kombinasyonu uygulanmıştır. Deneyin sonunda keratinosit ve fibroblast hücreleri uygulamalara farklı tepki gösterdi. Keratinosit hücrelerinde 48 ve 72 saat sonra artış gözlemlenirken fibroblast hücrelerinin üremelerinde azalma gerçekleşti. Fotobiyomodülasyonun yara iyileşmesini destekleme kabiliyeti, hücrelerin üremesindeki artışa bağlıdır. Elde ettiğimiz veriler ile sağlık alanlarında yara iyileşmesine katkı yapabileceğini düşünmekteyiz.

Purpose: Endometrial cancer is one of the most common gynecological cancers in the world. Glabridin is a main isoflavone in Glycyrrhiza glabra (licorice) root. It has several therapeutic effects such as anti-proliferative and anti-inflammatory. Long non-coding RNAs (LncRNAs) play a role in a variety of cellular processes, and their abnormal expression may contribute to tumor development and progression. In this study, the effects of glabridin on LncRNAs gene expression and viability of HEC-1B human endometrial cancer cell lines have been investigated. Materials and methods: Glabridin was applied to HEC-1B cells in concentrations of 1 μM, 10 μM, 20 μM, 40 μM, 60 μM, and 80 μM. Glabridin's effect on HEC-1B cell proliferation was also evaluated using MTS assay. Expression profiles of LncRNAs such as H19, RNU43, LNC-MYC-3:1 and ABCC5-AS1:1 were determined by real-time PCR. Results: Glabridin reduced the viability of HEC-1B cells in a time- and dose-dependent manner. The half maximal inhibitory concentration (IC50) dose in HEC-1B cells was detected to be 21.32 μM and 13.5 μM at the 24th and 48 hours, respectively. Glabridin has been observed to cause a significant decrease in the expression of H19 and RNU43 while increasing in the expression of LNC-MYC-3:1 and ABCC5-AS1:1. Conclusion: Glabridin could induce HEC-1B cell death by regulating LncRNAs expression. As a result, glabridin is a potential candidate for a more effective therapeutic agent against human endometrial cancer.

This study was designed for investigate of antioxidant, and anti-proliferative activities of acetone, methanol and water extracts of different parts (fresh and underground parts) of Cyclamen cilicium extracts. The antioxidant activities were determined using DPPH, ABTS free radical scavenging activities. Anti-proliferative activities of extracts in Caco-2 colon cancer cells were detected by XTT assay. The determination of contents was evaluated using total phenolics, flavonoid and tannin contents and found maximum values 3.69 ± 0.13 (mg GAE/g-extract), 18.48 ± 0.12 (mg QE/g- extract), 41.17 ± 0.44 (mg CE/g- extract) respectively. C. cilicium extracts exhibited anti-proliferative activity in Caco-2 colon cancer cells. IC50 values of methanol and water extracts of fresh part and water extract of underground part were determined as 35.3276 μg/mL, 42.1831 μg/mL and 64.6144 μg/mL at 24h, respectively. The results showed that C. cilicium is the source of natural antioxidants and may have potential use in pharmaceutical, health, biotechnology, agricultural industry.

Amaç: Ovaryum dokusu eksplant kültürü oluşturarak dokulardan göç eden stromal hücreleri izole etmeyi, ço ğaltmayı ve karakterize etmeyi hedefledik. Gereç ve Yöntem: 2 adet 4 haftalık (prepubertal) dişi Wistar Albino tipi sıçanların ovaryum dokularından eksplant kültürler oluşturuldu. Daha sonra prolifere olan overyan stromal hücrelere 2. pasajda (P2) flow sitometri analizi yapılarak CD29, CD54, CD90 (mezenkimal kök hücre yüzey antijeni) ve CD45 (hematopoetik kök hücre yüzey antijeni) ifadelerine bakıldı. Bulgular: Kültür ortamındaki hücrelerin proliferasyon yetenekleri ve morfolojik özellikleri seri pasajlar yapılarak incelendi. Stromal hücreler 2. günde dokulardan göç ederek kültür kaplarına yapıştılar ve çoğalıp 7. günde konflue (%70-80) oldular. Kültür ortam ındaki overyan stromal hücrelerin morfolojileri, faz kontrast mikroskobu ile incelendi ğinde iğ şeklinde ve fibroblast be nzeri hücre toplulukları olarak görüldü. Stromal hücre çoğalan kültür kaplarında mikroskop alt ında yap ılan sayımda s ırasıyla 2x106 ve 1.5x106 hücrenin ürediği gözlemlendi. İzole edilen overyan stromal hücrelerin flow sitometri analizlerinde CD54, CD90 ve CD45 yüzey antijenlerini eksprese ettikleri, CD29 yüzey antijenlerini ise eksprese etmedikleri belirlendi. Sonuç: Yapılan analizde kültür ortamında izole edip, ürettiğimiz overyan stromal hücrelerin hem hematopoetik hem de mezenkimal kök hücre belirteçlerinden bazılarını eksprese ettiklerini belirledik.

Purpose: The aim of this study was to determine the effects of Echinacoside on cell proliferation, invasion and mRNA expression changes of invasion-related genes in PC3 androgen-independent prostate cancer cells.Material and methods: The effect of Echinacoside on cell proliferation in PC3 cells was determined by XTT method. Anti-invasive efficacy was achieved using the transwell chamber. Total RNA isolation was performed by Trizol and cDNA was subsequently synthesized. mRNA expression changes in MMP2, MMP9, TIMP1, TIMP2 and TIMP3 were also performed in RT-PCR with SYBER Green.Results: In this study, the IC50 dose of Echinacoside in PC3 cells was determined as 55.21 μM at 48h. It was determined that echinacoside inhibited cell invasion in PC3 cells and reduced the invasion by 66% in the dose group. In addition, it was found statistically significant that Echinacoside increased TIMP1 mRNA expression 1.96 times, TIMP2 mRNA expression 2.60 times, while decreasing MMP2 expression 3.82 times and MMP9 mRNA expression 1.54 times in IC50 dose group according to control.Conclusion: It was revealed that echinacoside has an anti-proliferative effect on PC3 prostate cancer cells. It has also been shown that invasion-related genes can suppress invasion by regulating expression changes. With this study, preliminary data were presented in terms of detailed molecular biological studies to be carried out on echinacoside and its effect on prostate cancer.

Şu anda, GBM için iyileştirici tedavi seçenekleri yoktur ve 5 yıllık hayatta kalma oranı %5’den daha azdır. Monensin, ‘’Streptomyces cinnamonensis’’ den elde edilen antibakteriyal ve antiparazitik etkileri bilinen iyonofor bir antibiyotiktir. Literatürde monensinin GBM hücrelerinin apoptoz mekanizması üzerine etki gösterdiği bir çalışmaya rastlanmadığından yapılan bu çalışmanın amacı monensinin U373 GBM hücrelerinde apoptoz aracılı hücre proliferasyonu üzerine etkilerini araştırmaktır. Monensinin U373 hücre canlılığı üzerine etkileri XTT ile apoptoz üzerine etkileri ise RT-PCR ve Annexin V ile araştırılmıştır. Monensinin U373 GBM hücrelerinde IC50 değeri 48’inci saatte 4 µM olarak bulunmuştur. Monensin U373 GBM hücrelerinde apoptoz oranında 6 katlık bir artışa neden olmuştur. Bununla birlikte monensin kaspaz-10 gen ekspresyonunu arttırarak apoptozu anlamlı olarak aktive etmiştir. Sunulan çalışma monensinin GBM hücrelerinin kaspaz-10 aracılı apoptoz mekanizması üzerine etkilerini gösteren ilk çalışmadır. Currently, there are no curative treatment options for GBM and the 5-year survival rate is less than 5%. Therefore, further studies are needed to identify effective markers and therapeutic targets for GBM treatment. Monensin is an ionophore antibiotic obtained from Streptomyces cinnamonensis with known antibacterial and antiparasitic effects. In the literature, no study has been found that shows an effect of monensin on the apoptosis mechanism of GBM cells. The aim of the present study was to investigate the effects of monensin on apoptosis-mediated cell proliferation in U373 GBM cells. The effects of monensin on U373 cell viability were investigated by XTT and its effects on apoptosis were investigated by RT-PCR and Annexin V. The IC50 value of monensin in U373 GBM cells was 4 µM at 48 hours. Monensin caused a 6-fold increase in the rate of apoptosis in U373 GBM cells. Monensin also significantly triggered apoptosis by increasing caspase-10 gene expression. The present study is the first to demonstrate the effects of monensin on the caspase-10-mediated apoptosis mechanism of GBM cells. Our results suggest that monensin may be a therapeutic anticancer drug compound with potent apoptotic effects in GBM cancer.
