Abstract

The NC1 domain of the alpha 4 chain of type IV collagen was previously reported to exert anti‐tumor properties in a melanoma model and to inhibit angiogenesis. The minimal active sequence identified to date comprises 13 amino acids: QKISRCQVCVKYS. Unfortunately, this sequence is not soluble in aqueous media and requires prior dissolution in DMSO. The disulfide bridge (DB) that spontaneously forms in solution between two cysteine residues is crucial for its biological activity. The aim of this article was to study the impact of DMSO on the physicochemical properties of the QS‐13 peptide and to replace hydrophobic amino acids to enhance its water solubility. Using bioinformatics (GROMACS, VMD, and ProtParam) and programming (Python and RStudio) software programs, we demonstrated that DMSO could promote the formation of DBs, but it is not strictly necessary. Among the QS‐13 substituted peptides, some were demonstrated to display similar characteristics to the original peptide. 14. Dynamized ultra-low dilution of Ruta graveolensdisrupts plasma membrane organization and decreases migration of melanoma cancer cell

Abstract

ABSTRACTCutaneous melanoma is a cancer with a very poor prognosis mainly because of metastatic dissemination and therefore a deregulation of cell migration. Current therapies can benefit from complementary medicines as supportive care in oncology. In our study, we show that a dynamized ultra-low dilution of Ruta Graveolensleads to an in vitroinhibition of migration on fibronectin of B16F10 melanoma cells, as well as a decrease in metastatic dissemination in vivo. These effects appear to be due to a disruption of plasma membrane organization, with a change in cell and membrane stiffness, associated with a disorganization of the actin cytoskeleton and a modification of the lipid composition of the plasma membrane. Together, these results demonstrate, in in vitroand in vivomodels of cutaneous melanoma, an anti-cancer and anti-metastatic activity of ultra-low dynamized dilution of Ruta graveolensand reinforce its interest as complementary medicine in oncology. more...

Abstract

International audience; Background: LRP-1 is a multifunctional scavenger receptor belonging to the LDLR family. Due to its capacity to control pericellular levels of various growth factors and proteases, LRP-1 plays a crucial role in membrane proteome dynamics, which appears decisive for tumor progression. Methods: LRP-1 involvement in a TNBC model was assessed using an RNA interference strategy in MDA-MB-231 cells. In vivo, tumorigenic and angiogenic effects of LRP-1-repressed cells were evaluated using an orthotopic xenograft model and two angiogenic assays (Matrigel® plugs, CAM). DCE-MRI, FMT, and IHC were used to complete a tumor longitudinal follow-up and obtain morphological and functional vascular information. In vitro, HUVECs’ angiogenic potential was evaluated using a tumor secretome, subjected to a proteomic analysis to highlight LRP-1-dependant signaling pathways. Results: LRP-1 repression in MDA-MB-231 tumors led to a 60% growth delay because of, inter alia, morphological and functional vascular differences, confirmed by angiogenic models. In vitro, the LRP-1-repressed cells secretome restrained HUVECs’ angiogenic capabilities. A proteomics analysis revealed that LRP-1 supports tumor growth and angiogenesis by regulating TGF-β signaling and plasminogen/plasmin system. Conclusions: LRP-1, by its wide spectrum of interactions, emerges as an important matricellular player in the control of cancer-signaling events such as angiogenesis, by supporting tumor vascular morphology and functionality. more...

Abstract

The intensification of crop management have led to an impoverishment of the biological diversity, resulting in the loss of ecosystem services. In viticulture, flora could play an interesting role because it can compensate the negative impacts of vine monoculture. The flora biodiversity favours fauna biodiversity, sustaining especially the presence of pollinating insects in the grapevine inter row sown with grass or legumes and area around grapevine plots. These aspects have been poorly studied until now. A grapevine experiment set in Alsace at INRA Colmar in the frame of the DEPHY-EXPE PEPSVI project was used to address these questions. Between 2014 and 2016, floristic survey was carried out on 4 sites, within an area of 500 m2 within each plot. On those sites, different grapevine systems based on integrated, biological, or biodynamic production were tested. The “presence/absence” method implemented to characterize flora composition and richness on the grapevine row, the inter-row sown with grass, and the tilled inter-row. Results showed that Asteraceae, Fabaceae and Poaceae were the most represented flora families. Asteraceae and Fabaceae may provide pollination ecosystem services if they are well managed and allowed to flower. Moreover, Fabaceae family increases soil fertility by symbiotic fixation. The role of environmental factors, i.e., semi-natural area like hedge or forest edge close to vineyard, and anthropic factors, i.e., soil cover management, were also characterized and shown as impacted on flora composition and richness under grapevine rows and between inter- rows. Flora characterization will continue in 2017 and 2018 to validate the present results and to assess the evolution of flora composition and richness in different climatic conditions. more...

Abstract

International audience; Desialylation, governed by sialidases or neuraminidases, is strongly implicated in a wide range of human disorders, and accumulative data show that inhibition of neuraminidases, such as neuraminidases 1 sialidase, may be useful for managing atherosclerosis. Several studies have reported promising effects of oseltamivir phosphate, a widely used anti-influenza sialidase inhibitor, on human cancer cells, inflammation, and insulin resistance. In this study, we evaluated the effects of oseltamivir phosphate on atherosclerosis and thrombosis and potential liver toxicity in LDLR 2/2 mice fed with high-fat diet. Our results showed that oseltamivir phosphate significantly decreased plasma levels of LDL cholesterol and elastin fragmentation in aorta. However, no effect was observed on both atherosclerotic plaque size in aortic roots and chemically induced thrombosis in carotid arteries. Importantly, oseltamivir phosphate administration had adverse effects on the liver of mice and significantly increased messenger RNA expression levels of F4/80, interleukin-1b, transforming growth factor-b1, matrix metalloproteinase-12, and collagen. Taken together, our findings suggest that oseltamivir phosphate has limited benefits on atherosclero-sis and carotid thrombosis and may lead to adverse side effects on the liver with increased inflammation and fibrosis. more...

Abstract

Ce numéro est constitué d’articles de synthèse des projets DEPHY EXPE publiés à l’occasion du Colloque National DEPHY EXPE, qui s’est déroulé le 28 mai 2019 à l'Assemblée Permanente des Chambres d'Agriculture (Paris). Nous vous invitons à découvrir le bilan et les enseignements de 19 projets conduits sur la période 2013-2018.; PEPSVI research project (2013-2018) aims were to co-design, experiment and evaluate eleven new vine production systems. Vine productions systems were innovative because very low inputs were used. These systems were also under Integrated or Organic or Biodynamic certification. Systems innovations were (i) technics (i.e. using essential oils to reduce cupper use, 100% soil cover of the field) ; (ii) organizational (i.e. new decision rules to spray, Decision Aid tools) ; (iii) re-design (resistant grape varieties planted). Performances of the eleven new systems were evaluated about environment (Frenquency Treatment Index, Cupper rate), agronomy (yield, harvested berries quality, fungi damage, soil quality), social and economy. Results show that it is possible to have good performances of vineyard systems with a 0-89% -reduced TFI. TFI average reduction is 40% for all systems and studied years. Châtenois and Ingersheim are the most spectacular systems with a reduced TFI stabilized to 71%. 30. Genotyping by sequencing and genetic mapping of phenology-relevant and berry quality traits from two grapevine mapping populations

Abstract

The intensification of the vineyard practices led to an impoverishment of the biological diversity. In vineyard management, the reflection to recude pesticides uses concerns mainly the soil management of the vineyard, and often focuses on flora management in the inter-row. The goal of the present study is to gain more knowledge on the dynamic of vineyard flora, including relationships with environmental factors and soil practices. Assessment of floristic diversity was carried out for 5 fields of the research program PEPSVI in Alsace (France) on an area of 500 m2 within each of the fields. Soil management was either integrated or organic. Within each field, species richness was determined for the row (ur), the grassed inter-row (GIR), and the tilled inter-row (TIR) three times during each vine-growing season in 2014 and 2015. ANOVA tests were performed on data. First we observed an average of 54 diffecrent species in the fields per year and that there are no signifiicant differences between the different soil managements. The highest value belongs to organic soil management. The average species richness in organic fields is the highest in GIR (respectively 21 and 22 species in 2014 and 2015) and in UR (respectively 19 and 18 species in 2014 and 2015) and there is no significant difference between GIR and UR and between 2014 and 2015. The flora developed more considerably in the GIR (22 species) than UR (19 species) and less in TIR (16 species). The results of the study showed also that superficial tillage i.e. scraping or harrowing, helps flora emergence and increases species richness (21 species in average against 14 in average for the other soil managements). The environment has also to be taken into account. Surrounding vegetation of the field influences signigicantly the species richness, (30 more species in the year for the most favorable environment). Next steps of the study will be the analysis of distribution of flora families and Raunkiaer's life. more...
