2. Looking Towards Digital Equity and Inclusion: Exploring Perspectives and Barriers in School Districts

Abstract

In our increasingly digitized society, where technology use is ubiquitous, nearly all activities require access to and proficiency using digital tools. Proficient internet use may lead to the pursuit of higher education, launching a fulfilling career, or part of habits and healthy lifestyles where education is lifelong and life-wide. This study documented two school districts’ progress toward digital equity and inclusion. Promoting digital equity and inclusion in schools are important and complex issues that are not yet fully understood. Given federal legislation such as the National Educational Technology Plan and competitive grants tied to the Digital Equity Act, school districts and community based organizations are faced with developing and executing plans that consider the needs of vulnerable families, students, and communities. Through collaboration with school district personnel and openly discussing digital equity as a critical issue facing school districts nationwide, this research explored how two school districts are thinking about their digital equity and inclusion needs across their schools. Completed surveys captured multiple perspectives collected across the two school districts. Participants included district administrators, school principals, classroom teachers, and parents. Semi-structured interviews captured personal experiences from participants about digital equity and inclusion. Multiple barriers to digital equity and inclusion for students in schools, were documented in both surveys and interviews. Through a thematic analysis of the data collected, the study’s findings indicate a lack of coordinated efforts across schools within a district and classrooms, despite acknowledging the significance of digital equity. Through the analysis of data, the study yielded themes, cross-cutting concepts, and suggested actions aimed at lasting transformative change.

Abstract

In recent years, natural antisense transcripts (NATs) have been implicated in many aspects of eukaryotic gene expression including genomic imprinting, RNA interference, translational regulation, alternative splicing, X-inactivation and RNA editing. Moreover, there is growing evidence to suggest that antisense transcription might have a key role in a range of human diseases. Consequently, there have been several recent attempts to identify novel NATs. To date, ~2500 mammalian NATs have been found, indicating that antisense transcription might be a common mechanism of regulating gene expression in human cells. There are increasingly diverse ways in which antisense transcription can regulate gene expression and evidence for the involvement of NATs in human disease is emerging. A range of bioinformatic resources could be used to assist future antisense research.

20. Transport of IgA in rat salivary glands

Abstract

Transport of polymeric immunoglobulin A (plgA) in rat salivary glands has been investigated by combined morphological and biochemical techniques in vivo and in vitro. The distribution of IgA and its cellular receptor secretory component (SC) was observed by immunoperoxidase staining of cryosections from parotid and submaxillary gland, showing serous acinar cells are the site of IgA transport into saliva. Binding of horse radish peroxidase specific IgA to parotid serous acinar cells in vitro, observed by electron microscopy, shows that only the basolateral domain of acinar cells possesses exposed SC. A combination of new cell fractionation methods and standard western blotting techniques shows that SC present on basolateral plasma membrane of parotid acinar cells has a molecular weight (mwt) >100,000 and shows a high affinity for plgA in vitro. The existence of a 73,000 mwt SC occurring with plgA in cellular fractions of parotid gland suggest cleavage of SC occurs prior to secretion. The kinetics of plgA trancytosis was studied using isolated parotid acini. Bound plgA was secreted into the incubation medium as slgA, within thirty minutes of incubation at 37°C. Secretion of plgA was initially rapid but slowed over a 2hr period of incubation at 37°C. In addition to facilitating plgA transport serous acinar cells also synthesise and secrete a diverse range of other salivary proteins which are packaged into secretion granules and secreted directly through the apical plasma membrane. It is improbable that one complex secretory pathway facilitates both bulk secretion of salivary protein and transport of plgA. Therefore secreted proteins must be selectively segregated during secretion into saliva. Secretion of proteins from acinar cells in vitro shows proteins are released at two distinct rates.

Abstract

Background: Adult mental health in-patient units primarily provide a service for people deemed to be at significant risk to themselves or others, where treatment cannot be provided safely in the community. Whilst psychological interventions are indicated during episodes of acute mental distress, they are often psychoeducational and skills-based in nature. A common complaint amongst those admitted is the lack of psychological provision at a time of crisis when they most need to make sense of their difficulties Aims: This article reports on service users' experiences of open group cognitive behavioural therapy where participants choose the therapeutic targets. Method: A total of 75 patients admitted to acute in-patient wards over a 6-month period accessed open group cognitive therapy as part of routine care. Participants completed an evaluation questionnaire that measured their experiences of the group and the usefulness of them within an in-patient setting. Results: A total of 27 participants completed anonymous questionnaires (36%) and the results indicated that participants felt understood, respected and accepted within the group and felt that the group setting was helpful for sharing experiences. In addition, all participants reported that following the group they would be more likely to access psychological therapies in the future. Conclusions: Open group therapy where participants define the therapeutic targets each session is feasible and achievable on acute in-patient units and patients report finding this useful. Abstract

Abstract Background This paper describes an automated method for finding clusters of interconnected proteins in protein interaction networks and retrieving protein annotations associated with these clusters. Results Protein interaction graphs were separated into subgraphs of interconnected proteins, using the JUNG implementation of Girvan and Newman's Edge-Betweenness algorithm. Functions were sought for these subgraphs by detecting significant correlations with the distribution of Gene Ontology terms which had been used to annotate the proteins within each cluster. The method was implemented using freely available software (JUNG and the R statistical package). Protein clusters with significant correlations to functional annotations could be identified and included groups of proteins know to cooperate in cell metabolism. The method appears to be resilient against the presence of false positive interactions. Conclusion This method provides a useful tool for rapid screening of small to medium size protein interaction datasets.

Abstract

Trichinella spiralis is a muscle-specific parasitic worm that is uniquely intracellular. T. spiralis reprograms terminally differentiated skeletal muscle cells causing them to de-differentiate and re-enter the cell cycle, a process that cannot occur naturally in mammalian skeletal muscle cells, but one that holds great therapeutic potential. Although the host ubiquitin pathway is a common target for viruses and bacteria during infection, its role in parasite pathogenesis has been largely overlooked. Here we demonstrate that the secreted proteins of T. spiralis contain E2 Ub-conjugating and E3 Ub-ligase activity. The E2 activity is attributed to TsUBE2L3, a novel and conserved T. spiralis enzyme located in the secretory organ of the parasite during the muscle stages of infection. TsUBE2L3 cannot function with any T.spiralis secreted E3, but specifically binds to a panel of human RING E3 ligases, including the RBR E3 ARIH2 with which it interacts with a higher affinity than the mammalian ortholog UbcH7/UBE2L3. Expression of TsUBE2L3 in skeletal muscle cells causes a global downregulation in protein ubiquitination, most predominantly affecting motor, sarcomeric and extracellular matrix proteins, thus mediating their stabilization with regards to proteasomal degradation. This effect is not observed in the presence of the mammalian ortholog, suggesting functional divergence in the evolution of the parasite protein. These findings demonstrate the first example of host-parasite interactions via a parasite-derived Ub conjugating enzyme; an E2 that demonstrates a novel muscle protein stabilization function., Author Summary Parasitic worms often establish long-lasting infections in their hosts; tightly regulating their surroundings to strike a delicate balance between host cell modulation and protection that will ensure their replication. This is accomplished via the active secretion of parasite glycolipids and glycoproteins into the host. Trichinella spiralis, a parasitic nematode that infects skeletal muscle of mammals, birds and reptiles, is the only parasitic worm with a true intracellular stage. T. spiralis invade terminally differentiated myotubes, reprogramming them to de-differentiate and re-enter the cell cycle, a process that cannot occur naturally in mammalian skeletal muscle cells, but one that holds great therapeutic potential. We have identified and characterized a novel T. spiralis secreted protein that, despite a high level of sequence identity, appears to have evolved a different function to its host ortholog. This protein is an active Ub conjugating enzyme that binds to a panel of human E3 Ub ligases with higher affinity than the host ortholog. Furthermore, when expressed in skeletal muscle cells in culture, its presence uniquely leads to the stabilization of muscle-specific proteins via the downregulation of their ubiquitination.

Abstract

While climate variability is endemic to Southwest North America (SWNA), mounting evidence indicates the region is undergoing significant warming and becoming increasingly arid. Species are at or near their physiological limits at ecotone boundaries and are therefore particularly sensitive to climate change. Drought and warming associated tree mortality has been particularly acute in the semiarid forests and woodlands of the Jemez Mountains, New Mexico, USA, where ponderosa pine forests (Pinus ponderosa var. scopulorum) and piñon--juniper woodlands (Pinus edulis and Juniperus monosperma) have been subject to cambium-feeding pine beetle (Dendroctonus spp., Ips spp) attacks and increased wildfire activity and severity. Grazing and fire suppression have also impacted fire regimes leading to altered species composition and distribution. Projected warming and drought conditions in the 21st century will likely foster rapid (i.e. <5 year) ecotone shifts in semiarid ponderosa pine forests and piñon-juniper woodlands. This study analyzed ponderosa pine ecotone characteristics within a 2100 to 2200 m. range of the Vallecita watershed of the Jemez Mountains. Identification of sample sites was accomplished using remote sensing Landsat imagery, a moderate resolution earth observation data, coupled with a Geographic Information System (GIS) embedded semi--automated land cover classification method for raster-based analysis. Field procedures devised for this study determined past vegetation elements, current vegetation structure and composition, and present successional trajectory. The results of this research established baseline conditions and suggest the study area is undergoing a climate generated compositional shift from ponderosa pine dominated sites to piñon-juniper woodlands.
