The green synthesis of silver nanoparticles offers a non-toxic, eco-friendly, economical, and efficient solution against various microorganisms. The study aimed to investigate the utilization of cocoa bean shells for the production of silver nanoparticles through green synthesis, besides its characterization and evaluation against foodborne pathogens. To this end, total phenolic compounds, individual bioactive compounds, and antioxidant activity were evaluated in the aqueous extract of the cocoa bean shell. In addition, the size and stability of the silver nanoparticles were investigated (zeta potential and dynamic light scattering), morphology (scanning electron microscopy), chemical composition (mid-infrared spectroscopy), and their effectiveness against Escherichia coli (ATCC 25922), Listeria monocytogenes (ATCC 19117), Staphylococcus aureus (ATCC 22923), Salmonella enteritidis (S64), Cronobacter sakazakii (ATCC 29004), and Klebsiella pneumoniae (ATCC 18883). The extract was rich in reducing agents and efficiently produced AgNPs with high surface area (74.51 ± 0.38 nm) and stability (− 17.97 ± 0.12 mV). The nanoparticles were also found to be effective against all tested pathogens, demonstrating the potential of agro-industrial waste to produce AgNPs for various industrial applications. This study evaluated the capability of a thymol nanoemulsion (nTHY) to reduce cross-contamination with Salmonella Enteritidis in ground chicken meat. First, an nTHY (~ 54 nm) was prepared in a high-shear speed system (rotor–stator) and proved stable, remaining below 100 nm for 30 days in storage at 4 °C. Then, a minimum inhibitory concentration assay was performed by combining S. Enteritidis with viable native bacteria from chicken meat. nTHY presented half the MIC that was observed for its non-nanoemulsified versions for Salmonella Enteritidis (MIC = 0.31 mg/mL) and combined with native chicken bacteria (MIC = 0.625 mg/mL). A chitosan (CH) coating containing nTHY (6 mg/mL), equivalent to 10 × MIC (S. Enteritidis + native chicken bacteria), was prepared for in situ evaluation. Two groups of distinct chicken fillets were prepared for cross-contamination during grinding: chicken fillets experimentally contaminated with S. Enteritidis (~ 6.5 log CFU/g) and chicken fillets without S. Enteritidis but with the applied treatments (control, coated with CH, nTHY, and CH + nTHY). nTHY showed a bacteriostatic effect throughout the 7 days of storage. The CH + nTHY coating was the best treatment applied, reducing cross-contamination by up to 1.91 log CFU/g compared to uncoated samples. In mammals, enteric salmonellas can use tetrathionate (ttr), formed as a by-product from the inflammatory process in the intestine, as electron acceptor in anaerobic respiration, and it can fuel its energy metabolism by degrading the microbial fermentation product 1,2-propanediol. However, recent studies have shown that this mechanism is not important for Salmonella infection in the intestine of poultry, while it prolongs the persistence of Salmonella at systemic sites in this species. In the current study, we show that ΔttrApduA strains of Salmonella enterica have lower net survival within chicken-derived HD-11 macrophages, as CFU was only 2.3% (S. Enteritidis ΔttrApduA), 2.3% (S. Heidelberg ΔttrApduA), and 3.0% (S. Typhimurium ΔttrApduA) compared to wild-type strains after 24 h inside HD-11 macrophage cells. The difference was not related to increased lysis of macrophages, and deletion of ttrA and pduA did not impair the ability of the strains to grow anaerobically. Further studies are indicated to determine the reason why Salmonella ΔttrApduA strains survive less well inside macrophage cell lines. [ABSTRACT FROM AUTHOR]

Food‐related stresses such as heating and freezing may influence the biofilm formation ability of bacteria. This study aimed to investigate the main and interactive effects of food‐related stresses on the biofilm formation potential of Salmonella strains isolated from meat. Salmonella enteritidis, Salmonella typhi, and Salmonella typhimurium were subjected to osmotic, acid, heat, cold, and freezing stresses. The colorimetric microtiter plate method was used to measure the biofilm formation ability as a response to the stresses. Among the main effects, freezing time had the most significant effect on the biofilm formation responses of three Salmonella serovars. Freezing reduced the biofilm formation ability of Salmonella typhimurium and Salmonella enteritidis but increased that of Salmonella typhi (p < 0.05). Among the interaction effects, the most significant effect on the biofilm formation response of Salmonella typhi was the interaction between pH and heat, which had a negative effect on the biofilm formation response. In contrast, the interaction between osmotic pressure and cold stresses was the most significant interactive effect on the biofilm formation responses of Salmonella enteritidis and Salmonella typhimurium, which had an increasing effect. This study concluded that the food‐related stresses could change the biofilm formation capacity of Salmonella serotypes, and each serotype might show different biofilm formation abilities in response to different stresses. Phytochemicals-rich food-based botanicals including traditional or under-utilized plant-based ingredients can serve a dual functional role to help counter food contamination of bacterial origin, while also addressing the rise of diet-linked non-communicable chronic diseases (NCDs) such as type 2 diabetes, chronic hypertension and the associated oxidative stress. Hence the screening of these food-based botanicals for their phenolic content and profile, as well as antimicrobial, antioxidant, anti-hyperglycemic and anti-hypertensive properties has relevant merit. Using in vitro assay models, hot water extracts of different forms (slice, pickle, or powder) of amla (Phyllanthus emblica), clove (Syzygium aromaticum), kokum (Garcinia indica), and garlic (Allium sativum) were analyzed for their total soluble phenolic content (TSP) and phenolic profile as well as antimicrobial activity against strains of Salmonella Enteritidis, Listeria monocytogenes, and Escherichia coli that are associated with food-borne disease outbreaks. In addition, the antioxidant, anti-hyperglycemic and anti-hypertensive activity of the extracts were also determined using in vitro assay models, with the goal of establishing a dual functional role of the food safety and health protective benefits of these botanicals. A high baseline TSP content was observed in all the extracts and the major phenolic phytochemicals detected were gallic, cinnamic, ellagic, benzoic, dihydroxybenzoic, protocatechuic, and p-coumaric acid along with catechin and rutin. All extracts displayed significant antimicrobial activity against most of the bacterial strains tested and the antimicrobial activity was specific for each strain targeted in this study. Furthermore, significant antioxidant, anti-hyperglycemic and antihypertensive activity were observed among the botanical extracts, especially among the amla and kokum extracts. These results indicate that phytochemicals enriched botanicals, including amla and kokum, can be integrated into modern-day food preservation and dietary support strategies aimed at improving the food safety and health protective benefits of the food matrix. Invasive non-typhoidal Salmonella (iNTS) disease is an under-recognized high-burden disease causing major health and socioeconomic issues in sub-Saharan Africa (sSA), predominantly among immune-naïve infants and young children, including those with recognized comorbidities such as HIV infection. iNTS disease is primarily caused by Salmonella enterica serovar Typhimurium sequence type (ST) 313 and 'African-restricted clades' of Salmonella Enteritidis ST11 that have emerged across the African continent as a series of epidemics associated with acquisition of new antimicrobial resistance. Due to genotypes with a high prevalence of antimicrobial resistance and scarcity of therapeutic options, these NTS serovars are designated by the World Health Organization as a priority pathogen for research and development of interventions, including vaccines, to address and reduce NTS associated bacteremia and meningitis in sSA. Novel and traditional vaccine technologies are being applied to develop vaccines against iNTS disease, and the results of the first clinical trials in the infant target population should become available in the near future. The "Vaccine Value Profile" (VVP) addresses information related predominantly to invasive disease caused by Salmonella Enteritidis and Salmonella Typhimurium prevalent in sSA. Information is included on stand-alone iNTS disease candidate vaccines and candidate vaccines targeting iNTS disease combined with another invasive serotype, Salmonella Typhi, that is also common across sSA. Out of scope for the first version of this VVP is a wider discussion on either diarrheagenic NTS disease (dNTS) also associated with Salmonella Enteritidis and Salmonella Typhimurium or the development of a multivalent Salmonella vaccines targeting key serovars for use globally. This VVP for vaccines to prevent iNTS disease is intended to provide a high-level, holistic assessment of the information and data that are currently available to inform the potential public health, economic, and societal value of pipeline vaccines and vaccine-like products. Future versions of this VVP will be updated to reflect ongoing activities such as vaccine development strategies and a "Full Vaccine Value Assessment" that will inform the value proposition of an iNTS disease vaccine. This VVP was developed by a working group of subject matter experts from academia, non-profit organizations, public private partnerships, and multi-lateral organizations, and in collaboration with stakeholders from the World Health Organization African Region. All contributors have extensive expertise on various elements of the iNTS disease VVP and collectively aimed to identify current research and knowledge gaps. Background: European hedgehogs (Erinaceus europaeus) are widely distributed across Europe. They may play an important role by spreading zoonotic bacteria in the environment and to humans and animals. The aim of our work was to study the prevalence and characteristics of the most important foodborne bacterial pathogens in wild hedgehogs. Results: Faecal samples from 148 hospitalised wild hedgehogs originating from the Helsinki region in southern Finland were studied. Foodborne pathogens were detected in 60% of the hedgehogs by PCR. Listeria (26%) and STEC (26%) were the most common foodborne pathogens. Salmonella, Yersinia, and Campylobacter were detected in 18%, 16%, and 7% of hedgehogs, respectively. Salmonella and Yersinia were highly susceptible to the tested antimicrobials. Salmonella Enteritidis and Listeria monocytogenes 2a were the most common types found in hedgehogs. All S. Enteritidis belonged to one sequence type (ST11), forming four clusters of closely related isolates. L. monocytogenes was genetically more diverse than Salmonella, belonging to 11 STs. C. jejuni ST45 and ST677, Y. pseudotuberculosis O:1 of ST9 and ST42, and Y. enterocolitica O:9 of ST139 were also found. Conclusions: Our study shows that wild European hedgehogs should be considered an important source of foodborne pathogens, and appropriate hygiene measures after any contact with hedgehogs and strict biosecurity around farms are therefore important. The purpose of this study was to identify the saponin and phenolic components in root extracts of Saponaria officinalis, a widespread species, found in Cyprus. A total of six major saponins, including gypsogenin and gypsogenic acid derivatives, as well as saponariosides C, D, and E, were identified using UHPLC/Q-TOF-MS analysis, with gypsogenin derivatives being the most common saponins detected through quantitative analysis. A total of six phenolic compounds were also identified, including rutin, quercetin galactoside, syringic acid, apigenin, protocatechuic, and vanillic acid. In addition to their saponin and phenolic contents, the root extracts were prepared through different extraction methods, and their biological activity was assessed. All samples demonstrated antioxidant capacity, as well as antibacterial activity, against four bacterial strains (Escherichia coli, Staphylococcus aureus, Enterococcus faecalis, and Salmonella enteritidis), with the acetone extract presenting higher susceptibility. The evaluation of anticancer activity in A375 (human malignant melanoma), HeLa (human cervical epithelioid carcinoma), and HaCaT (healthy human keratinocytes) cell lines revealed that the acetone extract of S. officinalis extract demonstrated a significant inhibitory effect on the proliferation of A375 cells in a concentration-dependent manner. None of the extracts demonstrated anti-neurotoxic potential against Aβ25–35 cytotoxic peptides. The results of this study support previous findings that reveal that the Saponaria species are an excellent natural source of biologically active compounds with antioxidant, antimicrobial, and anticancer properties. The escalating incidence of foodborne salmonellosis poses a significant global threat to food safety and public health. As antibiotic resistance in Salmonella continues to rise, there is growing interest in bacteriophages as potential alternatives. In this study, we isolated, characterized, and evaluated the biocontrol efficacy of lytic phage L223 in chicken meat. Phage L223 demonstrated robust stability across a broad range of temperatures (20–70 °C) and pH levels (2–11) and exhibited a restricted host range targeting Salmonella spp., notably Salmonella Typhimurium and Salmonella Enteritidis. Characterization of L223 revealed a short latent period of 30 min and a substantial burst size of 515 PFU/cell. Genomic analysis classified L223 within the Caudoviricetes class, Guernseyvirinae subfamily and Jerseyvirus genus, with a dsDNA genome size of 44,321 bp and 47.9% GC content, featuring 72 coding sequences devoid of antimicrobial resistance, virulence factors, toxins, and tRNA genes. Application of L223 significantly (p < 0.005) reduced Salmonella Typhimurium ATCC 14,028 counts by 1.24, 2.17, and 1.55 log CFU/piece after 2, 4, and 6 h of incubation, respectively, in experimentally contaminated chicken breast samples. These findings highlight the potential of Salmonella phage L223 as a promising biocontrol agent for mitigating Salmonella contamination in food products, emphasizing its relevance for enhancing food safety protocols. Summary: This study delved into the antimicrobial potential of hydrolysates from adzuki bean and mung bean protein concentrates, created using Alcalase and Flavourzyme enzymes. Targeting common foodborne pathogens (Salmonella Typhimurium, Salmonella Enteritidis, Vibrio parahaemolyticus, Staphylococcus aureus, and Bacillus subtilis) and Escherichia coli, the research underscored the crucial role of peptidase in generating antibacterial peptides. Fractions F2 of Flavourzyme hydrolysed adzuki bean (ABF) and F4 of Flavourzyme hydrolysed mung bean (MBF) showed the highest antibacterial activity against S. Typhimurium and S. aureus, respectively, whereas F2 of Alcalase hydrolysed mung bean (MBA) exhibited very poor activity against S. aureus. TripleTOF LC–MS/MS analysis revealed small cationic antibacterial peptides, with lysine being prominent against S. aureus and proline–proline against S. Typhimurium. These findings highlighted the significant contribution of protease enzymes in fostering antibacterial peptide production, offering valuable insights for developing effective antimicrobial agents in the context of food safety. Fallopia japonica (FJ), an invasive plant species known for its rich bioactive compounds, has been used for centuries in traditional Chinese medicine. Despite its significant beekeeping potential, this aspect of FJ remains underexplored. This research aims to investigate the antimicrobial and antibiofilm properties of FJ plants and honey. Notably, this study is the first to identify individual phenolic compounds in both FJ plant tissues and FJ honey, highlighting resveratrol as a marker of FJ honey. The study tested inhibitory activity against seven bacterial strains: Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, Bacillus cereus, Salmonella enteritidis, and the yeast Candida albicans. Disk diffusion and microdilution methods were used to assess antimicrobial activity, while the crystal violet staining test evaluated antibiofilm activity. Results showed that FJ plant tissues and honey exhibited strong inhibition, particularly against Gram-negative bacterial strains. The most significant inhibition of biofilm formation, by both FJ plant tissues and honey, was observed against Staphylococcus aureus and Escherichia coli. A significant positive correlation was found between antimicrobial activity and individual polyphenols, especially resveratrol. The antibacterial and antibiofilm potential of FJ plant tissues and honey suggests promising applications in sustainable beekeeping. At present, there are no data in the scientific literature on studies aimed at characterizing Passiflora caerulea L. growing in Bulgaria. The present study aimed to investigate the metabolic profile and elemental composition of the leaves and pulp of this Passiflora, as well as to evaluate the antioxidant, antimicrobial and anti-inflammatory activities of its leaf and pulp extracts. The results showed that the pulp predominantly contained the essential amino acid histidine (7.81 mg g−1), while it was absent in the leaves, with the highest concentration being tryptophan (8.30 mg g−1). Of the fatty acids, palmitoleic acid predominated both in the pulp and in the leaves. A major sterol component was β-sitosterol. Fructose (7.50%) was the predominant sugar in the pulp, while for the leaves, it was glucose—1.51%. Seven elements were identified: sodium, potassium, iron, magnesium, manganese, copper and zinc. The highest concentrations of K and Mg were in the pulp (23,946 mg kg−1 and 1890 mg kg−1) and leaves (36,179 mg kg−1 and 5064 mg kg−1). According to the DPPH, FRAP and CUPRAC methods, the highest values for antioxidant activity were found in 70% ethanolic extracts of the leaves, while for the ABTS method, the highest value was found in 50% ethanolic extracts. In the pulp, for all four methods, the highest values were determined at 50% ethanolic extracts. Regarding the antibacterial activity, the 50% ethanolic leaf extracts were more effective against the Gram-positive bacteria. At the same time, the 70% ethanolic leaf extract was more effective against Gram-negative bacteria such as Salmonella enteritidis ATCC 13076. The leaf extracts exhibited higher anti-inflammatory activity than the extracts prepared from the pulp. The obtained results revealed that P. caerulea is a plant that can be successfully applied as an active ingredient in various nutritional supplements or cosmetic products. [ABSTRACT FROM AUTHOR]

Glycoconjugation is a well-established technology for vaccine development: linkage of the polysaccharide (PS) antigen to an appropriate carrier protein overcomes the limitations of PS T-independent antigens, making them effective in infants and providing immunological memory. Glycoconjugate vaccines have been successful in reducing the burden of different diseases globally. However, many pathogens still require a vaccine, and many of them display a variety of glycans on their surface that have been proposed as key antigens for the development of high-valency glycoconjugate vaccines. CDAP chemistry represents a generic conjugation strategy that is easily applied to PS with different structures. This chemistry utilizes common groups to a large range of PS and proteins, e.g., hydroxyl groups on the PS and amino groups on the protein. Here, new fast analytical tools to study CDAP reaction have been developed, and reaction conditions for PS activation and conjugation have been extensively investigated. Mathematical models have been built to identify reaction conditions to generate conjugates with wanted characteristics and successfully applied to a large number of bacterial PSs from different pathogens, e.g., Klebsiella pneumoniae, Salmonella Paratyphi A, Salmonella Enteritidis, Salmonella Typhimurium, Shighella sonnei and Shigella flexneri. Furthermore, using Salmonella Paratyphi A O-antigen and CRM197 as models, a design of experiment approach has been used to study the impact of conjugation conditions and conjugate features on immunogenicity in rabbits. Salmonella enterica serovar Enteritidis (S. Enteritidis) is the most predominant serovar in poultry and eggs, and it poses a significant threat to public health worldwide as it is a leading cause of salmonellosis in humans, which is transmitted through the consumption of contaminated poultry products like eggs. The objective of this study was to evaluate the presence of S. Enteritidis in household chicken eggs collected from Erzurum Province, Türkiye. A total of 168 household chicken eggs were collected from 168 small family poultry farms. Salmonella spp. was not isolated from any of the examined egg internal contents; however, successful isolation and identification of Salmonella spp. were achieved in 4 of the shell samples. Further characterization of the identified Salmonella spp. isolates was performed at the serovar level using 16S rDNA sequence analysis, and all 4 isolates were identified as S. Enteritidis. In conclusion, this study highlights the inherent risk of S. Enteritidis contamination in household chicken eggs and emphasizes the vital significance of implementing stringent food safety measures to safeguard consumer well-being and ensure the protection of public health.

Abstract Salmonella Enteritidis is a foodborne enteric pathogen that infects humans and animals, and bacterial small RNAs (sRNAs) have been found to play critical roles in the virulence of Salmonella. However, the specific mechanism by which sRNAs contribute to the host–pathogen interaction in the intestine remains unclear. In this study, a combination of transcriptomic and proteomic analysis was used to investigate the effect of a particular sRNA known as Salmonella adhesive‐associated sRNA (SaaS) on the intestinal damage induced by Salmonella. The results revealed that SaaS sRNA could suppress the host's inflammatory response through various gene sets, mainly including tumor necrosis factor (TNF)‐α signaling via NF‐κB, IL‐6 JAK‐STAT3 signaling, and interferon gamma responses. This suppressive effect was further confirmed by ELISA analysis, which showed the lowest levels of TNF‐α, IL‐6, and interferon gamma (IFN‐γ). Meanwhile, the work has identified several crucial factors, peptidoglycan recognition protein short, transferrin receptor protein 1, and corticosteroid 11‐beta‐dehydrogenase isozyme 2, that are specifically targeted by SaaS to regulate inflammatory response, neutrophils activity, and diseases. Furthermore, combining hematologic analysis and correlation analysis, this study demonstrated that SaaS led to a decrease in the percentage of neutrophils and revealed a strong correlation between the three crucial factors and various markers of inflammation and intestinal barrier function. These findings provide a comprehensive understanding of how SaaS sRNA modulates Salmonella invasion in the animal intestine and further present a strategy on controlling foodborne Salmonella based on the new targets, sRNAs.

Abstract Phage therapy holds promise as an alternative to antibiotics for combating multidrug-resistant bacteria. However, host bacteria can quickly produce progeny that are resistant to phage infection. In this study, we investigated the mechanisms of bacterial resistance to phage infection. We found that Rsm1, a mutant strain of Salmonella enteritidis (S. enteritidis) sm140, exhibited resistance to phage Psm140, which was originally capable of lysing its host at sm140. Whole genome sequencing analysis revealed a single nucleotide mutation at position 520 (C → T) in the rfbD gene of Rsm1, resulting in broken lipopolysaccharides (LPS), which is caused by the replacement of CAG coding glutamine with a stop codon TAG. The knockout of rfbD in the sm140ΔrfbD strain caused a subsequent loss of sensitivity toward phages. Furthermore, the reintroduction of rfbD in Rsm1 restored phage sensitivity. Moreover, polymerase chain reaction (PCR) amplification of rfbD in 25 resistant strains revealed a high percentage mutation rate of 64% within the rfbD locus. We assessed the fitness of four bacteria strains and found that the acquisition of phage resistance resulted in slower bacterial growth, faster sedimentation velocity, and increased environmental sensitivity (pH, temperature, and antibiotic sensitivity). In short, bacteria mutants lose some of their abilities while gaining resistance to phage infection, which may be a general survival strategy of bacteria against phages. This study is the first to report phage resistance caused by rfbD mutation, providing a new perspective for the research on phage therapy and drug-resistant mechanisms.

Salmonellosis represents a significant economic and public health concern for the poultry industry in Africa, leading to substantial economic losses due to mortality, reduced productivity, and food safety problems. However, comprehensive information on the burden of poultry salmonellosis at the continental level is scarce. To address this gap, a systematic review and meta-analysis were conducted to consolidate information on the prevalence and circulating serotypes of poultry salmonellosis in African countries. This involved the selection and review of 130 articles published between 1984 and 2021. A detailed systematic review protocol was structured according to Cochrane STROBE and PRISMA statement guideline. From the 130 selected articles from 23 different African countries, the overall pooled prevalence estimate (PPE) of poultry salmonellosis in Africa was found to be 14.4% (95% CI = 0.145–0.151). Cameroon reported the highest PPE at 71.9%. The PPE was notably high in meat and meat products at 23%. The number of research papers reporting poultry salmonellosis in Africa has shown a threefold increase from 1984 to 2021. Salmonella Enteritidis and Typhimurium were the two most prevalent serotypes reported in 18 African countries. Besides, Salmonella Kentucky, Virchow, Gallinarum, and Pullorum were also widely reported. Western Africa had the highest diversity of reported Salmonella serotypes (141), in contrast to southern Africa, which reported only 27 different serotypes. In conclusion, poultry salmonellosis is highly prevalent across Africa, with a variety of known serotypes circulating throughout the continent. Consequently, it is crucial to implement strategic plans for the prevention and control of Salmonella in Africa. RESEARCH HIGHLIGHTS The pooled sample prevalence of poultry salmonellosis in Africa is high (14.4%). The highest PPE was recorded in meat and meat products. The analysis included serovars, antibiotic resistance genes (ARGs), virulence factors (VFs), sequence types (STs), plasmid replicons, and genetic relationships. We also assessed pathogenicity of the isolates causing BSIs through growth, biofilm formation, and anti-serum killing assays.Results: WGS analysis identified 13 Salmonella serovars, with four responsible for BSIs. S. Enteritidis was the most prevalent serovar, involved in 19 (50.0%) cases. BSIs were caused by 17S. Enteritidis, two S. Typhimurium, two S. Munster and one S. Diguel. Of the 38 isolates, 27 (71.1%) exhibited high resistance to ampicillin, and 24 (63.2%) to ampicillin/sulbactam. Thirty-six types of ARGs were identified, with blaTEM-1B (n = 25, 65.8%) being the most frequent. Ten plasmid replicons were found; the combination of IncFIB(S)-IncFII(S)-IncX1 was the most common in S. Enteritidis (94.7%). Fifteen STs were identified, among which ST11 was the most prevalent and clonally disseminated, primarily responsible for BSIs. A total of 333 different VFs were detected, 177 of which were common across all strains. No significant differences were observed between the BSI and non-BSI isolates in terms of resistance rates, ARGs, plasmid replicons, and VFs, except for seven VFs. No strong pathogenicity was observed in the BSI-causing isolates.Conclusion: BSIs were predominantly caused by clonally disseminated S. Enteritidis ST11, the majority of which carried multiple ARGs, VFs and plasmid replicons. This study provides the first data on clonally disseminated S. Vaginitis, a prevalent gynecological condition in women, is mainly caused by an imbalance in the vaginal micro-ecology. The two most common types of vaginitis are vaginal bacteriosis and vulvovaginal candidiasis, triggered by the virulent Gardnerella vaginalis and Candida albicans, respectively. In this study, a strain capable of inhibiting G. vaginalis and C. albicans was screened from vaginal secretions and identified as Lactobacillus gasseri based on 16S rRNA sequences. The strain, named L. gasseri VHProbi E09, could inhibit the growth of G. vaginalis and C. albicans under co-culture conditions by 99.07% ± 0.26% and 99.95% ± 0.01%, respectively. In addition, it could significantly inhibit the adhesion of these pathogens to vaginal epithelial cells. The strain further showed the ability to inhibit the enteropathogenic bacteria Escherichia coli and Salmonella enteritidis, to tolerate artificial gastric and intestinal fluids and to adhere to intestinal Caco-2 cells. These results suggest that L. gasseri VHProbi E09 holds promise for clinical trials and animal studies whether administered orally or directly into the vagina. Whole-genome analysis also revealed a genome consisting of 1752 genes for L. gasseri VHProbi E09, with subsequent analyses identifying seven genes related to adhesion and three genes related to bacteriocins. These adhesion- and bacteriocin-related genes provide a theoretical basis for understanding the mechanism of bacterial inhibition of the strain. The research conducted in this study suggests that L. gasseri VHProbi E09 may be considered as a potential probiotic, and further research can delve deeper into its efficacy as an agent which can restore a healthy vaginal ecosystem. Background: Salmonella enteritidis (S. enteritidis), a zoonotic pathogen with a broad host range, presents a substantial threat to global public health safety. Vaccination stands as an effective strategy for the prevention and control of S. enteritidis infection, highlighting an immediate clinical need for the creation of safe and efficient attenuated live vaccines. Methods: In this study, a S. enteritidis peptidoglycan-associated lipoprotein (pal) gene deletion strain (Δpal), was constructed. To assess its virulence, we conducted experiments on biofilm formation capability, motility, as well as cell and mouse infection. Subsequently, we evaluated the immune-protective effect of Δpal. Results: It was discovered that deletion of the pal gene reduced the biofilm formation capability and motility of S. enteritidis. Cell infection experiments revealed that the Δpal strain exhibited significantly decreased abilities in invasion, adhesion, and intracellular survival, with downregulation of virulence gene expression, including mgtC, invH, spvB, sipA, sipB, ssaV, csgA, and pipB. Mouse infection experiments showed that the LD50 of Δpal increased by 104 times, and its colonization ability in mouse tissue organs was significantly reduced. The results indicated that the pal gene severely affected the virulence of S. enteritidis. Further, immunogenicity evaluation of Δpal showed a significant enhancement in the lymphocyte transformation proliferation capability of immunized mice, producing high titers of specific IgG and IgA, suggesting that Δpal possesses good immunogenicity. Challenge protection tests demonstrated that the strain could provide 100% immune protection against wild-type strains in mice. Discussion: This study proves that the pal gene influences the virulence of S. enteritidis, and Δpal could serve as a candidate strain for attenuated live vaccines, laying the foundation for the development of attenuated live vaccines against Salmonella. Introduction: Salmonella is one of the most common causes of food-borne outbreaks and infection worldwide. Non-typhoid Salmonella (NTS) infections are common and remain a significant public health problem among important bacterial foodborne diseases. The current study aimed to establish the Non typhoid Salmonella infection and antimicrobial resistance status among selected patients at Morogoro Regional Referral Hospital (MRRH), Morogoro Region, Tanzania, to inform clinical care management and public health interventions. Methodology: A cross-sectional study was conducted using medical records and samples were collected from hospitalised and outpatients between October and December 2021. A total of 153 participants were enrolled in the study and 132 consented to being sampled. The collected samples were analysed using standard microbiological techniques. The isolates were subjected to molecular genotyping, where Polymerase Chain Reaction (PCR) was performed targeting the 16S rDNA gene. PCR products were then submitted for sequencing to establish phylogenetic relatedness. Results: The phylogenetic analysis identified two Salmonella serovars; Salmonella Enteritidis and Salmonella Typhimurium. The isolates were from four adults and seven children patients. The isolates were tested against six antimicrobial agents: tetracycline, trimethoprim, gentamycin, ciprofloxacin, ampicillin and cefotaxime. Further antimicrobial assays were performed by screening 10 antimicrobial resistance genes using PCR. Overall, the highest resistance was observed in ampicillin (100%), whereas the lowest resistance was recorded for ciprofloxacin and gentamicin (9.1%). In addition, four (36.4%) of the isolates were resistant to cefotaxime and three (27.3%) to tetracycline and trimethoprim. The isolates also exhibit the presence of resistance genes for sulfamethoxazole 1&2, tetracycline (tet) A&B, Beta-lactamaseCTXM, Beta-lactamaseTEM, Beta-lactamaseSHV, Gentamycine, Acra and acc3-1 in different occurrences. In conclusion, the overall prevalence of Salmonella species in Morogoro region was 8.3% (11/132) with Salmonella Enteritidis and Salmonella Typhimurium being the only serovars detected from adults and children stool samples. Conclusion: Our investigation showed that both children and adults had been exposed to Salmonella spp. However, the occurrence of NTS was higher in children (5.3% (7/132) compared to adults (3.0% (4/132). To stop zoonotic infections and the development of antimicrobial resistance in the community, this calls for Infection Prevention and Control (IPC) and stewardship programmes on rational use of antimicrobials in both health facilities and at the community level. Author summary: Typhoid fever is claimed to prevail in Morogoro region. Most patients with stomach pain and or diarrhoea go for a diagnosis of typhoid despite the existence of other etiological agents causing the same clinical signs. Sample culture and isolation of bacteria have been a gold-standard method for the detection of disease-causing agents. However, this approach is rarely made by most hospitals, with the exception of regional and district hospitals in the public sector and big hospitals in the private sector. They always rely on the widal test, which suffers from significant limitations in its sensitivity and specificity. It is, therefore, difficult to identify appropriate etiological agents and assess their susceptibility to antimicrobials. Because of that, the contribution of non-typhoid salmonellosis is not featured. The authors have assessed the occurrence of non-typhoid Salmonella in hospitalized and outpatients of all age groups at Morogoro Regional Hospital as a modal to establish its contribution to gastrointestinal infections in the region through both conventional and molecular approaches. It was observed that the infection rate was high in children, and the no-typhoid Salmonella serovars had considerable resistance to the commonly used antimicrobial. Salmonella enteritidis is a main pathogen responsible for sporadic outbreaks of gastroenteritis, and therefore is an important public health problem. This study investigated the drug resistance and genomic characteristics of S. enteritidis isolated from clinical and food sources in Huzhou, Zhejiang Province, China, from February 1, 2021, to December 30, 2023. In total, 43 S. enteritidis strains isolated during the study period were subjected to virulence gene, drug resistance gene, genetic correlation, antibiotic resistance, and multilocus sequence typing analyses. All 43 isolates were identified as ST11, and contained 108 virulence-related genes. Drug sensitivity analysis of the 43 isolates showed resistance rates of 100% to nalidixic acid and 90.70% to ampicillin and ampicillin/sulbactam. Multidrug resistance is a serious issue, with 81.40% of strains resistant to three or more antibacterial drugs. Genome sequencing indicated that S. enteritidis possessed 23 drug resistance genes, of which 14 were common to all 43 isolates. Phylogenetic analysis based on core genome single-nucleotide polymorphisms divided the 43 S. enteritidis strains into three clusters, with the 10 samples from an outbreak forming an independent branch located in cluster 3. Summary: Salmonella contamination of fish after harvest is a significant food safety risk. To control this risk, it was aimed to reduce Salmonella Enteritidis in sea bass by helium and air plasma. Considering that preserving the fresh properties of sea bass is important for industrial applicability, determining the sensory effects of plasma treatments was also aimed. Since air plasma would provide a cheaper and easier alternative to helium plasma, their effects were compared. Additionally, the in vitro effects of these treatments on Salmonella Enteritidis were also investigated. Air and helium plasma completely inhibited the initial load of S. Enteritidis (2.79 Log CFU) inoculated on agar surface after 10 min treatment. Then, the inoculated sea bass samples were exposed to air or helium plasma for durations ranging from 1 to 15 min. Air and helium plasma treatments for 10 min provided 0.90 log cfu g−1 and 1.13 Log CFU/g reductions, respectively, but longer treatments did not lead to further significant reductions. Sensory scores were all above 7 (good) when samples were treated with air or helium plasma for 1–10 min, but longer treatments resulted in lower scores. The reduction of S. Enteritidis and changes in sensory characteristics were interpreted together and it was concluded that 10 min of air or helium plasma provided maximum reduction without causing any significant sensory changes. Objectives: Lichens are complex symbiotic organisms that generate various bioactive compounds with significant therapeutic value. We investigated the chemical composition and bioactivity of the acetone extract of the Algerian lichen Physconia venusta (Ach.) poet. Materials and Methods: Phytochemical screening was performed using gas chromatography-mass spectrometry (GC-MS). The antibacterial activity was assessed against Escherichia coli, Pseudomonas aeruginosa, Salmonella enteritidis, Salmonella typhi, Staphylococcus aureus, Listeria monocytogenes, and Bacillus subtilis using an agar diffusion test with the determination of the minimal inhibition concentration (MIC), while the antioxidant activity was determined using different chemical methods (DPPH, ABTS, CUPRAC, reducing power, superoxide anion scavenging, ß-carotene bleaching, and metal chelate). In addition, cytotoxic activity was tested using Artemia salina (Brine shrimp) bioassay. Results: The studied extract exhibited intense antibacterial activity against E. coli and S. aureus with inhibition diameters of 28 ± 0.01 and 22 ± 0.01 mm, respectively, with a MIC value of 6.25 mg/mL and a selectivity index of 2.8. The obtained extract showed different antioxidant trends depending on the selected assay. GC-MS analysis revealed many secondary metabolites. The application of bacteriophages (BP) could be a possible method to control poultry salmonellosis. However, the sensitivity of field isolates to different phages can restrict their application in the poultry industry. In this survey, the sensitivity of 20 Salmonella field isolates, including 10 S. Typhimurium and 10 S. Enteritidis, to a bacteriophage cocktail was evaluated. The investigated BP cocktail had a promising lytic effect on all field isolates in this survey. The lytic activity of the BP cocktail was retained at different pH values (5.5–8) and temperatures (25°C and 37°C for 48 h; 55°C for 10 min). Physico-chemical resistance of the BP cocktail indicated that it has the potential to be used as a feed additive to control Salmonella contamination. The BP cocktail in feed contaminated with S. Enteritidis and S. Typhimurium showed a significant reduction effect on colonization of Salmonella in the liver, crop, spleen, and caeca of the broilers at 7, 14 and 21 days. Bacteriophage (BP) cocktail was partially resistant to different temperatures and pH values. The BP cocktail showed lytic effects on different Salmonella isolates. This study investigated the effectiveness of plasma‐activated water (PAW) and plasma‐activated hydrogen peroxide (PAHP) in reducing egg‐associated pathogens. The antimicrobial activity of these solutions against Salmonella Enteritidis, Campylobacter jejuni, and Staphylococcus aureus on eggs was evaluated at different plasma treatment durations. The results demonstrated that increasing the duration of plasma treatment enhanced the antimicrobial efficacy of both PAW and PAHP. The bacterial counts of the egg‐associated pathogens significantly decreased from 7.61, 7.59, and 7.54 log (CFU/egg) to 5.4 and 3.09, 5.36 and 3.11, and 5.08 and 3.73 log (CFU/egg) for PAW and PAHP, respectively. The characteristics of the plasma solutions, including electrical conductivity, pH, H2O2, NO3¯, and NO2¯, synergistically acted as antimicrobial agents in both PAW and PAHP treatments. The storage study revealed that PAHP treatment had no adverse effects on the eggs' pH, albumen and yolk color, Haugh unit, and yolk index. However, it did result in reduced eggshell strength and compromised cuticle integrity. Overall, this study demonstrates the successful application of PAW and PAHP in effectively inhibiting egg‐associated pathogens while preserving essential egg quality attributes. Further research is needed to optimize the treatment conditions and investigate the long‐term effects of PAW and PAHP on eggs in larger‐scale applications. SUMMARY The poultry-housing environment plays a significant role in the transmission and persistence of the egg-associated pathogen Salmonella Enteritidis in laying flocks. The commercial egg industry is in the midst of a transition toward cage-free housing, but the food safety ramifications of this shift are not yet certain. The present study assessed internal organ colonization by Salmonella Enteritidis in layer pullets reared in cage-free housing and infected at two different ages. Groups of 280 pullets were transferred from the rearing facility (at 9 wk of age in one trial and 15 wk in another) to a containment facility with four isolation rooms simulating commercial cage-free barns with perches and nest boxes (70 birds/room). Twenty-four pullets in each room were orally inoculated with Salmonella Enteritidis immediately after placement in the containment facility. At 1–2 wk postinoculation in each trial, samples of liver, spleen, and intestinal tract were collected from all birds in two rooms for bacteriologic culturing to detect Salmonella Enteritidis. At 21–22 wk of age, samples of spleen, ovary, and intestinal tract were similarly collected and tested from all birds in the remaining two rooms. Among samples collected at 1–2 wk postinoculation, Salmonella Enteritidis was isolated significantly more often from groups of pullets infected initially at 15 wk of age than from those infected at 9 wk (61% vs. 38% of livers, 59% vs. 31% of spleens, and 84% vs. 57% of intestines). Among samples collected at 21–22 wk of age, the frequency of recovery of Salmonella Enteritidis was again significantly greater in birds infected at 15 wk of age than in those infected at 9 wk (16% vs. 6% of spleens, 9% vs. 1% of ovaries, and 26% vs. 10% of intestines). These data suggest that Salmonella Enteritidis infections introduced into flocks during the later stages of pullet rearing have greater potential to persist into the early phase of egg production. The rodents play a significant role in the transmission of Salmonella between farms and regions. The present study aimed to compare the virulence of Salmonella enteritidis isolated from fecal samples of laying hens and murine within the same poultry house but different regions in Libya using Vivo-quantitative measurement of invasiveness (chicken intestinal loop model). A total of 540 cloacal swabs from laying hens (Hy-line brown chickens) aged 36 weeks and 200 batches of murine fecal pellets were collected from the same poultry house at Gaser Bin Gisher and Furnag regions in Libya. The samples were passed on pre-enrichment broth (Buffered Peptone Water) and enrichment broths (Rappaport Vassiliadis, Selenite broth, and tetrathionate), then the samples were cultured onto Xylose Lysine Deoxycholate agar, brilliant green agar, Salmonella Shigella agar, and Hektoen enteric agar. Single colonies were selected and stained by gram stain and tested biochemically using analytical profile index (API) 20 tests. Salmonella enteritidis was isolated from all the collected samples. The invasion of Salmonella enteritidis isolated from laying hens and murine feces was significantly higher in the anterior inoculation position compared to the posterior position of jejunum in both regions. The account of Salmonella enteritidis isolated from laying feces of hens and murine at Gaser Bengasher region was significantly higher than that isolated from the AlFurnge region. In the present study, the rodents act only as mechanical transmitters without affecting Salmonella invasiveness capacity. Furthermore, the invasion of Salmonella enteritidis depends on the inoculation position in the jejunum. Moreover, the invasiveness variation of Salmonella enteritidis isolated from the Gaser Bengasher and AlFurnge regions could be attributed to the presence of different Salmonella strains in the studied area. Salmonella enteritidis isolated from poultry and murine in the current study was sensitive to gentamicin, ciprofloxacin, and enrofloxacin and resistant to doxycycline, chloramphenicol, sulfafurazol, and ampicillin. Salmonella Enteritidis, Escherichia coli, and Campylobacter jejuni are among the most common foodborne pathogens worldwide, and poultry products are strongly associated with foodborne pathogen outbreaks. These pathogens are capable of producing biofilms on several surfaces used in the food processing industry, including polyethylene and stainless steel. However, studies on multi-species biofilms are rare. Therefore, this study aimed to develop predictive mathematical models to simulate the adhesion and removal of multispecies biofilms. All combinations of microorganisms resulted in biofilm formation with differences in bacterial counts. E. coli showed the greatest ability to adhere to both surfaces, followed by S. Enteritidis and C. jejuni. The incubation time and temperature did not influence adhesion. Biofilm removal was effective with citric acid and benzalkonium chloride but not with rhamnolipid. Among the generated models, 46 presented a significant coefficient of determination (R2), with the highest R2 being 0.88. In recent years, there has been an increasing interest in antibacterial biopolymeric films. Among the different approaches for tuning the release kinetic of antibacterial compounds, the use of natural fillers allows for this purpose while optimizing the processability and the mechanical properties of the products. In this work, the effect of three different concentrations of biochar (BC) was investigated on the morphological, rheological, mechanical, and thermal properties of Polybutylene adipate terephthalate/Carvacrol/BC (PBAT/CV/BC) ternary biocomposites. The films were fabricated by means of melt mixing and compression molding and compared to PBAT/BC samples. The carvacrol kinetic release was evaluated as a function of the BC concentration in the ternary system. Results highlighted that BC allows tuning the properties of PBAT and of PBAT/CV samples and modifying the release kinetic of CV. Finally, the antimicrobial analysis revealed that PBAT/CV films exhibited excellent antimicrobial properties against Escherichia coli, Listeria monocytogenes, Salmonella Enteritidis, and Staphylococcus aureus. In the present study, new series of benzoxazolin-2-one linked to a variety of hydrazones and azoles were synthesized and assessed for their antibacterial properties against different bacterial microorganisms. All the synthesized target compounds were characterized by 1H NMR, 13C NMR and IR spectroscopy, and elemental analysis as well. The antibacterial activity of the synthesized compounds was evaluated according to the bacteriostatic and bactericidal activity against the tested pathogen strains by determining the minimum inhibition (MIC) and minimum bactericidal (MBC) concentrations and MBC/MIC ratios. The MIC was evaluated by the broth dilution and the MBC was evaluated by plating methods. The in vitro analysis suggested that some compounds, namely, amide, 5-chlorobenzimidazole, hydrazones with a 3-chloro substitution on the additional phenyl ring, and hydrazones with 2-furyl and 5-nitro-2-furyl substituents, demonstrated wide antibacterial activity against Escherichia coli, Bacillus subtilis, Staphylococcus aureus, and Salmonella Enteritidis. The most sensitive strains appeared to be Gram-negative E. coli and Gram-positive B. subtilis, while S. aureus showed some resistance. The most resistant pathogen was found to be S. enteritidis. The remaining compounds demonstrated moderate to low antibacterial potential. Once a silo has been opened, the silage inside will face challenges such as aerobic deterioration, rot, and contamination. Biocontrol bacteria, as a kind of biological antiseptic, are highly effective and natural and are gaining increasing attention. This study aimed to screen a strain with anti-microbial activity against silage spoilage microorganisms and examine its effects on the fermentation quality, aerobic stability, in vitro digestion, and methane production of silage. Lactic acid bacteria, pathogenic and rot-causing microorganisms, were used as indicators to screen the strains for putrefactive silage. The bacteriostatic spectrum, growth performance, and tolerance to the silage environment of the strain were tested. A strain named D-2 was screened from rotten whole-plant corn silage and identified as Bacillus velezensis through physiological and biochemical tests as well as 16S rDNA sequencing. This study found that D-2 exhibits antibacterial effects on several microorganisms, including Escherichia coli, Staphylococcus aureus, Salmonella enteritidis, Aspergillus niger, Saccharomyces cerevisiae, Fusarium oxysporum, and Fusarium graminearum. However, it has no adverse effect on Lactobacillus reuteri, Enterococcus faecium, or Lactobacillus casei. D-2 can attain a stable stage within 10 h and withstand temperatures of up to 70 °C. Moreover, this study found that D-2 had a high survival rate of over 97% after 48 h in a lactic acid environment with pH 4. Freshly chopped whole-plant corn was inoculated without or with D-2 and ensiled for 60 days. The results show that D-2 inoculations increase the content of water-soluble carbohydrates, acetic acid, and propionic acid in the silage and decrease the number of yeasts and molds, the NH4+-N/TN ratio, and the pH. We also found that fermenting whole-plant corn with D-2 significantly increased the in vitro digestibility and the propionic acid content, while also significantly inhibiting methane production. After being exposed to air for 10 days, D-2 can still effectively reduce the total number of yeasts and molds, prevent the decrease in lactic acid bacteria, and inhibit the increase in the pH and NH4+-N/TN ratio of silage products. Overall, D-2 is resistant to pathogenic and rot-causing microorganisms, allowing for easy adaptation to silage production conditions. D-2 can effectively improve aerobic stability and reduce losses in the nutritional value of silage, indicating possible applications for the prevention of silage rot and methane production. Biodegradable chitosan-based films containing different concentrations of jatobá fruit peel extract (0.0%, 0.04%, 0.07%, 0.1% and 0.3%) were prepared and evaluated for their antioxidant and antimicrobial activities, physicochemical, morphological and thermal properties. The extract from jatoba fruit peel was obtained and incorporated into formulations of chitosan films. The films were characterized for thickness, moisture, solubility, water vapor permeability, optical properties, color and morphology. Also, thermogravimetric analysis, differential scanning calorimetry, Fourier transform infrared spectroscopy, in vitro antioxidant activity and antimicrobial activity were performed. The addition of extract at increasing concentrations increased the in vitro antioxidant activity by about 9–60 times compared with the film without extract. Films with lower extract concentrations were more effective at inhibiting nine bacterial strains, with emphasis on Escherichia coli (95.0% inhibition), Bacillus cereus (92.5% inhibition), Bacillus subtilis (96.5% inhibition), Pseudomonas aeruginosa (94.4% inhibition), Salmonella Typhimurium (90.4% inhibition) and Salmonella Enteritidis (93.9% inhibition). Addition of the extract improved the physical properties of the film, such as moisture, solubility, WVP and luminosity. The thermal stability of the films was slightly reduced with addition of extract at increasing concentrations. Films incorporating jatobá fruit peel extract showed potential utility as 'green' bioactive, biodegradable packaging, an alternative to packaging containing petroleum derivatives. This study explores the potential probiotic properties of yeasts isolated from various Chilean honeys, focusing on Ulmo, Quillay, and Mountain honeys. Six yeast strains were identified, including Zygosaccharomyces rouxii, Candida sp., Schizosaccharomyces pombe, Rhodosporidiobolus ruineniae, Clavispora lusitaniae, and Metschnikowia chrysoperlae. Phenotypic characterization involved assessing their fermentative performance, ethanol and hops resistance, and cross-resistance. Ethanol concentration emerged as a limiting factor in their fermentative performance. The probiotic potential of these yeasts was evaluated based on resistance to high temperatures, low pH, auto-aggregation capacity, survival in simulated in vitro digestion (INFOGEST method), and antimicrobial activity against pathogens like Escherichia coli, Staphylococcus aureus, and Salmonella enteritidis. Three yeasts, Zygosaccharomyces rouxii, Schizosaccharomyces pombe, and Metschnikowia chrysoperlae, exhibited potential probiotic characteristics by maintaining cell concentrations exceeding 106 CFU/mL after in vitro digestion. They demonstrated fermentative abilities and resistance to ethanol and hops, suggesting their potential as starter cultures in beer production. Despite revealing promising probiotic and technological aspects, further research is necessary to ascertain their viability in producing fermented foods. This study underscores the innovative potential of honey as a source for new probiotic microorganisms and highlights the need for comprehensive investigations into their practical applications in the food industry. This study aimed to obtain and characterize an oil-in-water nanoemulsion (NE) loaded with an in vitro optimized bactericidal essential oil blend of 50% oregano, 40% thyme, and 10% lemongrass and to evaluate its potential at three different concentrations (0.5%, 1%, and 2%) in the inactivation of Escherichia coli, Staphylococcus aureus, and Salmonella enterica serotype Enteritidis inoculated in rainbow trout fillets stored at 4 °C for 9 days. Regarding the NE, the nanometric size (<100 nm) with low polydispersion (0.17 ± 0.02) was successfully obtained through ultrasound at 2.09 W/cm2. Considering the three concentrations used, S. Enteritidis was the most susceptible. On the other hand, comparing the concentrations used, the NE at 2% showed better activity, reducing S. Enteritidis, E. coli, and S. aureus by 0.33, 0.20, and 0.73 log CFU/g, respectively, in the trout fillets. Infections cause significant complications in surgical cases involving fractures and implants. Biomaterials, particularly bioactive glass, are known for their biocompatibility and tissue regeneration potential. The integration of bactericidal properties with bioactive glasses could prove pivotal in preventing infections during bone and wound regeneration. The LZS (Li2O–ZrO2–Si2O) glass system, recognized for its potential for bone regeneration via scaffolds, is a candidate for further exploration of its antimicrobial capabilities. This study evaluated the antimicrobial properties of LZS bioactive glass and glass‐ceramic against various pathogens (Staphylococcus aureus, Escherichia coli, Candida albicans, Pseudomonas aeruginosa, and Salmonella enteritidis) using the spread‐plate method. The LZS glass exhibited antibacterial effects against all the tested microorganisms, with minimum bactericidal concentrations (MBC) varying between 50 and 150 mg/mL. The bactericidal effects of the biomaterials were attributed to ion release from the glass matrix, which increased the pH of the solution and effectively eliminated microorganisms. In addition, this study found no evidence of DNA damage, and the LZS systems exhibited no cytotoxic or genotoxic effects at the tested concentrations. The results showed that the investigated antibacterial glass and glass‐ceramic have potential applications in bone regeneration or wound healing, especially in scenarios where bacterial proliferation is a concern. Simple Summary: Salmonella enterica ser. Enteritidis (S. Enteritidis) is widely found in chickens and eggs and has the potential to induce human illness. Infection with Salmonella enteritidis triggers inflammatory responses in the gut and body, leading to decreased production performance of poultry. Sodium butyrate (SB) is an alternative antibiotic used as an acidifier that can be transformed into butyric acid (BA) in the avian alimentary canal. BA, a short-chain fatty acid, is often used extensively in poultry production against Salmonella. Since an unpleasant odor can affect feed intake, coated sodium butyrate (CSB) is more widely used. However, the effects of CSB on intestinal health in hens infected with S. Enteritidis have been less reported. Therefore, this study aimed to investigate the effects of long-term dietary supplementation with CSB on the intestinal health and colonization of cecum Salmonella from laying hens infected with Salmonella enteritidis. The obtained results revealed that long-term dietary CSB improved the gut morphology and alleviated intestinal injury and the colonization of cecum Salmonella of laying hens infected with S. Enteritidis. Moreover, long-term supplementation of CSB in laying hens resulted in a better intestinal barrier, which effectively protected the intestinal health of laying hens and reduced Salmonella challenges. Salmonella enterica ser. Enteritidis (S. Enteritidis) is widely found in chickens and eggs, and it can potentially induce human illness. The investigation in this study centers on the impacts of long-term dietary supplementation with coated sodium butyrate (CSB) on intestinal well-being and the colonization of cecum Salmonella in laying hens infected with S. Enteritidis. We segregated a total of 120 Lohmann laying hens aged 51 weeks into four treatment categories: 0 (CON), 300 (CSB1), 500 (CSB2), and 800 (CSB3) mg/kg of CSB, supplemented with CSB from the first day of the experiment. A 24-week observation process was carried out for each laying hen. The S. Enteritidis was orally administered to all chickens on the morning of the first and third days of week 22 of the trial. After the S. Enteritidis challenge, egg production decreased the most in the CON group. Compared to the CON group, the three doses of CSB significantly improved egg production after the S. Enteritidis challenge (PANOVA < 0.05). S. Enteritidis challenge increased plasma DAO activity, but CSB supplementation reduced plasma DAO activity (Plinear < 0.05). The S. Enteritidis challenge disrupted intestinal villi morphology; compared to the CON group, the three dosages of CSB resulted in an increase in villus height (VH) and the ratio of villus height to crypt depth (V/C) in the duodenum, jejunum, and ileum of infected laying hens (Plinear < 0.05), with a significant increase in jejunal villus height (PANOVA < 0.05). A decrease in ileal crypt depth was also observed (Plinear < 0.05). CSB2 and CSB3 markedly increased the content of butyric acid in the cecum (PANOVA < 0.05). Additionally, in contrast to those in the CON group, the propionic acid content in the CSB supplementation group increased (Plinear < 0.05). Compared with those in the CON group, mRNA relative expression of the IL-6 and IL-1β in jejunum (Plinear < 0.05) and mRNA relative expression of the IL-1β in ileum (PANOVA < 0.05) were significantly lower, and mRNA relative expression of the IL-10 in ileum (Plinear < 0.05) were significantly higher in the CSB group. In addition, in contrast to the CON group, the CSB supplementation group significantly upregulated mRNA relative expression of the ZO-1 and CLDN1 (PANOVA < 0.05). Additionally, CSB supplementation reduced the number of Salmonella and increased the number of Lactobacilli in the cecum (Plinear < 0.05) and tended to increase the total bacteria count (Plinear = 0.069) and reduce the E. coli count (Plinear = 0.081). BACKGROUND: The shortcomings of synthetic pesticides are becoming increasingly evident. In the pursuit of looking for alternative, environmentally friendly pesticides, the potential use of two naturally occurring sesquiterpenes, α‐bisabolol and bisabolol oxide A, as bactericides or acaricides, was investigated against three gram‐positive and three gram‐negative bacteria, and the eggs and larvae of the cattle tick Hyalomma scupense, respectively. The primary targeted site of synthetic insecticides is acetylcholinesterase (AChE), so the inhibitory activity of both sesquiterpenes was evaluated. Then, molecular docking and molecular mechanics generalized Born surface area (MM‐GBSA) studies were performed to gain insight into their binding mode in the AChE active site. RESULTS: Bisabolol oxide A showed good activity against Staphylococcus aureus, Escherichia coli, and Salmonella enteritidis with inhibition zones ranging from 16 to 34 mm and minimal inhibitory concentration (CIM) of 3.125 mg mL−1. Ovicidal and larvicidal assays revealed promising activity of α‐bisabolol and bisabolol oxide A against H. scupense, with bisabolol oxide A being more effective against larval ticks with lethal concentration (LC50) value of 0.78%. Both compounds also displayed potent anti‐AChE activity with inhibition concentration (IC50) values of 37.09 and 28.14 μg mL−1, respectively. Furthermore, α‐bisabolol and bisabolol oxide A exhibited good and comparable docking scores (−7.289 and −7.139 Kcal mol−1, respectively) and were found to accommodate in the active‐site gorge of AChE via hydrogen bonding and hydrophobic interactions. CONCLUSION: Bisabolol oxide A and, to a lesser degree, α‐bisabolol are active against bacteria and ectoparasites and may represent an economical and sustainable alternative to toxic synthetic pesticides to control pathogens. © 2024 Society of Chemical Industry. Background and aim: Different Salmonella serotypes are considered one of the most important food pathogens in the world. Poultry meat and eggs are the primary carriers of Salmonella in human populations. This study aimed to estimate the Salmonella enteritidis and Salmonella typhimurium contamination rates of retail hen and quail eggs in Karaj, Iran. Moreover, the antimicrobial resistance patterns of the strains were evaluated, and the efficiency of the standard culture method and multiplex polymerase chain reaction (m‐PCR) were compared. Materials and methods: In this descriptive cross‐sectional study over 1 year (Jan–Dec 2022), 150 commercial and 150 backyard hen eggs and 300 commercial quail eggs, without cracks and fractures, were collected randomly from best selling groceries in Karaj city. All samples were examined for Salmonella contamination independently by standard culture and m‐PCR approaches. A standard disc diffusion method was employed to assess the antimicrobial susceptibility of the strains against 18 antimicrobial agents. Results: Out of 300 examined eggs, 2 S. enteritidis strains were isolated from the shell of backyard hen eggs. The same serotype was also detected in the contents of one of these two eggs. One S. typhimurium was isolated from the shell of a commercial hen egg. Overall, the Salmonella contamination of the shell and contents was 1% and 0.3%, respectively. Salmonella was not isolated from the eggshells or the contents of the quail eggs. There was complete agreement between the results of m‐PCR and the standard culture methods. Among the 18 tested antibiotics, the highest resistance was recorded for colistin (100%), followed by nalidixic acid (75%). Conclusion: As most Salmonella spp. are associated with human food poisoning, continuous surveillance is required to effectively reduce the risk posed by contaminated poultry eggs. Thermal death kinetics of Salmonella Enteritidis in ground chicken in the presence of ribwort, sorrel and yarrow extracts mixture (1%) was investigated by fitting the inactivation curves to the log–linear and Weibull models. The thermal inactivation curves were fitted well to both models (r2 ≥ 0.94). The times required for 4–log reduction in the S. Enteritidis population were shortened by 47–64% in ground chicken supplemented with the herbal extract mixture (p < 0.05). The 4–log reduction times calculated using the Weibull model were 0.13–13.6% longer than those extrapolated with the log–linear model. The z values in the ground chicken samples with and without herbal extract were 5.08°C (r2 = 0.97) and 5.57°C (r2= 0.94), respectively. The results suggest that the mixture of these extracts increased the thermal sensitivity of S. Enteritidis significantly at tested temperatures. In addition, using the Weibull model to calculate thermal death kinetics may reduce the underestimation of the thermal reduction times. Practical applications: This research aimed to investigate the effects of an herbal extract mixture consisting of ribwort, sorrel, and yarrow on the thermal resistance of Salmonella Enteritidis in ground chicken. While these plants are recognized for their antimicrobial and antioxidant properties, there are not many studies conducted to explore their effects on foodborne pathogens in the food matrix. As far as we know, this is the first study that examines the impact of these plants on the thermal resistance of S. Enteritidis in ground chicken. We determined the thermal death kinetics of S. Enteritidis in ground chicken with and without the herbal extract mixture using curve fitting to log‐linear and Weibull models. Salmonella Enteritidis was the primary foodborne pathogen responsible for acute gastroenteritis. The growing ceftriaxone resistance poses a significant threat to public health. Infection with S. Enteritidis has emerged as a major public health concern, particularly in developing countries. However, research on ceftriaxone-resistant S. Enteritidis (CRO-RSE) remains limited, particularly concerning its resistance mechanism, plasmid structure, and transmission characteristics. This study aims to address these gaps comprehensively. We collected 235 S. Enteritidis isolates from Hangzhou First People's Hospital between 2010 and 2020. Among these, 8.51% (20/235) exhibited resistance to ceftriaxone. Whole-genome analysis revealed that 20 CRO-RSE isolates harbored blaCTX-M-55 or blaCTX-M-14 on the plasmid. Moreover, the dissemination of the blaCTX-M-type gene was associated with IS26 and ISEcp1. Plasmid fusion entailing the integration of the p1 plasmid with antibiotic resistance genes and the p2 (pSEV) virulence plasmid was observed in certain CRO-RSE. Additionally, the structural analysis of the plasmids unveiled two types carrying the blaCTX-M-type gene: type A with multiple replicons and type B with IncI1 (Alpha) replicon. Type B plasmids exhibited superior adaptability and stability compared to type A plasmids within Enterobacteriaceae. Interestingly, although the type B (S808-p1) plasmid displayed the potential to spread to Acinetobacter baumannii, it failed to maintain stability in this species. [ABSTRACT FROM AUTHOR]

Bacitracin Methylene Disalicylate (BMD), as a feed additive to poultry diets, enhances digestion, prevents Salmonella enteritidis (SE) colonization, and treats current infections. The objective of this study was to utilize a quantitative proteomic approach to determine the effect of BMD feed additive on broiler chickens challenged with SE in the spleen proteome. At 1 d of age, chicks were randomly allocated into four groups: control with and without SE challenge (CON, n = 60; CON-SE, n = 60), BMD with and without SE challenge (BMD, n = 60; BMD-SE, n = 60). Birds in the CON-SE and BMD-SE treatment were administered SE inoculum by oral gavage. On day three and day seven post-gavage, the spleen was collected aseptically from birds in each treatment group (CON, n = 4/day; CON-SE, n = 4/day; BMD, n = 4/day; BMD-SE, n = 4/day). Proteomic analysis by liquid chromatography-tandem mass spectrometry (LC-MS/MS) showed an increased abundance of 115 proteins and decreased of 77 due to the BMD. Proteins that decreased in abundance were enriched for fibrinogen complex and extracellular space, whereas proteins that increased in abundance were enriched for proteasome-mediated ubiquitin-dependent protein catabolic process and mitochondrion. Analysis of the interaction between BMD and the Salmonella challenge found 230 differentially abundant proteins including proteins associated with RNA binding, spliceosome, protein transport, and cell adhesion among the upregulated proteins, and those associated with protein folding, carbon metabolism, biosynthesis of nucleotide sugars, response to oxidative stress, positive regulation of NIK/NF-kappaB signaling, and inflammatory response among the downregulated proteins. The impact of BMD treatment on spleen proteome indicates an anti-apoptotic effect. BMD also modified the response of the spleen to the SE challenge with a marked decrease in proteins that prompt cytokine synthesis and an increase in proteins involved in the selective removal of unfolded proteins. Human salmonellosis cases are often caused by Salmonella serovars Enteritidis and Typhimurium and associated with the consumption of eggs and egg products. Many countries therefore implemented general surveillance programmes on pullet and layer farms. The identification of risk factors for Salmonella infection may be used to improve the performance of these surveillance programmes. The aims of this study were therefore to determine 1) whether local farm density is a risk factor for the infection of pullet and layer farms by Salmonella Enteritidis and Typhimurium and 2) whether the sampling effort of surveillance programmes can be reduced by accounting for this risk factor, while still providing sufficient control of these serovars. We assessed the importance of local farm density as a risk factor by fitting transmission kernels to Israeli surveillance data during the period from June 2017 to April 2019. The analysis shows that the risk of infection by serovars Enteritidis and Typhimurium significantly increased if infected farms were present within a radius of approximately 4 km and 0.3 km, respectively. We subsequently optimized a surveillance programme that subdivided layer farms into low and high risk groups based on the local farm density with and allowed the sampling frequency to vary between these groups. In this design, the pullet farms were always sampled one week prior to pullet distribution. Our analysis shows that the risk-based surveillance programme is able to keep the between-farm R0 of serovars Enteritidis and Typhimurium below 1 for all pullet and layer farms, while reducing the sampling effort by 32% compared to the currently implemented surveillance programme in Israel. The results of our study therefore indicate that local farm density is an important risk factor for infection of pullet and layer farms by Salmonella Enteritidis and Typhimurium and can be used to improve the performance of surveillance programmes. Introduction: Microbiota plays a pivotal role in promoting the health and wellbeing of poultry. Essential oils (EOs) serve as an alternative solution for modulating poultry microbiota. This study aimed to investigate, using amplicon sequencing, the effect of a complex and well-defined combination of EOs feed supplement on both ileal and caecal broiler microbiota, within the context of Salmonella and Campylobacter intestinal colonization. Material and methods: For this experiment, 150-day-old Ross chicks were randomly allocated to two groups: T+ (feed supplementation with EO mix 500 g/t) and T- (non-supplemented). At day 7, 30 birds from each group were orally inoculated with 106 CFU/bird of a Salmonella enteritidis and transferred to the second room, forming the following groups: TS+ (30 challenged birds receiving infeed EOmix at 500g/t) and TS- (30 challenged birds receiving a non-supplemented control feed). At day 14, the remaining birds in the first room were orally inoculated with 103 CFU/bird of two strains of Campylobacter jejuni, resulting in the formation of groups T+C+and T-C+. Birds were sacrificed at day 7, D10, D14, D17, and D21. Ileal and caecal microbiota samples were analyzed using Illumina MiSeq sequencing. At D7 and D14, ileal alpha diversity was higher for treated birds (p<0.05). Results and discussion: No significant differences between groups were observed in caecal alpha diversity (p>0.05). The ileal beta diversity exhibited differences between groups at D7 (p < 0.008), D10 (p = 0.029), D14 (p = 0.001) and D17 (p = 0.018), but not at D21 (p = 0.54). For all time points, the analysis indicated that 6 biomarkers were negatively impacted, while 10 biomarkers were positively impacted. Sellimonas and Weissella returned the lowest (negative) and highest (positive) coefficient, respectively. At each time point, treatments influenced caecal microbiota beta diversity (p < 0.001); 31 genera were associated with T+: 10 Ruminoccocaceae genera were alternatively more abundant and less abundant from D7, 7 Lachnospiraceae genera were alternatively more and less abundant from D10, 6 Oscillospiraceae genera were variable depending on the date and 4 Enterobacteriaceae differed from D7. During all the experiment, Campylobacter decreased in treated birds (p < 0.05). This study showed that EO mix modulates ileal and caecal microbiota composition both before and during challenge conditions, increasing alpha diversity, especially in ileum during the early stages of chick life. Foodborne Salmonella serovars are important facultative intracellular pathogens that cause gastroenteritis in humans. Four strains from three of the more predominant Salmonella serovars in poultry were studied: Typhimurium, Enteritidis, and Heidelberg. Gentamicin susceptibility was determined using an agar disc diffusion test and minimum inhibitory concentration (MIC) assays for S. Typhimurium ATCC 14028 and S. Heidelberg ARI-14. Both strains were susceptible to gentamicin in disc diffusion. The MIC of gentamicin was approximately 125 mg/ml for all strains tested. These strains' adhesion and invasion abilities were determined with two different cell lines, a human intestinal epithelial cell line (Caco-2) as well as a chicken macrophage cell line (HD11). Attachment percentages for each Salmonella strain were greater than the strain's ability to invade cells. Similar attachment percentages to Caco-2 cells were observed for S. Typhimurium and S. Heidelberg. Attachment percentages were lower in HD11 cells than in Caco-2 cells, although Salmonella exhibited higher apparent HD11 invasion, likely from HD11 phagocytosis. Salmonella Enteritidis showed lower rates of adhesion and invasion in HD11 cells compared to Salmonella Typhimurium. Salmonella Enteritidis and Salmonella Typhimurium are the most common serovars observed in human salmonellosis while contaminated poultry products are the major source of Salmonella transmission to humans. Therefore, high pathogenicity of poultry originated S. Enteritidis and S. Typhimurium strains poses a serious risk to human health. In this study, we investigated the virulence genes of S. Enteritidis and S. Typhimurium strains isolated from litter and environmental samples of broiler chicken flocks. SipA, sipD, sopB, sopD, sopE, sopE2, sitC, sifA, ssaR, spvC, and pefA genes were investigated in a total of 137 strains consisting of 105 S. Enteritidis and 32 S. Typhimurium. Nine strains (6.6%) had all genes. No negative strain was detected for all genes. SopE was found in all strains (100.0%). SitC (89.1%), ssaR (83.9%), sipA (70.1%), sipD (73.0%), sopE2 (68.6%), spvC (68.6%), and pefA (73.0%) were also highly prevalent. Noticeable differences were observed between serovars in terms of sopE2, spvC and pefA prevalence: 77.1%, 80% and 82.9%, respectively, of S. Enteritidis strains were sopE2, spvC and pefA positive while 40.6%, 31.3% and 40.6% of S. Typhimurium strains were positive. This finding indicates that S. Enteritidis is more frequent than S. Typhimurium in poultry populations due to its higher virulence. Based on virulence gene distribution, the strains were divided into 44 different virulence genotypes, with the major genotype 4 (15.3%) carrying 8 of the 11 genes. The majority of strains (75.9%) were positive for at least 6 genes. S. Enteritidis and S. Typhimurium strains were highly virulent and pose a threat as zoonotic infection agents. Objective: Although it is thought that human stomach acidity would inhibit microorganisms, when food poisoning cases are considered, it is seen that especially during consumption of meat and meat products, the pH in the stomach rises and the fat layer creates a protective effect on bacteria, allowing microorganisms to transfer to intestine, and subsequently cause foodborne illnesses. The aim of this study was to determine whether beverage intake with meals influences why some individuals get sick, while others do not in food poisoning outbreak cases. Methods: The effect of coke, ayran and shalgam, which are popularly consumed beverages along with meals in Turkey, was examined on the survival of Salmonella enteritidis, Listeria monocytogenes. Escherichia coli O157: H7 and cocktail of these pathogens in simulated gastric fluid (SGF). Doner, a worldwide popular fast-food product, was used as the food sample. Results: Shalgam was found to be noteworthy that provided the greatest reduction in pathogen count. It was observed that in vitro consumption of one serving (300 mL) of shalgam, coke, and ayran along with contaminated doner, reduced the pathogenic bacteria count in SGF up to 2.6, 2.25 and 1.6 log CFU/g, respectively. The results showed that consuming those beverages alongside one portion (100 g) of low contaminated (~101 CFU/g) doner might eliminate the possibility of a healthy person having foodborne illness due to aforementioned bacteria. Conclusion: It can be concluded that, in food poisoning cases, not only the personal immune system, but also the type and amount of beverage consumed along with food, as well as the amount of food intake are effective. Eight new ruthenium complexes (C1, C2 (Ru-benzene or Ru-toluene complexes with L1, 3-amino-2-chloropyridine); C3, C4 (Ru-benzene or Ru-toluene complexes with L2, 2-amino-5-chloropyridine); C5, C6 (Ru-benzene or Ru-toluene complexes with L3, 3-acetylpyridine); and C7, C8 (Ru-benzene or Ru-toluene complexes with L4, 4-acetylpyridine) were synthesized. The chemical structures and purity of the compounds were confirmed using standard analytical methods such as 1H,13C NMR and IR spectroscopy, mass spectrometry, and elemental analysis, and their electrochemical behavior was evaluated using cyclic voltammetry experiments. The stability of the compounds in dimethyl sulfoxide solution was confirmed using 1H NMR spectroscopy. Finally, the antimicrobial potential of two complex precursors (CP1, [Ru(η6-benzene)Cl(μ-Cl)]2 and CP2, [Ru(η6-toluene)Cl(μ-Cl)]2), four ligands (L1–L4), and eight corresponding complexes (C1–C8) was screened against nine pathogenic microorganisms and two reference bacterial strains using the micro-well dilution method. The highest antibacterial activity (0.63 mg/mL) was for C5 against Staphylococcus aureus, Bacillus cereus, Escherichia coli, and Salmonella enteritidis, while C8 displayed more pronounced antimicrobial activity against Candida albicans (MIC/MMC = 0.31 mg/mL). Subsequently, the studied complexes underwent a detailed examination utilizing the DFT methodology, acquiring important quantum-chemical descriptors. Background: Probiotics are beneficial for animal health and new potential probiotics need to be characterized for their prospective use in improving animal health. In this study, 32 bacterial strains were isolated from a Norwegian forest cat (castrated, 12 years old) and a Persian cat (castrated, 10 years old), which were privately owned and had indoor access. Results: Lactobacillus rhamnosus CACC612 (CACC612) and Bifidobacterium animalis subsp. lactis CACC789 (CACC789) were selected as potential probiotics; characterization of the two strains showed equivalent acid tolerance, similar cell adhesion rates on the HT-29 monolayer cell line, and superior bile tolerance compared to Lactobacillus rhamnosus GG (LGG). Subsequently, they exhibited inhibitory effects against a broad spectrum of pathogenic bacteria, including E. coli (KCTC 2617), Salmonella Derby (NCCP 12,238), Salmonella Enteritidis (NCCP 14,546), Salmonella Typhimurium (NCCP 10,328), Clostridium difficile JCM 1296T. From evaluating host effects, the viability of the feline macrophage cell line (Fcwf-4) increased with the treatment of CACC612 or CACC789 (P < 0.05). The induced expression of immune-related genes such as IFN-γ, IL1β, IL2, IL4, and TNF-α by immune stimulation was significantly attenuated by the treatment of CACC612 or CACC789 (P < 0.05). When 52 clinical factors of sera from 21 healthy cats were analyzed using partial least squares discriminant analysis (PLS-DA), the animals were obviously clustered before and after feeding with CACC612 or CACC789. In addition, hemoglobin and mean corpuscular hemoglobin concentration (MCHC) significantly increased after CACC612 feeding (P < 0.05). Conclusions: In this study, feline-originated probiotics were newly characterized and their potentially probiotic effects were evaluated.
