The earlier studies have reported that grape seed extract exhibits broad-spectrum pharmacological activities. Therefore, studying the hepatoprotective effects and elucidation of mechanisms of action of the Indian Variety, Manjari Medika grape seed extract (GSE), may give an insight into therapeutic benefits. Methotrexate (MTX) is the first-line pharmacological therapy for different rheumatic diseases. The major adverse events such as hepatotoxicity are evident even in the low doses used for the treatment. The present study investigated the role of MTX on hepatic damage in murine liver and the plausible protective effects of the Indian grape variety, Manjari Medika grape seed extract, in ameliorating it.Methods and Results: To assess the hepatological modulation, mice were divided into eight groups to investigate the ameliorative potential of this GSE (75 and 125 mg/kg) and correlate the experimental findings. The active components of the extract were assessed through UPLC-(ESI)-QToF-MS analysis. On the other hand, various biochemical and immunological indices were carried out to correlate the experimental data. The result demonstrated that the prophylactic administration of GSE reduced MTX-induced hepatic toxicity indices, which subsequently restored the hepatic morphological architecture. Moreover, the application of GSE in a dual dosage (75 and 125 mg/kg) suppressed MTX-induced reactive oxygen species generation, followed by lipid peroxidation and cellular nitrite formation. MTX-induced inflammasome activation through the redox-assisted cascade of TLR4/NF-κB signaling was further reduced by applying the GSE. The results showed that the activation of cytoprotective transcription factor Nrf2 enhanced the level of endogenous antioxidants. Furthermore, through the regulation of TLR4/NF-κB and Nrf2/HO-1 axis, this extract could reduce the MTX-mediated hepatic damage.Conclusion: Our findings suggest that Manjari Medika seed extract could be used as a therapeutic agent to relieve the side effects of MTX and other hepatic disorders.Graphical Abstract: Keywords: hepatic fibrosis, Kupffer cells, Manjari Medika grape seed extract, proanthocyanidins, inflammasome, antioxidant response

7. Sustainable Utilization of Textile Dyeing Sludge and Coal Fly Ash by Brick Production Through Traditional Kilns

The fundamental purpose of this study was to evaluate the technical feasibility of incorporating fly ash (FA) and dyeing sludge (DS) in the production of brick. An attempt was taken to replace 10% to 100% clay by DS and FA in brick-making by volume. A brick firing kiln was used to burn the uniform-shaped bricks after replacing clay with DS and FA. Size and shape, hardness, soundness, water absorption, efflorescence, dry density, loss of ignition, firing shrinkage, specific gravity, compressive strength, and leaching tests were carried out to study the properties of these bricks. The compressive strength of the brink ranged from 6.25 MPa to 0.33 MPa and indicates a decreasing pattern in strength with the increase in the volume of DS and FA. Only 18.8% water absorption capacity was found in control bricks without DS and FA, while a maximum absorption of 40.19% was found for a particular combination of DS and FA. Similarly, dry density decreased with the increase in the volume of DS and FA. Besides, efflorescence in bricks was found within the allowable limits for certain combinations of DS and FA, which exceeded the allowable limits for other combinations. The presence of heavy metals (Ni, Zn, Cr, Cu, and Pb) in the extraction solution was insignificant. Based on the results of this study, we recommend that up to 10% clay can be substituted with DS and FA without substantially affecting the quality of bricks.

8. Impact of Pedagogical Intervention on Early Childhood Professionals' Emotional Availability to Children with Different Temperament Characteristics

- Abstract
The purpose of this study was to evaluate the impact of pedagogical intervention on early childhood education professionals' emotional availability to children with different temperament characteristics. Participants were 136 children (intervention group = 87; control group = 49) aged 1-6 years from 16 early childhood and care centres in Finland. The PedaSens intervention was a 9-month programme that included theory and video-based training for the professionals. Adult-child interaction was assessed with the Emotional Availability Scales before and after the intervention, and children's temperament was assessed with the Early Childhood Behavior Questionnaire. According to results, early childhood professionals' emotional availability increased in the intervention group, especially to children with high levels of activity and pleasure seeking and with low levels of attentional focusing. We argue that the quality of interaction in early childhood education and care can be increased by targeting professional trainings to support children with different temperament characteristics.

Melatonin (MLT) controls high-fat diet-induced NASH in the murine model by modulating NLRP3 mediated inflammation. P2X7R-mediated inflammasome activation is reported in several inflammatory models including NASH.Objective: The role of MLT in P2X7R-mediated inflammation in the NASH model has not yet been explored. The present study investigated the role of MLT in amending high-fat diet-induced nonalcoholic steatohepatitis in the murine liver.Methods: To evaluate the hepatological changes, mice were divided into four groups to investigate the improvement potential of this MLT (10 and 20 mg/kg) and to assess the experimental findings. Histology, biochemical assays, ELISA, FACS analysis, Western blotting, and IF were performed to assess the physical and molecular changes upon melatonin treatment.Results: The result demonstrated that MLT administration reduced HFD (high-fat diet)-induced non-alcoholic steatohepatitic indices, which successively restored the hepatic morphological architecture and other pathophysiological features too. Moreover, the application of MLT suppressed HFD-induced activation of the inflammasome and through TLR4/NF-κB signaling. Herein, we report that MLT significantly suppresses P2X7R expression and calcium influx along with inflammasome in both in vitro and in vivo. The docking study revealed a strong binding affinity of MLT with P2X7R. Moreover, the results also showed that the Nrf2 level was boosted which may normalize the expression of antioxidant proteins that safeguard against oxidative damage triggered by inflammation. Furthermore, some matrix metalloproteinases like MMP 2 and MMP 9 were repressed and TIMP-1 level was increased, which also signifies that MLT could improve liver fibrosis in this model.Conclusion: Based on our findings, this study may conclude that MLT could be used as a therapeutic agent in the high-fat diet-induced NASH model as it has persuasive anti-inflammatory potential.Graphical Abstract: Keywords: melatonin, NASH, NAFLD, inflammasome, P2X7, inflammation, Kupffer cells

In this article we, proposed a new two parameter distribution called inverted power modified Lindley distribution. The main objective is to introduce an extension to inverted modified Lindley distribution as an alternative to the inverted exponential, inverted gamma and inverted modified Lindley distributions, respectively. The proposed distribution is more flexible than the above mentioned distributions in terms of its hazard rate function. In the part of estimation of the proposed model, we first utilize the maximum likelihood (ML) estimator and parametric bootstrap confidence intervals, viz., standard bootstrap, percentile bootstrap, bias-corrected percentile (BCPB), bias-corrected accelerated bootstrap (BCAB) from the classical point of view as well the Bayesian estimation under different loss functions, squared error loss function, modified squared error loss function, and Bayes credible interval as to obtain the model parameter based on order statistics. A simulation study is carried out to check the efficiency of the classical and the Bayes estimators in terms of mean squared errors and posterior risks, respectively. Two real life data sets, have been analyzed for order statistics to demonstrate how the proposed methods may work in practice. [ABSTRACT FROM AUTHOR]

BACKGROUND: Students from Bangladesh pursuing STEM education often encounter obstacles when tackling diverse mathematical problems within various educational settings. Frequently, they find themselves lacking the essential prerequisite knowledge and strong foundational skills necessary to engage with the teaching and learning resources utilized at the undergraduate level, resulting in a significant number of students needing to seek readmission annually. OBJECTIVE: The objective of this study is to explore the determinants of academic achievement among university undergraduates majoring in mathematics in Bangladesh. Employing a mixed-method research approach, the study combines quantitative and qualitative data analysis to examine the viewpoints of both students and educators concerning these factors. The authors primarily emphasize classifying the factors that impact the efficacy of mathematics pedagogical methods. METHODOLOGY: The study is structured into three phases: i. An initial exploratory qualitative survey. ii. A quantitative triangulation survey. iii. Followed by explanatory semi-structured interviews. FINDINGS: To begin, the initial qualitative survey identified significant factors that contribute to students' achievements and setbacks in mathematics. Subsequently, the quantitative analysis verified both similarities and distinctions in the perspectives of students and educators. Furthermore, the correlation coefficient analysis revealed that male students frequently exhibit inconsistency and a lack of enthusiasm for studying, resulting in subpar performance. Conversely, female students frequently cited challenges like the difficulty of connecting mathematical theories to real-world applications, heavy course loads, and limited resources as reasons for their academic difficulties. - Abstract
Background: Accurate assessments of current and future fertility-including overall trends and changing population age structures across countries and regions-are essential to help plan for the profound social, economic, environmental, and geopolitical challenges that these changes will bring. Estimates and projections of fertility are necessary to inform policies involving resource and health-care needs, labour supply, education, gender equality, and family planning and support. The Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2021 produced up-to-date and comprehensive demographic assessments of key fertility indicators at global, regional, and national levels from 1950 to 2021 and forecast fertility metrics to 2100 based on a reference scenario and key policy-dependent alternative scenarios. Methods: To estimate fertility indicators from 1950 to 2021, mixed-effects regression models and spatiotemporal Gaussian process regression were used to synthesise data from 8709 country-years of vital and sample registrations, 1455 surveys and censuses, and 150 other sources, and to generate age-specific fertility rates (ASFRs) for 5-year age groups from age 10 years to 54 years. ASFRs were summed across age groups to produce estimates of total fertility rate (TFR). Livebirths were calculated by multiplying ASFR and age-specific female population, then summing across ages 10-54 years. To forecast future fertility up to 2100, our Institute for Health Metrics and Evaluation (IHME) forecasting model was based on projections of completed cohort fertility at age 50 years (CCF50; the average number of children born over time to females from a specified birth cohort), which yields more stable and accurate measures of fertility than directly modelling TFR. The other ten deal with Bapa Paloi, the stupid one, who is constantly being admonished by his wife. Sangumang and Bapa Paloi live in the upper-world, and mythological tales are told of their exploits. In addition to the 20 tales, a modern version of a Bapa Paloi tale is presented, written by the Rev. Munte Saha himself. The scene of this modern tale is not the upper-world but the present island of Kalimantan.

The controlled synthesis of two‐dimensional nanocrystals of graphene particles instead of amorphous carbon quantum dots is highly challenging and requires tremendous efforts to optimize the reaction conditions. It has been observed that as a significant bio‐compatible zero‐dimensional material, graphene quantum dots are considered as a novel material for opto‐electronics, energy, biological and environmental applications. However, very few are known about their utilization in photocatalytic degradation of pesticides, especially organochlorine pesticides which are resistant to removal by conventional wastewater treatment methods. In this work, graphene quantum dots were produced from rice husk at relatively low temperatures, resulting in an average particle size of 3.3 nm. The high resolution transmission electron microscopy clearly showed the formation of spherical quantum dots having lattice fringes and the selected area electron diffraction pattern showed the crystallinity of the graphene quantum dots. A study of photodegradation of three highly toxic organochlorine pesticides namely, dichlorodiphenyl trichloro ethane, cypermethrin and fenvalerate were carried out using as‐synthesized graphene quantum dots from rice husk. Interestingly, all these pesticides were found to be degraded around 60 % to 70 % under visible light conditions. It confirmed that synthesized graphene quantum dots from rice husk can act as prominent photocatalyst for the degradation of pesticides and other pollutants. [ABSTRACT FROM AUTHOR]

Rich in antioxidants, the peel and seed of pomegranate have previously demonstrated protection against oxidative-stress-related diseases, including cardiovascular disorders, diabetes, and cancer.Purpose: This study was designed to investigate the ameliorative role of pomegranate peel extract–stabilized gold nanoparticle (PPE-AuNP) on streptozotocin (STZ)-induced DN in an experimental murine model.Methods: Following the reduction methods, AuNP was prepared using the pomegranate peel ellagitannins and characterized by particle size, physical appearance, and morphological architecture. Modulatory potential of PPE-AuNP was examined through the plethora of biochemical and high throughput techniques, flow cytometry, immunoblotting, and immunofluorescence.Results: The animals treated with PPE-AuNP markedly reduced the fasting blood glucose, renal toxicity indices, and serum TC and TG in a hyperglycemic condition. As evident from an increased level of plasma insulin level, PPE-AuNP normalized the STZ-induced pancreatic β-cell dysfunction. The STZ-mediated suppression of endogenous antioxidant response was restored by the PPE-AuNP treatment, which reduced the generation of LPO as well as iROS. Furthermore, the hyperglycemia-mediated augmentation of protein glycation, followed by the NOX4/p-47phox activation, diminished with the application of PPE-AuNP. The histological and immunohistochemical findings showed the protective efficacy of PPE-AuNP in reducing STZ-induced glomerular sclerosis and renal fibrosis. In addition, it reduced proinflammatory burden through the modulation of the MAPK/NF-κB/STAT3/cytokine axis. Simultaneously, PI3K/AKT-guided Nrf2 activation was evident upon the PPE-AuNP application, which enhanced the antioxidant response and maintained hyperglycemic homeostasis.Conclusion: The findings indicate that the use of PPE-AuNPs might act as an economic therapeutic remedy for alleviating DN. Abstract: Mitogen-activated protein kinase (MAPK) phosphatase-3 (MKP-3) and its substrates (extracellular signal-regulated kinase [ERK] and p38) play an important role in pathophysiological mechanisms of acute postoperative and chronic neuropathic pain in the spinal cord. This study aimed to understand the role of MKP-3 and its target MAPKs at the site of surgical incision in nociceptive behavior. Wild-type (WT) and MKP-3 knockout (KO) mice underwent unilateral plantar hind paw incision. Mechanical allodynia was assessed by using von Frey filaments. Peripheral ERK-1/2 and p38 phosphorylation were measured by Western blot. Cell infiltration was determined using hematoxylin and eosin histological staining. Peripheral phosphorylated ERK-1/2 (p-ERK-1/2) inhibition was performed in MKP-3 KO mice. In WT mice, mechanical hypersensitivity was observed on postoperative day 1 (0.69±0.17 g baseline vs 0.13±0.08 g day 1), which resolved normally by postoperative day 12 (0.46±0.08 g, N=6). In MKP-3 KO mice, this hypersensitivity persisted at least 12 days after surgery (0.19±0.06 g; N=6). KO mice displayed higher numbers of infiltrating cells (51.4±6 cells/0.1 mm2) than WT mice (8.7±1.2 cells/0.1 mm2) on postoperative day 1 (vs 5–6 cells/0.1 mm2 at baseline) that returned to baseline 12 days after surgery (10–12 cells/0.1 mm2). In WT mice, peripheral p-p38 and p-ERK-1/2 expression increased (5- and 3-fold, respectively) on postoperative days 1 and 5, and returned to basal levels 7–12 days after surgery (N=3 per group). Peripheral p-p38 levels in MKP-3 KO mice followed a similar expression pattern as WT mice. Peripheral p-ERK-1/2 levels in MKP-3 KO mice remained elevated 12 days after surgery (2.5-fold, N=3 per group). Administration of PD98059 (MEK inhibitor, N=8, vehicle N=9) reduced p-ERK-1/2 expression in the incised tissue and blocked hypersensitivity in MKP-3 KO mice (N=6). 35. A Deep Learning-Based Method for Predicting Out-of-Field Radiotherapy Dose for Pediatric Wilms Tumor Patients.

Unintended out-of-field dose has been implicated as an important contributor to late health effects following radiotherapy. However, there are gaps in knowledge about organ dose tolerance. Furthermore, the out-of-field dose cannot be estimated accurately by the treatment planning system, but must instead be calculated by time-consuming Monte Carlo radiation transport simulation. Therefore, to support clinical treatment optimization and research on late health effects following radiotherapy, we sought to demonstrate a deep learning approach to predict 3D whole-body dose maps for pediatric patients. We used a database of Monte Carlo-derived whole-body dose maps which were previously reconstructed for 4716 pediatric patients in the National Wilms Tumor Study late effects cohort. The patients in this cohort tended to receive standardized treatments with the field borders determined by anatomical structures. To train and test our model we identified subsets of patients who received a 6 MV AP-PA beam configuration to the abdomen (with or without kidney block), left flank, right flank, or chest (90% patients used for training, 10% for testing). A customized neural network was trained separately for each field configuration with the dose map being normalized by the prescribed dose. The input to our model was: (1) a CT image of a whole-body phantom matched to the patient based on height and weight; (2) a RT structure file containing organ contours of interest; (3) the normalized dose map; and (4) the patient's height, weight, and gender. Next, we extracted multi-level and multi-scale feature maps, the predicted dose map file, from the customized deep neural network final layers. Predicted doses were compared voxel-to-voxel with original doses using different statistical metrics. Dose-volume histograms were made for each organ at risk to visualize differences between actual and predicted doses. The absolute dose map for a patient was calculated by multiplying the predicted normalized dose map by the patient's radiotherapy prescribed dose. The dose maps for patients receiving radiation to more than one location could be predicted separately and added together to get the total from the entire plan. Organ dose was calculated by applying organ contour masks to the predicted dose map. Our quantitative analysis of the test dataset yielded a structural similarity index of 0.89 ± 0.02 (mean ± SD) Confidence Interval (CI): [0.86, 0.90], mean absolute error of 2.57 ± 0.49 Gy (CI: [2.17, 3.40]), and root mean square error of 9.28 ± 1.97 Gy (CI: [7.54, 12.40]). Our results suggest that the algorithm is functional, although further refinement is necessary as error rates for some patients exceed expectations. We are also exploring extending this work to multi-class-based dose prediction considering the additional photon treatment energies (e.g., 4 MV, 10 MV, 16 MV, and Co-60). [ABSTRACT FROM AUTHOR]

- Abstract
Spinel-type LiMn1.5Ni0.5O₄ has been paid temendrous consideration as an electrode material because of its low cost, high voltage, and stabilized electrochemical performance. Here, we demonstrate the mechanism of iron (Fe) integration into LiMn1.5Ni0.5O₄ via solution methods followed by calcination at a high temparature, as an efficient electrocatalyst for water splitting. Various microscopic and structural characterizations of the crystal structure affirmed the integration of Fe into the LiMn1.5Ni0.5O₄ lattice and the constitution of the cubic LiMn1.38Fe0.12Ni0.5O₄ crystal. Local structure analysis around Fe by extended X-ray absorption fine structure (EXAFS) showed Fe3+ ions in a six-coordinated octahedral environment, demonstrating incorporation of Fe as a substitute at the Mn site in the LiMn1.5Ni0.5O₄ host. EXAFS also confirmed that the perfectly ordered LiMn1.5Ni0.5O₄ spinel structure becomes disturbed by the fractional cationic substitution and also stabilizes the LiMn1.5Ni0.5O₄ structure with structural disorder of the Ni²⁺ and Mn⁴⁺ ions in the 16d octahedral sites by Fe²⁺ and Fe³⁺ ions. However, we have found that Mn³⁺ ion production from the redox reaction between Mn⁴⁺ and Fe²⁺ influences the electronic conductivity significantly, resulting in improved electrochemical oxygen evolution reaction (OER) activity for the LiMn1.38Fe0.12Ni0.5O4 structure. Surface-enhanced Fe in LiMn1.38Fe0.12Ni0.5O₄ serves as the electrocatalytic active site for OER, which was verified by the density functional theory study.

- Abstract
Modern aquaculture practices fundamentally need upgrades under two aspects: i. eco-sustainability and ii. high quality of productions. Crustaceans may be both subjects of aquaculture productions and dangerous parasites of fish, to be chemically controlled in production plants. Salmon aquaculture, under this point of view, exhibits several issues, because of the high impacts of parasitic copepods: seventeen species of Caligidae copepods represent possible threats for fish and the environment. The drugs used to fight salmon louse are quite toxic and inefficient. For this reason, periodic treatments are needed to assure sufficient growth and survival of salmons, but they produce damages to the benthic and planktonic communities neighbour to the cages, in particular to crustacean’s communities. In addition, various economically important decapod crustaceans may be damaged by the presence of pesticides, so producing also an economic loss to the fishery activities. In this chapter, the main issues related to the use of pesticides in salmon aquaculture practices are reported, and the results of a newer investigation on natural compounds derived from cyanobacteria are exposed, as a promising tool to obtain an eco-sustainable control of parasitic copepods, maximizing the health of salmons and avoiding damage to other economically or ecologically important crustaceans.

43. Algal volatiles – the overlooked chemical language of aquatic primary producers

- Abstract
Volatiles are important ‘infochemicals’ that play a crucial role in structuring life on our planet, fulfilling diverse functions in natural and artificial systems. Algae contribute significant quantities to the global budget of volatiles, but the ecological roles of aquatic volatiles are not well understood. In this review, we discuss the current knowledge of volatile compounds from freshwater and marine microalgae and marine macroalgae, with a focus on their ecological roles. We highlight the multiple reported functions of biogenic volatiles, ranging from intraspecific communication for reproduction, intra-bloom signalling and antioxidant functions, to various interspecific signal exchanges that may allow herbivores to locate them and function in defence against competitors and predators. Beyond reviewing our current understanding, we specifically highlight major knowledge gaps and emerging questions for algal volatile research. These novel perspectives have the potential to improve our understanding of aquatic ecosystems and thus need to be addressed in future research. Filling these gaps and addressing these questions will facilitate humanity's efforts to exploit aquatic volatiles in various applications.

- Abstract
Chemical communication via infochemicals plays a pivotal role in ecological interactions, allowing organisms to sense their environment, locate predators, food, habitats, or mates. A growing number of studies suggest that climate change-associated stressors can modify these chemically mediated interactions, causing info-disruption that scales up to the ecosystem level. However, our understanding of the underlying mechanisms is scarce. Evidenced by a range of examples, we illustrate in this opinion piece that climate change affects different realms in similar patterns, from molecular to ecosystem-wide levels. We assess the importance of different stressors for terrestrial, freshwater, and marine ecosystems and propose a systematic approach to address highlighted knowledge gaps and cross-disciplinary research avenues.

Background: For a long older age and the presence of alarm features are regarded as indications for prompt endoscopy in patients with dyspepsia. We aimed to find out the value of alarm features in diagnosing serious organic upper gastrointestinal lesions in patients with dyspepsia. Material & methods: In this observational study clinical variables and endoscopic findings of consecutive patients with dyspepsia were recorded in a semi-structured questionnaire. Univariate and multivariate logistic regression was done stepwise to identify predictors for endoscopic findings. A simplified predictor model was built with the age and the presence of any predictor alarm feature to find out the diagnostic accuracy of this model for the significant endoscopic lesion. Results: Among 304 patients (M=134, F=170) one or more alarm features were present in 193 cases (63.5%). Significant organic lesions were found in 84(27.6%) cases. Age >45 years (OR 2.608), abdominal lump (OR 4.489) and family history of upper gastrointestinal cancer (OR 3.880) were found as independent predictors of major endoscopic findings. Using a simplified predictor model of age e" 45 years or the presence of any predictive alarm feature, sensitivity, specificity, positive predictive value, and negative predictive value were 32.4 %, 82.5 %, 79.8% and 36.4% respectively for a significant endoscopic lesion. For upper gastrointestinal cancers, these values were 6%, 100.0%, 100.0% and 47.3% respectively. Conclusion: The predictive value of the age and the presence of alarm features alone are not optimal for significant endoscopic findings in patients with dyspepsia. A newer and more accurate predictive model is a time demand for organic UGI lesions, especially for malignancies. [ABSTRACT FROM AUTHOR]
