Abstract

In this paper, an enhanced Virtual Element Method (VEM) formulation is proposed for plane elasticity. It is based on the improvement of the strain representation within the element, without altering the degree of the displacement interpolating functions on the element boundary. The idea is to fully exploit polygonal elements with a high number of sides, a peculiar VEM feature, characterized by many displacement degrees of freedom on the element boundary, even if a low interpolation order is assumed over each side. The proposed approach is framed within a generalization of the classic VEM formulation, obtained by introducing an energy norm in the projection operator definition. Although such generalization may mainly appear to have a formal value, it allows to effectively point out the mechanical meaning of the quantities involved in the projection operator definition and to drive the selection of the enhanced representations. Various enhancements are proposed and tested through several numerical examples. Numerical results successfully show the capability of the enhanced VEM formulation to (i) considerably increase accuracy (with respect to standard VEM) while keeping the optimal convergence rate, (ii) bypass the need of stabilization terms in many practical cases, (iii) obtain natural serendipity elements in many practical cases, and (vi) effectively treat also nearly incompressible materials., Comment: 27 pages, 6 figures

Abstract

Aims: The heterozygous phospholamban (PLN) mutation R14del (PLN R14del+/−) is associated with a severe arrhythmogenic cardiomyopathy (ACM) developing in the adult. “Superinhibition” of SERCA2a by PLN R14del is widely assumed to underlie the pathogenesis, but alternative mechanisms such abnormal energy metabolism have also been reported. This work aims to (1) to evaluate Ca2+ dynamics and energy metabolism in a transgenic (TG) mouse model of the mutation prior to cardiomyopathy development; (2) to test whether they are causally connected. Methods: Ca2+ dynamics, energy metabolism parameters, reporters of mitochondrial integrity, energy, and redox homeostasis were measured in ventricular myocytes of 8–12 weeks-old, phenotypically silent, TG mice. Mutation effects were compared to pharmacological PLN antagonism and analyzed during modulation of sarcoplasmic reticulum (SR) and cytosolic Ca2+ compartments. Transcripts and proteins of relevant signaling pathways were evaluated. Results: The mutation was characterized by hyperdynamic Ca2+ handling, compatible with a loss of SERCA2a inhibition by PLN. All components of energy metabolism were depressed; myocyte energy charge was preserved under quiescence but reduced during stimulation. Cytosolic Ca2+ buffering or SERCA2a blockade reduced O2 consumption with larger effect in the mutant. Signaling changes suggest cellular adaptation to perturbed Ca2+ dynamics and response to stress. Conclusions: (1) PLN R14del+/− loses its ability to inhibit SERCA2a, which argues against SERCA2a superinhibition as a pathogenetic mechanism; (2) depressed energy metabolism, its enhanced dependency on Ca2+ and activation of signaling responses point to an early involvement of metabolic stress in the pathogenesis of this ACM model.

11. Framing survivor speech : locating the victim/survivor in media accounts of sexual violence

Abstract

This thesis evaluates the role of survivor speech in discussions of sexual violence taking place in the media. By undertaking a case study analysis of the media coverage of three cases of 'stranger rape', child sexual exploitation and historic child sexual abuse respectively, the discursive contexts within which victim/survivors are giving their account of the experience are examined. The study focused on a selected set of discursive parameters which establish the specific contexts in which the voices of victims/survivors are presented to media audiences. These parameters are collated as discursive frames, within which each victim's/survivor's experience is formulated by the voices partaking in the conversation on sexual violence in the media. The identification of the frames of sexual violence was preceded by an analysis of the naming processes employed by these voices. Analysis of the case studies was focused on multimodal texts from print and broadcast media which offered input in the form of the voices of victims/survivors. Texts were sampled mostly from genres of current affairs and news features. This thesis also reviews the suitability of existing epistemologies and research methodologies for the examination of media-based conversations of sexual violence, focusing on distinctly feminist approaches of discourse analysis. By uncovering the discursive contexts which victims/survivors have to negotiate each time they speak out about their experience in the media, the analysis in this thesis is able to contribute a media studies-based perspective to debates about the process of surviving sexual violence and the corresponding positionality of the terms 'victim' and 'survivor'.

Abstract

Modeling human neuronal properties in physiological and pathological conditions is essential to identify novel potential drugs and to explore pathological mechanisms of neurological diseases. For this purpose, we generated a three-dimensional (3D) neuronal culture, by employing the readily available human neuroblastoma SH-SY5Y cell line, and a new differentiation protocol. The entire differentiation process occurred in a matrix and lasted 47 days, with 7 days of pre-differentiation phase and 40 days of differentiation, and allowed the development of a 3D culture in conditions consistent with the physiological environment. Neurons in the culture were electrically active, were able to establish functional networks, and showed features of cholinergic neurons. Hence here we provide an easily accessible, reproducible, and suitable culture method that might empower studies on synaptic function, vesicle trafficking, and metabolism, which sustain neuronal activity and cerebral circuits. Moreover, this novel differentiation protocol could represent a promising cellular tool to study physiological cellular processes, such as migration, differentiation, maturation, and to develop novel therapeutic approaches.

Abstract

Iatrogenic urinary tract injury (IUTI) is a severe complication of emergency digestive surgery. It can lead to increased postoperative morbidity and mortality and have a long-term impact on the quality of life. The reported incidence of IUTIs varies greatly among the studies, ranging from 0.3 to 1.5%. Given the high volume of emergency digestive surgery performed worldwide, there is a need for well-defined and effective strategies to prevent and manage IUTIs. Currently, there is a lack of consensus regarding the prevention, detection, and management of IUTIs in the emergency setting. The present guidelines, promoted by the World Society of Emergency Surgery (WSES), were developed following a systematic review of the literature and an international expert panel discussion. The primary aim of these WSES guidelines is to provide evidence-based recommendations to support clinicians and surgeons in the prevention, detection, and management of IUTIs during emergency digestive surgery. The following key aspects were considered: (1) effectiveness of preventive interventions for IUTIs during emergency digestive surgery; (2) intra-operative detection of IUTIs and appropriate management strategies; (3) postoperative detection of IUTIs and appropriate management strategies and timing; and (4) effectiveness of antibiotic therapy (including type and duration) in case of IUTIs.

Abstract

Urinary tract infections (UTIs) present a formidable challenge in the care of individuals affected by multiple sclerosis (MS). Lower urinary tract dysfunction is a prevalent issue among MS patients, predisposing them to an elevated risk of UTIs. When left untreated, UTIs can further exacerbate the already compromised quality of life in individuals with MS. The diagnosis and management of UTIs in MS patients necessitate a careful clinical evaluation. The objective of this review is to delineate preventive strategies and current and developing therapeutic approaches for preventing and treating UTIs associated with urinary dysfunction, catheterization, and upper urinary tract infections in patients with MS. Effectively addressing UTIs and urinary tract dysfunction in individuals with multiple sclerosis calls for a comprehensive, interdisciplinary approach. [ABSTRACT FROM AUTHOR]

Abstract

Three-dimensional cancer models, such as spheroids, are increasingly being used to study cancer metabolism because they can better recapitulate the molecular and physiological aspects of the tumor architecture than conventional monolayer cultures. Although Agilent Seahorse XFe96 (Agilent Technologies, Santa Clara, CA, United States) is a valuable technology for studying metabolic alterations occurring in cancer cells, its application to three-dimensional cultures is still poorly optimized. We present a reliable and reproducible workflow for the Seahorse metabolic analysis of three-dimensional cultures. An optimized protocol enables the formation of spheroids highly regular in shape and homogenous in size, reducing variability in metabolic parameters among the experimental replicates, both under basal and drug treatment conditions. High-resolution imaging allows the calculation of the number of viable cells in each spheroid, the normalization of metabolic parameters on a per-cell basis, and grouping of the spheroids as a function of their size. Multivariate statistical tests on metabolic parameters determined by the Mito Stress test on two breast cancer cell lines show that metabolic differences among the studied spheroids are mostly related to the cell line rather than to the size of the spheroid. The optimized workflow allows high-resolution metabolic characterization of three-dimensional cultures, their comparison with monolayer cultures, and may aid in the design and interpretation of (multi)drug protocols.

Abstract

Cocoa beans are one of the largest cultivated crops all over the world, producing large amount of by-products. For this reason, it is necessary to valorise cocoa by-products to obtain valuable source of bioactive compounds. In this paper, a pressurized hot water extraction process for recovery of theobromine and caffeine from cocoa by-product was developed and optimized. The extraction was carried out on ASE and all parameters affected by extraction efficiency and antioxidant capacity were optimized by a chemometric approach. Theobromine and caffeine were quantified by UPLC-UV (283 nm), whereas antioxidant capacity was evaluated by in vitro assays (ABTS and DPPH). By applying the PHWE under optimized conditions (ethanol 15%, temperature 90 °C, 5 cycles and static time 6 min), the extraction efficiency increased by 156% for theobromine and 160% for caffeine in comparison with the results obtained using ultrasound assisted liquid extraction. The effect of PHWE extract on cell viability of colorectal and breast cancer cell lines was also tested. The application of PHWE to selective recovery of theobromine and caffeine from cocoa shell by-products is a green, automatic, and rapid method, representing a valid alternative to conventional extraction methods to obtain ingredients for food industries.

Contains fulltext : 296327.pdf (Publisher’s version ) (Open Access), Iatrogenic urinary tract injury (IUTI) is a severe complication of emergency digestive surgery. It can lead to increased postoperative morbidity and mortality and have a long-term impact on the quality of life. The reported incidence of IUTIs varies greatly among the studies, ranging from 0.3 to 1.5%. Given the high volume of emergency digestive surgery performed worldwide, there is a need for well-defined and effective strategies to prevent and manage IUTIs. Currently, there is a lack of consensus regarding the prevention, detection, and management of IUTIs in the emergency setting. The present guidelines, promoted by the World Society of Emergency Surgery (WSES), were developed following a systematic review of the literature and an international expert panel discussion. The primary aim of these WSES guidelines is to provide evidence-based recommendations to support clinicians and surgeons in the prevention, detection, and management of IUTIs during emergency digestive surgery. The following key aspects were considered: (1) effectiveness of preventive interventions for IUTIs during emergency digestive surgery; (2) intra-operative detection of IUTIs and appropriate management strategies; (3) postoperative detection of IUTIs and appropriate management strategies and timing; and (4) effectiveness of antibiotic therapy (including type and duration) in case of IUTIs.
