Divertor tiles after Joint European Torus-ITER like wall (JET-ILW) campaigns and dust collected after JET-C and JET-ILW operation were examined by a set of complementary techniques (full combustion and radiography) to determine the total, specific and areal tritium activities, poloidal tritium distribution in the divertor and the presence of that isotope in individual dust particles. In the divertor tiles, the majority of tritium is detected in the surface region and, the areal activities in the ILW divertor are in the 0.5–12 kBq cm ^−2 range. The activity in the ILW dust is associated mainly with the presence of carbon particles being a legacy from the JET-C operation. The total tritium activities show significant differences between the JET operation with ILW and the earlier phase with the carbon wall (JET-C) indicating that tritium retention has been significantly decreased in the operation with ILW.

Tritium (T) distributions on tungsten (W)-coated plasma-facing tiles used in the third ITER-like wall campaign (2015–2016) of the Joint European Torus (JET) were examined by means of an imaging plate technique and β-ray induced x-ray spectrometry, and they were compared with the distributions after the second (2013–2014) campaign. Strong enrichment of T in beryllium (Be) deposition layers was observed after the second campaign. In contrast, T distributions after the third campaign was more uniform though Be deposition layers were visually recognized. The one of the possible explanations is enhanced desorption of T from Be deposition layers due to higher tile temperatures caused by higher energy input in the third campaign. Nondestructive analysis of tritium (T) distribution was performed by means of imaging plate technique on specimens cut from the Be limiters, W-coated carbon tiles and bulk W lamellae retrieved from the JET tokamak after the first and third experimental campaigns with the ITER-like wall. Afterwards, analyses were continued using X-ray photoelectron spectroscopy, microscopy techniques and thermal desorption spectroscopy. Co-deposits formed on the W-coated tiles in the 1st campaign showed large T retention because of high carbon content reaching up to 50 atomic %, while the carbon fraction in co-deposits after the 3rd campaign was distinctly lower. The T retention of the plasma-facing surface of the bulk W tile was smaller than that of the W-coated tiles by a factor of 20, while deposition of small amount of T was found at the side surfaces facing to the gaps in a lamella structure. The correlation of T distributions with surface morphology and the discharge conditions is discussed.

Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. Methods and results: A total of 52,014 patients with newly diagnosed AF were enrolled globally; of these, 1388 patients were recruited from 26 sites within India (2012–2016). 7. Delivery options for sublingual immunotherapy for allergic rhinoconjunctivitis: clinical considerations for North America

Background: Sublingual immunotherapy (SLIT) can be delivered via tablets (SLIT-T) or aqueous drops (SLIT-D). SLIT-D dosing recommendations using North American extracts were published in 2015. We review the 2015 recommendations in the context of recent research, and compare and contrast dosing, efficacy, safety, adherence, and cost of SLIT-T and SLIT-D for allergic rhinoconjunctivitis (ARC) in North America. Methods: Randomized controlled trials (RCT) of SLIT-D and SLIT-T trials were identified by a systematic PubMed search through March 1, 2022. Results: Dose-finding studies have been conducted for all approved SLIT-T; efficacy in North American populations was demonstrated in 11 RCTs. Approved SLIT-T are uniform internationally. Few dose-finding studies for SLIT-D have been conducted using North American extracts; efficacy was demonstrated in 2 RCTs. Extrapolation of dosing from SLIT-D studies conducted with extracts from other geographic regions is unreliable. Since the 2015 SLIT-D dosing recommendations, no new RCTs of SLIT-D have been conducted with North American extracts, whereas 6 SLIT-T RCTs have since been conducted in North America. Local allergic reactions are the most common adverse events with SLIT-T and SLIT-D, but both can induce systemic allergic reactions. Adherence to SLIT-D and SLIT-T remains a challenge. Patients must pay for SLIT-D directly, whereas SLIT-T is usually covered by insurance. Conclusion: As part of shared decision-making, patients should be informed about the scientific evidence supporting the use of SLIT-T and SLIT-D for ARC.

Chronic rhinosinusitis (CRS) is known to affect around 5 % of the total population, with major impact on the quality of life of those severely affected (1). Despite a substantial burden on individuals, society and health economies, CRS often remains underdiagnosed, under-estimated and under-treated (2). RNA polymerase III (Pol III) transcribes medium-sized non-coding RNAs (collectively termed Pol III genes). Emerging diverse roles of Pol III genes suggest that individual Pol III genes are exquisitely regulated by transcription and epigenetic factors. Here we report global Pol III expression/methylation profiles and molecular mechanisms of Pol III regulation that have not been as extensively studied, using nc886 as a representative Pol III gene. In a human mammary epithelial cell system that recapitulates early breast tumorigenesis, the fraction of actively transcribed Pol III genes increases reaching a plateau during immortalization. Hyper-methylation of Pol III genes inhibits Pol III binding to DNA via inducing repressed chromatin and is a determinant for the Pol III repertoire. When Pol III genes are hypo-methylated, MYC amplifies their transcription, regardless of its recognition DNA motif. Thus, Pol III expression during tumorigenesis is delineated by methylation and magnified by MYC.

Surface characterization of bulk tungsten tiles (W lamellae) used during the first campaign of JET with the ITER-Like Wall (JET-ILW) was performed by means of microscopy and tritium imaging techniques. This is the first report regarding very detailed structural studies of W lamellae from the JET-ILW divertor. A special feature of the W lamellae installed in JET is the intrinsic network of micro-cracks detected on surfaces of the as-manufactured material. Analyses of different W lamellae samples on the plasma facing surface reveal two types of surface structures caused by plasma impact: areas with strong erosion and regions of mild plasma interaction. In regions of strong erosion, a thin modified layer (thickness of ~20 nm) with a high density of defects including bubble-like structures has been formed. In this study, energetic helium (He) ion irradiation was performed to obtain bulk He distribution in tungsten (W) materials, concurrent with damage introduction at high temperature. Then, deuterium (D) implantation and thermal desorption spectrometry were performed to evaluate D retention. At the same time, the surface tritium (T) concentration and depth distribution were evaluated by imaging plate (IP) and β-ray induced X-ray spectroscopy (BIXS) measurements after mixed D-T gas exposure. Numerical simulations were applied to evaluate changes in binding energies, diffusion depths, and trapping sites under different irradiation conditions. The results showed that weak trapping sites with higher concentration, such as vacancies, were produced during only energetic He+ irradiation events, leading to enhancement of D retention. Fe3+-He+ simultaneous irradiation promoted the formation of HexVy complexes, which reduced the concentration of vacancy trapping sites and changed the stress field around defects, leading to the suppression of D trapping behavior. From the reduced effects of D retention caused by HexVy complexes at higher temperatures, the results suggested that defect recovery was the dominant mechanism. With increasing damage level at higher temperatures, more weak trapping sites, such as dislocations and vacancies sites, were produced, leading to a more dominant influence on D retention than HexVy complex effects. It was also found that HexVy complexes prevented D diffusion to the bulk and that simulation results showed that the damage level had little impact on D diffusion depth.

Acrolein (ACR) is a highly reactive α, β-unsaturated carbonyl compound that is related to several diabetic vascular complications. ACR, a ubiquitous aldehyde pollutant, is a product of incomplete combustion reactions and a major component of cigarette smoke. ACR is also endogenously generated during lipid peroxidation. Glycosylphosphatidylinositol (GPI)-anchored proteins is a membrane bound protein, which have a variety of cellular functions. It is reported that some of them showed the reduced level under diabetic conditions. In this study, we explored whether ACR may affect GPI anchor related gene expression in human umbilical vein endothelial cells (HUVECs) by modulation of miRNA expression that play a significant role in the post-transcriptional regulation of genes. We performed global gene expression analysis, acquired altered expression levels of 27 miRNAs and 21 mRNAs that are relevant to GPI anchored proteins, and observed their anti-correlations. Gene Ontology (GO) terms that are closely associated with GPI anchor biosynthesis were highest when analysis was based on biological processes characterized by enriched GO categories. Our results revealed that ACR may control miRNAs to regulate genes involving in GPI anchor biosynthesis in human endothelial cells.

We investigate the influence of post-rolling heat treatment on the microstructural evolution and subsequent mechanical properties of a roll-bonded stainless steel (STS)-Al–Mg 3-ply clad sheet. Intermetallic compound (IMC) phases, γ (Mg 17 Al 12 ) and β (Mg 2 Al 3 ), were generated at the interface between the constituent Mg and Al alloys. The thickness of the IMC layers increased to more than 10 μm with increasing annealing time up to 3 h at 300 °C. Upon extending the annealing time to 12 h, the β phase at the interface between Al and Mg hardened significantly, which was verified by investigating the local mechanical properties of the γ and β phases using nanoindentation. Furthermore, the mechanical properties of the three constituent alloys and the 3-ply clad sheets were systematically investigated by uniaxial tensile and flexural bending tests. Taking both tensile strength and elongation into account, the 3-ply sample annealed for 1 h at 300 °C exhibited acceptable overall mechanical properties. The mechanism for retarding interfacial delamination in the direction perpendicular to the transverse loading was also confirmed by observation from the side surfaces of the 3-point-bending-tested specimens. Abrupt longitudinal crack propagation was observed within the annealing-hardened β phase, followed by the generation of short lateral cracks in the bending sample annealed for 3 h, indicating that the overannealing-induced brittleness of a specific IMC phase at the interface may be detrimental to overall mechanical properties of the roll-bonded 3-ply clad sheet adopted in this study.

Summary Background Glucocorticoids (GCs) affect the pathophysiology of sebaceous glands, causing development or exacerbation of acne. The availability of GCs is regulated by isoenzymes of 11β-hydroxysteroid dehydrogenase (11βHSD) at tissue-specific levels. 11βHSD type 1 (HSD11β1) is a reductase, catalysing the conversion of cortisone to active cortisol, and is highly expressed in liver and adipose tissue. Recently, HSD11β1 was observed in human skin in keratinocytes and fibroblasts. Objectives To investigate the expression of HSD11β1 in sebaceous glands of normal and acne-involved skin, and to examine the role of HSD11β1 in GC-induced lipid synthesis and toll-like receptor 2 (TLR2) expression in sebocytes. Methods Expression of HSD11β1 was examined by immunohistochemistry in acne lesional skin and normal skin of healthy volunteers. The cultured SZ95 sebocytes were treated with dexamethasone, and the lipid synthesis and mRNA levels of sterol regulatory element binding protein 1 (SREBP-1) and TLR2 were determined. Use of an HSD11β1 inhibitor and the small interference RNA (siRNA) approach were used to investigate the role of HSD11β1 on the GC regulation of sebocyte functions. Results HSD11β1 was expressed in human sebaceous glands and upregulated in acne lesional skin. HSD11β1 mRNA was enhanced by dexamethasone and cytokines in SZ95 sebocytes. Dexamethasone enhanced lipid synthesis, partially through the transcriptional induction of SREBP-1, and also by increasing TLR2 mRNA levels. Inhibition of HSD11β1 by PF-915275 or siRNA significantly inhibited the GC-induced lipid synthesis and the mRNA expression of SREBP-1 and TLR2. Conclusions Our results indicate that HSD11β1 plays a key role in the modulation of GC action on sebocytes, including sebum production and TLR2-mediated inflammation, thereby influencing the pathogenesis of acne.

Background: There have been few clinical studies of the role of regulatory T cells (Tregs) in halo formation of halo nevus. Objective: To evaluate the clinicopathologic features and the presence of Tregs in halo nevi. Methods: We analyzed 30 halo nevi and performed immunohistochemical analysis using antibodies against CD4, CD8, CD25 and Foxp3. We also performed double immunohistochemical staining for Foxp3 and CD25. Results: We found significant increases in Foxp3+ Tregs, and the shorter the halo nevus duration, the more Foxp3+ Tregs were detected. Also, the ratio of Foxp3 to CD8 T cells was increased in early stages of halo nevi. Double immunohistochemical staining suggested that the Tregs in the halo nevi were CD25+Foxp3+ T cells. Conclusions: Foxp3+ Tregs were greatly increased in the halo nevi. The shorter the halo nevi duration, the more Foxp3+ Tregs were involved in the earlier developmental stages of halo nevi.

In the working population, muscle fatigue and musculoskeletal discomfort are common, which, in the case of insufficient recovery may lead to musculoskeletal pain. Workers suffering from musculoskeletal pains need to be rehabilitated for recovery. Isokinetic testing has been used in physical strengthening, rehabilitation and post-operative orthopedic surgery. Frequency analysis of electromyography (EMG) signals using the mean frequency (MNF) has been widely used to characterize muscle fatigue. During isokinetic contractions, EMG signals present strong nonstationarities. Hilbert-Haung transform (HHT) and autoregressive (AR) model have been known more suitable than Fourier or wavelet transform for nonstationary signals. Moreover, several analyses have been performed within each active phase during isokinetic contractions. Thus, the aims of this study were i) to determine which one was better suitable for the analysis of MNF between HHT and AR model during repetitive maximum isokinetic extensions and ii) to investigate whether the analysis could be repeated for sequential fixed epoch lengths. Seven healthy volunteers (five males and two females) performed isokinetic knee extensions at and until 50% of the maximum peak torque was reached. Surface EMG signals were recorded from the rectus femoris of the right thigh. An algorithm detecting the onset and offset of EMG signals was applied to extract each active phase of the muscle. Following the results, slopes from the least-square error linear regression of MNF values showed that muscle fatigue of all subjects occurred. The AR model is better suited than HHT for estimating MNF from nonstationary EMG signals during isokinetic knee extensions. Moreover, the linear regression can be extracted from MNF values calculated by sequential fixed epoch lengths (p> 0.0I).

We report a precise measurement of the masses of the $\Xi_c(2645)$ and $\Xi_c(2815)$ baryons using a data sample of 414 fb$^{-1}$ collected by the Belle collaboration at the KEKB $e^+ e^-$ collider. The states $\Xi_c(2645)^{0,+}$ are observed in the $\Xi_c^{+,0}\pi^{-,+}$ decay modes, while the $\Xi_c(2815)^{0,+}$ are reconstructed in the $\Xi_c(2645)^{+,0}\pi^{-,+}$ decay modes. The following mass splittings are determined: $m_{\Xi_c(2645)^+} - m_{\Xi_c(2645)^0} = (-0.1 \pm 0.3 ({\rm stat}) \pm 0.6 {(\rm syst})) {\rm MeV}/{\rm c}^2$ and $m_{\Xi_c(2815)^+} - m_{\Xi_c(2815)^0} = (-3.4 \pm 1.9 ({\rm stat}) \pm 0.9 {(\rm syst})) {\rm MeV}/{\rm c}^2$ with a much better precision than the current world averages. We also observe a new decay mode, $\Xi_c(2980)^{0,+} \to \Xi_c(2645)^{+,0}\pi^{-,+}$.

We report a high-statistics measurement of the relative branching fraction Br(D0 --> pi+pi-pi0)/Br(D0 --> K-pi+pi0). A 357 inverse fb data sample collected with the Belle detector at the KEKB asymmetric-energy e+e- collider was used for the analysis. The relative branching fraction Br(D0 --> pi+pi-pi0)/Br(D0 --> K-pi+pi0) is determined with an accuracy comparable to the latest world average value.

We report on the observation of LaGuerre-Gaussian (LG) beam profiles of photonic quantum ring (PQR) hole-type lasers. Naturally formed LG beams are highly valuable for optical tweezering applications, like transporting or manipulating DNA molecules. Various devices with different injection currents and hole diameters have been analyzed.

The geomicrobiological control of chromium behavior in Cr-contaminated sediment by indigenous bacteria was investigated under aerobic conditions with respect to the initial Cr(VI) concentration, electron donor, and pH condition. Sediment samples were collected from a stream which was contaminated with chromium from nearby pigment manufacturing. In experiments using enriched cultures of the indigenous bacteria from the sediment samples, 31% of an initial concentration of Cr(VI) of 13 mg/L, equivalent to the exchangeable Cr(VI) concentration in the sediment, was reduced by the bacteria. Furthermore, the removal efficiency of Cr(VI) by indigenous bacteria was increased with external supply of electron donor in the form of glucose, lactate or acetate. In the range of pH 3–9, the removal efficiencies of Cr(VI) were 31% to 41%. The removal efficiency of Cr(VI) was highest at pH 3, while at pH 7, the pH of the stream sediment in the field, 34% of dissolved Cr(VI) was reduced. The indigenous bacteria can mediate reduction of Cr(VI) to Cr(III), and chromate/dichromate pollution in the field. Environmental factors such as pH and the type of electron donor were found to influence the microbial control of Cr(VI) reduction.
