To predict primary failure of infliximab (IFX) therapy in Crohn's disease (CD) and to identify patients who maintain long-term effectiveness to IFX is currently not feasible. Some genetic variations are proposed as potential biomarkers.We assessed a set of single nucleotide polymorphisms (SNPs) in genes related to the IFX mechanism of action and the presence of HLA-DQA1 * 05 allele on the primary response and long-term durability in CD patients.A multi-centre cross-sectional study of IFX-exposed adult patients with CD was undertaken. Treatment persistence and time to failure were co-primary endpoints. DNA from the 131 patients was genotyped. Association between SNPs and clinical variables with IFX persistence was assessed.Failure to IFX was documented in 65 (49.6%) out of 131 patients. IFX persistence was associated either with carrying the TT genotype in ADAM17 rs10929587 (ORa=0.2; 95%CI=0.1-0.8; p = 0.021), or the CC genotype in SLCO1C1 rs3794271 (ORa=0.2; 95%CI=0.1-0.7; p = 0.008), according to multivariate logistic regression. In contrast, previous bowel resection increased the risk of IFX failure (ORa=2.8; 95%CI=1.1-7.3; p = 0.025). Cox regression analysis confirmed these findings and also identified IL23R rs10489629-TT (HRa 0.41; 95%CI=0.22-0.75; p = 0.004) and concomitant immunosuppressants (HRa 0.46; 95%CI=0.27-0.77; p = 0.003) as protection from IFX failure. However, no association between HLA-DQA1 * 05 allele and persistence of IFX therapy was found, with similar failure rates among carriers and non-carriers (52.8% vs. 47.4%, respectively; p = 0.544).SNPs rs10929587-TT in ADAM17, rs10489629-TT in IL23R and rs3794271-CC in SLCO1C1, together with no previous bowel surgery and concomitant immunosuppression, were identified as protection from failure to IFX.

Valorar en individuos con lesión medular traumática (LMT) la relación entre la mortalidad y la necesidad de UCI y las alteraciones objetivadas mediante resonancia magnética (RM) precoz, analizando alteraciones parenquimatosas, disrupción de ligamentos vertebrales (DLV) y compresión del cordón medular (CCM). Diseño. Estudio retrospectivo. Ámbito. Hospital de tercer nivel, unidad de lesionados medulares y UCI. Pacientes. Individuos con LMT aguda entre los años 2010 y 2019. Intervenciones. Análisis de RM realizada en las primeras 72 horas. Variables de interés. Ingreso en UCI y mortalidad. Resultados. Recogidos 269 casos. El patrón que se asoció a una mayor mortalidad fue la hemorragia (16,7%) por 12,5% de los edemas a un nivel y 6,5% de los edemas a múltiples niveles (p = 0,125). Lo mismo aconteció con los ingresos en UCI: 69,0% en hemorragia por 60,2% en edema múltiple y 46,3% en edemas cortos (p = 0,018). Con respecto a la CCM, la mortalidad fue del 13,4% con 59,2% de ingresos en UCI por 2,2% y 42,2% de quienes no presentaban compresión (p = 0,020 y p = 0,003). To assess in individuals with traumatic spinal cord injury (TSCI) the relationship between mortality and need for ICU and early magnetic resonance imaging (MRI), analyzing spinal parenchymal alterations, disruption of vertebral ligaments (DVL) and spinal cord compression (SCC). Design. Retrospective study. Setting. Third-level hospital, Spinal Cord Injury Unit and ICU. Patients. Individuals with acute TSCI between 2010 and 2019. Intervention. Analysis of MRI performed in the first 72 h. Variables of interest. Admission to ICU and mortality. Results. 269 cases collected. The pattern that demonstrated higher mortality was cord hemorrhage (16.7%) for 12.5% of single-level edema and 6.5% of multilevel edema (p = 0.125). The same happened with ICU admissions: 69.0% in hemorrhage, 60.2% in multilevel edema and 46.3% in short edema (p = 0.018). Analyzing CCM, mortality was 13.4% with 59.2% of ICU admissions, for 2.2% and 42.2% of individuals without cord compression (p = 0.020 and p = 0.003). The figures of death and ICU admission among cord injuries with DVL were 15.0% and 67.3%, for 6.2% and 44.4% of the individuals without DLV (p < 0.001 and p = 0.013). Conclusions. The presence of spinal cord hemorrhage, SCC and DVL was associated with a higher admission in ICU. A significant increase in mortality was observed in cases with SCC and DVL.

On 15 September 2021 , during one of the experimen tal bottom trawls, ca rried out at a speed of 3.5 knot, SGB noticed a medium-sized dark petrel. It was feeding on discards from the fishing operation in the company of Northern Fulmars, Sooty Shearwaters, Great Shearwaters, Northern Gannets and Lesser Black-backed Gulls Larus fuscus. SGB identified it as White-chinned Petrel Procellaria aequinoctialis and took severa l photographs during the two hours that it spent around the vessel. Ultra high dose rate electron beams also known as FLASH radiotherapy is becoming of importance in several preclinial cancer treatment studies. However, due to the dose rate used during the irradiation sessions, no real time dose monitoring device exists to date. In this work, we present the development of a beam current transformer (BCT), from the choice of the ferromagnetic component and the realisation of the shielding to the design of a front-end electronics based on a trans-impedance circuit in order to perform a low noise optimization of the detector. The BCT prototype is able to monitor a beam current range from 1.2 μA to 200 mA with a rise time constant better than 20 ns and a droop rate of the signal below 0.05% · μs-1. Preliminary in-situ measurements are also presented. The goal is to combine the BCT system which measure in real time the beam current, to an ionisation chamber monitoring the beam shape and position in order to provide a reliable dose monitoring system.

Standardized relative abundance indices of albacore (Thunnus alalunga Bonnaterre, 1788) caught by the Spanish surface longline (LLALB) in the western Mediterranean Sea were estimated for the period 2009-2019. Yearly standardized CPUE were estimated through Generalized Linear Mixed Effects Models (GLMM) under a negative binomial error distribution assumption. The main factors in the standardization analysis were year and season (quarter). The index shows an increasing trend from the beginning of the series (2009) to a maximum in 2011; following a decrease up to 2013, and a relatively stable trend fluctuating around a level three and a half times lower compared to the maximum abundance for the period 2013-2019.

Over recent years, the field of cell and gene therapy has witnessed rapid growth due to the demonstrated benefits of using living cells as therapeutic agents in a broad range of clinical studies and trials. Bioprocess economic models (BEMs) are fundamental tools for guiding decision-making in bioprocess design, being capable of supporting process optimization and helping to reduce production costs. These tools are particularly important when it comes to guiding manufacturing decisions and increasing the likelihood of market acceptance of cell-based therapies, which are often cost-prohibitive because of high resource and quality control costs. Not only this, but the inherent biological variability of their underlying bioprocesses makes them particularly susceptible to unforeseen costs arising from failed or delayed production batches. The present work reviews important concepts concerning the development of bioprocesses for stem cell therapy products and highlights the valuable role which BEMs can play in this endeavor. Additionally, some theoretical concepts relevant to the building and structuring of BEMs are explored. Finally, a comprehensive review of the existent BEMs so far reported in the scientific literature for stem cell-related bioprocesses is provided to showcase their potential usefulness.

Technology parks promote science, technology and innovation through their components, such as high-tech companies, universities and incubators. They show a significant increase in the number of units, especially in emerging countries such as Brazil. Unlike the industrial and logistic parks that, due to functional requirements, have their pre-determined and standardized architectural and urban characteristics, technological parks have in the innovation, their main characteristic, the possibility of proposing more suitable spaces for urban innovation. The Roof Mosaic is an approach that seeks self-sufficiency in urban areas from the food-energy-water nexus, using rooftops to produce food, renewable energy and rainwater harvesting. The integration between technology parks and Roof Mosaic can be advantageous since their roofs have a lower slope and greater design flexibility than other parks and can, therefore, contribute to the sustainable development of cities. The aim of this paper is to evaluate the potential of technology parks when implementing the Roof Mosaic. A case study underway was used, which is the Federal University of Rio de Janeiro (UFRJ) Technology Park (Rio de Janeiro, Brazil). The results from the case study show the potential for: 100% food self-sufficiency using hydroponic production of lettuces and tomatoes, meeting the demands of approximately 3,300 people; 45.2% for energy generated from photovoltaic panels; and 43% for water reusing rainwater harvesting. The potential of avoided CO2 emissions using rainwater for non-potable purposes, food production and renewable energy, presented of nearly 805 tons of avoided CO2 equivalents.

Despite the recent progresses in the development of inert diets, the rearing of early life stages of aquatic organisms still depends on the use of live feeds. Within live feeds, Artemia sp. and rotifer (Brachionus plicatilis) are widely used in the rearing of marine larvae due to their high availability and acceptance by a large number of species. Nonetheless, both live preys naturally possess low content of long-chain polyunsaturated fatty acids (LC-PUFA) such as 20:5n-3 (EPA), and 22:6n-3 (DHA), which are essential fatty acids (EFA) for proper development of marine fish larvae. In this respect, enrichment of live preys is used to tailor its lipid composition towards the nutritional needs of marine larvae. To improve the design of live prey enrichment protocols, it is advisable to unveil the metabolic fate of fatty acids (FA) during the enrichment process. Los parásitos que pueden afectar a los peces son numerosos e incluyen miembros de distintos grupos zoológicos. Estas parasitosis pueden ocurrir tanto en poblaciones salvajes como cultivadas, y pueden llegar a causar alerta en los consumidores por posibles efectos en la salud humana. En este sentido hay que destacar que los resultados obtenidos en proyectos actualmente en ejecución muestran una ausencia total de este tipo de parásitos en las poblaciones cultivadas.

Aims: The present study was designed to investigate the dose-response relationship of muscular performance and anaerobic power to whole-body vibration (WBV). Methods: Nine recreationally males were subjected to five experimental conditions: A) Squat exercise (SE) without WBV; B) WBV1 [31.55 m.s-2]; C) WBV2 [159.73 m.s-2]; (D) WBV3 [319.45 m.s-2]; and E) WBV4 [567.91 m.s-2]. Before and after the experimental procedures, muscle performance was assessed by a vertical jump test and handgrip strength test and anaerobic power by Wingate anaerobic test (WT). Results: WBV2 and WBV3 increased jump relative power [Δ=0.92 ± 0.89 W/Kg (+1.97%) and Δ=1.29 ±1.79 W/Kg (+2.77%), respectively; P=0.02; effect size= 0.92; power= 0.87] and height [Δ=0.96 ± 0.73 cm (2.80%) and Δ=1.61 ± 2.36 cm (4,57%), respectively; P= < 0.01; effect size= 0.96; power= 0.99] compared to the SE [Δ=-1.28 ± 0.81 cm (-3.55%)]. There was no dose-response relationship of handgrip strength to WBV. WBV2 improved WT relative power (power/body weight) compared to the other experimental conditions [Δ= 0.61 ± 0.36 W/Kg (+ 5.25%); P < 0.01; ; effect size= 0.98; power= 0.99], and WT relative work was higher in WBV3 [Δ= 1.86 ± 5.46 KJ/Kg (+ 0.70%)condition compared to WBV1[Δ= -6.71 ± 4.03 KJ/Kg (- 2.49%) ( P= 0.03). Conclusion: These results suggest that lower limb muscular performance and anaerobic power are responsive to vibration accelerations WBV2 (159.73 m.s-2) and WBV3 (319.45 m.s-2).

Objectives We wished to assess the potential impact of directing EC management based on molecular classification, and the projected cost implications of molecular subtype-directed care. Methods Surgical staging, treatment, surveillance, and hereditary cancer program(HCP) referrals were assessed for all ECs managed in a single calendar year (2016) across 24 Canadian centers. Variation of practice was recorded, as well as where a change in management would be projected and associated cost implications of that change based on molecular subtype assignment. Results Data from 862 patients revealed wide variation in surgical staging, with lymph node dissection(LND) performed in 61% of ECs (range 25–100%), including 38% LND in Gr1ECs (0–100%). Adjuvant therapy (type, when/if given) and cancer surveillance (frequency, site e.g., community vs. cancer center) was inconsistent within and across centers for both early-and late-stage disease. Molecular classification identified 29% MMRd ECs (n=247) but only 8% of these women had been referred to HCP. 30 women who did consult HCP were MMRproficient. 38% of MMRd ECs had no LND and 43% did not receive radiation. 16% and 18% of p53abn ECs had no LND or omentectomy respectively, and only 58% received chemotherapy. De-escalation of treatment in early-stage POLEmut and NSMP ECs (n=63 treated, where molecular classification would direct observation or brachytherapy-only) would have reduced costs by $348,896–407,830CAD or $5538–6466CAD per patient. Conclusions There is currently profound variation in practice for all aspects of EC management with implications to patients and health systems. Molecular classification can provide consistency in care and direct biologically-informed management.

Excessive sweating and body odors in many cultures can cause negative perceptions of an individual and in many cases is related to poor hygiene. Personal hygiene products have been developed with the intention of preventing these undesirable issues. The aim of this paper is to review the main active ingredients used in marketed deodorant and antiperspirant formulations as well as to identify new strategies and future methods to optimize such products and prevent malodor. PubMed and ScienceDirect databases were used to search for studies reporting the use of deodorants and antiperspirants, the compounds used in the formulations, their mechanisms of action and associated controversies, as well as new trends and approaches in the area. Even today, we are still using well-known and established actives such as triclosan and aluminum salts, and these are still the most used compounds in deodorants with bactericidal and antiperspirant properties. These substances have been on the market for more than 40 years, and still there are many questions concerning the safety of both actives. There is a general increased interest globally for lifestyles that focus on sustainability and more natural products such as plant sources and the use of, for example, essential oils. The research that focuses in the area of antiperspirants and deodorants is now more focused on studies of the armpit biochemistry and function and control of the microbiota present in this area. Other possible areas of interest are biotechnological solutions and finding new compounds that will interfere with the biochemistry of the process of sweat decomposition. Further approaches include formulations with probiotics which would maintain the balance of axillary microbiota.

Treatment options are limited for patients with advanced melanoma who have progressed on checkpoint inhibitors and targeted therapies such as BRAF/MEK inhibitors (if BRAF-V600E mutated). Adoptive cell therapy utilizing tumor-infiltrating lymphocytes (TIL) has shown antitumor efficacy with durable responses in heavily pretreated melanoma patients. Safety and efficacy of lifileucel, a centrally manufactured cryopreserved autologous TIL therapy assessed by both investigator and an independent review committee (IRC), are presented.

International audience; In this work, we report on systematic Monte Carlo (MC) studies for the FRACAS apparatus, a large acceptance mass spectrometer that will be used to measure the fragmentation cross sections of 12C ions for hadrontherapy. The apparatus, placed in a 100 mbar reaction chamber, will be made of a beam monitor, trackers surrounding a magnet and a Time-Of-Flight (TOF) wall. In order to determine the required performances of the trackers, Geant4 simulations of the whole system and in-house developed algorithms were used. While keeping the beam monitor and TOF-wall positions fixed, the effects of the tracker positions and spatial resolutions on the trajectory reconstruction and mass identification efficiencies have been extracted. An optimal configuration was found where the upstream trackers should be located 6 cm away from the target and spaced 4 cm apart whereas their spatial resolutions should be close to 100 \textmu m. The positions of the downstream trackers will have to be changed according to the beam energy to preserve high identification efficiencies. Their spatial resolutions, even though of a lesser importance compared to the upstream trackers, should be around 1 mm or better. In this optimal configuration, an overall fragment identification efficiency above 90% has been obtained for beam energies ranging from 100 to 400 MeV/nucleon.
