The study of the orientation of three-jet events from e+ e- –> Z0 –> multi-hadrons is presented, in particular the polar angle distributions of the thrust axis and of the normal to the three-jet plane, and the azimuthal correlations between the hadron plane and the one defined by the beam and thrust axes. The data are compared with results at lower energy and with QCD predictions. Good agreement with QCD predictions is observed. The scalar gluon theory is excluded by the data.

High Energy Physics::Phenomenology ,High Energy Physics::Experiment - Abstract
Using a data sample of 115000 hadronic Z0 decays, a search was performed for the production of excited charged leptons decaying into an ordinary lepton and one photon. Pair production of excited leptons of mass below 45 GeV/c2 is excluded. From an analysis of the lepton-photon mass spectra this experiment is also able to exclude single production of these particles for masses that reach up to 85 GeV/c2, if the compositeness scale is below 1 TeV. © 1992 Springer-Verlag.

High Energy Physics::Experiment - Abstract
From measurements of the cross sections for e+ e - –> hadrons and the cross sections and forward-backward charge-asymmetries for e+ e- –> e+ e-, mu+ mu- and tau+ tau- at several centre-of-mass energies around the Z0 pole with the DELPHI apparatus, using approximately 150000 hadronic and leptonic events from 1989 and 1990, one determines the following Z0 parameters: the mass and total width M(Z) = 91.177 +/- 0.022 GeV, GAMMA(Z) = 2.465 +/- 0.020 GeV, the hadronic and leptonic partial widths GAMMA(h) = 1.726 +/- 0.019 GeV, GAMMA(t) = 83.4 +/- 0.8 MeV, the invisible width GAMMA(inv) = 488 +/- 17 MeV, the ratio of hadronic over leptonic partial widths R(Z)z = 20.70 +/- 0.29 and the Born level hadronic peak cross section sigma-0 = 41.84 +/- 0.45 nb. A flavour-independent measurement of the leptonic cross section gives very consistent results to those presented above (GAMMA(t) = 83.7 +/- 0.8 MeV). From these results the number of light neutrino species is determined to be N(nu) = 2.94 +/0. 10. The individual leptonic widths obtained are: GAMMA(e) = 82.4 +/- 1.2 MeV, GAMMA(mu) = 86.9 +/- 2.1 MeV and GAMMA(tau) = 82.7 +/- 2.4 MeV. Assuming universality, the squared vector and axial-vector couplings of the Z0 to charged leptons are: VBAR(t)2 = 0.0003 +/- 0.0010 and ABAR(l)2 = 0.2508 +/- 0.0027. These values correspond to the electroweak parameters: rho(eff) = 1.003 +/- 0.011 and sin2-theta(W)eff = 0.241 +/- 0.009. Within the Minimal Standard Model (MSM), the results can be expressed in terms of a single parameter: sin2-theta(W)MSBAR = 0.2338 +/- 0.0027. All these values are in good agreement with the predictions of the MSM. Fits yield 43 < m(top) < 215 GeV at the 95% level. Finally, the measured values of GAMMA(Z) and GAMMA(inv), are used to derive lower mass bounds for possible new particles.

Astrophysics::High Energy Astrophysical Phenomena ,High Energy Physics::Experiment - Abstract
The total and differential cross-sections for the reaction e+e- –> gamma-gamma(gamma) are measured at centre of mass energies around 91 GeV using an integrated luminosity of 4.7 pb-1. The agreement with the QED prediction is good. Consequently there is no evidence for non-standard channels which would have the same experimental signature. The lower limits on the QED cutoff parameters are LAMBDA+ > 113 GeV and LAMBDA- > 95 GeV. An upper limit on the effective coupling between a possible excited electron and the gamma is derived. At 95% confidence level the branching ratios for Z0 decay into pi-0-gamma, eta-gamma and gamma-gamma-gamma are below 1.5 x 10(-4), 2.8 x 10(-4) and 1.4 x 10(-4) respectively.

High Energy Physics::Experiment ,Nuclear Experiment - Abstract
Measurements of the cross section and forward-backward asymmetry for the reaction e+e- –> mu+mu- using the DELPHI detector at LEP are presented. The data come from a scan around the Z0 peak at seven centre of mass energies, giving a sample of 3858 events in the polar angle region 22-degrees < theta < 158-degrees. From a fit to the cross section for 43-degrees < theta < 137-degrees, a polar angle region for which the absolute efficiency has been determined, the square root of the product of the Z0 –> e+e- and Z0 –> mu+mu- partial widths is determined to be (GAMMA-e-GAMMA-mu)1/2 = 85.0 +/- 0.9 (stat.) +/- 0.8 (syst.) MeV. From this measurement of the partial width, the value of the effective weak mixing angle is determined to be sin2(theta-w) = 0.2267 +/- 0.0037. The ratio of the hadronic to muon pair partial widths is found to be GAMMA-h/GAMMA-mu = 19.89 +/- 0.40 (stat.) +/- 0.19 (syst.). The forward-backward asymmetry at the resonance peak energy E(CMS) = 91.22 GeV is found to be A(FB) = 0.028 +/- 0.020 (stat.) +/- 0.005 (syst.). From a combined fit to the cross section and forward-backward asymmetry data, the products of the electron and muon vector and axial-vector coupling constants are determined to be V(e)V-mu = 0.0024 +/- 0.0015 (stat.) +/- 0.0004 (syst.) and A(e)A-mu = 0.253 +/- 0.003 (stat.) +/- 0.003 (syst.). These results are in good agreement with the expectations of the minimal standard model.

High Energy Physics::Experiment ,Nuclear Experiment - Abstract
This paper presents an analysis of the multiplicity distributions of charged particles produced in Z0 hadronic decays in the DELPHI detector. It is based on a sample of 25364 events. The average multiplicity is =20.71±0.04(stat)±0.77(syst) and the dispersion D=6.28±0.03(stat)±0.43(syst). The data are compared with the results at lower energies and with the predictions of phenomenological models. The Lund parton shower model describes the data reasonably well. The multiplicity distributions show approximate KNO-scaling. They also show positive forward-backward correlations that are strongest in the central region of rapidity and for particles of opposite charge. © 1991 Springer-Verlag.

High Energy Physics::Experiment ,Nuclear Experiment - Abstract
We have studied the energy-energy angular correlations in hadronic final states from Z0 decay using the DELPHI detector at LEP. From a comparison with Monte Carlo calculations based on the exact second order QCD matrix element and string fragmentation we find that Lambda-MS(5) = 104(-20)+25(stat.)-20(+25)(syst.)-00(+30)(theor.) MeV, which corresponds to alpha-s(91 GeV) = 0.106 +/- 0.003(stat.) +/- 0.003(syst.)-0.000(+0.003)(theor.). The theoretical error stems from different choices for the renormalization scale of alpha-s. In the Monte Carlo simulation the scale of alphA-s as well as the fragmentation parameters have been optimized to described reasonably well as aspects of multihadron production.

A determination of the partial width gamma-cc-BAR of the Z0 boson into charm quark pairs is presented, based on a total sample of 36 900 Z0 hadronic decays measured with the DELPHI detector at the LEP collider. The production rate of ccBAR events is derived from the inclusive analysis of charged pions coming from the decay of charmed meson D*+ –> D0-pi+ and D*- –> DBAR0-pi- where the pi- +/- is constrained by kinematics to have a low p(T) with respect to the jet axis. The probability to procedure these pi- +/- from D* +/- decay in ccBAR events is taken to be 0.31 +/- 0.05 as measured at square-root s = 10.55 GeV. The measured relative partial width GAMMA-ccBAR/GAMMA-h = 0.162 +/- 0.030 (stat.) +/- 0.050(syst.) is in good agreement with the standard model value of 0.171. Together with our previous measurement of the total hadronic width GAMMA-h this implies GAMMA-ccBAR = 282 +/- 53(stat.) +/- 88(syst.) MeV.

Quantifying the influence of human activities, such as reservoir building, water abstraction, and land use change, on hydrology is crucial for sustainable future water management, especially during drought. Model-based methods are very time-consuming to set up and require a good understanding of human processes and time series of water abstraction, land use change, and water infrastructure and management, which often are not available. Therefore, observation-based methods are being developed that give an indication of the direction and magnitude of the human influence on hydrological drought based on limited data. We suggest adding to those methods a “paired-catchment” approach, based on the classic hydrology approach that was developed in the 1920s for assessing the impact of land cover treatment on water quantity and quality. When applying the paired-catchment approach to long-term pre-existing human influences trying to detect an influence on extreme events such as droughts, a good catchment selection is crucial. The disturbed catchment needs to be paired with a catchment that is similar in all aspects except for the human activity under study, in that way isolating the effect of that specific activity. In this paper, we present a framework for selecting suitable paired catchments for the study of the human influence on hydrological drought. Essential elements in this framework are the availability of qualitative information on the human activity under study (type, timing, and magnitude), and the similarity of climate, geology, and other human influences between the catchments. We show the application of the framework on two contrasting case studies, one impacted by groundwater abstraction and one with a water transfer from another region. Applying the paired-catchment approach showed how the groundwater abstraction aggravated streamflow drought by more than 200 % for some metrics (total drought duration and total drought deficit) and the water transfer alleviated droughts with 25 % to 80 %, dependent on the metric. Benefits of the paired-catchment approach are that climate variability between pre- and post-disturbance periods does not have to be considered as the same time periods are used for analysis, and that it avoids assumptions considered when partly or fully relying on simulation modelling. Limitations of the approach are that finding a suitable catchment pair can be very challenging, often no pre-disturbance records are available to establish the natural difference between the catchments, and long time series of hydrological data are needed to robustly detect the effect of the human activities on hydrological drought. We suggest that the approach can be used for a first estimate of the human influence on hydrological drought, to steer campaigns to collect more data, and to complement and improve other existing methods (e.g. model-based or large-sample approaches).

Despite the increasing influence of human activities on water resources in our current Anthropocene era, the impacts of these activities on the duration, rate and timing of the recovery of drought events, known as the drought termination phase, remain unknown. Here, we present the first assessment of how different human activities (i.e. water abstractions, reservoirs, water transfers) affect drought termination. Six case studies in Europe were used to analyse the human influence on streamflow drought termination characteristics. For all case studies, we compared the drought and drought termination characteristics derived from a human-influenced time series of streamflow (observation data) and a naturalised time series (modelled data) for the same period. Overall, results clearly demonstrate the influence of human activities on drought terminations in all the studied catchments. Groundwater abstractions, reservoirs and mixed influences were all found to increase the average duration of drought termination, whereas water transfers into the catchment decreased drought termination duration. Results also show that average drought termination rates increased in all case studies due to the human influence. Furthermore, start and end months of the termination phase were more skewed to certain months in human-influenced data than in the naturalised situation. Future research could extend this new knowledge by looking to add further case studies and covering different human activities to gain a wider understanding on how human actions modify hydrological droughts and their recovery. Furthering this work could also help to improve the forecasting of drought recovery in the Anthropocene, which is important for informing drought management decisions.

In the current human-modified world, or Anthropocene, the state of water stores and fluxes has become dependent on human as well as natural processes. Water deficits (or droughts) are the result of a complex interaction between meteorological anomalies, land surface processes, and human inflows, outflows, and storage changes. Our current inability to adequately analyse and manage drought in many places points to gaps in our understanding and to inadequate data and tools. The Anthropocene requires a new framework for drought definitions and research. Drought definitions need to be revisited to explicitly include human processes driving and modifying soil moisture drought and hydrological drought development. We give recommendations for robust drought definitions to clarify timescales of drought and prevent confusion with related terms such as water scarcity and overexploitation. Additionally, our understanding and analysis of drought need to move from single driver to multiple drivers and from uni-directional to multi-directional. We identify research gaps and propose analysis approaches on (1) drivers, (2) modifiers, (3) impacts, (4) feedbacks, and (5) changing the baseline of drought in the Anthropocene. The most pressing research questions are related to the attribution of drought to its causes, to linking drought impacts to drought characteristics, and to societal adaptation and responses to drought. Example questions include (i) What are the dominant drivers of drought in different parts of the world? (ii) How do human modifications of drought enhance or alleviate drought severity? (iii) How do impacts of drought depend on the physical characteristics of drought vs. the vulnerability of people or the environment? (iv) To what extent are physical and human drought processes coupled, and can feedback loops be identified and altered to lessen or mitigate drought? (v) How should we adapt our drought analysis to accommodate changes in the normal situation (i.e. what are considered normal or reference conditions) over time? Answering these questions requires exploration of qualitative and quantitative data as well as mixed modelling approaches. The challenges related to drought research and management in the Anthropocene are not unique to drought, but do require urgent attention. We give recommendations drawn from the fields of flood research, ecology, water management, and water resources studies. The framework presented here provides a holistic view on drought in the Anthropocene, which will help improve management strategies for mitigating the severity and reducing the impacts of droughts in future.

Despite improvements in functional rehabilitation secondary to better control of scar and contractures, aesthetic rehabilitation of the extensively burned face has remained a difficult problem. This study was undertaken to evaluate both technique and aesthetic results of early excision and split thickness autografting (STAG) of full skin thickness face burns. Twenty-five patients with full skin thickness face burns were operated on between days 4 and 14 post-burn. Thirteen patients had excision and STAG in one stage. Twelve patients had a two-stage procedure-excision and coverage with a biological dressing followed 24-72 h later by STAG. Seven of these patients had a pressure dressing in the form of a silicone face mask applied at the second stage. Early cosmetic results were encouraging in all patients. Twenty-five per cent of patients later required either contracture release or skin resurfacing. Preliminary results are encouraging and warrant evaluation by surgeons at other centres. When early excision of full skin thickness face burns is undertaken, cautious optimism as to the ultimate aesthetic result, both by the surgeon and the patient, is advisable.

The multiplicity distributions of charged particles in restricted rapidity intervals in Z0 hadronic decays measured by the DELPHI detector are presented. The data reveal a shoulder structure, best visible for intervals of intermediate size, i.e. for rapidity limits around +/- 1.5. The whole set of distributions including the shoulder structure is reproduced by the Lund Parton Shower model. The structure is found to be due to important contributions from 3- and 4-jet events with a hard gluon jet. A different model, based on the concept of independently produced groups of particles, “clans”, fluctuating both in number per event and particle content per clan, has also been used to analyse the present data. The results show that for each interval of rapidity the average number of clans per event is approximately the same as at lower energies.

The CP-violating asymmetry a(sl)(s), is studied using semileptonic decays of B-s(0) and (B) over bar (0)(s) mesons produced in pp collisions at a centre-of-mass energy of 7 TeV at the LHC, exploiting a data sample corresponding to an integrated luminosity of 1.0 fb(-1). The reconstructed final states are D-s(+/-)mu(+/-)(s), with the D-s(+/-) particle decaying in the phi pi(+/-) mode. The D-s(+/-)mu(+/-)(s) yields are summed over B-s(0) and (B) over bar (0)(s) initial states, and integrated with respect to decay time. Data-driven methods are used to measure efficiency ratios. We obtain a(sl)(s) = (-0.06 +/- 0.50 +/- 0.36)%, where the first uncertainty is statistical and the second systematic. (C) 2013 The Authors. Published by Elsevier B.V. All rights reserved.

- Abstract
Primary ciliary dyskinesia (PCD) is a rare genetic disorder characterised by impaired mucociliary clearance leading to irreversible lung damage. In contrast to other rare lung diseases like cystic fibrosis (CF), there are only few clinical trials and limited evidence-based treatments. Management is mainly based on expert opinions and treatment is challenging due to a wide range of clinical manifestations and disease severity. To improve clinical and translational research and facilitate development of new treatments, the clinical trial network for PCD (PCD-CTN) was founded in 2020 under the framework of the European Reference Network (ERN)-LUNG PCD Core. Applications from European PCD sites interested in participating in the PCD-CTN were requested. Inclusion criteria consisted of patient numbers, membership of ERN-LUNG PCD Core, use of associated standards of care, experience in PCD and/or CF clinical research, resources to run clinical trials, good clinical practice (GCP) certifications and institutional support. So far, applications from 22 trial sites in 18 European countries have been approved, including >1400 adult and >1600 paediatric individuals with PCD. The PCD-CTN is headed by a coordinating centre and consists of a steering and executive committee, a data safety monitoring board and committees for protocol review, training and standardisation. A strong association with patient organisations and industrial companies are further cornerstones. All participating trial sites agreed on a code of conduct. As CTNs from other diseases have demonstrated successfully, this newly formed PCD-CTN operates to establish evidence-based treatments for this orphan disease and to bring new personalised treatment approaches to patients., Competing Interests: Conflict of interest: J. Raidt declares grants for Clinical Research Unit “Male Germ Cells: from Genes to Function” (CRU 326) – Subproject RA3522/1-1 from the Deutsche Forschungsgemeinschaft, in connection with the present manuscript; that they are a holder of patent WO 2020/165352 A1 (Treatment of ciliopathies); and an unpaid role as a Deputy Director of PCD-CTN. - Abstract
Background: Studies in separate cohorts suggest possible discrepancies between inhaled medicines supplied (median 50-60%) and medicines used (median 30-40%). We performed the first study that directly compares CF medicine supply against use to identify the cost of excess medicines supply., Methods: This cross-sectional study included participants from 12 UK adult centres with ≥1 year of continuous adherence data from data-logging nebulisers. Medicine supply was measured as medication possession ratio (MPR) for a 1-year period from the first suitable supply date. Medicine use was measured as electronic data capture (EDC) adherence over the same period. The cost of excess medicines was calculated as whole excess box(es) supplied after accounting for the discrepancy between EDC adherence and MPR with 20% contingency., Results: Among 275 participants, 133 (48.4%) were females and mean age was 30 years (95% CI 29-31 years). Median EDC adherence was 57% (IQR 23-86%), median MPR was 74% (IQR 46-96%) and the discrepancy between measures was median 14% (IQR 2-29%). Even with 20% contingency, mean potential cost of excess medicines was £1,124 (95% CI £855-1,394), ranging from £183 (95% CI £29-338) for EDC adherence ≥80% to £2,017 (95% CI £1,507-2,526) for EDC adherence <50%., Conclusions: This study provides a conservative estimate of excess inhaled medicines supply cost among adults with CF in the UK. The excess supply cost was highest among those with lowest EDC adherence, highlighting the importance of adherence support and supplying medicine according to actual use. MPR provides information about medicine supply but over-estimates actual medicine use., (Copyright © 2021. Published by Elsevier B.V.)

- Abstract
Background: Effects of unit design and shift worked on stress in neonatal intensive care unit (NICU) nurses have not been fully studied., Purpose: To compare stress in NICU nurses who work in single-family room (SFR) or open bay (OBY) units and on nonrotating day or night shift., Methods: Full-time registered nurses (RNs) (n = 72) from a 42-bed SFR and a 131-bed OBY NICU participated in this comparative cross-sectional study. The Nurse Stress Scale (NSS) and within-shift repeated salivary cortisol levels were used to measure stress. The relationship between NSS score and salivary cortisol level was examined using multiple linear regression. Salivary cortisol levels of day versus night shift were compared with mixed-effects linear models., Results: NSS scores were similar for SFR and OBY units (P = .672) and day versus night shift (P = .606). Changes in cortisol level over time (P = .764) and final cortisol level (P = .883) for SFR versus OBY were not significantly different after controlling for shift. Salivary cortisol level of day-shift nurses decreased significantly over time compared with night-shift nurses (P < .001). The final cortisol level was significantly higher for night-shift compared with day-shift nurses (P < .001)., Implications for Practice: Psychological (NSS) and physiologic (salivary cortisol) stress of NICU nurses is similar in established SFR and OBY units. Cortisol levels are higher at the end of shift in nurses who work night shift and may reflect increased physiologic stress., Implications for Research: Strategies are needed for reducing stress in NICU nurses who work night shift., Competing Interests: The authors declare no conflicts of interest., (Copyright © 2021 by The National Association of Neonatal Nurses.)

- Abstract
Background: Lung clearance index (LCI) is a measure of abnormal ventilation distribution derived from the multiple breath inert gas washout (MBW) technique. We aimed to determine the clinical utility of LCI in non-CF bronchiectasis, and to assess two novel MBW parameters that distinguish between increases in LCI due to specific ventilation inequality (LCIvent) and increased respiratory dead space (LCIds)., Methods: Forty-three patients with non-CF bronchiectasis and 18 healthy control subjects underwent MBW using the sulphur hexafluoride wash-in technique, and data from 40 adults with CF were re-analysed. LCIvent and LCIds were calculated using a theoretical two-compartment lung model, and represent the proportional increase in LCI above its ideal value due to specific ventilation inequality and increased respiratory dead space, respectively., Results: LCI was significantly raised in patients with non-CF bronchiectasis compared to healthy controls (9.99 versus 7.28, p < 0.01), and discriminated well between these two groups (area under receiver operating curve = 0.90, versus 0.83 for forced expiratory volume in one second [% predicted]). LCI, LCIvent and LCIds were repeatable (intraclass correlation coefficient > 0.75), and correlated significantly with measures of spirometric airflow obstruction., Conclusion: LCI is repeatable, discriminatory, and is associated with spirometric airflow obstruction in patients with non-CF bronchiectasis. LCIvent and LCIds are a practical and repeatable alternative to phase III slope analysis and may allow a further level of mechanistic information to be extracted from the MBW test in patients with severe ventilation heterogeneity.

- Abstract
Introduction: Deaths from suicide reached a peak in Sri Lanka in 1995. Several interventions reduced the suicide rate of 48.7 per 100,000 in 1995 to 23 per 100,000 in 2006, though it is still a major socioeconomic problem. All suicides have to be reported to the Inquirer of Sudden Death (ISD) or 'Coroner', according to the Criminal Procedure Code., Method: All deaths where a verdict of 'suicide' was given after an inquest at the Coroner's Court, Colombo, in 2006 were studied. Close relations or friends who attended the inquest were interviewed by medically qualified research assistants. Age, sex, marital and occupational status, level of education, living circumstances and method and reasons for the suicide were studied., Results: During 2006, 151 deaths from suicide were documented, of which 93 (62%) were men. The majority (47%) were aged between 20 and 29 years. One-third of the victims was unemployed. At the time of committing suicide, 75% were living with family; 89 (59%) were married and 46 (31%) were single. Poisoning was the cause of death in 66 (44%), 48 (70%) of which were due to pesticides. Burns caused 51 (34%) deaths. Other common causes of death included hanging (11%), jumping in front of a train (7%) and drowning (3%). The commonest reason for suicide was dispute with the spouse/marital disharmony (30%). Other reasons were dispute with parents (8%), financial matters (7%), organic diseases (7%), alcoholism (7%), psychiatric illnesses (6%) and disputes in love affairs (5%). In 29 cases (19%), no definite reason for the suicide was evident., Discussion: Self-poisoning and self-immolation were the commonest methods used to commit suicide. Marital disharmony was the main reason (30%). Psychiatric illnesses were responsible for only 6%. Future interventional activities should include secure access and restriction of the availability of pesticides and drugs, empowering people to manage anger and conflicts, and recognition and treatment of alcoholism and psychiatric illnesses. The success story of the reduction in the incidence of suicides in Sri Lanka should be a lesson to many developing countries where suicide is a major socioeconomic and health issue.

- Abstract
Macrophages in the airways form an important element of immune defense and inflammation. We analyzed induced sputum from airways of patients with CF for the types of macrophages present, their receptor expression, and phagocytic function. In samples from patients and age-matched controls, macrophages were analyzed by multicolor flow cytometry, scavenger receptor expression was studied at the protein and mRNA level, and receptor function was investigated using fluorescent particles. In adult patients with CF, we discovered a pronounced expansion of the small CD14+ DR+ CD68weak+ macrophages to 73 +/- 18% compared with 16 +/- 8% in healthy controls. Expression of the MARCO and CD206 (mannose receptor) was strongly reduced at the mRNA and protein level in sputum macrophages. Antibody-blocking studies showed that MARCO mediates phagocytosis of unopsonized particles. In line with reduced MARCO expression, sputum macrophages in CF showed a deficient uptake of particles (23+/-9% of cells) compared with healthy controls (71+/-15%). The deficiency of MARCO expression in the predominant small sputum macrophages in CF may lead to impaired clearance of inhaled particles with increased inflammation and damage to the CF lung.

- Abstract
Interleukin (IL)-8, the C-X-C chemokine, is a potent neutrophil chemoattractant that has been implicated in a number of inflammatory airway diseases such as cystic fibrosis. Here we tested the hypothesis that bradykinin, an inflammatory mediator and chloride secretagogue, would increase IL-8 generation in airway epithelial cells through autocrine generation of endogenous prostanoids. Bradykinin increased IL-8 generation in both a non-cystic fibrosis (A549) and cystic fibrosis epithelial cell line (CFTE29) that was inhibited by the nonselective cyclooxygenase (COX) inhibitor indomethacin and the COX-2 selective inhibitor NS-398. COX-2 was the only isoform of COX expressed in both cell lines. Furthermore, the COX substrate arachidonic acid and exogenous prostaglandin E(2) both increased IL-8 release in A549 cells. These results suggest that bradykinin may contribute to neutrophilic inflammation in the airway by generation of IL-8 from airway epithelial cells. The dependence of this response on endogenous production of prostanoids by COX-2 suggests that selective COX-2 inhibitors may have a role in the treatment of airway diseases characterized by neutrophilic inflammation such as cystic fibrosis or chronic obstructive pulmonary disease.

- Abstract
The in vitro production of juvenile hormones (JH) was investigated by using corpora allata (CA) of larvae and corpora cardiaca-corpora allata (CC-CA) complexes of adult females of the fall armyworm Spodoptera frugiperda. In female moths, JH release was high compared to that in 5th and 6th instar larvae. Concentrations of 0.11-0.12 mM methionine, 180-200 mM Na(+), 5.8-8.3 mM K(+), 10-50 mM Ca(2+) and a pH range of 5.7-6.3 yielded optimal incorporation of L-[methyl-(3)H] methionine in vitro by CC-CA complexes. The highest hourly incorporation occurred during a 9-h incubation period following a 1.5-h lag-phase. JH release from CC-CA complexes of adult females was shown to be age-dependent with a peak value on day 2 (approx. 4 pmol h(-1) CA(-1)). By a combination of reversed phase (RP)- and normal phase (NP)-high performance liquid chromatography (HPLC), two major labelled products released by the complex were separated. One compound co-migrated with chemically synthesized JH II diol, the second compound with JH III diol. Only traces of JH II and III could be detected in some samples. Gland extracts also contained both the major radiolabelled products. Double labelling experiments using [3H]methionine and [14C]acetate confirmed their de novo synthesis in CC-CA complexes of female moths. The nature of chemically synthesized reference JH III diol was proved by LC-MS (ESI mass spectrometry) and 1H-NMR (nuclear magnetic resonance spectroscopy).

- Abstract
Studies were undertaken to investigate vitellogenesis and its regulation in female adults of the fall armyworm, Spodoptera frugiperda. A single female-specific protein, likely to be the S. frugiperda vitellogenin (Vg), appeared approximately 5 h after adult eclosion in the hemolymph of virgin females. The concentration of the protein increased with age as sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) revealed. A protein with the same relative molecular mass was also present in egg extracts, but absent from hemolymph samples from male moths. The relative molecular mass of the designated S. frugiperda Vg was determined as 164.5+/-2.5 kDa. Vitellogenic oocytes became visible 36-48 h after emergence and egg deposition began on day 3 of adult life. Vg could not be detected in the hemolymph of females decapitated directly after eclosion. When decapitated virgin females were injected with the JH-mimic methoprene (MP), the level of Vg was comparable to that in non-decapitated moths, indicating that vitellogenesis in S. frugiperda depends on juvenile hormone (JH). However, the number of vitellogenic oocytes was somewhat lower than in non-decapitated virgin females. Injection of 20-hydroxyecdysone (20E) promoted Vg production to a similar extent in decapitated female moths, but in contrast to methoprene injection, treatment with 20E never resulted in the production of vitellogenic oocytes. In vitro cultivated ovaries of adult females dissected directly after eclosion produced lower amounts of ecdysteroids than those isolated on day 1 after emergence. Our results suggest a crucial role for 20E in the induction of vitellogenesis in the noctuid S. frugiperda, while JH seems to be essential for the continued uptake of Vg by developing oocytes and may trigger 20E biosynthesis in the ovary.

- Abstract
Although guanosine 3',5'-cyclic monophosphate (cGMP) acts as a relaxant second messenger, the regulation of intracellular cGMP has not been comprehensively studied in human airway smooth muscle. We studied the production of cGMP by cultured human airway smooth muscle cells (HASMC) after stimulation with activators of soluble guanylyl cyclase [sodium nitroprusside (SNP) and S-nitroso-N-acetylpenicillamine (SNAP)] and particulate guanylyl cyclase [atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP), C-type natriuretic peptide (CNP), and Escherichia coli heat stable enterotoxin (STa)]. cGMP was measured by enzyme-linked immunosorbent assay. Both SNP (10(-6) to 10(-3) M) and SNAP (10(-6) to 10(-3) M) caused concentration-dependent elevation of cGMP in the presence of the nonselective phosphodiesterase (PDE) inhibitor 3-isobutyl-1-methylxanthine (10(-3) M), with cGMP increasing 6- and 15-fold in response to SNP and SNAP, respectively, at the highest concentration tested (10(-3) M). The increases in cGMP in response to SNP (5 x 10(-5) M) and SNAP (10(-5) M) were inhibited by hemoglobin (Hb; 5 x 10(-5) M), a nitric oxide scavenger, and methylene blue (MB; 5 x 10(-4) M), an inhibitor of guanylyl cyclase. cGMP accumulation after SNAP was abolished by both Hb and MB. The response to SNP was inhibited by 79% with Hb and was abolished with MB. ANP, BNP, and CNP (10(-9) to 10(-5) M) + phosphoramidon (10(-6) M) caused a concentration-dependent elevation in cGMP with an order of potency ANP > BNP > CNP. cGMP formation in the presence of the highest concentration of the most potent natriuretic peptide (10(-5) M ANP) was two- to threefold greater than with the highest concentration of SNAP. The increase in cGMP seen with natriuretic peptides was similar in the presence or absence of phosphoramidon, a neutral endopeptidase (NEP) inhibitor, suggesting that NEP is not playing a role in modulating the effect of natriuretic peptides in HASMC. STa (400 IU/ml) had no effect on cGMP levels. SNAP- and ANP-induced cGMP accumulation was increased by the selective type V PDE inhibitors SKF-96231 and zaprinast, suggesting that type V PDE is responsible for cGMP breakdown in HASMC. These results suggest that cultured HASMC contain both soluble and particulate guanylyl cyclases. The order of potency of the natriuretic peptides ANP > BNP > CNP suggests that type A particulate membrane-bound guanylate cyclase predominates.

- Abstract
The basolateral Na+/K+ ATPase plays a critical role in sodium reabsorption across airway epithelium. Nasal epithelium shows increased Na+/K+ ATPase activity in cystic fibrosis (CF) but Na+/K+ ATPase has not been characterized in human lung epithelium or compared in CF and normal lung. We measured 3[H] ouabain binding and Na+/K+ ATPase activity in human tracheal epithelium and compared Na/K ATPase activity in bronchial epithelium in CF and control subjects. In tracheal epithelium Na+/K+ ATPase pumps were abundant and of high 3[H] ouabain affinity (Kd 4.7 nM, Bmax 38 pmol/mg) and Na+/K+ ATPase activity was 55 +/- 8 nmol/mg protein/min. Bronchial epithelial Na+/K+ ATPase activity was twofold higher in CF patients than in controls. The increased Na+/K+ ATPase activity may contribute to the increased sodium reabsorption seen in cystic fibrosis.
