This is the first time that continuous assessment of CH4 is reported in parallel with measurements of O2 consumption and CO2 production inside a whole room indirect calorimeter in humans and over 24 h. We provide a detailed description of the whole system and its parts. We carried out studies of reliability and validity of the whole system and its parts. CH4 is released in humans during daily activities.

Ocean warming is increasing the incidence, scale, and severity of global-scale coral bleaching and mortality, culminating in the third global coral bleaching event that occurred during record marine heatwaves of 2014-2017. While local effects of these events have been widely reported, the global implications remain unknown. Analysis of 15,066 reef surveys during 2014-2017 revealed that 80% of surveyed reefs experienced significant coral bleaching and 35% experienced significant coral mortality. The global extent of significant coral bleaching and mortality was assessed by extrapolating results from reef surveys using comprehensive remote-sensing data of regional heat stress. This model predicted that 51% of the world’s coral reefs suffered significant bleaching and 15% significant mortality, surpassing damage from any prior global bleaching event. These observations demonstrate that global warming’s widespread damage to coral reefs is accelerating and underscores the threat anthropogenic climate change poses for the irreversible transformation of these essential ecosystems.

The extension of life span driven by 40% caloric restriction (CR) in rodents causes trade-offs in growth, reproduction, and immune defense that make it difficult to identify therapeutically relevant CR-mimetic targets. We report that about 14% CR for 2 years in healthy humans improved thymopoiesis and was correlated with mobilization of intrathymic ectopic lipid. CR-induced transcriptional reprogramming in adipose tissue implicated pathways regulating mitochondrial bioenergetics, anti-inflammatory responses, and longevity. Expression of the gene Pla2g7 encoding platelet activating factor acetyl hydrolase (PLA2G7) is inhibited in humans undergoing CR. Deletion of Pla2g7 in mice showed decreased thymic lipoatrophy, protection against age-related inflammation, lowered NLRP3 inflammasome activation, and improved metabolic health. Therefore, the reduction of PLA2G7 may mediate the immunometabolic effects of CR and could potentially be harnessed to lower inflammation and extend the health span.

On 22 May 2017 Salman Abedi detonated an improvised explosive device in the Manchester Arena resulting in 23 deaths (including the attacker). This was the deadliest terrorist attack on UK soil since the 2005 London bombings, but was only one of five mass casualty terrorist attacks in the UK in 2017. Preparation for mass casualty incidents (MCI) is obligatory, involving such methods as multiagency tabletop exercises, mock hospital exercises, as well as simulation and training for clinicians in managing the injuries that would be anticipated in such an event. Even in the best prepared units, such an incident will pose significant challenges due to the unpredictable nature of these events with respect to timing and number of casualties. Following an MCI, local and national reviews are undertaken to assess the effectiveness of the response, but also to identify areas where lessons can be learnt and to disseminate these to allow inclusion in future planning. We present the experience following a mass casualty terrorist incident along with a number of lessons learnt from this event.

Background The optimal timing of cholecystectomy for patients admitted with acute gallbladder pathology is unclear. Some studies have shown that emergency cholecystectomy during the index admission can reduce length of hospital stay with similar rates of conversion to open surgery, complications and mortality compared with a ‘delayed’ operation following discharge. Others have reported that cholecystectomy during the index acute admission results in higher morbidity, extended length of stay and increased costs. This study examined the cost-effectiveness of emergency versus delayed cholecystectomy for acute benign gallbladder disease. Methods Using data from a prospective population-based cohort study examining the outcomes of cholecystectomy in the UK and Ireland, a model-based cost–utility analysis was conducted from the perspective of the UK National Health Service, with a 1-year time horizon for costs and outcomes. Probabilistic sensitivity analysis was used to investigate the impact of parameter uncertainty on the results obtained from the model. Results Emergency cholecystectomy was found to be less costly (£4570 versus £4720; €5484 versus €5664) and more effective (0·8868 versus 0·8662 QALYs) than delayed cholecystectomy. Probabilistic sensitivity analysis showed that the emergency strategy is more than 60 per cent likely to be cost-effective across willingness-to-pay values for the QALY from £0 to £100 000 (€0–120 000). Conclusion Emergency cholecystectomy is less costly and more effective than delayed cholecystectomy. This approach is likely to be beneficial to patients in terms of improved health outcomes and to the healthcare provider owing to the reduced costs.

Five taxa of talitrid amphipods were found in the archipelago of Bermuda, of which three were recorded there for the first time. Four of these are supralittoral wrack generalists: Platorchestia monodi BOLD:AAB3402, (a unique Molecular Operational Taxonomic Unit according to the Barcode Index Number system), a related species recognized by molecular methods, Platorchestia platensis BOLD:AAA2949, Mexorchestia carpenteri carpenteri BOLD:AAC1491 and Tethorchestia antillensis; and one a terrestrial leaf-litter generalist: Talitroides alluaudi. A key is provided to discriminate between the formally described talitrids of Bermuda. Organic chemicals discharged in urban wastewater from industrial and domestic sources, or those entering through atmospheric deposition onto paved areas via surface run-off, are predominantly lipophilic in nature and therefore become concentrated in sewage sludge, with potential implications for the agricultural use of sludge as a soil improver. Biodegradation occurs to varying degrees during wastewater and sludge treatment processes. However, residues will probably still be present in the resulting sludge and can vary from trace values of several micrograms per kilogram up to approximately 1 per cent in the dry solids for certain bulk chemicals, such as linear alkylbenzene sulphonate, which is widely used as a surfactant in detergent formulations. However, the review of the scientific literature on the potential environmental and health impacts of organic contaminants (OCs) in sludge indicates that the presence of a compound in sludge, or of seemingly large amounts of certain compounds used in bulk volumes domestically and by industry, does not necessarily constitute a hazard when the material is recycled to farmland. Furthermore, the chemical quality of sludge is continually improving and concentrations of potentially harmful and persistent organic compounds have declined to background values. Thus, recycling sewage sludge on farmland is not constrained by concentrations of OCs found in contemporary sewage sludges. A number of issues, while unlikely to be significant for agricultural utilization, require further investigation and include: (i) the impacts of chlorinated paraffins on the food chain and human health, (ii) the risk assessment of the plasticizer di(2-ethylhexyl)phthalate, a bulk chemical present in large amounts in sludge, (iii) the microbiological risk assessment of antibiotic-resistant micro-organisms in sewage sludge and sludge-amended agricultural soil, and (iv) the potential significance of personal-care products (e.g. triclosan), pharmaceuticals and endocrine-disrupting compounds in sludge on soil quality and human health.

To examine five available software packages for the assessment of abdominal adipose tissue with magnetic resonance imaging, compare their features and assess the reliability of measurement results. Feature evaluation and test–retest reliability of softwares (NIHImage, SliceOmatic, Analyze, HippoFat and EasyVision) used in manual, semi-automated or automated segmentation of abdominal adipose tissue. A random sample of 15 obese adults with type 2 diabetes. Axial T1-weighted spin echo images centered at vertebral bodies of L2–L3 were acquired at 1.5 T. Five software packages were evaluated (NIHImage, SliceOmatic, Analyze, HippoFat and EasyVision), comparing manual, semi-automated and automated segmentation approaches. Images were segmented into cross-sectional area (CSA), and the areas of visceral (VAT) and subcutaneous adipose tissue (SAT). Ease of learning and use and the design of the graphical user interface (GUI) were rated. Intra-observer accuracy and agreement between the software packages were calculated using intra-class correlation. Intra-class correlation coefficient was used to obtain test–retest reliability. Three of the five evaluated programs offered a semi-automated technique to segment the images based on histogram values or a user-defined threshold. One software package allowed manual delineation only. One fully automated program demonstrated the drawbacks of uncritical automated processing. The semi-automated approaches reduced variability and measurement error, and improved reproducibility. There was no significant difference in the intra-observer agreement in SAT and CSA. The VAT measurements showed significantly lower test–retest reliability. There were some differences between the software packages in qualitative aspects, such as user friendliness. Four out of five packages provided essentially the same results with respect to the inter- and intra-rater reproducibility. Our results using SliceOmatic, Analyze or NIHImage were comparable and could be used interchangeably. Newly developed fully automated approaches should be compared to one of the examined software packages.

Argon ions were implanted into n-type 6H-SiC epitaxial layers at 600°C. Postimplantation annealing was carried out at 1,600°C for 5 min in an Ar ambient. Four implantation-induced defect levels were observed at EC-0.28 eV, EC-0.34 eV, EC-0.46 eV, and EC-0.62 eV by deep level transient spectroscopy. The defect center at EC-0.28 eV is correlated with ED1/ED2 and with ID5. The defect at EC-0.46 eV with a capture cross section of 7.8 × 10−16 cm2 is correlated with E1/E2, while the defect at EC-0.62 eV with a capture cross section of 2.6 × 10−14 cm2 is correlated with Z1/Z2. Photo deep level transient spectroscopy was also used to study these defects. Upon illumination, the amplitudes of the deep level transient spectroscopy (DLTS) peaks increased considerably. Two emission components of Z1/Z2 were revealed: one fast and the other slow. The fast component could only be observed with a narrow rate window. In addition, a new defect was observed on the low-temperature side of the defect at EC-0.28 eV when the sample was illuminated.

Aim: To determine whether application of a periodontal dressing stopped pain arising from dentine hypersensitivity, objectively assessed with evaporative and thermal stimuli and recorded with Visual Analogue Scale scoring (VAS). Materials and methods: 22 subjects completed the single-centre, subject-blind, stratified, randomized, split-mouth study, with a minimum of two sensitive teeth, in at least two different quadrants, displaying a response of ≥30 mm with VAS to evaporative stimulus. One tooth in two different quadrants was identified and randomized to test or control groups. A dressing was applied to all the sensitive teeth in the test quadrant, and either side of the chosen sensitive tooth on the control side. The test teeth were then stimulated for hypersensitivity using evaporative stimuli and then using thermal stimuli. The United Kingdom National Health Service treats both elective and emergency patients and seeks to provide high quality care, free at the point of delivery. Equal numbers of emergency and elective general surgical procedures are performed, yet surgical training prioritisation and organisation of NHS institutions is predicated upon elective care. The increasing ratio of emergency general surgery consultant posts compared to traditional sub-specialities has yet to be addressed. How should the capability gap be bridged to equip motivated, skilled surgeons of the future to deliver a high standard of emergency surgical care? The aim was to address both training requirements for the acquisition of necessary emergency general surgery skills, and the formation of job plans for trainee and consultant posts to meet the current and future requirements of the NHS. Twenty nine trainees and a consultant emergency general surgeon convened as a Working Group at The Association of Surgeons in Training Conference, 2015, to generate a united consensus statement to the training requirement and delivery of emergency general surgery provision by future general surgeons. Unscheduled general surgical care provision, emergency general surgery, trauma competence, training to meet NHS requirements, consultant job planning and future training challenges arose as key themes. Recommendations have been made from these themes in light of published evidence. Careful workforce planning, education, training and fellowship opportunities will provide well-trained enthusiastic individuals to meet public and societal need.

We describe a digital system that allows people to have an interactive conversation with a human storyteller (a Holocaust survivor) who has recorded a number of dialogue contributions, including many compelling narratives of his experiences and thoughts. The goal is to preserve as much as possible of the experience of face-to-face interaction. The survivor’s stories, answers to common questions, and testimony are recorded in high fidelity, and then delivered interactively to an audience as responses to spoken questions. People can ask questions and receive answers on a broad range of topics including the survivor’s experiences before, after and during the war, his attitudes and philosophy. Evaluation results show that most user questions can be addressed by the system, and that audiences are highly engaged with the resulting interaction.

Aims: A single-centre, randomized single-blind parallel study was undertaken to compare staining seen with three brushing regimens and to determine subject perception of side effects such as staining and effects on taste. Methods: This 6-week parallel study used 157 volunteers who were randomized into one of three treatment groups: (i) brushing in the morning and evening with a normal dose of a 1% chlorhexidine gel, (ii) brushing with a low dose of chlorhexidine gel in the evening and a whitening dentifrice in the morning, and (iii) brushing with a standard fluoride paste in the morning and evening. Following home usage of their allocated products, the study volunteers returned after 3 and 6 weeks to record the amount of stain present. After the 6-week period, subject perception of taste and stain acceptability was determined using a questionnaire. Differentiation of embryonic stem (ES) cells generally occurs after formation of three-dimensional cell aggregates, known as embryoid bodies (EBs). This differentiation occurs following suspension culturing of EBs in media containing a high (25 mM) glucose concentration. Although high-glucose-containing media is used for maintenance and proliferation of ES cells, it has not been demonstrated whether this is a necessary requirement for EB development. To address this, we examined the growth and differentiation of EBs established in 0-mM, 5.5-mM (physiological), and 25-mM (high) glucose concentrations, through morphometric analysis and examination of gene and protein expression. The effect on EB development of supplementation with basic fibroblast growth factor (FGF2) was also studied. We report that the greatest rate of EB growth occurs in 5.5 mM glucose media. A morphological study of EBs over 104 days duration under glucose-containing conditions demonstrated the development of all three major embryonic cell types. The difference from normal human development was obvious in the lack of rostrocaudal control by the notochord. In the latest stages of development, the main tissue observed appeared to be cartilage and cells of a mesodermal lineage. We conclude that physiological glucose concentrations are suitable for the culturing of EBs, that the addition of FGF2 enhances the temporal expression of genes including POU5F1, nestin, FOXA2, ONECUT1, NEUROD1, PAX6, and insulin, and that EBs can be cultured in vitro for long periods, allowing for further examination of developmental processes. developmental biology, early development, embryo, gene regulation

A high spatial resolution Ar–Ar dating study of compositionally zoned micas using UV laser extraction has been used to investigate the effect of composition and compositional boundaries upon argon diffusion in mica. The crystals are characterised by muscovite cores and Li–F–Rb–Cs-rich lepidolite rims produced by the interaction of a residual melt-vapour phase with the original muscovite during the late stages of pegmatite crystallisation. Single crystals exhibit dramatic intra-grain age variations, with a maximum range of 2,880–2,117 Ma. Backscattered electron images combined with UV-laser analyses reveal that apparent ages younger than 2,600 Ma are directly associated with the compositionally distinct rims, suggesting that Ar diffusion was faster in the lepidolite than in muscovite. Although it is not possible to be fully quantitative using the present data, it seems that argon diffusion rates in lepidolite are similar to those in biotite. Major rearrangements of the crystal structure needed to accommodate high concentrations of lithium, fluorine and large interlayer cations (Rb+, Cs+) in muscovite account for lower argon retentivity of the crystal rims. In most cases the age profiles show no disturbance at the compositional boundary, despite the transition from a di-octahedral to tri-octahedral mica, indicating that in general the boundaries do not act as fast diffusion pathways. However, in some cases there is a clear drop in ages at the compositional boundary, which acts as an effective sub-grain boundary. The difference between the two types may be related to the internal structure of the compositional boundaries or their degree of contact with the grain boundary network of the rock.

Summary Biosolids are used extensively as effective soil conditioners and organic fertilisers in both farming and reclamation of ex-industrial and brownfield sites. However, there has been little attempt to introduce biosolids into amenity horticultural practices. A field trial with Betula pendula Roth. (Silver Birch) carried out at the Imperial College London Field Centre, Silwood Park, Ascot has demonstrated the nutrient value and soil conditioning properties of biosolids to trees. In addition to organic matter, they also provide substantial amounts of plant available nitrogen (N), phosphate (P), sulphur (S) and micronutrients. Composted biosolids also contain significant amounts of potassium (K). Enhanced treated biosolids have potential for application to areas where there is significant public activity, as these biosolids are treated to destroy microbial pathogens. It is widely accepted that increasing adiposity is associated with insulin resistance and increased risk of type II diabetes. The predominant paradigm used to explain this link is the portal/visceral hypothesis. This hypothesis proposes that increased adiposity, particularly in the visceral depots, leads to increased free-fatty acid flux and inhibition of insulin-action via Randle's effect in insulin-sensitive tissues.In this review, limitations of this paradigm will be discussed and two other paradigms that may explain established links between adiposity and insulin resistance/diabetes will be presented. (A) Ectopic fat storage syndrome. Three lines of evidence support this concept. Firstly, failure to develop adequate adipose tissue mass (also known as 'lipodystrophy') results in severe insulin resistance and diabetes. This is thought to be the result of ectopic storage of lipid into liver, skeletal muscle and the pancreatic insulin-secreting beta cell. Secondly, most obese patients also shunt lipid into the skeletal muscle, the liver and probably the beta cell. The importance of this finding is exemplified by several studies demonstrating that the degree of lipid infiltration into skeletal muscle and liver highly correlates with insulin resistance. Thirdly, increased fat cell size is highly associated with insulin resistance and the development of diabetes. Increased fat cell size may represent the failure of the adipose tissue mass to expand and therefore to accommodate an increased energy influx. Taken together, these observations support the 'acquired lipodystrophy' hypothesis as a link between adiposity and insulin resistance. Ectopic fat deposition is therefore the result of additive or synergistic effects including increased dietary intake, decreased fat oxidation and impaired adipogenesis. (B) Endocrine paradigm. This concept was developed in parallel with the 'ectopic fat storage syndrome' hypothesis. Adipose tissue secretes a variety of endocrine hormones such as leptin, interleukin-6, angiotensin II, adiponectin and resistin. From this viewpoint, adipose tissue plays a critical role as an endocrine gland, secreting numerous factors with potent effects on the metabolism of distant tissues.The novel paradigms of ectopic fat and fat cell as an endocrine organ probably will constitute a new framework for the study of the links between our obesigenic environment and the risk of developing diabetes.

Early reports suggested that resistin is associated with obesity and insulin resistance in rodents. However, subsequent studies have not supported these findings. To our knowledge, the present study is the first assessment in human subjects of serum resistin and insulin sensitivity by the insulin clamp technique. Thirty-eight nonobese subjects [age, 23 ± 4 yr; body mass index (BMI), 25.4 ± 4.3 kg/m2], 12 obese subjects (age, 54 ± 8 yr; BMI, 33.0 ± 2.5 kg/m2), and 22 obese subjects with type 2 diabetes (age, 59 ± 7 yr; BMI, 34.0 ± 2.4 kg/m2) were studied. Serum resistin concentrations were not different among nonobese (4.1 ± 1.7 ng/ml), obese (4.2 ± 1.6 ng/ml), and obese diabetic subjects (3.7 ± 1.2 ng/ml), and were not significantly correlated to glucose disposal rate during a hyperinsulinemic glucose clamp across groups. Serum resistin was, however, inversely related to insulin sensitivity in nonobese subjects only (r = −0.35; P = 0.05), although this association was lost after adjusting for percent body fat. Serum resistin was not related to percent fat, BMI, or fat cell size. A strong correlation was observed between serum resistin and resistin mRNA expression from abdominal sc adipose tissue in a separate group of obese subjects (r = 0.62; P < 0.01; n = 56). Although the exact function of resistin is unknown, we demonstrated only a weak relationship between resistin and insulin sensitivity in nonobese subjects, indicating that resistin is unlikely to be a major link between obesity and insulin resistance in humans.

U-Pb isotopic analyses using TIMS and Laser Ablation-Multi Collector-ICP-MS were carried out on columbite-tantalite minerals from three suites of rare-element (Li, Cs, Ta) pegmatites in the Superior Province of Canada. Creeping bentgrass (Agrostis stolonifera L.) is a high-value turfgrass species that offers opportunities for western Canadian seed companies and producers. The objective of this study was to evaluate creeping bentgrass seed yield and the relationship between seedhead density and seed yield across a diverse range of cultivars. A series of four trials was established in Manitoba between 1992 and 1994 and included 18th Green, Cobra, Emerald, National, Penneagle, Pennlinks, Putter, Southshore, UM86-01 and UM86-02. Seed production and seedhead density were measured for 2–3 yr at each location. Additional yield component measurements included harvest index and thousand seed weight. The three cultivars developed within the region consistently showed high seed yield, seedhead density and harvest index and were numerically ranked in the top four for seed yield across all 11 location years. UM86-01 was in the highest grouping for seed yield across all harvests with yields ranging from 312 to 582 kg ha-1. Seedhead density was strongly related to seed yield (highly significant in five of six comparisons) and therefore has the potential to be a valuable indicator of relative seed yield in creeping bentgrass. These results indicate that creeping bentgrass seed yield is variable across cultivars and that seedhead density can serve as a good predictor of seed production potential across cultivars. A range of native grass species evolved under the environmental extremes of a continental climate in the northern Great Plains, but most have not been evaluated for their suitability as turf. The objective of this research was to evaluate the turfgrass potential of a range of native grasses under three mowing heights. Twelve species (a total of 28 entries) were evaluated under three mowing heights (62, 38, and 18 mm), for turf quality, color, and density at two locations in Manitoba, Canada. The species evaluated included alpine bluegrass (Poa alpina L.), alkali grass [Puccinellia nuttalliana (Schult.) Hitchc.], alpine fescue [Festuca ovina L. var. brachyphylla (Schult. & Schult. f.) Piper, syn. F. brachyphylla Schult. & Schult. f.], blue grama [Bouteloua gracilis (Kunth) Lag. ex Griffiths], buffalograss {Buchloe dactyloides (Nutt.) Engelm. [= Bouteloua dactyloides (Nutt.) Columbus]} Canada bluegrass (Poa compressa L.), fescue spp., fowl bluegrass (Poa palustris L.), Idaho bentgrass (Agrostis idahoensis Nash), marsh muhly [Muhlenbergia racemosa (Michx.) Britton et al.], prairie junegrass [Koeleria macrantha (Ledeb.) Schult.], rough hairgrass {Agrostis scabra Willd. [= A. hyemalis var. scabra (Willd.) H. L. Blomq.]), side-oats grama [Bouteloua curtipendula (Michx.) Torr.], and tufted hairgrass [Deschampsia cespitosa (L.) P. Beauv.]. Entries showed similar results across all mowing heights. 'Bad River' blue grama, Minnesota ecotype blue grama, and 'Barkoel' prairie junegrass entries showed high quality ratings across all years and locations. Blue grama, a warm-season grass, was drought tolerant and maintained consistent green color throughout the growing season. Although a number of the species showed promise, most entries will require a breeding and selection program before release as low-maintenance turfgrasses.

Nitrogen doped 4H–SiC and 6H–SiC epitaxial layers with net doping concentration of 1.5×1015 cm−3 were implanted with either Al, B, or Ar ions at 600 °C. The energy of the ions was 160 keV and at a dose of 2×1016 cm−2. After annealing at 1600 °C for 5–60 min, Schottky diodes were fabricated on the samples. The measured junction capacitance of ion implanted samples varies with both temperature and frequency. The thermal activation energy determined from the frequency break ωB ranges from 183 to 202 meV. The Ar implanted samples remain n-type and show no capacitive dispersion. Similarly, the junction capacitance of samples doped with Al does not exhibit dispersion. It is suggested that the implantation induced defects play a role in the observed dispersion. The data show that the series resistance is insignificant and does not account for the observed dispersion.

This paper describes a soil-incubation study which was carried out to examine the effects of (a) phosphorus removal during the treatment of domestic sewage and (b) treating sewage sludge to an enhanced status, on the phosphorus fertiliser value of biosolids which are used in agriculture. Phosphorus-enriched and conventional dewatered digested biosolids, thermally-dried pellets and granules, and digested and thermally-hydrolysed liquid sludges were incorporated into two soil types with contrasting physico-chemical properties. Plant-available phosphorus in soil was assessed after incubation by a standard chemical-extraction procedure. Sludge from biological-P removal had the highest phosphorus extract,ability in soil whereas iron dosing slightly reduced or had no effect on phosphorus release from conventionally-treated biosolids, depending upon the type of soil. High-temperature drying significantly reduced the extractable-P content in sludge by 20-60%, compared with dewatered digested cake, and release from thermally-dried biosolids declined further with iron enrichment. Extractable-P recovery was greater from all types of biosolids when mixed with calcareous soil, compared with a loamy sand. A preliminary investigation of phosphorus mineralogy in biosolids, using advanced analytical techniques, is also described.

The continuing development of adapted alfalfa (Medicago sativa L.) cultivars that combine high yield, disease resistance, winterhardiness, and grazing tolerance is of great value to the beef cattle industry in western Canada. This study evaluated alternative management strategies and plant measurements for assessing the grazing tolerance of alfalfa cultivars over 3 yr. Seven hay-type, four pasture-type, and one dual-purpose (hay or graze) cultivars were established in pure stands and in mixture with meadow bromegrass (Bromus riparius Rehm. syn.B. biebersteinii Roem & Schult). Stands were defoliated under continuous or rotational stocking with beef cattle (Bos taurus). Plants we reassessed for winterhardiness, percent alfalfa within row, percent alfalfa cover, basal area and species composition. Continuous stocking resulted in consistent and significant differences between cultivars, whether persistence was measured as percent alfalfa within row, percent cover and basal area, in both pure and mixed stands (P < 0.05). High correlations between measurement techniques (r = 0.74 to 0.99; P < 0.05) confirmed the value of alfalfa within row and alfalfa cover as time and labour efficient methods to evaluate grazing persistence. Competition between alfalfa and meadow bromegrass resulted in a significant confounding effect under rotational stocking (P < 0.05). Winter injury was identified as an important criteria to evaluate for grazing persistence. The most grazing-tolerant genotypes were selected for subsequent breeding efforts to combine winterhardiness, grazing tolerance, and high productivity. This research showed that grazing method, grass competition, and severe winter conditions strongly influence differential survival of alfalfa cultivars under grazing in western Canada. Organogenesis requires tissue interactions to initiate the cascade of inductive and repressive signals necessary for normal organ development. Tissue interactions initiate the pancreatic lineage within the primitive foregut endodermal epithelium and continue to direct the morphogenesis and differentiation of the endocrine, exocrine and ductal portions of the pancreas. An understanding of the mechanisms controlling pancreatic growth would enable the development of alternative therapies for diseases such as type 1 diabetes.

Single-year hay alfalfas (Medicago sativa L.), berseem (Trifolium alexandrinum L.) and red clovers (Trifolium pratense L.), chickling vetch (Lathyrus sativus L.) and lentil (Lens culinaris Medik.) were evaluated for rotational yield and N benefits to the following first-year wheat (Triticum aestivum L.) and second-year barley (Hordeum vulgare L.) crops. Field experiments were initiated in 1997 and 1998 on a Riverdale silty clay soil at Winnipeg, Manitoba. Yield and N content of the following wheat crop were increased following legumes compared to wheat following a canola control. Wheat yield and N content averaged 2955 kg ha–1 and 76.1 kg ha–1, respectively, following the chickling vetch and lentil, 2456 kg ha–1 and 56.4 kg ha–1 following single-year hay legumes, compared with 1706 kg ha–1 and 37.9 kg ha–1 following canola. Non-dormant alfalfas (dormancy rating of eight or greater) contributed to larger grain yields than the dormant alfalfas only in the first year of each experiment. The chickling vetch and lentil provided similar or higher subsequent crop yields and N content for 2 yr compared to a canola control or fallow treatment. This study shows that some increase in yield can be achieved by using a single-year alfalfa hay crop instead of fallow; however, exclusive green manuring of chickling vetch and lentil crops can produce the most increase in yield and N uptake in subsequent crops. BACKGROUND/AIMS The aim of the study was to investigate the clinical and microbiological effects of azithromycin as an adjunct to the non-surgical treatment of periodontitis in adults. Azithromycin is an antibiotic which is taken up by phagocytes and is released over long periods in inflamed tissue but requires a total of only three doses of 500 mg to produce its therapeutic effect. METHOD 46 patients were treated in a double-blind placebo-controlled study with assessments at weeks 0, 1, 2, 3, 6, 10 and 22. Throughout the trial measurements were made of plaque, gingival bleeding, calculus, probing pocket depths and bleeding on probing. Microbiological sampling was carried out from a selected pocket >or=6 mm at each visit. The regime employed consisted of OHI, scaling and root planing at weeks 0, 1 and 2 with reinforcement of OHI and minimal scaling at weeks 6, 10 and 22. Patients were randomly assigned to receive either azithromycin, (A), or placebo capsules, (C), 500 mg, 1x daily for 3 days at week 2. 44 patients completed the study. Mean pocket depths were analysed using analysis of covariance in 3 groups with initial pocket depth values of 1-3 mm, 4-5 mm and >or=6 mm. RESULTS The results of the microbiology have been reported in a separate paper. The clinical data showed that by week 22 a lower % of pockets initially >5 mm deep remained above that level in the 23 patients taking azithromycin (A), than the 21 taking the placebo (C), (A, 5.6%; C, 23.3%). Also at week 22, for pockets initially 4 mm or more, the test group had fewer pockets >3 mm deep (A, 26.1%; C, 44.3%), fewer failing to improve in probing depth (A, 6.6%; C, 21.6%) and fewer continuing to bleed on probing (A, 46.9%; C, 55.6%) when compared with the control group. Layered oxide materials having alternating repeated layer thicknesses of 10 nm or less are difficult to make, especially with sharp interfaces. Nanostructured thin films having repeated layers of two different oxide materials were obtained by using pulsed laser deposition and two independent stationary targets consisting of Al 2 O 3 and BaTiO 3 . Desired thicknesses were achieved by using a specific number of pulses from a 248-nm KrF excimer laser, at an energy of 450 mJ / pulse , a galvanometer mirror system, and a background pressure of oxygen. Trends in material properties were identified by systematically varying the number of pulses for multiple nanostructured thin films and comparing the resulting properties measured using in-situ spectroscopic ellipsometry and ex-situ capacitance measurements, including relative permittivity and loss. Four films were deposited with a goal of having 0.25-, 1-, 4-, and 10-nm thick layers, and each ∼ 220 nm thick. Ellipsometry data were modeled in situ to calculate thickness, n and k . A representative transmission electron microscopy measurement was also collected for the 10-nm sample with corresponding x-ray photoelectron spectroscopy and energy disperive x-ray spectroscopy. Ellipsometry and capacitance measurements were all performed on each of the samples, with one sample having calculated impedance greater than 30 GOhm at 0.001 Hz.

In the past years, the number of mindfulness-based intervention and prevention programs has increased steadily. In order to achieve the intended program outcomes, program implementers need to understand the essential and indispensable components that define a program's success. This chapter describes the complex process of identifying the core components of a mindfulness and yoga program for urban early adolescents through the systematic study of fidelity of implementation of the intervention. The authors illustrate the CORE Process [(C) Conceptualize Core Components; (O) Operationalize and measure; (R) Run analyses and Review implementation findings; and (E) Enhance and refine], based on data gained from a mindfulness and yoga intervention study conducted as a community-academic partnership in Baltimore city.

BackgroundLisfranc injury is rare and the diagnosis maybe easily missed. This study reviews the experience of a single centre.MethodsA prospective review of patients with Lisfranc injuries presenting to a single surgeon with a specialist interest in foot and ankle surgery over a one year period.ResultsFive patients were identified-four men and one woman with a median age of 31 years (range 22-50 years). Presentation was a mean of 25 days after injury (range 3-56 days). The left foot was affected in three cases and the right in two. There was joint diastasis in four patients and fracture-dislocation in one. Three patients presented early and were treated by internal fixation and two presented late and were managed conservatively. Mean follow-up was eight months (range 4.5-12 months). Surgery resulted in a return to work by 6 months with no symptoms. The two patients managed conservatively continued to experience pain at 12 months and were unable to return to their original occupations.ConclusionInjury to the Lisfranc joint should be excluded in any foot injury. Early diagnosis and internal fixation appears to result in an earlier return to work when compared to nonoperative management.

Nitrate potentially has a major impact on the quality of groundwater for potable supply. Conventional treatment methods to control nitrate problems in abstracted groundwater are expensive, in terms of unit costs and environmental impact. Utilizing the natural denitrifying capacity of aquifers offers an alternative approach to nitrate management based on in-situ bioremediation, which may have a number of strategic advantages including financial benefits and environmental sustainability. This paper reviews a range of literature from different scientific disciplines, regarding the physical and biological factors controlling denitrification in fissured aquifers, to assess whether in-situ bioremediation of nitrate in Chalk groundwaters is technically feasible. The principal conclusion is that, despite the dual-porosity operating in this type of aquifer, nitrate removal by engineered, in-situ bioremediation is potentially a viable and practicable method for treating groundwater. Process optimization could be achieved by investment in a programme of fundamental research in a number of key areas. The review highlights the need for a combined experimental and modelling approach, to satisfy the data and predictive requirements for configuring in-situ bioremediation systems at the field-scale. It also provides the necessary conceptual basis for developing a combined macroscale hydrogeological and biochemical model of denitrification activity in Chalk aquifers.

Alfalfa (Medicago sativa L.) cultivar development programs routinely incorporate resistance to fungal diseases that limit forage yield, quality and stand longevity. "Hay molding", caused by saprophytic fungi, reduces forage quality during the post-harvest period, but genetic resistance has not been reported. The objective was to determine the response to selection for post-harvest fungal resistance in alfalfa. Initially, 1144 genotypes from 22 alfalfa cultivars were screened for post-harvest fungal resistance with a combination of Aspergillus species. Forty resistant and 40 susceptible genotypes were selected by measuring percentage of leaf area with fungal coverage. Intraocular lymphoma is a rare tumour, the two main types being primary central nervous system non-Hodgkin's lymphoma (CNS-NHL) and, less commonly, systemic lymphoma that has spread to involve the eye. We present a case of a systemic NHL resulting in a serpiginous choroidopathy. To our knowledge, this presentation has not previously been reported. The diagnosis of this case was made based on the temporal relationship between the clinical activity of the choroidopathy and its response to chemotherapy. Diagnosis of intraocular lymphoma on vitreous biopsy is notoriously difficult, and this case is presented along with a review of the literature regarding vitreous biopsy and diagnostic techniques.

Plant breeders have long sought to improve grazing tolerance of alfalfa without sacrificing the beneficial yield and quality attributes of this species. Most efforts have focussed on selecting for traits (e.g., creeping rootedness) related to grazing tolerance and/or simulated grazing, but these efforts failed to account for the multiple stresses caused by grazing animals. Trait selection often led to sacrifices in yield and other desirable characteristics resulting in cultivars that were not robust across grazing management systems and environments. An innovative selection procedure was recently developed at the University of Georgia which incorporated intensive grazing with continuous stocking by beef cattle. The development of "Alfagraze" using this procedure showed that grazing tolerance and high yields can be incorporated into the same cultivar, along with consistent performance across grazing management systems and environments. Subsequent research has shown that grazing tolerance can be improved within elite, high-yielding, multiple-pest-resistant cultivars and breeding populations. Selection using intensive grazing with continuous stocking has been summarised in a "Standard Test Protocol" that is now being successfully used by public and private alfalfa breeders and in cultivar evaluation programs in the USA, Canada, and other countries. In this study, the role of the hepatobiliary system in the early pathogenesis ofSalmonella enteritidisinfection was investigated in a rat model. Intravenous (i.v.) challenge with lipopolysaccharide (LPS) has previously been shown to enhance the translocation of normal gut flora. We first confirmed that LPS can similarly promote the invasion ofS. enteritidis. Oral infection of outbred Australian Albino Wistar rats with 106to 107CFU ofS. enteritidisled to widespread tissue invasion after days. If animals were similarly challenged after intravenous administration ofS. enteritidisLPS (3 to 900 μg/kg of body weight), significant invasion of the livers and mesenteric lymph nodes (MLN) occurred within 24 h, with invasion of the liver increasing in a dose-dependent fashion (P< 0.01). If bile was prevented from reaching the intestine by bile duct ligation or cannulation, bacterial invasion of the liver and MLN was almost totally abrogated (P< 0.001). As i.v. challenge with LPS could induce the delivery of inflammatory mediators into the bile, biliary tumor necrosis factor alpha (TNF-α) concentrations were measured by bioassay. Biliary concentrations of TNF-α rose shortly after LPS challenge, peaked with a mean concentration of 27.0 ng/ml at around 1 h postchallenge, and returned to baseline levels (3.1 ng/ml) after 2.5 h. Although TNF-α cannot be directly implicated in the invasion process, we conclude that the invasiveness of the enteric pathogenS. enteritidisis enhanced by the presence of LPS in the blood and that this enhanced invasion is at least in part a consequence of the delivery of inflammatory mediators to the gastrointestinal tract by the hepatobiliary system.

Temperature dependent Hall effect, optical admittance spectroscopy, and optical absorption measurements of semi-insulating bulk 4H–SiC are reported. Both intentionally vanadium doped material and commercial grade semi-insulating material were investigated. The carrier concentration versus inverse temperature results from Hall effect measurements up to 1000 K indicated the samples were dominated by one of two deep levels near midgap. In addition to the deep donor level of substitutional vanadium, Ec−1.6 eV, we observed another level at Ec−1.1 eV in some samples, indicating that levels other than the vanadium donor can pin the Fermi level in semi-insulating SiC. Optical admittance measurements on the semi-insulating material indicate the presence of levels at Ec−1.73 and 1.18 eV that were previously observed in conducting samples with this technique and we attribute these levels to the same defects producing the 1.1 and 1.6 eV levels seen by Hall effect. Secondary ion mass spectroscopy measurements of dopant and impurity concentrations are reported. Even though vanadium is present in all of these samples, along with other impurities we are at present unable to definitively identify the 1.1 eV level.

The parity violation parameters Ab and Ac of the Zb anti-b and Zc anti-c couplings have been measured directly, using the polar angle dependence of the Z0-pole polarized cross sections. Bottom and charmed hadrons were tagged via semileptonic decays. Both the muon and electron identification algorithms take advantage of new multivariate techniques, incorporating for the first time information from the SLD Cerenkov Ring Imaging Detector. Based on the 1993-95 SLD sample of 150,000 Z0 decays produced with highly polarized electron beams, we measure: Ab = 0.910 ± 0.068(stat) ± 0.037(syst) Ac = 0.642 ± 0.110(stat) ± 0.063(syst).

We present a direct measurement of the parity-violating asymmetry A{sub b} in the Z{sup 0} to b anti-b coupling using a new technique to distinguish the b and anti-b quarks using charged kaons from B decays. The Z{sup 0} bosons are produced in e{sup +}e{sup -} collisions at the SLC with longitudinally polarized electrons. b anti-b events are selected using a secondary vertex mass tag and A{sub b} is determined from the left-right forward-backward asymmetry. From the 1994-1995 data sample, selected from 100,000 hadronic Z{sup 0} decays, we obtain A{sub b} = 0.855 {+-} 0. 088{sub stat} {+-} 0.102{sub sys}.

Shallow impurities are the principal means of affecting the electrical properties of semiconductors in order to induce desired characteristics. They can be used to isolate a region by introducing carriers of opposite charge, or they can be used to enhance the number of carriers of a particular type. Thermal admittance spectroscopy has been used to determine the activation energies of the principal p-type dopants, Al and B, in 4H and 6H-SiC, and temperature dependent Hall effect measurements were used to study the shallow B acceptors in 6H-SiC. The accept or species B and Al occupy inequivalent lattice sites in the Si sublattice, and would be expected to exhibit distinct energy levels for each site in analogy to the well known donor energy levels of N. Activation energies for B in 6H-SiC were found to be Eh=Ev+0.27 eV, Ek1=Ev+0.31 eV, and Ek2=Ev+0.38 eV. Al acceptors in 4H-SiC were found to exhibit two energy levels at Eh=Ev+0.212 eV and Ek=Ev+0.266 eV.

We present a direct measurement of the parity-violation parameter ${A}_{b}$ using a self-calibrating track-charge technique. In the SLAC Linear Collider Large Detector (SLD) experiment we observe hadronic decays of ${Z}^{0}$ bosons produced in collisions between longitudinally polarized electrons and unpolarized positrons at the SLAC Linear Collider. A sample of $b\overline{b}$ events is selected using the topologically reconstructed mass of $B$ hadrons. From our 1993--1995 sample of approximately 150 000 hadronic ${Z}^{0}$ decays, we obtain ${A}_{b}\phantom{\rule{0ex}{0ex}}=\phantom{\rule{0ex}{0ex}}0.911\ifmmode\pm\else\textpm\fi{}0.045(\mathrm{stat})\ifmmode\pm\else\textpm\fi{}0.045(\mathrm{syst})$.

Forage deterioration due to field and storage fungi represents a major economic loss for hay producers. A series of experiments was conducted to develop a methodology for screening alfalfa plants for resistance to post-harvest fungal colonization. Pure cultures of Aspergillus glaucus, Aspergillus repens, Aspergillus flavus, Aspergillus versicolor and Aspergillus fumigatus, isolated from alfalfa forage sampled during field wilting and storage, were established and maintained to produce a 105 spore mL−1 suspension containing an equal proportion of spores from each Aspergillus spp. Alfalfa leaves were dipped in concentrations of propionic acid solution to simulate levels of resistance to fungal activity. Fully-developed trifoliate leaves were placed adaxial side down onto petri plates containing a dichloran (2 mg mL−1) and glycerol (18% vol/vol) agar. The Aspergillus spp. spore suspension was sprayed onto each petri plate and the plates were incubated under dark conditions at 25 °C and 70% relative humidity. Plates were monitored daily for percent of leaf area colonized. The random screening of a small population of cloned alfalfa genotypes using this screening procedure indicated that variation in post-harvest resistance to fungal attack does exist for greenhouse and field-grown plants harvested at an early vegetative stage. We report a new measurement of Rb = Gamma(Z0->b-bbar) / Gamma(Z0->hadrons) using a double tag technique, where the b hemisphere selection is based on the reconstructed mass of the B hadron decay vertex. The measurement was performed using a sample of 130k hadronic Z0 events, collected with the SLD at the SLC. The method utilizes the 3-D vertexing abilities of the CCD pixel vertex detector and the small stable SLC beams to obtain a high b-tagging efficiency and purity.
