1. New Safety and Technical Challenges and Operational Experience on the JET First Trace Tritium Experiment

Abstract: Trace Tritium Experiments' (TTE) were successfully performed on JET in 2003. The Campaign marked the first use of tritium in JET plasmas since the Deuterium-Tritium Experiment (DTE1) Campaign in 1997, and was the first use of tritium in experiments under the EFDA organisation with the UKAEA as JET Operator. The safety and regulatory preparations for the experiment were extensive. Since JET has been operated by the UKAEA the operations have followed the model of a licensed nuclear site. The safe operation of the JET torus is demonstrated in a safety case. Key Safety Management Requirement (KSMR) and Key Safety Related Equipment (KSRE) are identified in the Safety Case for DT operation. The safe operation of the torus is within the bounds of, and under the control of, an Authority to Operate (ATO). New technical challenges were presented by the need to inject and account for small quantities of tritium in very short pulses (∼80ms), with an accurate time stamp. The safety and operational management of the campaign are described. Valuable lessons were learned which would help in running future experiments. It is concluded that JET is in a strong position to run future trace tritium and full DT discharges.

Abstract: Two sets of wide-field neurons extend neurites into the fly's optic lamina, where monopolar cells receive photoreceptor input. They exhibit immunoreactivity to antibodies raised against either 5-hydroxytryptamine or the crustacean peptide PDH, respectively. Both are proposed whole-field neuromodulators of vision, apparently regulating a circadian rhythm of monopolar cell size. Seeking functional correlates, we have re-examined the electroretinogram for circadian rhythmicity, and for responses to locally injected 5-hydroxytryptamine and peptide. Long-term electroretinogram recordings from Calliphora entrained to a light/dark cycle and then transferred to constant darkness, uncovered a gradual, modest increase during the subjective night in the electroretinogram's ON- and OFF-transients, from the lamina's monopolar cells. Five to twenty nl of 5-hydroxytryptamine (10(-3) mol.1(-1)) injected into the head haemolymph strongly enhanced the electroretinogram transients, an action reversed by 5-hydroxytryptamine antagonists. Injected into the eye, 5-hydroxytryptamine (10(-4) mol.1(-1)) had the opposite effect; the rapid onset there suggests direct action, whilst the opposing effect from haemolymph injection suggests a different receptor site. Pigment-dispersing hormone (2.2 x 10(-5) mol.1(-1)) injected into the haemolymph increased the electroretinogram transients along a biphasic course, with a slow partial recovery; injected into the eye, it lacked effect.

4. Detection of Airborne Sound by a Cockroach ‘Vibration Detector’: A Possible Missing Link in Insect Auditory Evolution

Abstract: Extracellular recordings from nerve 5 in metathoracic legs of Periplaneta americana disclose a sense organ that is extremely responsive to vibration but also detects sound (best response near 1.8 kHz) with a sensitivity similar to some insect auditory organs. The energy required from an auditory signal for a criterion response is similar or even smaller than for an optimal vibratory input. Responses originate in the subgenual organs (SGO) in the proximal tibiae, and cross-modal adaptation indicates that the same cells respond to both vibration and sound. Sound is picked up directly on some internal structure, not via sound-induced substratum vibration. Adaptation at different frequencies discloses no frequency-selectivity in the SGO. The nerve response is a burst of synchronized impulses at a frequency, fR, of approximately 300 Hz, that is practically invariant both with sound intensity and within the burst, suggesting that fR might represent some underlying resonance phenomenon, either of the SGO or of air in the tracheal system. The latter possibility is ruled out by observations made while the tracheae are perfused with He–O2. Similar responses can be recorded from the pro-and mesothoracic legs. Although Periplaneta is thought to be deaf and appears to ignore loud tones presented to the home colony, a more sensitive assay detects small leg movements in response to sound, confirming the presence of a functional auditory sense. The SGO is suspended from an expansion of the leg trachea, which may function to enhance sensitivity to vibration. This linkage preadapts the SGO to detect airborne sound transmitted in the tracheal system, and contact vibration may also stimulate the system in part by deforming the tracheae. It is proposed that auditory organs of crickets evolved from an ancestral SGO that already possessed dual responsiveness by subsequently developing effective vibration-isolating filters.

Abstract: Following photo-ablation of receptor cells in the retina of the housefly's compound eye, their synaptic terminals degenerate with a timecourse which we have followed over 8 d. Degeneration deprives the monopolar interneurons in the first optic neuropile, the lamina, of their main synaptic input. Simultaneously it deprives one monopolar interneuron (L2) of one of its synaptic targets, as L2 makes numerous feedback synaptic contacts at which it is pre-synaptic upon receptor terminals. Because the feedback synapses are dyadic, input still remains available to the second element post-synaptic at the dyad, which does not degenerate. This element is T1, a higher-order interneuron from the next most proximal neuropile (the medulla). Some of the original feedback synaptic sites soon disappear as a consequence of the photo-ablation, but their loss is partly offset by the production of new synaptic contacts. The new pre-synaptic ribbons resemble those at the original sites except for being smaller. The sites are, moreover, monadic, with T1 now the sole post-synaptic partner. These results show that interneurons in the fly's lamina retain a dynamic capacity for synaptogenesis throughout much of adult life, normally a few weeks in Musca, and that during this synaptogenesis they re-enact the same cell preferences expressed earlier in development.

Abstract: In the first optic neuropile of the housefly Musca, photoreceptor terminals innervate fixed clusters of interneurons, one of which is the monopolar cell L2; L2's synapses in turn feed back upon the terminals. We examined the ultrastructure of these feedback synapses following degeneration of their normal targets, the receptor terminals; this was accomplished by photo-ablating the receptor cells after intraretinal injections of sulforhodamine. Even when all the terminals degenerated, their deafferentated target cells, including L2, remained structurally intact for at least 14 d. Despite this lack of obvious trans-synaptic degeneration, L2's synaptic connections did alter. Presynaptic organelles of the feedback synapses, synaptic ribbons and associated synaptic vesicles, soon appeared in L2's cytoplasm, separating from their site of attachment at the presynaptic membrane by invagination. Similar free-floating organelles and vesicles also occurred in another monopolar cell, L4. They were also occasionally encountered in L2, in normal, newly emerged flies at a time when a naturally occurring loss of feedback synapses is greatest. We interpret the process of internalization that forms these floating ribbons to be the first step in synaptic loss which occurs spontaneously, and that the rate is enhanced in L2 when its main synaptic targets, the receptor terminals, degenerate.

Abstract: All known brachyceran flies have inherited the same trapezoidal pattern of rhabdomeres in the ommatidia of the compound eye, different from that of their presumed nematoceran ancestors. The projection pattern of axons to lamina cartridges needs to match this in order to generate efficient neural superposition vision. This pattern has been traced in serial sections or via stimulated dye uptake in two families that diverged from the dipteran stem line more than 200 million years ago (Rhagionidae, Stratiomyidae) and in one family of intermediate age (Dolichopodidae). All show the same asymmetric projection as modern muscoid families and a concordantly 180 degrees-twisted axon bundle, suggesting that evolution to the 'modern' versions of both optics and projection coincided closely in evolutionary time. The basic developmental plan of the cartridge has also been retained since the origin of the Brachycera, but the synaptic participation of at least one axon (that of photoreceptor R7) varies markedly between species.

9. 22q13 deletion syndrome

Abstract: We have recently collected clinical information on 37 individuals with deletion of 22q13 and compared the features of these individuals with 24 previously reported cases. The features most frequently associated with this deletion are global developmental delay, generalized hypotonia, absent or severely delayed speech, and normal to advanced growth. Minor anomalies include dolicocephaly, abnormal ears, ptosis, dysplastic toenails, and relatively large hands. As with many terminal deletions involving pale G-band regions, the deletion can be extremely subtle and can go undetected on routine cytogenetic analysis. In fact, 32% of the individuals in our study had previous chromosome analyses that failed to detect the deletion. Eight of 37 individuals had deletion of 22q13 secondary to an unbalanced chromosome translocation. In the newborn, this deletion should be considered in cases of hypotonia for which other common causes have been excluded. In the older child, this syndrome should be suspected in individuals with normal growth, profound developmental delay, absent or delayed speech, and minor dysmorphic features. We recommend high-resolution chromosome analysis and fluorescence in situ hybridization studies, or molecular analysis to exclude this diagnosis.

10. Primitive, crustacean-like state of blood-brain barrier in the eye of the apterygote insect Petrobius (Archaeognatha) determined from uptake of fluorescent tracers

Abstract: Compound eyes of insects in 16 orders were tested for the presence of a blood-retina barrier (BRB) by injecting the hemolymph with Procion yellow, which was excluded from the eye in all Neoptera but not in two apterygotes. A primitive apterygote (Petrobius, Machilidae) was investigated further. Epifluorescence observations with small dyes Lucifer yellow (LY) and sulforhodamine 101 (SR) confirmed uptake by the eye within 3 min of injection. LY and SR both penetrated the eye, particularly the cornea, examined in sections. Uptake was quantified by microfluorometry, yielding entry half-times (t(1/2)) of 1-1.4 min, fitting predictions for a model where tracer uptake is limited by passive diffusion. A much larger fluorescent dextran entered at a similar rate (t(1/2) = 1.70 +/- 0.77 min; n = 22), too fast to be diffusion-limited, pointing to an active process, probably flushing of hemolymph through the retina. This is not an artifact associated with tracer injection and may be the natural result of circulatory pressures. Microfluorometry gave a first estimate of hemolymph volume (2.9% of body weight), of hemolymph mixing time (t(0.95) = 77 min); the eyes' receptive fields were also determined. All results point to a primitive crustacean-like condition in Petrobius, with open access of hemolymph to the eye and no BRB. An evolutionary hypothesis is suggested to explain how a primitive central nervous system barrier later extended to cut off the eye in Neoptera, in the face of access problems for respiratory gases and metabolites.

11. The link between substance abuse and posttraumatic stress disorder in women. A research review

Abstract: Research has documented a high incidence of comorbid post-traumatic stress disorder (PTSD) and substance abuse. Women substance abusers, in particular, show high rates of this dual diagnosis (30% to 59%), most commonly deriving from a history of repetitive childhood physical and/or sexual assault. Rates for men are two to three times lower and typically stem from combat or crime trauma. Patients with both disorders are characterized by high severity on a multitude of psychological and treatment variables and use of the most severe drugs (cocaine and opioids). Treatment research on women is limited but suggests the possibility of retaining patients and achieving positive outcomes.

12. Changes in geometrical and biomechanical properties of immature male and female rat tibia

Abstract: Tibial geometry and mechanical properties in male and female rapidly growing rats were measured to provide comparative data for spaceflight, exercise, or disease experiments that use immature rats as an animal model. A total of 68 immature Sprague-Dawley rats (43-58 d of age) were divided into 8 groups according to age and sex. Tibial length and middiaphysial cross-sectional data were analyzed, and tibial mechanical properties were obtained via three-point bending tests to failure. Results indicated that tibial geometry and mechanical properties were changed significantly during this 15-d period of rapid growth, with male rats having greater increases than female rats. Both male and female tibial lengths nevertheless increased significantly during this growth period, as did middiaphysical cross-sectional area. Tibial flexural rigidity increased significantly for both sexes during this growth period. The male tibiae changed primarily in structural characteristics, but some material properties also changed with age. In the female rats, however, the major changes in mechanical properties of the tibia were attributable only to structural changes in the bone.

Abstract: A comparative ultrastructural study of photoreceptor synapses formed upon homologous postsynaptic neurones in insects has been made by using serial-section electron microscopy in representative Diptera from a monophyletic series of 14 families. At all of the synaptic contacts there is a presynaptic dense bar, surmounted in phylogenetically more recent families by a presynaptic platform. Opposite the bar lies a pair of postsynaptic elements that invariably originate one each from two unique monopolar neurones L1 and L2. Both elements contain increasingly elaborate cisternae in more recent flies. Within the phylogenetic series, the postsynaptic ensemble itself changes from the original dyad to a tetradic configuration in more recent Muscomorpha by the addition of two new postsynaptic elements from an amacrine cell. This transition occurs once only in the series, which, gauged by the fossil record, covers divergences from the stem line extending back greater than 200 million years.

Abstract: The plasma concentrations, tissue distribution and excretion of prostaglandin A1 (PGA1) and related metabolites have been determined in rats, following the intravenous injection of a single dose of PGA1-5,6-3H. Urinary and faecal excretion accounted for averages of 25 and 43%, of the administered dose of PGA1, respectively. Oxidative cleavage of the carboxyl side chain of PGA1 appeared to be a major metabolic pathway in the rat. PGA1-5,6-3H was deemed unsuitable for metabolism studies in man, in view of the significant loss of tritium label from the prostaglandin.

15. Survey of lameness in British dairy cattle

Abstract: During 1977 a survey was undertaken by practising veterinary surgeons of the incidence of different lesions causing lameness in dairy cattle. Forty-eight veterinary practices participated and they treated 7526 cases of lameness in 1821 herds. The average incidence of lameness among all cows was 5.5 per cent and the annual incidence among practices ranged from 1.8 to 11.8 per cent. Most lesions (88.3 per cent) occurred in the feet; the commonest were foul of the foot (16.7 per cent), white line abscess (15.6 per cent), sole ulcer (13.6 per cent), punctured sole and pus (10.4 per cent) and underrun heel (8.7 per cent). These proportions varied in different parts of the country. Eighty-four per cent of foot lesions occurred in the hind feet and of these lesions 85 per cent occurred in the outer claw; 42 per cent of foot lesions occurred in abnormally shaped claws. The remaining 11.7 per cent of lesions occurred in the legs and trunk, 76 per cent of which occurred in the hindlimb. Trauma was the main cause of leg lesions, which occurred most frequently in the joints and ligaments.

Abstract: SUMMARYStrains of Babesia divergens of human and bovine origin were isolated from the same locality in Scotland. A morphometric comparison was then made between Babesia divergens parasites in blood smears made from groups of 3 splenectomized calves reacting to the inoculation of these strains. The various forms taken by the parasites of each strain were seen to be divisible into 7 categories and representatives of each category were found for each strain. More amoeboid forms were found in blood smears from calves infected with the strain of bovine origin but otherwise the frequencies of occurrence of different forms coincided. The dimensions of 3 forms of the parasites of each strain were compared and theparasites of human origin were found to be significantly larger. The reactions of the groups of calves to inoculation of the 2 strains of B. divergens were seen to be very similar, although inoculation of the bovine strain had a slightly greater effect on the animals' haematology as indicated by an increase in erythrocyte mean corpuscular volume.

Circulating concentrations of glucose, propionate, lactate and pyruvate, and net exchange of these compounds across the liver and gut, were measured in lactating and non-lactating dairy cows (a) in the normal fed state, (b) before, during and after intravenous infusion of an aqueous solution of glucose, propionate or lactate (lactating cows only) in fed animals, and (c) before and during 6 days of food deprivation. 2. In the normal fed state, gut output of propionate, hepatic output of glucose and hepatic uptake of lactate were all higher in the lactating group. There was a net uptake of pyruvate across the liver in the lactating cows and a net output in the non-lactating cows. In the lactating cows there was a net uptake of lactate and pyruvate by the splanchnic bed (i.e. gut and liver combined). 3. In the lactating cows, the glucose and propionate infusions had the following effects: decrease in net hepatic uptake of lactate; a switch in pyruvate exchange across the liver from uptake to output; suppression of uptake of lactate and pyruvate by the splanchnic bed; increase in the magnitude of the liver (propionate uptake)/(glucose output) ratio. Lactate infusion did not affect hepatic propionate uptake. 4. Food deprivation increased hepatic extraction of lactate and pyruvate and decreased the liver (propionate uptake)/(glucose output) ratio in both groups. 5. It is concluded that mechanisms exist to ensure an inverse relationship between the availability to the cow of glucose or propionate and utilization by the splanchnic bed of endogenously derived lactate and pyruvate.

Abstract: Thermodilatometric, dynamic thermomechanical, and light-microscopic analyses were done on the anterior and posterior regions of the rat knee menisci to correlate regional differences in morphology and extracellular matrix composition with regional mechanical behavior. Following the administration of a general anesthetic, menisci were excised from 12 young female Sprague-Dawley rats. During thermodilatometric and thermomechanical testing, tissue temperature was increased at a constant rate of 3.5 degrees C/min from 30 to 100 degrees C. Light microscopy revealed regional differences in cell density and proteoglycan content. The anterior horn was significantly heavier (greater than 87%) and thicker (greater than 60%) than the posterior region. During thermal analyses, both the anterior and posterior horns decreased in tissue thickness as the temperature increased from 30 to 73 degrees C. After 73 degrees C, however, the posterior horn expanded significantly, whereas the anterior remained in a comparatively contracted state. The rates of linear contraction and expansion of the posterior horn were seven times those of the anterior horn, and the stiffness of the anterior horn was significantly greater than the posterior horn.

Abstract: The urinary metabolites of arbaprostil-3H in the male rat were profiled, isolated, and purified. Their structures were deduced by gas chromatography/mass spectrometry (GC/MS) studies after conversion to the methyl ester-methoxime-trimethylsilyl ether derivatives, aided by GC with simultaneous radioactivity monitoring. The identified metabolites accounted for over 91% of the urinary excretion products. beta-oxidation of the carboxy side-chain of arbaprostil to 15-methyl-tetranor PGE1 appeared to be the most significant metabolic pathway. Conversion to the dinor A and B derivatives and further beta-oxidation of these to the 15-methyl-tetranor A and B metabolites also appeared to occur. C-19-hydroxylated tetranor A and B derivatives of arbaprostil-3H were excreted in the urine. No conjugated urinary metabolites were evident.

Abstract: 1. Electrical measurements indicate that there is a local barrier to extracellular current flow in the outer part of the locust compound eye, and that neurons must interact amongst each other because of this structure (Shaw 1975). An attempt to confirm this structure independently is described here, visually with dye tracers, and more quantitatively with radioisotopes. 2. Dyes injected into the blood (haemolymph) rapidly appear in a parallel array of capillaries which transfix the neural layers anterio-posteriorly just below the basement membrane of the retina, but dyes cannot be detected within the retina or the optic lobe neuropile even after many hours. 3. Procion dyes infiltrated through a hole in the cornea spread throughout the retina, but appear to travel no further centrally than the basement membrane, although pigmented glial cells extending down to the capillaries take up the stain. In this region below the basement membrane, dye movement both into and out of the eye seems to be retarded, in agreement with the electrical measurements. 4. 3H-inulin and3H-polyethylene glycol (PEG) both diffuse only very slowly from the blood into the eye, and finally indicate an exchangeable space of about 3% of the eye volume, believed to be extracellular. The electrical measurements require a similar space. 5. Theoretical uptake curves for the unrestricted diffusion of inulin and PEG have half times of about 9 and 4 min, respectively. The half times of the curves best fitting the actual measurements are 4.5 and 1.6 h, indicating that diffusion is much retarded. If as believed, this retardation occurs only in the basal retina, the diffusion coefficients of the two tracers in this region have to be reduced by about a hundred to fit the uptake curves. 6. The smaller tracers sorbitol, mannitol, sucrose and sulphate all indicate an unrealistically large space in the eye and (except sulphate) leg muscle, a space which does not equilibrate with the blood (except with sulphate). This suggests extensive penetration of the cells in these tissues by the tracers or their metabolites. Autoradiography of3H-sucrose in the retina shows this too. 7. The uptake of sucrose into the eye is similar to that predicted for diffusion followed by removal of tracer by cellular absorption. The uptake rate measured for sorbitol, and that estimated for saccharides to explain the observed metabolic rate, are much too large to be explained by diffusion then uptake. This suggests that a rapid bypass route exists for some compounds across the barrier, to handle metabolic needs. The basal pigmented glial cells are properly positioned, and take up both sucrose and Procion Yellow. 8. Taken overall, the results confirm the electrical measurements in suggesting that the retina is sealed off from the blood by a basal barrier, which greatly retards diffusional access to the eye. The isolation of the retina from the lamina synaptic region should be several times more severe.

Certain pairs are interpreted as different cells in the same ommatidium because they have markedly different planes of maximum sensitivity to polarized light, but identical angular sensitivities. 3. This rules out artefactual reflection effects as the basis of polarized light discrimination, and points to the rhabdom of each cell as an individual polarization analyser. 4. The irregular angular sensitivities sometimes recorded are interpreted as coming from one cell, not as a simultaneous recording of the responses of several cells. 5. When one cell of a pair is polarized by current applied through the electrode, coupling between cells, as evidenced by the small potential seen in the other cell of the pair, is low. 6. The observation of non-coincidence of “bumps” at low light levels, and the independence of the polarized light responses at high intensities provide “natural” tests which show coupling is low in physiological conditions.

Abstract: The individual water intakes of 16 (four groups of four) lactating dairy cows, measured on 7 consecutive days, were significantly correlated with dry-matter (DM) intakes (range 4·6 to 14·4 kg/day) and milk yields (range 13·7 to 30·0 kg/day). There was no significant correlation with DM content of the feed (range 833 to 898 g/kg), body weight (range 400 to 620 kg) or mean air temperature (range 7·0 to 20·0°C). Applying a multiple regression analysis to the data, the following relationship was obtained: water intake (kg/day) = 2·15 (± 0·415) × DM intake (kg/day)+0·73 (± 0·197) × milk yield (kg/day) + 12·3 (+ 5·57). After adjusting the data for variations in DM intake and milk yield there was no significant difference between the mean daily water intakes of individual cows. Abstract: Electron microscopy-style fixation followed by epoxy plastic embedding is often now the method of choice for preparing tissue even for light microscopy; I have found it excellent for fluorescence, autoradiographic and conventional histology (Shaw 1972, 1977). Sections more than about five microns thick can be cut on a really sharp steel knife if the plastic is reasonably soft (Stretton and Kravitz 1973, Shaw 1972), but this is much easier and knife marks are reduced if extra-wide glass knives are used on a special-purpose intermediate microtome like the Sorvall JB-4. Recent budgetary restrictions made us defer purchase of such a microtome, and some alternative had to be devised. I report here a simple but rugged adapter for glass knives which replaces the steel knife in a conventional Leitz rotary microtome and allows thin plastic sections to be cut as easily as with a more sophisticated cutter. Abstract: The morphological and mechanical responses of tibia and humerus were assessed in growing male rats after a 1-wk spaceflight aboard NASA Spacelab 3. In contrast to flights of longer duration, changes in middiaphysial cross-sectional morphology were minimal. Inhibition of longitudinal growth was not found in the tibia but was apparent in the humerus. The normal age-related increase in tibial middiaphysial density was not observed in the flight animals. Three-point bending tests indicated that a 1-wk spaceflight impeded the maturation of bone strength and stiffness, with the effects more pronounced in the tibia than in the humerus. Material property alterations in bone thus overshadowed morphological factors in determining the bone's mechanical response. It is likely that deprivation of normal weight-bearing loads was a major factor contributing to the observed changes, but endocrine and other local factors must also be considered.

Abstract: Three groups of four lactating cows received a subcutaneous injection of 0 . 05, 0 . 10 and 0 . 15 mg Se/kg body weighty respectively administered as sodium selenate. A fourth group was injected with saline. In all the cows injected with sodium selenate, the concentration of Se in blood increased rapidly and was significantly higher than in control cows for two days in the group receiving the lowest dose and for 182 days (the duration of the experiment) in the two other groups. The activity of glutathione peroxidase in blood increased slowly in all cows injected with sodium selenate and was significantly greater than in control cows after 15, 22 and 29 days respectively, and remained significantly greater for 63, 91 and 182 days respectively. In a second experiment a single subcutaneous injection of 0 . 15 mg Se/kg body weight had no effect on the mean milk yield of 37 animals (19 . 1 kg/day) compared with the milk yield of a similar group of control animals (19 . 1 kg/day) during 70 days. The concentration of Se in milk was significantly higher on the first (168 microgram/litre) and second (69 microgram/litre) day after injection than in control animals (mean 26 microgram/litre).

The region discussed is the retina proper and first synaptic neuropile, the lamina, and thus encompasses visual processing up to the level of third-order visual neurones. A number of reviews give either wider or fuller accounts of the same area: those of Laughlin (1980) and Järvilehto (1984) are particularly wide-ranging, Shaw (1981) discusses the neural connections in detail, whilst Meinertzhagen & Frohlich (1983) and Meinertzhagen (1984) provide introductions to aspects of neural development. A recent NATO conference volume features this area (Ali, 1984).

Abstract: The cellular mechanisms by which nervous systems evolve to match evolutionary changes occurring in the rest of the body remain largely unexplored. In a distal visual neuropil of a previously unexamined ancient dipteran family, Stratiomyidae, homologues of all of the periodic neurons known already from more recent Diptera can be recognized, occupying the same locations within the unit structure. This points to extreme developmental stasis for more than 200 million years, conserving both cell identity and position. The arborizations that some neurons make also have remained conservative, but others show marked differences between families in both size and branching patterns. At the electron-microscopical level, extensive differences in synaptic connectivity are found, some sufficient to radically redefine the systems roles of particular neurons. The findings bear out an earlier prediction that changes in the connectivity matrix linking conserved neurons may have been a major factor in implementing evolutionary change in the nervous system.

Abstract: A single dose of arbaprostil-11 beta-3H (4 micrograms/kg) was administered orally to male rats. A maximum plasma radioactivity concentration equivalent to 2.5 to 2.8 nanograms of the prostaglandin per ml was reached at 30 minutes and was maintained until 120 minutes after drug administration. The plasma drug disappearance half-life was 2.6 hours. These results along with data from tissue distribution studies suggested a rapid uptake of radiolabeled arbaprostil by the glandular stomach tissue followed by an apparent zero-order release of drug-related radioactivity from this tissue "reservoir" into the plasma. Drug-related radioactivity was excreted rapidly, with 96 to 99% of the urinary excretion and 82 to 97% of the fecal excretion being completed within 24 hours. A total of 49.6 +/- 3.5% of the orally administered dose was excreted in the urine and 46.7 +/- 3.9% in the feces. No radioactive residues were detected in the animals at the end of the 120 hour specimen collection period. The metabolic stability of the 11 beta-tritium label and the suitability of arbaprostil-3H for use in human studies was demonstrated.

Abstract: Musca and related flies have three main photoreceptor subsystems. The R1-6 group has short axons that terminate in the cartridges of the first optic neuropile, the lamina. The cartridges are bypassed by the longer axons of R7 and R8, which run together to terminate at different levels in the underlying medulla neuropile. The present account describes a shallow, previously unidentified zone in the lamina within which R7/8 make glancing contact with R1-6. At the distal border of the cartridge over no more than 3-4 microns depth, the tangentially directed short axon of R6 squeezes between the pair from R7 and R8, forming quite large areas of mutual contact (approximately 7 microns2). Less frequently, R1 is contacted. At least some of these sites contain smaller membrane specialisations indistinguishable from the more numerous gap junctions found more proximally that interconnect the terminals of R1-6. The R7/8 junctions with R6 are of comparable size (0.15 micron 2) and likewise possess symmetrical membrane densities. They provide proposed pathways for direct electrical interaction to account for observed electrical input from R7/8 to the R1-6 subsystem. In two cases R7/8 was possibly postsynaptic to R1-6 at a multiple-contact synapse, but even if functional, these sites were so rare that they are unlikely to have much operational significance.

Abstract: Since the last IAEA conference, the scientific programme of JET has focused on the qualification of the integrated operating scenarios for ITER and on physics issues essential for the consolidation of design choices and the efficient exploitation of ITER. Particular attention has been given to the characterization of the edge plasma, pedestal energy and edge localized modes (ELMs), and their impact on plasma facing components (PFCs). Various ELM mitigation techniques have been assessed for all ITER operating scenarios using active methods such as resonant magnetic field perturbation, rapid variation of the radial field and pellet pacing. Abstract: A project dedicated to the enhancement of the JET vertical stabilization system was launched in 2006, including an upgrade of the Power Supply of the Radial Field Amplifier, of hardware and software of the vertical stabilization control system. The main aim was to double the JET capability in stabilising high current plasmas when subject to perturbations, in particular large Edge Localised Modes. We present here the results of first plasma operation with the new Enhanced Radial Field Amplifier and its data acquisition and control system, focussing on the benefits of an approach based on phased commissioning, modelling and offline algorithm validation.
