1. An intensive exercise-based training program reduces prefrontal activity during usual walking in patients with Parkinson’s disease

Abstract: Introduction: Parkinson’s disease (PD) leads to a progressive loss of locomotor automaticity. Consequently, PD patients rely more on executive resources for the control of gait, resulting in increased prefrontal activity while walking. Exercise-based training programs may improve automaticity of walking and reduce prefrontal activity in this population. This study aimed to assess the effect of an intensive multidisciplinary exercise-based training program on prefrontal activity and gait performance during usual walking in PD patients. Method: Fourteen patients (mean age: 67 ± 9; disease duration: 6 ± 5 years; Hoehn and Yahr score: 1.9 ± 0.6) were included in this study. They were assessed in ON stage at three different times at 5-week intervals: two times before the training program (T0 and T1) and once after the training program (T2). Gait performance (stride time, speed, stride length, cadence, and their respective coefficient of variation) and cortical activity in the dorsolateral prefrontal cortex (DLPFC) using functional near infrared spectroscopy (fNIRS) were measured during usual walking. Results: Patients had reduced cortical activity of the DLPFC at T2 compared to T1 (p = 0.003). Patients had shorter stride time at T2 compared to T1 (p = 0.025) and tended to have longer stride length at T2 than at T1 (p = 0.056). Conclusion: The training program led to positive effects on prefrontal activity and gait performance. Reduced prefrontal activity during usual walking after training program suggests that patients may have a greater reserve capacity to face more challenging walking conditions. Further studies will investigate the effect of this training on cortical activity during dual-task walking..

2. Analysis of copy number variation in men with non‐obstructive azoospermia

Abstract: Recent findings demonstrate that single nucleotide variants can cause non-obstructive azoospermia (NOA). In contrast, copy number variants (CNVs) were only analysed in few studies in infertile men. Some have reported a higher prevalence of CNVs in infertile versus fertile men.This study aimed to elucidate if CNVs are associated with NOA.We performed array-based comparative genomic hybridisation (aCGH) in 37 men with meiotic arrest, 194 men with Sertoli cell-only phenotype, and 21 control men. We filtered our data for deletions affecting genes and prioritised the affected genes according to the literature search. Prevalence of CNVs was compared between all groups. Exome data of 2,030 men were screened to detect further genetic variants in prioritised genes. Modelling was performed for the protein encoded by the novel candidate gene TEKT5 and we stained for TEKT5 in human testicular tissue.We determined the cause of infertility in two individuals with homozygous deletions of SYCE1 and in one individual with a heterozygous deletion of SYCE1 combined with a likely pathogenic missense variant on the second allele. We detected heterozygous deletions affecting MLH3, EIF2B2, SLX4, CLPP and TEKT5, in one subject each. CNVs were not detected more frequently in infertile men compared with controls.While SYCE1 and MLH3 encode known meiosis-specific proteins, much less is known about the proteins encoded by the other identified candidate genes, warranting further analyses. We were able to identify the cause of infertility in one out of the 231 infertile men by aCGH and in two men by using exome sequencing data.As aCGH and exome sequencing are both expensive methods, combining both in a clinical routine is not an effective strategy. Instead, using CNV calling from exome data has recently become more precise, potentially making aCGH dispensable.

Abstract: Executive functions are important for successful accomplishment of walking tasks, particularly during a dual task. Over the past few years, several studies investigated prefrontal cortex activity under different walking conditions in older adults with functional near infrared spectroscopy (fNIRS). However, little is known about changes in dorsolateral prefrontal cortex (DLPFC) activity during walking in the early stages of aging. The main objective of this study was to compare changes in DLPFC activity during simple and dual task walking across three different age groups. Twenty-five young (age range = 18-37), twenty-five youngest-old (age range = 55-65), and twenty-five older adults (age range = 67-87) participated in this study. Main results showed that, during simple task walking, older adults had increased DLPFC activity with equivalent walking performance. This increased mainly concerned the right hemisphere. During dual task walking, older adults had increased right DLPFC activity but seemed to have enough resources to maintain their performance during DT walking. This result supports the idea that compensation mechanisms, due to loss of automaticity of walking in aging, appear already during simple task walking. Measuring cortical activity with fNIRS during a simple task walking might be used as valuable indicator for identifying individuals at risk of falling.

Abstract: Prolonged cycling exercise performance in normoxia is limited because of both peripheral and central neuromuscular impairments. It has been reported that cerebral perturbations are greater during short-duration exercise in hypoxia compared with normoxia. The purpose of this study was to test the hypothesis that central deficits are accentuated in hypoxia compared with normoxia during prolonged (three bouts of 80 min separated by 25 min) whole-body exercise at the same relative intensity.Ten subjects performed two sessions consisting of three 80-min cycling bouts at 45% of their relative maximal aerobic power in normoxia and hypoxia (FiO2 = 0.12). Before exercise and after each bout, maximal voluntary force, voluntary activation assessed with nerve stimulation and transcranial magnetic stimulation, corticospinal excitability (motor evoked potential), intracortical inhibition (cortical silent period), and electrical (M-wave) and contractile (twitch and doublet peak forces) properties of the knee extensors were measured. Prefrontal and motor cortical oxygenation was also recorded during each cycling bout in both conditions.A significant but similar force reduction (≈-22%) was observed at the end of exercise in normoxia and hypoxia. The modifications of voluntary activation assessed with transcranial magnetic stimulation and nerve stimulation, motor evoked potential, cortical silent period, and M-wave were also similar in both conditions. However, cerebral oxygenation was reduced in hypoxia compared with normoxia.These findings show that when performed at the same relative low intensity, prolonged exercise does not induce greater supraspinal fatigue in hypoxia compared with normoxia. Despite lower absolute exercise intensities in hypoxia, reduced brain O2 availability might contribute to similar amounts of central fatigue compared with normoxia.

Abstract: International audience; The aim of the present study was to compare the influence of three rest intervals of 30, 60 and 180 seconds randomly applied between two sets of five isokinetic contractions of flexor-extensor knee muscles in a cohort composed of 14 men (32.5 ± 8 years). The comparison of the isokinetic values measured during the first set of each evaluation session showed no significant differences (0.55 < P < 0.80). The analysis of the time and rest interval interaction demonstrated no significant difference between the isokinetic measures obtained after the rest intervals for peak torque of the quadriceps and of the hamstrings and for the mean power of the quadriceps and the hamstrings. These findings raise questions about a strict energetic approach and a future research should analyze the interaction of force production and fatigue phenomena in relation to the rest interval.

Abstract: The central nervous system can play a critical role in limiting exercise performance during hypoxic conditions. Hypocapnia, which is associated with hypoxia-induced hyperventilation, may affect cerebral perfusion. We hypothesized that CO2 clamping during hypoxic isometric knee extensions would improve cerebral oxygenation and reduce central fatigue.Fifteen healthy men (mean ± SD: age, 25 ± 8 yr; body mass, 72 ± 11 kg; height, 179 ± 7 cm) performed intermittent isometric knee extensions at ∼50% of maximal voluntary contraction to task failure in normoxia, hypoxia with CO2 clamping (arterial O2 saturation, 80% ± 2%; end-tidal CO2 partial pressure, 40 ± 2 mm Hg), and hypoxia without CO2 clamping (arterial O2 saturation, 80% ± 3%). Transcranial magnetic stimulation and femoral nerve electrical stimulation were used to assess central and peripheral determinants of fatigue. Prefrontal cortex and quadriceps femoris oxygenation were monitored by multichannel near-infrared spectroscopy.Exercise duration was reduced to a similar extent in hypoxia with CO2 clamping (997 ± 460 s) or hypoxia without CO2 clamping (929 ± 412 s) compared to normoxia (1473 ± 876 s; P0.001). Prefrontal cortex and quadriceps oxygenation were increased (+5.3 ± 8.6 and +2.6 ± 3.0 μmol·cm at task failure, respectively; P0.01) during hypoxia with CO2 clamping compared to hypoxia without CO2 clamping. Transcranial magnetic stimulation maximal voluntary activation decreased to a greater extent at task failure in hypoxia without CO2 clamping (-18% ± 8%) compared to hypoxia with CO2 clamping (-9% ± 9%; P0.01) and normoxia (-10% ± 7%; P0.05). Conversely, exercise-induced peripheral fatigue was larger in hypoxia with CO2 clamping than in hypoxia without CO2 clamping (e.g., Db10-to-Db100 ratio of 0.54 ± 0.12 and 0.63 ± 0.11 at task failure, respectively; P0.05).The results demonstrate that CO2 clamping can alter central and peripheral mechanisms that contribute to neuromuscular fatigue during hypoxic isometric knee extensions in men. Hypocapnia impairs cerebral oxygenation and central drive but exerts a protective effect against fatigability in muscles.

Abstract: While there are clear effects of exercise on brain activity measured subsequent to the bout of physical activity, it is also important to elucidate the nature of the changes that occur in the brain during exercise, because these changes may be linked to sensorimotor performance. Characterization of the cortical contribution to motor control in humans is important for a better understanding of the pathophysiology of sensorimotor deficits after an injury to the central nervous system. This chapter addresses (1) how brain activation patterns change in response to various levels of exercise intensity, (2) in what extend the severity of neuromuscular fatigue modulates brain activation patterns in healthy persons, (3) and how the brain networks adapt to cerebrovascular injury (Stroke) and rehabilitation program to relearn movement.

Abstract: The Region Languedoc-Roussillon is the umbrella organisation for an interconnected and integrated project on AHA covering the 3 pillars of the European Innovation Partnership on Active and Healthy Ageing. All sub-activities (A1: electronic pharmaceutical file, A2: falls prevention initiative, A3: frailty, B3: chronic respiratory diseases, chronic diseases with comorbidities, oral health and hepatitis virus C chronic infection, C2 and D4 active and independent living and handicap) are included in MACVIA-LR that has a strong political commitment and includes all stakeholders (public, private, patients, policy makers). It is one of the Reference Sites of the European Innovation Partnership on Active and Healthy Ageing built around chronic diseases, ageing and handicap. The framework of MACVIA-LR has the vision that the prevention and management of CDs is essential for AHA promotion and for the reduction of handicap. The main objective of MACVIA-LR is to develop innovative solutions for a network of Living Labs in order to improve the care of patients affected by CDs in the Languedoc-Roussillon area and to disseminate the innovation.

Abstract: International audience; Falls represent a major cause of burden and death in older adults. The MACVIA-LR (fighting chronic diseases for active and healthy ageing in Languedoc-Roussillon) falls initiative is a cross-cutting Living Lab based on the European Innovation Partnership on Active and Healthy Ageing (EIP on AHA) at the regional level. It covers all action plans (A1, A2, A3, B3, C2 and D4) of the EIP on AHA. The Living Lab will: (i) raise awareness, (ii) drive attitude change with respect to falls and falls prevention and (iii) improve access to information on falls prevention. It aims to reduce fall-related hospitalisations by 30% in 2020 in the région Languedoc-Roussillon (LR). It includes: (i) a falls prevention clinic for the assessment of subjects (with a falls risk) in the two teaching hospitals of Montpellier and Nîmes, (ii) the Regional Ageing Institute (CARSAT-LR), (iii) the Low Vision Institute of Nîmes, (iv) the Lattes Social Welfare Centre, (v) the home automation Institut méditerranéen des métiers de longévité, (vi) teaching, training and coaching as well as (vii) industrial partnership. Two care pathways have been developed (stroke and falls) and are being applied to remote rural areas of the région. The MACVIA-falls prevention initiative is an output of the EIP on AHA.

Abstract: Cardiorespiratory fitness has been shown to protect and enhance cognitive and brain functions, but little is known about the cortical mechanisms that underlie these changes in older adults. In this study, functional near infrared spectroscopy (fNIRS) was used to investigate variations in oxyhemoglobin [HbO2] and in deoxyhemoglobin [HHb] in the dorsolateral prefrontal cortex (DLPFC) during the performance of an executive control task in older women with different levels of cardiorespiratory fitness (VO2max). Thirty-four women aged 60-77 years were classified as high-fit and low-fit based on VO2max measures. They all performed a control counting (CNT) task and the Random Number Generation (RNG) task at two different paces (1 number/1 s and 1 number/1.5 s), allowing to manipulate task difficulty, while hemodynamic responses in the bilateral DLPFCs were recorded using continuous-wave NIRS. The behavioral data revealed that the high-fit women showed significantly better performance on the RNG tasks compared with the low-fit women. The high-fit women showed significant increases in [HbO2] responses in both left and right DLPFCs during the RNG task, while the low-fit women showed significantly less activation in the right DLPFC compared with the right DLPFC of the high-fit women and compared with their own left DLPFC. At the level of the whole sample, increases in the [HbO2] responses in the right DLPFC were found to mediate in part the relationship between VO2max level and executive performance during the RNG task at 1.5 s but not at 1 s. These results provide support for the cardiorespiratory fitness hypothesis and suggest that higher levels of aerobic fitness in older women are related to increased cerebral oxygen supply to the DLPFC, sustaining better cognitive performance.

19. Déterminants des différentes phases de la cinétique de la consommation d’oxygène chez l’homme

Abstract: Resume Objectifs – L’objectif de cette revue est de faire le point sur les connaissances concernant les determinants des differentes phases de la cinetique de la consommation d’oxygene V O 2 lors d’exercice a charge constante. Actualites – A ce jour, la signification de la phase rapide et de la phase de composante lente de la cinetique de V O 2 n’est pas encore clairement etablie. Des travaux experimentaux recents indiquent que l’ajustement initial du metabolisme aerobie represente l’action mutuelle des facteurs qui determinent l’approvisionnement en O 2 aux mitochondries, l’activite enzymatique et l’inertie metabolique des muscles actifs, plutot qu’un systeme de controle exclusif. Le developpement de la phase de composante lente trouve son origine principalement a la peripherie ou a lieu le travail musculaire avec une influence du type de fibres impliquees dans l’exercice. Perspectives et projets – De nombreux modeles (animal, petites masses musculaires) ont ete utilises pour savoir si l’apport en O 2 ou l’inertie metabolique determine la phase rapide. Il reste a preciser les facteurs qui regulent les differentes etapes constitutives du transfert de l’O 2 de l’air pulmonaire jusqu’a l’utilisation de l’O 2 pour des exercices globaux chez l’homme. Enfin la mesure conjointe du V O 2 des muscles actifs et des facteurs actuels de la composante lente reste a determiner afin de preciser l’etiologie de ce phenomene.

20. Relationships Between Aerobic Energy Cost, Performance and Kinematic Parameters in Roller Ski Skating - Aerobic Energy Cost of Roller Ski Skating

Abstract: The aims of this study were to test the hypotheses that 1) the aerobic energy cost of roller ski skating (C s ) is significantly related to level of performance and 2) a significant part of inter-individual differences of C s can be explained by kinematic parameters of skating locomotion. Oxygen uptake, kinematics of the knee and ankle joint, EMG of the vastus lateralis (VL) and gastrocnemius lateralis (GL) muscles, and roller ski velocity were recorded in 13 skiers who roller skied at 19.0 ± 0.1 km x h -1 . C s was found to be 2.51 ± 0.35] × kg -1 × m -1 and significantly correlated with the skiers' level of performance (r = 0.61; p < 0.05). Significant relationships were found between C s and 1) knee angular amplitude (r = 0.75; p < 0.01) during the concentric phase, 2) VL average EMG during the concentric phase (r = 0.72; p < 0.01) and 3) VL and GL average EMG during the eccentric phase (p < 0.05). The results of this study showed that a significant part of performance could be explained by the aerobic energy cost of locomotion in skating. It can also be suggested that differences in upper/lower body utilization and/or in mechanical efficiency may explain the differences in C s .

21. Decrease in Oxygen Uptake at the End of a High-Intensity Submaximal Running in Humans

Abstract: The purpose of the present study was to examine oxygen consumption (VO(2) ) kinetics during severe-intensity running exercise through a four-phase model that considered a decrease in VO(2) at the end of the exercise in light of previous research in which this decrease was only noticed. After determination of maximal oxygen consumption VO(2) max), thirteen highly trained males performed a square-wave running to exhaustion at approximately 95 % of VO(2) max on a level treadmill. VO(2) and ventilatory gas exchange variables were determined breath-by-breath. Computerised non-linear regression techniques incorporating exponential and linear terms were used to describe VO(2) and ventilatory gas exchange variable responses. In contrast with the classical 3-component model that describes the increase in VO(2) for severe-intensity exercise, we observed a 4(th) phase characterised by a significant decrease in VO(2) before exhaustion (slope of VO(2)-time relationship significantly different from a zero value, p < 0.01) in 7 out of 13 subjects. Following a time delay of 31 +/- 44 s after the decrease in VO(2), a significant decrease of minute ventilation (V(E)) was present for 6 of the 7 subjects (p < 0.02). During the exercise for the subjects who decreased V(E), a reduction of 288 +/- 169 ml in tidal volume was associated with an increase of 10.2 +/- 2.4 min(-1) in breathing frequency. These data suggest that the respiratory system might be stressed more for some endurance-trained athletes. The specific link between reduced VO(2) and reduced V(E) remains to be explored.

22. Impaired oxygen uptake kinetics in the first high-level athlete with Hb Hope: a case study

Abstract: Hemoglobin (Hb) Hope is a beta-globin chain variant with reduced oxygen (O₂) affinity, known to induce anemia. This usually leads to limitations in O₂uptake (VO₂) and exercise tolerance. We studied the case of a high-level female athlete with Hb Hope. She had been selected for cross-country races from 13 yrs onward, then was a national junior champion in 400-m race, and finally failed to win any cross-country races as an adult. Hematological analysis revealed normal red blood cell indices and Hb level (12.3 g.dL⁻¹). Incremental exercise showed peak work rate (WR), VO(2max) and gas exchange threshold (GET) within normal ranges for healthy females. Constant WR testing at 90% of GET showed that kinetics of pulmonary VO₂included the presence of a slow component. This was in disagreement with the data on VO₂kinetics response to exercise intensities below GET. Phase 2 parameters, time constant (τ2, 31 s), time delay (TD2, 39 s), amplitude (A2, 780 ml.min⁻¹), and gain in VO₂(ΔVO₂ .ΔWR-1, 9.2 ml.min-1.W⁻¹) were within normal ranges. Phase 3 showed a slow component similar to that reported in severe exercise. The absence of anemia and the normality of phase 2 suggested normal O₂delivery and oxidative metabolism in exercising muscles. In contrast, phase 3 suggested poor aerobic capacity and limited exercise tolerance. Abstract: Although the existence of a central pattern generator system modulated by sensory information has become broadly accepted in the control of gait, many findings indicate that the cortex also plays a role of primary importance in human walking (Miyai et al., 2001; Gwin et al., 2011; Petersen et al., 2012). Examining the neural control of gait in humans requires recording cortical activity during gait. Direct evidence for cortical involvement in human locomotion comes from neuroimaging studies using position emission tomography (la Fougere et al., 2010), electroencephalography (EEG, Gwin et al., 2011) and functional near-infrared spectroscopy (fNIRS, Miyai et al., 2001) methods. Among possible neuroimaging methods, fNIRS is ideally suited to perform brain imaging during walking as it represents several advantages over other methods (Perrey, 2008).

Abstract: The aim of the study was to test the ability to estimate the power output (PO) and heart rate (HR) associated with ''anaerobic threshold'' levels for triathletes by means of a 30-min perceptive individual time trial (PITT30).Thirteen triathletes (8 males and 5 females) performed an incremental exercise test to estimate maximal parameters such as oxygen uptake, power output and heart rate. From this incremental exercise test, the individual anaerobic threshold (IAT) and ventilatory threshold (VT) for all subjects were estimated. Then, the subjects completed a PITT30 at self-selected work intensity on a stationary ergometer equipped with the SRM Training System. Mean values of PO, HR, and pedalling cadence were recorded continuously between the 5th and the 30th min of the test.Significant correlations were observed between the mean PO recorded during PITT30 and PO measured at IAT (r=0.88; p0.0001) and at VT (r=0.89; p0.0001). Furthermore, bias and limits of agreement confirm the degree of association between the 3However, PITT30 over-estimated HR values compared to the values obtained at IAT and VT.It was concluded that, for triathletes, mean PO measured with PITT30 allows a partial valid estimation of PO associated with 2 known methods of ''anaerobic threshold'' determination. The application of PITT30 may offer a useful tool for athletes and coaches to estimate the ''anaerobic threshold'' in order to control accurately the training effects.

Abstract: During angiogenesis, microvascular endothelial cells (ECs) secrete proteinases that permit penetration of the vascular basement membrane as well as the interstitial extracellular matrix. This study tested the hypothesis that cathepsin S (Cat S) contributes to angiogenesis. Treatment of cultured ECs with inflammatory cytokines or angiogenic factors stimulated the expression of Cat S, whereas inhibition of Cat S activity reduced microtubule formation by impairing cell invasion. ECs from Cat S–deficient mice showed reduced collagenolytic activity and impaired invasion of collagens type I and IV. Cat S–deficient mice displayed defective microvessel development during wound repair. This abnormal angiogenesis occurred despite normal vascular endothelial growth factor and basic fibroblast growth factor levels, implying an essential role for extracellular matrix degradation by Cat S during microvessel formation. These results demonstrate a novel function of endothelium-derived Cat S in angiogenesis.

28. Relationships between aerobic energy cost, performance and kinematic parameters in roller ski skating

Abstract: The aims of this study were to test the hypotheses that 1) the aerobic energy cost of roller ski skating (CS) is significantly related to level of performance and 2) a significant part of inter-individual differences of CS can be explained by kinematic parameters of skating locomotion. Oxygen uptake, kinematics of the knee and ankle joint, EMG of the vastus lateralis (VL) and gastrocnemius lateralis (GL) muscles, and roller ski velocity were recorded in 13 skiers who roller skied at 19.0 +/- 0.1 km x h(-1). CS was found to be 2.51 +/- 0.35 J x kg(-1) x m(-1) and significantly correlated with the skiers' level of performance (r = 0.61; p0.05). Significant relationships were found between CS and 1) knee angular amplitude (r = 0.75; p0.01) during the concentric phase, 2) VL average EMG during the concentric phase (r = 0.72; p0.01) and 3) VL and GL average EMG during the eccentric phase (p0.05). The results of this study showed that a significant part of performance could be explained by the aerobic energy cost of locomotion in skating. It can also be suggested that differences in upper/lower body utilization and/or in mechanical efficiency may explain the differences in CS.

29. Field testing of VO2peak in cross-country skiers with portable breath-by-breath system

Abstract: Peak aerobic exercise responses were determined in 8 competitive cross-country skiers during field-testing and during an incremental treadmill test (TM) to determine peak oxygen uptake (VáO2peak), heart rate, and oxygen pulse. Field testing consisted of maximal effort roller skiing using diagonal stride (DS) and double poling (DP) techniques. VáO2 was measured during roller skiing by the portable Cosmed K4b2 breath-by-breath system. The portable system was compared against a standard laboratory mixing box system, and no differences were found across the full range of VáO2. During DS, VáO2peak and oxygen pulse were not different from TM, whereas peak HR was lower. With DP, VáO2peak and oxygen pulse were less than DS or TM, whereas peak HR was less than with TM but not different from DS. These results demonstrate the validity and utility of the portable breath-by-breath system for measurement of aerobic energy demand of athletes as they carry out their normal activity.

Abstract: The purposes of this study were 1) to examine the occurrence of the stretch-shortening cycle (SSC) in the three principal techniques of ski skating and 2) to test the hypothesis that differences in SSC behaviour may explain the choice of a technique when the grade is altered. Ten highly skilled male cross-country skiers roller skied at 5.3+/-0.1 m x s(-1) using the V1 skate (V1), V2-alternate (V2A), and V2 skate (V2) techniques. Knee and ankle joint kinematics, EMG of the vastus lateralis (VL) and gastrocnemius lateralis (GL) muscles, and roller ski velocity were recorded during the last 40 s of each bout of roller skiing. The existence of an EMG signal during the eccentric phase prior to the thrust, as well as the lack of significant delay between the two phases, showed that SSC occurs in the three techniques of roller ski skating with the exception of 50% of the subjects at the weak side knee with V1. The VL stretching velocity over the eccentric phase was higher (p < 0.005) with V2 and V2A than V1, indicating that less elastic energy was stored in the lower limbs extensor muscles with V1. This could partly explain why this technique is not used on flat terrains during races. However, no differences of average EMG activity or IEMG existed between techniques for either GL or VL during the concentric phase, suggesting that the theoretical advantage due to the SSC behaviour is minor in roller ski skating.

Abstract: Two receptor pathways are thought to mediate the hepatic clearance of chylomicron remnants, (i) the low density lipoprotein receptor (LDLR) pathway and (ii) non-LDLR pathway. The current study was undertaken to quantitatively assess the contribution of each receptor pathway to hepatic catabolism of chylomicron remnants, by using mice that are deficient in apolipoprotein E (apoE) (apoE(-/-)), the LDLR (LDLR(-/-)), and both (apoE(-/-);LDLR(-/-)). Vitamin A fat tolerance tests showed that the area under the curves of the plasma excursions of retinyl ester in the LDLR(-/-), apoE(-/-), and apoE(-/-);LDLR(-/-) mice were 4, 12, and 12 times larger than those in wild-type mice. The retinyl ester accumulated in the plasma of the LDLR(-/-) mice was distributed in larger subfractions of triglyceride-rich lipoproteins, chylomicrons through very low density lipoprotein-C. These results indicate that the LDLR constitutes the major pathway for the clearance of retinyl ester. In support of this, agarose gel electrophoresis revealed that an oral fat load resulted in retention of chylomicrons in the LDLR(-/-) mice, which was not seen in wild-type mice. The observation that the apoE(-/-) mice showed larger retinyl ester excursion than LDLR(-/-) mice indicates that an apoE-dependent non-LDLR pathway is involved in the rest of the clearance of the retinyl ester. Together, we conclude that the LDLR pathway plays a significant role in the chylomicron remnant metabolism in mice fed a normal chow.

32. Faster oxygen uptake kinetics at the onset of submaximal cycling exercise following 4 weeks recombinant human erythropoietin (r-HuEPO) treatment.

Abstract: We tested the hypothesis that prolonged administration of moderate doses of recombinant human erythropoietin (r-HuEPO) accelerates the initial rate of rise in pulmonary O2 uptake (VO2) in response to submaximal exercise and increases the maximal rate of O2 uptake (VO2,max). Sixteen endurance-trained athletes were divided into two groups: r-HuEPO- (n=9) or placebo-treated (n=7). r-HuEPO or placebo (saline) injections were given s.c. 3 times a week for 4 weeks. Exercise testing, before and after the 4 weeks, comprised incremental maximal tests and several transitions from rest to 10-min cycling exercise at 65% VO2,max. VO2 was measured breath-by-breath during all tests. In the r-HuEPO group, resting haemoglobin concentration (+9.6%) and haematocrit (+8.3%), as well as VO2,max (+7.0%) and power output (+7.2%) increased significantly (P<0.05) after the 4 weeks, whereas no change was observed in the control group. The time constant of the primary VO2 response was significantly faster (+18%) after the 4 weeks r-HuEPO treatment than before (mean±SD; 29.3±4.5 vs. 35.7±7.4 s, respectively, P<0.05) but was unaffected in the placebo group (34.5±7.3 vs. 33.4±7.9 s). Collectively, our findings suggest that r-HuEPO contributes both to an acceleration of the dynamic response of VO2 to submaximal exercise and to an increase in maximal exercise capacity. 33. Is the &Vdot;O(2) slow component dependent on progressive recruitment of fast-twitch fibers in trained runners?

Abstract: The goal of this study was to use spectral analysis of EMG data to test the hypothesis that the O(2) uptake (&Vdot;O(2)) slow component is due to a recruitment of fast fibers. Thirteen runners carried out a treadmill test with a constant speed, corresponding to 95% of the velocity associated with maximal &Vdot;O(2). The &Vdot;O(2) response was fit with the classical model including three exponential functions. Electrical activity of six lower limb muscles (vastus lateralis, soleus, and gastrocnemius of both sides) was measured using electromyogram surface electrodes. Mean power frequency (MPF) was used to study the kinetics of the electromyogram discharge frequency. Three main results were observed: 1) a common pattern of the MPF kinetics in the six muscles studied was noted; 2) MPF decreased in the first part of the exercise, followed by an increase for all the muscles studied, but only the vastus lateralis, and gastrocnemius muscles of both sides increased significantly (P < 0.05); and 3) the beginning of the MPF increase of the four muscles mentioned above corresponded with the beginning of the slow component. Our results suggest a progression in the average frequency of the motor unit discharge toward the high frequencies, which coheres with the hypothesis of the progressive recruitment of fast-twitch fibers during the &Vdot;O(2) slow component. However, this interpretation must be taken with caution because MPF is the result of a balance between several phenomena.

It covers the 3 pillars of the European Innovation Partnership on Active and Healthy Ageing (EIP on AHA): (A) Prevention and health promotion, (B) Care and cure, (C) and (D) Active and independent living of elderly people. All sub-activities (poly-pharmacy, falls prevention initiative, prevention of frailty, chronic respiratory diseases, chronic diseases with multimorbidities, chronic infectious diseases, active and independent living and disability) have been included in MACVIA-LR which has a strong political commitment and involves all stakeholders (public, private, patients, policy makers) including CARSAT-LR and the Eurobiomed cluster. It is a Reference Site of the EIP on AHA. The framework of MACVIA-LR has the vision that the prevention and management of chronic diseases is essential for the promotion of AHA and for the reduction of handicap. The main objectives of MACVIA-LR are: (i) to develop innovative solutions for a network of Living labs in order to reduce avoidable hospitalisations and loss of autonomy while improving quality of life, (ii) to disseminate the innovation. The three years of MACVIA-LR activities are reported in this paper.

Abstract: It has been proposed that interbrain synchrony (IBS) may help to elucidate the neural mechanisms underpinning teamwork. As hyperscanning studies have provided abundant findings on IBS in team environments, the current review aims to synthesize the findings of hyperscanning studies in a way that is relevant to the teamwork research. A systematic review was conducted. Included studies were classified according to the IPO (i.e. input, process, output) model of teamwork. Three multi-level meta-analyses were performed to quantify the associations between IBS and the three IPO variables. The methodology followed PRISMA guidelines and the protocol was pre-registered (https://osf.io/7h8sa/). Of the 229 studies, 41 were included, representing 1326 teams. Abstract: Background: In the last 5 years since our last systematic review, a significant number of articles have been published on the technical aspects of muscle near-infrared spectroscopy (NIRS), the interpretation of the signals and the benefits of using the NIRS technique to measure the physiological status of muscles and to determine the workload of working muscles., Objectives: Considering the consistent number of studies on the application of muscle oximetry in sports science published over the last 5 years, the objectives of this updated systematic review were to highlight the applications of muscle oximetry in the assessment of skeletal muscle oxidative performance in sports activities and to emphasize how this technology has been applied to exercise and training over the last 5 years. In addition, some recent instrumental developments will be briefly summarized., Methods: Preferred Reporting Items for Systematic Reviews guidelines were followed in a systematic fashion to search, appraise and synthesize existing literature on this topic. Electronic databases such as Scopus, MEDLINE/PubMed and SPORTDiscus were searched from March 2017 up to March 2023. Potential inclusions were screened against eligibility criteria relating to recreationally trained to elite athletes, with or without training programmes, who must have assessed physiological variables monitored by commercial oximeters or NIRS instrumentation., Results: Of the identified records, 191 studies regrouping 3435 participants, met the eligibility criteria. This systematic review highlighted a number of key findings in 37 domains of sport activities. Overall, NIRS information can be used as a meaningful marker of skeletal muscle oxidative capacity and can become one of the primary monitoring tools in practice in conjunction with, or in comparison with, heart rate or mechanical power indices in diverse exercise contexts and across different types of training and interventions., Conclusions: Although the feasibility and success of the use of muscle oximetry in sports science is well documented, there is still a need for further instrumental development to overcome current instrumental limitations. 40. Eccentric cycling involves greater mental demand and cortical activation of the frontoparietal network.

Abstract: Eccentric, compared to concentric exercise, is proposed to involve different neuro-motor processing strategies and a higher level of mental demand. This study compared eccentric and concentric cycling at matched perceived effort and torque for the mental demand and related-cortical activation patterns. Nineteen men (30 ± 6 years) performed four different 5-min cycling conditions at 30 RPM on a semi-recumbent isokinetic cycle ergometer: (1) concentric at a moderate perceived effort (23 on the CR100® scale) without torque feedback; (2) concentric and (3) eccentric at the same average torque produced in the first condition; and (4) eccentric at the same moderate perceived effort than the first concentric condition. The conditions two to four were randomized. After each condition, mental demand was monitored using the NASA Task Load Index scale. Changes in oxy-(O 2 Hb) and deoxy-(HHb) hemoglobin during exercise were measured over both prefrontal cortices and the right parietal lobe from a 15-probe layout using a continuous-wave NIRS system. Mental demand was significantly higher during eccentric compared to concentric cycling (+52%, p = 0.012) and when the exercise intensity was fixed by the torque rather than the perceived effort (+70%, p < 0.001). For both torque- or perceived effort-matched exercises, O 2 Hb increased significantly (p < 0.001) in the left and right prefrontal cortices, and right parietal lobe, and HHb decreased in the left, and right, prefrontal cortices during eccentric compared to concentric cycling. Abstract: Introduction: Dementia is a neurological disorder associated with aging that can cause a loss of cognitive functions, impacting daily life. Alzheimer's disease (AD) is the most common cause of dementia, accounting for 50-70% of cases, while frontotemporal dementia (FTD) affects social skills and personality. Electroencephalography (EEG) provides an effective tool to study the effects of AD on the brain., Methods: In this study, we propose to use shallow neural networks applied to two sets of features: spectral-temporal and functional connectivity using four methods. We compare three supervised machine learning techniques to the CNN models to classify EEG signals of AD / FTD and control cases. Abstract: The emergence of consciousness is one of biology's biggest mysteries. During the past two decades, a major effort has been made to identify the neural correlates of consciousness, but in comparison, little is known about the physiological mechanisms underlying first-person subjective experience. Attention is considered the gateway of information to consciousness. Recent work suggests that the breathing phase (i.e., inhalation vs. exhalation) modulates attention, in such a way that attention directed toward exteroceptive information would increase during inhalation. One key hypothesis emerging from this work is that inhalation would improve perceptual awareness and near-threshold decision-making. The present study directly tested this hypothesis. We recorded the breathing rhythms of 30 humans performing a near-threshold decision-making task, in which they had to decide whether a liminal Gabor was tilted to the right or the left (objective decision task) and then to rate their perceptual awareness of the Gabor orientation (subjective decision task). In line with our hypothesis, the data revealed that, relative to exhalation, inhalation improves perceptual awareness and speeds up objective decision-making, without impairing accuracy. Overall, the present study builds on timely questions regarding the physiological mechanisms underlying consciousness and shows that breathing shapes the emergence of subjective experience and decision-making. NEW & NOTEWORTHY Breathing is a ubiquitous biological rhythm in animal life. However, little is known about its effect on consciousness and decision-making. Here, we measured the respiratory rhythm of humans performing a near-threshold discrimination experiment. We show that inhalation, compared with exhalation, improves perceptual awareness and accelerates decision-making while leaving accuracy unaffected.

45. Predictive modeling of the ratings of perceived exertion during training and competition in professional soccer players.

Abstract: Objectives: Evaluate the ability of predicting the ratings of perceived exertion from the external load variables in professional soccer players through a chronological perspective (i.e., past features values are considered additional features) through machine learning models by considering the playing position., Design: Prospective cohort study., Methods: Thirty-eight elite soccer players aged 19-27 years were observed during 151 training sessions, 44 matches across a full season. External load variables (58 derived from Global Positioning System and 30 from accelerometers) and the internal load derived from ratings of perceived exertion were collected for each player and each session and match. Machine learning models (linear regression, K-NN, decision trees, random forest, elastic net regression, XGBoost) were compared and interpreted in order to deepen the relationship between external load variables and ratings of perceived exertion according to the player position in a predictive perspective., Results: Application of the machine learning models on the dataset provided enough predictive power to reduce the Root Mean Squared Error of 60 % from dummy predictions. The most accurate models (Root Mean Squared Error ≈ 1.1 for random forest and = 1 for XGBoost) highlighted a memory effect in subsequent ratings of perceived exertion values. Abstract: Healthy aging leads to poorer performance in upper limb (UL) daily living movements. Understanding the neural correlates linked with UL functional movements may help to better understand how healthy aging affects motor control. Two non-invasive neuroimaging methods allow for monitoring the movement-related brain activity: functional near-infrared spectroscopy (fNIRS) and electroencephalography (EEG), respectively based on the hemodynamic response and electrical activity of brain regions. Coupled, they provide a better spatiotemporal mapping. The aim of this study was to evaluate the effect of healthy aging on the bilateral sensorimotor (SM1) activation patterns of functional proximal UL movements. Twenty-one young and 21 old healthy participants realized two unilateral proximal UL movements during: i) a paced reaching target task and ii) a circular steering task to capture the speed-accuracy trade-off. Combined fNIRS-EEG system was synchronised with movement capture system to record SM1 activation while moving. The circular steering task performance was significantly lower for the older group. The rate of increase in hemodynamic response was longer in the older group with no difference on the amplitude of fNIRS signal for the two tasks. The EEG results showed aging related reduction of the alpha-beta rhythms synchronisation but no desynchronisation modification. In conclusion, this study uncovers the age-related changes in brain electrical and hemodynamic response patterns in the bilateral sensorimotor network during two functional proximal UL movements using two complementary neuroimaging methods. 49. Probing the Promises of Noninvasive Transcranial Electrical Stimulation for Boosting Mental Performance in Sports.

Abstract: While the importance of physical abilities is noncontested to perform in elite sport, more focus has recently been turned toward cognitive processes involved in sport performance. Practicing any sport requires a high demand of cognitive functioning including, but not limited to, decision-making, processing speed, working memory, perceptual processing, motor functioning, and attention. Noninvasive transcranial electrical stimulation (tES) has recently attracted considerable scientific interest due to its ability to modulate brain functioning. Neuromodulation apparently improves cognitive functions engaged in sports performance. This opinion manuscript aimed to reveal that tES is likely an adjunct ergogenic resource for improving cognitive processes, counteracting mental fatigue, and managing anxiety in elite athletes. Nevertheless, the first evidence is insufficient to guarantee its real effectiveness and benefits. All tES techniques could be add-ons to make performance-related cognitive functions more efficient and obtain better results. Modulating inhibitory control through tES over the frontal cortex might largely contribute to the improvement of mental performance. Nevertheless, studies in elite athletes are required to assess the long-term effects of tES application as an ergogenic aid in conjunction with other training methods (e.g., neurofeedback, mental imagery) where cognitive abilities are trainable.

50. Stable decoding of working memory load through frequency bands.

Abstract: Numerous studies have shown that working memory modulates every frequency band's power in the human brain. Yet, the question of how the highly distributed working memory adapts to external demands remains unresolved. Here, we explored frequency band modulations underlying working memory load, taking executive control under account. We hypothesized that synchronizations underlying various cognitive functions may be sequenced in time to avoid interference and that transient modulation of decoding accuracy of task difficulty would vary with increasing difficulty. We recorded whole scalp EEG data from 12 healthy participants, while they performed a visuo-spatial n-back task with three conditions of increasing difficulty, after an initial learning phase. We analyzed evoked spectral perturbations and time-resolved decoding of individual synchronization. Surprisingly, our results provide evidence for persistent decoding above the level-of-chance (83.17% AUC) for combined frequency bands. In fact, the decoding accuracy was higher for the combined than for isolated frequency bands (AUC from 65.93% to 74.30%). However, in line with our hypothesis, frequency band clusters transiently emerged in parieto-occipital regions within two separate time windows for alpha-/beta-band (relative synchronization from approximately 200 to 600 ms) and for the delta-/theta-band (relative desynchronization from approximately 600 to 1000 ms). Overall, these findings highlight concurrent sustained and transient measurable features of working memory load. This could reflect the emergence of stability within and between functional networks of the complex working memory system. In turn, this process allows energy savings to cope with external demands.
