Arctodiaptomus dorsalis has been reported in the Philippines as an invasive species, which originated from Central America. Apart from its wide distribution in the United States and Central America, it is now widely distributed in many lakes in the Philippine islands, such as Luzon Island and southwestern Mindanao, making it an ideal specimen for studies on population dynamics and population genetics. Recently, new techniques have been emerging as important zooplankton monitoring tools, such as metabarcoding and the use of environmental DNA/environmental RNA. However, these would only be useful if good-quality reference DNA sequences are available before utilizing these techniques. This study sequenced the nearly complete nuclear ribosomal DNA (nrDNA) of A. dorsalis from 202 individuals from Lake Taal, Batangas. The total length was 5887 bp, which includes the partial 18S sequences, and the complete 5.8S, ITS and 28S sequences with an organization that is typical for any eukaryotic organism. This is the first record of a nearly complete nrDNA sequence for freshwater calanoid copepods and is considered an important stepping stone for further molecular studies in freshwater Copepoda.

We report on the development of a chemical mechanical planarization (CMP) process for thick damascene Ta structures with pattern feature sizes down to 100 nm. This CMP process is the core of the fabrication sequence for scalable superconducting integrated circuits at 300 mm wafer scale. This work has established the elements of the various CMP-related design rules that can be followed by a designer for the layout of circuits that include Ta-based coplanar waveguide resonators, capacitors, and interconnects for tantalum-based qubits and single flux quantum (SFQ) circuits. The fabrication of these structures utilizes 193 nm optical lithography, along with 300 mm process tools for dielectric deposition, reactive ion etch, wet-clean, CMP and in-line metrology, all tools typical for a 300 mm wafer CMOS foundry. Process development was guided by measurements of physical and electrical characteristics of the planarized structures. Physical characterization such as atomic force microscopy across the 300 mm wafer surface showed local topography was less than 5 nm. Electrical characterization confirmed low leakage at room temperature, and less than 12% within wafer sheet resistance variation, for damascene Ta line-widths ranging from 100 nm to 3 {\mu}m. Food web studies rely heavily on the use of traditional gut content analysis or the fairly popular stable isotope analysis rather than fatty acid analysis (FAA) despite its straightforward process, and ability to identify and characterize more diverse trophic pathways. In this study, we employed fatty acid (FA) biomarkers as a preliminary attempt to trace and characterize trophic pathways in food webs of four tropical lakes of volcanic origin in Luzon Island, Philippines - three clustered maar lakes (Yambo, Pandin and Calibato) and a large caldera lake (Taal). These lakes have a long history of human disturbance but limited ecological data. Knowledge of basal food sources and existing trophic dynamics of organisms are generally non-existent. Particulate organic matter (POM), zooplankton, and fish species were collected in August 2019 from the study lakes as representative of three trophic guilds. Non-metric multidimensional scaling (NMDS) and principal component analysis (PCA) were conducted to analyse FA profiles and characterize trophic relationships between representative organisms. For the POM, within lakes comparison of taxon-specific FA profiles showed a significant difference between the surface and near bottom depths, with the former dominated by photoautorophs and the latter by chemotrophs, suggesting the ability of FAA to effectively delineate between micro-organisms. Between lakes comparison also showed significant difference between the caldera and maar lakes, with the latter containing higher composition of bacterial FA, reminiscent of the considerably smaller lakes’ response to the impact of unmitigated organic loadings from anthropogenic activities. Taken together with the primary consumers’ FA profiles, analysis confirmed the ability of FAA to discriminate between FA profile sources. PCA explained >70% of the variance in the FA compositions for three trophic guilds in the two deepest lakes, which delineated both zooplankton and fish species food selectivity in each lake, alluding to FAA’s capacity to characterize dietary reliance of various species in an environment with numerous food sources. Although certain limitations were encountered, such as the specificity of the sampling depths for POM, and the small sample size of the representative species of the third trophic level, this study demonstrated the effectiveness of FAA as a powerful ecological tool for disentangling intricate lake food webs comprising various food sources. Overall, this study provided baseline information on basal food sources and trophic pathways of representative organisms from four tropical lakes. Taken together, FAA studies have wide application in understanding food webs, including anthropogenically-threatened lake ecosystems.

The CRYSTAL EYE detector is proposed as a space-based X and gamma ray all-sky monitor to be active from 10 keV up to 30 MeV. In its full scale configuration, it consists in a 40 cm diameter hemisphere, made by 112 pixels, with an overall weight lower than 50 kg, wide Field Of View (FOV, about 6 sr), full sky coverage and very large effective area (about 6 times higher than Fermi-GBM at 1 MeV) in the energy range of interest. Each pixel consists of two layers of scintillating LYSO crystals, read out by arrays of Silicon PhotoMultipliers (SiPMs), equipped with a segmented anticoincidence detector for charged Cosmic Ray (CR) identification and hard X-ray detection. The primary scientific goals include the observation of transient X and gamma flashes from Gamma Ray Bursts (GRBs), Gravitational Wave (GW) follow up, Supernovae (SN) explosions, etc. and stable gamma-ray source observation in the MeV energy range. The pioneering design optimizes these observations in terms of localization of the source and timing. By using specific triggers for charged particles, solar flares and space weather phenomena could also be studied. A pathfinder mission is foreseen onboard of the Space Rider vehicle run by European Space Agency (ESA), allowing technology tests, qualification and both deep space and Earth observation during the mission. We here present the CRYSTAL EYE technology.

Reservoirs are continuous waterscapes that exhibit hydrodynamic characteristics of both lakes and rivers with water flow. These dynamic habitats provide a novel chance to explore the effects of metacommunity dynamics on community structure in relation to environmental, spatial, and temporal processes. We investigated the spatial distribution of spring zooplankton communities in a large reservoir in southern China. We intensively sampled along a longitudinal gradient during early and full spring. We compared the relationship of environmental and spatial variables to the community structure of three zooplankton groups: rotifers, cladocerans, and copepods. The overall composition and abundance of zooplankton groups across longitudinal zones became more heterogeneous with increasing environmental heterogeneity across sampling periods. Multi-level ANOVA and PERMANOVA results showed that rather than homogenize the environment, water flow in late spring increased spatial heterogeneity in the environment and increased the importance of species sorting in shaping the zooplankton community. However, variation partitioning analysis showed that the influence of environmental and spatial variables also varied across the three zooplankton groups likely due to their differences in both physiological and morphological adaptation. Our study shows that metacommunity processes are important in structuring zooplankton communities in reservoir waterscapes that are influenced by flow.

A massive invasion of the Neotropical calanoid copepod Arctodiaptomus dorsalis (Marsh, 1907) has been taking place in the Philippines since at least 1993. This copepod’s fecundity is known to depend on food concentration, increasing temperature appears to have detrimental effects on its hatching success and survivorship, but parameters that are truly predictive of the successful integration and propagation of this species in particular bodies of water have not yet been identified. We conducted a spatial analysis of morphometric features and reproductive capacity, the latter proxied by clutch size (CS), of different populations of A. dorsalis inhabiting 11 tropical lakes in the Philippines. Correlations between either morphometry or CS and selected water quality parameters were investigated based on collections and measurements made from December 2018 to April 2019. Means of morphometric measurements differed significantly (P < 0.001) among the populations of A. dorsalis in all lakes. Total length (TL) of adult males, TL of non-ovigerous adult females, and CS of ovigerous females all showed significant (P < 0.001) inverse correlations with lake depth, accounting for about 30% of the observed variability among lakes (r = –0.55, –0.55, and –0.58, respectively). Shallow lakes, which are more homogenous in terms of nutrient distribution throughout the water column, thus provide a more suitable habitat than deep lakes for A. dorsalis and provides a partial explanation of the widespread occurrence and abundance of the species in the shallower, more eutrophic lakes in the Philippines.

During the COVID-19 pandemic, wastewater-based epidemiology (WBE) has been engaged to complement medical surveillance and in some cases to also act as an early diagnosis indicator of viral spreading in the community. Most efforts worldwide by the scientific community and commercial companies focus on the formulation of protocols for SARS-CoV-2 analysis in wastewater and approaches addressing the quantitative relationship between WBE and medical surveillance are lacking. In the present study, a mathematical model is developed which uses as input the number of daily positive medical tests together with the highly non-linear shedding rate curve of individuals to estimate the evolution of global virus shedding rate in wastewater along calendar days. A comprehensive parametric study by the model using as input actual medical surveillance and WBE data for the city of Thessaloniki (similar to 700,000 inhabitants, North Greece) during the outbreak of November 2020 reveals the conditions under which WBE can be used as an early warning tool for predicting pandemic outbreaks. It is shown that early warning capacity is different along the days of an outbreak and depends strongly on the number of days apart between the day of maximum shedding rate of infected individuals in their disease cycle and the day of their medical testing. The present data indicate for Thessaloniki an average early warning capacity of around 2 days. Moreover, the data imply that there exists a proportion between unreported cases (asymptomatic persons with mild symptoms that do not seek medical advice) and reported cases. The proportion increases with the number of reported cases. The early detection capacity of WBE improves substantially in the presence of an increasing number of unreported cases. For Thessaloniki at the peak of the pandemic in mid-November 2020, the number of unreported cases reached a maximum around 4 times the number of reported cases. (C) 2021 The Authors. Considered as some of the most devastating complications, Cutibacterium acnes (C. acnes)-related osteomyelitis are among the hardest infections to diagnose and treat. Mesenchymal stem cells (MSCs) secrete number of immunomodulatory and antimicrobial soluble factors, making them an attractive treatment for bacterial infection. In this study, we examined MSCs/C. acnes interaction and analyzed the subsequent MSCs and bacteria's behaviors. Human bone marrow-derived MSCs were infected by C. acnes clinical strain harvested from non-infected bone site. Following 3 h of interaction, around 4% of bacteria were found in the intracellular compartment. Infected MSCs increased the secretion of prostaglandin E2 and indolamine 2,3 dioxygenase immunomodulatory mediators. Viable intracellular bacteria analyzed by infrared spectroscopy and atomic force microscopy revealed deep modifications in the wall features. In comparison with unchallenged bacteria, the viable intracellular bacteria showed (i) an increase in biofilm formation on orthopaedical-based materials, (ii) an increase in the invasiveness of osteoblasts and (iii) persistence in macrophage, suggesting the acquisition of virulence factors. Overall, these results showed a direct impact of C. acnes on bone marrow-derived MSCs, suggesting that blocking the C. acnes/MSCs interactions may represent an important new approach to manage chronic osteomyelitis infections. Statement of significance The interaction of bone commensal C. acnes with bone marrow mesenchymal stem cells induces modifications in C. acnes wall characteristics. These bacteria increased (i) the biofilm formation on orthopaedical-based materials, (ii) the invasiveness of bone forming cells and (iii) the resistance to macrophage clearance through the modification of the wall nano-features and/or the increase in catalase production.

Periodic repetition of right heart catheterization (RHC) in pulmonary arterial hypertension (PAH) can be challenging. We evaluated the correlation between RHC and cardiopulmonary exercise test (CPET) aiming at CPET use as a potential noninvasive tool for hemodynamic burden evaluation. The center-periphery hypothesis (CPH) predicts that peripheral populations will have lower genetic variation than those at the center of a species’ distribution. However, ecological margins do not always coincide with geographical edges when topographies are diverse. Historical climate changes can also strongly affect genetic variation. Here, we examined genetic variation in Phyllodiaptomus tunguidus, a calanoid copepod endemic to the north-eastern Oriental. This species was predicted to exhibit a complex pattern of genetic variation across its range due to the diverse topographies and stable climate history of the north-eastern Oriental. To test this, we used geographic distance to the center of the distribution, current ecological suitability, and climate during the last glacial maximum as geographical, ecological, and historical factors, respectively, in our analyses. We measured genetic diversity and population differentiation using mitochondrial and nuclear markers. This showed that P. tunguidus had 3 refugia during the last glacial maximum (LGM). Such a pattern of multiple refugia complicates the determination of the center and periphery of spatial genetic diversity. Both regression models and redundancy analyses failed to support the CPH. Instead, they showed that geographical, ecological, and historical factors together shaped population genetic structure in this species. Ecological factors explained significantly more genetic variation than did geographical and historical factors—however, all three factors interacted significantly to affect the pattern of genetic variation. The results extend our understanding of the CPH and the extent to which it can explain genetic variation across populations.

Body size is a functional trait that influences the overall biology and ecology of an organism. Studying the shape of size–frequency distributions and size variability within different scales, approximates the influence of large-scale ecological and evolutionary processes on a species. In this study we examine the patterns of distribution and variability of body size among freshwater Cladocera across different taxonomic levels, geographic distribution and habitat association. Using extensive literature data, we show the global distribution of body size in freshwater Cladocera. Hierarchical models were used to assess the effect of different categorical variables on size variability. Our results show that almost all size–frequency distributions were skewed right in all categories. The hierarchical model showed that taxonomic affiliation contributes the most to size variability in our dataset, suggesting that size might be a conserved trait. Large genera (≥1mm) have larger estimated variability compared to smaller genera. In general, our observations on size–frequency distributions and size variability show a brief insight in the varying advantages of adaptive body size in this group of organisms in both biology (physiology) and ecology (competition and co-existence). Thus, body size is a trait important to the survival and continuing evolution of Cladocera.

The original description of Diaptomus vexillifer Brehm, 1933, endemic to Lake Danao (Leyte Island), was elementary and lacking taxonomically satisfactory characters. In this paper, we redescribe the Philippine-endemic genus Filipinodiaptomus Lai, Mamaril Sr. & Fernando, 1979 and the species D. vexillifer collected from its type locality, using light and scanning electron microscopy and an analysis of the mtCOI gene and the ITS of the nuclear 18S operon. We found that D. vexillifer is the second species under this endemic genus. A neotype and paraneotypes are hereby designated. Also a detailed morphological comparison is made between Filipinodiaptomus vexillifer (Brehm, 1933) comb. nov. and Filipinodiaptomus insulanus (Wright, 1928). Their taxonomic characters, interspecies relationship, and biogeography are likewise discussed.

Polyurethanes (PU) are a broad class of polymers that offer good solvent compatibility and a wide range of properties that can be used to generate microfluidic layers. Here, we report the first characterization of a commercially available Shore 80D polyurethane (Ultraclear&trade, 480N) for biomicrofluidic applications. Studies included comparing optical clarity with Polydimethylsiloxane (PDMS) and using high-fidelity replica molding to produce solid PU structures from the millimeter to nanometer scales. Additionally, we report the first use of NanoAccel&trade, treatment in Accelerated Neutral Atom Beam (ANAB) mode to permanently roughen the surface of PU and improve the adhesion of breast cancer cells (MDA-MB-231) on PU. Surface energy measurements using Owens-Wendt equations indicate an increase in polar and total surface energy due to ANAB treatment. Fourier-transform infrared (FTIR) spectroscopy in attenuated total reflectance (ATR) mode was used to demonstrate that the treatment does not introduce any new types of functional groups on the surface of Ultraclear&trade, PU. Finally, applicability in rapid prototyping for biomicrofluidics was demonstrated by utilizing a 3D-printing-based replica molding strategy to create PU microfluidic layers. These layers were sealed to polystyrene (PS) bases to produce PU-PS microfluidic chips. Ultraclear&trade, PU can serve as a clear and castable alternative to PDMS in biomicrofluidic studies.

Crosstalk between mesenchymal stem cells (MSCs) and bacteria plays an important role in regulating the regenerative capacities of MSCs, fighting infections, modulating immune responses and maintaining tissue homeostasis. Commensal Cutibacterium acnes (C. acnes) bacterium becomes an opportunistic pathogen causing implant-associated infections. Herein, we examined MSCs/C. acnes interaction and analysed the subsequent bacteria and MSCs behaviours following infection. Human bone marrow derived MSCs were infected by two clinical and one laboratory C. acnes strains. Following 3h of interaction, all bacterial strains were able to invade MSCs. Viable intracellular bacteria acquired virulence factors by increasing biofilm formation and/or by affecting macrophage phagocytosis. Although the direct and indirect (through neutrophil stimulation) antibacterial effects of the MSCs secretome were not enhanced following C. acnes infection, ELISA analysis revealed that C. acnes clinical strains are able to license MSCs to become immunosuppressive cell-like by increasing the secretion of IL-6, IL-8, PGE-2, VEGF, TGF-β and HGF. Overall, these results showed a direct impact of C. acnes on bone marrow derived MSCs, providing new insights into the development of C. acnes during implant-associated infections. STATEMENT OF SIGNIFICANCE: The originality of this work relies on the study of relationship between human bone marrow derived mesenchymal stem cells (MSCs) phenotype and C. acnes clinical strains virulence following cell infection. Our major results showed that C. acnes are able to invade MSCs, inducing a transition of commensal to an opportunistic pathogen behaviour. Although the direct and indirect antibacterial effects were not enhanced following C. acnes infection, secretome analysis revealed that C. acnes clinical strains were able to license MSCs to become immunosuppressive and anti-fibrotic cell-like. These results showed a direct impact of C. acnes on bone marrow derived MSCs, providing new insights into the development of C. acnes during associated implant infections.

Background: Human cases of West Nile virus (WNV) infection are recorded since 2010 in Greece, with seasonal outbreaks occurring almost annually. Enhanced surveillance has been implemented since 2010, to promptly characterise cases’ temporal and geographical distribution and inform authorities for implementation of appropriate measures (mosquito control, health education, blood safety). Aim: We describe the epidemiology of WNV human infections in Greece focusing on the 2018 season. Methods: The National Public Health Organization advised physicians to test all suspect WNV infection cases and refer samples to reference laboratories. Laboratories notified diagnosed cases on a daily basis. Treating physicians, patients, and infected blood donors were interviewed within 48hours after diagnosis and the probable infection location was identified. Hospitalised cases were followed up until discharge. Results: A total of 317 autochthonous WNV infection cases were diagnosed in 2018. Among them, 243 cases had neuroinvasive disease (WNND), representing a 23% increase of WNND cases compared with 2010, the previous most intense season. There were 51 deaths. Cases started occurring from week22, earlier than usual. Both rural and urban areas were affected, with 86 (26% of the total) municipalities belonging to seven (54% of the total) regions recording cases. Two major epicentres were identified in Attica and Central Macedonia regions. Conclusions: The largest number of human cases of WNV infection ever recorded in Greece occurred in 2018, with a wide geographical distribution, suggesting intense virus circulation. With the observation of the gravitational wave event of August 17th 2017 the multi-messenger astronomy era has definitely begun. With the opening of this new panorama, it is necessary to have new instruments and a perfect coordination of the existing observatories. Crystal Eye is a detector aimed at the exploration of the electromagnetic counterpart of the gravitational waves. Such events generated by neutron stars' mergers are associated with gamma-ray bursts (GRB). At present, there are few instruments in orbit able to detect photons in the energy range going from tens of keV up to few MeV. These instruments belong to two different old observation concepts: the all sky monitors (ASM) and the telescopes. The detector we propose is a crossover technology, the Crystal Eye: a wide field of view observatory in the energy range from 10 keV to 10 MeV with a pixelated structure. A pathfinder will be launched with Space RIDER in 2022. We here present the preliminary results of the characterization of the first pixel.

Despite the increasing interest in studying Cladocera and Copepoda in Philippine freshwaters, there is a need to update our knowledge on its taxonomy and distribution in highly urbanized areas, such as Metropolitan Manila. This paper presents an updated listing of freshwater microcrustaceans and their distribution in Metropolitan Manila, considering the continued deterioration of water quality and increased urbanization in many areas since the last comprehensive study on these taxa in 1950s. We collected water samples from 33 freshwater sites in Metropolitan Manila and 23 of which were found to contain microcrustacean zooplankton. A total of 13 species were identified including two new locality records for C. cornuta and D. dubium in Pasig River. All 8 cladocerans, we identified have already recorded in previous studies while 3 cyclopoid copepods are new records for Metropolitan Manila. Furthermore, 16% of all known freshwater microcrustacean zooplankton species in the Philippines are found in Metropolitan Manila, including the Luzon-endemic Filipinodiaptomus insulanus collected from man-made reservoirs in less-disturbed habitats (an urban wildlife park, golf course and eco-park). Other identified species such as Thermocyclops taihokuensis may serve as indicator for high levels of nutrient. These results point to the importance of monitoring urban aquatic biodiversity for their potential in determining aquatic ecosystem health and of maintaining urban aquatic sanctuaries which may serve as alternative habitat for flora and fauna in rapidly developing urban centers such as Metropolitan Manila.
