Abstract Background Surfactant is a well-established therapy for preterm neonates affected by respiratory distress syndrome (RDS). The goals of different methods of surfactant administration are to reduce the duration of mechanical ventilation and the severity of bronchopulmonary dysplasia (BPD); however, the optimal administration method remains unknown. This study compares the effectiveness of the INtubate-RECruit-SURfactant-Extubate (IN-REC-SUR-E) technique with the less-invasive surfactant administration (LISA) technique, in increasing BPD-free survival of preterm infants. This is an international unblinded multicenter randomized controlled study in which preterm infants will be randomized into two groups to receive IN-REC-SUR-E or LISA surfactant administration. Methods In this study, 382 infants born at 24+0–27+6 weeks’ gestation, not intubated in the delivery room and failing nasal continuous positive airway pressure (nCPAP) or nasal intermittent positive pressure ventilation (NIPPV) during the first 24 h of life, will be randomized 1:1 to receive IN-REC-SUR-E or LISA surfactant administration. The primary outcome is a composite outcome of death or BPD at 36 weeks’ postmenstrual age. The secondary outcomes are BPD at 36 weeks’ postmenstrual age; death; pulse oximetry/fraction of inspired oxygen; severe intraventricular hemorrhage; pneumothorax; duration of respiratory support and oxygen therapy; pulmonary hemorrhage; patent ductus arteriosus undergoing treatment; percentage of infants receiving more doses of surfactant; periventricular leukomalacia, severe retinopathy of prematurity, necrotizing enterocolitis, sepsis; total in-hospital stay; systemic postnatal steroids; neurodevelopmental outcomes; and respiratory function testing at 24 months of age. Randomization will be centrally provided using both stratification and permuted blocks with random block sizes and block order. Stratification factors will include center and gestational age (24+0 to 25+6 weeks or 26+0 to 27+6 weeks). Analyses will be conducted in both intention-to-treat and per-protocol populations, utilizing a log-binomial regression model that corrects for stratification factors to estimate the adjusted relative risk (RR). Discussion This trial is designed to provide robust data on the best method of surfactant administration in spontaneously breathing preterm infants born at 24+0–27+6 weeks’ gestation affected by RDS and failing nCPAP or NIPPV during the first 24 h of life, comparing IN-REC-SUR-E to LISA technique, in increasing BPD-free survival at 36 weeks’ postmenstrual age of life. Microplastics pollution differentially impacts coral reef systems, by threatening corals physically, through physiological distress and by increasing diseases. However, most of the studies to date have focused on scleractinian corals. The present work reports for the first time the patterns of microplastic ingestion and adhesion by the alcyonacean Coelogorgia palmosa. Feeding and adhesion tests were carried out with various concentrations of polyethylene microbeads. Results showed a wide range of surface adhesion, ranging from 3 to 1573 microbeads per coral fragment, suggesting that adhesion driven by mucus is the main mechanism of microplastic trapping. Polyethylene was ingested by 60% of coral fragments, and the average number of ingested microbeads was much lower compared to scleractinian corals. Considering the ecological importance of soft corals in coral reef ecosystems, specific attention regarding microplastic pollution effects on this taxon is recommended.

Awareness of plastic pollution in marine habitats, such as coral reefs, has grown in recent years. Several studies have shown that tiny particles resulting from plastic breakdown, especially microplastics, can potentially harm corals. However, to date, there is very little evidence regarding the impact that nanoplastics (<1 μm) can have on the physiology and health of corals, particularly soft corals. In this study, we exposed the soft coral Pinnigorgia flava to two concentrations (0.1 and 1 mg/L) of secondary nanoplastics-specifically nanofibers obtained from the photodegradation of polypropylene nonwoven fabrics-and their related leachates, to evaluate the coral's cellular response through the analysis of antioxidant enzyme activities (SOD, CAT, GST, GR). Chemo-physical characterization of the nano-aggregates displayed an average size of 224.3 ± 8.1 nm, while GC-MS analyses of the leachates showed a variety of mono- and dicarboxylic acids. Although both nanoplastic treatments generated a cellular oxidative stress response, the physical interaction with secondary plastic fiber nano-aggregates affected cellular homeostasis more than the chemical interaction with the released compounds, triggering a stronger antioxidant response. The activity of all antioxidant enzymes increased with higher nanofiber concentrations, while this trend was not consistently observed for the leachates. Overall, SOD and CAT were the two most responsive antioxidant enzymes in cellular detoxification. Coral diseases are an increasing threat to continuously degrading reefs as disease abundance and virulence are expected to increase in the future. Active conservation measures, specifically coral restoration projects, are increasingly being implemented worldwide. Yet little is known about the implications of disease occurrence in a restoration context. This study describes white syndrome pathogenesis in 2 coral genera, farmed in 2 mid-water rope nurseries located in a reef and a lagoon habitat in the Maldives. Over a 112 d monitoring period, disease metrics were obtained from 336 Pocillopora and 96 Acropora fragments to assess the impacts of unmitigated disease progression in coral nurseries. Disease had low prevalence (2.2%) and incidence (0.007) in reef-farmed Pocillopora, but survival (91%) was significantly reduced in comparison with the unaffected lagoon-farmed Pocillopora (99%). In contrast, lagoon-farmed Acropora showed high disease prevalence (78.5%) and incidence (0.064), and a lower survival (79%) than disease-unaffected reef-farmed Acropora (98%). These findings had distinct implications for coral gardening success. While Pocillopora was considered suitable for outplanting, especially since subsequent mitigation interventions were successful, the diseased, lagoon-reared Acropora posed a potential risk to the restoration site and was unsuitable for outplanting following 1 yr of farming effort. Our findings demonstrate that unmitigated diseases can cause major setbacks to restoration success. Coral gardening projects are likely to be particularly susceptible; therefore, disease monitoring and risk management are imperative. Since there is currently a lack of available diagnostic and mitigative tools, this study aims to provide case study data for restoration practitioners.

The Mediterranean is known for its marine biodiversity, especially gorgonian forests. Unfortunately, these are experiencing rapid declines due to climate change, manifested by repeated marine heat waves resulting in mass mortality events since the early 1990 s. To better understand why gorgonians are declining, more systematic approaches to investigate the exact causes are needed, and pathology may aid in this goal. We described gross and microscopic pathology of tissue loss in three key gorgonian species in the Mediterranean region, Paramuricea clavata, Eunicella cavolini, and Leptogorgia sarmentosa, that were all experiencing various degrees of acute to subacute tissue loss characterized by exposed axial skeleton sometimes partly colonized by epibionts and thinning of adjacent tissues. The most significant variety of lesions was seen in P. clavata followed by L. sarmentosa and E. cavolini. For all species, dissociation of gastrodermal cells was the dominant microscopic lesion followed by necrosis of the gastrodermis. Ciliates invading gastrodermis and associated with necrosis of polyps were seen only in E. cavolini. Epidermal tissue loss was seen only in L. sarmentosa, while P. clavata was distinguished by a prominent inflammatory response and unidentified dark round structures within the tentacle epidermis and gastrodermis with no host response. Coral reefs are lately suffering a fast decline in biodiversity due to the coupled effect of climate change and disease outbreaks, which in recent decades have been reported with higher frequency and shorter intervals. Limited studies have been conducted on coral diseases in the Maldives resulting in the impossibility of assessing the temporal trend in their dynamics. In this context, we evaluated the change in the distribution, prevalence, and host range of 4 diseases, namely black band disease (BBD), brown band disease (BrB), skeletal eroding band (SEB) and white syndrome (WS), in the reef system around Thudufushi Island after an interval of 12 yr since the last assessment. In this period, the overall disease prevalence increased, except for BrB, with SEB showing the most severe increase in 2022 in comparison to 2010. The overall average prevalence of coral diseases is approximately 2%, indicating an increase of about 0.7% since 2010. Diseased coral colonies were found in all the investigated sites, with the east site being the most affected and SEB emerging as the most prevalent disease across all the investigated sites. The affected colonies belong to 13 genera, with Psammocora genus showing the highest overall mean disease prevalence. This study depicted a basic temporal trend in disease prevalence that confirms an increase in coral diseases in the region and calls for a dedicated national monitoring protocol to better understand and predict future coral disease dynamics at regional scales.

Background and Aim: First, to compare somatosensory evoked potentials (SEPs) in preterm newborns without major brain injury studied at term equivalent age (TEA) with a term historical control group. Second, to investigate the impact of pain exposure during the first 28 days after birth on SEPs. Third, to evaluate the association between SEPs and Bayley-III at 2 years corrected age (CA)., Methods: Infants born at <32 weeks' gestational age (GA) were studied with continuous-SEPs. First, SEP differences between preterm and term infants were analyzed. Second, regression analyses were conducted to explore the association between SEPs and painful procedures, and then between SEPs and neurodevelopment., Results: 86 preterm infants were prospectively enrolled. Preterm infants exhibited prolonged N1 latencies, central conduction times (CCTs), lower N1-P1 amplitudes, and more recurrently abnormal SEPs compared to term infants. Higher pain exposure predicted longer N1 latency and slower CCT (all p < 0.005), adjusting for clinical risk factors. Younger GA and postmenstrual age (PMA) at SEP recording were associated with longer N1 latency and lower N1-P1 amplitude (all p < 0.005). A normal SEP at TEA positively predicted cognitive outcome at 2 years CA ( p < 0.005)., Conclusion: Pain exposure and prematurity were risk factors for altered SEP parameters at TEA. SEPs predicted cognitive outcome.

At present the information regarding the occurrence of human pharmaceuticals (PhaCs) in coral reefs and their potential impacts on the associated fauna is limited. To optimize the collection of data in these delicate environments, we employed a solid-phase microextraction (bioSPME) and liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) procedure that enabled in vivo determinations in soft corals. Specifically, we researched the antibiotics Ofloxacin Sulfamethoxazole and Clarithromycin, the anti-inflammatory Diclofenac Propyphenazone Ketoprofen and Amisulpride, the neuroactive compounds Gabapentin-lactam, the beta-blocker Metoprolol and the antiepileptic Carbamazepine. Reproducibility was between 2.1% and 9.9% and method detection limits LODs) were between 0.2 and 1.6 ng/g and LOQs between 0.8 and 5.4 mg/g. The method was then applied to establish a baseline for the occurrence of these compounds in the Maldivian archipelago. Colonies of Sarcophyton sp. and Sinularia sp. were sampled along an inner-outer reef transect. Five of the ten targeted PhaCs were identified, and 40% of the surveyed coral colonies showed the occurrence of at least one of the selected compounds. The highest concentrations were found inside the atoll rim. Oxoflacin (9.5 ± 3.9 ng/g) and Ketoprofen (4.5 ± 2.3 ng/g) were the compounds with the highest average concentrations. Outside the atoll rim, only one sample showed contamination levels above the detection limit. Sharks are particularly susceptible to bioaccumulation due to their life history characteristics and trophic position within marine ecosystems. Despite this, studies of bioaccumulation cover only a small proportion of extant species. In this study we report concentrations of trace elements and heavy metals in blood samples of Sphyrna lewini for the first time. We report high concentrations of several trace elements and heavy metals, with concentrations of some elements exceeding the limit determined safe for human consumption. High elemental concentrations may reflect biochemical differences between blood plasma and other tissues; however, they may also be symptomatic of high levels of exposure triggered by anthropogenic activities. We also provide evidence of elemental accumulation through ontogeny, the nature of which differs from that previously reported. Capitate hydrozoans are a morphologically and ecologically diverse hydrozoan suborder, currently including about 200 species. Being grouped in two clades, Corynida and Zancleida, these hydrozoans still show a number of taxonomic uncertainties at the species, genus and family levels. Many Capitata species established symbiotic relationships with other benthic organisms, including bryozoans, other cnidarians, molluscs and poriferans, as well as with planktonic dinoflagellates for mixotrophic relationships and with bacteria for thiotrophic ectosymbioses. Our study aimed at providing an updated and comprehensive phylogeny reconstruction of the suborder, at modelling the evolution of selected morphological and ecological characters, and at testing evolutionary relationships between the symbiotic lifestyle and the other characters, by integrating taxonomic, ecological and evolutionary data. The phylogenetic hypotheses here presented shed light on the evolutionary relationships within Capitata, with most families and genera being recovered as monophyletic. The genus Zanclea and family Zancleidae, however, were divided into four divergent clades, requiring the establishment of the new genus Apatizanclea and the new combinations for species in Zanclea and Halocoryne genera. The ancestral state reconstructions revealed that symbiosis arose multiple times in the evolutionary history of the Capitata, and that homoplasy is a common phenomenon in the group. Correlations were found between the evolution of symbiosis and morphological characters, such as the perisarc. Overall, our results highlighted that the use of genetic data and a complete knowledge of the life cycles are strongly needed to disentangle taxonomic and systematic issues in capitate hydrozoans. Finally, the colonization of tropical habitat appears to have influenced the evolution of a symbiotic lifestyle, playing important roles in the evolution of the group., (© 2023 The Authors. Microplastic pollution is an increasing threat to coral reefs, which are already strongly challenged by climate change-related heat stress. Although it is known that scleractinian corals can ingest microplastic, little is known about their egestion and how microplastic exposure may impair corals at physiological and cellular levels. In addition, the effects of microplastic pollution at current environmental concentration have been little investigated to date, particularly in corals already impacted by heat stress. In this study, the combined effects of these environmental threats on Pocillopora damicornis were investigated from a physical and cellular perspective. Colonies were exposed to three concentrations of polyethylene microplastic beads (no microplastic beads: [No MP], 1 mg/L: [Low MP]; 10 mg/L: [High MP]), and two different temperatures (25 °C and 30 °C) for 72 h. No visual signs of stress in corals, such as abnormal mucus production and polyp extroflection, were recorded. At [Low MP], beads adhered to colonies were ingested but were also egested. Moreover, thermally stressed colonies showed a lower adhesion and higher egestion of microplastic beads. Coral bleaching was observed with an increase in temperature and microplastic bead concentration, as indicated by a general decrease in chlorophyll concentration and Symbiodiniaceae density. An increase in lipid peroxidation was measured in colonies exposed to [Low MP] and [High MP] and an up-regulation of stress response gene hsp70 was observed due to the synergistic interaction of both stressors. Overall, our findings showed that heat stress still represents the main threat to P. damicornis, while the effect of microplastics on coral health and physiology may be minor, especially at control temperature. However, microplastics could exacerbate the effect of thermal stress on cellular homeostasis, even at [Low MP]. The Mediterranean Sea's biodiversity is declining due to climate change and human activities, with plastics and emerging contaminants (ECs) posing significant threats. This study assessed phthalic acid esters (PAEs) and active pharmaceutical ingredients (APIs) occurrence in four anthozoan species (Cladocora caespitosa, Eunicella cavolini, Madracis pharensis, Parazoanthus axinellae) using solid phase microextraction (SPME) and liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). All specimens were contaminated with at least one contaminant, reaching maximum values of 57.3 ng/g for the ∑PAEs and 64.2 ng/g (wet weight) for ∑APIs, with dibutyl phthalate and Ketoprofen being the most abundant. P. axinellae was the most contaminated species, indicating higher susceptibility to bioaccumulation, while the other three species showed two-fold lower concentrations. Moreover, the potential adverse effects of these contaminants on anthozoans have been discussed. Phthalates are widely employed plasticizers blended to plastic polymers that, during plastic aging and weathering are prone to leach in the surrounding environment. Thus, phthalates were proposed to indirectly evaluate MPs contamination in marine environments, with still uncertain and scarce data, particularly for wildlife. This study investigates simultaneously microplastics (MPs) and phthalates (PAEs) occurrence in wild Actinia equina and Anemonia viridis, two common and edible sea anemone species. Both species had a 100 % frequency of MPs occurrence, with similar average concentrations. PAEs were detected in 70 % of samples, with concentrations up to 150 ng/g in A. equina and 144.3 ng/g for A. viridis. MPs and PAEs present in sea anemone tissues appear to reflect seawater plastic contamination conditions in the study area. Objective: The present study aimed to explore first the impact of perinatal risk factors on flash-VEP waves and morphology in a group of preterm infants studied at term equivalent age (TEA). Second, to correlate VEP morphology with neurological outcome at 2 years corrected age (CA)., Methods: Infants with a gestational age (GA) at birth <32 weeks, without major brain injury, were enrolled. Multivariate regression analyses were performed, and the models were run separately for each dependent variable N2, P2, N3 latencies and P2 amplitude. Logistic regression was applied to study N4 component (present/absent) and VEP morphology (regular/irregular). The predictors were GA, bronchopulmonary dysplasia (BPD), postmenstrual age at VEP registration, cumulative morphine and fentanyl dose, and painful procedures. Lastly, linear regression models were performed to assess the relation between the Bayley-III cognitive and motor scores at 2 years CA and VEP morphology, in relation to GA, BPD, painful procedures and cumulative morphine dose., Results: Eighty infants were enrolled. Morphine was the predictor of N2 (R 2  = 0.09, p  = 0.006), P2 (R 2  = 0.11, p  = 0.002), and N3 (R 2  = 0.13, p  = 0.003) latencies. Younger GA was associated with lower amplitude (R 2  = 0.05, p  = 0.029). None of the independent variables predicted the presence of N4 component, nor VEP morphology in the logistic analysis. VEP morphology was not associated with cognitive and motor scores at 2 years., Conclusions: Morphine treatment and prematurity were risk factors for altered VEPs parameters at TEA. Verified records of the bull shark Carcharhinus leucas are lacking in the Maldives. This study provides the first confirmed evidence of 23 sightings observed from 2013 to 2023 in the central and southern atolls of this archipelago. Most of the sightings occurred in close proximity to inhabited areas, where food waste is often discarded into the water, or in several dive sites, suggesting the presence of this species in different locations around central and southern atolls. Although further research is required to fully investigate the C. leucas population in the Maldives, this report documents and confirms its presence in this region., (© 2023 The Authors. Massive coral bleaching episodes induced by thermal stress are one of the first causes of coral death worldwide. Overproduction of reactive oxygen species (ROS) has been identified as one of the potential causes of symbiosis breakdown between polyps and algae in corals during extreme heat wave events. Here, we propose a new strategy for mitigating heat effects by delivering underwater an antioxidant to the corals. We fabricated zein/polyvinylpyrrolidone (PVP)-based biocomposite films laden with the strong and natural antioxidant curcumin as an advanced coral bleaching remediation tool. Biocomposites' mechanical, water contact angle (WCA), swelling, and release properties can be tuned thanks to different supramolecular rearrangements that occur by varying the zein/PVP weight ratio. Following immersion in seawater, the biocomposites became soft hydrogels that did not affect the coral's health in the short (24 h) and long periods (15 days). Laboratory bleaching experiments at 29 and 33 °C showed that coral colonies of Stylophora pistillata coated with the biocomposites had ameliorated conditions in terms of morphological aspects, chlorophyll content, and enzymatic activity compared to untreated colonies and did not bleach. Finally, biochemical oxygen demand (BOD) confirmed the full biodegradability of the biocomposites, showing a low potential environmental impact in the case of open-field application. These insights may pave the way for new frontiers in mitigating extreme coral bleaching events by combining natural antioxidants and biocomposites.

Phytochromes are red-light photoreceptors that regulate a wide range of physiological processes in plants, fungi and bacteria. Canonical bacteriophytochromes are photosensory histidine kinases that undergo light-dependent autophosphorylation, thereby regulating cellular responses to red light via two-component signaling pathways. However, the molecular mechanism of kinase activation remains elusive for bacteriophytochromes. In particular, the directionality of autophosphorylation is still an open question in these dimeric photoreceptor kinases. In this work, we perform histidine kinase assays on two tandem bacteriophytochromes RpBphP2 and RpBphP3 from the photosynthetic bacterium Rhodopseudomonas palustris. By examining the kinase activities of full-length bacteriophytochromes and two loss-of-function mutants under different light conditions, we demonstrate that RpBphP2 and RpBphP3 undergo light-dependent trans-phosphorylation between protomers in both homodimeric and heterodimeric forms. We have further determined the crystal structure of the histidine kinase domains of RpBphP2 at 3.19 Å resolution. Conventional wisdom holds that pre-event planning is a key factor in effective disaster response. In assessing the response to the Coronavirus Disease 2019 (COVID-19) pandemic, it is necessary to evaluate the extent to which emergency management agencies were prepared to respond to a pandemic, particularly given the unusual nature of this event, ie, scope, scale, and length of response. While emergency management agencies at every level of government have been involved in the COVID-19 response, state-level governments have taken on a prominent and atypical lead-ership role. This study assesses the extent and role that emergency management agencies planned for a pandemic scenario. Understanding the extent to which state-level emergency management agencies planned for an event like the COVID-19 pandemic and what they anticipated their roles to provide insight for future revisions in pandemic planning. This study addresses two related research questions: RQ 1: To what extent did state-level emergency management agencies account for a pandemic in emergency management response plans prior to COVID-19? RQ 2: What was the planned role of state-level emergency management agencies in the response to a pandemic? An analysis of state-level emergency management plans found that, although all states with available emergency management response plans included pandemics, there was significant variation in the extent of the inclusion, and the role prescribed for emergency management. Public health and emergency management response plans were congruent with respect to the planned role of emergency management.

The presence of microplastics in the world's oceans and their effects on marine habitats are highly concerning. As suspension-feeders, corals are very exposed to microplastics, compromising the health of coral reef ecosystems. In this study we surveyed for the first time the presence of microplastics in Maldivian reef-building corals. Aiming to determine the influence of exposure and depth on microplastic distribution, analyses were carried out on 38 individuals belonging to three different species. 58% of the investigated colonies resulted contaminated with particles within the 25-150 μm size range. The maximum concentration was encountered in a Pocillopora verrucosa colony sampled from a shallow inner reef (8.9 particles/g of coral). No significant differences in microplastic concentration were observed between different depths, exposures, sites and species. The bioconcentration of dimethyl phthalate (DMP) diethyl phthalate (DEP) dibutyl phthalate (DBP) butyl benzyl phthalate (BBzP), di-(2-ethy hexyl) phthalates (DEHP), mono-butyl phthalate (MBP), mono-benzyl phthalate (MBzP), mono-(2-ethy hexyl) phthalate (MEHP) in the soft corals Coelogorgia palmosa, Sinularia sp., Sarcophyton glaucum, and Lobophytum sp. was investigated. Specimens were cultured in a microcosm environment built-up at the Genova Aquarium and analyses were carried out by in vivo SPME-LC-MS/MS. The distributions of the phthalates among the four surveyed species resulted significantly different. Calculated bioconcentration factors (BCFs) showed values spanning over two orders of magnitude, from a minimum of log10 BCF DEP  = 1.0 in Sarcophyton glaucum to a maximum of log10 BCF DBP  = 3,9 calculated for Coelogorgia palmosa. Moreover, the calculated BCFs of the long chain phthalates resulted up to three orders of magnitude lower than theoretically predicted (from logK ow ), whereas BCF of short chain phthalates resulted higher. The general public's understanding of disasters is influenced by the portrayal of such events in popular culture. Disaster films have remained a core attraction in this regard. A systematic assessment of the most recent disaster film cycle (that is, from January 2000-December 2019) is warranted, therefore, to gain insights into the current landscape of the genre and to comprehend better the imagery that people encounter onscreen. This study evaluated 173 disaster films and found that most depict natural hazards and global catastrophes despite films about monsters and smaller-scale disasters being the most popular. It provides a foundation for future research on the relevance of disaster films to disaster scholarship and emergency management practice. Coral restoration initiatives are gaining significant momentum in a global effort to enhance the recovery of degraded coral reefs. However, the implementation and upkeep of coral nurseries are particularly demanding, so that unforeseen breaks in maintenance operations might jeopardize well-established projects. In the last 2 years, the COVID-19 pandemic has resulted in a temporary yet prolonged abandonment of several coral gardening infrastructures worldwide, including remote localities. Here we provide a first assessment of the potential impacts of monitoring and maintenance breakdown in a suite of coral restoration projects (based on floating rope nurseries) in Colombia, Seychelles, and Maldives. Our study comprises nine nurseries from six locations, hosting a total of 3,554 fragments belonging to three coral genera, that were left unsupervised for a period spanning from 29 to 61 weeks. Floating nursery structures experienced various levels of damage, and total fragment survival spanned from 40 to 95% among projects, with Pocillopora showing the highest survival rate in all locations present. Overall, our study shows that, under certain conditions, abandoned coral nurseries can remain functional for several months without suffering critical failure from biofouling and hydrodynamism. Still, even where gardening infrastructures were only marginally affected, the unavoidable interruptions in data collection have slowed down ongoing project progress, diminishing previous investments and reducing future funding opportunities. The need for comprehensive and effective coral restoration projects, as part of a broader conservation management strategy, is accelerating in the face of coral reef ecosystem decline. This study aims to expand the currently limited knowledge base for restoration techniques in the Maldives by testing the performance of mid-water rope nurseries in a lagoon and a reef habitat. We examined whether different coral farming habitats impacted fragment survival, health and growth of two coral genera and how the occurrence of mutualistic fauna, predation and disease influenced coral rearing success. Two nurseries were stocked with a total of 448 Pocillopora verrucosa and 96 Acropora spp. fragments, divided into different groups (four Pocillopora groups: lagoon nursery at 5 m; reef nursery at 5, 10 and 15 m; two Acropora groups: lagoon nursery at 5 m and reef nursery at 5 m). Eight fragment replicates from the same donor colony ( Pocillopora genets: N  = 14, Acropora genets N  = 6) were used in each group and monitored for one year. Our results show that fragment survival was high in both farming habitats (>90%), with P. verrucosa surviving significantly better in the lagoon and Acropora spp. surviving and growing significantly faster in the reef nursery. P. verrucosa growth rates were similar between reef and lagoon habitat. Different rearing depths in the reef nursery had no impact on the survival of P. verrucosa but coral growth decreased considerably with depth, reducing fragments' ecological volume augmentation and growth rates by almost half from 5 to 15 m depth. Further, higher fish predation rates on fragments were recorded on the reef, which did not impact overall nursery performance. Mutualistic fauna, which correlated positively with fragment survival, was more frequently observed in the lagoon nursery. The occurrence of disease was noted in both habitats, even though implications for fragment health were more severe in the lagoon. Overall, our study demonstrates that lagoon and reef nurseries are suitable for rearing large numbers of coral fragments for transplantation. Nevertheless, we recommend considering the specific environmental conditions of the farming habitat, in particular water quality and year-round accessibility, in each case and to adjust the coral farming strategy accordingly. The octocoral family Alcyoniidae represents a rich source of bioactive substances with intriguing and unique structural features. This review aims to provide an updated overview of the compounds isolated from Alcyoniidae and displaying potential cytotoxic activity. In order to allow a better comparison among the bioactive compounds, we focused on molecules evaluated in vitro by using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, by far the most widely used method to analyze cell proliferation and viability. Specifically, we surveyed the last thirty years of research, finding 153 papers reporting on 344 compounds with proven cytotoxicity. The data were organized in tables to provide a ranking of the most active compounds, to be exploited for the selection of the most promising candidates for further screening and pre-clinical evaluation as anti-cancer agents. Specifically, we found that (22 S ,24 S )-24-methyl-22,25-epoxyfurost-5-ene-3β,20β-diol ( 16 ), 3β,11-dihydroxy-24-methylene-9,11-secocholestan-5-en-9-one ( 23 ), (24 S )-ergostane-3β,5α,6β,25 tetraol ( 146 ), sinulerectadione ( 227 ), sinulerectol C ( 229 ), and cladieunicellin I ( 277 ) exhibited stronger cytotoxicity than their respective positive control and that their mechanism of action has not yet been further investigated.

The hydrozoan family Cladocorynidae inhabits tropical to temperate waters and comprises the two genera Pteroclava and Cladocoryne. Pteroclava lives in association with some octocorals and hydrozoans, whereas Cladocoryne is more generalist in terms of substrate choice. This work provides a thorough morpho-molecular reassessment of the Cladocorynidae by presenting the first well-supported phylogeny of the family based on the analyses of three mitochondrial and four nuclear markers. Notably, the two nominal genera were confirmed to be monophyletic and both morphological and genetic data led to the formal description of a new genus exclusively associated with octocorals, Pseudozanclea gen. nov. Maggioni & Montano. Accordingly, the diagnosis of the family was updated. The ancestral state reconstruction of selected characters revealed that the symbiosis with octocorals likely appeared in the most recent common ancestor of Pteroclava and Pseudozanclea. Additionally, the presence of euryteles aggregation in the polyp stage and the exumbrellar nematocyst pouches with euryteles represent synapomorphies of all cladocorynid taxa and probably emerged in their most recent common ancestor. The analysis of several Pteroclava krempfi colonies from Indo-Pacific and Caribbean localities associated with several host octocorals revealed a high intra-specific genetic variability. Single- and multi-locus species delimitations resulted in three to five species hypotheses, but the statistical analysis of morphometric data showed only limited distinction among the clades of P. krempfi. However, P. krempfi clades showed differences in both host specificity, mostly at the octocoral family level, and geographic distribution, with one clade found exclusively in the Caribbean Sea and the others found in the Indo-Pacific., (© 2021 The Authors. Ecosystems face both local hazards, such as over-exploitation, and global hazards, such as climate change. Since the impact of local hazards attenuates with distance from humans, local extinction risk should decrease with remoteness, making faraway areas safe havens for biodiversity. However, isolation and reduced anthropogenic disturbance may increase ecological specialization in remote communities, and hence their vulnerability to secondary effects of diversity loss propagating through networks of interacting species. We show this to be true for reef fish communities across the globe. An increase in fish-coral dependency with the distance of coral reefs from human settlements, paired with the far-reaching impacts of global hazards, increases the risk of fish species loss, counteracting the benefits of remoteness. Hotspots of fish risk from fish-coral dependency are distinct from those caused by direct human impacts, increasing the number of risk hotspots by ~30% globally. These findings might apply to other ecosystems on Earth and depict a world where no place, no matter how remote, is safe for biodiversity, calling for a reconsideration of global conservation priorities., (© 2021. Malaria elimination efforts in Peru have dramatically reduced the incidence of cases in the Amazon Basin. To achieve the elimination, the detection of asymptomatic and submicroscopic carriers becomes a priority. Therefore, efforts should focus on tests sensitive enough to detect low-density parasitemia, deployable to resource-limited areas and affordable for large screening purposes. In this study, we assessed the performance of the Malachite-Green LAMP (MG-LAMP) using heat-treated DNA extraction (Boil & Spin; B&S MG-LAMP) on 283 whole blood samples collected from 9 different sites in Loreto, Peru and compared its performance to expert and field microscopy. A real-time PCR assay was used to quantify the parasite density. In addition, we explored a modified version of the B&S MG-LAMP for detection of submicroscopic infection in 500 samples and compared the turnaround time and cost of the MG-LAMP with microscopy. Compared to expert microscopy, the genus B&S MG-LAMP had a sensitivity of 99.4% (95%CI: 96.9%- 100%) and specificity of 97.1% (95%CI: 91.9%- 99.4%). The P. vivax specific B&S MG-LAMP had a sensitivity of 99.4% (96.6%- 100%) and specificity of 99.2% (95.5%- 100%) and the P. falciparum assay had a sensitivity of 100% (95%CI: 78.2%- 100%) and specificity of 99.3% (95%CI: 97.3%- 99.8%). The modified genus B&S MG-LAMP assay detected eight submicroscopic malaria cases (1.6%) which the species-specific assays did not identify. The turnaround time of B&S MG-LAMP was faster than expert microscopy with as many as 60 samples being processed per day by field technicians with limited training and utilizing a simple heat-block. The modified B&S MG-LAMP offers a simple and sensitive molecular test of choice for the detection of submicroscopic infections that can be used for mass screening in resources limited facilities in endemic settings nearing elimination and where a deployable test is required., Competing Interests: KAB is an employee of Vysnova and reports personal fees outside the submitted work.
