Zusammenfassung Einleitung: Messverfahren zur Bestimmung von Beinvolumina objektivieren im klinischen Alltag Therapieverläufe und werden häufig im Rahmen klinischer Studien benötigt. Als Gold-standard bei der Volumenmessung gilt derzeit die Wasserplethysmographie. Sie ist bei vielen klinischen Studien Mittel der Wahl.Ziel der Studie war es, ein berührungsloses optisches Messverfahren „Venotrain Image 3D” mit dem Goldstandard, der Wasserplethysmographie, zur Beurteilung von Beinvolumina bzw. der Volumenveränderung im zeitlichen Verlauf zu vergleichen. Wir untersuch-ten, ob sich „Venotrain Image 3D“ als Instrument der Volumenmessung im untersuchten Kollektiv eignet. Patienten und Methoden: Untersucht wurden 21 schwangere Frauen im Alter von 27 bis 42 Jahren (MW 33,4 ± 4,4 Jahre). Bei Einschluss in die Studie befanden sich die Studienteilnehmerinnen in der 12. bis 28. Schwangerschaftswoche. Sie wurden prospektiv über 16 Wochen untersucht. Bis zur Entbindung wur-den die Patientinnen in 4-wöchigen Abstän-den sowie 3 Monate postpartum bezüglich der Beinvolumina mittels „Venotrain Image 3D” und zur Unterschenkelvolumenbestim-mung mittels Wasserplethysmographie untersucht. Dabei werden folgende Kompartimente erfasst: Wasserplethysmographie: Fuß- und Unterschenkelverdrängung bis zu einer Höhe von 28 cm oberhalb der Fußsohle; „Image 3D“: automatische Auffindung Unterschenkel, Beginn: Extremalstelle cB bis Patellaspalt; Oberschenkel: Beginn: Patellaspalt bis Extremalstelle cG Ergebnisse: Da unterschiedliche Kompartimente vermessen wurden, eignen sich die gemessenen Absolutvolumina nur bedingt als Parameter zum Methodenvergleich. Die Korrelation der Messergebnisse beider Methoden betrug bei Betrachtung der Volumenveränderung im zeitlichen Verlauf r=0,78, p Schlussfolgerung: Die „Image 3D“-Methode zur Bestimmung der Beinvolumina korreliert sehr eng mit den Ergebnissen der Wasserplethysmographie hinsichtlich relativer und absoluter Unterschenkelvolumina. Bei den Absolutvolumina kommt es jedoch zur systematischen Abweichung bei großen Unterschenkelvolumina. Dabei stellt die Unterschenkellänge die entscheidende Einflussgröße dar. Beide Methoden messen dabei unterschiedliche Kompartimente des Unterschenkels, sodass je nach Fragestel-lung die eine der anderen Methode überlegen zu sein scheint.

This article addresses the strategic network planning for international automotive manufacturers, in particular of premium cars. The focus is on the product to plant allocation and capacity expansion decisions for a given network design with fixed plant locations. A mixed integer program minimizing the net present value (NPV) of all capital expenditures and operational cost while incorporating flexibility of a network by a specific allocation structure (“chain”) is formulated. Computational illustrations on the influence of flexible allocation structures on the NPV are demonstrated considering changes in demand, exchange rates and total available capacity.

We and others have demonstrated already that TRAIL (TNF-related apoptosis-inducing ligand) is a very promising candidate for molecular targeted anticancer therapy, especially when combined with ionizing radiation or other DNA-damaging agents. Agonist monoclonal antibodies that activate and are specific for the death signaling TRAIL receptors are an alternative method to stimulate the programmed cell death pathway. Phase 1 clinical trials have subsequently been conducted and shown a very good tolerability of these antibodies. In order to assess the efficacy of TRAIL receptor stimulation to induce cell death by this alternate method, we studied the combination of the agonistic-TRAIL receptor antibodies HGS-ETR1 and HGS-ETR2 with radiation in vitro and in vivo. Induction of apoptosis after combined treatment with TRAIL receptor antibodies HGS-ETR1 and/or HGS-ETR2 (0.01, 0.1, 1.0 mg/ml) and irradiation with 2, 5 or 10 Gy was determined by fluorescence microscopy and Western blot analysis of caspase-8 and PARP. The colorectal tumour cell lines Colo 205, HCT 116 and HCT-15 were used for in vitro experiments. Growth delay experiments were performed with combined treatment with fractionated irradiation (days 1-5 and 3 Gy single dose/day) and the receptor antibodies (intraperitonially, three different concentrations, application on days 1, 4 and 8) on Colo 205 xenograft-bearing NMRI (nu/nu) nude mice. HGS-ETR1 and HGS-ETR2 induced apoptotic cell death in a dose-dependent fashion and significantly increased cell death in combination with irradiation in vitro when compared to either irradiation or antibody treatment alone. The efficacy of the combined treatment seems to be at least partially Bax-dependent. Similar to the results from cell culture experiments, in vivo experiments demonstrated a dose-dependent delay in tumour growth after combined treatment. In vivo, in the Colo205 xenograft model, HGS-ETR2 revealed a higher activity than HGS-ETR1. This is the first study to demonstrate significant efficacy of combined treatment with the monoclonal agonistic TRAIL receptor antibodies HGS-ETR1 and HGS-ETR2 and ionising radiation in in vitro and in vivo models. We postulate that HGS-ETR1 and HGS-ETR2 will be very promising new agents in the field of molecular targeted multi-modality anticancer therapy.
