1. Identification and management of infections caused by Giardia sp., Trichuris sp. and Demodex sp. in captive Brazilian porcupines (Coendou prehensilis)

Abstract: The Brazilian porcupine (Coendou prehensilis, Rodentia, Erethizontidae) is an arboreal South American nocturnal rodent. Switzerland is home to one of the largest captive colonies in Europe. In June 2022, most of the animals in this colony showed severe diarrhoea, and Giardia sp. cysts were detected. All the animals were treated with metronidazole (75 mg/animal/day orally) for five days, repeating after two weeks. The diarrhoea continued, sometimes containing blood, and further analyses revealed Giardia sp. cysts and Trichuris sp. eggs with a particular undulating eggshell in pooled samples. The soil layer of some enclosures was removed to thoroughly clean and disinfect the underlying concrete floor. The animals were treated with fenbendazole (50 mg/kg/day orally) for 5 days repeating after three days. Giardia sp. cysts were not further detected. However, Trichuris sp. eggs were detected in branch bark samples and in six animals 2–3 months after treatment. The treatment with fenbendazole was repeated and no further Trichuris sp. eggs were detected. A 18S rRNA fragment consensus sequence showed 98.58% identity with Trichuris fossor. The Trichuris sp. in C. prehensilis may represent a new species, specific for arboreal porcupines. Demodex mites were observed in faecal flotations and thereafter in skin scrapings from five animals (four of them being family-related). A 16S consensus sequence showed 86.4% identity with other Demodex species. The animals were initially treated with moxidectin (0.4 and 0.8 mg/kg orally) and afterwards with sarolaner (10 mg/animal) but the treatments were not completely effective since in control scrapings, two animals evidenced few non-motile mites. An individual susceptibility and poor immunological control of the infection is suggested. Treatment with fenbendazole was effective against Giardia sp. and Trichuris sp. infections; however, reinfections may occur if the enclosures and tree branches are not deep cleaned and disinfected or replaced.

2. Switzerland-wide Neospora caninum seroprevalence in female cattle and identification of risk factors for infection

Abstract: IntroductionNeospora caninum is an important cause of abortion in cattle worldwide. Infection in cattle occurs horizontally by ingestion of oocysts shed by canids or vertically, from an infected dam to the fetus, and may result in abortion, stillbirth, or birth of seropositive offspring. The control of bovine neosporosis is difficult and costly. The objectives of this study were to estimate the current nationwide seroprevalence of N. caninum infections in Swiss cattle and to assess risk factors for infection with this parasite.MethodsWe conducted a cross-sectional study with cattle farms randomly selected and stratified according to population size, resulting in a sample of 780 female cattle. The cattle originated from 161 farms distributed over all Switzerland. The serum samples were tested for antibodies against N. caninum using a commercial ELISA and if inconclusive, retested using an in-house immunoblot technique. To collect farm parameters relevant to N. caninum transmission and prevention, farm owners were mailed a questionnaire which addressed topics putatively related to N. caninum infection such as husbandry, history of abortion, and presence of dogs on farm. Univariate analysis by generalized linear mixed model (with animal seropositivity as outcome variable) and logistic regression modeling (with farm seropositivity as outcome variable) was conducted on farm parameters investigated in the questionnaire.ResultsBy ELISA and immunoblot, 4.2% (33/780) of cattle sera yielded positive results. At the farm level, 16.2% (26/161) of the sampled farms had at least one seropositive animal. The return rate of the valid questionnaires was 54.0%. At the animal level, odds for farm seropositivity were 3.8 times higher when rodents had been recorded by the farmer as a problem on the farm. At the farm-level, two protective factors were identified: rearing of replacement heifers and feeding of concentrated feed.ConclusionWe recorded a low seroprevalence of N. caninum in a random sample of Swiss cattle representative for the years 2017–2018. Based on a questionnaire survey, we could identify risk and protective factors for infection with N. caninum, however their biological relevance needs to be confirmed in further studies.

3. Dynamics of Gastro-Intestinal Strongyle Parasites in a Group of Translocated, Wild-Captured Asiatic Wild Asses in Kazakhstan

Abstract: Asiatic wild ass (Kulan, Equus hemionus) population range and numbers became severely reduced and a reintroduction project is currently aiming to re-establish a population in the Central Steppe of Kazakhstan. Pre-emptive deworming is often recommended for equid translocations but eliminating parasites prior to translocation could cause disruptions in a balanced host-parasite relationship, adding an additional stressor to an already stressful intervention involving capture, transport, and adaptation to a new environment. Following a disease risk assessment, we decided against pre-emptive deworming and focused on monitoring the first group of nine translocated kulan in a large acclimatization enclosure prior to release. Over the 5-month acclimatization period, we regularly collected fecal samples and analyzed the shedding intensity of gastro-intestinal parasite eggs, obtained time budgets through behavioral observations, and visually assessed body condition. We identified strongyles (Strongylinae and Cyathostominae) and pinworms (Oxyuris equi) in fecal samples. All individuals shed strongyle eggs and two of the nine individuals had higher shedding intensities, but rarely reached levels for which deworming is recommended. All kulan appeared healthy throughout the observation period, aggressive interactions were very rare, and time budgets were very similar and dominated by feeding. Our results suggest that in translocation projects where the risk of introducing new parasites is minimal, pre-emptive treatment in wild equids can be replaced with non-invasive monitoring during the acclimatization period. We acknowledge that the small number of kulan, the large size of the enclosure, and the low temperatures during the animals stay in the acclimatization enclosure may all have reduced infestation pressure.

Abstract: Leptospirosis is a worldwide bacterial zoonosis which incidence is expected to increase in conjunction with global change. In urban ecosystems, synanthropic rats are the key source of Leptospira infection in humans and other animals. Risk assessment and prediction of human leptospirosis require investigations of the environment associated with the bacteria and infection patterns in the reservoir hosts. The objective of this study was to address the prevalence of mixed Leptospira infection in the lungs and kidneys of brown rats captured in three sites of the city centre of Vienna, Austria, between 2016 and 2018. A total of 96 brown rats were examined for the presence of Leptospira using PCR. Occurrence of mixed Leptospira infections was explored through next-generation sequencing (NGS). A logistic regression model was built to predict the individual infection status using morphological and land-use data. Overall, the prevalence of Leptospira interrogans in the kidney was 25% but varied among sites (0–36%). We did not evidence any pulmonary nor mixed infections. Host body mass and sex were strong predictors of Leptospira carriage in the sampled rats (relative variable importance (RVI) = 0.98 and 0.89, respectively) while the presence of water affected it moderately (RVI = 0.44). Our findings demonstrate that NGS is an unbiased approach to the direct characterisation of mixed leptospiral infections that could provide further insights into the ecology of Leptospira. Future surveillance programmes should consider the use of rats as sentinels for the early detection of emerging pathogenic Leptospira in urban ecosystems.

Abstract: Scanning superconducting quantum interference device (SQUID) microscopy is a magnetic imaging technique combining high-field sensitivity with nanometer-scale spatial resolution. State-of-the-art SQUID-on-tip probes are now playing an important role in mapping correlation phenomena, such as superconductivity and magnetism, which have recently been observed in two-dimensional van der Waals materials. Here, we demonstrate a scanning probe that combines the magnetic and thermal imaging provided by an on-tip SQUID with the tip-sample distance control and topographic contrast of a non-contact atomic force microscope (AFM). We pattern the nanometer-scale SQUID, including its weak-link Josephson junctions, via focused ion beam milling at the apex of a cantilever coated with Nb, yielding a sensor with an effective diameter of 365 nm, field sensitivity of 9.5 $\text{nT}/\sqrt{\text{Hz}}$ and thermal sensitivity of 620 $\text{nK}/\sqrt{\text{Hz}}$, operating in magnetic fields up to 1.0 T. Abstract: In response to the COVID-19 pandemic, governments have implemented a wide range of non-pharmaceutical interventions (NPIs). Monitoring and documenting government strategies during the COVID-19 crisis is crucial to understand the progression of the epidemic. Following a content analysis strategy of existing public information sources, we developed a specific hierarchical coding scheme for NPIs. We generated a comprehensive structured dataset of government interventions and their respective timelines of implementation. To improve transparency and motivate collaborative validation process, information sources are shared via an open library. We also provide codes that enable users to visualise the dataset. Standardization and structure of the dataset facilitate inter-country comparison and the assessment of the impacts of different NPI categories on the epidemic parameters, population health indicators, the economy, and human rights, among others. This dataset provides an in-depth insight of the government strategies and can be a valuable tool for developing relevant preparedness plans for pandemic. Abstract: Although black (Rattus rattus) and brown (Rattus norvegicus) rats are among the most widespread synanthropic wild rodents, there is a surprising scarcity of knowledge about their ecology in the urban ecosystem. In particular, relatively few studies have investigated their helminth species diversity in such habitat. We followed the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analysis) guideline to synthesize the existing published literature regarding the helminth fauna of urban rats in developed countries (North America, Europe, Australia, New Zealand and Japan). We aimed at describing the species diversity and richness of urban rat helminths, the species prevalence and associations, the methods of investigation, the pathological changes observed in the hosts, the risk factors of infection and the public health significance of rat-borne helminthiases. Twenty-three scientific papers published between 1946 and 2019 were reviewed, half of them were conducted in Europe. Twenty-five helminth species and eight genera were described from the liver, digestive tract, lungs and muscles of urban rats. The most commonly reported parasite was Calodium hepaticum. Prevalence and risk factors of helminth infection in urban rats varied greatly between studies. Observed pathological findings in the rat host were generally minor, except for C. hepaticum. Several rat helminths can parasitize humans and are therefore of public health significance. The lack of references to identification keys and the rare use of molecular tools for species confirmation represent the main limitation of these studies. Knowledge gap on this topic and the needs for future research are discussed. Abstract: The beauty production cross section for deep inelastic scattering events with at least one hard jet in the Breit frame together with a muon has been measured, for photon virtualities Q^2 > 2 GeV^2, with the ZEUS detector at HERA using integrated luminosity of 72 pb^-1. The total visible cross section is sigma_b-bbar (ep -> e jet mu X) = 40.9 +- 5.7 (stat.) +6.0 -4.4 (syst.) pb. The next-to-leading order QCD prediction lies about 2.5 standard deviations below the data. The differential cross sections are in general consistent with the NLO QCD predictions; however at low values of Q^2, Bjorken x, and muon transverse momentum, and high values of jet transverse energy and muon pseudorapidity, the prediction is about two standard deviations below the data., 18 pages, 4 figures

Abstract: A resonance search has been made in the K0s p and K0s pbar invariant-mass spectrum measured with the ZEUS detector at HERA using an integrated luminosity of 121 pb-1. The search was performed in the central rapidity region of inclusive deep inelastic scattering at an ep centre-of-mass energy of 300-318 GeV for exchanged photon virtuality, Q2, above 1 GeV2. Recent results from fixed-target experiments give evidence for a narrow baryon resonance decaying to K+ n and K0s p, interpreted as a pentaquark. The results presented here support the existence of such state, with a mass of 1521.5+/-1.5(stat.)^{+2.8}_{-1.7}(syst.) MeV and a Gaussian width consistent with the experimental resolution of 2 MeV. The signal is visible at high Q2 and, for Q2>20 GeV2, contains 221+/-48 events. The probability of a similar signal anywhere in the range 1500-1560 MeV arising from fluctuations of the background is below 6x10^{-5}., 4 eps figures, 15 pages, 1 table, accepted by Physics Letters B

Abstract: The production of D^*+, D^0, D^+, D_s^+ and Lambda_c^+ charm hadrons and their antiparticles in ep scattering at HERA was measured with the ZEUS detector using an integrated luminosity of 79 pb^-1. The measurement has been performed in the photoproduction regime with the exchanged-photon virtuality Q^2 < 1 GeV^2 and for photon-proton centre-of-mass energies in the range 130 < W < 300 GeV. The charm hadrons were reconstructed in the range of transverse momentum p_T(D, Lambda_c) > 3.8 GeV and pseudorapidity |eta(D, Lambda_c)| < 1.6. The production cross sections were used to determine the ratio of neutral and charged D-meson production rates, R_u/d, the strangeness-suppression factor, gamma_s, and the fraction of charged D mesons produced in a vector state, P_v^d. The measured R_u/d and gamma_s values agree with those obtained in deep inelastic scattering and in e^+e^- annihilations. The measured P_v^d value is smaller than, but consistent with, the previous measurements. The fractions of c quarks hadronising as a particular charm hadron, f(c -> D, Lambda_c), were derived in the given kinematic range. The measured open-charm fragmentation fractions are consistent with previous results, although the measured f(c -> D^*+) is smaller and f(c -> Lambda_c^+) is larger than those obtained in e^+e^- annihilations. Abstract: The ZEUS inclusive differential cross-section data from HERA, for charged and neutral current processes taken with e+ and e- beams, together with differential cross-section data on inclusive jet production in e+ p scattering and dijet production in \gamma p scattering, have been used in a new NLO QCD analysis to extract the parton distribution functions of the proton. The input of jet data constrains the gluon and allows an accurate extraction of \alpha_s(M_Z) at NLO; \alpha_s(M_Z) = 0.1183 \pm 0.0028(exp.) \pm 0.0008(model) An additional uncertainty from the choice of scales is estimated as \pm 0.005. Abstract: A search for lepton-flavor-violating interactions $e p \to \mu X $ and $e p\to \tau X $ has been performed with the ZEUS detector using the entire HERA I data sample, corresponding to an integrated luminosity of 130 pb^{-1}. The data were taken at center-of-mass energies, $\sqrt{s}$, of 300 and 318 GeV. No evidence of lepton-flavor violation was found, and constraints were derived on leptoquarks (LQs) that could mediate such interactions. For LQ masses below $\sqrt{s}$, limits were set on $\lambda_{eq_1} \sqrt{\beta_{\ell q}}$, where $\lambda_{eq_1}$ is the coupling of the LQ to an electron and a first-generation quark $q_1$, and $\beta_{\ell q}$ is the branching ratio of the LQ to the final-state lepton $\ell$ ($\mu$ or $\tau$) and a quark $q$. For LQ masses much larger than $\sqrt{s}$, limits were set on the four-fermion interaction term $\lambda_{e q_\alpha} \lambda_{\ell q_\beta} / M_{\mathrm{LQ}}^2$ for LQs that couple to an electron and a quark $q_\alpha$ and to a lepton $\ell$ and a quark $q_\beta$, where $\alpha$ and $\beta$ are quark generation indices. Some of the limits are also applicable to lepton-flavor-violating processes mediated by squarks in $R$-Parity-violating supersymmetric models. Abstract: Multijet production rates in neutral current deep inelastic scattering have been measured in the range of exchanged boson virtualities 10 < Q2 < 5000 GeV2. The data were taken at the ep collider HERA with centre-of-mass energy sqrt(s) = 318 GeV using the ZEUS detector and correspond to an integrated luminosity of 82.2 pb-1. Jets were identified in the Breit frame using the k_T cluster algorithm in the longitudinally invariant inclusive mode. Measurements of differential dijet and trijet cross sections are presented as functions of jet transverse energy E_{T,B}{jet}, pseudorapidity eta_{LAB}{jet} and Q2 with E_{T,B}{jet} > 5 GeV and -1 < eta_{LAB}{jet} < 2.5. Next-to-leading-order QCD calculations describe the data well. The value of the strong coupling constant alpha_s(M_Z), determined from the ratio of the trijet to dijet cross sections, is alpha_s(M_Z) = 0.1179 pm 0.0013(stat.) {+0.0028}_{-0.0046}(exp.) {+0.0064}_{-0.0046}(th.), 22 pages, 5 figures

17. 3TC+ PI dual therapy during pregnancy for PMTCT of HIV-1-naïve or pretreated women

Abstract: Purpose of the study : Perinatal transmission of HIV has fallen dramatically in countries where access to antiretrovirals is available, placing the focus of research on safety. The aim of this study is to assess whether in naive or pretreated HIV-1 women, a dual, zidovudine sparing regimen (3TC+ PI) initiated during pregnancy can control maternal viral load while preventing maternal and infant toxicities related to in utero exposure to zidovudine. Methods : We performed a retrospective, descriptive study between January 2006 and 2012. Seventeen naive and 28 pretreated women who received dual therapy (3TC+PI) during pregnancy were included in the studied group and compared to 49 women on standard triple therapy (3TC/ ZDV+ PI), who delivered in the same time interval, at the same hospital. The primary endpoint was the % of women with viral loads (VL) ≤ 50 cp/ml at delivery. Secondary endpoints included % of women with VL ≤400 cp/ml after one month of dual therapy and of women who remained under dual therapy until delivery and achieved a VL ≤50 cp/ml at delivery. We also compared safety outcomes during pregnancy and until 18 months in children. Summary of results : Groups had indistinguishable median VL at day zero (200 vs. 1680, p=0.29) and gestational ages at initiation of therapy (19.6 vs 18 weeks, p=0.18). At initiation of dual therapy (in 80% of cases: 3TC+LPV/r; ATV/r or ATV), the median VL was 21,692 cp/ml (6589 to 31,269) for naive women and 43 cp/ml (20 to 200) for pretreated women. In intent-to-treat analysis of the dual therapy group, 41/45 women (91.11%, 95% CI 78.7 to 97.5%) achieved their primary endpoint. The table presents proportions of viral success at primary and secondary endpoints in the dual (N=45) and control (N=49) groups. Groups had similar proportions of cesarean section (28.8% vs 34.7%, p=0.65) and of prematurity and changes in ART due to intolerance. There was one case of MTCT for the controls and none in the dual therapy group. Newborns with prenatal ZDV exposure had higher proportions of clinical syndromes at birth (50% vs 25%, p=0.02) and SAE in the first 18 months of life (90.3% vs 68.2%, p=0.05) and worse haematological values at birth, with lower levels of haemoglobin (15.6 vs 17.5 g/dl, p=0.05). Conclusions : The 3TC+ PI dual therapy strategy in naive or pretreated women achieved a satisfactory virologic effectiveness and resulted in less SAE in the first 18 months of life and better haematological parameters at birth for children. 18. In-situ combination of SEMPA, STM, and FIB for magnetic imaging and nanoscale structuring

Abstract: In the field of spintronics and its application in data storage and processing, research on nanomagnetism attains more and more attention. Therefore, we have constructed a versatile UHV instrumentation for the investigation of magnetism and magnetotransport phenomena on the nanometer scale, the nanospintronics cluster tool. The system consists of three interconnected vacuum chambers. The first chamber is a preparation unit containing 7 evaporators for thin film deposition, RHEED and LEED for structural analysis, and MOKE, AES and XPS to investigate magnetic and chemical properties of the films. The second chamber houses a low temperature scanning tunneling microscope (STM) that we plan to operate in a spin-resolved mode in order to image the magnetic structure on an atomic scale in a temperature range from 4 to 300 K. The third part of the UHV system combines a scanning electron microscope with polarization analysis (SEMPA) with a focused ion beam (FIB). Thus, it is possible to create objects on the nanometer scale and analyze the magnetic properties of their surfaces in-situ. The sample stage of the SEM can be cooled down to 30 K and features electrical contacts for in-situ four-point measurements and the investigation of spin-torque driven magnetization dynamics.

Abstract: Cross sections for e(+)p neutral current deep inelastic scattering have been measured at a center-of-mass energy of roots=318 GeV with the ZEUS detector at HERA using an integrated luminosity of 63.2 pb(-1). The double-differential cross section, d(2)sigma/dxdQ(2), is presented for 200 GeV2 lt Q(2) lt 30 000 GeV2 and for 0.005 lt x lt 0.65. The single- differential cross sections dsigma/dQ(2), dsigma/dx and dsigma/dy are presented for Q(2) gt 200 GeV2. The effect of Z- boson-exchange is seen in dsigma/dx measured for Q(2) gt 10 000 GeV2. The data presented here were combined with ZEUS e(+)p neutral current data taken at roots=300 GeV and the structure function F-2(em) was extracted. All results agree well with the predictions of the Standard Model.

Abstract: The photoproduction of bottom quarks in events with two jets and a muon has been measured with the ZEUS detector at HERA using an integrated luminosity of 110 ${\mathrm{pb}}^{\mathrm{\ensuremath{-}}1}$. The fraction of jets containing b quarks was extracted from the transverse momentum distribution of the muon relative to the closest jet. Differential cross sections for bottom production as a function of the transverse momentum and pseudorapidity of the muon, of the associated jet and of ${x}_{\ensuremath{\gamma}}^{\mathrm{jets}},$ the fraction of the photon's momentum participating in the hard process, are compared with MC models and QCD predictions made at next-to-leading order. The latter give a good description of the data.

Abstract: A search for events containing isolated tau leptons and large missing transverse momentum, not originating from the tau decay, has been performed with the ZEUS detector at the electron-proton collider HERA, using 130 pb^-1 of integrated luminosity. A search was made for isolated tracks coming from hadronic tau decays. Observables based on the internal jet structure were exploited to discriminate between tau decays and quark- or gluon-induced jets. Three tau candidates were found, while 0.40 +0.12 -0.13 were expected from Standard Model processes, such as charged current deep inelastic scattering and single W-boson production. To search for heavy-particle decays, a more restrictive selection was applied to isolate tau leptons produced together with a hadronic final state with high transverse momentum. Abstract: The subjet multiplicity has been measured in neutral current e+p interactions at Q**2 > 125 GeV**2 with the ZEUS detector at HERA using an integrated luminosity of 38.6 pb-1. Jets were identified in the laboratory frame using the longitudinally invariant K_T cluster algorithm. The number of jet-like substructures within jets, known as the subjet multiplicity, is defined as the number of clusters resolved in a jet by reapplying the jet algorithm at a smaller resolution scale y_cut. Measurements of the mean subjet multiplicity, < n_sbj >, for jets with transverse energies E_T,jet >15 GeV are presented. Next-to-leading-order perturbative QCD calculations describe the measurements well. The value of alpha_s(M_Z), determined from < n_sbj > at y_cut=10**-2 for jets with 25 < E_T,jet < 71 GeV, is alpha_s (M_Z) = 0.1187 +/- 0.0017 (stat.) +0.0024 / -0.0009 (syst.) +0.0093 / -0.0076 (th.)., Comment: 27 pages, 4 figures, 3 tables; submitted to Phys. 29. Subacute thyroiditis - Is it really linked to viral infection? Retrospective hospital patient registry study.

Abstract: Objective: Subacute thyroiditis (SAT) is a painful inflammatory disorder of the thyroid gland, which - after a phase of thyrotoxicosis - leads to transient, or less frequently permanent hypothyroidism. Apart from a strong association with specific HLA alleles, the causes are uncertain. Viral disease has been hypothesised as a trigger, with Enteroviruses, namely Echoviruses and Coxsackieviruses, showing a seasonal distribution that coincides with the incidence of SAT. In the first year of the COVID-19 pandemic, strict hygiene measures led to a sharp decline in infections and thus offered the opportunity to test this hypothesis., Methods: We analysed national registry data of hospitalised patients from Germany during the years 2015 to 2022 (Federal statistical Office (Destatis), Wiesbaden, Germany) and surveillance data on infectious diseases from the same years (clinical-virology.net and RKI). Statistical analysis includes modelling of seasonality by month, polynomial autoregression and Granger causality to assess dependency of future SAT frequencies from past ones, and association of virus incidence to SAT frequency, respectively., Results: Our study confirms previously described epidemiological findings with higher incidence in females and a seasonal peak in late summer coinciding with the seasonality of enteroviruses until 2019. In 2020, the pattern remained unchanged, except for the marked reduction of enteroviruses and other pathogens (except SARS-CoV-2) due to hygienic measures. Moreover, the SAT seasonality in the years 2021 and 2022 was apparently unaltered through the COVID-19 pandemic., Conclusions: Our study provides strong evidence that despite their seasonal pattern, Echoviruses and Coxsackieviruses are not the cause of SAT. Moreover, no other analysed virus (including Influenza A and B, Parainfluenza, Rhinovirus, Human Coronaviruses including SARS-CoV-2) showed any association., (© The Author(s) 2025. 30. HBV shows different levels of adaptation to HLA class I-associated selection pressure correlating with markers of replication.

Abstract: Background & Aims: Immune responses by CD8 T cells are essential for control of HBV replication. Although selection of escape mutations in CD8 T-cell epitopes has been previously described in HBV infection, its overall influence on HBV sequence diversity and correlation with markers of HBV replication remain unclear., Methods: Whole-genome sequencing was applied to HBV isolates from 532 patients with chronic HBV infection and high-resolution HLA class I genotyping. Using a Bayesian model (HAMdetector) for identification of HLA-associated mutational states (HAMs), the frequency and location of residues under CD8 T-cell selection pressure were determined and the levels of adaptation of individual isolates were quantified., Results: Using previously published thresholds for the identification of HAMs, a total of 295 residues showed evidence of CD8 T-cell escape, the majority of which were located in previously unidentified epitopes. Interestingly, HAMs were highly enriched in the HBV core protein compared to all other proteins. When individual HBV isolates were compared, different levels of adaptation to HLA class I immune pressure were noted. The level of adaptation increased with patient age and correlated with markers of replication, with low levels of adaptation in HBeAg-positive infection. Furthermore, the levels of adaptation negatively correlated with HBV viral load and HBsAg levels, consistent with high levels of HLA class I-associated selection pressure in patients with low replication levels., Conclusions: HBV sequence diversity is shaped by HLA class I-associated selection pressure with the HBV core protein being a predominant target of selection. Importantly, different levels of adaptation to immune pressure were observed between HBV infection stages, which need to be considered in the context of T-cell-based therapies., Impact and Implications: The immune response mediated by CD8 T cells plays a critical role in controlling HBV infection and shows promise for therapeutic strategies aimed at achieving a functional cure. This study demonstrates that mutational escape within CD8 T-cell epitopes is common in HBV and represents a key factor in the failure of immune control. Notably, the HBV core protein emerges as the primary target of CD8 T-cell selection pressure. 31. Trajectories and predictive significance of inflammatory parameters for clinical outcome in COVID-19 patients treated with tocilizumab.

Abstract: Purpose: The IL-6 receptor inhibitor tocilizumab reduces mortality and morbidity in severe cases of COVID-19 through its effects on hyperinflammation and was approved as adjuvant therapy. Since tocilizumab changes the levels of inflammatory markers, we aimed to describe these changes in patients treated with tocilizumab, analyse their value in predicting death and bacterial superinfection and determine their influence on mortality rates., Methods: A retrospective analysis of 76 patients who were treated with tocilizumab for severe COVID-19 in 2020 and 2021 was conducted. Inflammatory markers (IL-6, C-reactive protein (CRP), procalcitonin) were documented before and up to seven days after tocilizumab administration., Results: The overall mortality was 25% and 53.8% in patients who required invasive respiratory support. Deceased patients had higher baseline IL-6 (p = 0.026) and peak IL-6 levels after tocilizumab vs those who survived (p < 0.0001). A peak IL-6 value > 1000 pg/dl after tocilizumab administration was a good predictor of mortality (AUC = 0.812). Of the deceased patients 41.1% had a renewed CRP increase after an initial decrease following tocilizumab administration, compared to 7.1% of the surviving patients (p = 0.0011). Documented bacterial superinfections were observed in 35.5% (27/76) of patients, of whom 48.1% (13/27) died., Conclusion: CRP-decline and IL-6 increase after tocilizumab treatment occurs regularly. An increase of IL-6 levels exceeding tenfold of baseline IL-6 levels, an absolute peak of 1000 pg/ml or a renewed increase of CRP are associated with higher mortality. Suppressed CRP synthesis can impede the diagnosis of bacterial superinfections, thus increasing the risk for complications., (© 2024. The Author(s).)

Abstract: Purpose: Prolonged shedding of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been observed in immunocompromised hosts. Early monotherapy with direct-acting antivirals or monoclonal antibodies, as recommended by the international guidelines, does not prevent this with certainty. Dual therapies may therefore have a synergistic effect., Methods: This retrospective, multicentre study compared treatment strategies for corona virus disease-19 (COVID-19) with combinations of nirmatrelvir/ritonavir, remdesivir, molnupiravir, and/ or mABs during the Omicron surge. Co-primary endpoints were prolonged viral shedding (≥ 10 6 copies/ml at day 21 after treatment initiation) and days with SARS-CoV-2 viral load ≥ 10 6 copies/ml. Therapeutic strategies and risk groups were compared using odds ratios and Fisher's tests or Kaplan-Meier analysis and long-rank tests. Multivariable regression analysis was performed., Results: 144 patients were included with a median duration of SARS-CoV-2 viral load ≥ 10 6 copies/ml of 8.0 days (IQR 6.0-15.3). Underlying haematological malignancies (HM) (p = 0.03) and treatment initiation later than five days after diagnosis (p < 0.01) were significantly associated with longer viral shedding. Prolonged viral shedding was observed in 14.6% (n = 21/144), particularly in patients with underlying HM (OR 3.5; 95% CI 1.2-9.9; p = 0.02). Clinical courses of COVID-19 were mild to moderate with only few adverse effects potentially related to combination treatment., Conclusion: Early combination treatment of COVID-19 effectively prevented prolonged viral shedding in 85.6% of cases. Considering the rapid viral clearance rates and low toxicity, individualized dual therapy approaches may be beneficial in high-risk patients., (© 2023. The Author(s).)

In addition, we conducted a literature search on published cases of malaria-associated sHLH and systematically analyzed the literature regarding clinical and diagnostic criteria., Results: We obtained data from 13 centers treating 1461 malaria cases with different Plasmodium species, of which 5 patients (0.34%) also were diagnosed with sHLH. The literature search revealed detailed case reports from further 51 patients and case series comprising the description of further 24 patients with malaria-associated sHLH. Most cases (48/80; 60%) were reported from Asia. The median time interval between onset of malaria symptoms and hospital admission was 7 days. Severe complications of sHLH were documented in 36% (20/56) of patients, including two patients with multiple organ failure in our case series. Only 41% (23/56) of patients received specific treatment for sHLH, nevertheless the mortality rate (CFR) of 5% is lower compared to the CFR reported for sHLH triggered by other infectious diseases (e.g., 25% in sHLH due to EBV infection)., Conclusion: Malaria-associated sHLH appears to have a comparatively good prognosis but may still represent an underdiagnosed and potentially fatal complication of malaria, especially in resource-poor settings., (© 2023. 34. Dynamics of Virological and Clinical Response Parameters of Bulevirtide Treatment for Hepatitis D: Real-World Data.

Abstract: Background and Aims: The entry inhibitor bulevirtide represents the first specific treatment for hepatitis-D virus (HDV)-infected patients. In clinical trials, around 80% of patients achieve normalization of alanine aminotransferase (ALT) with about 60% virological response after 1 year, but little is known about the dynamics of responses and clinical predictors of treatment outcomes. We report our single-center data from 15 patients and describe response dynamics, clinical outcomes, and predictive factors for treatment response., Methods: Retrospective data from 15 patients have been analyzed at our department who started treatment with bulevirtide between 10/2020 and 08/2022. According to our standard procedures, laboratory parameters were controlled monthly; transient elastography was performed every 3 months, and the treatment duration was 12 months., Results: Treatment response rates after 1 year of treatment were similar to published data from clinical trials. ALT normalization usually occurs between months 2-6 of treatment, followed by a virological response after ≥6 months. Patients with more severe hepatitis at the start of treatment were less likely to respond in the first year of treatment. Loss of HDV-RNA was observed in one-third of patients after ≥1 year of treatment. Low body mass index and high alpha-fetoprotein at baseline were possible predictors of a delayed treatment response., Conclusion: Bulevirtide is a safe treatment option for HDV, leading to a fast hepatological response. Of note, decrease in transaminases precedes virological response. Patients with high viral load and ALT levels respond slower, but nonresponders (as classified by Food and Drug Administration criteria) still show a reduction in viremia. Abstract: Magnons, the quantum-mechanical fundamental excitations of magnetic solids, are bosons whose number does not need to be conserved in scattering processes. Microwave-induced parametric magnon processes, often called Suhl instabilities, have been believed to occur in magnetic thin films only, where quasi-continuous magnon bands exist. Here, we reveal the existence of such nonlinear magnon-magnon scattering processes and their coherence in ensembles of magnetic nanostructures known as artificial spin ice. We find that these systems exhibit effective scattering processes akin to those observed in continuous magnetic thin films. We utilize a combined microwave and microfocused Brillouin light scattering measurement approach to investigate the evolution of their modes. Scattering events occur between resonance frequencies that are determined by each nanomagnet's mode volume and profile. Comparison with numerical simulations reveals that frequency doubling is enabled by exciting a subset of nanomagnets that, in turn, act as nanosized antennas, an effect that is akin to scattering in continuous films. Abstract: Background: Monoclonal antibodies (mAbs) that target severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are predominantly less effective against Omicron variants. Immunocompromised patients often experience prolonged viral shedding, resulting in an increased risk of viral escape., Methods: In an observational, prospective cohort, 57 patients infected with Omicron variants who received sotrovimab alone or in combination with remdesivir were followed. The study end points were a decrease in SARS-CoV-2 RNA <106 copies/mL in nasopharyngeal swabs at day 21 and the emergence of escape mutations at days 7, 14, and 21 after sotrovimab administration. All SARS-CoV-2 samples were analyzed using whole-genome sequencing. Individual variants within the quasispecies were subsequently quantified and further characterized using a pseudovirus neutralization assay., Results: The majority of patients (43 of 57, 75.4%) were immunodeficient, predominantly due to immunosuppression after organ transplantation or hematologic malignancies. Infections by Omicron/BA.1 comprised 82.5%, while 17.5% were infected by Omicron/BA.2. Twenty-one days after sotrovimab administration, 12 of 43 (27.9%) immunodeficient patients had prolonged viral shedding compared with 1 of 14 (7.1%) immunocompetent patients (P = .011). Viral spike protein mutations, some specific for Omicron (e.g., P337S and/or E340D/V), emerged in 14 of 43 (32.6%) immunodeficient patients, substantially reducing sensitivity to sotrovimab in a pseudovirus neutralization assay. Combination therapy with remdesivir significantly reduced emergence of escape variants., Conclusions: Immunocompromised patients face a considerable risk of prolonged viral shedding and emergence of escape mutations after early therapy with sotrovimab. Abstract: Molecular testing of SARS-CoV-2 RNA is essential during the pandemic. Here, we compared the results of different respiratory specimens including anterior nasal swabs, pharyngeal swabs, saliva swabs, and gargle lavage samples to nasopharyngeal swabs on two automated SARS-CoV-2 test systems. Samples were collected and tested simultaneously from a total of 36 hospitalized symptomatic COVID-19 patients. Detection and quantification of SARS-CoV-2 was performed on cobas®6800 (Roche) and NeuMoDx™ (Qiagen) systems. Both assays showed reliable detection and quantification of SARS-CoV-2 RNA, with nasopharyngeal swabs showing the highest sensitivity. SARS-CoV-2 RNA concentrations in other respiratory specimens were lower (mean 2.5 log10 copies/ml) or even undetectable in up to 20%. These data clearly indicate that not all respiratory materials are equally suitable for the management of hospitalized patients, especially, in the late phase of COVID-19, when the viral phase subsides and inflammation becomes the predominant factor, making detection of even lower viral loads increasingly important., (Copyright © 2022 The Authors. Abstract: Establishing the optimal treatment for COVID-19 patients remains challenging. Specifically, immunocompromised and pre-diseased patients are at high risk for severe disease course and face limited therapeutic options. Convalescent plasma (CP) has been considered as therapeutic approach, but reliable data are lacking, especially for high-risk patients. We performed a retrospective analysis of 55 hospitalized COVID-19 patients from University Hospital Duesseldorf (UKD) at high risk for disease progression, in a substantial proportion due to immunosuppression from cancer, solid organ transplantation, autoimmune disease, dialysis. A matched-pairs analysis (1:4) was performed with 220 patients from the Lean European Open Survey on SARS-CoV-2-infected Patients (LEOSS) who were treated or not treated with CP. Both cohorts had high mortality (UKD 41.8%, LEOSS 34.1%). A matched-pairs analysis showed no significant effect on mortality. CP administration before the formation of pulmonary infiltrates showed the lowest mortality in both cohorts (10%), whereas mortality in the complicated phase was 27.8%. CP administration during the critical phase revealed the highest mortality: UKD 60.9%, LEOSS 48.3%. In our cohort of COVID-19 patients with severe comorbidities CP did not significantly reduce mortality in a retrospective matched-pairs analysis. 39. Prevalence of H. pylori among asymptomatic HIV-positive and negative individuals in Central Ethiopia and efficacy of eradication therapy.

Abstract: Objectives: Helicobacter pylori is a widespread pathogen and major contributor to dyspeptic disease and gastric cancer. Although the interaction between HIV and H. pylori infection is not well investigated, previous studies have suggested a decreased prevalence of H. pylori and limited efficacy of eradication therapy in HIV-positive individuals. Therefore, the objectives of this study were to describe the prevalence of H. pylori infection according to HIV status and analyze the efficacy of eradication therapy in Ethiopia., Methods: A prospective, randomized, interventional study was performed involving HIV-positive and negative participants presenting to the Asella Referral and Teaching Hospital in Central Ethiopia between March and June 2017. A stool antigen test was used as a screening tool for H. pylori infection . Randomly selected patients received triple eradication therapy., Results: The cumulative H. pylori prevalence was 77.3% (392/507): 78.8% (241/306) among HIV-positive individuals versus 75.1% (151/201) among HIV-negative individuals ( P = 0.386). Twenty-five HIV-positive and 26 HIV-negative H. pylori -infected participants were randomized to receive standard triple therapy; three of them were lost to follow-up (one HIV-positive, two HIV-negative). The total eradication rate was 50.0%: 62.5% (15/24) among those HIV-negative versus 37.5% (9/24) among those HIV-positive [Au?1]., Conclusions: A high prevalence of H. pylori was observed among HIV-positive and negative individuals in Central Ethiopia. Abstract: Purpose: Castleman's disease (CD) is a well-established entity but there is a lack of available data regarding the management and therapy of HIV- and HHV-8-positive multicentric CD (MCD). We provide our own single-center experience with HIV-associated MCD., Methods: We performed a retrospective, descriptive study on a cohort of patients with MCD, diagnosed and admitted to the infectious diseases or intensive care unit in the University Hospital Düsseldorf between 2008 and 2018. Included patients had a previous or new HIV diagnosis and clinical signs resembling MCD with evidence of HHV-8 replication or histological diagnosis for MCD., Results: Nine male patients were included in the study. All patients were treated with Rituximab after diagnosis of MCD, with six of them acquiring resolution of symptoms. Three patients received tocilizumab additionally. Other treatment options included: splenectomy (2/9), valganciclovir (2/9), vincristine and siltuximab (1/9), ruxolitinib and Cytosorb ® (2/9). The relapse rate was 44% (4/9) and the survival rate 87.5% after 1 year (8/9) and 71.4% after 3 years (5/7)., Conclusion: The most effective first-line therapy and retreatment option remains rituximab. The effectiveness of other treatment options like splenectomy or different immunotherapeutic approaches requires confirmation in larger-scale studies., (© 2021. 42. Spin-Wave Dynamics and Symmetry Breaking in an Artificial Spin Ice.

Abstract: Artificial spin ices are periodic arrangements of interacting nanomagnets which allow investigating emergent phenomena in the presence of geometric frustration. Recently, it has been shown that artificial spin ices can be used as building blocks for creating functional materials, such as magnonic crystals. We investigate the magnetization dynamics in a system exhibiting anisotropic magnetostatic interactions owing to locally broken structural inversion symmetry. We find a rich spin-wave spectrum and investigate its evolution in an external magnetic field. We determine the evolution of individual modes, from building blocks up to larger arrays, highlighting the role of symmetry breaking in defining the mode profiles. Moreover, we demonstrate that the mode spectra exhibit signatures of long-range interactions in the system. These results contribute to the understanding of magnetization dynamics in spin ices beyond the kagome and square ice geometries and are relevant for the realization of reconfigurable magnonic crystals based on spin ices.

43. Comparison of Three Cellular Assays to Predict the Course of CMV Infection in Liver Transplant Recipients.

Abstract: To estimate protection from cytomegalovirus (CMV) replication after solid organ transplantation, CMV serology has been considered insufficient and thus CMV immunity is increasingly assessed by cellular in vitro methods. We compared two commercially available IFN-γ ELISpot assays (T-Track CMV and T-SPOT .CMV ) and an IFN-γ ELISA (QuantiFERON-CMV). Currently, there is no study comparing these three assays. The assays were performed in 56 liver transplant recipients at the end of antiviral prophylaxis and one month thereafter. In CMV high- or intermediate-risk patients the two ELISpot assays showed significant correlation ( p < 0.0001, r > 0.6) but the correlation of the ELISpot assays with QuantiFERON-CMV was weaker. Results of both ELISpot assays were similarly predictive of protection from CMV-DNAemia ≥500 copies/mL [CMV pp65 T-SPOT. CMV at the end of prophylaxis: area under curve (AUC) = 0.744, cut-off 142 spot forming units (SFU), sensitivity set to 100%, specificity 46%; CMV IE-1 T-Track CMV at month 1: AUC = 0.762, cut-off 3.5 SFU, sensitivity set to 100%, specificity 59%]. The QuantiFERON-CMV assay was inferior, reaching a specificity of 23% when setting the sensitivity to 100%. In conclusion, both CMV-specific ELISpot assays appear suitable to assess protection from CMV infection/reactivation in liver transplant recipients.

44. Time-resolved imaging of three-dimensional nanoscale magnetization dynamics.

Abstract: Understanding and control of the dynamic response of magnetic materials with a three-dimensional magnetization distribution is important both fundamentally and for technological applications. From a fundamental point of view, the internal magnetic structure and dynamics in bulk materials still need to be mapped 1 , including the dynamic properties of topological structures such as vortices 2 , magnetic singularities 3 or skyrmion lattices 4 . From a technological point of view, the response of inductive materials to magnetic fields and spin-polarized currents is essential for magnetic sensors and data storage devices 5 . Here, we demonstrate time-resolved magnetic laminography, a pump-probe technique, which offers access to the temporal evolution of a three-dimensional magnetic microdisc with nanoscale resolution, and with a synchrotron-limited temporal resolution of 70 ps. We image the dynamic response to a 500 MHz magnetic field of the complex three-dimensional magnetization in a two-phase bulk magnet with a lateral spatial resolution of 50 nm. This is achieved with a stroboscopic measurement consisting of eight time steps evenly spaced over 2 ns. These measurements map the spatial transition between domain wall motion and the dynamics of a uniform magnetic domain that is attributed to variations in the magnetization state across the phase boundary. Our technique, which probes three-dimensional magnetic structures with temporal resolution, enables the experimental investigation of functionalities arising from dynamic phenomena in bulk and three-dimensional patterned nanomagnets 6 .

45. Stray-Field Imaging of a Chiral Artificial Spin Ice during Magnetization Reversal.

Abstract: Artificial spin ices are a class of metamaterials consisting of magnetostatically coupled nanomagnets. Their interactions give rise to emergent behavior, which has the potential to be harnessed for the creation of functional materials. Consequently, the ability to map the stray field of such systems can be decisive for gaining an understanding of their properties. Here, we use a scanning nanometer-scale superconducting quantum interference device (SQUID) to image the magnetic stray field distribution of an artificial spin ice system exhibiting structural chirality as a function of applied magnetic fields at 4.2 K. The images reveal that the magnetostatic interaction gives rise to a measurable bending of the magnetization at the edges of the nanomagnets. Micromagnetic simulations predict that, owing to the structural chirality of the system, this edge bending is asymmetric in the presence of an external field and gives rise to a preferred direction for the reversal of the magnetization. This effect is not captured by models assuming a uniform magnetization. Our technique thus provides a promising means for understanding the collective response of artificial spin ices and their interactions.

46. Unexpected field-induced dynamics in magnetostrictive microstructured elements under isotropic strain.

Abstract: We investigated the influence of an isotropic strain on the magnetization dynamics of microstructured magnetostrictive Co 40 Fe 40 B 20 (CoFeB) elements with time-resolved scanning transmission x-ray microscopy. We observed that the application of isotropic strain leads to changes in the behavior of the microstructured magnetostrictive elements that cannot be fully explained by the volume magnetostriction term. Therefore, our results prompt for an alternative explanation to the current models used for the interpretation of the influence of mechanical strain on the dynamical processes of magnetostrictive materials.

A cut-off of 19.5 spot forming units (SFU)/200,000 lymphocytes for the T-Track-CMV IE-1 (AUC = 0.802, sensitivity 45%, specificity 100%) and 495 SFU/200,000 lymphocytes for the T-Track-CMV pp65 (AUC = 0.617, sensitivity 11%, specificity 100%) was defined to assess protection against reactivation. The QuantiFERON-CMV performed modestly (AUC = 0.477, cut-off 85.1 IU/ml)., Conclusions: The QuantiFERON-CMV and T-Track-CMV enable the functional assessment of CMV-specific CMI in KTx recipients. 49. Emergent dynamic chirality in a thermally driven artificial spin ratchet.

Abstract: Modern nanofabrication techniques have opened the possibility to create novel functional materials, whose properties transcend those of their constituent elements. In particular, tuning the magnetostatic interactions in geometrically frustrated arrangements of nanoelements called artificial spin ice can lead to specific collective behaviour, including emergent magnetic monopoles, charge screening and transport, as well as magnonic response. Here, we demonstrate a spin-ice-based active material in which energy is converted into unidirectional dynamics. Using X-ray photoemission electron microscopy we show that the collective rotation of the average magnetization proceeds in a unique sense during thermal relaxation. Our simulations demonstrate that this emergent chiral behaviour is driven by the topology of the magnetostatic field at the edges of the nanomagnet array, resulting in an asymmetric energy landscape. In addition, a bias field can be used to modify the sense of rotation of the average magnetization. This opens the possibility of implementing a magnetic Brownian ratchet, which may find applications in novel nanoscale devices, such as magnetic nanomotors, actuators, sensors or memory cells.

Abstract: Geometrical frustration occurs when entities in a system, subject to given lattice constraints, are hindered to simultaneously minimize their local interactions. In magnetism, systems incorporating geometrical frustration are fascinating, as their behavior is not only hard to predict, but also leads to the emergence of exotic states of matter. Here, we provide a first look into an artificial frustrated system, the dipolar trident lattice, where the balance of competing interactions between nearest-neighbor magnetic moments can be directly controlled, thus allowing versatile tuning of geometrical frustration and manipulation of ground state configurations. Our findings not only provide the basis for future studies on the low-temperature physics of the dipolar trident lattice, but also demonstrate how this frustration-by-design concept can deliver magnetically frustrated metamaterials.Artificial magnetic nanostructures enable the study of competing frustrated interactions with more control over the system parameters than is possible in magnetic materials. Farhan et al. present a two-dimensional lattice geometry where the frustration can be controlled by tuning the unit cell parameters.
