1. Broadband metamaterial polarizers with high extinction ratio for high-precision terahertz spectroscopic polarimetry

Abstract

The demand for precise polarizers is increasing to investigate the polarization characteristics of materials non-invasively in the terahertz region. Recently, to address the low extinction ratio and fragile nature of conventional wire-grid polarizers, plasmonic structures and metasurfaces have been proposed. However, the challenge of achieving low transmittance compared to a high extinction ratio, along with the bulky structure due to a thick substrate, remains to be addressed. Here, we present high-efficiency broadband metamaterial polarizers consisting of cross-aligned double-layers of subwavelength metallic slit arrays, leveraging the extraordinary optical transmission and funneling effects. We obtained extinction ratios exceeding 70 dB over a broad frequency range, from 0.2 to 2.5 THz, reaching a maximum extinction ratio of ∼90 dB at 0.7 THz. To investigate the influence of high extinction ratio polarizers on actual measurement results, we measured a non-Hermitian metasurface with asymmetric polarization conversion and analyzed them using the Jones matrix formalism. The results confirmed that the extinction ratio of the polarizer has a significant impact on precise polarization-dependent measurements, especially on cross-polarization measurements. The enhanced performance of our polarizers offers significant potential for sensitive THz systems, paving the way for advancements in polarization analysis of emerging materials and chiral sensing.

Abstract

Abstract While the properties of the observed Higgs boson agree with the Standard Model predictions, the hierarchy of fermion masses lacks an explanation within the model. In this work, we consider a fresh approach to this problem, involving a different Higgs doublet responsible for each quark mass. We construct a model with a gauged, non-anomalous U(1) family symmetry that fixes which fermion couples to which doublet with an O $$ \mathcal{O} $$ (1) Yukawa coupling. The hierarchy of masses is generated by the hierarchy of vacuum expectation values of the Higgs fields. The model generically predicts a light, weakly coupled pseudoscalar. We verify that the model satisfies constraints from flavour changing neutral currents, Higgs phenomenology and electroweak precision tests.

Abstract

Contamination of surface waters with microbiological pollutants is a major concern to public health. Although long-term and high-frequency Escherichia coli (E. coli) monitoring can help prevent diseases from fecal pathogenic microorganisms, such monitoring is time-consuming and expensive. Process-driven models are an alternative means for estimating concentrations of fecal pathogens. However, process-based modeling still has limitations in improving the model accuracy because of the complexity of relationships among hydrological and environmental variables. With the rise of data availability and computation power, the use of data-driven models is increasing. In this study, we simulated fate and transport of E. coli in a 0.6 km2 tropical headwater catchment located in the Lao People's Democratic Republic (Lao PDR) using a deep-learning model and a process-based model. The deep learning model was built using the long short-term memory (LSTM) methodology, whereas the process-based model was constructed using the Hydrological Simulation Program–FORTRAN (HSPF). First, we calibrated both models for surface as well as for subsurface flow. Then, we simulated the E. coli transport with 6 min time steps with both the HSPF and LSTM models. The LSTM provided accurate results for surface and subsurface flow with 0.51 and 0.64 of the Nash–Sutcliffe efficiency (NSE) values, respectively. In contrast, the NSE values yielded by the HSPF were −0.7 and 0.59 for surface and subsurface flow. The simulated E. coli concentrations from LSTM provided the NSE of 0.35, whereas the HSPF gave an unacceptable performance with an NSE value of −3.01 due to the limitations of HSPF in capturing the dynamics of E. coli with land-use change. The simulated E. coli concentration showed the rise and drop patterns corresponding to annual changes in land use. This study showcases the application of deep-learning-based models as an efficient alternative to process-based models for E. coli fate and transport simulation at the catchment scale.

Abstract

The effect of additives on the densification behavior and mechanical properties of pure and additive (Zr, B and Mg)-added silica ceramics were investigated for their application to the matrix phase of a silica fiber reinforced silica (SiO2/SiO2f) composite. The additives affected the rate of densification and crystallization (or transformation) of the amorphous silica. Among the compositions, pure silica ceramics sintered at 900°C for 1 h showed the maximum flexural strength. Based on the results, SiO2/SiO2f was fabricated by a repeated vacuum-assisted infiltration method followed by the heat treatment at 900°C for 1 h. The relative density of the composite was 78.2% with a flexural strength of 22.4 MPa. Fractography revealed that the composite was damaged by strong bonding at the fiber/matrix interface and the fracture of fiber.

Abstract

Ocean surface represents roughly 70 % of the Earth's surface, playing a large role in the partitioning of the energy flow within the climate system. The ocean surface albedo (OSA) is an important parameter in this partitioning because it governs the amount of energy penetrating into the ocean or reflected towards space. The old OSA schemes in the ARPEGE-Climat and LMDZ models only resolve the latitudinal dependence in an ad hoc way without an accurate representation of the solar zenith angle dependence. Here, we propose a new interactive OSA scheme suited for Earth system models, which enables coupling between Earth system model components like surface ocean waves and marine biogeochemistry. This scheme resolves spectrally the various contributions of the surface for direct and diffuse solar radiation. The implementation of this scheme in two Earth system models leads to substantial improvements in simulated OSA. At the local scale, models using the interactive OSA scheme better replicate the day-to-day distribution of OSA derived from ground-based observations in contrast to old schemes. At global scale, the improved representation of OSA for diffuse radiation reduces model biases by up to 80 % over the tropical oceans, reducing annual-mean model–data error in surface upwelling shortwave radiation by up to 7 W m−2 over this domain. The spatial correlation coefficient between modeled and observed OSA at monthly resolution has been increased from 0.1 to 0.8. Despite its complexity, this interactive OSA scheme is computationally efficient for enabling precise OSA calculation without penalizing the elapsed model time.

Abstract

The body index [body mass, body mass index (BMI), waist circumference and waist-height ratio] and functional fitness (aerobic capacity, muscular strength, balance, flexibility and functional ability) of people with Down syndrome (DS) are substantially lower than those in the general population, as DS has an overall negative impact on health.This study explored the effect of a 24-week basketball training programme on the body composition and functional fitness of adults with DS.Twenty-two adults with DS (24 ± 6 years; 25 ± 4 kg/mHip circumference and waist-height ratio were statistically different at baseline between the intervention and control groups. After the 24-week basketball training programme, the results showed significant differences between the exercise and control groups for hip circumference, body mass, BMI, waist circumference, hip circumference, waist-height ratio, flexibility, aerobic capacity, static and dynamic balance, balance and basketball functional ability (P 0.05). The exercise group showed significant improvements in all variables of body composition and functional fitness (P 0.05) except for the waist-hip ratio. There was no statistical significance between all physical composition and functional health variables in the control group except for the standing long jump (P 0.05), with effect sizes ranging from small to large.The basketball training programme improved body composition and functional fitness of adults with DS.

Abstract

The thermo-mechanical response of shock-initiated energetic materials (EMs) is highly influenced by their microstructures, presenting an opportunity to engineer EM microstructures in a “materials-by-design” framework. However, the current design practice is limited, as a large ensemble of simulations is required to construct the complex EM structure-property-performance linkages. We present the physics-aware recurrent convolutional (PARC) neural network, a deep learning algorithm capable of learning the mesoscale thermo-mechanics of EM from a modest number of high-resolution direct numerical simulations (DNS). Validation results demonstrated that PARC could predict the themo-mechanical response of shocked EMs with comparable accuracy to DNS but with notably less computation time. The physics-awareness of PARC enhances its modeling capabilities and generalizability, especially when challenged in unseen prediction scenarios. We also demonstrate that visualizing the artificial neurons at PARC can shed light on important aspects of EM thermos-mechanics and provide an additional lens for conceptualizing EM.

Abstract

Dissolving microneedle patches have been extensively studied in the field of cosmetics comparison with topical cosmetics focusing on the delivery of active ingredients. Nevertheless, the skin improvement effect of hyaluronic acid, which is mainly used as a backbone material for dissolving microneedle, was not analyzed. In this study, adenosine encapsulated high and low molecular weight hyaluronic acid dissolving microneedle patch (Ad-HMN and Ad-LMN) were evaluated with respect to skin wrinkling, dermal density, elasticity, and safety in a clinical test on the crow's feet area.Clinical efficacy and safety tests were performed for 12 weeks on twenty three female subjects with wrinkles around their eyes. The Ad-HMN and Ad-LMN patch were applied once every 3 days, in the evening, for 8 weeks to the designated crow's feet area. Skin wrinkling, dermal density, and elasticity were measured by using PRIMOSBoth Ad-HMN and Ad-LMN groups showed statistically significant efficacy for almost all parameters. The Ad-HMN patch had better effect on the mean depth of biggest wrinkles, maximum depth of biggest wrinkles, dermal density, and skin elasticity than the Ad-LMN patch. No adverse effects were observed in either group during the test period.In the clinical efficacy test of four skin-improvement parameters, the Ad-HMN patch showed the better effect than the Ad-LMN patch with the similar adenosine dose.Les patches à micro-aiguilles dissolvantes ont fait l’objet d’études approfondies dans le domaine de la cosmétique en comparaison avec la cosmétique topique axée sur la diffusion de principes actifs. Cependant, l’effet améliorant de l’acide hyaluronique sur la peau, principalement utilisée comme matière de base pour la dissolution des micro-aiguilles, n’a pas été analysé. Dans la présente étude, les patchs à micro-éguilles dissolvant l’acide hyaluronique à poids moléculaire élevé et faible, encapsulé dans l’adénosine (Ad-HMN et Ad-LMN) ont été analysés par rapport à la formation des rides, la densité cutanée, l’élasticité et la sécurité d’emploi lors d’un test clinique sur la zone de pattes d’oie. MÉTHODES: Des tests d’efficacité et de sécurité cliniques ont été réalisés pendant 12 semaines sur 23 sujets féminins ayant des rides autour des yeux. Les patchs Ad-HMN et Ad-LMN ont été appliqués une fois tous les trois jours, le soir, pendant huit semaines, dans la zone à pattes d’oie désignée. La formation de rides, la densité cutanée et l’élasticité ont été mesurées à l’aide de PRIMOS® premium, Dermascan® C, Cutometer® MPA580 et Corneometer® CM825, respectivement. RÉSULTATS: Les deux groupes Ad-HMN et Ad-LMN ont présenté une efficacité statistiquement significative pour la quasi-totalité des paramètres. Le patch Ad-HMN a eu un meilleur effet sur la profondeur moyenne des rides les plus importantes, la profondeur maximale des rides les plus importantes, la densité cutanée et l’élasticité de la peau par rapport au patch Ad-LMN. Aucun effet indésirable n’a été observé dans aucun des groupes pendant la période d’essai.Lors du test d’efficacité clinique des quatre paramètres d’amélioration de la peau, le patch Ad-HMN a eu un meilleur effet que le patch Ad-LMN avec la dose d’adénosine similaire.

26. A broadband high-brightness quantum-dot double solid immersion lens single photon source

Abstract

High-brightness single photon sources (SPSs) are key components for practical quantum information processing systems. Although the performances of recently reported high-brightness SPSs are excellent, it remains challenging to match the emission wavelength of a quantum dot (QD) to the cavity since the high-Q cavity structures have narrow spectral bandwidths. Here, we propose a highly bright and broadband QD SPS that can be deterministically fabricated with a simple yet precise method. The optimized GaAs-polymer double solid immersion lens structure is capable of a brightness of 88% at 0.5 NA and has an operation band of 65 nm with a brightness of over 80% from numerical simulations. Experimentally, we achieved a brightness of 51.6% ± 2% and pure single photon emission [g(2)(0) = 0.029 ± 0.005] at saturation. We believe that our result can pave the way to a practical high-brightness QD SPS, considering its simple QD geometry together with its low cost and precise deterministic fabrication without using expensive and complicated e-beam lithography and dry etching processes.

28. Abstract TP194: Radiation And Stroke Death Risk In Diffuse Large B-cell Lymphoma Patients, A Population Based Study

Abstract

Objective: Radiation (RT) is associated with vascular changes, which carotid studies have linked with stroke risk. Previous studies have reported an increased risk of stroke following RT for squamous cancers of head & neck (HN), or Hodgkin’s lymphomas, though with variations in dose and anatomical radiation targets. Hence the effects of low dose radiation to only the HN remains unclear. We used a national database with long-term follow up to quantify the association of RT and stroke death (SD) in patients with diffuse large B cell lymphoma (DLBCL) of the HN. Materials/Methods: Patients 30 or older with DLBCL, stage I-II of a known primary site besides CNS or mediastinum from 1983-2013 were obtained for analysis from the SEER database. Cause of death coding was filtered for Cerebrovascular Disease. Chi square, T-test, and Kaplan-Meier analysis were performed with log-rank test and Cox accounting for age, sex, RT, and year of diagnosis. Results: 33945 patients met inclusion criteria with a median follow up of 44 months. SD occurred in 410 patients in median time 62 months. RT was given in 3855 of 8312 (46%) HN and 7686 of 25633 (30%) of non-HN patients. The 10 year-SD rate was 2.2 / 2.8 % (p=0.77) for HN receiving/ not receiving RT (noRT), and 1.8 / 2.1 % for non-HN patients (p=0.40). When combining both RT and noRT patients, HN patients did show a trend towards higher SD risk compared with non-HN patients, 2.5 vs 2.0%, (p=0.053), though this did not appear to be driven by RT. On univariate analysis of HN patients, there was no difference in the effects of RT on the SD rate in any subgroup. Multivariate analysis did not identify any increased risk of SD associated with RT, either (HR = 0.87, p=0.63). Conclusions: This study did not identify any increased risk of stroke death from RT in patients with early stage DLBCL of the head & neck, although there was a trend suggesting stroke rates may be higher in patients with disease located in head & neck compared to other sites regardless of whether they received RT. Higher chemotherapy doses for non-RT patients may be a relevant confounding variable. Confirmation with pooled analyses and meta-analysis may be useful.

Abstract

We propose a novel method to fabricate small apertures in a metal film using polymethyl methacrylate (PMMA). A PMMA/SiO2 double layer is employed to perform a negative tone lift-off with SiO2 as a sacrificial layer. Through this method, we succeeded in making sub-50 nm holes in a metal film using well-established individual fabrication steps. The proposed method will be more reliable and economical compared to the hydrogen silsesquioxane/PMMA bilayer method.

Abstract

Bipolar disorder is a heritable mental illness with complex etiology. We performed a genome-wide association study of 41,917 bipolar disorder cases and 371,549 controls of European ancestry, which identified 64 associated genomic loci. Bipolar disorder risk alleles were enriched in genes in synaptic signaling pathways and brain-expressed genes, particularly those with high specificity of expression in neurons of the prefrontal cortex and hippocampus. Significant signal enrichment was found in genes encoding targets of antipsychotics, calcium channel blockers, antiepileptics and anesthetics. Integrating expression quantitative trait locus data implicated 15 genes robustly linked to bipolar disorder via gene expression, encoding druggable targets such as HTR6, MCHR1, DCLK3 and FURIN. Analyses of bipolar disorder subtypes indicated high but imperfect genetic correlation between bipolar disorder type I and II and identified additional associated loci. Together, these results advance our understanding of the biological etiology of bipolar disorder, identify novel therapeutic leads and prioritize genes for functional follow-up studies. © 2021, The Author(s), under exclusive licence to Springer Nature America, Inc.

Abstract

In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide. Our ultimate goal is to encourage intellectual and technical innovation in the field.

Abstract

Objective The Paris System (TPS), which was recently introduced, emphasised the key features of malignant urine cytology: a high nuclear to cytoplasmic ratio, nuclear hyperchromasia, irregular nuclear membranes and coarse chromatin. Although other diagnostic features have been described, the diagnostic significance of such features and their application to TPS have not been fully defined for urinary tract washing specimens. Methods A total of 142 cases of urinary tract washing specimens with corresponding surgical pathology samples were examined for the key features of TPS and 13 previously described features. The diagnostic performance of TPS and our proposed modification of TPS was compared with that of the current system. Results In addition to the key features of TPS, in the present study, high-grade urothelial carcinoma (HGUC) frequently exhibited tumour diathesis, a ragged edge of urothelial cell groups, anisonucleosis, India ink nuclei, apoptotic bodies and pleomorphism. As anisonucleosis and India ink nuclei remained independent predictors of HGUC for the multivariate analysis, they were used to modify TPS. The reporting rate of the atypical urothelial cell (AUC) category decreased from 25.3% in the current system to 14.8% in TPS and 11.3% in our proposed modification of TPS. The sensitivity increased from 59.4% in the current system to 70.8% in TPS and 90.0% in this study. The diagnostic accuracy increased from 0.786 in the current system and 0.754 in TPS to 0.859 in this study. Conclusions TPS is a useful diagnostic system for urinary tract washing specimens by decreasing the number of AUC cases and increasing sensitivity. In this study, anisonucleosis and India ink nuclei improved the diagnostic accuracy of HGUC.

Abstract

Purpose The number of citations that an article has received reflects its impact on a particular research area. The aim of this study was to identify the 100 most-cited articles focused on ultrasound (US) imaging and to analyze the characteristics of these articles. Methods We determined the 100 most-cited articles on US imaging via the Web of Science database, using the search term. The following parameters were used to analyze the characteristics of the 100 most-cited articles: publication year, journal, journal impact factor, number of citations and annual citations, authors, department, institution, country, type of article, and topic. Results The number of citations for the 100 most-cited articles ranged from 1849 to 341 (median: 442.0) and the number of annual citations ranged from 108.0 to 8.1 (median: 22.1). The majority of articles were published in 1990 – 1999 (39 %), published in radiology journals (20 %), originated in the United States (45 %), were clinical observation studies (67 %), and dealt with the vessels (35 %). The Department of Internal Medicine at the University of California and the Research Institute of Public Health at the University of Kuopio (n = 4 each) were the leading institutions and Salonen JT and Salonen R (n = 4 each) were the most prolific authors. Conclusion Our study presents a detailed list and analysis of the 100 most-cited US articles, which provides a unique insight into the historical development in this field.

Abstract

A modified series combining transformer incorporating an autotransformer for the dual-mode operation to improve efficiency performance in the low-output power region is presented. The dual-mode CMOS power amplifier (PA) with the proposed reconfigurable transformer is fabricated using 40-nm CMOS technology. Compared with other dual-mode WLAN CMOS PAs, the proposed PA achieves high-efficiency performance in both high-power and low-power modes; in dual-mode operation, the PA has average output power levels of 19.2 and 15.2 dBm with power added efficiencies of 17.3 and 14.4%, respectively, while satisfying the error vector magnitude requirements of WLAN 802.11g signals.

Abstract

The integration of single quantum dots (QDs) into a planar Fabry–Perot microcavity has been established as a direct and viable approach to vertically steer photons emitted from the quantum emitters, resulting in a strong increase in the source brightness, which becomes particularly evident when a lens with a low numerical aperture is used. However, the spectral bandwidth of QD–microcavity structures is limited and determined by their intrinsic quality factor; these structures are, thus, not ideal for the extraction of entangled photon pairs or for studies of exciton dynamics. We have found that, when a deterministic low-index solid immersion lens is placed on top of the QD in a QD–microcavity structure, the structure exhibits an enhancement in the bandwidth to 27 nm and a source brightness of 23%. The solid immersion lens is deterministically fabricated via two-photon absorption and can be remade several times without perturbing the QD, ensuring that the QD's intrinsic properties are preserved and ensuring its long-term reliability.

Abstract

Background In contrast to interscalene block, there was little information regarding the analgesic efficacy of supraclavicular block for shoulder surgery. This study aimed to compare the analgesic efficacy and side effects of interscalene and supraclavicular blocks for shoulder surgery. Methods Patients scheduled for shoulder surgery were assigned to receive either ultrasound-guided interscalene (n = 25) or supraclavicular block (n = 24) with 20 ml of 0.375% ropivacaine. We assessed the duration of post-operative analgesia as a primary outcome and pain scores, supplemental analgesia, diaphragmatic excursion, motor block, fingertip numbness, side effects, and patient satisfaction as secondary outcomes. Results The duration of post-operative analgesia was not statistically different between groups: 868 (800–1440) min for supraclavicular block vs. 800 (731–922) min for interscalene block (median difference −85 min, 95% CI, −283 to 3 min, P = 0.095). The incidence of diaphragmatic paresis was significantly lower in the supraclavicular block group compared with that in the interscalene block group, both at 30 min after the block (66.7% vs. 92%, P = 0.021) and in the post-anaesthesia care unit (62.5% vs. 92%, P = 0.024). Motor block was higher in the supraclavicular block group in the post-anaesthesia care unit, however, not at 24 h. Other secondary outcomes were similar for both groups. Conclusions This study showed no statistically significant difference in the duration of post-operative analgesia between the supraclavicular and interscalene blocks. However, the supraclavicular block was associated with a lower incidence of diaphragmatic paresis compared with that of the interscalene block after shoulder surgery.

Abstract

Background Over two decades of observations in the field in South East Asia and Hawai‘i suggest that majority of the commercial dairy herds are of black hair coat. Hence a simple study to determine the accuracy of the observation was conducted with two large dairy herds in Hawaii in the mid-1990s. Methods A retrospective study on longevity of Holstein cattle in the tropics was conducted using DairyComp-305 lactation information coupled with phenotypic evaluation of hair coat color in two large dairy farms. Cows were classified into 3 groups: a) black (B, >90%); b) black/white (BW, 50:50) and c) white (W, >90%). Cows with other hair coat distribution were excluded from the study. In farm A, 211 out of 970 cows were identified having 4 or more lactations. In farm B, 690 out of 1,350 cows were identified with 2 or more lactations for the study. Results The regression analyses and the Wilcoxon-Log-rank test for survival probability showed that Holstein cattle with 90% black hair coat had greater longevity compared to Holstein cattle with 90% white hair coat. Conclusions This study suggests that longevity of Holstein cattle in tropical regions was influenced by hair coat color and characteristics.

Abstract

A TRACE/PARCS model has been developed to analyze anticipated transient without SCRAM (ATWS) events for a boiling water reactor (BWR) operating in the maximum extended load line limit analysis-plus (MELLLA+) expanded operating domain. The MELLLA+ domain expands allowable operation in the power/flow map of a BWR to low flow rates at high power conditions. Such operation exacerbates the likelihood of large amplitude power/flow oscillations during certain ATWS scenarios. The analysis shows that large amplitude power/flow oscillations, both core-wide and out-of-phase, arise following the establishment of natural circulation flow in the reactor pressure vessel (RPV) after the trip of the recirculation pumps and an increase in core inlet subcooling. The analysis also indicates a mechanism by which the fuel may experience heat-up that could result in localized fuel damage. TRACE predicts the heat-up to occur when the cladding surface temperature exceeds the minimum stable film boiling temperature after periodic cycles of dryout and rewet; and the fuel becomes “locked” into a film boiling regime. Further, the analysis demonstrates the effectiveness of the simulated manual operator actions to suppress the instability.

Abstract

Using conventional deposition techniques, we demonstrate a method to fabricate ultra-smooth 10 nm silver films without using a wetting layer or co-depositing another material. The argon working pressure plays a crucial role in achieving an excellent surface flatness for silver films deposited by DC magnetron sputtering on an InP substrate. The formation of ultra-smooth silver thin films is very sensitive to the argon pressure. At the optimum deposition condition, a uniform silver film with an rms surface roughness of 0.81 nm has been achieved.

48. First Report of Leaf Spot Caused by Cercospora fukushiana on New Guinea Impatiens in Korea

Abstract

New Guinea impatiens, Impatiens hawkeri W. Bull, is widely cultivated as a potted plant and garden plant. In July 2013, hundreds of young plants (cv. Fanfare) showing symptoms of leaf spot with approximately 50% incidence were found in polyethylene tunnels in Yongin City, Korea. Leaf spots were circular to oblong, reaching 6 mm or more in diameter. The spots were initially uniformly brown to reddish brown, turning gray with reddish brown margin. Diseased plants defoliated prematurely and were abandoned without marketing due to signs of discoloration and yellowing on leaves. A cercosporoid fungus was consistently observed in association with disease symptoms. Stromata were brown, small, and composed of a few swollen hyphal cells. Conidiophores were emerging through the cuticle, fasciculate (n = 2 to 20), olivaceous to brown, paler toward the apex, straight to mildly curved, geniculate, 30 to 260 μm long, 3.5 to 5 μm wide, 1- to 6-septate, and with conspicuous conidial scars. Conidia were hyaline and acicular. Smaller conidia were straight and longer conidia were mildly curved. Conidia were subacute to obtuse at the apex, truncate to obconically truncate at the base, 2- to 18-septate, 30 to 320 × 3.5 to 5.5 μm, and with thickened, darkened hila at the base. Morphological characteristics of the fungus were consistent with the previous reports of Cercospora fukushiana (Matsuura) W. Yamam. (1). Voucher specimens were housed in the Korea University herbarium (KUS). An isolate from KUS-F27438 was deposited in the Korean Agricultural Culture Collection (Accession No. KACC47640). Fungal DNA was extracted with DNeasy Plant Mini Kits (Qiagen Inc., Valencia, CA). The complete internal transcribed spacer (ITS) region of rDNA was amplified with the primers ITS1/ITS4 (4) and sequenced. The resulting sequence of 497 bp was deposited in GenBank (Accession No. KJ620981). This showed >99% similarity with sequence of C. fukushiana (EF600954) on I. balsamina from Korea. Isolate of KACC47640 was used in the pathogenicity tests. Hyphal suspensions were prepared by grinding 3-week-old colonies grown on PDA with distilled water using a mortar and pestle. Five plants were inoculated with hyphal suspensions and five plants were sprayed with sterile distilled water. The plants were covered with plastic bags to maintain a relative humidity of 100% for 24 h and then transferred to a 25 ± 2°C greenhouse with a 12-h photoperiod. Typical symptoms of necrotic spots appeared on the inoculated leaves 10 days after inoculation, and were identical to the symptoms observed in the field. C. fukushiana was re-isolated from symptomatic leaf tissues, confirming Koch's postulates. No symptoms were observed on water-inoculated control plants. Previously, leaf spots of Impatiens spp. associated with C. apii, C. balsaminae, and C. fukushiana have been reported (1,2,3). To our knowledge, this is the first report of C. fukushiana on I. hawkeri in Korea. Our observations in the nurseries of I. hawkeri suggest that low humidity with good ventilation as well as plant hygiene in greenhouses might be main strategies for preventing this disease. 50. A Dual Phosphodiesterase Inhibitor, Zardaverine (Type 3/4), Enhances Motility of Frozen-thawed Boar Sperm

Abstract

Cryopreserved semen is useful for animal breeding via artificial insemination (AI); however, the use of frozen-thawed boar sperm is limited due to cryodamage.The goal of this study was to improve post-thaw motility of boar semen by supplementing the thawing medium with a phosphodiesterase inhibitor, Zardaverine.Thawed boar semen samples were treated with different concentrations of Zardaverine (0, 20, 50, 75, 100 µM) and the motility was evaluated using a computer-assisted sperm analyser. Toxic effects (sperm viability, DNA integrity, mitochondrial activity) were examined by eosin-nigrosin staining, terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay and MitoTracker.Sperm motility values included curvilinear velocity, rectilinear speed, average value, linearity index, straightness index, and progressive motility. The kinetic values were significantly higher with the 50 uM Zardaverine supplementation compared to non-treated control. Furthermore, there were no toxic effects of the Zardaverine treatment.The dual phosphodiesterase inhibitor (type 3/4) Zardaverine significantly enhanced the motility of thawed spermatozoa without adverse effects.
