Research on the diacylglycerol acyltransferase 1 (DGAT1) K232A marker in cattle shows inconsistent results across regions, largely due to small sample sizes, limited genetic variation, and data restricted to few lactations, which complicates establishing a reliable genotype–phenotype correlation. This research aimed to determine the effect of the K232A polymorphism of the bovine DGAT1 gene on milk production and quality traits in dairy cattle. We used 1104 cattle, including 828 Holstein Friesian and 276 Jersey cows. The analysis utilized extensive data from six lactations of cows raised on four commercial dairy farms. We genotyped the population using the polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) technique and Sanger sequencing for verification. We then evaluated the 305 d and test-day milk yields as well as fat and protein yields and percentages. The number of inseminations per conception and calving ease were also assessed as reproduction indices. Genotype–phenotype associations were quantified using linear mixed models. The AA genotype was absent in Jersey cows, and the heterozygous genotype was predominant in both breeds. The K232A marker was significantly associated with test-day milk yield, fat, and protein content in Jersey cows. Further, it substantially affected the fat percentage of milk in Holstein Friesian cows (p<0.001). We found that the KK genotype is highly desirable for milk quality and especially fat content. This comprehensive assessment demonstrated that the KK genotype of the DGAT1 K232A polymorphism significantly influenced fat and protein contents in dairy cattle.

Leptin (LEP), alternatively recognized as the obesity gene, influences food consumption, energy balance, and lipid metabolism. Additionally, it plays a crucial role in energy metabolism, and variations in the LEP gene have been studied extensively among various livestock species. These investigations have unveiled correlations with traits such as meat quality, milk yield and composition, and growth characteristics. Nevertheless, the existing knowledge regarding its correlation with growth and reproductive traits in goats is comparatively limited, particularly when juxtaposed with studies of cattle. Hence, the objective of the current study was to investigate the relationship between polymorphisms in the intron 2 (Sau3AI) and exon 2 (HinfI) regions of the LEP gene and growth and reproductive traits in Saanen goats. The study comprised 185 Saanen goats in total. The PCR-RFLP (polymerase chain reaction restriction fragment length polymorphism) technique was employed to genotype these polymorphisms. Population genetic analysis included the number of effective alleles, heterozygosity, polymorphism information content, and compatibility with the Hardy–Weinberg equilibrium. The general linear model procedure's least-squares methods were used for the statistical analysis. The SNP (single-nucleotide polymorphism) located in exon 2 (LEP-HinfI) was monomorphic. Remarkably, the SNP located in intron 2 (LEP-Sau3AI) was associated with birth weight (p) and average daily weight gain (p). In this context, the AA genotype demonstrated higher birth weight and daily live weight gain means compared to other genotypes. No significant differences were observed in litter size, total weight gain, or morphometric measurements. The caprine LEP-Sau3AI polymorphism situated within intron 2 proved influential in traits critical for the profitability and sustainability of goat breeding. The findings of this study provide novel and valuable information for future research on the LEP gene in goats.

The relationship between the index values used to evaluate the genomic value and essential markers such as casein genes provides important information at the herd level. Therefore, this study aimed to determine the association between casein gene genotypes and the indices of genetic merit in Holstein Friesian cattle. A total of 805 cows were genotyped using the Affymetrix® Axiom® array system. We used data composed of the total performance index (TPI) and net merit (NM) values as well as the predicted transmitting abilities (PTAs) indices, including milk, fat, fat percentage, protein, protein percentage, combined fat and protein (CFP), productive life (PL), somatic cell score (SCS), daughter pregnancy rate (DPR), livability (LV), udder composite (UDC), and feet–legs composite (FLC) of each animal. The statistical analysis consisted of a one-way analysis of variance (ANOVA) test followed by Tukey's test. The A2A2 and the AB genotypes were predominant in the CSN2 and CSN3 genes, respectively. The A2A2 animals were found to have higher TPI and NM values. Moreover, they exhibited higher PTA values for proteins, CFP, and PL. On the other hand, the A1A1 genotype was significantly associated with the highest UDC. Concerning the CSN3 haplotypes, the BB animals had higher protein percentage and PL than alternative haplotypes. The AA and AB haplotypes were found to be significantly associated with the highest SCS and DPR values, respectively. In addition, the BE haplotype had the highest NM. Selection procedures focusing on casein genes in dairy cattle are becoming increasingly common worldwide, especially for A2 milk. However, herd-based dynamics are also fundamental to providing a desired genetic merit for the animals. This study may be valuable for further analyses regarding selection decisions using the breeding values of candidate animals in commercial dairy herds.

This study was made to determine the effects of genotype and housing system on physical quality characteristics of breast and leg meat of broilers under experimental conditions. The 150 slow-growing and 150 fast-growing 1 d old chicks were divided into three sub-groups with indoor raised slatted plastic floor, indoor concrete floor with rice hull litter, and free-range housing systems (2 genotype groups × 3 housing systems). All birds were offered the same diet and were housed in similar conditions until they were 56 d old. At slaughter, 10 birds from each main group were selected randomly to determine the quality characteristics of the meat. In total, 60 breast meat pieces (pectoralis major muscle) and 60 legs of the chickens were used for meat quality analysis including pH, shear force, and colour characteristics such as lightness (L∗), redness (a∗), yellowness (b∗), saturation (C∗), and hue angle (h∗). The pH of breast meat was significantly affected by genotype and housing system (P and P). There were significant genotype × housing system interactions for pH (P and P) and shear force values (P and P) of leg and breast meat. There were no significant effects of genotype and housing system on leg and breast meat colour properties except for effects of genotype on redness (a∗) of breast meat (p) and effects of housing on redness of leg meat colour (p). Slow-growing chickens and chickens housed in deep litter had a higher redness (darker) value of breast and leg meat colour compared to fast-growing birds and free range and slatted floor. In conclusion, it can be said that fast-growing broilers may be more appropriate for slatted plastic floor housing and slow-growing broilers may be more suitable for a free-range housing system, but further research on factors affecting meat quality would be very beneficial, especially in slow-growing broilers.

The objective of this study was to determine the association of single nucleotide polymorphisms (SNPs) in selected candidate genes with fattening performance traits in a commercial cattle herd. Fifteen SNPs in 12 candidate genes (LEP, FABP4, DGAT1, TG, IGF1, IGF1R, MYF5, LGB, CAPN1, CAST, GHR, and OLR1) were evaluated in 296 purebred Holstein–Friesian bulls using PCR-RFLP (polymerase chain reaction – restriction fragment length polymorphism). Associations between each segregating SNP and genetic merit for fattening performance were quantified using linear mixed models. Traits included in the study were fattening period, final weight, dry matter intake, feed conversion rate, and average daily weight gain. Apart from the general determination of the above-mentioned traits, each trait was evaluated based on the fattening periods between five selected target body weights (W1 = 100 kg, W2 = 200 kg, W3 = 300 kg, W4 = 400 kg, W5 = 450 kg). All markers with the exception of CAPN1 530, IGF1R, TG, and DGAT1 were associated with at least one of the traits. Furthermore, novel associations were observed for LEP × GHR, IGF1 × LEP, FABP4 3691 × FABP4 2834, and FAP4 3533 × LEP interactions. The results of this study confirm some previously reported associations. Moreover, novel associations have been identified, which may be incorporated into breeding programs to improve fattening performance.

The objective of this study was to determine the association of single nucleotide polymorphisms (SNPs) with carcass characteristics and meat quality traits in selected candidate genes in Holstein bulls. Five SNPs in four genes, i.e. calpain 1 (CAPN1), calpastatin (CAST), leptin (LEP) and growth hormone receptor (GHR), were genotyped in 400 purebred bulls using PCR-RFLP. Statistically significant associations were as follows: CAPN1 G316A with live weight, carcass weight, back fat thickness, m. longissimus thoracis et lumborum area and carcass measurements; CAPN1 V530I with pH and L∗; CAST S20T with live weight, inner chest depth and b∗ value; and GHR with ph, a∗ and h∗. In addition, significant genotypic interactions were observed for dressing percentage (LEP A80V × CAST S20T), pH (CAPN1 V530I × GHR S555G and LEP A80V × GHR S555G) and rump width (CAPN1 V530I × CAST S20T). There was no association between the LEP A80V marker and any of the traits evaluated, nor was there any association of the tested SNPs with chest width, C∗ and marbling score. The present results could therefore be indicative for future studies on meat yield and quality.

In this study we aimed to analyse the trauma cases admitted to hospital and died in the 3-month follow-up. Methods: The cases that admitted to various departments due to trauma in one year period and died in 3 month follow-up were analysed retrospectively. Demographics, causes of trauma, type of arrivals, arrival vital signs and admitted clinic data were saved. The data obtained were evaluated. Results: For one year period 551 cases were admitted to hospital and 35 of them (6.35%) were died in follow up period. The mean age was 54.38 (min:11–max:90) and male/female ratio was 1,7. The most common causes were fall (n = 14), firearm wounds (n = 7) and traffic accidents (n = 4). 15 (42.35%) cases were brought by ambulance. 16 (45.7%) cases were unstable at arrival and it is identified that 11 (68.75%) of them were died in early period. The most common admitted services were neurosurgery (n = 14), orthopaedics (n = 9) and intensive care unit (n = 8). It is determined that 7 (20%) cases were died in early period (1 week) and other 24 (68.5%) cases were died in first one month period. Discussion and conclusion: It is observed that early period mortality proportion was low in all trauma mortalities and more patients were died due to late complications. It can be considered as this result is an indirect indicator of the effectiveness of early resuscitation applications.

Pulmonary pseudocyst is an uncommon condition that may occur after blunt thoracic trauma. Traumatic pulmonary pseudocysts develop in less than 3% of patients after blunt thoracic trauma. In this study we share a “formation of a pulmonary pseudocyst following a blunt thoracic trauma” case. Case presentation: A 34-year-old male patient suffering from a motor vehicle accident, who had admitted to another medical unit, was referred to our ED for advanced treatment after the first intervention. Due to wide hemopneumothorax, bilateral tube thoracostomy had been performed when patient arrived. Thorax CT revealed a pulmonary pseudocyst in the right lung beside diffuse bilateral pulmonary contusions, hemopneumothorax and rib fracture. Following necessary medical treatments, patient was admitted to the intensive care unit. After three months with clinical and radiological follow-up, traumatic pulmonary pseudocyst in the right lung was fully healed. Discussion and Conclusion: Pulmonary pseudocysts are air cysts that may occur in the lung parenchyma after blunt thoracic trauma. Around 85% of cases reported in the literature involved young and male patients. Tears occurring in the lung parenchyma are one of the underlying mechanisms of pseudocyst formation. In these cases there is no need for further treatment of these cysts which are known to recede spontaneously unless no complication occurs.

Pelvic apophysis avulsion fractures in adolescent athletes are rare injuries and these fractures occur on the growth cartilage on the apophysis plates. Case presentation: A 17-year-old male patient admitted to ED suffering from sharp pain in the right hip and difficulty in walking after kicking ball severely during playing football. On physical examination, there was tenderness in the right thigh consistent with spina iliaca anterior superior (SIAS). Hip movements were within normal range except pain during movement. Neurovascular examination was within normal range. On Pelvic X-ray revealed a crescent shaped avulsion fracture of the right SIAS. The patient was treated conservatively and discharged with recommendations. Discussion and conclusion: The epiphyses and the apophyses are the weakest part of all the skeletal system. In these regions, avulsion fractures may occur with sudden and severe spasm of the muscles. Avulsion fractures of SIAS are mostly treated conservatively unless there is more than 2 cm fragment separated and non-union case in which case surgery is recommended. This type of injuries can be easily overlooked or misdiagnosed due to history of trauma free. It may also effects

The purpose of this study was to determine whether there is a correlation between CPAP usage and corneal thickness in patients with sleep disordered breathing. Full-night polysomnography (PSG) recordings were collected. Ten patients had undergone PSG recordings with continuous positive airway pressure (CPAP), and seven patients had undergone PSG recordings without CPAP. We measured corneal thickness by ultrasonic pachymeter before sleep and ten minutes after waking. We also measured visual acuity with a routine ophthalmologic eye chart before and after sleep. We asked patients to fill out a post-sleep questionnaire to get their subjective opinions. In the without-CPAP group, corneal thickness increased significantly during sleep in both eyes (left, p = 0.0025; right, p<0.0001). In the with-CPAP group, corneal thickness did not increase significantly (p>0.05 for both left and right cornea). There was no significant difference in visual acuity tests (p>0.05 for both left and right eye) between the two groups. According to our results, there is a significant increase in corneal thickness in the without-CPAP group. Our data show that a low percentage of Rapid Eye Movement (REM) sleep may cause an increase in corneal thickness, which can indicate poor corneal oxygenation. In fact, many sleep-disordered breathing (SDB) patients have low REM. Since a contact lens may cause low corneal oxygenation, SDB patients with contact lenses should be monitored carefully for their corneal thickness.

Background: In the present study, we want to demonstrate the correlation between obstructive sleep apnea syndrome (OSAS) whose independent effect on carotid artery intima-media thickness (IMT) was demonstrated, with Framingham risk score (FRS) showing the overall cardiovascular risk., Methods: IMT of the carotid artery was measured with ultrasonography and 10-year risk of coronary heart disease (CHD) was defined with FRS in 90 consecutive patients referred to our sleep clinic and who underwent polysomnography (PSG), with vascular risk factors and without a clinical atherosclerotic disease., Results: IMT and FRS were found to be statistically significantly increased in the severe OSAS group compared to the other two groups. Carotid IMT was found to be significantly positively correlated with, apnea-hypopnea index (AHI), oxygen desaturation index (ODI) and time duration with oxygen saturation (SpO2) <90%, and negatively correlated with minimum oxygen saturation at sleep (minimum SpO2) and mean SpO2. In control and mild OSAS group IMT and FRS have significantly positive correlation (r: 0.501, P: 0.027; r: 0.625, P<0.001), while in severe OSAS group no significant correlation was detected between IMT and FRS (r: 0.321, P: 0.06). In the regression analysis AHI and ODI were found to be an independent predictor of carotid IMT. ODI was found to have an independent effect on the progression of atherosclerosis., Conclusions: Increased carotid IMT in severe OSAS group could not be explained with the classical risk factors. In this respect, FRS might be insufficient to determine correctly the cardiovascular risk and protection strategies against the disease in OSAS patients.

YKL-40 (chitinase-3-like-1) has been introduced as a marker of inflammation in asthma. The aim of this study was to determine the role of YKL-40 in asthma and to evaluate the relationship between YKL-40 and asthma severity.In the study, 60 non-smoker asthma patients without additional diseases (aged between 20-60 years, female: 34) were grouped [Group I: Well controlled asthma patients (n: 30), Group II: Patients during acute exacerbation of asthma (n: 30)]. Healthy non-smoker female individuals were included in Group III (n: 30) as a control group. The level of serum YKL-40 of all groups were determined by ELISA. Also, serum YKL- 40 level was correlated with age, asthma duration in years, body mass index (BMI), forced expiratory volume in first second/ forced vital capacity (FEV1/FVC, %), FEV1 (%), and total IgE levels of asthma patients. Mean serum YKL-40 level was highest in patients during acute exacerbation of asthma (36.36±10.49 ng/ml) while mean serum YKL-40 level was the lowest (13.20±5.60 ng/ml) in the control group. There was a negative significant correlation between the serum YKL-40 levels and FEV1 (%) in patients during acute exacerbation of asthma. There were no significant correlations between the serum YKL-40 levels and other variables in group II.We found that increased serum YKL-40 levels may be used as a marker for evaluation of asthma severity and genetic polimorphism.
