Abstract

The Swedish micro-satellite Astrid-2, designed for studies in magnetosperic physics, was launched into orbit on 10 December 1998 from the Russian cosmodrome Plesetsk. It was injected into a circular orbit at 1000 km and at 83 degrees inclination. The satellite carried, among other instruments, a double Langmuir Probe instrument called LINDA (Langmuir INterferometer and Density instrument for Astrid-2). The scientific goals of this instrument, as well as the technical design and possible modes of operation, are described. LINDA consists of two lightweight deployable boom systems, each carrying a small spherical probe. With these probes, separated by 2.9 meters, and in combination with a high sampling rate, it was possible to discriminate temporal structures (waves) from spatial structures. An on-board memory made it possible to collect data also at times when there was no ground contact. Plasma density and electron temperature data from all magnetic latitudes and for all seasons have been collected.Key words. International audience; The Radio and Plasma Waves (RPW) instrument on the ESA Solar Orbiter mission is designed to measure in situ magnetic and electric fields and waves from the continuum up to several hundred kHz. The RPW also observes solar and heliospheric radio emissions up to 16 MHz. It was switched on and its antennae were successfully deployed two days after the launch of Solar Orbiter on February 10, 2020. Since then, the instrument has acquired enough data to make it possible to assess its performance and the electromagnetic disturbances it experiences. In this article, we assess its scientific performance and present the first RPW observations. In particular, we focus on a statistical analysis of the first observations of interplanetary dust by the instrument’s Thermal Noise Receiver. We also review the electro-magnetic disturbances that RPW suffers, especially those which potential users of the instrument data should be aware of before starting their research work.

Abstract

The Radio and Plasma Waves (RPW) instrument on the ESA Solar Orbiter mission is described in this paper. This instrument is designed to measure in-situ magnetic and electric fields and waves from the continuous to a few hundreds of kHz. RPW will also observe solar radio emissions up to 16 MHz. The RPW instrument is of primary importance to the Solar Orbiter mission and science requirements since it is essential to answer three of the four mission overarching science objectives. Abstract

This paper describes the design of MEFISTO (Mercury Electric Field In-Situ Tool) and WPT (Wire Probe Antenna) electric field sensors for Plasma Wave Investigation (PWI) on the BepiColombo Mio spacecraft (Mercury Magnetospheric Orbiter, MMO). The two sensors will enable the first observations of electric fields, plasma waves and radio waves in and around the Hermean magnetosphere and exosphere. MEFISTO and WPT are dipole antennas with 31.6 m tip-to-tip length. Each antenna element has a spherical probe at each end of the wire (15 m length). They are extended orthogonally in the spin plane of the spacecraft and enable measurements of the electric field in the frequency range of DC to 10 MHz by the connection to two sets of receivers, EWO for a lower frequency range and SORBET for higher frequencies. In the initial operations after the launch (20 Oct. 2018), we succeeded to confirm the health of both antennas and to release the launch lock of the WPT. After Mercury orbit insertion planned at the end of 2025, both sensors will be fully deployed and activate full operations of the PWI electric field measurements.

Abstract

Albino mice were injected intraperitoneally with Ehrlich ascites carcinoma. Experimental animals and intact controls were killed at 2–3-day-intervals for 16 days after injection. Samples were taken from the kidney for histochemical estimation of the noradrenaline content of perivascular sympathetic nerve fibres and for evalution of the juxtaglomerular granularity index. Noradrenaline was estimated biochemically in the kidney, heart and spleen. We found histochemical signs of noradrenaline depletion that was maximal about 7 days after injection and most pronounced in the smallest arterial blood vessels. Noradrenaline levels were normal 12 days after injection, but later were again subnormal. The biochemical assay showed a reversible 10 to 30 per cent noradrenaline depletion in the kidney and heart. Corresponding findings in the spleen were of doubtful validity because of a considerable increase in spleen weight. Different possible causes of these phenomena are discussed.

Abstract

The Swedish micro-satellite Astrid-2, designed for studies in magnetosperic physics, was launched into orbit on 10 December 1998 from the Russian cosmodrome Plesetsk. It was injected into a circular orbit at 1000 km and at 83 degrees inclination. The satellite carried, among other instruments, a double Langmuir Probe instrument called LINDA (Langmuir INterferometer and Density instrument for Astrid-2). The scientific goals of this instrument, as well as the technical design and possible modes of operation, are described. LINDA consists of two lightweight deployable boom systems, each carrying a small spherical probe. With these probes, separated by 2.9 meters, and in combination with a high sampling rate, it was possible to discriminate temporal structures (waves) from spatial structures. An on-board memory made it possible to collect data also at times when there was no ground contact. Plasma density and electron temperature data from all magnetic latitudes and for all seasons have been collected.Key words. Abstract

The Wave Experiment, F4, on the Swedish/German satellite Freja, is designed to measure the electric wave fields up to 4 MHz, the magnetic wave fields up to 16 kHz and the plasma density and its relative variations up to 2 kHz. Six wave signals and four density probe signals can be measured simultaneously. The wave forms of all signals are transmitted to ground without any analysis onboard. The limited TM allocation does not allow continuous sampling of the wave signals, so normally the measurements are made in snapshots of various lengths dependent on sampling frequency, etc. Continuous sampling can be made for shorter time periods by using a 6 Mbyte memory as a buffer.

To assess possible interactions between inflammation and activation of the anticoagulant protein C system during post-ischemic reperfusion protein C, APC, neutrophil L-selectin expression and myocardial myeloperoxidase activity (MPO) were measured in 19 patients undergoing cardiopulmonary bypass. After reperfusion for 10 min, APC to protein C ratio (APC/PC) increased from pre-reperfusion value 1.43 +/- 0.12 (mean +/- SEM) to 2.25 +/- 0.29, p = 0.015. Negative correlations were observed between APC/PC and MPO activity (Spearman r -0.64, p = 0.007) and APC/PC and neutrophil L-selectin expression (r = -0.7, p = 0.007, demonstrating that post-ishemic protein C activation was associated with decreased neutrophil tissue sequestration. Thus, physiological protein C activation may be involved in regulation of the inflammatory injury during reperfusion of human ischemic coronary circulation.

10. Transhepatic neutrophil and monocyte activation during clinical liver transplantation

During experimental liver transplantation, neutrophil sequestration results in increased oxygen free radical production and correlates inversely with graft viability. Neutrophil activation in clinical liver transplantation is poorly understood.We assessed leukocyte sequestration and transhepatic differences of neutrophil and monocyte CD11b expression, neutrophil free radical production, and plasma concentrations of interleukin 6 and interleukin 8 in nine patients during liver transplantation.Significant hepatic neutrophil sequestration occurred during initial graft rewarming with portal blood, after inferior vena cava declamping, and after hepatic artery declamping (all P0.05). A positive transhepatic difference (i.e., outcoming - ingoing) in CD11b expression of neutrophils was observed after portal vein declamping (51+/-32 relative fluorescence unit [RFU]) and in CD11b expression of monocytes during initial graft rewarming (67+/-86 RFU, both P0.05). A transcoronary increase in both unstimulated (74+/-80 RFU) and N-formyl-methionyl-leucylphenylalanine-stimulated (112+/-168 RFU) neutrophil free radical production took place after hepatic artery declamping (both P0.05). A negative transcoronary difference of interleukin 6 occurred during initial graft rewarming (-192+/-176 pg/ml) and a positive difference of interleukin 8 occurred after hepatic artery declamping (17+/-23 pg/ml, both P0.05).Hepatic sequestration and transhepatic activation of neutrophils, and hepatic production of interleukin 8 occur during clinical liver transplantation. A splanchnic influx of interleukin 6 occurs to the graft, possibly modulating neutrophil-mediated graft reperfusion injury.

12. Systemic inflammatory response syndrome without systemic inflammation in acutely ill patients admitted to hospital in a medical emergency

Abstract

Criteria of the systemic inflammatory response syndrome (SIRS) are known to include patients without systemic inflammation. Our aim was to explore additional markers of inflammation that would distinguish SIRS patients with systemic inflammation from patients without inflammation. The study included 100 acutely ill patients with SIRS. Peripheral blood neutrophil and monocyte CD11b expression, serum interleukin-6, interleukin-1beta, tumour necrosis factor-alpha and C-reactive protein were determined, and severity of inflammation was evaluated by systemic inflammation composite score based on CD11b expression, C-reactive protein and cytokine levels. Levels of CD11b expression, C-reactive protein and interleukin-6 were higher in sepsis patients than in SIRS patients who met two criteria (SIRS2 group) or three criteria of SIRS (SIRS3 group). The systemic inflammation composite score of SIRS2 patients (median 1.5; range 0-8, n=56) was lower than that of SIRS3 patients (3.5; range 0-9, n=14, P=0.013) and that of sepsis patients (5.0; range 3-10, n=19, P0.001). The systemic inflammation composite score was 0 in 13/94 patients. In 81 patients in whom systemic inflammation composite scores exceeded 1, interleukin-6 was increased in 64 (79.0%), C-reactive protein in 59 (72.8%) and CD11b in 50 (61.7%). None of these markers, when used alone, identified all patients but at least one marker was positive in each patient. A method employing CD45-gating of blast cells was evaluated for the flow cytometric detection of residual disease in CD19- and myeloid antigen-positive acute leukemia in morphologic remission. In the normal bone marrow, CD45-gating identified at least three populations of immature cells, one of which appeared to retain a minute fraction of CD19- and CD13/33-antigen co-expressing cells. In acute leukemia, CD45 expression separated the blast cell population from the normal marrow cell populations. In the majority of patients with CD19- and myeloid antigen-positive acute leukemia, subpopulations of blast cells with this mixed phenotype were detected during morphologic remission.

Abstract

Most approaches to demonstrating immunoglobulin heavy chain gene rearrangements are relatively laborious for routine follow-up of acute lymphoblastic leukemia (ALL). Here the use of a simple polymerase chain reaction (PCR) approach to monitor ALL disease activity has been validated. In the dilution experiments the method revealed a detection sensitivity 0.5% clonal cells in a background of 99.5% normal cells. To validate the immunoglobulin heavy chain gene PCH (IgH-PCR) in practice, we monitored the disease activity of 26 adult ALL patients showing a B-cell lineage component in immunophenotyping at the diagnosis of the disease. In 18 of those 26 patients, an IgH-PCR product could be demonstrated in the samples taken either at diagnosis or in relapse. These 18 patients were followed with a total of 158 consecutive samples by IgH-PCR. The mean follow-up time for the IgH-PCR-positive patients was 13.6 months (range 4 to 26 months). Eleven of these patients underwent altogether 18 relapses. In nine patients (81.8%), ten relapses (55.6%) could be predicted using the IgH-PCR approach. The mean time of IgH-PCR clonality detection, preceding a cytologic relapse, was 9.1 weeks (range 1.0 to 30.7 weeks). It seems that in three patients the predictive value of the IgH-PCR was remarkable, showing a repetitive positivity in spite of a cytologic remission, even one year prior to the relapse. We find that IgH-PCR provides a straightforward additional tool for monitoring B-cell lineage ALL. Due to the straightforward technical performance the method has low running costs and it is thus suitable for a routine service laboratory. Even if a negative finding in IgH-PCR does not rule out a forthcoming relapse in the patient, a positive finding is a definitive warning signal. All of the patients that showed an IgH-PCR clonality in the follow-up samples relapsed sooner or later.

Abstract

It is well documented that the calcitonin gene area in the short arm of chromosome 11 is hypermethylated in most acute leukemias as well as in chronic lymphatic leukemia. In contrast, the gene is normally methylated during the chronic phase of the chronic myeloid leukemia but turns hypermethylated as the disease escalates. As the methylation of the calcitonin gene correlates with the disease activity in chronic myeloid leukemia, it seemed worthwhile to study the gene methylation in other premalignant hematologic conditions with a potential to terminate in fulminant acute leukemia. We report here on the calcitonin gene methylation in patients with myelodysplastic syndromes (MDS) using a methylation sensitive restriction enzyme HpaII and standard Southern blotting techniques. Bone marrow aspirates from a total of 26 MDS patients were studied. In 24 of these patients, the calcitonin gene was hypermethylated. There was no correlation between the methylation status and the morphological stage of the disease. All six patients with a blast count5% had a hypermethylated gene. Of the 19 patients with a blast count5%, 17 were hypermethylated only two having normal methylation status of the gene. It appears that the hypermethylation of the calcitonin gene area in the short arm of chromosome 11 may be an early event in the pathogenesis of the myelodysplastic syndromes. The methylation analysis may thus be of value as a diagnostic tool in MDS but an abnormal methylation pattern does not seem to have a direct relation with the degree of blast infiltration.

The results show that the receptor-occupancy and receptor-inhibition relationship at the motor endplate is incompatible with a reserve of ‘spare receptors’ greater than about 20% of the total.

Abstract

Ultrasound guided fine-needle biopsy of mass lesions affecting the hepatobiliary tract in 67 patients is reported. A free-hand approach was used without a special biopsy transducer. All aspirates were sufficient and relevant. A correct cytologic diagnosis was reached in 91 per cent of the 57 malignant cases. In addition, malignancy was correctly suggested in 5 cases. In 33 per cent of the cases with unknown primary tumor, cytology determined the origin of the malignancy. No complications occurred.

Abstract

A family has previously been described in which four members with Pelger-Huët (P-H) anomaly suffered from recurrent attacks of abdominal pain and fever, while one member, whose polymorphonuclear leucocytes (PMNs) were also hyposegmented, was asymptomatic. We studied chemotaxis, chemokinesis and spontaneous locomotion of PMNs in the three surviving symptomatic sisters, in their asymptomatic brother and in two asymptomatic members of another family with P-H anomaly. The spontaneous migration of the PMNs of the three sisters was significantly slower both under agarose and in a membrane filter than that of the PMNs of the asymptomatic patients with P-H anomaly. Chemotactic and chemokinetic locomotion of the PMNs of the symptomatic sisters was also slow. Our results suggests that the impaired chemotaxis was due to a defect in the intrinsic locomotor capacity of PMNs rather than in their deformability or their responsiveness to the chemotactic stimulus.

Abstract

The effects of the depolarizing agent, batrachotoxin (BTX), and of various analogs were studied on rat phrenic nerve-diaphragm muscle preparations at 37 degrees C. The structural modifications of BTX included: 1) replacement of the 20alpha-pyrrole-3-carboxylate moiety; 2) alterations of substituents on the pyrrole moiety; 3) clevage of the 3alpha, 9alpha-hemiketal linkage; and 4) quaternization of the tertiary nitrogen of BTX. All of the compounds except batrachotoxinin A (BTX-A), which lacks the 20alpha-substituent, depolarized the postsynaptic membrane, transiently increased the frequency of spontaneous transmitter release to 400 to 600 sec- minus 1 and finally produced blockade of the directly and indirectly elicited muscle twitches. Of the compounds tested, only BTX-A potentiated the muscle twitches. The concentration which elicits a 50% depolarization of the muscle membrane in 1 hour was determined for all the compounds except for BTX-A and for dihydrobatrachotoxin which lacks the 3alpha, 9alpha-hemiketal linkage; these two analogs never depolarized the postsynaptic membrane by more than 10 to 15%. BTX, the 20alpha-2, 4, 5-trimethylpyrrole-3-carboxylate of BTX-A and the 20alpha-ester of BTX-A with 2-ethyl-4-methylpyrrole-3-carboxylic acid (homobatrachotoxin) were the three most potent toxins with doses of 4.5, 12 and 18 times 10- minus 9 M eliciting a 50% membrane depolarization in 1 hour. The quaternary derivative of BTX, the 20alpha-4, 5-dimethylpyrrole-3-carboxylate of BTX-A and 20alpha-2,4-dimethyl-5-acetylpyrrole-3-carboxylate of BTX-A were 24-, 65- and 110-fold less potent than BTX as depolarizing agents, whereas the 20alpha-p-bromobenzoate of BTX-A was 220-fold less potent. Each of these derivatives had the ability to increase sodium permeability since the increase in spontaneous miniature end-plate potential frequency and membrane depolarization were reversed by tetrodotoxin or by reducing the external sodium concentration. BTX was found to be more effective at alkaline pH (pH 9.0), at which it exists almost entirely in the un-ionized form, than at physiological or acidic pH(6.0). The results indicate that the analogs of BTX act by a mechanism similar to that of the parent compound, but that their potency differs and certain compounds may have a more selective action on either the pre- or postsynaptic membrane. For maximal depolarizing activity, a substituted pyrrole moiety is necessary at the 20alpha-position of BTX-A and 3alpha, 9alpha-hemiketal linkage must remain intact providing rigidity for the pentacyclic steroid nucleus.

Abstract

Membrane fluidity was studied by electron-spin-resonance techniques in human En(a-) erythrocytes that lack the major membrane sialoglycoprotein, glycophorin A. By using stearic acid spin labels with a doxyl group in the C-12 or C-15 positions, we demonstrated that the hydrophobic core in these cells was more fluid than in normal cells. Surface-located regions in isolated En(a-) membranes, when probed with stearic acid labelled in the C-5 position, appeared more stable than in normal membranes. In isolated En(a-) membranes, protein motion was decreased when probed with a nitroxide derivative of maleimide. After incubation with anti-(glycophorin A) antibodies protein motion and membrane fluidity were increased in normal membranes. This effect was observed also after spectrin depletion, which by itself increased protein motion but decreased membrane fluidity in the hydrophobic core of the membrane. The results show that membrane proteins influence the fluidity of membrane lipids.

1 The uptake of radiocalcium by nerve-ending particles isolated from rat brain was studied in vitro by means of a rapid lanthanum quenching technique. 2 The observed uptake fits a theoretical three-compartment model with two separate uptake phases, a fast, initial phase followed by a late, slow phase. This holds true during control conditions as well as during high-potassium stimulation. 3 The uptake as a function of the external calcium concentration can be described in terms of Michaelis-Menten kinetics during high-potassium stimulation. Under control conditions the fit is clearly applicable but statistically not as good as during potassium stimulation. 4 The affinity for the uptake of calcium remains unchanged under control conditions while during high-potassium stimulation the affinity drastically decreases during the late, slow phase of uptake. 5 During high-potassium stimulation the maximal velocity of calcium uptake is twice that during control conditions. This holds true for both the fast and the slow phases of the uptake. 6 Mg2+ has an inhibitory effect on the uptake, the inhibition being more effective during high-potassium stimulation. Tetrodotoxin has a slight inhibitory effect additional to that extended by Mg2+ during the initial phase of uptake into high potassium stimulated synaptosomes.

Abstract

The differential blocking effect of bupivacaine and 2-chloroprocaine was studied on isolated rabbit cervical sympathetic trunks (B and C fibres) and phrenic nerves (A fibres). The B fibres (myelinated, preganglionic) were more sensitive than C fibres (unmyelinated, postganglionic) to nerve block by bupivacaine and 2-chloroprocaine, as assessed by changes in action potential amplitude and latency. The blocking action of both bupivacaine and 2-chloroprocaine caused a decrease in amplitude in the B fibres approximately twice as great as that produced in the C fibres. In similar experimental conditions, the fastest conducting A fibres (myelinated, motor) were least affected. Bupivacaine 200 mumol litre-1 completely blocked all B fibres in 7 min and C fibres in 10 min. The average action potential amplitude of A fibres at that time was 78%. 2-Chloroprocaine 300 mumol litre-1 completely blocked B fibres in 4 min and C fibres in 15 min. At 15 min the average action potential amplitude of the A fibres was still approximately 35%. Although these two local anaesthetics differ structurally and physico-chemically, the rates of block of the different fibres were in the same order in vitro.

Abstract

Mice injected intraperitoneally with Ehrlich ascites carcinoma were killed after varying intervals 2 to 21 days after injection. Tissue specimens from the kidney, spleen and heart were examined histochemically for mono-amines in the adrenergic fibres and the noradrenaline content of the kidney was chemically determined. The mono-amine-indicating fluorescense was subjectively estimated as absent or faint in a greater number of slides, and correspondingly as clear in a smaller number of slides in the ascitic group than in the controls, the difference being highly significant. This applied to the adrenergic fibres of the blood vessels of the kidney, to the arterioles of the spleen and to the adrenergic fibres of the myocardium. A highly significant reduction in the chemically determined content of noradrenaline was found in the kidney of ascitic mice as compared with the controls.

Publisher Summary The 5-hydroxytryptamine (5-HT) is stored in the mast cells, as well as in the nervous system in intracellular storage granules. There are, however, great differences between the mast cell granules and the 5-HT-storing synaptic vesicles. The mast cell granule has a diameter of some 500 nm, while the small granular (dense-cored) vesicles are about 30–60 nm. Vesicles with a diameter up to 200 nm were reported after homogenization isolated 5-HT-storing vesicles with a diameter of 50–130 nm. Normal mast cells handle the amine in a similar way—that is, at low concentrations of exogenous 5-HT, the amine is taken up by an active mechanism, while at high concentrations of 5-HT, the amine is incorporated by a passive mechanism. Mast cells contain large amounts of histamine and, in rat and mouse, 5-HT. Because of the high endogenous content of 5-HT and the ease with which the cells can be handled, the uptake and storage of 5-HT in normal mast cells show that the mast cell serves as a useful comparative model for the adrenergic neurons.
