Activation of cannabinoid 1 receptors (CB1 R) modulates multiple behaviours, including exploration, motor coordination and response to psychostimulants. It is known that CB1 R expressed by either excitatory or inhibitory neurons mediates different behavioural responses to CB1 R activation, yet the involvement of CB1 R expressed by medium spiny neurons (MSNs), the neuronal subpopulation that expresses the highest level of CB1 R in the CNS, remains unknown. We report a new genetically modified mouse line that expresses functional CB1 R in MSN on a CB1 R knockout (KO) background (CB1 R(MSN) mice). The absence of cannabimimetic responses measured in CB1 R KO mice was not rescued in CB1 R(MSN) mice, nor was decreased spontaneous locomotion, impaired instrumental behaviour or reduced amphetamine-triggered hyperlocomotion measured in CB1 R KO mice. Significantly, reduced novel environment exploration of an open field and absence of amphetamine sensitization (AS) measured in CB1 R KO mice were fully rescued in CB1 R(MSN) mice. Impaired motor coordination in CB1 R KO mice measured on the Rotarod was partially rescued in CB1 R(MSN) mice. Thus, CB1 R expressed by MSN control exploration, motor coordination, and AS. Our study demonstrates a new functional roles for cell specific CB1 R expression and their causal link in the control of specific behaviors.

Background: Parkinsonism is caused by dopamine (DA) insufficiency and results in a hypokinetic movement disorder. Treatment with L-Dopa can restore DA availability and improve motor function, but patients can develop L-Dopa-induced dyskinesia (LID), a secondary hyperkinetic movement disorder. The mechanism underlying LID remains unknown, and new treatments are needed. Experiments in mice have shown that DA deficiency promotes an imbalance between striatal acetylcholine (ACh) and DA that contributes to motor dysfunction. While treatment with L-Dopa improves DA availability, it promotes a paradoxical rise in striatal ACh and a further increase in the ACh to DA ratio may promote LID. Methods: We used conditional Slc6a3DTR/+ mice to model progressive DA deficiency and the β-adrenergic receptor (β-AR) antagonist propranolol to limit the activity of striatal cholinergic interneurons (ChIs). DA-deficient mice were treated with L-Dopa and the dopa decarboxylase inhibitor benserazide. LID and motor performance were assessed by rotarod, balance beam, and open field testing. Electrophysiological experiments characterized the effects of β-AR ligands on striatal ChIs. Results: LID was observed in a subset of DA-deficient mice. Treatment with propranolol relieved LID and motor hyperactivity. Electrophysiological experiments showed that β-ARs can effectively modulate ChI firing. Conclusions: The work suggests that pharmacological modulation of ChIs by β-ARs might provide a therapeutic option for managing LID.

Summary Many learned responses depend on the coordinated activation and inhibition of synaptic pathways in the striatum. Local dopamine neurotransmission acts in concert with a variety of neurotransmitters to regulate cortical, thalamic, and limbic excitatory inputs to drive the direct and indirect striatal spiny projection neuron outputs that determine the activity, sequence, and timing of learned behaviors. We review recent advances in the characterization of stereotyped neuronal and operant responses that predict and then obtain rewards. These depend on the local release of dopamine at discrete times during behavioral sequences, which, acting with glutamate, provides a presynaptic filter to select which excitatory synapses are inhibited and which signals pass to indirect pathway circuits. This is followed by dopamine-dependent activation of specific direct pathway circuits to procure a reward. These steps may provide a means by which higher organisms learn behaviors in response to feedback from the environment.

Introduction Surgical site infection (SSI) is one of the most common healthcare-associated infections (HAIs). However, there is a lack of data available about SSI in children worldwide, especially from low-income and middle-income countries. This study aimed to estimate the incidence of SSI in children and associations between SSI and morbidity across human development settings. Methods A multicentre, international, prospective, validated cohort study of children aged under 16 years undergoing clean-contaminated, contaminated or dirty gastrointestinal surgery. Any hospital in the world providing paediatric surgery was eligible to contribute data between January and July 2016. The primary outcome was the incidence of SSI by 30 days. Relationships between explanatory variables and SSI were examined using multilevel logistic regression. Countries were stratified into high development, middle development and low development groups using the United Nations Human Development Index (HDI). Results Of 1159 children across 181 hospitals in 51 countries, 523 (45·1%) children were from high HDI, 397 (34·2%) from middle HDI and 239 (20·6%) from low HDI countries. The 30-day SSI rate was 6.3% (33/523) in high HDI, 12·8% (51/397) in middle HDI and 24·7% (59/239) in low HDI countries. SSI was associated with higher incidence of 30-day mortality, intervention, organ-space infection and other HAIs, with the highest rates seen in low HDI countries. Median length of stay in patients who had an SSI was longer (7.0 days), compared with 3.0 days in patients who did not have an SSI. Use of laparoscopy was associated with significantly lower SSI rates, even after accounting for HDI. Conclusion The odds of SSI in children is nearly four times greater in low HDI compared with high HDI countries. Policies to reduce SSI should be prioritised as part of the wider global agenda. ©

Summary Motor and cognitive functions depend on the coordinated interactions between dopamine (DA) and acetylcholine (ACh) at striatal synapses. Increased ACh availability was assumed to accompany DA deficiency based on the outcome of pharmacological treatments and measurements in animals that were critically depleted of DA. Using Slc6a3DTR/+ diphtheria-toxin-sensitive mice, we demonstrate that a progressive and L-dopa-responsive DA deficiency reduces ACh availability and the transcription of hyperpolarization-activated cation (HCN) channels that encode the spike timing of ACh-releasing tonically active striatal interneurons (ChIs). Although the production and release of ACh and DA are reduced, the preponderance of ACh over DA contributes to the motor deficit. The increase in striatal ACh relative to DA is heightened via D1-type DA receptors that activate ChIs in response to DA release from residual axons. These results suggest that stabilizing the expression of HCN channels may improve ACh-DA reciprocity and motor function in Parkinson’s disease (PD). Video Abstract Download : Download video (19MB)

The diagnosis and management of movement disorders in children can be improved by understanding the pathways, neurons, ion channels, and receptors involved in motor learning and control. In this review, we use a localization approach to examine the anatomy, physiology, and circuitry of the basal ganglia and highlight the mechanisms that underlie some of the major movement disorders seen in children. We begin by reviewing the connections between the basal ganglia and the thalamus and cortex. We address the basic clinical definitions of movement disorders and then place diseases within an anatomic/physiologic framework that highlights our current understanding of basal ganglia function. We discuss how new pharmacological, behavioral, and electrophysiological approaches may benefit the child by modifying synaptic function. A better understanding of the mechanisms underlying movement disorders allows improved diagnostic and treatment decisions.

Small fluctuations in striatal glutamate and dopamine are required to establish goal-directed behaviors and motor learning, while large changes appear to underlie many neuropsychological disorders, including drug dependence and Parkinson’s disease. A better understanding of how variations in neurotransmitter availability can modify striatal circuitry will lead to new therapeutic targets for these disorders. Here, we examined dopamine-induced plasticity in prefrontal cortical projections to the nucleus accumbens core. We combined behavioral measures of male mice, presynaptic optical studies of glutamate release kinetics from prefrontal cortical projections, and postsynaptic electrophysiological recordings of spiny projection neurons within the nucleus accumbens core. Our data show that repeated amphetamine promotes long-lasting but reversible changes along the corticoaccumbal pathway. In saline-treated mice, coincident cortical stimulation and dopamine release promoted presynaptic filtering by depressing exocytosis from glutamatergic boutons with a low-probability of release. The repeated use of amphetamine caused a frequency-dependent, progressive, and long-lasting depression in corticoaccumbal activity during withdrawal. This chronic presynaptic depression was relieved by a drug challenge which potentiated glutamate release from synapses with a low-probability of release. D1 receptors generated this synaptic potentiation, which corresponded with the degree of locomotor sensitization in individual mice. By reversing the synaptic depression, drug reinstatement may promote allostasis by returning corticoaccumbal activity to a more stable and normalized state. Therefore, dopamine-induced synaptic filtering of excitatory signals entering the nucleus accumbens core in novice mice and paradoxical excitation of the corticoaccumbal pathway during drug reinstatement may encode motor learning, habit formation, and dependence.

The striatum is a critical component of the brain that controls motor, reward, and executive function. This ancient and phylogenetically conserved structure forms a central hub where rapid instinctive, reflexive movements and behaviors in response to sensory stimulation or the retrieval of emotional memory intersect with slower planned motor movements and rational behaviors. This review emphasizes two distinct pathways that begin in the thalamus and converge in the striatum to differentially affect movements, behaviors, and decision making. The convergence of excitatory glutamatergic activity from the thalamus and cortex, along with dopamine release in response to novel stimulation, provide the basis for motor learning, reward seeking, and habit formation. We outline how the rules derived through research on neural pathways may enhance the predictability of reflexive actions and rational responses studied in behavioral economics.

Parkinson’s disease (PD) is primarily associated with the degeneration of midbrain dopamine neurons, but it is now appreciated that pathological processes like Lewy-body inclusions and cell loss affect several other brain regions, including the central lateral (CL) and centromedian/parafascicular (CM/PF) thalamic regions. These thalamic glutamatergic neurons provide a non-cortical excitatory input to the dorsal striatum, a major projection field of dopamine neurons. To determine how thalamostriatal signaling may contribute to cognitive and motor abnormalities found in PD, we used a viral vector approach to generate mice with loss of thalamostriatal glutamate signaling specifically restricted to the dorsal striatum (CAV2Cre-Slc17a6lox/lox mice). We measured motor function and behaviors corresponding to cognitive domains (visuospatial function, attention, executive function, and working memory) affected in PD. CAV2Cre-Slc17a6lox/lox mice were impaired in motor coordination tasks such as the rotarod and beam-walk tests compared with controls (CAV2Cre-Slc17a6+/+ mice). They did not demonstrate much cognitive impairment in the Morris water maze or a water U-maze, but had slower processing reaction times in those tests and in a two-way active avoidance task. These mice could model an aspect of bradyphrenia, the slowness of thought that is often seen in patients with PD and other neurological disorders., Brain connectivity: Interrupting cognitive processing speed Mice in which glutamate signaling from the thalamus to dorsal striatum has been genetically inactivated mimic the slowness of thought that is often observed in patients with Parkinson’s disease (PD). The midbrain and striatum are the brain regions that are most affected in PD, however, it is increasingly recognized that cell loss in other areas of the brain also contribute to disease symptoms. Martin Darvas at the University of Washington, Seattle, USA, and colleagues found that disrupting the excitatory input from thalamic projection neurons into the dorsal striatum affected motor coordination and balance in mice. Although these mice did not have significant impairments in spatial learning and memory, they were slower at reacting to cues and executing learned behaviors suggesting that they could be used to test new approaches for treating this specific cognitive symptom of PD.

Modulation of corticostriatal synaptic activity by dopamine is required for normal sensorimotor behaviors. After loss of nigrostriatal dopamine axons in Parkinson's disease, l-3,4-dihydroxyphenlalanine (l-DOPA) and dopamine D2-like receptor agonists are used as replacement therapy, although these drugs also trigger sensitized sensorimotor responses including dyskinesias and impulse control disorders. In mice, we lesioned dopamine projections to the left dorsal striatum and assayed unilateral sensorimotor deficits with the corridor test as well as presynaptic corticostriatal activity with the synaptic vesicle probe, FM1-43. Sham-lesioned mice acquired food equivalently on both sides, while D2 receptor activation filtered the less active corticostriatal terminals, a response that required coincident co-activation of mGlu-R5 metabotropic glutamate and CB1 endocannabinoid receptors. Lesioned mice did not acquire food from their right, but overused that side following treatment with l-DOPA. Synaptic filtering on the lesioned side was abolished by either l-DOPA or a D2 receptor agonist, but when combined with a CB1 receptor antagonist, l-DOPA or D2 agonists normalized both synaptic filtering and behavior. Thus, high-pass filtering of corticostriatal synapses by the coordinated activation of D2, mGlu-R5, and CB1 receptors is required for normal sensorimotor response to environmental cues.

Brain copper accumulation commonly causes neurological symptoms in Wilson disease (WD). Many of the neurological manifestations that are present in WD involve movement disorders such as tremor, dysarthria, parkinsonism, dystonia, and gait impairment. Current de-coppering therapy provides in most cases only partial improvement of the neurological aspects of WD, and the use of medications to treat residual neurological symptoms is commonly required to improve patients’ quality of life. The evidence assessing the efficacy of different therapeutic options for movement disorders in WD is limited. This chapter aims to review the most common neurological manifestations in WD and the therapeutic options currently available.

Growing evidence points to the influence of social relationships on morbidity and mortality outcomes in later life. With the quantity and quality of individuals’ social relationships increasingly a concern for public health, this paper explores the scale and nature of inequalities that characterise the social relationships of people aged 50 and over in England. A central focus is on social isolation and loneliness. The paper draws on evidence from a multi-stage scoping review of inequalities in later life, conducted within the methodological framework suggested by Arksey and O’Malley (2005). The review explores inequalities in terms of personal social relationships that are associated with the characteristics of gender, age, ethnicity, socio-economic status, and place of residence. The review process assesses not only the quality and strength of evidence concerning unequal social relationships in later life, but also identifies promising opportunities for interventions to narrow inequalities in such relationships.

We establish a sample of 560 spectroscopically confirmed cluster members of MACS J1206.2- 0847 at z = 0.45 and utilize multi-wavelength and multi-component Sersic profile fitting to provide luminosities and sizes for the key structural components bulge and disk. While the difference between field and cluster galaxy properties are mostly due to a preference for cluster members to be early-type (quiescent, bulge-dominated), we see evidence for an outer disk fading and a sharp rise in the number of red disks with smaller effective radii at the tidally active cluster region around R200. Even though red disks are already virialized according to their velocity distribution, they are clearly not part of the old population found in the innermost region; they represent an important population of transitional objects in clusters.

Objective Prenatal cocaine exposure (PCE) can cause persistent neuropsychological and motor abnormalities in affected children, but the physiological consequences of PCE remain unclear. Conclusions drawn from clinical studies can sometimes be confounded by polysubstance abuse and nutritional deprivation. However, existing observations suggest that cocaine exposure in utero, as in adults, increases synaptic dopamine and promotes enduring dopamine-dependent plasticity at striatal synapses, altering behaviors and basal ganglia function. Methods We used a combination of behavioral measures, electrophysiology, optical imaging, and biochemical and electrochemical recordings to examine corticostriatal activity in adolescent mice exposed to cocaine in utero. Results We show that PCE caused abnormal dopamine-dependent behaviors, including heightened excitation following stress and blunted locomotor augmentation after repeated treatment with amphetamine. These abnormal behaviors were consistent with abnormal γ-aminobutyric acid (GABA) interneuron function, which promoted a reversible depression in corticostriatal activity. PCE hyperpolarized and reduced tonic GABA currents in both fast-spiking and persistent low-threshold spiking type GABA interneurons to increase tonic inhibition at GABAB receptors on presynaptic corticostriatal terminals. Although D2 receptors paradoxically increased glutamate release following PCE, normal corticostriatal modulation by dopamine was reestablished with a GABAA receptor antagonist. Interpretation The dynamic alterations at corticostriatal synapses that occur in response to PCE parallel the reported effects of repeated psychostimulants in mature animals, but differ in being specifically generated through GABAergic mechanisms. Our results indicate approaches that normalize GABA and D2 receptor-dependent synaptic plasticity may be useful for treating the behavioral effects of PCE and other developmental disorders that are generated through abnormal GABAergic signaling. Interactions between dopamine and glutamate signalling within the nucleus accumbens core are required for behavioural reinforcement and habit formation. Dopamine modulates excitatory glutamatergic signals from the prefrontal cortex, but the precise mechanism has not been identified. We combined optical and electrophysiology recordings in murine slice preparations from CB1 receptor-null mice and green fluorescent protein hemizygotic bacterial artificial chromosome transgenic mice to show how dopamine regulates glutamatergic synapses specific to the striatonigral and striatopallidal basal ganglia pathways. At low cortical frequencies, dopamine D1 receptors promote glutamate release to both D1 and D2 receptor-expressing medium spiny neurons while D2 receptors specifically inhibit excitatory inputs to D2 receptor-expressing cells by decreasing exocytosis from cortical terminals with a low probability of release. At higher cortical stimulation frequencies, this dopaminergic modulation of presynaptic activity is occluded by adenosine and endocannabinoids. Glutamatergic inputs to both D1 and D2 receptor-bearing medium spiny neurons are inhibited by adenosine, released upon activation of NMDA and AMPA receptors and adenylyl cyclase in D1 receptor-expressing cells. Excitatory inputs to D2 receptor-expressing cells are specifically inhibited by endocannabinoids, whose release is dependent on D2 and group 1 metabotropic glutamate receptors. The convergence of excitatory and inhibitory modulation of corticoaccumbal activity by dopamine, adenosine and endocannabinoids creates subsets of corticoaccumbal inputs, selectively and temporally reinforces strong cortical signals through the striatonigral pathway while inhibiting the weak, and may provide a mechanism whereby continued attention might be focused on behaviourally salient information.

We present a detailed investigation into the effects of galaxy environment on their star formation rates (SFR) using galaxies observed in the Galaxy and Mass Assembly Survey (GAMA). We use three independent volume-limited samples of galaxies within z < 0.2 and Mr < -17.8. We investigate the known SFR-density relationship and explore in detail the dependence of SFR on stellar mass and density. We show that the SFR-density trend is only visible when we include the passive galaxy population along with the star-forming population. This SFR-density relation is absent when we consider only the star-forming population of galaxies, consistent with previous work. While there is a strong dependence of the EWH?a on density we find, as in previous studies, that these trends are largely due to the passive galaxy population and this relationship is absent when considering a "star-forming" sample of galaxies. We find that stellar mass has the strongest influence on SFR and EWH?a with the environment having no significant effect on the star-formation properties of the star forming population. We also show that the SFR-density relationship is absent for both early and late-type star-forming galaxies. We conclude that the stellar mass has the largest impact on the current SFR of a galaxy, and any environmental effect is not detectable. The observation that the trends with density are due to the changing morphology fraction with density implies that the timescales must be very short for any quenching of the SFR in infalling galaxies. Alternatively galaxies may in fact undergo predominantly in-situ evolution where the infall and quenching of galaxies from the field into dense environments is not the dominant evolutionary mode.

Phasic dopamine transmission encodes the value of reward-predictive stimuli and influences both learning and decision-making. Altered dopamine signaling is associated with psychiatric conditions characterized by risky choices such as pathological gambling. These observations highlight the importance of understanding how dopamine neuron activity is modulated. While excitatory drive onto dopamine neurons is critical for generating phasic dopamine responses, emerging evidence suggests that inhibitory signaling also modulates these responses. To address the functional importance of inhibitory signaling in dopamine neurons, we generated mice lacking the β3 subunit of the GABAA receptor specifically in dopamine neurons (β3-KO mice) and examined their behavior in tasks that assessed appetitive learning, aversive learning, and risk preference. Dopamine neurons in midbrain slices from β3-KO mice exhibited attenuated GABA-evoked inhibitory post-synaptic currents. Furthermore, electrical stimulation of excitatory afferents to dopamine neurons elicited more dopamine release in the nucleus accumbens of β3-KO mice as measured by fast-scan cyclic voltammetry. β3-KO mice were more active than controls when given morphine, which correlated with potential compensatory upregulation of GABAergic tone onto dopamine neurons. β3-KO mice learned faster in two food-reinforced learning paradigms, but extinguished their learned behavior normally. Enhanced learning was specific for appetitive tasks, as aversive learning was unaffected in β3-KO mice. Finally, we found that β3-KO mice had enhanced risk preference in a probabilistic selection task that required mice to choose between a small certain reward and a larger uncertain reward. Collectively, these findings identify a selective role for GABAA signaling in dopamine neurons in appetitive learning and decision-making.

Charcot-Marie-Tooth disease (CMT) is one of the most common inherited neurological disorders, affecting 36 in 100,000 people. CMT type 1A (hereditary motor and sensory neuropathy) is the most frequent form of this disease, affecting 60 to 80% of the CMT population, but its diagnosis may be delayed because of inconsistent clinical signs and symptoms and a considerable variability in age at onset. Here, we report on four children (aged 10-17y) who presented with neuromuscular hip dysplasia and other orthopedic abnormalities but were only later diagnosed with CMT 1A. Hip dysplasia may be the initial clinical sign in CMT, so children with late-manifesting hip disease (i.e. age >8y) should be examined for signs of peripheral neuropathy, particularly when presenting with a 'waddling' or broad-based gait.

The performance of x-ray capillary lenses has been evaluated. The tests were carried out using an x-ray tube set-up. A single glass capillary with tapered inner channel, a monolithic glass polycapillary, and an in-house manufactured single metallic capillary with parabolic inner channel were characterized in terms of gain, spatial resolution, and element detection limits. The spatial resolution of a confocal set-up utilizing a monolithic glass polycapillary and a polycapillary conical collimator has also been measured. The highest gain of about 2500 was observed for the glass polycapillary. The maximum gain achieved with the single glass capillary was equal to about 25, and the gain of the metallic capillary was only slightly greater than 1. For the glass capillary and polycapillary lenses, significant filtering of the higher-energy photons (energy > 8 keV) was observed. The lowest relative detection limits were obtained with an ordinary cylindrical collimator and the polycapillary lens. Similar absolute detection limits were achieved with the use of the polycapillary and single capillary lenses. A relation between the ratios of the detection limits of elements achieved with different x-ray lenses and the lens parameters (spatial resolution and gain) has been proposed and was verified experimentally. The monolithic polycapillary lens was found to be an optimum focusing device for an x-ray tube-based scanning spectrometer. This type of x-ray lens can be coupled with a polycapillary conical collimator or a polycapillary half-lens to make a confocal x-ray microscope capable of depth profiling with a spatial resolution equal to about 30 micrometers. A portable focused-beam XRF spectrometer was designed, constructed, and manufactured. The spectrometer allows to detect and perform analysis of chemical elements from Na upwards. The system is equipped with a compact vacuum chamber to reduce absorption of both the excitation and the fluorescence radiation in air. A low power Pd-anode tube operated up to 50 kV and 1 mA with a point focus of 400 μm is used as excitation source. A polycapillary lens with a spot size of about 160 μm, or a collimator with a 1 mm inner diameter can be used alternatively for either focusing or collimating the primary beam. The fluorescence radiation is collected by an Si drift detector with an active area of 10 mm2 and equipped with an 8 μm Be entrance window. A compact vacuum chamber was designed to house the X-ray beam optics and the detector snout. The chamber is attached to the X-ray tube and can be pumped down to 0.1 mbar. A Kapton™ window of 7.5 μm thickness allows to locate the investigated spot at about 1–2 mm distance outside of the chamber, thus minimizing absorption losses in the excitation and X-ray fluorescence radiation paths. Two lasers pointers are mounted inside the chamber. The laser beams cross at a point outside the chamber in front of the entrance window and coincide with the focal spot of the polycapillary. This paper reports some preliminary results obtained from an in situ analysis of bronze samples as well as a comparison of these data with those given by other laboratory spectrometers and the reference values provided by the Italian bronze foundry Venturi Arte Bologna, Italy.

Owing to the increased demands from society for reliable/certified analytical results, x-ray fluorescence laboratories face an urgent need for quality management system (QMS) implementation, in particular complying with the general requirements for the competence of testing and calibration laboratories of the latest approved ISO standard (ISO/IEC 17025: 2005). Some specific features of energy dispersive x-ray fluorescence (EDXRF) techniques require a careful interpretation of some concepts related to method validation and quality control. This paper presents some thoughts on how to assess the traceability of the results and to evaluate the characteristics of performance of the analytical methods, including linearity, working range, precision, trueness and detection limits. Typical examples of EDXRF analytical techniques are provided for illustration, as well as some recommendations to quantify the uncertainty of the results. Copyright © 2007 John Wiley & Sons, Ltd.

Low-energy accelerators have in the past produced a major part of our current knowledge of nuclear physics. Today they are mainly used for applied research and industrial applications. In view of this, the International Atomic Energy Agency (IAEA) has during recent years initiated several Coordinated Research Projects (CRPs) concerning Ion Beam Analysis of Materials, Accelerator Mass Spectrometry and Nuclear Microprobe Techniques. The CRPs involve laboratories from developing as well as developed Member States, networking on a common topic coordinated by the IAEA. In order to facilitate networking, the IAEA has recently published the “World Survey of Accelerator Based Analytical Techniques” available on the Internet and as a CD-ROM. The IAEA maintains also a beamline at a 6 MV Van de Graaff accelerator in the Ru đ jer Boskovic Institute, Zagreb, Croatia. Small and medium power accelerator driven spallation neutron sources will become more important as many small neutron producing research reactors are approaching the end of their useful working life. The IAEA has, within its Department for Nuclear Sciences and Applications, a programme on the Effective Utilization of Accelerators. This programme helps Member States, in particular developing Member States, in finding new areas of applications for their low and medium energy accelerators through increased participation in activities such as Coordinated Research Projects, Technical Meetings and Conferences. This paper describes the IAEA's current programme on accelerator utilization and proposed future activities.

This paper describes the specific philosophy behind the functioning of the IAEA XRF Laboratory at Seibersdorf and its role in the XRF community. Some examples of the research and development activities and the results obtained are given. They include methodological development and construction of XRF instruments in order to extend the applicability range of the XRF technique, particularly in support of applications of the analytical technique in developing IAEA member states. The contribution of the laboratory to the training in methodology and applications of XRF techniques and to development of QA/QC procedures is also emphasised. Six radioactive particles stemming from Thule area (NW-Greenland) were investigated by gamma-ray and L X-ray spectrometry based on radioactive disintegration, scanning electron microscopy coupled with energy-dispersive and wavelength-dispersive X-ray spectrometer, synchrotron radiation based techniques as microscopic X-ray fluorescence, microscopic X-ray absorption near-edge structure (A-XANES) as well as combined X-ray absorption and fluorescence microtomography. Additionally, one particle from Mururoa atoll was examined by microtomography. From the results obtained, it was found out that the U and Pu were mixed in the particles. The U/Pu intensity ratios in the Thule particles varied between 0.05 and 0.36. The results from the microtomography showed that U/Pu ratio was not homogeneously distributed. The 241 Am/ 238+239+240 Pu activity ratios varied between 0.13 and 0.17, indicating that the particles originate from different source terms. The oxidation states of U and Pu as determined by A-XANES showed that U(IV) is the preponderant species and for Pu, two types of particles could be evidenced. One set had about 90% Pu(IV) while in the other the ratio Pu(IV)/Pu(VI) was about one third. D 2005 Elsevier B.V. All rights reserved.

X-ray tube based, micro-beam X-ray fluorescence scanning spectrometer has been equipped with two energy dispersive X-ray detectors. The two-detector configuration allows for simultaneous collection of X-ray fluorescence (XRF) and transmitted X-ray beam signals with a spatial resolution in the range of 10–50 μm, depending on the X-ray focussing element in use. The XRF signal is collected with a standard, liquid nitrogen cooled Si(Li) detector. The X-ray beam transmitted through the sample is acquired with a thermoelectrically cooled, silicon drift (SD) detector. The data acquisition is carried out in a fully automatic way under control of the SPECTOR-LOCATOR software. The software controls the scanning procedure and X-ray spectra acquisition during the scan. The energy dispersive X-ray spectra collected at every “pixel” are stored for off-line processing. For selected regions of interest (ROI’s), the element maps are constructed and displayed on-line. The spectrometer has been used for mapping elemental distributions and for performing 2D- and 3D-tomograpic imaging of minute objects in X-ray absorption and in X-ray fluorescence mode. A unique feature of the described system is simultaneous utilization of the two detectors, Si(Li) and SD, which adds new options for quantitative analysis and data interpretation. Examples of elemental mapping and 3D tomographic imaging as well as the advanced features of the SPECTOR-LOCATOR measurement control and data acquisition software are presented in this work.

The energy-dispersive x-ray fluorescence (EDXRF) technique has limitations in the quantitative analysis of light elements (low-Z analytes with Z < 10), for many reasons. This work, however, circumvents the problem through an a priori determination of low-Z analytes, representative of plant-based samples. The main purpose of this work was to characterize the major elements in the dark matrix of some plant-based samples (including biomonitors) using Rutherford backscattering spectrometry (RBS), and the results provided as a generalized input for EDXRF analysis. The derived stoichiometry and mass ratio for the moss, lichen, and cotton cellulose samples analyzed were found to be similar and close to C 7 H 10 O 5 , with an average matrix of C = 49.8%, H = 4.0% and O = 45.8%. Quantitative analysis of plant-based reference material IAEA-336 (lichen) was subsequently carried out. Use of the a priori determined dark matrix elements (from one-time RBS spectrometry) extended the scope of applicability of the EDXRF quantitative methods used, and improved accuracy in the elemental analysis of plant-based samples. The results obtained were in good agreement with the reference values.
