Antibacterial coatings on biomedical instruments are of great interest because they can suppress bacterial colonization on these instruments. In this study, antibacterial polymeric thin coatings were deposited on teflon substrates using atmospheric pressure plasma polymerization from a propane-butane mixture. The plasma polymerization was performed by means of surface dielectric barrier discharge burning in nitrogen at atmospheric pressure. The chemical composition of plasma polymerized propane-butane films was studied by energy-dispersive X-ray spectroscopy (EDX) and FTIR. The film surface properties were studied by SEM and by surface energy measurement. The EDX analysis showed that the films consisted of carbon, nitrogen and oxygen from ambient air. The FTIR analysis confirmed, in particular, the presence of alkyl, nitrile, acetylene, imide and amine groups. The deposited films were hydrophilic with a water contact angle in the range of 13-23 degrees. The thin film deposited samples were highly active against both S. aureus and E. coli strains in general. On the other hand, the films were cytocompatible, reaching more than 80% of the cell viability threshold compared to reference polystyrene tissue.

Polyoxazoline thin coatings were deposited on glass substrates using atmospheric pressure plasma polymerization from 2-ethyl-2-oxazoline vapours. The plasma polymerization was performed in dielectric barrier discharge burning in nitrogen at atmospheric pressure. The thin films stable in aqueous environments were obtained at the deposition with increased substrate temperature, which was changed from 20 degrees C to 150 degrees C. The thin film deposited samples were highly active against both S. aureus and E. coli strains in general. The chemical composition of polyoxazoline films was studied by FTIR and XPS, the mechanical properties of films were studied by depth sensing indentation technique and by scratch tests. The film surface properties were studied by AFM and by surface energy measurement. After tuning the deposition parameters (i.e., monomer flow rate and substrate temperature), stable films, which resist bacterial biofilm formation and have cell-repellent properties, were achieved. Such antibiofouling polyoxazoline thin films can have many potential biomedical applications.

Efficient treatment of contaminated water in industrially viable volumes is still a challenging task. The hydrodynamic cavitation plasma jet (HCPJ) is a promising plasma source for industrial-scale generation of biologically active environments at high flow rates of several m(3)/h. The combined effect of a hydro-mechanical phenomenon consisting of hydrodynamic cavitation and electrical discharge in cavitation voids was found to be highly efficient for large-volume generation of reactive oxygen species, ultraviolet (UV) radiation, and electro-mechanical stress in a liquid environment. Here, the persistence of biocidal properties of HCPJ-activated water (i.e., plasma-activated water (PAW)) was tested by the study of algae and cyanobacteria inactivation. Algae and cyanobacteria cultivated in media containing PAW (1:1) were completely inactivated after 72 h from first exposure. The test was performed at a total power input of up to 0.5 kWh/m(3) at the treated liquid flow rate of 1 m(3)/h. A beneficial modification of our previous HCPJ design is described and thoroughly characterized with respect to the changes of hydrodynamic flow conditions as well as discharge performance and its optical characteristics. The modification proved to provide high biocidal activity of the resulting PAW, which confirms a strong potential for further design optimization of this promising water (liquid) plasma source.

Cyanobacterial water blooms represent toxicological, ecological and technological problems around the globe. When present in raw water used for drinking water production, one of the best strategies is to remove the cyanobacterial biomass gently before treatment, avoiding cell destruction and cyanotoxins release. This paper presents a new method for the removal of cyanobacterial biomass during drinking water pre-treatment that combines hydrodynamic cavitation with cold plasma discharge. Cavitation produces press stress that causes Microcystis gas vesicles to collapse. The cyanobacteria then sink, allowing for removal by sedimentation. The cyanobacteria showed no signs of revitalisation, even after seven days under optimal conditions with nutrient enrichment, as photosynthetic activity is negatively affected by hydrogen peroxide produced by plasma burnt in the cavitation cloud. Using this method, cyanobacteria can be removed in a single treatment, with no increase in microcystin concentration. This novel technology appears to be highly promising for continual treatment of raw water inflow in drinking water treatment plants and will also be of interest to those wishing to treat surface waters without the use of algaecides

Stable antimicrobial nisin layers were prepared on the carrying medium-polyvinyl alcohol (PVA) films, crosslinked by glutaric acid. Surface plasma dielectric coplanar surface barrier discharge (DCSBD) modification of polyvinyl alcohol was used to improve the hydrophilic properties and to provide better adhesion of biologically active peptide-nisin to the polymer. The surface modification of films was studied in correlation to their cross-linking degree. Nisin was attached directly from the salt solution of the commercial product. In order to achieve a stable layer, the initial nisin concentration and the following release were investigated using chromatographic methods. The uniformity and stability of the layers was evaluated by means of zeta potential measurements, and for the surface changes of hydrophilic character, the water contact angle measurements were provided. The nisin long-term stability on the PVA films was confirmed by tricine polyacrylamide gel electrophoresis (SDS-PAGE) and by antimicrobial assay. It was found that PVA can serve as a suitable carrying medium for nisin with tunable properties by plasma treatment and crosslinking degree. © 2018 by the authors.

Bio-interaktivní a bio-inertní materiály připravené pomocí depozice organokřemičitých vrstev v plazmatu na povrchu polykarbonátu byly studovány a popsány v tomto příspěvku. Dusík s malou příměsí hexamethyldisiloxanu byl použit jako plazmový plyn., Bio-interactive and bio-inert materials prepared by plasma deposition of organosilicon thin films on polycarbonate substrates were studied and described in this communication. The atmospheric pressure surface barrier discharge was applied. Nitrogen with small admixture of hexamethyldisiloxane (HMDSO) was used as plasma discharge gas. The zeta potential measurement followed by cell adhesion to polystyrene was used to find optimal values of stability of the cell suspension and cell adhesion to the tested surfaces. The samples of plasma deposited layers were characterized by contact angle measurement.

Diplomová práca sa zaoberá problematikou využitia prídavku nanočastíc na zlepšovanie vlastností cementových kompozitov. V teoretickej časti sú zhrnuté poznatky z výskumu v tejto oblasti so zameraním na metódy dispergácie nanočastíc a ich úpravu pre použitie v cementových kompozitoch. Praktická časť sa zameriava na porovnanie metód dispergácie a plazmovej úpravy vodných roztokov nanočastíc redukovaného grafén oxidu (rGO) z pohľadu procesu zhlukovania. Počas tejto práce bola vyvinutá metóda systematického optického/vizuálneho sledovania sedimentácie/zhlukovania na doplnenie sofistikovaných metód ako je spektrofotometria (UV/Vis) a elektrónová mikroskopia (SEM). Ďalej bol stanovení vplyv prídavku rGO na vlastnosti cementovej malty, v podobe vodných roztokov pripravených metódami dispergácie určenými v predošlej časti., The diploma thesis deals with the possibility of using the addition of nanoparticles to improve the properties of cement composites. The theoretical part summarizes the findings of research in this area with a focus on methods of dispersion of nanoparticles and their treatment for use in cement composites. The experimental part focuses on the comparison of methods of dispersion and plasma treatment of reduced graphene oxide (rGO) nanoparticle solutions from the point of view of the agglomeration process. During this work, a method of systematic optical/visual monitoring of sedimentation/agglomeration was developed to complement sophisticated methods such as spectrophotometry (UV/Vis) and electron microscopy (SEM). Furthermore, the effect of the addition of rGO on the properties of cement mortar, in the form of aqueous solutions prepared by the dispersion methods determined in the previous section, was investigated.

Předložená diplomová práce je zaměřena na studium dílčích inaktivačních mechanismů uplatňujících se při sterilizaci prokaryotických systémů v dielektrickém bariérovém výboji (DBD). Sterilizace je důležitou biomedicínskou i potravinářskou technologií a plazmová sterilizace je jednou z metod, které vyhovují požadavkům na sterilizaci teplotně senzitivních materiálů a materiálů citlivých na chemické látky. Pro opracování vzorků byl použit dielektrický bariérový výboj pracující za atmosférického tlaku. Výboj byl generován v argonu a dusíku, hustota plazmového výkonu pro argon činila 2725,93 mW cm–3, v případě použití dusíku to bylo 2325,93 mW cm–3. Jako prokaryotní mikroorganismus byla použita grampozitivní bakterie Bacillus subtilis a gramnegativní bakterie Escherichia coli. Vzorky byly naneseny na filtračním papíře Whatman 1. Byl posuzován vliv sterilizačního účinku UV záření, teploty, reaktivních částic a plazmového výboje, kde působí tyto agens dohromady. Účinky UV záření a teploty byly studovány odděleně. Vliv UV záření byl zjišťován za použití křemenného skla, jež UV záření propouští. Část vzorků byla při opracování zakryta sklem, část byla ponechána volně. Vzorky přikryté sklem byly vystaveny působení UV záření a teploty (tu nelze eliminovat), vzorky ponechány volně byly vystavené přímému působení plazmatu. Z výsledků je patrné, že u vzorků zakrytých sklem bylo dosaženo nižší inaktivace než u vzorků, které byly vystaveny přímému působení plazmatu. V plazmatu totiž na vzorky působí kromě UV záření a teploty také reaktivní částice. Při studiu vlivu teploty byly nejprve pomocí termočlánku změřeny teploty působící mezi elektrodami. Poté byly vzorky v termostatu vystaveny působení stejných teplot. Výsledky byly srovnány s výsledky při přímé expozici plazmatem a bylo zjištěno, že vliv teploty na inaktivaci mikroorganismů je velmi malý. Pomocí optické emisní spektroskopie byla provedena diagnostika plazmatu. During the plasma exposition selected samples were covered with the quartz window while other samples were directly exposed to the plasma. Covered samples were exposed to UV radiation and temperature (which cannot be eliminated), samples without quartz window were directly plasma exposed (treated). Results show that for covered samples the lower inactivation was reached than by the samples directly exposed to plasma. When studying the effect of temperature, the temperature between the DBD electrodes was measured by means of a thermocouple. Afterwards the samples were placed in an oven and exposed to the same temperature as was measured between the electrodes. By comparing the results of heat treated samples and plasma treated samples it can be assumed that the influence of the temperature during the sterilization process in DBD is very low. T

Ozon, jedna z forem kyslíku, je vysoce reaktivní molekulou, která je známa zejména díky svým dezinfekčním vlastnostem. Teoretická část této práce má za cíl shrnout poznatky o základních vlastnostech ozonu, technologických aplikacích, generaci tohoto plynu a mechanismech reakcí ozonu na povrchu některých materiálů. Experimentální část sestává z řady pokusů, které jsou zaměřeny na vznik a zánik ozonu na povrchu skleněné kyvety. The theoretical part of this work describes the fundamental properties of ozone, the technological applications, the synthesis of this gas and the mechanism of the reactions on the surface of some materials. The experimental part consists of a number of experiments that are focused on the formation and degradation of ozone on the surface of a glass cuvette. Ozone was, for our purposes, generated by DBD, its concentration was measured by absorption spectrometry.

Praktická časť sa zameriava na porovnanie metód dispergácie a plazmovej úpravy vodných roztokov nanočastíc redukovaného grafén oxidu (rGO) z pohľadu procesu zhlukovania. Počas tejto práce bola vyvinutá metóda systematického optického/vizuálneho sledovania sedimentácie/zhlukovania na doplnenie sofistikovaných metód ako je spektrofotometria (UV/Vis) a elektrónová mikroskopia (SEM). Ďalej bol stanovení vplyv prídavku rGO na vlastnosti cementovej malty, v podobe vodných roztokov pripravených metódami dispergácie určenými v predošlej časti., The diploma thesis deals with the possibility of using the addition of nanoparticles to improve the properties of cement composites. The theoretical part summarizes the findings of research in this area with a focus on methods of dispersion of nanoparticles and their treatment for use in cement composites. The experimental part focuses on the comparison of methods of dispersion and plasma treatment of reduced graphene oxide (rGO) nanoparticle solutions from the point of view of the agglomeration process. During this work, a method of systematic optical/visual monitoring of sedimentation/agglomeration was developed to complement sophisticated methods such as spectrophotometry (UV/Vis) and electron microscopy (SEM). Furthermore, the effect of the addition of rGO on the properties of cement mortar, in the form of aqueous solutions prepared by the dispersion methods determined in the previous section, was investigated.

Předložená diplomová práce je zaměřena na studium dílčích inaktivačních mechanismů uplatňujících se při sterilizaci prokaryotických systémů v dielektrickém bariérovém výboji (DBD). Sterilizace je důležitou biomedicínskou i potravinářskou technologií a plazmová sterilizace je jednou z metod, které vyhovují požadavkům na sterilizaci teplotně senzitivních materiálů a materiálů citlivých na chemické látky. Pro opracování vzorků byl použit dielektrický bariérový výboj pracující za atmosférického tlaku. Výboj byl generován v argonu a dusíku, hustota plazmového výkonu pro argon činila 2725,93 mW cm–3, v případě použití dusíku to bylo 2325,93 mW cm–3. Jako prokaryotní mikroorganismus byla použita grampozitivní bakterie Bacillus subtilis a gramnegativní bakterie Escherichia coli. Vzorky byly naneseny na filtračním papíře Whatman 1. Byl posuzován vliv sterilizačního účinku UV záření, teploty, reaktivních částic a plazmového výboje, kde působí tyto agens dohromady. Účinky UV záření a teploty byly studovány odděleně. Vliv UV záření byl zjišťován za použití křemenného skla, jež UV záření propouští. Část vzorků byla při opracování zakryta sklem, část byla ponechána volně. Vzorky přikryté sklem byly vystaveny působení UV záření a teploty (tu nelze eliminovat), vzorky ponechány volně byly vystavené přímému působení plazmatu. Z výsledků je patrné, že u vzorků zakrytých sklem bylo dosaženo nižší inaktivace než u vzorků, které byly vystaveny přímému působení plazmatu. V plazmatu totiž na vzorky působí kromě UV záření a teploty také reaktivní částice. Při studiu vlivu teploty byly nejprve pomocí termočlánku změřeny teploty působící mezi elektrodami. Poté byly vzorky v termostatu vystaveny působení stejných teplot. Výsledky byly srovnány s výsledky při přímé expozici plazmatem a bylo zjištěno, že vliv teploty na inaktivaci mikroorganismů je velmi malý. Pomocí optické emisní spektroskopie byla provedena diagnostika plazmatu. Vzorky vystavené působení DBD byly analyzovány za pomoci skenov, Presented master’s thesis is focused on the study of the elementary inactivation processes acting during sterilization of procaryotic systems in dielectric barrier discharge (DBD). Sterilization is an important biomedical and food-industry application and plasma sterilization is one of the methods, which are suitable for sterilization of heat and chemical sensitive materials. Biologically contaminated samples were treated in dielectric barrier discharge operated at atmospheric pressure. The discharge was generated in argon and in nitrogen. The plasma power density was 2725,93 mW cm–3 in argon and 2325,93 mW cm–3 in nitrogen. Gram positive bacteria Bacillus subtilis and gram negative bacteria Escherichia coli were used as a bioindicator. Bacteria were spread onto the surface of Whatman No.1 filtration paper. The influence of UV radiation, reactive species, heat and plasma discharge where the synergistic function of all of the agents was studied. Effects of UV radiation and temperature were studied separately. In order to separate the effect of UV radiation generated by DBD the quartz glass transmitting UV radiation was employed. During the plasma exposition selected samples were covered with the quartz window while other samples were directly exposed to the plasma. Covered samples were exposed to UV radiation and temperature (which cannot be eliminated), samples without quartz window were directly plasma exposed (treated). Results show that for covered samples the lower inactivation was reached than by the samples directly exposed to plasma. When studying the effect of temperature, the temperature between the DBD electrodes was measured by means of a thermocouple. Afterwards the samples were placed in an oven and exposed to the same temperature as was measured between the electrodes. By comparing the results of heat treated samples and plasma treated samples it can be assumed that the influence of the temperature during the sterilization process in DBD is very low. T
