Methods of spatiotemporal characterization of nonequilibrated polymer based matrices are still immature and imperfect. The purpose of the study was to develop the methodology for the spatiotemporal characterization of water transport and properties in alginate tablets under hydration. The regions of low water content were spatially and temporally sampled using Karl Fisher and Differential Scanning Callorimetry (spatial distribution of freezing/nonfreezing water) with spatial resolution of 1 mm. In the regions of high water content, where sampling was infeasible due to gel/sol consistency, magnetic resonance imaging (MRI) enabled characterization with an order of magnitude higher spatial resolution. The minimally hydrated layer (MHL), infiltration layer (IL) and fully hydrated layer (FHL) were identified in the unilaterally hydrated matrices. The MHL gained water from the first hour of incubation (5–10% w/w) and at 4 h total water content was 29–39% with nonfreezing pool of 28–29%. The water content in the IL was 45–47% and at 4 h it reached ~50% with the nonfreezing pool of 28% and T2 relaxation time &lt, 10 ms. The FHL consisted of gel and sol layer with water content of 85–86% with a nonfreezing pool of 11% at 4 h and T2 in the range 20–200 ms. Hybrid destructive/nondestructive analysis of alginate matrices under hydration was proposed. It allowed assessing the temporal changes of water distribution, its mobility and interaction with matrices in identified layers.

The purpose was to show, using destructive/nondestructive methods, that the interplay between water, tablet structure, and composition determine the unique spatiotemporal hydration pattern of polymer-based matrices. The tablets containing a 1:1 w/w mixture of sodium alginate with salicylic acid (ALG/SA) or sodium salicylate (ALG/SNA) were studied using Karl Fischer titration, differential scanning calorimetry, X-ray microtomography, and magnetic resonance imaging. As the principal results, matrix specific features were detected, e.g., “locking” of the internal part of the matrix (ALG/SA), existence of lamellar region associated with detection of free/freezing water (ALG/SA), existence of water penetrating the matrix forming specific region preceding infiltration layer (ALG/SNA), switch in the onset temperature of endothermic water peak associated with an increase in the fraction of non-freezing water weight per dry matrix weight in the infiltration layer (ALG/SNA). The existence of complicated spatiotemporal hydration patterns influenced by matrix composition and molecular properties of constituents has been demonstrated.

A catalytic and enantioselective preparation of the (S)-4-methyleneproline scaffold is described. The key reaction is a one-pot double allylic alkylation of an imine analogue of glycine in the presence of a chinchonidine-derived catalyst under phase transfer conditions. These 4-methylene substituted proline derivatives are versatile starting materials often used in medicinal chemistry. In particular, we have transformed tert-butyl (S)-4-methyleneprolinate (12) into the N-Boc-protected 5-azaspiro[2.4]heptane-6-carboxylic acid (1), a key element in the industrial synthesis of antiviral ledipasvir.

[spa] En la industria farmacéutica es común buscar procesos alternativos de producción de fármacos comerciales o a punto de entrar en el mercado. En este trabajo, y en colaboración con Esteve Química S.A., se han buscado rutas sintéticas novedosas para dos fármacos de interés, aprobados en 2017 y 2018 respectivamente. La ruta planteada hacia el primero requería la construcción de un indazol y su acoplamiento con un bromuro de arilo. Se lograron estas transformaciones con rendimientos aceptables, disponiendo así de un anillo aromático unido al indazol y con un grupo funcional (en para) potencialmente transformable en aldehído bencílico. Dicha transformación se consiguió con peryodinano de Dess-Martin. La construcción estereoselectiva del anillo de piperidina presente en la molécula a partir del aldehído no fue posible y se abandonó el proyecto. El segundo es un dipéptido doblemente arilado (con restos piridínicos), y con un enlace amida en el extremo C-terminal. Esto nos permitió dividir la molécula en 5 fragmentos comercialmente disponibles y con la estereoquímica deseada. Aunque se buscaba una ruta convergente, no fue posible porque el ácido piroglutámico arilado epimerizaba en los intentos de unión con el otro aminoácido. Por su parte, la arilación de la clorofenilglicina presentó problemas debido a la poca estabilidad del material de partida y la pérdida total de la estereoquímica en la mayoría de las pruebas. Se ensayaron muchas condiciones hasta dar con unas en las que se obtenía producto por un mecanismo similar a una SNAr asistida por Pd. No obstante, este producto resultó totalmente inerte ante los intentos de condensación con el ácido piroglutámico. En conjunto, los resultados negativos obligaban a replantear el orden de la secuencia, pero no se realizaron más pruebas. Por otro lado, es común la aparición de impurezas durante la producción de fármacos. La detección de una impureza de difícil separación durante uno de los procesos de producción de un fármaco en Esteve Química condujo a la necesidad de sintetizar dicho subproducto. La impureza era supuestamente una tetrahidroisoquinolina bencilada, y se planteó su síntesis a partir de un intermedio avanzado. Tras varios intentos de alquilación y acilación fue evidente la necesidad de proteger previamente el nitrógeno para evitar reacciones secundarias. La bromación de la posición aromática deseada, seguida de una bencilación de tipo Negishi (optimizada posteriormente) permitieron la introducción del grupo bencilo. La desprotección del grupo amino presentó complicaciones, pero se pudo entregar cantidad suficiente del producto. Además, se consolidó la metodología de bencilación descubierta al extenderla a diferentes sustratos. Finalmente, se quiso profundizar en la síntesis de tetrahidroisoquinolinas presentes en productos como el Moexipril ya que se disponía de cierta experiencia en reacciones catalíticas de activación C−H por aminas primarias o iminas que podían conducir a estas estructuras. Se ha realizado un estudio extenso sobre coordinación y metalación de sustratos del tipo piridin-2-ilmetaniminas que actúan como ligandos bidentados, tanto con paladio como con platino. Los resultados demuestran la necesidad de la presencia de dos grupos alquilo en α al nitrógeno para obtener resultados positivos. Con paladio sólo se consiguió metalar una de las iminas ensayadas sin dos metilos en α. Con platino, en cambio, fueron tres. Se comprobó que cambios en el anillo de piridina podían favorecer mucho el proceso de metalación: utilizar picolina o quinolina incrementaba la reactividad de las iminas frente al metal. Los mejores resultados con platino lo sitúan como posible sustituto al paladio en reacciones catalíticas que no funcionaron anteriormente., [eng] This work began with the search for new synthetic approaches to two drugs of interest in collaboration with Esteve Química S.A. The indazol moiety of the first drug was constructed in five steps, and its N2-arylation was performed with several compounds. However, it was not possible to form the chiral piperidine ring, and the project was abandoned. The retrosynthetic analysis of the second drug allowed the division of the molecule into five commercially available building blocks. The two amino acids bore the desired stereochemistry. The N-arylation of the pyroglutamic acid scaffold was achieved without many complications, whereas the N-arylation of the other amino acid required an extensive screening of conditions. Since we could not form the dipeptide and reach the final product, the whole synthetic path needed revision. We decided to move on to the next project, which concerned the synthesis of an impurity found during the industrial synthesis of a different molecule. After protecting the amino group of the tetrahydroisoquinoline moiety, bromination of the desired aromatic position was performed. The benzylation was achieved through a microwave-assisted Negishi reaction, accomplishing the main objective of the chapter. Later, we carried out an optimisation of this process and checked the scope of the conditions by testing several aryl bromides. Previous works in our group led to the construction of tetrahydroisoquinoline rings through C−H activation. However, some structural limitations were found such as the necessity of two alkyl substituents next to the amino directing group. We have performed a study of the coordination and metallation process of imines with palladium and platinum. Using imines containing nitrogenated heterocycles that could act as bidentate ligands, the objective was to enhance the metallation reaction to overcome the structural limitations. Thus, the knowledge acquired could be used in the future to develop a new catalytic process towards the formation of tetrahydroisoquinolines. Besides obtaining four metallacycles without two alkyl groups adjacent to the nitrogen, we found out that picoline and quinoline were more reactive than pyridine.

The objective of these studies is to verify and validate the improvement in the inter-tablet coating uniformity for an industrially commercialized coated tablet, without involving changes in the approved registration dossier. Using the CPP (critical process parameters) determined from previous retrospective statistical analysis, the recommended working ranges are identified. Retrospective analysis showed that the design of experiments (DoE) provided an improved process variable configuration. Therefore, it is decided to study two critical parameters: Product temperature and drum speed, with an additional 22 experimental design. The quality results of the samples analyzed show that the aesthetic defects of the batches made with the new working ranges have been reduced. These results have also been corroborated with the 42 industrial batches manufactured with the new ranges. With the optimized parameters, tablets have been coated and the suitability of the model determined. The results demonstrated the overall reliability and effectiveness of the proposed Quality by Design approach and provides a useful tool to help optimize the industrial coating process. This study confirms that it is possible to optimize and validate the manufacturing process of an existing commercial product by means of a DoE with retrospective data. Therefore, no variation in the dossier is required.

[cat] Aquesta tesi doctoral està enfocada a la investigació de nous agents amb potencial activitat antitumoral per inhibició enzimàtica. S’ha descrit la implicació de la G6PDH en el desenvolupament i progressió del càncer degut a la seva participació en la síntesi d’àcids nucleics. Es busca la preparació de compostos que actuïn inhibint de forma més selectiva les cèl·lules canceroses, actuant sobre els enzims implicats en el seu metabolisme, com és la G6PDH, per tal de millorar l’eficàcia i reduir els efectes adversos dels tractaments dels que disposem actualment. S’ha dut a terme la preparació sintètica de tres series de compostos diferents. La primera pertany a les arilmetilenindolinona i derivats, la segona consta de sistemes tiofen-condensats, dissenyats a partir de modelització i amb canvis estructurals a partir de farmacomodulacions modulatives i finalment la tercera sèrie, la corresponent a les urees. Els compostos s’han preparat mitjançant reaccions clàssiques de química orgànica i s’han purificat posteriorment mitjançant tècniques de separació (cromatografia). L’elucidació estructural corresponent s’ha dut a terme mitjançant tècniques de Ressonància Magnètica Nuclear (RMN), Espectroscòpia de masses (EM) i d’Infraroig (IR). Finalment, s’ha dut a terme part de l’avaluació biològica d’alguns dels compostos preparats, utilitzant assajos in vitro de viabilitat cel·lular en diferents línies cel·lulars de leucèmia (TPH-1, MOLT3, NB4, HL-60, K562, KU812 i OCIAML-3) i de càncer de pròstata (PC-3) mitjançant la tècnica MTT. Per tal d’estudiar l’activitat inhibitòria enzimàtica, s’han realitzat assajos d’inhibició directa de la G6PDH, mesurada a partir de la producció d’NADPH., [eng] This PhD has been focused on the discovery of new compounds with antitumoral potential activity for inhibition of enzymes, like G6PDH. The implications of G6PDH for the development and progression of cancer due to its participation in nucleic acid synthesis have been described. Our objective was to prepare compounds that can be able to inhibit enzymes such as G6PDH that involved in metabolism of cancer cells with a good level of selectivity which have a perfect efficacy without significant side effects. Synthetic preparation of three different compounds has been carried out. The first one has arilmetilenindolinone and its derivatives. The second one contains thiophen systems and designed by computational modulator and changed structure from pharmacomodulations and finally, the third series of compounds is based on the urea-derivatives. Compounds have been prepared using classical organic chemistry reactions and further purification with separation techniques (chromatography). Corresponding structural elucidation by NMR, Mass and IR. Finally, part of the biological evaluation of some of the compounds have been prepared and carried out using in vitro assays based on cellular viability on different prostate (PC-3) and leukemic (TPH-1, MOLT3, NB4, HL-60, K562, KU812 and OCIAML-3) cell lines using the MTT assay. The direct inhibition of G6PDH measured by the production quantity of NADPH was used to study enzymatic inhibition activity.

Anther culture is a fast tool to obtain double haploid plant lines for breeding purposes. In rice, this procedure is commonly performed in two steps: i) induction of calli from anthers and ii) regeneration of plantlets from calli. It has been stated that genotype highly influences the anther culture efficiency, so the media used in each step should be optimized for each variety. In this study, we tested different media modifications of an efficient protocol optimized for a medium sized grain temperate japonica NRVC980385, used as a control, in a long grain temperate japonica rice variety (NRVC20120346), and two long grain tropical japonica varieties (303012 and 303013). We found that the addition of 150 mg l(−1) colchicine to the induction medium worked best for all genotypes except for NRVC20120346, whose best induction was obtained with the colchicine-free medium. Referring to regeneration, increased gelling agent in the medium provided the best rates in NRVC980385, improving our former NRVC980385-optimized anther culture protocol. Sorbitol fortified regeneration medium worked the best in the case of the long grain varieties. The presence of colchicine in the induction medium was also related to a higher obtention of double haploid plantlets. This study highlights that genotype is a key factor in the performance of rice anther culture. It has set a first anther culture study on long grain japonica varieties and optimizes the anther culture protocol for temperate japonica medium grain NRVC980385 with the use of colchicine and other additives that increase osmotic stress.

Treballs Finals de Grau d'Enginyeria Química, Facultat de Química, Universitat de Barcelona, Curs: 2017-2018, Tutors: Esther Chamarro Aguilera, Manel Vicente Buil, Oxolamine Citrate is an active ingredient of many antitussive pharmaceutical drugs. Last years, its demand has increased significantly, what has caused the necessity to increase its production. For that, it was studied experimentally the way to optimize the process that was conducting a company to produce the drug back then, determining the critical points and suggesting improvements. Through the laboratory research and implementing many modifications, it was achieved the way to increase its yield from 24% to 64%, thanks to the identification of impurities in the Oxolamine Citrate synthesis. For this reason, in this project is going to be studied how to transfer this optimized process to industrial scale. It is going to work in discontinuous mode. The process can be divided in two phases: Oxolamine Citrate synthesis and its purification. It is going to be studied the most feasible method to the synthesis phase, even if it is going to take into account the purification phase to know the batch time and the time that will take to obtain the requested production. The production that has to be satisfied is 100 tones per year. In order to achieve this quantity, it has been searched the way to get a balance between the production time and the minimum number of equipment required. Consequently, the equipment will be flexible, so when it is finished the annual production of Oxolamine Citrate, they will be able to be adapted to future processes. The equipment used is: for the synthesis phase, 4 vessels, all of them provided of a halfpipe jacket, an agitator and a condenser to assure the reflux of the liquid that could evaporate. Anyway, the capacities of these vessels are not the same, and in them are carried out different operations as can be reactions, extractions or crystallizations. In the case of the purification phase, it would be provided of a centrifuge, a rotary dryer and two micronization mills. It has been scheduled the production process, and has been dimensioned the equipment involved in the synthesis phase, apart from the control and automation system with its P&ID.Lastly, it has been achieved to produce the annual requirement of 100 tones, through the production of 46 batches of 2180 kg in a period of 8 weeks

[eng] Chiral amino alcohol derived lactams are a valuable scaffold for the enantioselective synthesis of alkaloids. In the present thesis the potential of this methodology was expanded and to demonstrate the synthetic utility of chiral tricyclic lactams obtainedthe enantioselective total synthesis complex cis-decahydroquinoline alkaloids was carried out. The alkaloids that were targeted for synthesis using the developed methodology were the lepadin alkaloids (marine alkaloids) and tricyclic cis-decahydroquinoline alkaloids such as cermizine B (Lycopodium alkaloids). To accomplish these objectives further studies on the functionalization of the decahydroquinoline system were required in order to install the appropriate substituents and functionalities for the synthesis of each target. During the present PhD thesis it was possible to complete the total synthesis of (+)-gephyrotoxin 287C, (−)-cermizine B, (−)-lepadin B and (+)-lepadin D and the formal total synthesis of (−)-lepadins A and C.

L'objectiu principal d'aquesta Tesi doctoral ha estat l’obtenció d’una via alternativa a les existents per a l'obtenció d'un intermedi clau, 1, en la síntesi d'un fàrmac anticoagulant. La nova via havia de ser novedosa i patentable per tal de poder obtenir el producte industrialment. Així doncs inicialment es va fer una cerca bibliogràfica per tal de conèixer els precedents sintètics existents i a partir d'aquests es van proposar una sèrie d’objectius més concrets: una primera aproximació per tal de sintetitzar el ciclohexà central de 1, una altra basada en una reacció de iodolactamització per tal de tenir un dels nitrògens del sistema cis-diamino ja introduït i una tercera basada en una aziridina com a intermedi clau tenint en aquest cas tambè ja un dels nitrògens presents en la molècula. La primera aproximació es basava en una reacció Diels-Alder com a etapa clau per a formar el ciclohexà central de la molècula objectiu. Tot i arribar a un intermedi força avançat, no es va poder arribar a producte final i es va haver de descartar aquesta aproximació. La segona via que es va provar consistia en una reacció de iodolactamització com a etapa clau seguida de la formació del derivat de Boc. A partir d’aquest intermedi es van proposar vàries alternatives per a substituir l’àtom de iode i introduir el segon nitrogen a la molècula, ja fos directament amb amoníac per exemple, o a travès d’un equivalent sintètic del grup amina com podia ser l’azida o bè el grup ftalimida. Tambè es va provar de formar intermedis tipus urea amb isocianats i introduïr així el segon nitrogen de l’anell de sis baules intramolecularment. Malauradament cap d’elles va permetre arribar a obtenir el producte objectiu 1 així que es va passar a estudiar la darrera aproximació. La tercera via proposada passava per un intermedi clau aziridínic que es formava per reacció d’una olefina amb una sulfamida amb catàlisi de rodi. Desprès de provar la reacció principal a partir de diferents substrats, es va trobar un intermedi de tipus lactona inesperat però que va ser clau en aquesta aproximació. Seguint una sèrie de reaccions senzilles partint de la lactona es va arribar a obtenir 1. Així doncs es va assolir l’objectiu d’obtenir una via per tal d’obtenir 1. Malaurdament degut a una reivindicació d’una patent que protegeix l’últim intermedi de la síntesi i la transformació d’aquest a 1, no serà possible sintetitzar industrialment 1 seguint aquest nou procès obtingut. Tot i això, es va decidir seguir les reaccions de l’aproximació que havia permès obtenir 1, però variant el grup dimetilamida per altres grups alternatius. Es va pensar en formar èsters activats amb el 2,2,2-trifluoroetanol, el fenol i la N,N- dietilhidroxilamina. Aquesta estratègia permetria arribar a obtenir la molècula objectiu però sense infringir la patent ja que, en la darrera etapa, seria on s’introduiria el grup dimetilamida de manera que no es passaria per l’intermedi reivindicat. Malauradament, amb cap dels grups alternatius utilitzats es va aconseguir arribar a l’objectiu degut a la formació d’ intermedis tipus aziridina, els quals no es van poder transformar a la sulfamida bicíclica desitjada de cinc baules., The main objective of this Thesis was to obtain a novel and alternative route to the existing key intermediate 1 in the synthesis of an anticoagulant drug. The new approach had to be innovative and patentable in order to produce the product industrially. First of all, we searched in the literature the existing synthetic precedents and from these we proposed a series of more specific objectives: a first approach to synthesize the central cyclohexane, another based on a iodolactamization reaction, having one of the nitrogens in the cis-diamino system already introduced, and a third approach based on aziridine as a key intermediate, in this case it has also already one of the nitrogens present in the molecule. The first approach was based on a Diels-Alder reaction as a key step to form the central cyclohexane target molecule. Despite reaching an advanced intermediate, this approach failed to reach the final product and had to discard it. The second route that was tested had a iodolactamization reaction as a key step and followed the synthesis of the Boc derivative of this product . From this intermediate we proposed several alternatives to replace the iodine atom and introduce the second nitrogen in the molecule, either directly with ammonia for example, or through a synthetic equivalent of the amino group for example with an azide or a phthalimide group. We also tried to form an urea intermediate with isocyanate and thus insert the second nitrogen in the six member ring intramolecularly. Unfortunately none of the methods allowed us to arrive at the desired product so we kept on studying the latest approach. The third proposed route passes through a key aziridine intermediate that was formed by the reaction of an olefin with a sulfamide using rhodium catalysts. After testing the main reaction from different substrates, we found a lactone intermediate which was key in this approach. Following a series of simple reactions starting from the lactone we get the objective molecule 1. The main objective was achieved, unfortunately, due to a claim of a patent that protects the last intermediate synthesis and transformation of this to 1, it will not be possible to synthesize 1 industrially following this new process. However, we decided to follow the reactions of the approach that had yielded 1, but varying the dimethylamide group to other alternative groups. We thought about forming activated esters with 2,2,2-trifluoroethanol, phenol and N,N- diethylhydroxylamine. This strategy would reach the target molecule obtained without infringing the patent because it was in the last stage, where it would introduce the dimethylamide group so the route would not pass through the intermediate claimed.

De la mateixa manera, es busca la preparació de compostos que actuïn sobre més d'una diana i de forma selectiva contra les cèl·lules canceroses per tal de millorar l'eficàcia dels furuts tractaments i els seus efectes adversos. S'ha dut a terme la preparació sintètica de tres sèries de compostos; la primera conté en la seva estructura els nuclis de diarilèter i diarilurea. La segona pren com a model el nucli del polifenol natural resveratrol, sobre el qual es fan modificacions estructurals, tant en els fenols com en el doble enllaç mitjançant la fixació de la configuració desitjada. Finalment, la tercera sèrie es basa en la preparació d'anàlegs de la combretastatina A4 mitjançant modificacions en els dos anells aromàtics i la fixació de la seva configuració mitjançant la introducció d'un cicle. Els compostos s'han preparat utilitzant reaccions clàssiques de química orgànica i posteriorment purificats mitjançant tècniques de separació (cromatografia o recristal·lització) i s'ha dut a terme l'elucidació estructural corresponent. Finalment, s'ha realitzat l'avaluació biològica d'un dels compostos preparats, així com també d'altres caps de sèrie del nostre grup de treball, utilitzant assajos in vitro de viabilitat cel·lular en cèl·lules HT29 de càncer de colon mitjançant la tècnica del MTT. Per tal d'estudiar l'activitat antiproliferativa dels compostos en models in vitro representatius de l'ambient tumoral, s'han utilitzat cultius monocapa en condicions de normòxia i hipòxia, així com també models 3D com els esferoids., [eng] This PhD is focused on the discovery of new compounds with potential antitumour activity, through the inhibition of enzymes that are essential in cancer development and progression, like KRAS. At the same time, compounds that exert their activity upon more than one target and selectively in cancer cells are of interest in order to improve treatment efficacy and diminish its side effects. Synthetic preparation of three new series of compounds has been carried out; the first one contains the core of a diarylether and diarylurea. The second takes as a model the scaffold from the natural polyphenol resveratrol, in which structural modifications are done in the phenol moieties and the double bond in order to fix the desired configuration. Finally, the third series is based in the preparation of combretastatin A4 analogues through modification in the two aromatic rings and the fixation of the configuration with the introduction of a cycle. Compounds have been prepared using classical organic chemistry reactions and further purification with separation techniques (chromatography or recrystallization). The consequent structural elucidation has also been performed. Finally, part of the biological evaluation of one of the compounds prepared, as well as other important leads from our group, has been carried out using in vitro assays based on cellular viability in colon cancer HT29 cells using the MTT technique. In order to study the antiproliferative activity of the different compounds in representative models of the tumour environment, monolayer cell cultures have been employed under normoxic and hypoxic conditions as well as 3D models like spheroids.

The manuscript of this Doctoral Thesis has been divided into five main parts: a general introduction into the calyciphylline A-type alkaloids followed by three research chapters and conclude by a review about the radical cyclization of trichloroacetamides. The general introduction provides an overview of the calyciphylline A-type alkaloids, the partial syntheses previously reported and as well as the group precedents in the field of the calyciphylline A-type alkaloids. The second chapter titled “Toward the Synthesis of Calyciphylline A-type alkaloids” describes our synthetic efforts toward the calyciphylline A-type alkaloids with a new synthetic approach. The synthesis started from the cyclohexanedione monoethylene acetal, followed by the 5-endo-trig radical cyclization of trichloroacetamides using Bu3SnH and AIBN as the initiator to give access to 3a-methyloctahydroindoles, the cornerstone of our strategy. After derivatization, several advanced tricyclic intermediates (ABC and ACD ring systems) were synthetized using Pd a-alkenylation of enolates, radical cyclization of trichloroacetamides and aldol condensation. Further functionalization allowed the synthesis of the tetracyclic ABCD ring core of calyciphylline A-type alkaloids via a 6-exo radical cyclization of a trichloroacetamide upon an enol acetate embedded in a tricyclic synthetic intermediate.In summary, it highlights the synthetic usefulness of radical cyclization procedures in structural demanding substrates with the formation of carbon-carbon bonds using dichloromethylcarbamoyl radicals upon electron-rich acceptors. The third chapter titled “Radical Cyclization in the Synthesis of cis-3-Methyloctahydroindol-6-ones” describes a generalization of the 5-endo-trig radical cyclization of trichloroacetamides to other haloacetamides and enynes showing the versatility of this process. It allowed an express synthesis of cis-3-methyloctahydroindoles, a motif embedded in. yuzurimine- and daphniglaucine-type alkaloids, by 5-endo-trig radical cyclizations of alkynes or methylhaloacetamides. The results obtained were interesting from the mechanistic point of view, since some unexpected reaction pathways were found in the radical cyclization processes. This project was performed in collaboration with Sergi Jansana. The fourth chapter titled “Synthesis of a-Chlorolactams by Cyanoborohydride-Mediated Radical Cyclizations of Trichloroacetamides” describes a new methodology to access a-chlorolactams in a simple and an efficient way using cheap NaBH3CN and easily available allylic and homoallylic secondary amines as starting materials. The standard conditions favored the diastereoselectivity toward the formation of the thermodynamic trans a-chlorolactam products and can be applied to non-activated, electron rich or electron poor alkenes. The process can be performed in a gram scale. And this methodology allowed the synthesis of a library of compounds sent for biological testing. This project was performed in collaboration with Bayer CropScience (Frankfurt am Main, Germany) under the supervision of Dr. Stephen Lindell and Dr. Harald Jakobi. The fifth chapter titled “Radical Cyclization of Trichloroacetamides: Synthesis of Lactams” gathers all the published results and methodologies involving trichloroacetamides in radical chemistry. Trichloroacetamides can act as radical precursors to synthesize nitrogen-containing heterocycles such as β-, γ-, and δ-lactams which are highly useful building blocks in the synthesis of nitrogen-containing natural products and bioactive molecules, in a variety of processes, mainly involving atom transfer radical cyclizations (ATRC), mediated by Cu(I) or Ru(II) catalysts, and the hydride reductive method, employing either Bu3SnH or (Me3Si)3SiH, or recently NaBH3CN. Additionally, amine-mediated single electron transfer cyclizations, as well as radical processes promoted by Ni, Fe, Mn, Ti, and Ag, have been developed.

[cat] Posar al mercat un nou fàrmac és un procés molt llarg i costós i que comporta un risc econòmic important per una empresa farmacèutica. En canvi, el desenvolupament d'un procediment millorat per a la producció d'un fàrmac ja conegut, un cop ha passat a ser genèric, és molt menys arriscat i genera propietat industrial valuosa i una possibilitat de negoci. El desenvolupament d’una nova ruta implica fomentar la innovació i la millora continuada en els processos de fabricació augmentant la competitivitat en els plans de producció, el que exigeix un coneixement exhaustiu i científic dels processos de fabricació. L’objectiu del present Projecte, en col·laboració amb Esteve Química, és el desenvolupament de rutes sintètiques potencialment patentables i industrialitzables d'un antivíric d’interès (o bé d'un intermedi avançat d'aquest) i d’un producte utilitzat per a malalties d’insuficiència cardíaca que es trobin ja en el mercat o en fases clíniques molt avançades. A la primera part d’aquest treball l’objectiu ha estat el desenvolupament de rutes sintètiques per a l’obtenció d’un fragment present en alguns antivírics. Es van explorar fins a sis rutes diferents per a arribar a l’estructura d’interès. S’ha trobat una aproximació sense precedents basada en la doble alquilació estereoselectiva de les imines de glicinats en condicions de transferència de fase. Hem aconseguit arribar a l’estructura desitjada amb un rendiment global del 29%. Tot i els bons resultats obtinguts, la síntesi desenvolupada no és clarament competitiva respecte a la alternativa descrita a la bibliografia, ja que, tot i que té millors rendiments i conté etapes fàcilment millorables en els processos d'escalat, els materials de partida resulten ser més cars. A la segona part d’aquest treball s’ha afrontat l’objectiu d’obtenir un compost utilitzat en el tractament de la insuficiència cardíaca, utilitzant una amida derivada de la pseudoefedrina com a etapa clau de la síntesi. Vam arribar a l’estructura d’interès en 5 etapes i amb un rendiment global del 14%, però la etapa clau del procés donà resultats irregulars i excessos diastereomèrics discrets. Aquests resultats podrien ser millorables, si es dugués a terme un estudi més exhaustiu de la reacció d’alquilació. La síntesi proposada per aquest compost és nova i patentable i probablement competitiva respecte als resultats que hi han descrits fins al moment a la bibliografia. En general, doncs, es pot dir que, en les dues parts d’aquests treball s’ha arribat als productes desitjats, a través de nous processos, amb rendiments del rang dels descrits a la bibliografia. Tot i això, la viabilitat industrial no està assegurada i són necessaris estudis més profunds i l’optimització d’algunes etapes., [eng] The aim of this project, in collaboration with Esteve Química is the development of potentially patentable and industrializable synthetic routes to an antiviral (or an advanced intermediate of it) and a drug used for diseases of heart failure. Both compounds are already on the market or in very advanced clinical stages. In the first part of this work the aim has been the development of synthetic routes to get a fragment present in some antivirals or an advanced intermediate thereof. We explored up to six different routes to afford the structure of interest. The more interesting has been an approach based on unprecedented stereoselective double alkylation of N-(diphenylmethylene)glycine tert-butyl ester under phase transfer conditions. We managed to reach the desired structure with a global yield of 29%. Despite the good results, this approach seems not competitive with report to the previous method described in the literature because, although it has better yields and contains easily improved stages in the scaling processes, the starting materials are more expensive. In the second part of this work we have faced the goal of obtaining a compound used in the treatment of heart failure, based on an alkylation reaction with a pseudoephedrine derivative amide as a key step of the synthesis. We got the structure of interest in five stages and an overal yield of 14%, but the key stage of the process gave erratic results and discrete diastereomeric excesses . These results could be improved if it carried out an exhaustive study of the alkylation reaction. Thus, the synthesis proposed would probably be competitive with respect to the results that have so far described in the literature. In general, we can say that in the two parts of this work has reached the desired products through new processes, yields range from those described in the literature. However, the industrial viability is not assured and deeper studies and some optimization stages are needed.

[cat] En el present treball es descriu la millora més important realitzada en el procés de la síntesi de les formidolides B i C. Aquesta millora es basa en un canvi d'estratègia per sintetitzar el nucli macrocíclic d'aquesta família de compostos. Els canvis principals han estat la utilització d'una metodologia diastereoselectiva per a la formació del nucli de tetrahidrofuran (THF), l'addició d'un al·lilstannà a un aldehid per donar exclusivament l'isòmer desitjat (Z)-1,5-anti i la millora en les condicions finals de ciclació. Tot això va suposar multiplicar per deu el rendiment descrit anteriorment., [eng] This paper describes the most important optimization carried out in the phormidolides B and C synthetic process. It consists in a change of strategy in order to synthesize the macrocyclic core of this family of compounds. The main changes have been the use of a diastereoselective methodology to construct the tetrahydrofuran (THF) core, a (Z)-1,5-anti allylstannane addition where the synthesis converges, and a notable improvement in the cyclization conditions. All this work has supposed a 10-fold increment in the overall yield of the synthesis.

[spa] El estudio trata de la síntesis y la actividad biológica de 4 antitumorales diferentes: Análogos de tetrahidro-[1,4]-dioxanisoquinolinas, de combretastatina A-4, de dioxancarbazol y del resveratrol. El estudio describe la estrategia sintética y la síntesis multietapas de esos compuestos potencialmente antitumorales. Varias tetrahidroisoquinolinas fueron sintetizadas, cinco de ellas fueron probadas biológicamente y presentan una actividad de inhibición de la proteína K-Ras prometedora. Dos de esos compuestos presentan una actividad anti antiangiogénica elevada y uno presenta actividades de antiosteoporosis. Dos derivados de pirazolona fueron sintetizados y probados biológicamente, uno de ellos muestra una inhibición muy elevada de la K-Ras, de antiangiogénesis y de antiosteoporosis e inhibe la fase G2 del ciclo celular y sus subfases. Se han preparado dos derivados de azolediona combretastatina A-4 que se evaluarán biológicamente. Se sintetizaron dos derivados de dioxancarbazol, uno de ellos presenta una actividad alta de la K-Ras y de antiangiogénesis. Finalmente, se prepararon y evaluaron biológicamente cuatros análogos del resveratrol. Los resultados biológicos muestran una inhibición moderada y selectiva de la K-ras en las líneas celulares estudiadas. Próximamente, el resto de los compuestos sintetizados se evaluarán biológicamente. Ensayos adicionales de inhibición de CDKs, de topoisomerasa II y de toxicidad serán llevados a cabo., [eng] The study deals with the synthesis and biological activity of 4 different antitumours: Tetrahydro-[1,4]-dioxanisoquinolines, combretastatin A-4, dioxancarbazoles and resveratrol analogues. This study describes the synthesis strategy and multi-step synthesis of these potential antitumour compounds. Various tetrahydroisoquinolines were synthesized, five of which were biologically tested and have a promising K-Ras inhibition activity. Two of them show a high antiangiogenesis activity, one of which also presents antiosteoporosis properties. Two pirazolone derivatives were synthetized and biologically tested, one of which shows a very high K-Ras inhibition, antiangiogenesis and antiosteoporosis activity and inhibits the G2 cell cycle phases and subphases. Two azoledione combretastatin A-4 derivatives were also synthesized and will be biologically tested. Two dioxancarbazoles were prepared and biologically tested, one of which shows a high K-Ras and angiogenesis inhibition. Finally, four resveratrol analogues were synthesized and biologically tested. Their biological results show a moderate and selective K-Ras inhibition on the studied cell lines. In the future, the rest of the synthesized compounds are to be biologically tested. Further CDKs and topoisomerase II inhibition trials as well as toxicity studies will be carried out.

[spa] Los fármacos derivados de nucleósidos son ampliamente utilizados en el tratamiento de enfermedades hematológicas, infecciones virales y enfermedades autoinmunes o inflamatorias. Estos fármacos necesitan de transportadores para ser internalizados en la célula y así, ser activos. Los transportadores implicados en la internalización de los nucleósidos y sus derivados son: los transportadores de nucleósidos concentrativos (CNTs) codificados por SLC28 y, los transportadores de nucleósidos equilibrativos (ENTs) codificados por SLC29. Otros transportadores, como los transportadores de cationes orgánicos (OCTs) codificados por SLC22, están implicados también en el transporte de algunos análogos usados en el tratamiento del SIDA. Los transportadores de nucleósdios y hOCT1 han sido detectados en células de sistema inmune, así como también, en epitelio polarizado donde se encuentran distribuidos asimétricamente mediando un flujo neto de nucleósidos y de sus derivados. Los transportadores de membrana pueden presentar polimorfismos alterando su habilidad de interacción con el sustrato. De todos los transportadores estudiados, hOCT1 es el más polimórfico, estando relacionadas sus variantes con la farmacocinética y farmacodinámica de algunos fármacos, como la metformina. En esta tesis mostramos la importancia que tienen estas variantes polimórficas, no solamente en la internalización de lamivudina, sino también en las interacciones de ésta con otros fármacos co-administrados. La infección de PBMCs con VIH mostró un aumento en la expresión de hOCT1, convirtiéndolos en mejor diana. Se descartó también la posibilidad de que los polimorfismos localizados en el loop extracelular, pudieran afectar la oligomerización del transportador. hOCT1 se ha identificado también como transportador de bendamustina, observándose a su vez, que sus variantes polimórficas modificaban la sensibilidad al fármaco. La sensibilidad de células de CLL ex vivo puede ser debida, en parte, a las variantes polimórficas que presentan. En el caso de los inhibidores de la ADN metiltransferasa (DNMT), observamos que los transportadores de nucleósidos son los responsables de la captación de zebularina pero no de decitabina, mediando un flujo de esta apical-basolateral en un modelo de epitelio polarizado. hOCT1 es capaz de mediar la extrusión de zebularina, en cambio, no lo son las variantes polimórficas estudiadas. Estos resultados estarían indicando que los polimorfismos serían responsables de la sensibilidad al tratamiento con zebularina. Para poder entender mejor la interacción transportador-sustrato, se generó un modelo preliminar de hCNT3 basado en el cristal de CNT de V. cholerae. En resumen, esta tesis muestra la importancia que tienen los transportadores y sus variantes polimórficas, en la biodisponibilidad y acción de los fármacos., [eng] Nucleoside-derived drugs are used for the treatment of haematological malignancies, viral infections and autoimmune or inflammatory diseases. These drugs require membrane transporters to be internalised and be active. Transporters implicated in nucleoside and nucleoside derivatives internalisation are concentrative nucleoside transporters (CNTs) encoded by SLC28 and equilibrative nucleoside transporters (ENTs) encoded by SLC29. Other transporters such as organic cation transporters (OCTs) encoded by SLC22 have been implicated in the transport of some antiviral nucleoside derivatives, too. Nucleoside transporters as well as hOCT1 have been detected in immune cells, as well as in polarised epithelia where they are asymmetrically distributed mediating a net flux of the natural nucleoside or the derivative. Membrane transporters can present polymorphic variants which can alter its ability to interact with the substrate. Of all studied transporters, hOCT1 is the most polymorphic, being its polymorphic variants implicated in pharmacokinetics and pharmacodynamics of some drugs used in clinics such as metformine among others. In this thesis we show the importance polymorphic variants of hOCT1 have not only in lamivudine uptake but also in drug-drug interactions with other drugs co-administrated with lamivudine. Infected PBMCs showed an up-regulation of hOCT1 expression which would made it a better lamivudine target. We also discard the possibility that polymorphisms located at the extracellular loop might affect oligomerization. hOCT1 has been also identified as a bendamustine transporter and hOCT1 polymorphic variants modulated bendamustine sensitivity. Bendamustine sensitivity in CLL cells ex vivo can partially be explained by the hOCT1 polymorphic variants they are carrying. In case of DNMT inhibitors, nucleoside transporters are responsible for zebularine internalisation but not decitabine and mediate a apical-basolateral flux in polarised epithelia. hOCT1 can mediate zebularine efflux which is impaired by the presence of polymorphic variants. In this case polymorphic variants would confer sensitivity to zebularine treatment. In order to better understand the transporter-substrates interaction a hCNT3 model has been generated based on the recent crystallised V.cholerae nucleoside transporter. This model has been preliminary proved experimentally showing, so far, to be a good model. In addition to its structural role in cell membranes, these lipids have also crucial functions in signal transduction and cell regulation. As a consequence of the important biological roles of sphingolipids, the synthesis of their analogs and the development of chemical inhibitors of sphingolipid enzymes are the object of current interest. Furthermore, it has been reported that slight modifications on the natural C1 (phyto)sphingosine scaffold or variations in its chain lengths can potentially alter the role and bioactivity of sphingolipid analogs. On the other hand, cyclitols are an important group of compounds due to their remarkable biological activities as well as their synthetic usefulness in the synthesis of other natural compounds or pharmaceuticals. Specially, the development of carbohydrate mimetics prompted primarily by their properties as glycosidase inhibitors, has led to a wide variety of novel structures by themselves or when considering them as key synthetic precursors of more complex molecules such as glycosphingolipids. For the above‐mentioned reasons, we developed a synthetic methodology towards sphingolipid analogs based on the regioselective ring‐opening reactions of N‐acylaziridines with different nucleophiles such as thiols or beta‐glycosil thiols, amines, phosphate derivatives or phosphorothioate derivatives. This approach is a short and flexible route for the synthesis of a variety of (phyto)sphingolipid analogs from aziridine derivatives and this method leads to molecules with different groups bonded to the C‐1 position of the sphingoid backbone from common intermediate aziridines by changing the nucleophile in the ring‐opening reaction step. Moreover, the diversity of compounds obtained is extended by introducing different N‐acyl groups at the nitrogen aziridine. Once, the desired sphingolipid analogs were obtained, a representative selection of them was tested in enzyme assays to identify inhibitors of mammalian sphingolipid metabolism enzymes, such as sphingomyelin synthase (SMS) and glucosylceramide synthase (GCS). Some of the tested analogs were potent inhibitors of GCS and none of them inhibited SMS at concentrations relevant for activity. In general, the inhibitory activity was higher for phytosphingosine analogs than their corresponding sphingosine analogs and the presence of the double bond between C4 and C5 of the sphingosine backbone in analogs was important for their bioactivity, since the dihydroceramide analogs were less active. In addition, a collection of (phyto)sphingolipid analogs were tested in an assay to identify inhibitors against yeast inositol phosphorylceramide synthase (IPCS). Generally, the inhibitory activities were lower than the inhibitions against GCS. However, one analog depicted selectivity against baker’s yeast IPCS since no inhibition of mammalian SMS and GCS was observed. For that reason, this analog showed potential properties as antifungal agent. Some (phyto)sphingolipid analogs were also tested as antigens for CD1d‐restricted iNKT cells and some of them were active, although this activity was lower when comparing with the endogenous glycolipid beta‐glucosylceramide. In particular, the most active compound was a nonglycosidic phytoceramide analog, but some glycosphingolipid analogs were also active in the activation of iNKT cells. However, these compounds were considered weak antigens. Finally, in order to obtain glycolipid analogs a synthetic methodology to obtain enantio‐ and diastereomerically pure galacto‐configured aziridines based on olefin aziridination reactions was examined. This consists in the use of nitrene precursors and a galactose related cyclohexene. The results obtained with this method showed potential for the preparation of the aziridine compounds., [spa] Los esfingolípidos (SL) son biomoléculas esenciales para las funciones fisiológicas de las células. El metabolismo anómalo o la expresión de SL específicos o glicoesfingolípidos (GSL) es el origen de diferentes enfermedades como las esfingolipidosis, causadas por la presencia de mutaciones en las enzimas que intervienen en la biosíntesis de SL o GSL. Debido a la importancia biológica de los SL, el desarrollo de inhibidores químicos de las enzimas que intervienen en el metabolismo de los SL de mamíferos y hongos, es un tema de gran interés científico. Por otro lado, cabe remarcar que los ciclitoles son un tipo de compuestos muy interesantes debido a su remarcable actividad biológica y a su utilidad como precursores sintéticos de moléculas más complejas como los GSL. Teniendo en cuenta las consideraciones comentadas anteriormente, se ha desarrollado una nueva metodología sintética para la obtención de análogos de SLs basada en las reacciones de apertura de Nacilaziridinas con diferentes nucleófilos como tioles, beta‐glicosil tioles, aminas, fosfatos y fosforotioatos. Esta aproximación es una ruta corta y flexible para la obtención de análogos lipídicos con diversidad estructural. Una selección representativa de los compuestos sintetizados se ensayó frente a la inhibición de enzimas del metabolismo de los mamíferos tales como la esfingomielin sintasa (SMS) y la glucosilceramida sintasa (GCS), y enzimas del metabolismo de los hongos como la inositol fosforilceramida sintasa (IPCS). Algunos de estos análogos resultaron ser inhibidores potentes de la GCS pero ninguno de ellos fue inhibidor de la SMS. Mientras que la inhibición frente a la IPCS en levaduras fue más baja en comparación con la inhibición de la GCS. A pesar de ello, uno de los análogos ensayados mostró selectividad frente a la IPCS ya que no se observó inhibición de la SMS y la GCS de mamíferos, mostrando así propiedades interesantes como agente antifúngico. De manera adicional, algunos análogos de (fito)esfingolípidos fueron ensayados como antígenos de células iNKT restringidas por la proteína CD1d y resultaron activos. Curiosamente, el compuesto más activo resultó ser un análogo de fitoceramida no glicosídico, aunque otros GSL también resultaron activos. Por otro lado, se desarrolló una metodología sintética para obtener aziridinas con configuración galacto (análogos de galactosa) enantiomericamente y diastereomericamente puras para obtener análogos de glicolípidos.

The reaction of a series of 1,2-diols with thionyl chloride led to bisnoradamantane sulfites in very good yields. The reaction has also been applied to related polycyclic scaffolds. The compounds have been tested for antiviral activity but none of them showed to be active. [cat] La present Tesi Doctoral es situa en el context de l’estudi d’estratègies sintètiques per a la formació d’anells en l’àrea heterocíclica. En particular, s’han avaluat les possibilitats sintètiques de la combinació de dues reaccions d’eficàcia prou reconeguda per a la formació d’enllaços carboni-carboni, la reacció de RCM i la reacció intramolecular de Heck d’halurs vinílics, per a la construcció de les estructures policíliques amb pont característiques dels alcaloides ervitsina i aparicina, així com també de la unitat superior de la vinorelbina. Els principals aconseguiments del treball i les conclusions més significatives que se’n deriven s’exposen a continuació: • La reacció de RCM de diens indòlics ha mostrat ser un bon procediment per a la formació d’anells de 8 baules fusionats amb el nucli indòlic. L’eficiència de la ciclació, combinada amb la facilitat de preparació dels substrats diènics a partir de compostos indòlics senzills, determina que l’estratègia pugui ser emprada com a via d’accés a les subestructures tricícliques tant de l’alcaloide aparicina com de la unitat superior de la vinorelbina. • L’aminació reductora d’indole-3-carbaldehids amb alquenilamines permet instal•lar cadenes d’alquenilaminometil a la posició 3 de l’heterocicle, d’interès per la preparació dels substrats diènics precursors de diversos azacicles per RCM. En el cas particular de 2-vinil-3-indolecarbaldehids, el procés d’aminació reductora no és compatible amb la presència d’un grup fenilsulfonil a l’àtom de nitrogen indòlic, ja que en comptes d’obtenir-se la corresponent amina secundària, s’observa la formació ràpida del nucli de tetrahidro-γ-carbolina. L’anulació és el resultat de l’addició conjugada de l’amina primària a l’agrupació d’aldehid γ, δ insaturat del substrat, seguida d’un procés d’aminació reductora intramolecular. • La metodologia de doble anulació RCM-Heck possibilita l’accés al sistema amb pont de 2-azabiciclo[4.3.1]decà característic de l’alcaloide ervitsina. En concret, la reacció de RCM quimioselectiva d’un triè indòlic genera un ciclohepta[b]indole, amb un doble enllaç adequat per al tancament subsegüent de l’anell de piperidina mitjançant un acoblament intramolecular de Heck amb l’halur vinílic, connectat al sistema anular a través de l’àtom de nitrogen. Si bé s’ha accedit al sistema tetracíclic de l’alcaloide que incorpora el substituent E-etilidè a la posició 20, l’estratègia no s’ha pogut estendre a la preparació de compostos tetracíclics funcionalitzats a la posició 3. • S’ha dut a terme la primera síntesi total de l’alcaloide aparicina mitjançant una via sintètica concisa que implica com a etapas clau: (i) la reacció de RCM d’un diè indòlic per construir l’anell central de 8 baules; (ii) la isomerització promoguda per base del doble enllaç resultant de la ciclació, amb desprotecció concomitant de l’àtom de nitrogen indòlic; i (iii) la ciclació de Heck d’un halur vinílic per completar la formació de l’esquelet amb pont d’1-azabiciclo[4.2.2]decà i al mateix temps introduïr els dos dobles enllaços exocíclics de l’alcaloide. • La inserció de l’agrupació de 2-etilpropè entre l’àtom de nitrogen alifàtic i la posició 4 d’un azocino[4,3-b]indole mitjançant una seqüència d’N-alquilació seguida de ciclació de Heck permet la formació del sistema amb pont d’1-azabiciclo[5.3.1]undecà característic de la unitat superior de la vinorelbina. Aquest resultat, juntament amb l’obtingut en el context de la síntesi de l’aparicina, posa de manifest la validesa dels acoblaments intramoleculars de Heck en els quals intervenen dobles enllaços inclosos en anells mitjans., [eng] The first part of this thesis work focus attention to: • The new synthetic route to the azocino[4,3-b]indole system relying on RCM of 2-allyl-3-(allyl-aminomethyl)indoles. The efficiency of the cyclization combined with the easy preparation of the dienic precursors from simple indolic derivates make this strategy attractive for the construccion of medium-sized indolo 2,3-fused carbo- and azacycles, which are scaffolds found in many bioactive compounds. • Our efforts in getting more azepinoindole systems leads to tetrahydro γ-carbolines. The reaction of N-(phenilsulfonyl)-2-vynil-3-indolecarbaldehydes with primary amines under mild reductive amination conditions leads to tetrahydro-γ-carbolines. The process can be suppressed by changing the protecting group at the indole nitrogen for a methoxymethil group, thus allowing the preparation of RCM substrates for azepinoindole synthesis. In the second part: • An efficient approach to the bridged framework of the indole alkaloid ervitsine have been done. The RCM reaction from a 2,3-disubstituted indole followed by a vynil halide Heck cyclazation upon the resulting cycloheptene ring gives short access to the Ervitsine alkaloid skeleton. • The first total synthesis of (±)-apparicine has been accomplished by a concise route employing a vynil halide Heck cyclization to close the bridged piperidine ring in the last synthetic step. • An efficient approach to the high subunit of Vinorelbin has been done. The subunit has been successfully assembled developing an indole-templated RMC followed by an intramolecular Heck cyclization.

The first total synthesis of the indole alkaloids (+/-)-aplicyanins A, B, and E, plus 17 analogues, all in racemic form, is reported. Modifications to the parent compound included changing the number of bromine substituents on the indole, the nature of the substituents on the indole nitrogen (H, Me, or OMe), and/or the oxidation level of the heterocyclic core tetrahydropyrimidine. Each compound was screened against three human tumor cell lines, and 14 of the newly synthesized compounds showed considerable cytotoxicity. The assay results were used to establish structure-activity relationships. These results suggest that the presence of the bromine at position 5 of the indole is critical to activity, as well as the acetyl group on the imine nitrogen does in some compounds.

Els compostos policíclics d'elevada simetria, a més del seu gran atractiu estètic i interès estructural, són substrats valuosos per a estudis mecanístics o bé com a intermedis en la síntesi de molècules orgàniques més complexes.Des de fa més de vint anys el grup d'investigació del Prof. Pelayo Camps del Departament de Famacologia i Química Terapèutica de la Universitat de Barcelona treballa en la síntesi i en la reactivitat de compostos policíclics. Concretament, en la present Tesi Doctoral s'ha dut a terme l'obtenció de l'1,3,5,7-tetraiodotriciclo[3.3.0.0/3,7]octà, un derivat bisnoradamantànic polisubstituït en les posicions cap de pont, com a precursor del compost triciclo[3.3.0.0/3,7]octa-1(5),3(7)-diè, que contindria dos dobles enllaços altament piramidalitzats perpendiculars entre si, com a un intermedi sintètic per a la preparació de compostos policíclics més complexes. Malauradament, tots els intents duts a terme per a obtenir aquest intermedi tant inestable van resultar infructuosos, degut probablement a la formació d'un enllaç intern a la molècula conduint a la formació d'un derivat [2.2.1]propel·lànic contingut dins l'esquelet de bisnoradamantà, extremadament reactiu, que conté dos àtoms de carboni amb geometria invertida. Per a verificar la formació d'aquest intermedi propel·lànic, es va dur a terme l'obtenció del (±)-1,3-diiodotriciclo[3.3.0.0/3,7]octà, un derivat bisnoradamantànic 1,3-disubstituït en les posicions cap de pont, a partir del qual mitjançant reaccions amb diferents agents reductors s'ha generat l'1,3-deshidrobisnoradamantà, un [2.2.1]propel·là amb un pont metilènic entre les posicions 2 i 5, altament reactiu, que en les condicions de les reaccions realitzades evolucionaria cap a diversos productes, molts dels quals han estat aïllats i plenament caracteritzats. Els càlculs teòrics DFT realitzats suggereixen també la formació d'aquest intermedi. Paral·lelament, s'ha sintetitzat una petita col·lecció de derivats bisnoradamantànics enantiopurs di- i tetrasubstituïts en les posicions cap de pont utilitzat l'(R)-N-fenilpantolactama, un auxiliar quiral desenvolupat en el nostre grup d'investigació. Aquests compostos són els primers exemples descrits de bisnoradamantans quirals enantiopurs que tenen la seva quiralitat a la diferent substitució en les posicions cap de pont. La puresa enantiomèrica dels compostos obtinguts s'ha determinat utilitzat CG quiral/EM en alguns d'aquests compostos., Polycyclic cage compounds are a unique class of organic compounds. They are useful substrates for mechanistic studies as well as intermediates in the synthesis of more complex organic molecules.For more than twenty years Prof. Pelayo Camps' research group at the Departament de Farmacologia i Química Terapèutica of the Universitat de Barcelona has been working on the synthesis and reactivity of polycyclic compunds. Specifically, in this Thesis the syntheses of 1,3,5,7-tetraiodotricyclo[3.3.0.0/3,7]octane was carried out, a bridgehead tetrasubstituted bisnoradamantane derivative, as a potential precursor of the tricyclo[3.3.0.0/3,7]oct-1(5),3(7)-diene. This compound, that features two highly pyramidalized carbon-carbon double bonds, may be useful as an intermediate in the syntheses of more complex polycyclic compounds. Unfortunately, we have no evidences in favor of the formation of this intermediate, probably because a very reactive [2.2.1]propellane derivative is generated. This intermediate has two carbon atoms with inverted geometries. To verify the formation of this propellane intermediate, the synthesis of (±)-1,3-diiodotricyclo[3.3.0.0/3,7]octane was carried out, a bridgehead 1,3-disubstituted bisnoradamantane derivative. From the reaction of this compound with different reductors evidences for the generation of 1,3-dehydrobisnoradamantane, a bridged, highly distorted and very reactive [2.2.1]propellane were found. Theoretical calculations also support the intermediacy of this derivative. On the other hand, a series of enantiomeric bridgehead di- and tetrasubstituted bisnoradamantane derivatives have been prepared by using (R)-N-phenylpantolactam, a chiral auxiliary developed by our research group. Although several bridgehead substituted chiral bisnoradamantane were known, efforts directed towards their preparation as single enantiomers had not been previously described. The enantiopurity of these compounds was established by chiral GC/MS.

La present Tesi doctoral s'emmarca dins d'un projecte de síntesi de productes naturals azapolicíclics.En el decurs de la mateixa, s'han desenvolupat diverses metodologies per tal d'assolir la síntesi d'1-azaspiro[4.5]dec-6-en-8-ones polifuncionalitzades de forma racèmica i enantiopura. Aquests sistemes espirànics s'han sintetitzat per reacció aldòlica d'un producte provinent del trencament oxidatiu d'un 1-azaspiro[4.4]noné enantiomèricament pur o d'un producte provinent de l'alquilació d'una prolina modificada. Alternativament també s'han assolit aquests sistemes per deshidrogenació de les corresponents cetones.La reacció més destacable en la ruta que ens ha conduit a la síntesi enantioselectiva de les d'1-azaspiro[4.5]dec-6-en-8-ones, és la reacció d'inserció d'un carbè alquilidènic a un enllaç C-H en posició 5. L'excés enantiomèric es pugué determinar per HPLC quiral. A partir d'aquestst sistemes d'1-azaspiro[4.5]decà amb els dos metilens veïns al carbonil diferenciats per la presència d'un doble enllaç, s'ha estudiat la seva utilitat sintètica en la preparació dels sistemes azatricíclics de l'FR901483 de manera enantio i regioselectiva. Tots els intents per a assolir el tancament de l'anell piperidínic de l'FR901483 utilitzant substrats aminats resultaren infructuosos llevat del que implica l'acoblament d'halurs de vinil sobre enolats de cetona catalitzada per pal·ladi malgrat aquesta no fou quimioselectiva ni conseqüentment regioselectiva. La raó de la dificultat en aquest procés probablement resideix en la dificultat de generar espècies electrofíliques en la posició  d'amines, per la seva inestabilitat a causa de la participació del nitrogen en processos col·laterals ja sigui de desalquilació, formació d'aziridines, descarbonilacions, etc. A les reaccions que impliquen l'ús d'una amida encara que aquestes no presenten el problema d'inestabilitat, apareix el problema dels possibles rotàmers que junt amb la conformació inicial que potser no adequada per experimentar un procés de ciclació malbarata els intents d'accés al sistema azatricíclic de l'FR901483. Solament la ciclació radicalària de tricloroacetamides com a proradicals utilitzant sililenolèters com a acceptors radicalaris permeté l'aïllament de compostos amb el nucli azatricíclic de l'FR901483, encara que el seu baix rendiment inhabilità el seu ús en el procés de síntesi total.Per altra banda, ens hem plantejat l'accés al nucli azatricíclic de les cilindricines que també contenen el sistema d'1-azaspiro[4.5]decà de referència sintetitzant en primera instància el sistema de decahidroquinolina fusionada en cis i posteriorment el sistema espirànic comentat.En aquest sentit, s'ha implantat una nova metodologia sintètica succinta i versàtil per a la preparació de decahidroquinolines i octahidroindoles funcionalitzats basada en la iodoaminociclació de bishomoal·lilamines. La reacción de 2-al·lil-N-benzilciclohexilamines amb NIS permet l'accés a 3-iododecahidroquinolines que per tractament amb alúmina condueixen a 3-hidroxidecahidroquinolines i/o 2-hidroximetiloctahidroindoles amb retenció de la configuració. S'observà que les 3-iododecahidroquinolines, obtingudes per iodoaminociclació de 2-al.lil-N-benzilciclohexilamines tenen una reactivitat diferent segons presenten una fusió cis o trans. Així, els derivats cis endo al tractar-los amb Al2O3 formen fàcilment sals d'aziridini que en un procés de control cinètic proporcionen octahidroindoles en un procés de contracció anular. Els derivats trans també formen sals d'aziridini, però en aquestes estructures el bescanvi del iode per hidroxil transcorre mantenint-se l'estructura de decahidroquinolina i la conseqüent retenció de la configuració relativa. En condicions de control termodinàmic, l'obertura de les sals d'aziridini provenint de cis-decahidroquinolines canvia de regioselectivitat i dóna lloc a cis-decahidroquinolines amb bescanvi funcional.Aquesta estratègia desenvolupada a l'última part de la tesi, obre una nova ruta sintètica per accedir als sistemes tricíclics de les ciclindricines i la fasicularina a través de les decahidroquinolines corresponents., The present thesis was realized as a part of a framework directed to the synthesis of azapolicyclic natural products, namely the immunosupressant FR901483 and the marine alkaloids cylindricines.During its development, we succesfully studied several methodologies to achieve the syntheses of polifunctionalized racemic and enantiopure 1-azaspiro[4.5]dec-6-en-8-ones. Afterwards, we studied the utility of these advanced intermediates in the synthesis of the azatricyclic system present in the immunosupressant FR901483 or in the one embodying cylindricines' skeleton. The most considerable results were obtained with the palladium catalized reaction of vinyl halides over ketone enolate. Also, radical reaction of trichloroacetamides as proradicals on sylylenolethers as radical acceptors was significant.In the last part, a new versatile and brief methodology on the iodoaminocyclation of bishomolylamines for the preparation of decahydroquinolines and octahydroindoles was achieved. The reaction of 2-allyl-N-benzylcyclohexilamines with NIS makes possible an entry to 3-iododecahydroquinolines which on treatment with alumina lead to 3-hydroxydecahydroquinolines and/or 2-hydroxymethyloctahydroindoles with retention of the configuration.This strategy opens a new synthetic route to the tricyclic system of fasicularin and cylindricines via the corresponding decahydroquinolines.

La importància de la quiralitat és ben coneguda principalment en relació al fet que la major part dels productes bioactius naturals són quirals i que les seves propietats fisiològiques i farmacològiques depenen del seu reconeixement per part de macromolècules diana quirals, les quals interaccionaran només amb l'enantiòmer del producte bioactiu d'una configuració absoluta determinada.Anàlogament, en el cas de fàrmacs quirals se solen observar diferències importants d'activitat entre els dos enantiòmers. L'enantiòmer més actiu d'un fàrmac quiral es denomina eutòmer i el menys actiu distòmer.Així, la legislació farmacèutica internacional és cada cop més restrictiva respecte a l'ús de fàrmacs com a barreges racèmiques. Realment, la utilització de fàrmacs enantiopurs és actualment un requisit fonamental per a pràcticament totes les indústries farmacèutiques. Així doncs, el desenvolupament de nous mètodes sintètics per obtenir compostos enantiopurs s'ha convertit en un punt clau per a la indústria farmacèutica. La síntesi asimètrica constitueix una de les principals estratègies per tenir accés a compostos enantiopurs, utilitzant auxiliars, reactius o catalitzadors quirals.Tenint en compte els excel·lents resultats dels auxiliars quirals (R)- i (S)-2a en reaccions de desracemització, resolució cinètica dinàmica en la reacció d'ALFA-haloèsters amb diversos nucleòfils i en reaccions de Diels-Alder asimètriques ens vam plantejar: 1. Posar a punt un procediment per obtenir noves pantolactames en forma racèmica, a partir de les quals es podrien preparar els corresponents enantiòmers pel procediment descrit per a (R)- i (S)-2a. 2. L'aplicació de l'(R)- o (S)-N-fenilpantolactama, (R)- o (S)-2a o d'alguna de les noves pantolactames en noves reaccions de desracemització d'àcids carboxílics alfa,alfa-disubstituïts com a etapes clau per a la síntesi en forma enantiopura de compostos amb potencial activitat farmacològica o de fàrmacs establerts com àcids alfa-aril-gamma-aminobutírics anàlegs del GABA (àcid gamma-aminobutíric) o l'antiepilèptic levetiracetam. 3. L'aplicació de l'(R)- o (S)-N-fenilpantolactama, (R)- o (S)-2a, o d'alguna de les noves pantolactames a addicions de Michael diastereoselectives amb acceptors contenint l'auxiliar quiral, com a etapa clau per a la síntesi dels dos enantiòmers del fàrmac baclofè.4. L'aplicació de l'(R)- o (S)-N-fenilpantolactama, (R)- o (S)-2a, o d'alguna de les noves pantolactames a cicloaddicions 1,3-dipolars diastereoselectives amb dipolaròfils contenint l'auxiliar quiral, com a etapa clau per a la síntesi en forma enantiopura d'N,O-psiconucleòsids amb potencial activitat antivírica., We have developed two alternative one-pot procedures for the preparation of N-substituted pantolactams in low to medium yields starting from pantolactone and primary amines under acid catalysis. In one procedure the reaction is performed in a pressure reactor at 250 ºC for 4h, while in the other one the reaction takes place in minutes under microwave irradiation. alpha-aryl-substituted GABA derivatives (R)- and (S)-23, 26 and 27 of potential biological interest have been prepared in enantiopure form (ee > 99%) from the readily available methyl arylacetates 16, 31 and 37, through a seven-step synthetic sequence, involving as the key step the deracemization of cyano acids ()-18, 33 and 39 using (R)- or (S)-N-phenylpantolactam as chiral auxiliaries. In sharp contrast with the reported absolute lack of efficiency of (R)-pantolactone as chiral auxiliary in the deracemization of this kind of cyano acids, the use of (R)- and (S)-N-phenylpantolactam led to the corresponding pantolactam esters in almost quantitative yields, with high levels of diastereoselectivity (dr up to 93:7), and with predictable sense of asymmetric induction. Moreover, pantolactam esters in hihgly diastereoenriched form (dr > 98:2 or 97:3) were obtained by column chromatography of the diastereoenriched crude materials resulting from the deracemization reactions. Hydrolysis of these esters under essentially nonracemizing conditions, followed by recrystallization of the resulting crude cyano acids or their cyclohexylammonium salts, and subsequent reisolation of the carboxylic acids in the last cases, led to enantiopure alpha-aryl-beta-cyano acids (R)- and (S)-18, 33 and 39, which were transformed in good yields into the N-Boc-alpha-aryl-substituted GABA derivatives (R)- and (S)-23, 26 and 27 through a standard procedure.We have developed a synthesis of levetiracetam by deracemization of racemic 2-bromobutyric acid 50 to the (R)-enantiomer with (S)-N-phenylpantolactam, followed by nucleophilic substitution with the sodium salt of 2-pyrrolidinone to give (S)-2-(2-oxopyrrolidin-1-yl)butyric acid, a known precursor of levetiracetam. The key points of the synthesis are: (1) the good diastereomeric ratio (9:1) in the esterification of ()-50 with the chiral auxiliary and the easy purification of the main diastereomer by column chromatography, (2) the LiOH/H2O2 hydrolysis of the diastereopure ester to give acid (R)-50 without racemization and (3) the SN2 transformation of (R)-50 to the immediate acid precursor of levetiracetam. This sequence has also been applied to the preparation of the enantiomer of levetiracetam.(R)- and (S)-baclofen hydrochloride have been prepared in enantiopure form (>99% ee) from the readily available racemic nitro acid (±)-71, through a three-step synthetic sequence, involving the resolution of the above acid via diastereomeric esters derived from the chiral auxiliaries (R)- and (S)-N-phenylpantolactam, (R)- and (S)-2a, followed by hydrolysis and hydrogenation (Ra-Ni) of the nitro group. The chiral auxiliaries were conveniently recovered in enantiopure form after the hydrolysis step.Finally, we have developed a synthesis of N,O-psiconucleosides involving as the key step the 1,3-dipolar cycloaddition of phenylnitrone and methyl phenyltioacrilate. The key points are: (1) esterification with the resolution agent, (R)- and (S)-N-phenylpantolactam, (R)- and (S)-2a , (2) nucleosidation with timine and 5-fluoruracile as nucleobases and the efficiently column cromatography to led the corresponding esters of pantolactam in diasteropure form (dr > 98:2), (3) reduction of transesterification products with Vitride and recovery of (R)- and (S)-N-phenylpantolactam with high yield and in enantiopure form (ee > 99%).

Els productes naturals són de gran importància en els sistemes de prevenció i de cura de les dolences i malalties de l'ésser humà. Així, un 50% dels fàrmacs actuals estan basats en productes naturals i un 80% de la població mundial depèn, encara avui, de la medicina tradicional. El procés de desenvolupament d'un nou fàrmac a partir d'un producte natural és llarg i complex. Les millores introduïdes recentment i de forma generalitzada a cadascuna de les etapes del procés han suposat una renovació de l'interès de la indústria farmacèutica per nous compostos naturals bioactius. La síntesi química té un paper rellevant en el procés de desenvolupament d'un nou fàrmac: la síntesi química del producte bioactiu permet assegurar la identitat del compost, així com assegurar la producció a gran escala per permetre la realització dels assaigs clínics i donar resposta a les demandes del mercat farmacèutic. La introducció de la química en fase sòlida ha estat una de les millores més importants en l'àrea de la síntesi química, permetent una exploració sintètica més ràpida i eficaç. Aquest canvi ha estat rellevant en la síntesi de pèptids i oligonucleòtids i, especialment determinant, en l'obtenció de depsipèptids cíclics produïts per microorganismes i d'interessant activitat i interès com a nous fàrmacs. Així, en la present tesi s'ha treballat en el desenvolupament de noves síntesis en fase sòlida de dos depsipèptids cíclics: la sirengotoxina i la tiocoralina, amb la finalitat de contribuir en la seva aplicació terapèutica.La sirengotoxina és un lipononadepsipèptid cíclic produït per una bactèria patògena dels arbres cítrics que ha mostrat una interessant activitat contra el paràsit de la Leishmania. En aquest projecte de fàrmac s'ha dissenyat un pèptid anàleg al pèptid natural que conserva els mateixos elements estructurals i la configuració de la cadena peptídico. S'ha obtingut mitjançant una estratègia compatible amb la síntesi del producte natural. En aquesta síntesi s'ha resolt l'obtenció de l'aminoàcid no proteïnogènic així com la seva correcta inserció a la cadena.La tiocoralina és un pèptid bicíclic simètric produït per un microorganisme marí aïllat del corall tou i que presenta una potent activitat com a antitumoral. En aquest projecte s'ha desenvolupat la síntesi d'un anàleg del pèptid natural mitjançant l'aproximació en fase sòlida. En aquest cas, també s'ha resolt la síntesi de dos aminoàcids no proteïnogènics, dues N-metil cisteïnes.Finalment, s'han aïllat nous agents antitumorals d'esponges marines recol·lectades a les costes australianes amb la finalitat d'obtenir nous projectes de fàrmacs així com per ampliar els coneixements en el procés de desenvolupament d'un nou fàrmac., "Natural Products as a source of new drugs: cyclic depsipeptides synthesis on solid-phase and isolation of antitumoral compounds from marine sponges"TEXT:Natural Products play an important role in the prevention and therapeutic system for human healthy. 50% of drugs in the market are based on natural products and 80% of world depends still from traditional medicine. The drug development process from natural sources is a long and complex way. Recently, several improvements in each step of this process have been determinant for a reinterest from pharmaceutical companies in these kinds of compounds.The chemical synthesis has a relevant role in the drug development process: it allows assuring about identity of compounds, obtaining the bioactive compound in large scale to develop the clinical trial and satisfy the pharmaceutical market demands. The introduction of solid-phase chemistry has been one of the most important improvements in chemical synthesis area because it allows a rapid and efficient exploration about strategic synthetic routes. It has been wide used in peptides and oligonucleotides synthesis and especially useful in cyclic depsipeptides produced by marine microorganism. In the present thesis, is described the development of solid-phase synthesis for two new cyclic depsipeptides, syrengotoxin and thiocoraline with the aim to contribute in its medical use.Syrengotoxin is a cyclic lipononadepsipeptide produced by a pathogen bacterium that shows interesting activity against Leishmania parasite. In this project, it has been designed a analogue peptide which preserve all structural features and the configuration of peptide chain respect natural peptide. The defined synthesis is compatible with syrengotoxin synthesis. It has been developed the non-natural amino acids presents in natural compound and its correct incorporation in the peptide chain.The thiocoraline is a symmetric biclyclic peptide which is produced by a marine microorganism and presents a potent activity as an antitumoral drug. In this project, it has been developed a solid-phase synthesis of a more resistant in plasma analogue. In this case it has been resolved the N-methyl non natural amino acids synthesis that are also present in natural compound.Lastly, it has been isolated new antitumoral compounds from marine sponges collected from south-east Australian coasts which the aim to obtain new drugs and improve the knowledge about drug development from natural sources.

Els 2-amino-1,2,3,4-tetrahidro-1-naftols poden considerar-se com anàlegs cíclics semirígids de les ariletanolamines adrenèrgiques, de les que deriven per incorporació de dos àtoms de carboni entre l'anell aromàtic i la posició alfa respecte a l'àtom de nitrogen.En el cas de la noradrenalina, o dels seus derivats N-alquilats, la modificació pot donar lloc a dos tipus d'estructures isòmeres no equivalents, segons quina sigui la posició "orto" de l'anell aromàtic que intervé en la formació del nou anell.Existeixen alguns precedents bibliogràfics de l'aplicació d'aquesta modificació a ariletanolarnines. Aixi, quan es va aplicar a la noradrenalina, o als seus derivats, s'obtingué, en un cas, un producte amb els hidroxils fenòlics situats en les posicions 6 i 7 que va resultar inactiu com a agonista beta-adrenèrgic. En canvi, els productes hidroxilats sobre les posicions 5 i 6 resultaren potents conlagonistes beta-adrenèrgics. Aquests productes estan mancats d'activitat alfa-adrenèrgica. En tots els casos es va veure que els productes d'estereoquímica "trans" eren molt més actius que els "eis".Per tal de determinar la importància de la posició dels grups hidroxil fenòlics per la interacció amb els receptors adrenèrgics, hem aplicat l'anterior modificació a fàrmacs agonistes monofenòlics, com són l'octopanlina i el metaraminol. Amb la síntesi i posterior assaig farrnacològic dels productes "1" a "6" pretenem trobar alguna relació que ens permeti deduir quina serà la disposició adoptada per l'anell aromàtic de la noradrenalina o de la isoprenalina en la interacció amb els receptors alfa i beta, respectivament.En tots els casos s'ha partit de la tetralona amb la substitució adient, obtinguda segons els mètodes ja descrits a la bibliografia. En una primera fase es va procedir a introduir l'àtom de nitrógeno; per fer-ho es disposava de dos mètodes diferents. El primer d'ells consisteix en la transposició denominada de Neber de l'O-tosilat d'oxima de la cetona de partida en medi alcalí d'etòxld potàssic. Després de la hidròlisi de l'azirina que resulta amb àcid clorhídric s'obté una alfa-aminocetona protegida en forma d'hidroclorur. L'altre mètode per a la introducció del nitrogen consisteix en la reacció de la cetona de partida amb un nitrit orgànic en rnedi alcalí de "t"-butòxid potàssic. Així s'obté una oximinocetona.Posteriorment es realitzen les modificacions funcionals convenients fins arribar als aminoalcohols desitjats. Els compostos isopropilats sobre el grup amino s'otingueren per tractament de l'amina primària amb acetona i posterior reducció amb borohidrur sòdic. Els productes finals de tipus fenòlic s'obtingueren per desprotecció dels èters benzílics per hidrogenòlisi en medi neutre a 200 psi de pressió d'hidrogen.La determinació de la configuració dels compostos es va realitzar en base a les dades de RMN. Quant a la conformació dels de l'estudi dels valors de constants d'acoblament protons de la zona alifàtica de la molècula es dedueix que el grup hidroxil es disposa pseudoequatorial i el grup amina equatorial. Aquesta disposició per als grups OH/NH permet l'establiment d'un pont d'hidrogen que estabilitza la conformació. La conformació coincideix amb l'obtinguda a partir d'estudis de difracció de raigs X per la isoprenalina.Com que no s'ha establert inequivocament si ariletanolamines i ariloxipropanolamines interaccionen de la mateixa manera amb el receptor beta, com a segon objectiu de la Tesi es dissenyen les estructures "41" i "42", combinación molecular del prenalterol (ariloxipropanolamina) amb el rimiterol (ariletanolamina) per tal d'estudiar si ambdós tipus de molècules estan sotmesos a les mateixes relacions estructura-activitat. RESULTATS D'ACTIVITAT FARMACOLÒGI('iAEls compostos "32" i "33" s'assajaren com a alfa i beta adrenèrgics en aorta, aurícula esquerra i tràquea de cobai, respectivament. Amb anterioritat als assaigs es reserpinitzaren els animals per evitar el possible efecte indrecte tant de partons com productes problema. Els valors d"activitat obtinguts semblen indicar que el neurotransmissor, la noradrenalina, adoptaria una conformació que disposés un dels hidroxils fenòlics oposat respecte al benzílic en interactuar el receptor beta. Per la interacción amb el receptor alfa, l'anell aromàtic del neurotransmissor hauria de girar 180°. Quant als productes assajats, assenyalen el guany en selectivitat que suposa la pèrdua de l'hidroxil en posició 6 respecte a productes dienòlics ja descrits en la bibliografia. El producte que pot presentar un interès més gran és el "33ª2, que podria constItuir-se en nou cap de sèrie per a l'obtenció de broncodilatadors exempts d'acció sobre el cor.Pel que fa als productes "41" i "42" s'assajaren només com a beta-adrenèrgics. Dels resultats sembla deduir-se que la modificació molecular és vàlida per.a les ariletanolamines, com el rimiterol, i que els comunica selectivitat beta-2, no és aplicable a les ariloxipropanolamines.D'altra banda, amb la síntesi de productes en què el grup hidroxil ocupa diferents posicions en l'anell aromàtic, es pretenia veure si la posició d'aquest és tan important com per als compostos de tipus aminotetrahidronaftol. Si Comparem les activitats dels compostos "eritro-41" i "eritro-42" s'observa la pèrdua d'activitat per canvi de posición del grup, però a més de ser poc significativa té caracter contrari al dels altres compostos en què l'important és l'hidroxil en "meta" respecte a la cadena. Per tant podem afirmar que l'anell aromàtic en ariletanolamines i ariloxipropanolamines agonistes no juga el mateix paper a l'hora d'interactuar amb el receptor beta.
