Abstract

Species of Diaporthe are considered important plant pathogens, saprobes, and endophytes on a wide range of plant hosts. Several species are well-known on grapevines, either as agents of pre- or post-harvest infections, including Phomopsis cane and leaf spot, cane bleaching, swelling arm and trunk cankers. In this study we explore the occurrence, diversity and pathogenicity of Diaporthe spp. associated with Vitis vinifera in major grape production areas of Europe and Israel, focusing on nurseries and vineyards. Surveys were conducted in Croatia, Czech Republic, France, Hungary, Israel, Italy, Spain and the UK. A total of 175 Diaporthe strains were isolated from asymptomatic and symptomatic shoots, branches and trunks. A multi-locus phylogeny was established based on five genomic loci (ITS, tef1, cal, his3 and tub2), and the morphological characters of the isolates were determined. Preliminary pathogenicity tests were performed on green grapevine shoots with representative isolates. The most commonly isolated species were D. eres and D. ampelina. Four new Diaporthe species described here as D. bohemiae, D. celeris, D. hispaniae and D. hungariae were found associated with affected vines. Pathogenicity tests revealed D. baccae, D. celeris, D. hispaniae and D. hungariae as pathogens of grapevines. No symptoms were caused by D. bohemiae. This study represents the first report of D. ambigua and D. baccae on grapevines in Europe. The present study improves our understanding of the species associated with several disease symptoms on V. vinifera plants, and provides useful information for effective disease management.

Abstract

Background: The fungal genus Aspergillus is of critical importance to humankind. Species include those with industrial applications, important pathogens of humans, animals and crops, a source of potent carcinogenic contaminants of food, and an important genetic model. The genome sequences of eight aspergilli have already been explored to investigate aspects of fungal biology, raising questions about evolution and specialization within this genus. Results: We have generated genome sequences for ten novel, highly diverse Aspergillus species and compared these in detail to sister and more distant genera. Comparative studies of key aspects of fungal biology, including primary and secondary metabolism, stress response, biomass degradation, and signal transduction, revealed both conservation and diversity among the species. Observed genomic differences were validated with experimental studies. This revealed several highlights, such as the potential for sex in asexual species, organic acid production genes being a key feature of black aspergilli, alternative approaches for degrading plant biomass, and indications for the genetic basis of stress response. A genome-wide phylogenetic analysis demonstrated in detail the relationship of the newly genome sequenced species with other aspergilli. Conclusions: Many aspects of biological differences between fungal species cannot be explained by current knowledge obtained from genome sequences. The comparative genomics and experimental study, presented here, allows for the first time a genus-wide view of the biological diversity of the aspergilli and in many, but not all, cases linked genome differences to phenotype. Insights gained could be exploited for biotechnological and medical applications of fungi. © 2017 The Author(s).

Abstract

This review summarises the current knowledge on the biochemical and physiological events that directly or indirectly alter the engagement of the cyanide-resistant alternative respiratory pathway in the cephalosporin C producer filamentous fungus Acremonium chrysogenum. Particular emphasis is placed on the role this activity plays in the overproduction of antibiotic, and also on the critical fermentation technology background that supports its operation.

Abstract

Methionine increased the intracellular glutathione (reduced) (GSH) pool and the specific gamma-glutamyltransferase (gamma-GT) activity in the cephalosporin C (CPC) producer Acremonium chrysogenum. The accelerated turnover of GSH might be indicative of the existence of a functioning gamma-glutamate cycle, and might supply the CPC biosynthetic machinery with L-cysteine. The gamma-GT was not subject to nitrogen metabolic repression but was more active in cells exposed to different oxidative-stress-generating agents. Exogenous cysteine hindered both the uptake of methionine and the induction of gamma-GT, and was not beneficial for CPC production. Abstract

Quinol oxidation inhibitors (QoIs) are one of the most important classes of fungicides used in agriculture. They block electron transfer between cytochrome b and cytochrome c1, thereby impeding the production of ATP via oxidative phosphorylation. QoI fungicides are generally at high risk of provoking resistance in fungal phytopathogens. Resistance has been reported in more than thirty species, amongst others, in Botrytis cinerea. In various QoI-resistant monosporic B. cinerea isolates from Hungary, a G-to-C point mutation was identified in the mitochondrial gene that encodes the QoI target, cytochrome b, resulting in a glycine to alanine substitution at position 143 (G143A). Analysis of Hungarian group I and group II strains further indicated the frequent occurrence of an additional group I-type intron in the cytb gene directly downstream of the glycine-143 codon. Mutual presence of distinct mitochondrial DNAs specifying different cytb alleles (heteroplasmy) has also been detected in monosporic strains. Remarkably, a number of group II field isolates were found to be highly resistant to azoxystrobin although they did not appear to carry the G-to-C mutation (G143A) generally associated with fungal QoI-resistance.
