Abstract

Mannheimia haemolytica-associated abomasitis has been clinically described as a cause of sudden death in lambs, but it is poorly characterized. We describe the pathological features of a severe fibrinonecrotizing abomasitis in 3 lambs that died suddenly. All 3 abomasums had a thickened submucosa due to edema and necrotic areas delimited by bands of degenerate neutrophils with slender nuclei (oat cells) and angiocentric distributions. The overlying mucosa was congested. Myriads of gram-negative coccobacilli were observed within the oat cell bands. M. haemolytica was isolated from the abomasum in all 3 animals and was serotyped as A2 in one of them. Pericarditis and pleuritis were observed in 2 of the lambs. Clostridium spp. were isolated in 1 lamb and detected by immunohistochemistry in the 3 animals, suggesting clostridial co-infection. M. haemolytica should be considered among the differential diagnoses of necrotizing abomasitis in lambs.

Abstract

Short response time is critical for future military medical operations in austere settings or remote areas. Such effective patient care at the point of injury can greatly benefit from the integration of semi-autonomous robotic systems. To achieve autonomy, robots would require massive libraries of maneuvers. While this is possible in controlled settings, obtaining surgical data in austere settings can be difficult. Hence, in this paper, we present the Dexterous Surgical Skill (DESK) database for knowledge transfer between robots. The peg transfer task was selected as it is one of 6 main tasks of laparoscopic training. Also, we provide a ML framework to evaluate novel transfer learning methodologies on this database. The collected DESK dataset comprises a set of surgical robotic skills using the four robotic platforms: Taurus II, simulated Taurus II, YuMi, and the da Vinci Research Kit. Then, we explored two different learning scenarios: no-transfer and domain-transfer. In the no-transfer scenario, the training and testing data were obtained from the same domain; whereas in the domain-transfer scenario, the training data is a blend of simulated and real robot data that is tested on a real robot. Using simulation data enhances the performance of the real robot where limited or no real data is available. The transfer model showed an accuracy of 81% for the YuMi robot when the ratio of real-to-simulated data was 22%-78%. For Taurus II and da Vinci robots, the model showed an accuracy of 97.5% and 93% respectively, training only with simulation data. Results indicate that simulation can be used to augment training data to enhance the performance of models in real scenarios. Abstract

Breast cancer (BCa) is a highly prevalent pathological condition (̴30% in women) with limited and subtype-dependent prognosis and therapeutic options. Therefore, BCa management might benefit from the identification of novel molecular elements with clinical potential. Since splicing process is gaining a great relevance in cancer, this work analysed the expression of multiple Spliceosome Components (SCs = 17) and Splicing Factors (SFs = 26) and found a drastic dysregulation in BCa (n = 69) vs. control (negative biopsies; n = 50) samples. Among all the components analysed, we highlight the upregulation of ESRP1 and down-regulation of PRPF8 and NOVA1 in BCa vs. control samples. Indeed, ESRP1 was specially overexpressed in triple-negative BCa (TNBCa) and associated with worse prognosis (i.e., higher BCa grade and lower overall survival), suggesting an association of ESRP1 with BCa aggressiveness. On the other hand, PRPF8 expression was generally downregulated in BCa with no associations to clinical characteristics, while NOVA1 expression was lower in TNBCa patients and highly aggressive tumours. Consistently, NOVA1 overexpression in vitro reduced functional parameters of aggressiveness in ER-/PR- cell lines (MDA-MB-231 and BT-549) but not in ER+/PR+ cells (MCF7), suggesting a critical role of NOVA1 in subtype-specific BCa. Finally, the in vitro pharmacological inhibition of splicing machinery using pladienolide B decreased aggressiveness features in all the BCa cell lines, showing a subtype-independent inhibitory potential, but being relatively innocuous in normal-like breast cells. These results demonstrate the profound dysregulation of the splicing machinery in BCa and their potential as source of promising diagnosis/prognosis markers, as well as valuable therapeutic targets for BCa. Abstract

Land tenure security (LTS) is important for achieving many sustainable development goals but its influence on forest cover is mixed. The uncertain relationship between LTS and forests is driven, in part, by the moderating influence of other drivers of deforestation. In this paper we illustrate this complex relationship between LTS and forest cover for individual private landholders in the Colombian Amazon. We use household surveys and econometric analysis with matching techniques to examine whether formal land titles and perceptions of LTS influence forest cover. We explore how the effect of a land title on forest cover is moderated by perceptions of LTS, time to markets, and participation in a conservation program. We find that more secure land tenure, on average, has a statistically significant and negative influence on forest cover in our sample. The negative association between LTS and forest cover is stronger when landholders perceive they have secure tenure and are closer to markets. However, we find the negative relationship between land title and forest cover goes away when a landholder participates in a conservation program. While our cross-sectional data and quasi-experimental methods cannot lead to causal statements, our results are in line with many recent studies in the Amazon region, and our household-level data provides important insight regarding drivers of deforestation that moderate the relationship between land title and forest cover. The article discusses the challenges in diagnosing and managing allergies to nonspecific lipid transfer protein (nsLTP) and the varying degrees of clinical severity in patients. A study was conducted to determine the minimum amount of peach nsLTP required to induce an allergic reaction during a food challenge test. The research found that around 24.5 μg of Pru p 3 (equivalent to 2.5 g of peach with peel) was needed to trigger symptoms, highlighting the importance of standardizing diagnostic tests for better clinical management.
