Bu bildiride bir buzağıda karşılaşılan doğmasal dermal melanom olgusunun klinik ve histopatolojik olarak değerlendirilmesi amaçlandı. Olgumuzu Kafkas Üniversitesi Veteriner Fakültesi Cerrahi Kliniğine getirilen, Simental ırkı, 4 aylık, erkek bir buzağı oluşturdu. Anamnez bilgilerinden, boynun sağ tarafında doğuştan var olan fındık büyüklüğündeki oluşumun hayvanın büyümesi ile birlikte hacmini arttırdığı öğrenildi. Klinik muayenede boyun derisi ile bağlantılı 20x16x9 cm boyutlarında, sert kıvamda ve koyu renkte bir kitle saptandı. Operasyonla uzaklaştırılan kitlenin histopatolojik incelenmesinde hücresel tip melanom olduğu belirlendi. Postoperatif iki yıllık süreçte nüks ile karşılaşılmadı., In the present case clinical and histopathological findings of a congenital dermal melanoma in a calf was reported. A 4-months-old, Simmental breed male calf was admitted to the Veterinary Surgery Clinics of the Faculty of Veterinary Medicine, University of Kafkas. Anamnesis revealed presence of a hazelnut sized mass on the right neck region as it was born which was growing continuously since birth. Clinical examination showed a dark coloured, hard mass attached to the skin of the neck region with a size of 20x16x9cm. Histopathological examination of the mass following surgical removal showed that the mass was a melanoma of cellular type. Postoperative follow up for two years showed no recurrence.

In this presentation it was aimed to report a case of mucocele (sialocele) and dermal collagenous hyperplasia in a 3-day-old, male Montafon calf admitted to the University of Kafkas, Faculty of Veterinary Medicine, Department of Surgery. Case history revealed a mass under the tongue and a swelling on the ventral part of the neck in the calf. Clinical examination showed a diffuse thickening of the ventral neck skin and a 4x4x3 cm sized, fluid filled sublingual mass which was removed surgically. The sublingual mass and biopsy material from the thickened part of the neck skin was sent for histopathological investigation. Histopathology revealed that the fluid filled sublingual mass was a mucocele (sialocele) and the dermal thickening was diagnosed dermal collagenous hyperplasia. Postoperative follow up for two years showed no recurrence of the sublingual mass. However, the collagenous hyperplasia on the neck region was persistent without any change in size., Bu bildiride Kafkas Üniversitesi Veteriner Fakültesi Cerrahi Kliniğine getirilen üç günlük, Montafon ırkı, erkek bir buzağıda karşılaşılan mukosel (sialosel) ve dermal kollajenöz hiperplazi olgusunun sunulması amaçlanmıştır. Hastanın anamnez bilgilerinden, doğuştan dil altında bir kitlenin varlığı ve boyunda şişkinlik olduğu öğrenildi. Klinik muayenede sublingual bölgede 4x4x3 cm boyutlarında, içi sıvı dolu bir kitle ve boynun ventralinde yaygın kalınlaşma tespit edildi. Dilaltındaki kitlenin ekstirpasyonu gerçekleştirildi. Boyun derisindeki kalınlaşma yaşamsal fonksiyonları etkilemediği için opere edilmedi. Biyopsi materyali alınarak histopatolojik incelemeye tabi tutulan dilaltındaki kitlenin mukosel, boyundan alınan deri parçasının ise dermal kollajenöz hiperplazi olduğu saptandı. Postoperatif iki yıllık süreçte dilaltında herhangi yeni bir oluşuma rastlanmadığı, boynun ventralindeki kollajenöz hiperplazinin ise mevcut durumunu koruduğu öğrenildi.

Amaç: Bu in vitro çalışmada, kızılcık meyva ekstraktının sisplatin indüklü nefrotoksisiteye karşı koruyucu etkisinin değerlendirilmesi amaçlanmıştır.Materyal ve Metot: Çalışmada Afrika yeşil maymun böbrek epitel hücreleri (Vero), 100 mg/mL kızılcık meyva ekstraktı, 50 ?mol/L sisplatin veya 50 ?mol/L sisplatin + 100 mg/mL kızılcık meyva ekstraktı ile 4 saat inkübe edildi. Herhangi bir ekleme yapılmayan, sadece hücre bulunan kuyucuklar kontrol grubu olarak kullanıldı. Hücre canlılığı, 3-(4,5-dimetil-thiazole-2-yl)-2,5-difenil tetrazolium bromide tayini ile belirlendi. Kültür medyumları toplandı ve santrifüj edildi. Malondialdehit (MDA), glutatyon (GSH), superoksit dismutaz (SOD) ve glutatyon peroksidaz (GPx) analizleri yapıldı.Bulgular: Sisplatine maruz bırakılan hücrelerde hücre canlılılığı %42, sisplatinle birlikte kızılcık meyva ekstraktı uygulanan hücrelerde ise hücre canlılılığı %59 bulundu. Sisplatin uygulanan hücrelerde hücresel hasar oranı yüksekti. Bununla birlikte kızılcık meyva ekstraktı ile kombine sisplatin grubunda, sisplatinin zararlı etkileri anlamlı derecede azalmıştı. Sisplatin grubu, kontrol grubu, kızılcık grubu ve sisplatin + kızılcık grupları ile karşılaştırıldığında MDA konsantrasyonu önemli decede yüksek, GSH konsantrasyonu, GPx ve SOD enzim aktiviteleri anlamlı derecede düşük bulundu (p < 0.05).Sonuç: Bu çalışma sisplatin ile uyarılan in vitro oksidatif hasar üzerine kızılcık meyva ektraktının koruyucu etkileri olduğunu göstermiştir, Objective: The aim of this study was to evaluate the protective effects of cornelian cherry fruit extract against cisplatin-induced nephrotoxicity in vitro.Materials and methods: African green monkey kidney epithelial cells (Vero) were incubated with 100 mg/mL of cornelian cherry fruit extract, 50 ?mol/L of cisplatin or 50 ?mol/L of cisplatin plus 100 mg/mL of cornelian cherry fruit extract for 4 h. The wells containing cells without any supplementation served as control. Cell viability was determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazoliumbromide assay. Culture mediums were collected, centrifuged and analyzed for malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD) and glutathione peroxidase (GPx)Results: The cell viability was 59% in cells co-treated with cisplatin and cornelian cherry fruit extract simultaneously and 42% in cisplatin treated cells. The cellular damage ratio was elevated in cells treated with cisplatin. However, when cisplatin combined with cornelian cherry fruit extract the deleterious effects of cisplatin were significantly decreased. Amaç Bu çalışmada, tarımda bir insektisit olarak kullanılan malathion’un neden olduğu oksidasyon parametreleri üzerine Onosma nigricaule (Boraginaceae)’den elde edilen bitkisel özütün böbrek dokusunda leptin üzerine etkisinin immunohistokimyasal olarak araştırılması amaçlandı. Materyal-Metot Çalışmamızda, toplam 48 adet erkek fare kullanıldı. Gruplar; kontrol (grup 1), malathion’un taşıt maddesi olan mısır yağı (grup 2), bitki özütünün taşıt maddesi olan serum fizyolojik (grup 3), malathion (grup 4), Onosma nigricaule (grup 5), ve Onosma nigricaule + malathion grubu (grup 6) olmak üzere toplam 6 grup olarak belirlendi. Böbreklerden alınan kesitlere leptin immunoreaktivitesini belirlemek için Streptavidin-Biotin-Peroksidaz Kompleks tekniği uygulandı. Hazırlanan kesitlere histolojik incelemeler için hematoksilen-eosin ve Crossman’ın üçlü boyama yöntemi kullanıldı. Bulgular Kontrol grubu ile taşıt kontrol grupları olan serum fizyolojik grubu ve mısır yağı grupları ile Onosma nigricaule, malathion ve Onosma nigricaule + malathion gruplarının böbrek dokularında herhangi bir lezyon tespit edilmedi. Diğer yandan, kontrol, mısır yağı, serum ve malathion grubundaki farelerin tubulus proksimalis ve tubulus distalis epitel hücrelerinde güçlü leptin immunoreaktivite görülmesine karşın Onosma nigricaule ve Onosma nigricaule + malathion grubu farelerde ise zayıf leptin immunoreaktivitesi tespit edildi. Sonuç Fareler ile yaptığımız çalışmada, yurdumuzda insektisit olarak yaygın kullanılan malathion’un fare böbreğinde leptin aktivitelerini etkilediği görülmüştür. Aim The purpose of this study was to determine the effects of the plant extract, which is obtained from Onosma nigricaule due to the oxidation parameters caused in mice by Malathion that is used as an insecticide in agriculture, on leptin in renal tissues in an immunohistochemical way. Material-Methods A total of 48 male mice were used in our study. 6 groups were determined as the control (group 1), maize oil which is the transport agent of malathion (group 2), normal saline which is the transport agent of plant extract (group 3), Onosma nigricaule (group 4), malathion (group 5) and Onosma nigricaule plant extract + malathion (group 6) group. The localization of leptin in the renal tissue was determined using the method of Streptavidin-BiotinPeroxidase. Hematoxylin-eosin and triple staining methods were used for histological and pathological examinations. Results No lesion was determined in renal tissues of the control group, normal saline group, maize oil group, Onosma nigricaule malathion and onosma nigricaule + malathion groups. Leptin immunoreactivity was determined with a weak intensity in renal tubulus proximalis and distalis epithelial cells of mice in the onosma nigricaule group, and onosma nigricaule + malathion group and with a high intensity in tubulus proximalis of other groups. Conclusion This study conducted on mice revealed that malathion, which is commonly used as insecticide in Turkey, affects leptin activities in renal tissue of mice.

In this study, the effect of capsaicin on the immunohistochemical distribution of cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) were examined in liver of the rat at the pubertal age.Thirty rats (Sprague-Dawley) aging 50 days old were used in this study. Rats were divided into three groups; experimental, sham and control group. Experimental group (n = 10) rats, was injected 1 mg / kg doses of capsaicin (10% ethanol, 1% tween, containing 80% distilled water mixture) subcutaneously every day for a period of one week. Sham group (n = 10) rats, were subcutaneously injected with a mixture containing 10% ethanol and 1% tween within 80% distilled water. The control group made no application. A week after the last injection, rats of three groups body was weighed and euthanized with ether anesthesia and liver tissues were taken. Body weights compared within the groups.The tissue localization of COX-1 and COX-2 was analysed by immunohistochemical method.Streptavidin-biotin-peroxidase complex technique was applied.COX-1 immunoreactivity experimental, sham and control groups were observed in centralis vein and the environment Kiernan centrilobuler region. Immunoreactivity was found in similar concentrations in all three groups. COX-2 is a experimental, sham and control groups were observed in centralis vein and Kiernan centrilobuler region in the portal area, Çalışmamızda, puberte döneminde capsaicin uygulanan sıçanların karaciğerinde siklooksijenaz-1 (COX-1) ve siklooksijenaz-2 (COX-2)’nin immunohistokimyasal lokalizasyonu incelendi. Çalışmada 50 günlük 30 erkek sıçan (Sprague-Dawley ırkı) kullanıldı. Sıçanlar deneme, sham ve kontrol olmak üzere 3 gruba ayrıldı. Deneme grubundaki (n=10) sıçanlara subkutan yolla her gün 1 mg/kg dozda capsaicin, % 10 ethanol, % 1 tween, % 80 distile su içeren karışım bir hafta süre ile enjekte edildi. Sham grubunu (n=10) oluşturan sıçanlara da aynı deneme grubundaki sıçanlarda olduğu gibi subkutan yolla CAP yerine sadece % 10 ethanol, % 1 tween, % 80 distile su içeren karışım subkutan olarak yapıldı. Kontrol grubuna (n=10) ise herhangi bir uygulama yapılmadı. Bir haftanın sonunda her üç gruptaki hayvanların vücut ağırlıkları tartıldıktan sonra eter ile ötenazi edildi ve karaciğer dokuları alındı. Grupların canlı ağırlıkları karşılaştırıldı. COX-1 ve COX-2 lokalizasyonu, immunohistokimyasal yöntemle tespit edildi. Streptavidin-Biotin-Peroksidaz Kompleks tekniği uygulandı. COX-1’in immunoreaktivitesi deneme, sham ve kontrol gruplarında vena sentralis ve Kiernan aralığı çevresindeki hepatositlerin sitoplazmasında gözlendi. Her üç grupta da immunoreaktivite birbirine benzer yoğunlukta bulundu. COX-2 ise deneme, sham ve kontrol gruplarında vena sentralisler etrafında ve Kiernan aralığına kadar olan portal alanda gözlemlendi

White muscle disease (WMD) is a world-wide nutritional disease of all animal species characterised by degeneration of skeletal and cardiac muscles. In the present study we determined the cardiac troponin I (cTnI) and inducible nitric oxide synthase (iNOS) expression in hearts of the lambs with WMD (n = 15). Eight clinically healthy lambs served as control. Creatine kinase (CK), creatine kinase MB (CK-MB), aspartate aminotransferase (AST), and lactate dehydrogenase (LDH) activities were analysed for both groups. cTnI levels were measured by a commercially available ELISA kit. Mean cTnI (10.49 ± 0.25 ng/ml), CK (261.16 ± 38.61 U/l), CK-MB (214.16 ± 12.71 U/l), AST (125.83 ± 1.05), and LDH (228.77 ± 14.86 U/l) concentrations were higher in WMD cases. Immunohistochemistry revealed definitive loss of cTnI expression in all cases. None of the 8 controls showed loss of the cTnI expression. However, iNOS immunoreactivity was augmented in degenerated cardiac myocytes and macrophages infiltrating to the interstitium compared to controls. The results of the study suggested that cTnI antibody might successfully be used as a sensitive test in the diagnosis of myocardial injury. iNOS expression augments in cardiac myocytes in lambs with WMD, most likely through influencing the signalling pathways to iNOS induction.

This study was aimed at determining the immunolocalization of VEGF and FGF-2 in the quail oviduct. The avidinbiotin peroxidase method was applied as an immunohistochemical technique. Immunostaining for VEGF and FGF-2 was rather intense in the luminal epithelium of caudal region of the infundibulum and of the isthmus , and it was observed that immunostaining was localized mostly in the apical cytoplasm. The epithelial cells lining the isthmus displayed diffuse cytoplasmic reactions for VEGF and FGF-2. While the reaction for VEGF was very intense, the reaction for FGF-2 was very weak in the luminal epithelium and moderate in the invaginated epithelium. In the magnum, VEGF (+) cells were present in the epithelium lining the mucosal folds, which were situated in regions with higher activity of the proprial glands. In the uterus and vagina, moderate to weak immunostaining for FGF-2 was detected in only the smooth muscle of the muscularis layer. The expression of FGF-2 and VEGF in the oviduct of healthy quails demonstrates that the reproductive physiology of these animals is regulated not only by hormonal mechanisms, but also by other molecular mechanisms., Bu çalışma bıldırcın oviduktunda VEGF ve FGF-2’nin immunolokalizasyonunu belirlemek amacıyla yapıldı. İmmunohistokimyasal yöntem olarak avidin-biotin peroksidaz metodu uygulandı. VEGF ve FGF-2 immunboyanması infindibulumun kaudal bölümünün ve istmus bölümünün lumenini sınırlandıran epitelde oldukça yoğundu ve çoğunlukla apikal sitoplazmada lokalizeydi. İstmusu çevreleyen epitel hücrelerinde VEGF ve FGF-2 için diffuz sitoplazmik reaksiyon gözlendi. Ancak bu reaksiyon VEGF için çok yoğun, FGF-2 için ise lumen epitelinde çok zayıf, invagine olan epitel bölgelerinde ise orta yoğunluktaydı. Magnum bölümünde, lamina propriyada bulunan bezlerin daha aktif olduğu bölgelerdeki mukoza dürümlerini örten epitelde VEGF (+) hücrelere rastlandı. Uterus ve vagina bölümlerinde sadece muskularis katmanındaki düz kas dokusunda FGF-2 için orta ve zayıf derecede immunboyanma gözlendi. FGF-2 ve VEGF’nin sağlıklı bıldırcın oviduktunda ekspresse edilmeleri, bu canlılarda üreme fizyolojisinin kontrolünde sadece hormonal mekanizmaların değil farklı diğer bazı moleküler mekanizmaların da bulunduğunu göstermektedir.

Bu olgunun materyalini iştahsızlık, zayıflama ve kronik timpani şikâyeti ile getirilen 4 yaşlı melez bir inek oluşturdu. Hastanın rutin klinik muayenesinden sonra deneysel laparatomi kararı verildi. Deneysel laparatomide rumen serozasında büyüklükleri 2-5 mm arasında değişen küçük ve parlak granulamatöz lezyonlara rastlandı. Hayvandan klinik biyokimya, hematolojik ve tümör belirteçlerinin incelenmesi için kan örneği ve laparatomide ise lezyonlardan biyopsi alındı. Hastalığın teşhisinden sonra hayvan kesime sevk edildi. Kesim esnasında yapılan makroskobik incelemelerde daha önce rumen serozasında belirlenen lezyonların benzerlerinin böbrek, dalak ve peritonda da yaygın şekilde yer aldığı tespit edildi. Yapılan histopatolojik, hematolojik, biyokimyasal ve tümor belirteci incelemeleri sonucunda; olguya “Diffuz mezotelyom” teşhisi konuldu

Purpose of this study was to repair achilles tendon rupture with protection of the integrity of the tendon without the need for the joint immobilization and prevent the complications reported with other techniques. For this purpose, mostly used techniques, Krackow, Modified Kessleer, Bunnel techniques were compared with the newly developed technique using the objective histologic and biomechanic criteria. Depending on the preliminary results of the pull-out tests, 6 tendons were included at each group, consisting of four groups totally. Operations were performed under the anesthesia of xylazine hydrochlorur, bupivacain HCI. At group 1, imlant prepared from the flexible polyethylene material was used. Krackow technique at group 2, Modified Kessler technique at group 3, Bunnell technique at group 4 were utilized. Rabbits were euthanised at 6th week postoperatively by use of overdose of sodium pentobarbital (50 mg/kg). While there was no difference between groups at histological analysis, at biomechanical analysis ultimate load to failure was found to be higher than all other techniques. Depending on these results, newly described technique can be applied clinically and may help to solve this orthopaedic problem., Bu çalışmayla eklemleri immobilize etme gereği olmadan ve tendon bütünlüğü mümkün olduğunca korunarak aşil tendon rüptürlerinin onarılması ve diğer tekniklerde bildirilen komplikasyonların önemli bir bölümünün önlenmesi amaçlandı. Bunun için günümüzde en sık kullanılan Krackow, Modifiye Kessler ve Bunnell sütür tekniklerinin uygulandığı üç ayrı kontrol grubu ve geliştirilen tekniğe ait sonuçlar biyomekanik objektif kriterlere göre karşılaştırılması yapıldı. Demonstrasyon olgularından elde edilen çekme deneylerinin sonuçları dikkate alınarak her grupta 6 denek olacak şekilde 4 ayrı grup oluşturuldu. Operasyonlar xylazine hydroklorür (Rompun 2% -Bayer) sedasyonunu izleyerek bupivacain HCI ( Marcaine 0,05- Eczacıbaşı) ile elde edilen spinal anestezi altında gerçekleştirildi. I. Grupta üzerinde delikler açılarak önceden hazırlanan fleksibl polyetilen materyalden hazırlanmış implant kullanıldı. II. grupta Krackow, III. grupta Modifiye Kessler, IV. grupta ise Bunnell dikiş teknikleri uygulanarak rüptür onarıldı. Denekler postoperatif 6.haftada overdose sodium penthobarbital (50 mg/kg) kullanılarak ötenazi edildi. Histopatolojik açıdan gruplar arasında önemli bir fark saptanamazken, biyomekanik test sonuçları dikkate alındığında geliştirilen tekniğe göre hem tendonun maksimum dayanacağı yük hem de uzama miktarı literatürdeki bütün tekniklere göre oldukça yüksek bulundu. Bu sonuç geliştirilen tekniğin maksimum dayanım sınırından sonra bile belirli bir süre daha yük taşımaya devam ettiğini göstermektedir. Buradan elde edilen sonuçlardan aynı tekniğin insan modellerinde de uygulanabileceği kanısına varılırken, tekniğin insan modellerinde başarılı olması durumunda bu alanda çok önemli bir ortopedik problemin sorunsuz bir şekilde çözüme kavuşturulabileceği söylenebilir.

The present study examined the mammary glands and supramammary lymph nodes of cows with a history of abortion due to Brucella abortus, and presents the histopathological, immunohistochemical, and bacteriological results. The study included 11 Brucella-seropositive cows that aborted during their first gestation. Histopathologically, the mammary glands showed lymphoplasmacytic and histiocytic interstitial mastitis. The lymph nodes had lymphofollicular hyperplasia and medullary plasmacytosis. Immunohistochemistry results showed brucellar antigens, predominantly in the cytoplasm of macrophages, and neutrophils in the intralobular interstitium and periductal stroma in the udders of 3 cows. Desquamated alveolar epithelium also indicated intense immunopositivity. In the lymph nodes, macrophages containing the antigen were frequently observed in the medullary region. B. abortus biotype 3 was isolated from the udders and supramammary lymph nodes of 4 cows. Although the most specific procedure for diagnosing the disease is isolation of the causative organism, for suspected cases in which the bacteriologic culture is negative or the material is fixed in formalin, immunohistochemistry may be used as a diagnostic tool for the detection of Brucella organisms., Bu çalışmada, Brucella abortus’a bağlı atık yapmış ineklerin meme bezi ve supramammar lenf düğümleri patolojik, immunohistokimyasal ve bakteriyolojik olarak değerlendirildi. Çalışma materyalini, ilk gebeliklerinde atık yapmış Brucella-seropozitif11 inek oluşturdu. Histopatolojik olarak, meme bezlerinde lenfo-plazmositik ve histiyositik intersitisyel mastitis gözlendi. Lenf düğümlerinde folliküler hiperplazi ve medullar plazmasitozis görüldü. İmmunohistokimyasal olarak, 3 ineğin meme bezinde intralobular intersitisyum ve periduktal stromada özellikle makrofaj ve nötrofil lökositlerin sitoplazmasında brucella antijeni tespit edildi. Ayrıca, alveolar lumende deskuamatif alveolar epitelyumun şiddetli immunopozitif sitoplazmik reaksiyonu gözlendi. Lenfdüğümlerinde, antijen bulunan makrofajlara sıklıkla medullar bölgede rastlandı. Dört ineğin meme bezi ve supramammar lenf düğümlerinden B. abortus biyotip 3 izole edildi. Brusellozisin kesin tanısında en güvenilir yöntem etken izolasyonu olmakla beraber, bakteriyel kültürün negatif olduğu veya dokunun formalin ile tespit edildiği durumlarda, brucella antijenlerinin ortaya konulması için immunohistokimyasal yöntemlerin teşhis aracı olarak kullanılabileceği sonucuna varılmıştır.

Erdoğan, Hidayet Metin ( Aksaray, Yazar ), This study was designed to determine carnitine concentrations in newborn calves with suspected septicaemia, healthy calves, and colostrum samples as well as to detect alterations in blood carnitine concentrations after colostrum intake. Carnitine concentrations were detected in both neonatal calves with suspected septicaemia (n = 195) and healthy calves (n =10) as well as in colostrum/milk samples from their dams (n = 20). The mean carnitine concentration on the 2nd day after colostrum intake (32.61 μmol/L) was significantly higher than that measured before colostrum intake (17.61 μmol/L) (P < 0.05). The striking result was significantly lower mean carnitine concentration detected in calves with suspected septicaemia (17.11 μmol/L), compared to healthy calves (24.92 μmol/L) (P < 0.001). Apart from a sudden increase on day 7, no significant alteration was observed in colostrum/milk carnitine concentrations throughout the postpartum period. The results indicated that colostrum contained carnitine, which passed into the bloodstream of the calf by passive colostral transfer, and that a dramatic reduction in blood carnitine concentrations of calves with suspected septicaemia existed. The results might be of help in dealing with septicaemia suspected neonatal calves through both measurement of blood carnitine and, in response, to supplement such cases with carnitine.

Amaç: Bu çalışma, köpek ve kedi yumuşak doku sarkomlarında ve farelerde 3-MCA ile indüklenen rhabdomyosarkomlarda PDGFA, PDGFR-alfa, integrin alfa v ile E-selektin ekspresyonları ile tadalafilin kemoterapötik etkilerinin değerlendirilmesi amaçlandı.Materyal ve Metot: Çalışmada, 25 (14 kedi ve 11 köpek) olgu incelendi. Ayrıca, 60 deney faresinde tümör oluşturuldu. Fareler 6 gruba ayrıldı. Üç grup kontrol grupları olarak kullanıldı, diğer üç grupta tümör oluşturuldu; bunlardan birisi, tadalafil ile intraperitoneal ve biri de intratümöral olarak 14 gün süreyle tedavi edildi. Köpek, kedi ve fare tümörlerinin klasifikasyonun için bir dizi antikor kullanıldı. Daha sonra tümörler, PDGFA, PDGFR-alfa, integrin alfa v ve E-selektin ile boyandı.Bulgular: Kedilerde 13 tümör enjeksiyon bölgesi fibrosarkomu, bir tanesi mikzoid liposarkom, köpeklerde ise 10'u fibrosarkom, biri ise mikzosarkom olarak teşhis edildi. Kedi ve köpek tümörlerinde PDGFA, PDGFR-alfa, integrin alfa v ekspresyonlarının yüksek E-selectin salınımının ise düşük olduğu belirlendi. Farelerde, sırası ile PDGFA, PDGFR-alfa, integrin alpha v, ve E-selectin salınımlarının %88, %58, %96 ve %46 olduğu görüldü. IP grubundaki tümör hücrelerinde PCNA ekspresyonunun daha differensiye oldukları ve PCNA salınımının daha düşük olduğu görüldü.Sonuç: Köpek ve kedi yumuşak doku sarkomları ile 3-MCA ile oluşturulan fare sarkomlarında tirozin kinazlar ile ECAM'ın arttığı görüldü. Tadalafilin ise 3-MCA ile oluşturulan RMS'larda önemli bir terapötik etkisinin olmadığı belirlendiAnahtar Kelimeler: Yumuşak doku sarkomları, 3-MCA, tadalafil, PDGF, İntegrin, E-selektin Aim: The objective of the current study was to determine the expression of PDGFA, PDGFR-alpha, integrin alpha v and E-selectin in the canine and feline soft tissue sarcomas and in 3-MCA induced rhabdomyosarcomas in mice, and evaluation of chemotherapeutic effects of tadalafil.Materials and Methods: The study was conducted on the 25 (14 cats & 11 dogs) cases of soft tissue sarcomas. Furthermore, tumor induced in 60 mice. Three groups served as control while tumor was induced in other 3 groups; one was tumor control and others were treated with tadalafil intraperitoneally or intratumorally for 14 days. A panel of antibodies was used to classify the tumors of dog, cat and mice. Later tissue sections were stained with PDGFA, PDGFR-alpha, integrin alpha v and E-selectin. Results: Thirteen tumors in cats were diagnosed as injection site fibrosarcoma and 1 as myxoid liposarcoma while 10 tumors in dogs were fibrosarcoma, 1 was myxosarcoma. PDGFA, PDGFR-alpha, integrin alpha v was upregulated in cat and dog tumors while E-selectin expression was low. In mice, PDGFA, PDGFR-alpha, integrin alpha v and E-selectin expression was observed in 88% and 58%, 96% and 46% tumors, respectively. Tumor cells in IP group was more differentiated and showed less PCNA expression.Conclusion: Tyrosine kinases and ECAM are upregulated in canine & feline soft tissue sarcomas and 3-MCA induced RMS and tadalafil does not have significant chemotherapeutic effects against 3-MCA induced RMS at the dose rate used in the current study.Keywords: Soft tissue sarcomas, 3-MCA, tadalafil, PDGFs, Integrin, E-selectin 144
