Infectious laryngotracheitis virus (ILTV) causes an acute and highly contagious respiratory disease in poultry. Live-attenuated vaccines are generally used to control and prevent infectious laryngotracheitis (ILT). However, these vaccines can revert to a virulent form due to multiple passages and thereby become an ILT source. Hence, monitoring of ILTV in the field through molecular characterization is critically important for controlling infection and differentiating circulating isolates. In this study, we genotypically characterized and phylogenetically analyzed eight ILTV isolates from chicken flocks located in four different cities of Turkey between 2019 and 2022. For all isolates, we analyzed two regions of the infected cell protein 4 gene (ICP4-1 and ICP4-2) and the thymidine kinase (TK) gene. The isolates were 100%, 100%, and 99.8-100% identical to each other in the ICP4-1 and ICP4-2 gene fragments and the TK gene, respectively. None of the ICP4 sequences had a deletion at nt 272-283, confirming that they were field isolates. None of the isolates were predicted to have a T252M mutation in the thymidine kinase, suggesting that they have low virulence. The isolates were 100%, 99.36%, and 99.91% identical to Turkish ILTV isolates in their ICP4-1, ICP4-2, and TK gene region, respectively. Phylogenetic analysis based on the ICP4-1 and TK genes confirmed that the ILTV isolates are closely related to Turkish ILTV isolates. This suggests that these ILTVs were endemic isolates, which in turn suggests that the ILTV isolates circulating in Turkey were evolutionarily close, originated from the field, and had low virulence., (© 2024. In this study, it was aimed to determine the effects of using UV light instead of formaldehyde in disinfection of hatching eggs on the microbial load, shell and yolk compositions, embryo development, hatching results and chick characteristics. 552 hatching eggs were divided into formaldehyde fumigation and disinfection with UV groups. After disinfection, bacteriological growth was not observed in groups. While the difference between the groups in terms of hatching results was insignificant, it was determined that the incubation period in the UV group was shorter than that of formaldehyde group. On the hatching day, relative chick weight and weight of bursa Fabricus were found to be higher in the UV group  . UV light can be used as an alternative to formaldehyde as disinfection of hatching eggs because it is easy to use and does not cause any negative effects on egg shell structure, embryo development, hatching results and chick characteristics.

Proteus mirabilis is a common enteric bacterium in livestock and humans. The increase and spread of the antimicrobial resistant P. mirabilis is considered alarming worldwide. Transmission mainly occurs through consumption of contaminated poultry products. We investigated antimicrobial resistance (AMR) and virulence markers in broiler chicken-originated P. mirabilis isolates from 380 fecal samples. Phenotypic AMR test was performed against seventeen different antimicrobials. Genotypic AMR test was performed to detect sixteen different AMR genes. The samples were also tested for the presence of eight different virulence genes and biofilm formation. P. mirabilis was isolated in 11% of the samples, with significantly high multidrug-resistant (MDR) prevalence (63%). All isolates were resistant to tetracycline (100%). The combined disc method indicated that all isolates were of extended-spectrum beta-lactamase (ESBL) producers, which was compatible with the high blaTEM prevalence (95%). This was associated with blaTEM being responsible for more than 80% of ampicillin resistance in enteric pathogens. The absence of phenotypically carbapenem-resistant isolates was compatible with the very low prevalences of blaOXA (2%) and blaNDM (0%). All isolates were positive for pmfA, atfA, hpmA, and zapA (100%) virulence genes, while biofilm formation rate (85%) indicated high adherence abilities of the isolates.

This study investigated five strains of each serotype of Salmonella Agona, Salmonella Heidelberg, Salmonella Hindmarsh, Salmonella Kouka, Salmonella Muenchen, Salmonella Ottmarchen, Salmonella Saintpaul and Salmonella II, isolated during the 2014-2017 period. Disc diffusion was used to identify the phenotypic profiles of antibiotic resistance to 12 antimicrobials while the presence of antibiotic resistance genes (ARGs) was detected by PCR. The most sensitive serotype was S. Kouka while the most resistant serotypes were S. Agona and S. Heidelberg. MDR was detected most frequently in S. Agona strains, followed by S. Saintpaul, S. Hindmarsch, and S. Ottmarchen. The samples were most susceptible to chloramphenicol and ceftazidime and most resistant to sulfonamide. The resistance genes were detected in phenotypically resistant strains. Among the tetracycline-resistant strains, tet (A) was the most prevalent gene. The results of this study highlight the importance of monitoring antibiotic resistance profiles and related genes, which can spread to form MDR bacteria. Salmonella spp., which significantly contribute to ARG dissemination, should be monitored constantly to protect the closely related health of humans, animals, and the environment. The level of antibiotic resistance observed in this study, even in rarely isolated Salmonella serotypes, also indicates the need for careful and selective use of antibiotics.

Climate change directly or indirectly affects the standard of living and quality of life for everyone, and so it has generated widespread interest and concern. Human awareness of global climate change, knowledge of the dimensions of the problem, and its consequences, are the first and most important steps towards a solution. However much the United Nations Convention on Climate Change (UNFCCC) and the Paris Agreement are seen as global efforts  to combat climate change, economic outcomes overshadow worries about climate change. Therefore, even if individual awareness seems small for such a large problem, it is as an effective first step. In this regard, a survey was conducted among 384 people living in the districts of Odunpazarı and Tepebaşı of Eskişehir province to determine their levels of awareness and knowledge of climate change. Survey participants were evaluated with regard to the district in which they lived and their gender, age group, education, membership in nongovernmental organizations (NGO), and income. Correlations between various determinant characteristics of the participants and their knowledge of climate change and readiness to pay more for goods produced without harming the environment are presented with χ 2 analysis, percentages, and proportions. No statistically significant correlation was found between knowledge of climate change of the participants and district, age, or gender, but a significant correlation was found between education and income levels, and NGO membership. There was no correlation between being willing to pay more for goods produced without harming the environment and district. No correlation was found between NGO membership and readiness to pay more for goods produced without harming the environment. Both central districts are close to each other and are very similar in terms of lifestyle, income, and the practices of local administrations regarding climate change and environmental problems. One of the factors affecting the hatching results and chick quality is the implementation of an appropriate disinfection program with effective disinfectants. This study was carried out to evaluate the effectiveness of oregano juice as a disinfection of hatching layer eggs as an alternative to formaldehyde. In the study, 1800 eggs collected from 48-week old Akbay white layer breeders were used. Eggs were divided into 3 equal disinfection groups (fumigation with formaldehyde, 50% oregano juice, and 100% oregano juice). In the study, the characteristics of the eggshell, microbial load, embryo development, egg weight loss, hatching results, chick weight and quality, and performance in the first 2 weeks were determined. No differences were observed among the disinfection groups in egg shell characteristics, microbial load in the shell, hatchability of fertile and set eggs, embryonic deaths, and body weight, body weight gain, and feed conversion ratio in female chicks. Egg weight loss was higher (P < 0.001) in the 50% oregano juice group (13.75%) compared to the formaldehyde fumigation group (12.11%). Chick quality score was found to be lower in the formaldehyde fumigation group (96.44) compared to the other two oregano juice groups (P < 0.001). The chicks hatched from the eggs in the formaldehyde fumigation group consumed more feed (119 g/week) in the first 2 weeks (P < 0.001). The use of 50% oregano juice in the disinfection of hatching eggs did not have any negative effects compared to the fumigation process with formaldehyde. Therefore, it is concluded that disinfection with 50% oregano juice was effective in reducing the microbial load in the egg shell as much as formaldehyde fumigation, and even it was superior in chick quality score., (© 2021. Hydrogen sulfide (H 2 S) detection is a screening method for distinguishing and identifying Salmonella strains from other bacteria in the intestine. Incidences of H 2 S-negative Salmonella have recently been reported in different countries. Although a high resistance rate against antimicrobial agents has been reported for H 2 S-positive Salmonella in many regions of the world, there is increasing evidence that high resistance to antibiotics has also increased in many H 2 S-negative Salmonella isolates. In this study, molecular characterisation of three H 2 S-negative Salmonella Havana, isolated from cloacal swab samples of broiler chickens, was performed. The phsA, phsB and phsC genes of the phs operon, which is responsible for hydrogen sulfide production, were amplified. Sequence analysis was then performed to identify mutations in the gene cluster. The antimicrobial resistance profiles of the isolates were determined by disc diffusion. Molecular characterisation was performed by multilocus sequence typing (MLST) and pulsed field gel electrophoresis (PFGE). The sequence analysis showed identified five point mutations in the phsA gene and one point mutation in the phsC gene in all isolates. The antibiotic resistance profile showed that the strains were resistant to cefoxitin and ceftazidime. MLST analysis showed that all strains belonged to sequence type (ST) 1621. This study is the first to report the H 2 S-negative S. Havana serotype.

Burkholderia spp. emerged as important pathogens in the airways of immunocompromised humans, especially those with cystic fibrosis (CF). Failure of identification with conventional techniques, high intrinsic resistance to most antibiotics and biofilm formation can cause difficulties in the treatment of these infections. The aim of this study was to identify Burkholderia spp. strains isolated from CF and non-CF patients with with routine microbiological methods, matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) and multilocus sequence analysis (MLSA), to determine of the antibiotic susceptibility and synergies, and to evaluate biofilm formation of these isolates. A total of 38 Burkholderia spp. (25 CF, 13 non-CF) from 26 patients were identified by biochemical, phenotypical and matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) and sequence types were revealed by multilocus sequence analysis (MLSA). Sequence types of isolates were identified using the PubMLST database. Characteristics of biofilm formation of clinical isolates were evaluated by microplate method. Antibiotic susceptibilities of ceftazidime, meropenem, trimethoprim-sulfamethoxazole (TMP-SXT) and levofloxacin were determined by broth microdilution method according to CLSI (2017) guidelines. Synergy tests were performed by checkerboard method. Clinical isolates were identified as Burkholderia cenocepacia (n= 16), Burkholderia contaminans (n= 11), Burkholderia gladioli (n= 4), Burkholderia dolosa (n= 4), Burkholderia multivorans (n= 2) and Burkholderia seminalis (n= 1). Sequence types of these isolates were determined as ST19, ST72, ST102, ST180, ST482, ST602, ST629, ST740, ST839 and ST1392. The correct identification at the species-level with MALDI-TOF MS was 94-100% for all isolates except B.contaminans. Biofilm formation among the identified species in the study was determined as 53% (n= 20). There was no statistical difference when the biofilm production was evaluated separately among Burkholderia species and biofilm production rates between CF (56%, 14/25) and non-CF (46%, 6/13) Burkholderia isolates (p> 0.05). Overall rates of resistance to ceftazidime, meropenem, TMP-SXT, and levofloxacin of the isolates were 35%, 66%, 50% and 40%, respectively. The antibiotic resistance against Burkholderia spp., isolates obtained from CF patients were more susceptible to ceftazidime, but no significant difference was found for other antibiotics. Synergy was determined between meropenem and TMP-SXT in two isolates. Antagonism was detected in 15 isolates, 12 of them were between meropenem and ceftazidime, three of them were between ceftazidime and TMP-SXT. Numerous resistance mechanisms may lead to higher resistance in this bacteria, whereas the antagonism between meropenem and ceftazidime in this study might be attributed to the expression of beta-lactamases. In this study, the distinctness of sequence types between Burkholderia spp. isolated from CF and non-CF patient, provided a better understanding about the importance of biofilm formation for the infections with these bacteria and emphasized that the management of therapy should be driven by the antibiotic test results.
