Oxidative stress is known to play a major role in the pathogenesis of inflammatory bowel diseases (IBDs), and, in particular, superoxide dismutase (SODs) defenses were shown to be weakened in patients suffering from IBDs. SOD mimics, also called SOD mimetics, as low-molecular-weight complexes reproducing the activity of SOD, constitute promising antioxidant catalytic metallodrugs in the context of IBDs. A Mn(II) complex SOD mimic (Mn1) based on an open-chain diaminoethane ligand exerting antioxidant and anti-inflammatory effects on an intestinal epithelial cellular model was shown to experience metal exchanges between the manganese center and metal ions present in the biological environment (such as Zn(II)) to some degrees. As the resulting complexes (mainly Zn(II)) were shown to be inactive, improving the kinetic inertness of Mn(II) complexes based on open-chain ligands is key to improve their bioactivity in a cellular context. We report here the study of three new Mn(II) complexes resulting from Mn1 functionalization with a cyclohexyl and/or a propyl group meant to limit, respectively, (a) metal exchanges and (b) deprotonation of an amine from the 1,2-diaminoethane central scaffold. The new manganese-based SOD mimics display a higher intrinsic SOD activity and also improved kinetic inertness in metal ion exchange processes (with Zn(II), Cu(II), Ni(II), and Co(II)). They were shown to provide anti-inflammatory and antioxidant effects in cells at lower doses than Mn1 (down to 10 μM). This improvement was due to their higher inertness against metal-assisted dissociation and not to different cellular overall accumulations. Based on its higher inertness, the SOD mimic containing both the propyl and the cyclohexyl moieties was suitable for intracellular detection and quantification by mass spectrometry, quantification, that was achieved by using a 13C-labeled Co-based analog of the SOD mimics as an external heavy standard.

Several preclinical studies have reported the presence of gadolinium (Gd) in different chemical forms in the brain, depending on the class (macrocyclic versus linear) of Gd-based contrast agent (GBCA) administered. The aim of this study was to identify, with a special focus on insoluble species, the speciation of Gd retained in the deep cerebellar nuclei (DCN) of rats administered repeatedly with gadoterate or gadodiamide 4 months after the last injection.Three groups (N = 6/group) of healthy female Sprague-Dawley rats (SPF/OFA rats; Charles River, L'Arbresle, France) received a cumulated dose of 50 mmol/kg (4 daily intravenous administrations of 2.5 mmol/kg, for 5 weeks, corresponding to 80-fold the usual clinical dose if adjusted for man) of gadoterate meglumine (macrocyclic) or gadodiamide (linear) or isotonic saline for the control group (4 daily intravenous administrations of 5 mL/kg, for 5 weeks). The animals were sacrificed 4 months after the last injection. Deep cerebellar nuclei were dissected and stored at -80°C before sample preparation. To provide enough tissue for sample preparation and further analysis using multiple techniques, DCN from each group of 6 rats were pooled. Gadolinium species were extracted in 2 consecutive steps with water and urea solution. The total Gd concentrations were determined by inductively coupled plasma mass spectrometry (ICP-MS). Soluble Gd species were analyzed by size-exclusion chromatography coupled to ICP-MS. The insoluble Gd species were analyzed by single-particle (SP) ICP-MS, nanoscale secondary ion mass spectroscopy (NanoSIMS), and scanning transmission electron microscopy with energy-dispersive X-ray spectroscopy (STEM-EDX) for elemental detection.The Gd concentrations in pooled DCN from animals treated with gadoterate or gadodiamide were 0.25 and 24.3 nmol/g, respectively. For gadoterate, the highest amount of Gd was found in the water-soluble fractions. It was present exclusively as low-molecular-weight compounds, most likely as the intact GBCA form. In the case of gadodiamide, the water-soluble fraction of DCN was composed of high-molecular-weight Gd species of approximately 440 kDa and contained only a tiny amount (less than 1%) of intact gadodiamide. Furthermore, the column recovery calculated for this fraction was incomplete, which suggested presence of labile complexes of dissociated Gd3+ with endogenous molecules. The highest amount of Gd was detected in the insoluble residue, which was demonstrated, by SP-ICP-MS, to be a particulate form of Gd. Two imaging techniques (NanoSIMS and STEM-EDX) allowed further characterization of these insoluble Gd species. Amorphous, spheroid structures of approximately 100-200 nm of sea urchin-like shape were detected. Furthermore, Gd was consistently colocalized with calcium, oxygen, and phosphorous, strongly suggesting the presence of structures composed of mixed Gd/Ca phosphates. No or occasional colocalization with iron and sulfur was observed.A dedicated analytical workflow produced original data on the speciation of Gd in DCN of rats repeatedly injected with GBCAs. The addition, in comparison with previous studies of Gd speciation in brain, of SP element detection and imaging techniques allowed a comprehensive speciation analysis approach. Whereas for gadoterate the main fraction of retained Gd was present as intact GBCA form in the soluble fractions, for linear gadodiamide, less than 10% of Gd could be solubilized and characterized using size-exclusion chromatography coupled to ICP-MS. The main Gd species detected in the soluble fractions were macromolecules of 440 kDa. One of them was speculated to be a Gd complex with iron-binding protein (ferritin). However, the major fraction of residual Gd was present as insoluble particulate species, very likely composed of mixed Gd/Ca phosphates. This comprehensive Gd speciation study provided important evidence for the dechelation of linear GBCAs and offered a deeper insight into the mechanisms of Gd deposition in the brain.

Quantitative speciation of low molecular weight iron complexes based on the in situ generation of isotopically-labelled species reflecting the natural iron speciation in coconut water as a model endosperm liquid.

The influence of the fermentation process on selenite metabolism by a probiotic Bifidobacterium longum DD98 and its consequent enrichment in selenium (Se) were studied. The effects of sodium selenite (Na2SeO3) concentration (18−400 μg/ml), feeding time (12, 16, and 24 h), and fermentation stage (secondary and tertiary fermentation) were evaluated by measuring (i) the total Se content and its distribution between the water-soluble metabolome fraction and the water-insoluble fraction; (ii) the total concentrations of the two principal Se compounds produced: selenomethionine (SeMet) and γ-glutamyl-selenomethionine (γ-Glu-SeMet), and (iii) the speciation of Se in the metabolite fraction. The results revealed that the fermentation process notably changed the Se incorporation into metabolites (γ-Glu-SeMet and free SeMet) and proteins (bound-SeMet) in B. longum DD98. In particular, the production of SeMet was negatively correlated to that of γ-Glu-SeMet when no red precipitate was seen in the bacteria. The study offers a tool for the control of the optimization of the fermentation process towards the desired molecular speciation of the incorporated Se and hence contributes to the production of Se-enriched probiotics with good qualities and bioactivities.

Many fields of basic and applied sciences, including geochronology, astronomy, metabolism, etc., rely on the ability of mass spectrometry to obtain isotope ratio measurements having a high degree of certainty. The inability to resolve difficult isobaric interferences plagues certain measurements. A combined atomic and molecular (CAM) ionization source has been interfaced to a high-field Orbitrap mass spectrometer to alleviate severe atomic, isobaric interferences. This work examines the geochronologically significant 87Sr and 87Rb isotope pair. The mass difference between 87Sr and 87Rb is approximately 0.3 mDa, requiring a minimum resolving power (R = m/Δm) of ∼290,000, a value ∼30× higher than available with sector-field elemental mass spectrometers. Under ultrahigh-resolution conditions, Sr isotope ratio accuracy and precision were evaluated using NIST Sr SRM 987, yielding precision values of 1.7M. At equal 87Sr and 87Rb intensities, 87Sr/86Sr was measured as 0.71294 (a relative error of only 0.37%) with a precision of 0.097% RSD, clearly reflecting the alleviation of the isobaric interference.

Thioredoxin (Trx) and Thioredoxin Reductase (TrxR) enzymes are essential constituents of the Trx system involved in the cellular redox regulation network. This mitochondrial apparatus, in recent years, has been recognized as an important modulator of tumor growth; hence specific inhibitors are developed to target TrxR/Trx for cancer treatment[1]. Trx and TrxR redox active sites are characterized by the presence, respectively, of two cysteine (WCGPCK) at the N-terminus and by a selenol thiol motif (GCUG, where U=SeCys) at the C-terminus. Both Cys and SeCys are Lewis soft base therefore Au(I) based compounds show high affinity for these targets. In this context, while TrxR has been largely explored as target of cytotoxic gold complexes [2], Trx has been neglected. Here, we selected and synthesized two (NHC)2-gold(I) complexes (Figure 1), able to cross mitochondrial membrane, and investigated by mass spectrometry their interaction with Trx in comparison with the Au(I) drug auranofin.

Selenium (Se) is an essential element being present in the form of the naturally occurring amino acid selenocysteine (Sec), 25 human proteins involved in different cellular pathways contain Sec. As the most potent intracellular soft Lewis base, selenocysteine (SeCys) is able to bind electron poor soft acids as heavy metals, of awareness for environmental and human toxicology, Hg ions bind Se by means of higher equilibrium constants than sulfur (ca. 106 times), therefore these values compensate the lower cellular abundance (105 times) of selenols compared to thiols[2]. In this communication we present a comparative reactivity study of Hg(I) and Hg(II) compounds with model peptides: vasopressin (AVP) hormone with antidiuretic and vasopressor actions and its Sec containing analogs. These peptides were synthesized either by standard solid phase peptide Fmoc or Boc protocols. The metal ion interaction with these peptides was investigated by RP- LC coupled with electrospray MS/MS detection (LC-MS/MS). We observed mono, bis and bridged peptide metallations as detailed in the Scheme. Taking into consideration the stability of Se-Hg bonds, our results support the hypothesis of a binding preference of Hg to Sec residues in selenoproteins.

International audience; Purpose: To date, the analysis of gadolinium (Gd) speciation in the brain of animals administered with macrocyclic and linear Gd-based contrast agents (GBCAs) has been limited to Gd soluble in mild buffers. Under such conditions, less than 30% of the brain tissue was solubilized and the extraction recoveries of GBCAs into the aqueous phase were poor, especially in the case of the linear GBCAs. The aim of this study was to find the conditions to solubilize the brain tissue (quasi-)completely while preserving the Gd species present. The subsequent analysis using size exclusion chromatography–inductively coupled plasma–mass spectrometry (SEC-ICP-MS) was intended to shed the light on the speciation of the additionally recovered Gd.Methods: Four groups of healthy female Sprague Dawley rats (SPF/OFA rats; Charles River, L'Arbresle, France) received randomly 5 intravenous injections (1 injection per week during 5 consecutive weeks) of either gadoterate meglumine, gadobenate dimeglumine, gadodiamide (cumulated dose of 12 mmol/kg), or no injection (control group). The animals were sacrifice 1 week (W1) after the last injection. Brain tissues were solubilized with urea solution, whereas tissues extracted with water served as controls. Total Gd concentrations were determined in the original brain tissue and its soluble and insoluble fractions by inductively coupled plasma–mass spectrometry (ICP-MS) to calculate the Gd accumulation and extraction efficiency. Size exclusion chromatography coupled to ICP-MS was used to monitor the speciation of Gd in the soluble fractions. The stability of GBCAs in the optimum conditions was monitored by spiking the brain samples from the untreated animals. The column recoveries were precisely determined in the purpose of the discrimination of weakly and strongly bound Gd complexes. The identity of the eluted species was explored by the evaluation of the molecular size and retention time matching with Gd chelates and ferritin standard. The speciation analyses were carried out for 2 different brain structures, cortex and cerebellum.Results: The combination of water and urea extractions (sequential extraction) managed to solubilize efficiently the brain tissue (97% ± 1%) while preserving the stability of the initially injected form of GBCA. For macrocyclic gadoterate, 97% ± 1% and 102% ± 3% of Gd initially present in the cortex and cerebellum were extracted to the soluble fraction. For gadobenate, similar amounts of Gd (49% ± 1% and 46% ± 4%) were recovered from cortex and cerebellum. For gadodiamide, 48% ± 2% of Gd was extracted from cortex and 34% ± 1% from cerebellum. These extraction efficiencies were higher than reported elsewhere. The SEC-ICP-MS and the column recovery determination proved that Gd present at low nmol/g levels in brain tissue was exclusively in the intact GBCA form in all the fractions of brain from the animals treated with gadoterate. For the linear GBCAs (gadobenate and gadodiamide), 3 Gd species of different hydrodynamic volumes were detected in the urea-soluble fraction: (1) larger than 660 kDa, (2) approximately 440 kDa, and (3) intact GBCAs. The species of 440 kDa corresponded, on the basis of the elution volume, to a Gd3+ complex with ferritin. Gd3+ was also demonstrated by SEC-ICP-MS to react with the ferritin standard in 100 mM ammonium acetate (pH 7.4). In contrast to macrocyclic gadoterate, for linear GBCAs, the column recovery was largely incomplete, suggesting the presence of free, hydrolyzed, or weakly bound Gd3+ with endogenous ligands.Conclusions: The sequential extraction of rat brain tissue with water and urea solution resulted in quasi-complete solubilization of the tissue and a considerable increase in the recoveries of Gd species in comparison with previous reports. The macrocyclic gadoterate was demonstrated to remain intact in the brain 1 week after administration to rats. The linear GBCAs gadobenate and gadodiamide underwent ligand exchange reactions resulting in the presence of a series of Gd3+ complexes of different strength with endogenous ligands. Ferritin was identified as 1 of the macromolecules reacting with Gd3+. For the linear GBCAs, 3% of the insoluble brain tissue was found to contain more than 50% of Gd in unidentified form(s).

International audience; The reactions of the medicinal gold(i) compound auranofin and its close analogues with vasopressin and the diselenide analogue were comparatively investigated by LC-electrospray MS/MS. Evidence is gained of the possible cleavage of the S-S and Se-Se bridges induced by Au(i). Notably, we found that, in the absence of reducing agents, the sulfur and selenium atoms are metallated only at high temperature (70 °C) with the preferential binding of gold to selenium. The reaction with the S-S bridge can take place at physiological temperature (37 °C) under reducing conditions. The implications of these results are discussed in the general frame of the reactivity of biologically relevant soft Lewis acids with peptides and proteins.

International audience; Asphaltenes are considered to be the most problematic components of heavy oils because they can self-aggregate which leads to precipitation and causes various problems during oil recovery, transportation, and refining. The contribution of the porphyrins present in asphaltenes to the aggregation was previously studied by gel permeation chromatography inductively coupled plasma-mass spectrometry (GPC-ICP MS). The molecular weight of asphaltene aggregates was shown to be increased by free metal-containing porphyrins (corresponding to the lower molecular weight fraction) interacting with the aggregate’s surfaces by weak forces. The characterization of free porphyrins within the asphaltenes is therefore for the understanding of the mechanism of the aggregation, coprecipitation, and demetalation processes. Here, we developed a method for the separation of free porphyrins from asphaltenes on the basis of their polarity using high-performance thin-layer chromatography (HPTLC). This technique, using disposable plates, is particularly well suited for asphaltene analysis since it eliminates the risk of clogging typical of column chromatography. Cellulose plates were used in this study. The lower polarity of their hydroxyl groups limit the irreversible adsorption and improves the detection limit by the ICP-MS. Two well-separated peaks were obtained from purified asphaltene (Asphaltene 2017; Asphaltene Characterization Interlaboratory Study for PetroPhase 2017. In Proceedings of the 18th International Conference on Pretroleum Phase Behavior and Fouling, Le Havre, France, June 11–15, 2017; Total, the University of Pau, and the University of Rouen-Normandy: Le Havre, France, 2017.) and its corresponding whole crude oil and C5 and C7 fractions. The distribution of vanadium due to migration was determined by laser ablation (LA) ICP MS. The eluted fraction contained the free porphyrins, whereas the major fraction did not migrate and corresponded to trapped porphyrins. A comparison with the signal obtained by UV densitometry allowed the ratio between the inorganic and organic material to be measured.

International audience; Many fundamental questions of astrophysics, biochemistry, and geology rely on the ability to accurately and precisely measure the mass and abundance of isotopes. Taken a step further, the capacity to perform such measurements on intact molecules provides insights into processes in diverse biological systems. Described here is the coupling of a combined atomic and molecular (CAM) ionization source, the liquid sampling-atmospheric pressure glow discharge (LS-APGD) microplasma, with a commercially available ThermoScientific Fusion Lumos mass spectrometer. Demonstrated for the first time is the ionization and isotopically resolved fingerprinting of a long-postulated, but never mass-spectrometrically observed, bi-metallic complex Hg:Se-cysteine. Such a complex has been implicated as having a role in observations of Hg detoxification by selenoproteins/amino acids. Demonstrated as well is the ability to mass spectrometrically-resolve the geochronologically important isobaric 87Sr and 87Rb species (Δm ~ 0.3 mDa, mass resolution m/Δm ≈ 1,700,000). The mass difference in this case reflects the beta-decay of the 87Rb to the stable Sr isotope. These two demonstrations highlight what may be a significant change in bioinorganic and atomic mass spectrometry, with impact expected across a broad spectrum of the physical, biological, and geological sciences.

International audience; Selenoprotein P (SELENOP) is an emerging marker of the nutritional status of selenium and of various diseases, however, its chemical characteristics still need to be investigated and methods for its accurate quantitation improved. SELENOP is unique among selenoproteins, as it contains multiple genetically encoded SeCys residues, whereas all the other characterized selenoproteins contain just one. SELENOP occurs in the form of multiple isoforms, truncated species and post-translationally modified variants which are relatively poorly characterized. The accurate quantification of SELENOP is contingent on the availability of specific primary standards and reference methods. Before recombinant SELENOP becomes available to be used as a primary standard, careful investigation of the characteristics of the SELENOP measured by electrospray MS and strict control of the recoveries at the various steps of the analytical procedures are strongly recommended. This review critically discusses the state-of-the-art of analytical approaches to the characterization and quantification of SELENOP. While immunoassays remain the standard for the determination of human and animal health status, because of their speed and simplicity, mass spectrometry techniques offer many attractive and complementary features that are highlighted and critically evaluated.

The objective of the present study was to evaluate of serum metal levels in COVID-19 patients with different disease severity, and to investigate the independent association between serum metal profile and markers of lung damage. The cohort of COVID-19 patients consisted of groups of subjects with mild, moderate, and severe illness, 50 examinees each. Forty-four healthy subjects of the respective age were involved in the current study as the control group. Serum metal levels were evaluated using inductively-coupled plasma mass-spectrometry. Examination of COVID-19 patients demonstrated that heart rate, respiratory rate, body temperature, C-reactive protein levels, as well as lung damage increased significantly with COVID-19 severity, whereas SpO2 decreased gradually. Increasing COVID-19 severity was also associated with a significant gradual decrease in serum Ca, Fe, Se, Zn levels as compared to controls, whereas serum Cu and especially Cu/Zn ratio were elevated. No significant group differences in serum Mg and Mn levels were observed. Serum Ca, Fe, Se, Zn correlated positively with SpO2, being inversely associated with fever, lung damage, and C-reactive protein concentrations. Opposite correlations were observed for Cu and Cu/Zn ratio. In regression models, serum Se levels were inversely associated with lung damage independently of other markers of disease severity, anthropometric, biochemical, and hemostatic parameters. Cu/Zn ratio was also considered as a significant predictor of lower SpO2 in adjusted regression models. Taken together, these findings demonstrated that metal metabolism significantly interferes with COVID-19 pathogenesis, although the causal relations as well as precise mechanisms are yet to be characterized.

Aluminum (Al) is considered as the most abundant metal in the Earth’s crust, being the third most abundant chemical element after oxygen and silicon. Intensive development of the Al industry (Brough and Jouhara, 2020) due to a wide use of the metal has resulted in a significant increase in environmental Al levels (Crisponi et al., 2012). The sources of human Al exposure may include diet (Tietz et al., 2019), being responsible for 95% of total body Al (Goulle and Grangeot-Keros, 2020), drinking water (Krupinska, 2020), air (Exley, 2013), as well as cosmetics (Crisponi et al., 2012, 2013) and medicinal drugs, namely antacids (Klotz et al., 2017). Involvement in Al processing industry may also result in occupational Al exposure (Skalny et al., 2018). Earlier studies demonstrated that vaccination could be considered as a source of Al exposure due to the presence of aluminium adjuvants that are currently not widely used thus reducing the risk of vaccine-associated Al exposure (Goulle and Grangeot-Keros, 2020). Being a non-essential element, Al was shown to be toxic for humans (Exley, 2013), resulting in adverse health effects (Crisponi et al., 2011) including bone pathology (Klein, 2019) and breast cancer (Darbre et al., 2013). Our data also demonstrated the association between obesity (Tinkov et al., 2019), laboratory markers of metabolic syndrome and Al exposure markers (Skalnaya et al., 2018). However, the existing data on adverse effects of Al exposure are limited (Krewski et al., 2007). Recent studies have demonstrated that the brain may be considered as the target for Al toxicity (Exley, 2014), resulting in neurodegenerative (Exley, 2013; Shaw et al., 2014) and neurodevelopmental disorders (Blaylock, 2012). Recent detailed studies by Exley and the coauthors have highlighted the association between brain Al accumulation and neurological disorders including Alzheimer’s disease, multiple sclerosis and autism spectrum disorder (Exley and Clarkson, 2020; Mirza et al., 2017; Mold et al., 2018). Speciation analysis using hyphenated techniques demonstrated that Al exposure induces a specific bioligand response in Al-exposed neuronal cells (Polec-Pawlak et al., 2004). However, the particular mechanisms of Al neurotoxicity and their role in Al-associated neurological disorders are still debatable (Morris et al., 2017). This chapter will provide an update on the particular mechanisms of Al neurotoxicity that may be used as targets for development of therapeutic strategies. Generally, the neurotoxic effect of Al exposure is mediated by its common toxic properties including prooxidant, proinflammatory, proapoptotic activity that are reported for a variety of cell lines and tissues, as well as more specific “neurotropic” effects namely interference with neurotransmitter metabolism and neuronal cytoskeleton.

Methylselenol (MeSeH) has been suggested to be a critical metabolite for anticancer activity of selenium, although the mechanisms underlying its activity remain to be fully established. The aim of this study was to identify metabolic pathways of MeSeH in Saccharomyces cerevisiae to decipher the mechanism of its toxicity. We first investigated in vitro the formation of MeSeH from methylseleninic acid (MSeA) or dimethyldiselenide. Determination of the equilibrium and rate constants of the reactions between glutathione (GSH) and these MeSeH precursors indicates that in the conditions that prevail in vivo, GSH can reduce the major part of MSeA or dimethyldiselenide into MeSeH. MeSeH can also be enzymatically produced by glutathione reductase or thioredoxin/thioredoxin reductase. Studies on the toxicity of MeSeH precursors (MSeA, dimethyldiselenide or a mixture of MSeA and GSH) in S. cerevisiae revealed that cytotoxicity and selenomethionine content were severely reduced in a met17 mutant devoid of O-acetylhomoserine sulfhydrylase. This suggests conversion of MeSeH into selenomethionine by this enzyme. Protein aggregation was observed in wild-type but not in met17 cells. Altogether, our findings support the view that MeSeH is toxic in S. cerevisiae because it is metabolized into selenomethionine which, in turn, induces toxic protein aggregation.

International audience; Coconut water (Cocos Nucifera) is shown to be a source of essential elements present in the form of low-molecular weight stable complexes known for their bio-availability. The total element concentrations were in the range of 0.2-2.7, 0.3-1, 3-14 and 0.5-2 ppm for Fe, Cu, Mn, and Zn, respectively, and varied as a function of the origin of the nut and its maturity. Speciation was investigated by size-exclusion chromatography-inductively coupled plasma mass spectrometry (ICP MS), and hydrophilic interaction liquid chromatography (HILIC)electrospray-Orbitrap MS. The metal species identified included: iron complexes with citrate and malate: Fe III (Cit) 3 (Mal), Fe III (Cit) 2 (Mal) 2 , Fe III (Mal) 2 , glutamine: Fe III (Glu) 2 and nicotianamine: Fe II (NA); copper complexes with phenylanine: Cu II (Phe) 2 and Cu II (Phe) 3 and nicotianamine: Cu II (NA); zinc complexes with citrate: Zn II (Cit) 2 and nicotianamine Zn II (NA) and manganese complex with asparagine Mn II (Asp) 2. The contributions of the individual species to the total elements concentrations could be estimated by HILIC-ICP MS.

The ability for all organisms to acquire metals from their environment is essential for life. To overcome the metal restriction imposed by the host’s nutritional immunity, bacterial pathogens exploits the use of small high metal affinity molecules called metallophores. Metallophores are first synthetized in the cytoplasm, then secreted into the medium where they sequester the metal. The metal-metallophore complex is then imported into the bacterium following binding to dedicated cell surface receptors. Recently, a new family of metallophores has been discovered in pathogenic bacteria called staphylopine in Staphylococcus aureus and pseudopaline in Pseudomonas aeruginosa. Here, we are expending the molecular understanding of pseudopaline secretion/recovery cycle across the double-membraned envelope of the Gram-negative bacterium Pseudomonas aeruginosa. We first revealed that pseudopaline is secreted in a two-step process including export across the inner membrane by the CntI exporter followed by a specific transport across the outer membrane by the MexAB-OprM efflux pump. Such involvement of MexAB-OprM in pseudopaline secretion, reveal a new natural function that extends its spectrum of functions and therefore reasserts its interest as antibacterial target. We then addressed the fate of the recovered metal-loaded pseudopaline by combining in vitro reconstitution experiments using radio-labeled pseudopaline subjected to bacterial lysates, and in vivo phenotyping in absence of pseudopaline transporters. Our data support the existence of a pseudopaline degradation/modification mechanism, possibly involved in metal release following pseudopaline recovery. All together our data allowed us to provide an improved molecular model of secretion, recovery and fate of this important metallophore by P. aeruginosa.IMPORTANCEPseudopaline is a broad spectrum metallophore produced and used by Pseudomonas aeruginosa to supply the bacterium in metal in metal scarce environments. Here we are investigating the pseudopaline transport/recovery cycle across the bacterial envelope. We are first demonstrating that pseudopaline secretion in the medium is achieved by a specific efflux pump, usually dedicated to the release of toxic compounds such as antibiotics, thus revealing a new natural function for this efflux pump reasserting its interest as antibacterial target. Additional experiments also revealing the existence of an intracellular pseudopaline degradation mechanism providing new clues to another obscure step of the pseudopaline cycle which is the intracellular metal liberation from the imported metal-pseudopaline complex. All together our data allowed us to disclose important aspects of the secretion, recovery and fate of this essential molecule used by P. aeruginosa to survive during infections thus constituting new potential targets for antibacterial development.

Mycosporines and mycosporine-like amino acids have been described as natural sunscreens and antioxidant compounds presenting a great potential for health and cosmetic applications. Herein, an untargeted screening approach for mycosporines and mycosporine-like amino acids (MAAs) was developed by the coupling of zwitterionic hydrophilic interaction liquid chromatography (HILIC) with multistage electrospray mass spectrometry MS2/MS3 using an Orbitrap analyzer and fragment ion search (FISh). This method was applied to study the mycosporine and MAA contents of five algae extracted using a 50% methanol solution and sonication. Candidate-MAAs were detected by mining eight characteristic fragment ions in their HILIC data-dependent MS2 mass spectrum. Their exact masses were measured with 3 ppm mass accuracy and their structures were elucidated on the basis of the MS3/MS4 mass spectra. The method developed was validated with a targeted analysis using an extract of Gymnogongrus devoniensis which confirmed the detection of 14 MAAs reported in the literature. In addition, 23 previously unreported MAAs were detected and the structures could be assigned for seven of them. The developed method was applied to the analysis of four algae: Gelidium sesquipedale, Halopithys incurva, Porphyra rosengurtii and Cystoseira tamariscifolia allowing the detection of MAAs, including some reported here for the first time.

International audience; The coupling of a combined atomic and molecular (CAM) ionization source, the liquid sampling-atmospheric pressure glow discharge (LS-APGD) microplasma to an Orbitrap Fusion Lumos Tribrid 1M mass spectrometer is described as a significant step towards the elimination of isobaric interferences in elemental/isotopic mass spectrometry. The developed setup permits broadband, elemental mass spectra to be easily acquired at a mass resolution (m/Δm) of 500 000. A new ion source housing and integrated control system provide enhanced stability and sensitivity of the microplasma ion source in comparison to the previous designs. Detailed evaluation of the ion sampling parameters provided insight into differences with other Orbitrap platforms leading to a 5-fold increase in resolution in elemental/isotopic analysis. The roles of data acquisition/ion processing parameters were evaluated with regards to the precision and accuracy of 235U/238U isotope ratios. Significantly, the precision of those measurements improves as a function of pre-set mass resolution up to a value of 1 000 000, where a precision of 0.086% RSD is obtained for uranium concentrations of 100 ng mL−1. Analytical response curves (log–log), acquired for the two uranium isotopes at a resolution of 120 000 were linear (R2 = 0.9969) over more than 5 orders of magnitude, with limits of detection of 1 pg mL−1 for U. Based on 60 μL injection volumes, this represents a mass of 70 fg 235U. These figures of merit satisfy International Target Values (ITVs) for Measurement Uncertainties in Safeguarding Nuclear Materials set by the International Atomic Energy Agency (IAEA). Ultimately, and totally unique from beam-type instruments, the move to the higher-resolution Orbitrap platform provides all of the benefits of high mass resolution, with no penalties in sensitivity, precision, or dynamic range for uranium analysis.

International audience; Ag, As, Cu, Pb and Zn were found to be the principal metallic contaminants of a post-mining area of Peru (Hualgayoc, Cajamarca). Study of metal distribution amongst roots, stems, and leaves of four indigenous hypertolerant plant species, Arenaria digyna, Puya sp., Hypericum laricifolium, Nicotiana thyrsiflora indicated significant translocation of Zn (0.6 < TF ≤ 10.0) and Cu (0.4 < TF ≤ 6.5) into aerial plant organs and substantial water-extractable fraction (20–60%) of these metals, except for A. digyna (root and stems). A study of the metal speciation by ultrahigh-performance size-exclusion (fast-SEC) and hydrophilic ion interaction (HILIC) liquid chromatography with dual ICP (inductively coupled plasma) and electrospray (ESI) Orbitrap MS detection revealed the presence of nicotianamine and deoxymugineic acid copper and zinc complexes in roots, stem and leaves of N. thyrsiflora and Puya sp., and nicotianamine alone in A. digyna. A previously unreported compound, dihydroxy-nicotianamine was identified as the most abundant Cu and Zn ligand in H. laricifolium. The presence of arsenobetaine and an arsenosugar was confirmed by ESI MS. Ag and Pb were hardly translocated to leaves and were found as high molecular species; one of the Pb-containing species co-eluted in fast-SEC-ICP MS with rhamnogalacturonan-II-Pb complex commonly found in in the walls of plants.

Their behavior in refining processes is critically dependent on the speciation. Recent analytical developments, especially on the level of coupled techniques and high-resolution mass spectrometry, start allowing the speciation of individual metal compounds in crude oil known for its extreme complexity. These developments include: (i) GC stationary phases of high thermal stability and the high-temperature interfaces with ICP-MS and TOF-MS; (ii) high-efficiency microcolumn gel-permeation chromatography with detection by sector-field ICP-MS; (iii) thin layer chromatography coupling with laser ablation ICP-MS detection; and (iv) two-dimensional separation protocols increasing the purity of heteroelement containing fractions. Progress in electrospray and atmospheric-pressure photoionization Fourier Transform MS allows resolutions of above 1,000,000 to be achieved making it possible to identify by accurate mass measurement individual sulfur and metal species directly in crude oils. © 2017 Elsevier B.V.

International audience; Liver and other tissues accumulate selenium (Se) when animals are supplemented with high dietary Se as inorganic Se. To further study selenometabolites in Se-deficient, Se-adequate, and high-Se liver, turkey poults were fed 0, 0.4, and 5 mg Se g À1 diet as Na 2 SeO 3 (Se(IV)) in a Se-deficient (0.005 mg Se g À1) diet for 28 days, and the effects of Se status determined using HPLC-ICP-MS and HPLC-ESI-MS/MS. No selenomethionine (SeMet) was detected in liver in turkeys fed either this true Se-deficient diet or supplemented with inorganic Se, showing that turkeys cannot synthesize SeMet de novo from inorganic Se. Selenocysteine (Sec) was also below the level of detection in Se-deficient liver, as expected in animals with negligible selenoprotein levels. Sec content in high Se liver only doubled as compared to Se-adequate liver, indicating that the 6-fold increase in liver Se was not due to increases in selenoproteins. What increased dramatically in high Se liver were low molecular weight (M W) selenometabolites: glutathione-, cysteine-and methyl-conjugates of the selenosugar, seleno-N-acetyl galactosamine (SeGalNac). Substantial Se in Se-adequate liver was present as selenosugars decorating general proteins via mixed-disulfide bonds. In high-Se liver, these ''selenosugar-decorated'' proteins comprised B50% of the Se in the water-soluble fraction, in addition to low M W selenometabolites. In summary, more Se is present as the selenosugar moiety in Se-adequate liver, mostly decorating general proteins, than is present as Sec in selenoproteins. With high Se supplementation, increased selenosugar formation occurs, further increasing selenosugar-decorated proteins, but also increasing selenosugar linked to low M W thiols. Significance to metallomics The study shows for the first time that supplementation of a monogastric animal with inorganic Se(IV) leads to the synthesis of selenocysteine only, with no synthesis of selenomethionine. Excess Se(IV) is metabolized to selenosugars which react with low molecular weight thiols and with cysteine-containing peptides and proteins present in liver. These ''selenosugar-decorated'' proteins account for half of the liver Se in Se-adequate as well as high-Se turkey liver. This comprehensive approach was possible owing to the state-of-the art analytical techniques based on the different HPLC modes coupled with Se-specific (ICP MS) and molecule specific (Orbitrap MS/MS) detection and chemical reduction and derivatization.

International audience; The aim of this study was to investigate the presence and chemical forms of residual gadolinium (Gd) in rat brain after a single dose of Gd-based contrast agent. METHODS: Four groups of healthy rats (2 sacrifice time-points, n = 10/group, 80 rats in total) were randomized to receive a single intravenous injection of 1 of the 3 Gd-based contrast agents (GBCAs) (gadoterate meglumine, gadobenate dimeglumine, or gadodiamide) or the same volume of 0.9% saline solution. The injected concentration was 0.6 mmol/kg, corresponding to a concentration of 0.1 mmol/kg in humans after body surface normalization between rats and humans (according to the US Food and Drug Administration recommendations). Animals were sacrificed at 2 washout times: 1 (M1) and 5 (M5) months after the injection. Total Gd concentrations were determined in cerebellum by inductively coupled plasma mass spectrometry. Gadolinium speciation was analyzed by size-exclusion chromatography coupled to inductively coupled plasma mass spectrometry after extraction from cerebellum. RESULTS: A single injection of a clinically relevant dose of GBCA resulted in the detectable presence of Gd in the cerebellum 1 and 5 months after injection. The cerebellar total Gd concentrations after administration of the least stable GBCA (gadodiamide) were significantly higher at both time-points (M1: 0.280 ± 0.060 nmol/g; M5: 0.193 ± 0.023 nmol/g) than those observed for macrocyclic gadoterate (M1: 0.019 ± 0.004 nmol/g, M5: 0.004 ± 0.002 nmol/g; P < 0.0001). Gadolinium concentrations after injection of gadobenate were significantly lower at both time-points (M1: 0.093 ± 0.020 nmol/g; M5: 0.067 ± 0.013 nmol/g; P < 0.05) than the Gd concentration measured after injection of gadodiamide. At the 5-month time-point, the Gd concentration in the gadoterate group was also significantly lower than the Gd concentration in the gadobenate group (P < 0.05). Gadolinium speciation analysis of the water-soluble fraction showed that, after injection of the macrocyclic gadoterate, Gd was still detected only in its intact, chelated form 5 months after injection. In contrast, after a single dose of linear GBCAs (gadobenate and gadodiamide), 2 different forms were detected: intact GBCA and Gd bound to soluble macromolecules (above 80 kDa). Elimination of the intact GBCA form was also observed between the first and fifth month, whereas the amount of Gd present in the macromolecular fraction remained constant 5 months after injection. CONCLUSIONS: A single injection of a clinically relevant dose of GBCA is sufficient to investigate long-term Gd retention in the cerebellar parenchyma. Administration of linear GBCAs (gadodiamide and gadobenate) resulted in higher residual Gd concentrations than administration of the macrocyclic gadoterate. Speciation analysis of the water-soluble fraction of cerebellum confirmed washout of intact GBCA over time. The quantity of Gd bound to macromolecules, observed only with linear GBCAs, remained constant 5 months after injection and is likely to represent a permanent deposition.

BTEX (benzene, toluene, ethylbenzene, and the different xylene isomers), known for carcinogenic and neurotoxic effects, are common environmental contaminants. The first step for the development of the bioremediation technologies is the detection of intense microbial degradation in contaminated waters in the quest for the most active bacterial strains. This requires the multispecies analysis for BTEX metabolites which are considered as markers of microbial degradation. A direct (50 &micro, L injection) HPLC&ndash, electrospray MS/MS analytical method was developed for the simultaneous analysis of 11 BTEX metabolites (o-, m-, p-toluic, salicylic, benzoate, benzyl, and phenyl succinic acids, 2-(1-phenylethyl)-, 2-(2-methylbenzyl), and 2-(3-methylbenzyl)-, 2-(4-methyl benzyl)-succinic acids) in bacterial cultures and ground waters down to 0.1 ng/mL. The optimization of the chromatographic conditions allowed for the resolution of position isomers of toluic and methylbenzyl-succinic acids. The stability of the analytes during sample storage tested in different conditions showed the instability of some of them when stored at room temperature. The feasibility of the method was demonstrated by the detection of all the investigated metabolites in a water sample of a deep aquifer hosting natural gas storage. A model laboratory study emphasized the importance of 2-(2-methylbenzyl)-succinic acid as a marker of anaerobic microbial degradation.

International audience; A sequential fractionation procedure based on (i) water extraction, (ii) hexane extraction, (iii) saccharification, and (iv) proteolysis was developed to provide the first ever data on the molecular distribution of iron in maize. This was completed by the operational determination of the iron bioavailability using an in-vitro simulated model for gastro-intestinal digestion. The coupling of hydrophilic interaction chromatography (HILIC) and size exclusion chromatography (SEC) with the parallel detection by inductively coupled plasma mass spectrometry (ICP-MS) and high resolution electrospray mass spectrometry (HR-ESI-MS) allowed the identification of water-soluble Fe(III)-mugineate, Fe(III)-(citrate)2, and Fe(III)2-(phytate)2. The procedures were applied to study some well characterized maize varieties having shown previously differences in iron bioavailability during cell culture and animal model feeding studies. The combined analytical methods developed in this work could unambiguously discriminate low from high Fe bioavailable seeds in these closely related maize varieties.

International audience; Selenium is an essential trace element which is incorporated in the form of a rare amino acid, the selenocysteine, into an important group of proteins, the selenoproteins. Among the twenty-five selenoprotein genes identified to date, several have important cellular functions in antioxidant defense, cell signaling and redox homeostasis. Many selenoproteins are regulated by the availability of selenium which mostly occurs in the form of water-soluble molecules, either organic (selenomethionine, selenocysteine, and selenoproteins) or inorganic (selenate or selenite). Recently, a mixture of selenitriglycerides, obtained by the reaction of selenite with sunflower oil at high temperature, referred to as Selol, was proposed as a novel non-toxic, highly bioavailable and active antioxidant and antineoplastic agent. Free selenite is not present in the final product since the two phases (water soluble and oil) are separated and the residual water-soluble selenite discarded. Here we compare the assimilation of selenium as Selol, selenite and selenate by various cancerous (LNCaP) or immortalized (HEK293 and PNT1A) cell lines. An approach combining analytical chemistry, molecular biology and biochemistry demonstrated that selenium from Selol was efficiently incorporated in selenoproteins in human cell lines, and thus produced the first ever evidence of the bioavailability of selenium from selenized lipids.

We report for the first time that antimicrobial Ag+ targets the glycolytic pathway through inhibiting the key enzyme of GAPDH., Silver has long been used as an antibacterial agent, yet its molecular targets remain largely unknown. Using a custom-designed coupling of gel electrophoresis with inductively coupled plasma mass spectrometry (GE-ICP-MS), we identified six silver-binding proteins in E. coli. The majority of the identified proteins are associated with the central carbon metabolism of E. coli. Among them, we unveil that GAPDH, an essential enzyme in glycolysis, serves as a vital target of Ag+ in E. coli for the first time. We demonstrate that silver inhibits the enzymatic function of GAPDH through targeting Cys149 in its catalytic site. The X-ray structure reveals that Ag+ coordinates to Cys149 and His176 with a quasi-linear geometry (S–Ag–N angle of 157°). And unexpectedly, two Ag+ ions coordinate to Cys288 in the non-catalytic site with weak argentophilic interaction (Ag···Ag distance of 2.9 Å). This is the first report on antimicrobial Ag+ targeting a key enzyme in the glycolytic pathway of E. coli. The findings expand our knowledge on the mode of action and bio-coordination chemistry of silver, particularly silver-targeting residues in proteins at the atomic level.

International audience; Single particle inductively coupled plasma mass spectrometry (SP-ICP-MS) has become a well-established technique for the detection, size characterization and quantification of inorganic nanoparticles but its use for the analysis of micro- and nanoparticles composed of carbon has been scarce. Here, the analysis of a microplastic suspensions by ICP-MS operated in single particle mode using microsecond dwell times is comprehensively discussed. The detection of polystyrene microparticles down to 1.2 μm was achieved by monitoring the 13C isotope. Plastic microparticles of up to 5 μm were completely volatized and their components atomized, which allowed the detection of microplastics, their quantification using aqueous dissolved carbon standards, and the measurement of the size-distribution of the detected particles. Limits of detection of 100 particles per milliliter were achieved for an acquisition time of 5 min. The method developed was applied to the screening of microplastics in personal care products and released from food packagings. The chemical identity of the detected microplastics was confirmed by attenuated total reflectance Fourier-transform infrared spectroscopy.

OBJECTIVES: Liver and other tissues accumulate selenium (Se) when animals are supplemented with high dietary Se as inorganic Se. Because the nature of this accumulated Se is not well characterized, we studied selenometabolomics in Se-deficient, Se-adequate, and high-Se liver. In high-Se liver, these “selenosugar-decorated” proteins comprised ∼50% of the Se in the water-soluble fraction, in addition to the low MW selenometabolites. CONCLUSIONS: In Se-adequate liver, far more Se is present as the selenosugar moiety, mostly decorating general proteins, than is present as Sec in selenoproteins. With high Se supplementation, increased selenosugar formation occurs, further increasing selenosugar-decorated proteins, but also increasing selenosugar linked to low MW thiols, leading to the formation of methyl-SeGalNac. This suggested pathway, underlying adaptation to high Se status in animals, needs further investigation including study of the potential of selenosugar compounds as biomarkers of high Se status. Palladium is recognized as a technologically critical element (TCE) because of its massive use in automobile exhaust gas catalytic converters. The release of Pd into the environment in the form of nanoparticles of various size and chemical composition requires an understanding of their metabolism by leaving organisms. We provide here for the first time a chemical speciation insight into the identity of the ligands produced or used by a plant Sinapis alba L. exposed in hydropony to Pd nanoparticles and soluble Pd (nitrate). The analytical method developed was based on the concept of 2D HPLC with parallel inductively coupled plasma mass spectrometry (ICP MS) and electrospray MS detection. Size exclusion chromatography – ICP MS of the plant extracts showed no difference between the speciation of Pd after the exposure to nanoparticles and after that to Pd2+ which indicated the reactivity and dissolution of Pd nanoparticles. A comparative investigation of the Pd speciation in a control plant extract spiked with Pd2+ and of an extract of a plant having metabolized palladium indicated the response of the Sinapis alba by the formation of a Pd–histidine complex. The complex was identified via Orbitrap MS; the HPLC-MS chromatogram produced two peaks at m/z 415.0341 each corresponding to a Pd–His2 complex. An investigation by ion-mobility MS revealed a difference in their collision cross section indicating that the complexes present varied in terms of spatial conformation. A number of other Pd complexes with different ligands (including nicotianamine) circulating in the plant were detected but these ligands were already observed in a control plant and their concentrations were not affected by the exposure to Pd.

International audience; We have examined the aggregation behavior of a typical atmospheric residue feedstock by gel permeation chromatography (GPC). The size profiles for compounds containing sulfur, vanadium, and nickel were determined on-line from elemental detection by inductively coupled plasma (ICP) mass spectrometry. Four fractions that vary in aggregation state were analyzed by positive atmospheric pressure photoionization (APPI) 9.4 tesla Fourier transform ion cyclotron resonance mass spectrometry (APPI FT-ICR MS). Results showed an inverse relationship between fraction aggregate size and monomer ion yield and revealed that aggregation tendency did not correlate with higher polar or aromatic species abundance. Aggregation in the atmospheric residue more closely correlated with increased relative abundance of larger and more aliphatic compounds. The molecular composition of the GPC aggregate fractions suggests that non-polar intermolecular forces between saturated, long-chain alkyl substituents contribute more to aggregation than pi-pi interactions.

International audience; The growing release of technology-critical elements (TCEs): Ga, Ge, In, Nb, Ta, Te, Tl, and most of the platinum group elements (PGEs), and rare earth elements (REEs) requires the understanding of their mobility, reactivity and chemical transformations in the environment which are critically dependent on their chemical form (speciation). The total concentrations of these elements at the trace levels make their speciation analysis challenging. Coupled techniques combining a high resolution chromatographic separation with ultrasensitive element-specific detection by inductively coupled plasma mass spectrometry (ICP MS) often offer the unique possibility of accessing the information on the identities and concentrations of individual metal species present in environmental samples. This paper reviews the advantages and limitations of these techniques to TCEs speciation and highlights the development trends in analytical methodology that may contribute to our understanding of the risks associated with the increasing environmental presence of TCEs. © 2017 Elsevier B.V.
