Abstract Microbes can decompose biodegradable plastics on land, rivers and seashore. However, it is unclear whether deep-sea microbes can degrade biodegradable plastics in the extreme environmental conditions of the seafloor. Here, we report microbial decomposition of representative biodegradable plastics (polyhydroxyalkanoates, biodegradable polyesters, and polysaccharide esters) at diverse deep-sea floor locations ranging in depth from 757 to 5552 m. The degradation of samples was evaluated in terms of weight loss, reduction in material thickness, and surface morphological changes. Poly(l-lactic acid) did not degrade at either shore or deep-sea sites, while other biodegradable polyesters, polyhydroxyalkanoates, and polysaccharide esters were degraded. The rate of degradation slowed with water depth. We analysed the plastic-associated microbial communities by 16S rRNA gene amplicon sequencing and metagenomics. Several dominant microorganisms carried genes potentially encoding plastic-degrading enzymes such as polyhydroxyalkanoate depolymerases and cutinases/polyesterases. Analysis of available metagenomic datasets indicated that these microorganisms are present in other deep-sea locations. Our results confirm that biodegradable plastics can be degraded by the action of microorganisms on the deep-sea floor, although with much less efficiency than in coastal settings.

Abstract In the brain, many regions work in a network-like association, yet it is not known how durable these associations are in terms of activity and could survive without structural connections. To assess the association or similarity between brain regions with a generating approach, this study evaluated the similarity of activities of neurons within each region after disconnecting between regions. The “generation” approach here refers to using a multi-layer LSTM (Long Short-Term Memory) model to learn the rules of activity generation in one region and then apply that knowledge to generate activity in other regions. Surprisingly, the results revealed that activity generation from one region to disconnected regions was possible with similar accuracy to generation between the same regions in many cases. Notably, firing rates and synchronization of firing between neuron pairs, often used as neuronal representations, could be reproduced with precision. Additionally, accuracies were associated with the relative angle between brain regions and the strength of the structural connections that initially connected them. This outcome enables us to look into trends governing non-uniformity of the cortex based on the potential to generate informative data and reduces the need for animal experiments.

Abstract Background This study aimed to conduct a preliminary evaluation of the effects of 6-methylsulfinylhexyl isothiocyanate (6-MSITC) contained in wasabi rhizomes on fatigue and sleep and to examine its safety through overdose study. Methods A total of 20 healthy volunteers who were experiencing daily fatigue were given powder containing 6-MSITC (4.8 mg/day of 6-MSITC) extracted from wasabi for 4 weeks. Then, fatigue, sleep, autonomic nervous functioning, stress, and immunity were evaluated. In addition, an overdose safety study of the extract powder (up to 16 mg/day of 6-MSITC for 4 weeks) was performed with 30 healthy volunteers in a double-blind, placebo-controlled method. Results The powder containing 6-MSITC did not improve fatigue after a mental task, but fatigue before the mental task, sleep, and mood were improved significantly after 4 weeks intake. No changes were observed in the autonomic nerve function, stress, or immune markers. In the overdose safety study, no changes in the parameters ​​or side effects were observed, and the results showed that high doses of the extract powder containing 6-MSITC is safe. Conclusion This study confirmed the possibility that this powder extracted from wasabi that contains 6-MSITC might improve fatigue and sleep. However, because the effectiveness evaluation in this study was a single-arm, open-label study and there was no placebo control group, these points must be considered when interpreting the results. Safety was confirmed in an overdose study of more than three times the amount compared to that in the efficacy evaluation study. In the future, further research should be conducted on its effectiveness for treating fatigue and sleep problems. Trial registration UMIN clinical trial registration system, UMIN000049913. Polychlorinated biphenyls (PCBs) persistently pollute marine ecosystems even though their production has been discontinued decades ago. Deep-sea organisms have been shown to accumulate PCBs and other persistent organic compounds; however, the extent of contamination and its effects remain poorly understood. Here, we measured PCB concentrations in separated organs of non-feeding bivalve clams belonging to Phreagena, a taxon representative of deep-sea chemosynthetic ecosystems. The highest concentrations of PCBs were detected in the ovaries. We also examined surface sediments of the clam habitat and observed concentrations comparable to those in the sediments of the seas around Japan, including shallow waters. The results showed that PCBs from the environment penetrate Phreagena clams through the body surface and accumulate in the female reproductive system. This raises concerns about a potentially severe direct impact on the survival of vulnerable and highly endemic deep-sea chemosynthetic bivalves.

With the high number of microplastic-like particles captured by net hauls including manta or neuston nets, it is often required to subsample in order to decrease sample volume for microplastic enumeration and analysis. Plankton splitter is commonly used to divide microplastic samples. However, current devices such as Folsom plankton splitter and Motoda box splitter have accuracy issues in separating highly buoyant microplastics, namely expanded polystyrene (EPS) as they tend to adhere to the inner walls. Inspired by an apple cutter, we have developed a simple radial splitter made of stainless steel that efficiently divides EPS microplastic samples into precise aliquots. With this simple device, we uniformly divided EPS microplastic samples from marine environments into eight aliquots with no significant differences. The device is a versatile tool to partition all buoyant microplastics including polypropylene and polyethylene microplastics. • The method developed facilitates the precise division of buoyant microplastics into equal aliquots. • The method is specifically effective in splitting expanded polystyrene particles with high buoyancy.

The microplastics inflow into the Arctic Ocean may increase environmental stress on the Arctic marine ecosystem on the Pacific side, where sea ice has been significantly reduced because of global warming. However, quantitative data on microplastics are very limited in the Beaufort Sea, which is covered by sea ice for most of the year, even in summer. We therefore observed microplastic concentrations over a wide area of the southern Beaufort Sea using a neuston net from 30 August to 10 September 2022 to estimate the total number (particle inventory) and mass (mass inventory) of microplastics in the entire water column. The particle inventory during the sampling period ranged from 937 to 28,081 pieces km−2 (mean ± standard deviation, 7570 ± 7600 pieces km−2). The mass inventory of microplastics ranged from 22 to 664 g km−2 (179 ± 180 g km−2). Relatively high microplastic inventories (>6500 pieces km−2) were observed off Utqiaġvik and the mouth of the Mackenzie River, suggesting that some microplastics originate not only in the Pacific but also in the Arctic. These values indicate that waters in the southern Beaufort Sea is contaminated with microplastics to the same order of magnitude as the Chukchi Sea.

Abstract Background Wasabi (Eutrema japonicum) is a common pungent spice used in Japan. 6-Methylsulfinylhexyl isothiocyanate (6-MSITC) found in the rhizome of wasabi has been shown to have anti-inflammatory and antioxidant effects, as well as improve neuroinflammation and memory. Therefore, we hypothesized that these effects would be beneficial for treating myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS). The present study was conducted to investigate the effectiveness of wasabi extract containing 6-MSITC on ME/CFS in an open-label trial. Methods Fifteen patients (3 males, 12 females, 20–58 years old) were orally administered wasabi extract (9.6 mg of 6-MSITC/day) for 12 weeks. The following parameters and test results were compared pre- and post-treatment: performance status (PS), self-rating questionnaires, pressure pain threshold (PPT) on the occiput, Trail Making test-A (TMT-A), and hemodynamic patterns determined by an active standing test. Results After treatment with 6-MSITC, PS improved significantly (p = 0.001). Although the scores on the 11-item Chalder Fatigue scale (CFS-11) and numerical rating scale (NRS) of fatigue did not show significant changes, subjective symptoms improved significantly, including headache frequency (4.1 to 3.0 times/week, p = 0.001) and myalgia (4.1 to 2.4 times/week, p = 0.019), NRS brain fog scores (5.7 to 4.5, p = 0.011), difficulty finding appropriate words (4.8 to 3.7, p = 0.015), photophobia (4.8 to 3.5, p = 0.008), and the Profile of Mood Status vigor score (46.9 to 50.0, p = 0.045). The PPT of the right occiput (17.3 to 21.3 kPa, p = 0.01) and TMT-A scores (53.0 to 38.1 s, p = 0.007) also changed, suggesting reduced pain sensitivity, and improved cognitive function, respectively. Orthostatic patterns determined by a standing test did not show remarkable changes. There were no serious adverse reactions. Conclusion This study suggests that 6-MSITC improves PS as well as subjective symptoms such as pain and cognitive dysfunction, and psychological vitality of patients with ME/CFS. It also improved cognitive performance and increased pain thresholds in these patients. 6-MSITC may be a promising therapeutic option especially for improving cognitive dysfunction associated with ME/CFS.

Summary: The accumulation of polybrominated diphenyl ethers (PBDEs) and benzotriazole UV stabilizers (BZT-UVs) were examined in the hadal amphipod Hirondellea gigas caught from a near-land trench off the Japan island (9200 m). H. gigas were collected from two distinct sites: one is located at the outlet of submarine canyons directly connected to land and the other is apart from the outlet and geographically isolated from the first site. The level of the PBDEs in H. gigas at the canyon outlet (mean 219 ng/g lipid weight (l.w.)) was significantly higher than that in the isolated site (mean 42 ng/g l.w.) and BZT-UVs were only detected within the first site (mean 1.5 ng/g wet weight). In addition to vertical transport from the surface water, near-land trenches associated with submarine canyons and troughs may have more complex influx of contaminants through horizontal transportation from the land, resulting in more severe contamination.

It is becoming obvious that the abundance of microplastics is increasing in worldwide oceans, raising concerns about their impact on marine ecosystems. Tiny plastic particles enter the body of marine organisms not only via oral ingestion but also through the body surface (e.g., gills or epidermis), but the mechanism of internalization into cells is poorly understood. In this study, we conducted experiments using deep-sea chemosynthetic mussels with limited feeding by exposing their gills to fluorescently labeled microplastic beads. We identified the gill cell types that preferentially internalized the beads and demonstrated the inhibitory effect of phagocytosis inhibitors on bead uptake. Furthermore, using correlative light-electron microscopy, we microhistologically verified that beads were enclosed within membrane-bound vacuoles. Our results indicated that microplastic particles were internalized into gill cells of deep-sea and coastal mussels by phagocytosis. This study highlights the need for further research on plastic contamination via the body surface to conserve the highly endemic and vulnerable deep-sea fauna and mitigate human health risks from consuming coastal bivalves.

Acute myocarditis is a rare but serious complication associated with mRNA-based coronavirus disease 2019 (COVID-19) vaccination. In this article, four COVID-19 mRNA vaccination induced myocarditis cases managed at our tertiary Medical Center have been discussed. Three patients had typical myocarditis. One patient suffered from atrioventricular block and heart failure, which required more intensive treatment, but eventually improved. Additionally, a review of cardiac magnetic resonance imaging (MRI) features related to the diagnosis of myocarditis showed that COVID-19 mRNA vaccine-associated myocarditis tend to have more late-gadolinium enhancement (LGE) accumulation in the inferior lateral wall direction. According to a report by the U.S. Centers for Disease Control and Prevention (CDC), the diagnosis of COVID-19 mRNA vaccine-associated myocarditis is based on clinical symptoms, altered myocardial enzymes, cardiac MRI finding, or histopathology. Cardiac MRI is relatively less invasive than myocardial biopsy and plays an important role in the diagnosis of myocarditis. This review may aid in the diagnosis of COVID-19 mRNA vaccine-associated myocarditis.

Extreme storms, such as tropical cyclones, are responsible for a significant portion of the plastic debris transported from land to sea yet little is known about the storm response of microplastics and other debris in offshore and open waters. To investigate this, we conducted floating plastic surveys in the center of Sagami Bay, Japan approximately 30 km from the coastline, before and after the passage of a typhoon. The concentrations (number of particles/km2) of micro- and mesoplastics were two orders of magnitude higher 1-day after the typhoon than the values recorded pre-typhoon and the mass (g/km2) of plastic particles (sum of micro- and mesoplastics) increased 1,300 times immediately after the storm. However, the remarkably high abundance of micro- and mesoplastics found at 1-day after the typhoon returned to the pre-typhoon levels in just 2 days. Model simulations also suggested that during an extreme storm a significant amount of micro- and mesoplastics can be rapidly swept away from coastal to open waters over a short period of time. To better estimate the annual load of plastics from land to sea it is important to consider the increase in leakages of plastic debris into the ocean associated with extreme storm events.

Marine ecosystems are continuously subjected to anthropogenic environmental pollution. Understanding the spread of pollution and the potential risks it poses to deep-sea ecosystems is important for developing better conservation measures. Here, we identified non-negligible levels of persistent organic pollutants in deep-sea chemosynthetic bivalves with limited or no filter feeding. The bivalves were collected from two sites: one located near a highly populated region and the other located relatively far from human activity. Analyses of samples collected nearly every decade in a period of 30 years suggested that environmental policy restrictions might be effective in reducing chemical pollution. However, the detection of contamination in deep-sea chemosynthetic animals suggests that the pollution could be spreading globally to chemosynthetic organisms with limited or no feeding. To protect these highly endemic and vulnerable deep-sea chemosynthetic ecosystems, our findings indicate that further research on chemical contamination and its effects on these ecosystems is required.

Removing non-plastic materials is a mandatory process for studying microplastics in environmental samples, and non-plastic materials, both inorganic and organic matter, are often removed chemically through sequential processes. In the multiple chemical treatment processes, the samples need to be collected and the reagent removed at the end of each chemical treatment before the samples are again exposed to a different reagent in a separate container. This leads to a loss of microplastics to some extent. Here, we developed a new, yet simple, small sieve made of stainless-steel that can fit in a laboratory beaker (e.g. 200 ml volume), allowing it to be transferred as-is between chemical treatments of environmental samples, even being soakable in a beaker of acid solution. The collection rates of microplastics were significantly higher in the small stainless-steel sieve than the commonly used filter method for different size of microplastic particles. The use of the new sieve means the processes of rinsing off and filtering samples can be abbreviated throughout the entire process of non-plastic matter removal from environmental samples, contributing to a lower chance of microplastic loss. The time consumed in the sieve method was also significantly lower than for the filtering method due to the elimination of the collection and rinsing steps, thus the use of this sieve can reduce processing time for the samples. Microplastics are abundant even on the deep-sea floor far from land and the ocean surface where human activities take place. To obtain samples of microplastics from the deep-sea floor, a research vessel and suitable sampling equipment, such as a multiple corer, a box corer, or a push corer manipulated by a remotely operated (ROV) or human occupied vehicle (HOV) are needed. Most such corers use sampling tubes made of plastic, such as polycarbonate, acrylic, or polyvinyl chloride. These plastic tubes are easily scratched by sediment particles, in particular during collection of coarse sandy sediments, and, consequently, the samples may become contaminated with plastic from the tube. Here, we report on the use of aluminum tubes with both a multiple corer and a push corer to prevent such plastic contamination. When compared with plastic tubes, aluminum tubes have the disadvantages of heavier weight and non-transparency. Plastic pollution has become one of the most emergent issues threating aquatic and terrestrial ecosystems. However, it is still challenging to rapidly detect small microplastics. Here, we present a method to rapidly detect microplastics using hyperspectral imaging in which we optimized a commercially available hyperspectral imaging system (Pika NIR-640, Resonon Inc., USA). The optimizations included: (1) changing the four-lamp assembly to a symmetrical set of converged-light near-infrared lamps that are placed sideways instead of above the sample stage; (2) adopting a macro-photography technique by applying an extension tube between the camera and the lens, and moving the lens of the hyperspectral camera to the imaging target (working distance of ~3 cm); (3) adjusting the exposure and aspect ratio by tuning the frame rate and scan speed of the imaging system. After optimization, the detection resolution of each pixel improved from 250 µm to 14.8 µm. With the optimized system, microplastics down to 100 µm in size were rapidly detected. This result is promising for the application of our new method in the accelerated detection of microplastics and will contribute to a better understanding of the microplastic pollution situation. • A commercially available hyperspectral imaging system is optimized for rapid detection of microplastics. • Infrared lamps are used and macro-photography technique adopted along with adjustments of exposure. • Microplastics down to 100 µm in size could be detected with the optimized system.

Abstract 3D visual mapping of the seafloor has found applications ranging from environment monitoring and survey of marine minerals to underwater archaeology and inspection of modern artificial structures. However, the attenuation of light is significantly more pronounced in water than in air or in space, and so in order to obtain underwater images in colour, it is typically necessary to be within 2–3 m of the seafloor. In addition to the high risk of collision when operating underwater vehicles at such low altitudes, the limited area of the seafloor covered in each image means large area surveys require a significant investment of time. In this research, we aim to increase the efficiency of mapping large areas of the seafloor by developing an underwater imaging system that can take colour images at ranges of more than 10 m, so that each image can cover a larger area, together with the necessary algorithms to automatically process the data it obtains. The system was deployed to map artificial hydrothermal vents in Iheya North field using the ROV Hyper-Dolphin in October 2012. The surveyed area is of particular interest to the research community, as multiple artificial vent holes were drilled during a mission in 2010, which locally impacted the flow of hydrothermal fluids. In this paper, we describe the methods used to process the data that the imaging system obtains and demonstrate how the mapping data can be used in quantitative studies of the seafloor. Habitats of Shinkaia crosnieri squat lobsters, which are abundant in the hydrothermally active areas, are identified in the maps and their population density calculated, and the amount of hydrothermal deposits that have grown on the artificial vent is derived from the mapping data. The work demonstrates how 3D visual mapping can be applied to benthic biology and geological studies.

Separating microplastics from marine and freshwater sediments is challenging, but necessary to determine their distribution, mass, and ecological impacts in benthic environments. Density separation is commonly used to extract microplastics from sediments by using heavy salt solutions, such as zinc chloride and sodium iodide. However, current devices/apparatus used for density separation, including glass beakers, funnels, upside-down funnel-shaped separators with a shut-off valve, etc., possess various shortcomings in terms of recovery rate, time consumption, and/or usability. In evaluating existing microplastic extraction methods using density separation, we identified the need for a device that allows rapid, simple, and efficient extraction of microplastics from a range of sediment types. We have developed a small glass separator, without a valve, taking a hint from an Utermöhl chamber. This new device is easy to clean and portable, yet enables rapid separation of microplastics from sediments. With this simple device, we recovered 94–98% of

The boundary zone between two different hydrological regimes is often a biologically enriched environment with distinct planktonic communities. In the center of the Amazon River basin, muddy white water of the Amazon River meets with black water of the Negro River, creating a conspicuous visible boundary spanning over 10 km along the Amazon River. Here, we tested the hypothesis that the confluence boundary between the white and black water rivers concentrates prey and is used as a feeding habitat for consumers by investigating the density, biomass and distribution of mesozooplankton and ichthyoplankton communities across the two rivers during the rainy season. Our results show that mean mesozooplankton density (2,730 inds. m−3) and biomass (4.8 mg m−3) were higher in the black-water river compared to the white-water river (959 inds. m−3; 2.4 mg m−3); however an exceptionally high mesozooplankton density was not observed in the confluence boundary. Nonetheless we found the highest density of ichthyoplankton in the confluence boundary (9.7 inds. m−3), being up to 9-fold higher than in adjacent rivers. The confluence between white and black waters is sandwiched by both environments with low (white water) and high (black water) zooplankton concentrations and by both environments with low (white water) and high (black water) predation pressures for fish larvae, and may function as a boundary layer that offers benefits of both high prey concentrations and low predation risk. This forms a plausible explanation for the high density of ichthyoplankton in the confluence zone of black and white water rivers.

There has been an increasing interest in seafloor exploitation such as mineral mining in deep-sea hydrothermal fields, but the environmental impact of anthropogenic disturbance to the seafloor is poorly known. In this study, the effect of such anthropogenic disturbance by scientific drilling operations (IODP Expedition 331) on seabed landscape and megafaunal habitation was surveyed for over 3 years using remotely operated vehicle video observation in a deep-sea hydrothermal field, the Iheya North field, in the Okinawa Trough. We focused on observations from a particular drilling site (Site C0014) where the most dynamic change of landscape and megafaunal habitation was observed among the drilling sites of IODP Exp. 331. No visible hydrothermal fluid discharge had been observed at the sedimentary seafloor at Site C0014, where Calyptogena clam colonies were known for more than 10 years, before the drilling event. After drilling commenced, the original Calyptogena colonies were completely buried by the drilling deposits. Several months after the drilling, diffusing high-temperature hydrothermal fluid began to discharge from the sedimentary subseafloor in the area of over 20 m from the drill holes, 'artificially' creating a new hydrothermal vent habitat. Widespread microbial mats developed on the seafloor with the diffusing hydrothermal fluids and the galatheid crab Shinkaia crosnieri endemic to vents dominated the new vent community. The previously soft, sedimentary seafloor was hardened probably due to barite/gypsum mineralization or silicification, becoming rough and undulated with many fissures after the drilling operation. Although the effects of the drilling operation on seabed landscape and megafaunal composition are probably confined to an area of maximally 30 m from the drill holes, the newly established hydrothermal vent ecosystem has already lasted 2 years and is like to continue to exist until the fluid discharge ceases and thus the ecosystem in the area has been altered for long-term.

Previous studies have suggested that Crown-of-Thorns starfish (COTS) larvae may be able to survive in the absence of abundant phytoplankton resources suggesting that they may be able to utilize alternative food sources. Here, we tested the hypothesis that COTS larvae are able to feed on coral-derived organic matter using labeled stable isotope tracers (13C and 15N). Our results show that coral-derived organic matter (coral mucus and associated microorganisms) can be assimilated by COTS larvae and may be an important alternative or additional food resource for COTS larvae through periods of low phytoplankton biomass. This additional food resource could potentially facilitate COTS outbreaks by reducing resource limitation.
