A certain wavelength of incident light is forbidden based on the optical periodicity related to the intervals and refractive indices of the comprising materials. For multi-bilayered films with periodicity, on–off switching of reflection is possible due to the presence or absence of a refractive index difference between two consisting films. Changes in the refractive index can be induced by stimuli-responsive orientation behaviors of liquid crystal (LC) molecules with birefringence. Azobenzene (Az) groups are employed for photo response and copolymerized with non-photo-responsive LC mesogen groups for cooperative orientation to improve light penetration by the dilution of Az concentration. LC mesogen groups are designed to have a high refractive index to improve the reflection contrast. The refractive indices and the orientation behaviors of the copolymer films are investigated. On–off switching of reflection for multi-bilayered films consisting of alternate Az-containing LC copolymers and poly vinyl alcohol is conducted using ultraviolet and visible light irradiation, and their reflection intensity and response speed are compared between the different LC mesogen types and molar ratios.

CRISPR–Cas9 enables recombination between homologous chromosome arms at predefined sites and also underscores the need for caution when applying CRISPR technologies in translational medicine., CRISPR–Cas9–based genome editing has transformed the life sciences, enabling virtually unlimited genetic manipulation of genomes: The RNA-guided Cas9 endonuclease cuts DNA at a specific target sequence and the resulting double-strand breaks are mended by one of the intrinsic cellular repair pathways. Imprecise double-strand repair will introduce random mutations such as indels or point mutations, whereas precise editing will restore or specifically edit the locus as mandated by an endogenous or exogenously provided template. Recent studies indicate that CRISPR-induced DNA cuts may also result in the exchange of genetic information between homologous chromosome arms. However, conclusive data of such recombination events in higher eukaryotes are lacking. Here, we show that in Drosophila, the detected Cas9-mediated editing events frequently resulted in germline-transmitted exchange of chromosome arms—often without indels. These findings demonstrate the feasibility of using the system for generating recombinants and also highlight an unforeseen risk of using CRISPR-Cas9 for therapeutic intervention.

A novel technique for the continuous extraction of nickel (Ni) from Ni-based superalloy scraps using molten zinc (Zn) has been proposed, and its feasibility was experimentally demonstrated. The newly developed approach allows for extraction of Ni metal directly from superalloy scraps with simultaneous separation of the Zn from the resulting Zn-Ni alloy. The optimal conditions for the extraction of Ni and separation of valuable elements such as rhenium (Re), tantalum (Ta), and tungsten (W) were determined by varying major process parameters including the reaction time and configuration of the reaction chamber. The proposed method has been successfully utilized for the production of the superalloy containing 62.8 mass pct of Ni and 15.5 mass pct of refractory metals (Re, W, and Ta). Under certain conditions, 41 pct of the Ni contained in the superalloy could be extracted at 1173 K (900 °C) over 48 hours, producing an alloy containing 84.0 mass pct of Ni and 0.2 mass pct of the refractory metals.

Multi-bilayered films with precisely controlled layer thickness were constructed for structural color and their reflection angular dependences were investigated. For wide angle structural color, it was intended to the macro irregularity of multi-bilayered films by randomly dispersed micron size multi-bilayered films in PDMS matrix. In addition, on-off switching of the reflection was examined with respect to the molecular orientation behavior of azobenzene chromophores by annealing and light irradiation.

With the purpose of developing a new process for recycling nickel (Ni) directly from superalloy scraps, a fundamental study on the extraction and separation of Ni was carried out using molten zinc (Zn) as the extraction medium. In order to examine the reaction between molten Zn and the Ni-based superalloy, superalloy samples and Zn shots were heated at 1173 K (900 °C) for 6 hours. After heating, the superalloy samples fully reacted with Zn and dissolved in molten Zn. The Zn-alloyed sample obtained by slow cooling consisted of two separated upper and lower phases. In the upper part of the sample, only Zn and the Zn-Ni alloys were found; in the lower part, an intermetallic alloy consisting of refractory metals such as rhenium (Re) and tantalum (Ta) was found. This result shows that Ni and refractory metals contained in the scrap can be separated by utilizing the density differences between the Zn-Ni alloy and the refractory metals in molten Zn. Vacuum treatment of the upper part of the Zn-alloyed sample at 1173 K (900 °C) reduced the concentration of Zn in the sample from 97.0 to 0.4 mass pct. Synthesis of chiral inorganic or hybrid nanomaterials through sol–gel transcription of chiral organic templates has attracted a great deal of interest for more than a decade. However, the chiral nature of these inorganic matrices has never been directly observed. For the first time, we report a direct evaluation of chirality on noncrystalline silica chiral nanoribbons by vibrational circular dichroism (VCD) measurements. Strong Cotton effect around 1150–1000 cm–1 from Si—O—Si asymmetric stretching vibration was observed. Surprisingly, calcination of these hybrid nanoribbons doubled the intensity of Cotton effects. On the basis of transmission electron microscopy observations, IR, VCD, NMR, and Raman spectroscopies, we demonstrate that the silica chirality originates from twisted siloxane network composed of chiral arrangement of the Si—O—Si bonds. Dielectric multilayer including azobenzene polymer liquid crystal acts as a one-dimensional photonic crystal and has optical-switchable reflection bands. The reflection intensity can be controlled by irradiation of ultraviolet (UV) or visible light, in which the optical switch is based on cis-trans photoisomerization. An increase in the number of layers prevents photoresponse in the multilayer including azobenzen layers because UV or visible light for control cis-trans isomerization hardly reach the bottom of the multilayer due to exponential decay of light intensity. In this study, the multilayer consisting of thin azobenzene layers is investigated. According to numerical calculations, the non-quarter-wave structured multilayer has multiwavelength reflection band. Angular dependence of reflection color from multiwavelength stopband is also studied using the XYZ color space. The non-quarter-wave multilayer suppresses reflection color change compared with conventional quarter-wave multilayer.

In this paper, the change in structural color of multi-bilayered films fabricated by stacking copolymers, PMAz–PMBP (m : n), consisting of azobenzene and biphenyl side chain groups, and polyvinyl alcohol (PVA) alternately, was investigated. The multi-bilayered films consisting of polyacrylates having an azobenzene side chain group and PVA were found to reflect a light of specific wavelength, and to show reversible change in the reflection intensity by thermal annealing and irradiation with non-polarized ultra violet (UV) light. However, a long irradiation time was required for the change in reflection intensity of the multi-bilayered films. Therefore, we synthesized PMAz–PMBP (m : n), consisting of azobenzene and biphenyl side chain group having no absorbance over 300 nm, and the PMAz–PMBP (m : n)/PVA multi-bilayered films were fabricated. All the multi-bilayered films showed reversible change in the reflection intensity by thermal annealing and irradiation with non-polarized UV light. The re-coloration speed of PMAz–PMBP (75 : 25)/PVA 20-multibilayered film was faster than others fabricated in this study. It is related to penetration depth of light into the films. The effect of the introduction of the biphenyl side groups in the copolymers on the response behavior of the change in the structural color will be discussed.

In this paper, photoresponsive behavior of multi-bilayered films having precisely controlled layer thickness prepared by stacking an azo-functionalized polymer liquid crystal, PMAzXAc, and polyvinyl alcohol alternatively, PVA, is described. The multi-bilayered films were found to reflect a light of specific wavelength depending on the layer thickness and refractive index, and showed the reversible change in the reflection intensity by irradiation with visible and UV lights. The change in the reflection intensity was brought about by change in the molecular orientation of PMAzXAc between an out-of-plane orientation and a photo-induced isotropic state, and was strongly dependent on the number of methylene spacer of PMAzXAc linking the azobenzene side group with the acrylate polymer main chain. PMAz6Ac with hexa-methylene spacer showed the largest change in the reflection intensity, while smaller change in the reflection intensity was observed for PMAzXAc having shorter or longer methylene spacer than 6. The effect of the methylene spacers on the photochemical change in the molecular orientation of azobenzene chromophores in the multi-bilayered films will be discussed.

Three type polymers containing the same azobenzene side chain group with different backbone structures, poly(DMAz6Fm), poly(DMAz6It) and poly(MAz6Ac) derived respectively from fumaric acid, itaconic acid and acrylic acid, were synthesized. The dependence of photo-response properties on the polymeric architecture was investigated for the solutions and films. By irradiation of the linearly polarized light (436 nm), uniaxial orientations of the thin films were induced and anisotropic properties were investigated by measurements of the order parameters and birefringence values.

Polyethylene imines (PEIs) with azobenzene side groups, PEI6M and IP-DAz, were synthesized and the effects of the substitution degree of the PEI backbone structures on the photoresponse properties and the orientation behaviors were investigated. The phase transition temperatures are closely related to thermal stability in orientational order. IP-DAz showed only the smectic phase in a much broader range between 53 °C and 170 °C and the thermal stability was significantly improved compared with PEI6M. Reversible changes in absorption spectra relating to photoisomerization behavior for both polymers were observed by UV and visible light irradiation. Uniaxial orientation for the selective direction was induced by irradiation by linearly polarized light and the anisotropic properties were investigated by measurements of the order parameter and birefringence. Rapid photoresponses for molecular orientations were demonstrated for both polymers. Although the order parameter of IP-DAz was lower than that of PEI6M, the birefringence of IP-DAz for in-plane orientation was as high as for PEI6M and is related to the stable orientational order. © 2013 Society of Chemical Industry

Modulation of the refractive index of a polymer was achieved by combining it with diamond nanoparticles (NDs). The increase in the refractive index was controlled by the amount of NDs added, according to the Lorentz-Lorenz equation. The refractive index of poly(vinyl alcohol) (PVA), which was used as the base polymer, increased from 1.52 to 1.88. A multilayer film consisting of alternating layers of ND-PVA composite and poly(methyl methacrylate) exhibited ca. 80% reflectance with 10 bilayers.

RhoA activity is transiently inhibited at the initial phase of integrin engagement, when Cdc42- and/or Rac1-mediated membrane spreading and ruffling predominantly occur. Paxillin, an integrin-assembly protein, has four major tyrosine phosphorylation sites, and the phosphorylation of Tyr31 and Tyr118 correlates with cell adhesion and migration. We found that mutation of Tyr31/118 caused enhanced activation of RhoA and premature formation of stress fibers with substantial loss of efficient membrane spreading and ruffling in adhesion and migration of NMuMG cells. These phenotypes were similar to those induced by RhoA(G14V) in parental cells, and could be abolished by expression of RhoA(T19N), Rac1(G12V), or p190RhoGAP in the mutant-expressing cells. Phosphorylated Tyr31/118 was found to bind to two src homology (SH)2 domains of p120RasGAP, with coprecipitation of endogenous paxillin with p120RasGAP. p190RhoGAP is known to be a major intracellular binding partner for the p120RasGAP SH2 domains. We found that Tyr31/118-phosphorylated paxillin competes with p190RhoGAP for binding to p120RasGAP, and provides evidence that p190RhoGAP freed from p120RasGAP efficiently suppresses RhoA activity during cell adhesion. We conclude that Tyr31/118-phosphorylated paxillin serves as a template for the localized suppression of RhoA activity and is necessary for efficient membrane spreading and ruffling in adhesion and migration of NMuMG cells.

Paxillin is a protein containing four LIM domains, and functions in integrin signaling. We report here that two transcripts are generated from the paxillin gene locus in Drosophila; one encodes a protein homolog of the vertebrate Paxillin (DPxn37), and the other a protein with only three LIM domains, partly encoded by its own specific exon (PDLP). At the myotendinous junctions of Drosophila embryos where integrins play important roles, both DPxn37 and PDLP are highly expressed with different patterns; DPxn37 is predominantly concentrated at the center of the junctions, whereas PDLP is highly enriched at neighboring sides of the junction centers, primarily expressed in the mesodermal myotubes. Northern blot analysis revealed that DPxn37 is ubiquitously expressed throughout the life cycle, whereas PDLP expression exhibits a biphasic pattern during development, largely concomitant with muscle generation and remodeling. Our results collectively reveal that a unique system exists in Drosophila for the generation of a novel type of LIM-only protein, highly expressed in the embryonic musculature, largely utilizing the Paxillin LIM domains.

Paxillin acts as an adaptor protein in integrin signaling. We have shown that paxillin exists in a relatively large cytoplasmic pool, including perinuclear areas, in addition to focal complexes formed at the cell periphery and focal adhesions formed underneath the cell. Several ADP-ribosylation factor (ARF) GTPase-activating proteins (GAPs; ARFGAPs) have been shown to associate with paxillin. We report here that Git2-short/KIAA0148 exhibits properties of a paxillin-associated ARFGAP and appears to be colocalized with paxillin, primarily at perinuclear areas. A fraction of Git2-short was also localized to actin-rich structures at the cell periphery. Unlike paxillin, however, Git2-short did not accumulate at focal adhesions underneath the cell. Git2-short is a short isoform of Git2, which is highly homologous to p95PKL, another paxillin-binding protein, and showed a weaker binding affinity toward paxillin than that of Git2. The ARFGAP activities of Git2 and Git2-short have been previously demonstrated in vitro, and we provided evidence that at least one ARF isoform, ARF1, is an intracellular substrate for the GAP activity of Git2-short. We also showed that Git2-short could antagonize several known ARF1-mediated phenotypes: overexpression of Git2-short, but not its GAP-inactive mutant, caused the redistribution of Golgi protein β-COP and reduced the amounts of paxillin-containing focal adhesions and actin stress fibers. Perinuclear localization of paxillin, which was sensitive to ARF inactivation, was also affected by Git2-short overexpression. On the other hand, paxillin localization to focal complexes at the cell periphery was unaffected or even augmented by Git2-short overexpression. Therefore, an ARFGAP protein weakly interacting with paxillin, Git2-short, exhibits pleiotropic functions involving the regulation of Golgi organization, actin cytoskeletal organization, and subcellular localization of paxillin, all of which need to be coordinately regulated during integrin-mediated cell adhesion and intracellular signaling.

The highly conserved and ubiquitously expressed 14-3-3 proteins regulate differentiation, cell cycle progression and apoptosis by binding intracellular phosphoproteins involved in signal transduction. By screening in vitro translated cDNA pools for the ability to bind 14-3-3, we identified a novel transcriptional co-activator, TAZ (transcriptional co-activator with PDZ-binding motif) as a 14-3-3-binding molecule. TAZ shares homology with Yes-associated protein (YAP), contains a WW domain and functions as a transcriptional co-activator by binding to the PPXY motif present on transcription factors. 14-3-3 binding requires TAZ phosphorylation on a single serine residue, resulting in the inhibition of TAZ transcriptional co-activation through 14-3-3-mediated nuclear export. The C-terminus of TAZ contains a highly conserved PDZ-binding motif that localizes TAZ into discrete nuclear foci and is essential for TAZ-stimulated gene transcription. TAZ uses this same motif to bind the PDZ domain-containing protein NHERF-2, a molecule that tethers plasma membrane ion channels and receptors to cytoskeletal actin. TAZ may link events at the plasma membrane and cytoskeleton to nuclear transcription in a manner that can be regulated by 14-3-3.

In this study, faster response speed of ON/OFF switching of multi-bilayered films containing azo-functionalized polymer liquid crystal and polyvinyl alcohol, PVA, is investigated. The multi-bilayered films were found to reflect a light of specific wavelength, and showed reversible change in the reflection intensity by irradiation with non-polarized visible light and UV light. The multi-bilayered film having high absorbance around 300-500 nm due to stacking several decades of azobenzene containing layer. So, the change in reflection intensity of multi-bilayered film takes long time by light irradiation. We synthesized polyacrylates copolymers with azobenzene and biphenyl side chains where biphenyl groups is having no absorbance at UV and visible region. Multi-bilayered films were fabricated by spin-coating method and the switching time of reflection was investigated comparing to the multi-bilayered film containing azobenzene homopolymer, the ON switching times of reflection for the copolymers were faster than homopolymer needing 900 s. It is attributed to that inducing UV light (λ = 365 nm) easily entered into the film by introducing of biphenyl groups.

alpha-2'-Deoxyadenosine (alpha) is a major adenine lesion produced by gamma-ray irradiation of DNA under anoxic conditions. In this study, single-stranded recombinant M13 vectors containing alpha were constructed and transfected into Escherichia coli to assess lethal and mutagenic effects of this lesion. The data for alpha were further compared with those obtained with M13 vectors containing normal A or a model abasic site (F) at the same site. The transfection assay revealed that alpha constituted a moderate block to DNA replication. The in vivo replication capacity to pass through alpha was approximately 20% relative to normal A, but 20-fold higher than that of F constituting an almost absolute replication block. Similar data were obtained by in vitro replication of oligonucleotide templates containing alpha or F by E.coli DNA polymerase I. The mutagenic consequence of replicating M13 DNA containing alpha was analyzed by direct DNA sequencing of progeny phage. Mutagenesis was totally targeted at the site of alpha introduced into the vector. Mutation was exclusively a single nucleotide deletion and no base substitutions were detected. The deletion frequency associated alpha was dependent on the 3'-nearest neighbor base: with the 3'-nearest neighbor base T mutation (deletion) frequency was 26%, whereas 1% with the 3'-nearest neighbor base G. A possible mechanism of the single nucleotide deletion associated with alpha is discussed on the basis of the misinsertion-strand slippage model.

A simultaneous quantitative analytical method for 13 stilbenoids including (−)-hopeaphenol (1), (+)-isohopeaphenol (2), hemsleyanol D (3), (−)-ampelopsin H (4), vaticanols A (5), E (6), and G (7), (+)-α-viniferin (8), pauciflorol A (9), hopeafuran (10), (−)-balanocarpol (11), (−)-ampelopsin A (12), and trans-resveratrol 10-C-β-d-glucopyranoside (13), and two dihydroisocoumarins, phayomphenols A1 (14) and A2 (15) in the extract of Shorea roxburghii (dipterocarpaceae) was developed. According to the established protocol, distributions of these 15 polyphenols (1–15) in the bark and wood parts of S. roxburghii and a related plant Cotylelobium melanoxylon were evaluated. In addition, the principal polyphenols (1, 2, 8, 13–15) exhibited hepatoprotective effects against d-galactosamine (d-galN)/lipopolysaccharide (LPS)-induced liver injury in mice at a dose of 100 or 200 mg/kg, p.o. To characterize the mechanisms of action, the isolates were examined in in vitro studies assessing their effects on (i) d-GalN-induced cytotoxicity in primary cultured mouse hepatocytes; (ii) LPS-induced nitric oxide (NO) production in mouse peritoneal macrophages; and (iii) tumor necrosis factor-α (TNF-α)-induced cytotoxicity in L929 cells. The mechanisms of action of these polyphenols (1, 2, and 8) were suggested to be dependent on the inhibition of LPS-induced macrophage activation and reduction of sensitivity of hepatocytes to TNF-α. However, none of the isolates reduced the cytotoxicity caused by d-GalN.

Pleocytosis of the cerebrospinal fluid is a key finding for the diagnosis of bacterial meningitis. Bacterial meningitis presenting in normal cerebrospinal fluid is rare in adult patients. We describe the case of a patient with pneumococcal meningitis without cerebrospinal fluid pleocytosis. This case suggests that immediate antibiotic therapy should be started when meningitis is suspected, even with normal cerebrospinal fluid findings. (See Figure.)

We use the Dpp morphogen gradient in the Drosophila wing disc as a model to address the fundamental question of how a gradient of a growth factor can produce uniform growth. We first show that proper expression and subcellular localization of components in the Fat tumor-suppressor pathway, which have been argued to depend on Dpp activity differences, are not reliant on the Dpp gradient. We next analyzed cell proliferation in discs with uniformly high Dpp or uniformly low Fat signaling activity and found that these pathways regulate growth in a complementary manner. While the Dpp mediator Brinker inhibits growth in the primordium primarily in the lateral regions, Fat represses growth mostly in the medial region. Together, our results indicate that the activities of both signaling pathways are regulated in a parallel rather than sequential manner and that uniform proliferation is achieved by their complementary action on growth.

The use of binary transcriptional systems offers many advantages for experimentally manipulating gene activity, as exemplified by the success of the Gal4/UAS system in Drosophila . To expand the number of applications, a second independent transactivator (TA) is desirable. Here, we present the optimization of an additional system based on LexA and show how it can be applied. We developed a series of LexA TAs, selectively suppressible via Gal80, that exhibit high transcriptional activity and low detrimental effects when expressed in vivo. In combination with Gal4, an appropriately selected LexA TA permits to program cells with a distinct balance and independent outputs of the two TAs. We demonstrate how the two systems can be combined for manipulating communicating cell populations, converting transient tissue-specific expression patterns into heritable, constitutive activities, and defining cell territories by intersecting TA expression domains. Finally, we describe a versatile enhancer trap system that allows swapping TA and generating mosaics composed of Gal4 and LexA TA-expressing cells. The optimized LexA system facilitates precise analyses of complex biological phenomena and signaling pathways in Drosophila .

Non-quarter-wave stacked dielectric multilayers including azobenzene polymer liquid crystal layers are investigated in this study. The azobenzene polymer liquid crystal has a photoinduced reversible refractive index based on photoisomerization. By using the reversible refractive-index change, the reflectance of a stopband can be controlled. In this system, the azobenzene molecules change their conformation when they are irradiated with ultraviolet (UV) or visible light. In general, stacking many layers of different thicknesses can produce broadband or multicolor reflections for a dielectric multilayer. However, in a multilayer having thick azobenzene layers, UV or visible light used for controlling photoisomerization hardly reaches the bottom part of the multilayer because the light is mainly absorbed at its top surface. To solve this problem, the dependence on the thickness ratio of the multilayer is investigated and a non-quarter-wave stacked multilayer having RGB reflections is experimentally demonstrated using thin azobenzene layers.

Previously we reported that AMAP2/PAG3/Papalpha/KIAA0400, a GTPase-activating protein (GAP), acts to antagonize Arf6 function when overexpressed, whereas it was shown to exhibit efficient GAP activities for other Arf isoforms in vitro. Here, we found that AMAP2, through its ArfGAP domain, binds to GTP-Arf6 but not to GDP-Arf6 or other Arfs irrespective of nucleotide status. The majority of AMAP2 was localized to intracellular tubulovesicular structures and redistributed to Arf6-enriched membrane areas upon Arf6 activation. In HeLa cells, Arf6 has been shown to be involved in the clathrin-independent endocytosis of Tac, but not the clathrin-dependent endocytosis of transferrin. We found that Arf6 silencing inhibited the internalization of Tac, but not transferrin, in HeLa cells. Internalization of Tac, but not transferrin, was also significantly inhibited by AMAP2 silencing and overexpression. AMAP2 was moreover found to bind to amphiphysin IIm, a component of the endocytic machinery, via its proline-rich domain. We propose that AMAP2 has dual mechanisms for its function; it exhibits efficient catalytic GAP activity for the class I and II Arfs and yet is involved in the cellular function of the class III Arf without immediate GAP activity. These dual mechanisms of AMAP2 may be important for the cellular function of GTP-Arf6.

A protein module called the WW domain recognizes and binds to a short oligopeptide called the PY motif, PPxY, to mediate protein-protein interactions. The PY motif is present in the transcription activation domains of a wide range of transcription factors including c-Jun, AP-2, NF-E2, C/EBPalpha and PEBP2/CBF, suggesting that it plays an important role in transcriptional activation. We show here that mutation of the PY motif in the subregion of the activation domain of the DNA-binding subunit of PEBP2, PEBP2alpha, abolishes its transactivation function. Using yeast two-hybrid screening, we demonstrate that Yes-associated protein (YAP) binds to the PY motif of PEBP2alpha through its WW domain. The C-terminal region of YAP fused to the DNA-binding domain of GAL4 showed transactivation as strong as that of GAL4-VP16. Exogenously expressed YAP conferred transcription-stimulating activity on the PY motif fused to the GAL4 DNA-binding domain as well as to native PEBP2alpha. The osteocalcin promoter was stimulated by exogenous PEBP2alphaA and a dominant negative form of YAP strongly inhibited this activity, suggesting YAP involvement in this promoter activity in vivo. These results indicate that the PY motif is a novel transcription activation domain that functions by recruiting YAP as a strong transcription activator to target genes.

The TGF-β homolog Decapentaplegic (Dpp) acts as a secreted morphogen in the Drosophila wing disc, and spreads through the target tissue in order to form a long range concentration gradient. Despite extensive studies, the mechanism by which the Dpp gradient is formed remains controversial. Two opposing mechanisms have been proposed: receptor-mediated transcytosis (RMT) and restricted extracellular diffusion (RED). In these scenarios the receptor for Dpp plays different roles. In the RMT model it is essential for endocytosis, re-secretion, and thus transport of Dpp, whereas in the RED model it merely modulates Dpp distribution by binding it at the cell surface for internalization and subsequent degradation. Here we analyzed the effect of receptor mutant clones on the Dpp profile in quantitative mathematical models representing transport by either RMT or RED. We then, using novel genetic tools, experimentally monitored the actual Dpp gradient in wing discs containing receptor gain-of-function and loss-of-function clones. Gain-of-function clones reveal that Dpp binds in vivo strongly to the type I receptor Thick veins, but not to the type II receptor Punt. Importantly, results with the loss-of-function clones then refute the RMT model for Dpp gradient formation, while supporting the RED model in which the majority of Dpp is not bound to Thick veins. Together our results show that receptor-mediated transcytosis cannot account for Dpp gradient formation, and support restricted extracellular diffusion as the main mechanism for Dpp dispersal. The properties of this mechanism, in which only a minority of Dpp is receptor-bound, may facilitate long-range distribution., Author Summary Morphogens are signaling molecules that trigger specific responses in cells in a concentration-dependent manner. The formation of morphogen gradients is essential for the patterning of tissues and organs. Decapentaplegic (Dpp) is the Drosophila homolog of the bone morphogenic proteins in vertebrates and forms a morphogen gradient along the anterior-posterior axis of the Drosophila wing imaginal disc, a single-cell layered epithelium. Dpp determines the growth and final size of the wing disc and serves as an ideal model system to study gradient formation. Despite extensive studies the mechanism by which morphogen gradients are established remains controversial. In the case of Dpp two mechanisms have been postulated, namely extracellular diffusion and receptor-mediated transcytosis. In the first model Dpp is suggested to move by diffusion through the extracellular matrix of a tissue, whereas in the latter model Dpp is transported through the cells by receptor-mediated uptake and re-secretion. In this work we combined novel genetic tools with mathematical modeling to discriminate between the two models. Our results suggest that the Dpp gradient forms following the extracellular diffusion mechanism. Moreover, our data suggest that the majority of the extracellular Dpp is free and not bound to its receptor, a property likely to play a role for the long-range gradient formation.

Many types of fiber‐optic sensors have been proposed and developed. In general, efforts have been made to detect a specific parameter of interest separately from other parameters. In the homodyne interferometric type of sensors, the fading of output caused by temperature variation is a serious problem, and several types of compensation schemes have been developed. In contrast, this paper describes the simultaneous sensing of acoustic wave and temperature change using a polarization‐maintaining fiber. The acoustic wave is detected by an optical heterodyne scheme and temperature change is detected by an optical homodyne scheme. The zeroth‐order diffracted light from a Bragg cell is directed into both the X and Y polarization axes of a polarization‐maintaining fiber. The homodyne interference of the X and Y components is utilized to detect the temperature variation. Simultaneous sensing of acoustic wave and temperature change using a polarization-maintaining fiber is experimentally investigated. Temperature change is detected by homodyne interference of the X and Y eigenmodes of the fiber, while acoustic wave is detected by heterodyne interference of the X component of the output light from the fiber and the reference beam. Simultaneous sensing was successfully demonstrated.

A heterodyne interference type detection scheme using only one polarization-maintaining fiber is experimentally investigated for underwater sound detection. A linear relation between sound pressure and output voltage has been obtained up to a sound pressure level of 188 dB re 1 µPa. It has been confirmed that an optical fiber hydrophone of this type has a good transient response and is useful for cavitation noise detection.
