Abstract

ITER envisages the use of two heating neutral beam injectors plus an optional one as part of the auxiliary heating and current drive system. The 16.5 MW expected neutral beam power per injector is several notches higher than worldwide existing facilities. A Neutral Beam Test Facility (NBTF) was established at Consorzio RFX, exploiting the synergy of two test beds, SPIDER and MITICA. SPIDER is dedicated to developing and characterizing large efficient negative ion sources at relevant parameters in ITER-like conditions: source and accelerator located in the same vacuum where the beam propagates, immunity to electromagnetic interferences of multiple radio-frequency (RF) antennas, avoidance of RF-induced discharges on the outside of the source. Three years of experiments on SPIDER have addressed to the necessary design modifications to enable full performances. The source is presently under a long shut-down phase to incorporate learnings from the experimental campaign. Parallelly, developments on MITICA, the full-scale prototype of the ITER NBI featuring a 1 MV accelerator and ion neutralization, are underway including manufacturing of in-vessel components, while power supplies and auxiliary plants are already under final testing and commissioning. Integration, commissioning and tests of the 1MV power supplies are essential for this first-of-kind system, unparalleled both in research and industry field. The integrated test to confirm 1MV output by combining invertor systems, DC generators and transmission lines extracted errors/accidents in some components. To realize a concrete system for ITER, solutions for the repair and the improvement of the system were developed. Hence, NBTF is emerging as a necessary facility, due to the large gap with existing injectors, effectively dedicated to identify issues and find solutions to enable successful ITER NBI operations in a time bound fashion.

The 16.5 MW expected neutral beam power per injector is several notches higher than worldwide existing facilities. In order to enable such development, a Neutral Beam Test Facility (NBTF) was established at Consorzio RFX, exploiting the synergy of two test beds, called SPIDER and MITICA. SPIDER is dedicated developing and characterizing large efficient negative ion sources at relevant parameters in ITER-like conditions: source and accelerator located in the same vacuum where the beam propagates, immunity to electromagnetic interferences of multiple radio-frequency (RF) antennas, avoidance of RF-induced discharges on the outside of the source. The source is presently under a long shut-down phase to incorporate learnings from the experimental campaign, in particular events/issues occurred during operation, which led to the identification of improvement opportunities/necessities (e.g. RF discharges, local burns, water leaks, other damages, configuration/design upgrades to maximize chances/margin to quest target parameters). Parallelly, developments on MITICA, the full-scale prototype of the ITER Neutral Beam Injector (NBI) featuring a 1 MV accelerator and ion neutralization, are underway including manufacturing of the beam source, accelerator and the beam line components, while power supplies and auxiliary plants, already installed, are under final testing and commissioning. Abstract

To reach fusion conditions and control the plasma configuration in ITER, the next step in tokamak fusion research, two neutral beam injectors (NBIs) will supply 16.5 MW each, by neutralizing accelerated negative hydrogen or deuterium ions. The requirements of ITER NBIs (40A/1 MeV D- ions for ≤1 h, 46A/870 keV H- ions for ≤1000 s) have never been simultaneously attained. So in the Neutral Beam Test Facility (NBTF, Consorzio RFX, Italy) the operation of the full-scale ITER NBI prototype (MITICA) will be tested and optimised up to full performances, focussing on accelerator (including voltage holding), beam optics, neutralisation, residual ion removal. The NBTF includes also the full-scale prototype of the ITER NBI source with 100 keV particle energy (SPIDER), for early investigation of: negative ion production and extraction, source uniformity, negative ion current density and beam optics. This paper will describe the main results of the first two years of SPIDER operation, devoted to characterizing plasma and beam parameters, including investigation of RF-plasma coupling efficiency and magnetic filter field effectiveness in reducing co-extracted electrons. SPIDER is progressing towards the first caesium injection, which aims at increasing the negative ion density. A major shutdown, planned for 2021, to solve the issues identified during the operation and to carry out programmed modifications, will be outlined. The installation of each MITICA power supply and auxiliary system is completed; in-vessel mechanical components are under procurement by Fusion for Energy (F4E). Integration, commissioning and test of the power supplies, procured by F4E and QST, as the Japanese Domestic Agency (JADA), will be presented. Abstract

Nitrogen pollution is an important environmental issue gaining traction in policy circles. However, there is little understanding of current nitrogen policies around the world: whether they account for nitrogen’s unique ability to exacerbate multiple environmental impacts or balance nitrogen’s dual role as an essential agricultural input and major pollutant. Here we assemble and analyse the first database of nitrogen policies generated by national and regional legislatures and government agencies, a collection of 2,726 policies across 186 countries derived from the ECOLEX database. The database covers all major environmental sinks (such as air, water and climate), economic sectors (including agriculture, wastewater and industry) and policy instruments (from market mechanisms to regulatory standards). We find that sink-centred policies are focused predominantly on water, mirroring the distribution of nitrogen’s global environmental and human health costs. However, policy integration across sinks is severely lacking, which heightens the risk of substituting one form of nitrogen pollution for another. Moreover, two-thirds of agricultural policies (ranging from broad sectoral programmes to nitrogen-specific measures) incentivize nitrogen use or manage its commerce, demonstrating the primacy of food production over environmental concerns.

Abstract

An altered metabolism is involved in the development of clear cell - renal cell carcinoma (ccRCC), and in this tumor many altered genes play a fundamental role in controlling cell metabolic activities. We delineated a large-scale metabolomic profile of human ccRCC, and integrated it with transcriptomic data to connect the variations in cancer metabolism with gene expression changes. Moreover, to better analyze the specific contribution of metabolic gene alterations potentially associated with tumorigenesis and tumor progression, we evaluated the transcription profile of primary renal tumor cells. Untargeted metabolomic analysis revealed a signature of an increased glucose uptake and utilization in ccRCC. In addition, metabolites related to pentose phosphate pathway were also altered in the tumor samples in association with changes in Krebs cycle intermediates and related metabolites. We identified NADH dehydrogenase (ubiquinone) 1 alpha subcomplex 4-like 2 (NDUFA4L2) as the most highly expressed gene in renal cancer cells and evaluated its role in sustaining angiogenesis, chemoresistance, and mitochondrial dysfunction. Finally, we showed that silencing of NDUFA4L2 affects cell viability, increases mitochondrial mass, and induces ROS generation in hypoxia.

Abstract

Clear cell Renal Cell Carcinoma (ccRCC) causes over 13,000 deaths each year, and about 20,000 new cases/year in Europe. In most cases, the causes are unknown and, most importantly, there are no reliable biomarkers for the diagnosis and prognosis of this disease. The search for sensitive biomarkers for early diagnosis and prognosis of clear cell Renal Cell Carcinoma (ccRCC) is currently a fast growing field. We carried out proteomics analysis of 93 urinary samples of healthy subjects (HS) and patients affected by ccRCC, prostate cancer (PCa) and chronic kidney disease (CKD), that was able to successfully distinguish each group. The most significant candidate biomarker was identified by mass spectrometry as Raf Kinase Inhibitor Protein (RKIP), a key regulator of cell signaling, already described in several cancer types as a metastasis suppressor. By combining ELISA, immunoblotting and tissue microarray, we demonstrated that, in ccRCC, urinary excretion of RKIP and its phosphorylated form (p-RKIP) reflected the tissue expression of these putative biomarkers. Baseline urinary RKIP, evaluated in an independent cohort of 56 ccRCC patients and 28 HS, successfully distinguished both groups and, most importantly, a cut-offvalue of 10 ng/mg/g Pr/uCr enabled a highly accurate prediction of Cancer-specific survival and Progression-free survival. Furthermore, p-RKIP was totally undetectable in both tissue and urine samples of ccRCC, showing a great potential for diagnostics purposes. Our data indicate that urinary RKIP encompasses both the unphosphorylated and the phosphorylated form and that their combined evaluation can help in the diagnosis and prognosis of ccRCC.

Abstract

Some basic dynamical aspects relevant to the carbon oxidation catalyzed by Pt(111) via the direct reaction mechanism are explored using a semiempirical PES derived from the spectroscopic constant of the chemisorptions species. The results concluded that the surface dynamics is quite complex, due to the activation of several adsorbed/desorption and sticking process that can complete with the CO formation in the gas-phase.

Abstract

Renal cell carcinoma (RCC) accounts for approximately 3% of adult malignancies, and clear cell RCC (ccRCC), that has a high metastatic index and high relapse rate, is the most common histological subtype. The identification of new biomarkers in ccRCC is fundamental for stratifying patients into prognostic risk groups and to guide therapy. The renoprotective antiaging gene, Klotho, has recently been found to work as a tumor suppressor in different human cancers. Here, we evaluated Klotho expression in tissue and serum of ccRCC patients and correlated it with disease progression. Tissue Klotho expression was studied by quantitative RT-PCR and immunohistochemistry. In addition, soluble serum Klotho levels were preoperatively measured in 160 patients who underwent nephrectomy for RCC with ELISA. Estimates of cancer-specific (CSS) and progression-free survival (PFS) were calculated according to the Kaplan-Meier method. Multivariate analysis was performed to identify the most significant variables for predicting CSS and PFS. Klotho protein levels were significantly decreased in RCC tissues compared with normal tissues (P<0.01) and the more advanced the disease, the more evident the down-regulation. This trend was also observed in serum samples. Statistically significant differences resulted between serum Klotho levels and tumor size (P = 0.003), Fuhrman grade (P = 0.007), and clinical stage (P = 0.0004). CSS and PFS were significantly shorter in patients with lower levels of Klotho (P<0.0001 and P = 0.0004, respectively). At multivariate analysis low serum levels of Klotho were independent adverse prognostic factors for CSS (HR = 2.11; P = 0.03) and PFS (HR = 2.18; P = 0.03).These results indicate that a decreased Klotho expression is correlated with RCC progression, and suggest a key role of declining Klotho in the onset of cancer metastasis.

Abstract

The interaction between a molecule and a solid surface can lead to a great variety of elementary processes such as elastic, inelastic and reactive. Of particular importance is the dissociative chemisorption of a diatomic molecule where a molecule chemisorbed at the surface in a specific roto-vibrational state (v,j) dissociates with the two atoms adsorbed or scattered into the gas-phase. Of great importance is also the atom recombination on surfaces. Here two atoms recombine thus forming a diatomic molecule that can be either chemisorbed or reflected in the gas-phase in a given internal energy state. Reactions (1)-(2) are very often the rate determining step of complex heterogeneous systems of interest in different branches of industrial and technological applications, as for example in the ammonia synthesis, hydrocarbon production, chemical vapour deposition, etching and thin solid film deposition via plasma, nuclear rector technologies. Both processes are of central importance in aerothermodynamics and the chemistry of interstellar media. Thus, the recombination of atomic O and N on silica and UHTC materials plays a central role for the thermal protection system of the space shuttles entering into the terrestrial atmosphere, whereas the recombination of hydrogen atoms on ice grains covered by carbon is, very likely, the main source of molecular hydrogen observed in the interstellar media. The interaction of chemical species with surfaces can lead to other non-reactive chemico-physical processes such as the inelastic processes and adsorption. The adsorption processes occur when the particle is trapped in a chemisorption site and its available energy is not enough to escape from the chemisorption potential well.
