Abstract: Major migration events in Holocene Eurasia have been characterized genetically at broad regional scales1-4. However, insights into the population dynamics in the contact zones are hampered by a lack of ancient genomic data sampled at high spatiotemporal resolution5-7. Here, to address this, we analysed shotgun-sequenced genomes from 100 skeletons spanning 7,300 years of the Mesolithic period, Neolithic period and Early Bronze Age in Denmark and integrated these with proxies for diet (13C and 15N content), mobility (87Sr/86Sr ratio) and vegetation cover (pollen). We observe that Danish Mesolithic individuals of the Maglemose, Kongemose and Ertebølle cultures form a distinct genetic cluster related to other Western European hunter-gatherers. Despite shifts in material culture they displayed genetic homogeneity from around 10,500 to 5,900 calibrated years before present, when Neolithic farmers with Anatolian-derived ancestry arrived. Although the Neolithic transition was delayed by more than a millennium relative to Central Europe, it was very abrupt and resulted in a population turnover with limited genetic contribution from local hunter-gatherers. The succeeding Neolithic population, associated with the Funnel Beaker culture, persisted for only about 1,000 years before immigrants with eastern Steppe-derived ancestry arrived. This second and equally rapid population replacement gave rise to the Single Grave culture with an ancestry profile more similar to present-day Danes. In our multiproxy dataset, these major demographic events are manifested as parallel shifts in genotype, phenotype, diet and land use.

Abstract: Western Eurasia witnessed several large-scale human migrations during the Holocene1-5. Here, to investigate the cross-continental effects of these migrations, we shotgun-sequenced 317 genomes-mainly from the Mesolithic and Neolithic periods-from across northern and western Eurasia. These were imputed alongside published data to obtain diploid genotypes from more than 1,600 ancient humans. Our analyses revealed a 'great divide' genomic boundary extending from the Black Sea to the Baltic. Mesolithic hunter-gatherers were highly genetically differentiated east and west of this zone, and the effect of the neolithization was equally disparate. Large-scale ancestry shifts occurred in the west as farming was introduced, including near-total replacement of hunter-gatherers in many areas, whereas no substantial ancestry shifts happened east of the zone during the same period. Similarly, relatedness decreased in the west from the Neolithic transition onwards, whereas, east of the Urals, relatedness remained high until around 4,000 BP, consistent with the persistence of localized groups of hunter-gatherers. The boundary dissolved when Yamnaya-related ancestry spread across western Eurasia around 5,000 BP, resulting in a second major turnover that reached most parts of Europe within a 1,000-year span. The genetic origin and fate of the Yamnaya have remained elusive, but we show that hunter-gatherers from the Middle Don region contributed ancestry to them. Yamnaya groups later admixed with individuals associated with the Globular Amphora culture before expanding into Europe. Similar turnovers occurred in western Siberia, where we report new genomic data from a 'Neolithic steppe' cline spanning the Siberian forest steppe to Lake Baikal. These prehistoric migrations had profound and lasting effects on the genetic diversity of Eurasian populations.

3. Pancreatic masses clinically diagnosed as tuberculosis: Case reports

Abstract: Pancreatic masses are commonly encountered in clinical practice, with concern for the possibility of cancer. Tissue sampling or outright surgical resection may be offered in this setting. However, surgery has been unnecessarily performed in patients with pancreatic masses that proved to be benign. Less invasive options for pancreatic masses that may be benign like tuberculosis should thus be explored. Three adult Filipino patients less than 60 years old presented with symptomatic pancreatic masses suspected of cancer on abdominal imaging studies. Two were smokers without a history of prior tuberculosis. Without any tissue sampling, anti-tuberculosis treatment was eventually given to all three patients due to concomitant diagnoses of extrapancreatic tuberculosis. Endoscopic ultrasound documentation of post-treatment resolution of pancreatic masses was noted in all cases. In endemic regions, although clinical diagnosis of tuberculosis may be possible for pancreatic masses, empiric treatment should still be a last-line option in cases where tissue sampling cannot be done.

8. Maximally Supersymmetric AdS Solutions and their Moduli Spaces

Abstract: We study maximally supersymmetric AdS$_D$ solutions of gauged supergravities in dimensions $D \geq 4$. We show that such solutions can only exist if the gauge group after spontaneous symmetry breaking is a product of two reductive groups $H_R \times H_\mathrm{mat}$, where $H_R$ is uniquely determined by the dimension D and the number of supersymmetries N while $H_\mathrm{mat}$ is unconstrained. This resembles the structure of the global symmetry groups of the holographically dual SCFTs, where $H_R$ is interpreted as the R-symmetry and $H_\mathrm{mat}$ as the flavor symmetry. Moreover, we discuss possible supersymmetry preserving continuous deformations, which correspond to the conformal manifolds of the dual SCFTs. Under the assumption that the scalar manifold of the supergravity is a symmetric space we derive general group theoretical conditions on these moduli. Using these results we determine the AdS solutions of all gauged supergravities with more than 16 real supercharges. We find that almost all of them do not have supersymmetry preserving deformations with the only exception being the maximal supergravity in five dimensions with a moduli space given by $SU(1,1)/U(1)$. 9. A Case of Skin Limited IgA Vasculitis in an 84-Year-Old Female.

Abstract: IgA vasculitis is a small vessel vasculitis and while it represents the most common form of vasculitis in pediatric populations, it is rare in adults. This manuscript describes a case of skin limited IgA vasculitis in an 84-yearold Caucasian female. The patient presented for bilateral lower extremity rash that occurred roughly one month following a viral upper respiratory illness. Her rash initially began as small petechial macules around her ankles and progressed to large confluent ecchymoses with scattered petechiae and necrosis over the course of a month. The patient underwent extensive clinical and laboratory analysis for suspected vasculitis which revealed elevated serum IgA, moderate hematuria, pyuria, and an asymptomatic urinary tract infection on urinalysis, but had otherwise normal results on autoimmune testing. The patient underwent tissues biopsies of her bilateral lower extremity rashes which revealed pathology suggestive of leukocytoclastic vasculitis of dermal vessels. Nephrology evaluation revealed no acute renal pathologies and thus biopsy was deferred. Given the result of skin biopsies, elevated serum IgA, and previous viral prodrome, the diagnosis of IgA vasculitis was confirmed. The patient was treated with 40 mg prednisone daily which was tapered over 6 weeks. Following completion of her steroid taper the rashes appeared to be healing well with only residual erythema and no signs of recurrent or worsening purpura. She continues to follow with her medical specialist for potential systemic involvement, which currently remains negative. [ABSTRACT FROM AUTHOR]

Abstract: Western Eurasia witnessed several large-scale human migrations during the Holocene1–5. Here, to investigate the cross-continental effects of these migrations, we shotgun-sequenced 317 genomes—mainly from the Mesolithic and Neolithic periods—from across northern and western Eurasia. These were imputed alongside published data to obtain diploid genotypes from more than 1,600 ancient humans. Our analyses revealed a ‘great divide’ genomic boundary extending from the Black Sea to the Baltic. Mesolithic hunter-gatherers were highly genetically differentiated east and west of this zone, and the effect of the neolithization was equally disparate. Large-scale ancestry shifts occurred in the west as farming was introduced, including near-total replacement of hunter-gatherers in many areas, whereas no substantial ancestry shifts happened east of the zone during the same period. Similarly, relatedness decreased in the west from the Neolithic transition onwards, whereas, east of the Urals, relatedness remained high until around 4,000 bp, consistent with the persistence of localized groups of hunter-gatherers. The boundary dissolved when Yamnaya-related ancestry spread across western Eurasia around 5,000 bp, resulting in a second major turnover that reached most parts of Europe within a 1,000-year span. The genetic origin and fate of the Yamnaya have remained elusive, but we show that hunter-gatherers from the Middle Don region contributed ancestry to them. Yamnaya groups later admixed with individuals associated with the Globular Amphora culture before expanding into Europe. Similar turnovers occurred in western Siberia, where we report new genomic data from a ‘Neolithic steppe’ cline spanning the Siberian forest steppe to Lake Baikal. These prehistoric migrations had profound and lasting effects on the genetic diversity of Eurasian populations.

Abstract: Major migration events in Holocene Eurasia have been characterized genetically at broad regional scales1–4. However, insights into the population dynamics in the contact zones are hampered by a lack of ancient genomic data sampled at high spatiotemporal resolution5–7. Here, to address this, we analysed shotgun-sequenced genomes from 100 skeletons spanning 7,300 years of the Mesolithic period, Neolithic period and Early Bronze Age in Denmark and integrated these with proxies for diet (13C and 15N content), mobility (87Sr/86Sr ratio) and vegetation cover (pollen). We observe that Danish Mesolithic individuals of the Maglemose, Kongemose and Ertebølle cultures form a distinct genetic cluster related to other Western European hunter-gatherers. Despite shifts in material culture they displayed genetic homogeneity from around 10,500 to 5,900 calibrated years before present, when Neolithic farmers with Anatolian-derived ancestry arrived. Although the Neolithic transition was delayed by more than a millennium relative to Central Europe, it was very abrupt and resulted in a population turnover with limited genetic contribution from local hunter-gatherers. The succeeding Neolithic population, associated with the Funnel Beaker culture, persisted for only about 1,000 years before immigrants with eastern Steppe-derived ancestry arrived. This second and equally rapid population replacement gave rise to the Single Grave culture with an ancestry profile more similar to present-day Danes. In our multiproxy dataset, these major demographic events are manifested as parallel shifts in genotype, phenotype, diet and land use.

Abstract: Western Eurasia witnessed several large-scale human migrations during the Holocene1,2,3,4,5. Here, to investigate the cross-continental effects of these migrations, we shotgun-sequenced 317 genomes—mainly from the Mesolithic and Neolithic periods—from across northern and western Eurasia. These were imputed alongside published data to obtain diploid genotypes from more than 1,600 ancient humans. Our analyses revealed a ‘great divide’ genomic boundary extending from the Black Sea to the Baltic. Mesolithic hunter-gatherers were highly genetically differentiated east and west of this zone, and the effect of the neolithization was equally disparate. Large-scale ancestry shifts occurred in the west as farming was introduced, including near-total replacement of hunter-gatherers in many areas, whereas no substantial ancestry shifts happened east of the zone during the same period. Similarly, relatedness decreased in the west from the Neolithic transition onwards, whereas, east of the Urals, relatedness remained high until around 4,000 BP, consistent with the persistence of localized groups of hunter-gatherers. The boundary dissolved when Yamnaya-related ancestry spread across western Eurasia around 5,000 BP, resulting in a second major turnover that reached most parts of Europe within a 1,000-year span. The genetic origin and fate of the Yamnaya have remained elusive, but we show that hunter-gatherers from the Middle Don region contributed ancestry to them. Yamnaya groups later admixed with individuals associated with the Globular Amphora culture before expanding into Europe. Similar turnovers occurred in western Siberia, where we report new genomic data from a ‘Neolithic steppe’ cline spanning the Siberian forest steppe to Lake Baikal. These prehistoric migrations had profound and lasting effects on the genetic diversity of Eurasian populations.

19. Improved Floral Characteristics of Salvia coccinea through Intraspecific Hybridization

Abstract: Salvia is the largest genus in the Lamiaceae with more than 1000 species. The species S. coccinea used in this study has naturalized in the southeastern United States and is an important plant for pollinators. This project aimed to improve phenotypic characteristics of S. coccinea for use in the landscape by selecting for increased petal size and unique petal color. Two elite accessions were selected for hybridization using the pedigree method. One selection displayed compact habit with bicolored coral and white flowers, while the other was slightly larger with solid red flowers. Selections were made based on improved flower color and larger petal size. The breeding program achieved a 25% increase in petal width and a more vivid petal color for the coral bicolored selections. Additionally, a 60% increase in petal width was achieved for red flowers. These novel selections are attractive plants for the landscape, displaying improved ornamental value and supporting local pollinator populations.

Abstract: Salvia is a genetically diverse genus in the Lamiaceae family, with hundreds of species distributed globally. With base chromosome numbers ranging from 6 to 19 and ploidy levels ranging from diploid to octoploid, the genus has been proposed to be subdivided based on molecular data rather than morphology. However, little is known about total DNA content across the genus. The DNA content of 141 Salvia genotypes were analyzed using flow cytometry. Samples of Salvia were stained with propidium iodide and compared with the internal standards Pisum sativum ‘Ctirad’ and Solanum lycopersicum ‘Stupické’ to generate estimations of DNA content. Holoploid 2C genome sizes of the analyzed Salvia ranged from 0.63 pg to 6.12 pg. DNA content showed a wide distribution across chromosome number, ploidy, and clade. The wide distribution of DNA content across the genus further indicates the diversity of Salvia and may be useful for future breeding efforts.

Abstract: During the Last Glacial Maximum, continental ice sheets isolated Beringia (northeast Siberia and northwest North America) from unglaciated North America. By around 15 to 14 thousand calibrated radiocarbon years before present (cal. kyr bp), glacial retreat opened an approximately 1,500-km-long corridor between the ice sheets. It remains unclear when plants and animals colonized this corridor and it became biologically viable for human migration. We obtained radiocarbon dates, pollen, macrofossils and metagenomic DNA from lake sediment cores in a bottleneck portion of the corridor. We find evidence of steppe vegetation, bison and mammoth by approximately 12.6 cal. kyr bp, followed by open forest, with evidence of moose and elk at about 11.5 cal. kyr bp, and boreal forest approximately 10 cal. kyr bp. Our findings reveal that the first Americans, whether Clovis or earlier groups in unglaciated North America before 12.6 cal. kyr bp, are unlikely to have travelled by this route into the Americas. However, later groups may have used this north-south passageway.

Abstract: The genus Pavonia is one of the largest genera in the Malvaceae species; it is mainly distributed in South America. Three species of Pavonia were identified based on different flower colors and potential for landscape use in the southeastern United States. These species produce a large amount of seed at the end of the blooming season, which is not ideal for ornamental use. To reduce the seed set, gamma irradiation was used for mutation induction and propensity to induce compactness and sterility. A preliminary study indicated that the seed of Pavonia hastata would germinate at irradiation rates up to 2000 Gy. Seeds of three species were treated with six different dose rates ranging from 0 Gy to 1000 Gy to determine the ideal rate for Pavonia breeding and how gamma irradiation affected seed germination. M1 (the first mutant generation) P. lasiopetala and P. missionum were sown in 2018 and planted in the field at the University of Georgia Durham Horticulture Farm on 1 May 2019, as were M2 (the second mutant generation) seeds of P. hastata. Seed germination in 2019 showed no significance due to treatment but significance due to species and species by treatment interaction. Field evaluation performed in 2019 indicated that height was not influenced by irradiation for any of the three species but that the width index was. Flower diameter and leaf area of P. missionum became smaller as the irradiation rate increased, but the other two species showed no trends. Chlorophyll mutations were observed on P. hastata at the 1500 Gy level, which has attractive traits for ornamental use.

Abstract: In this paper, we revisit one of the prototypical PT-symmetric oligomers, namely the trimer. We find all the relevant branches of "regular" solutions and analyze the bifurcations and instabilities thereof. Our work generalizes the formulation that was proposed recently in the case of dimers for the so-called "ghost states" of trimers, which we also identify and connect to symmetry-breaking bifurcations from the regular states. We also examine the dynamics of unstable trimers, as well as those of the ghost states in the parametric regime where the latter are found to exist. Finally, we present the current state of the art for optical experiments in PT-symmetric trimers, as well as experimental results in a gain-loss-gain three channel waveguide structure.

Abstract: Introduction Regardless of having effective vaccines against COVID-19, containment measures such as enhanced physical distancing and good practice of personal hygiene remain the mainstay of controlling the COVID-19 pandemic. Countries across Asia have imposed these containment measures to varying extents. However, residents in different countries would have a differing degree of compliance to these containment measures potentially due to differences in the level of awareness and motivation in the early phase of pandemic.Objectives In our study, we aimed to describe and correlate the level of knowledge and attitude with the level of compliance with personal hygiene and physical distancing practices among Asian countries in the early phase of pandemic.Methods A multinational cross-sectional study was carried out using electronic surveys between May and June 2020 across 14 geographical areas. Subjects aged 21 years and above were invited to participate through social media, word of mouth and electronic mail.Results Among the 2574 responses obtained, 762 (29.6%) participants were from East Asia and 1812 (70.4%) were from Southeast Asia (SEA). A greater proportion of participants from SEA will practise physical distancing as long as it takes (72.8% vs 60.6%). Having safe distancing practices such as standing more than 1 or 2 m apart (AdjOR 5.09 95% CI (1.08 to 24.01)) or more than 3 or 4 m apart (AdjOR 7.05 95% CI (1.32 to 37.67)), wearing a mask when they had influenza-like symptoms before the COVID-19 pandemic, preferring online news channels such as online news websites/applications (AdjOR 1.73 95% CI (1.21 to 2.49)) and social media (AdjOR 1.68 95% CI (1.13 to 2.50) as sources of obtaining information about COVID-19 and high psychological well-being (AdjOR 1.39 95% CI (1.04 to 1.87)) were independent factors associated with high compliance.Conclusions We found factors associated with high compliance behaviour against COVID-19 in the early phase of pandemic and it will be useful to consider them in risk assessment, communication and pandemic preparedness.

Abstract: Salvia uliginosa is a desirable ornamental shrub for the landscape with blue flowers and the ability to attract pollinators, but limited variation is commercially available in this species. Mutation breeding is a valuable tool to induce variation in ornamental species. However, many deleterious effects are associated with mutation breeding, including reduced rooting ability of vegetative cuttings. Cuttings of S. uliginosa were randomly assigned into groups of 10 and exposed to 0, 10, 20, 30, 40, or 50 Gy of gamma rays from a cobalt-60 source to determine an appropriate treatment rate. A follow-up experiment treated 25 S. uliginosa cuttings at 35 Gy to induce favorable mutations. Root quality, survival, and plant height were reduced at higher levels of gamma radiation in the M1V1. However, rooting ability was not impacted in M1V2 selections. Additionally, one mutant was isolated from the 35 Gy treatment with variegated leaves for a mutation rate of 4%. Our research determined a treatment rate that induced a dominant mutation in S. uliginosa while minimizing the deleterious influence of gamma radiation. [ABSTRACT FROM AUTHOR]

Abstract: Interest in plant species that support pollinator health has been increasing in recent years. As a result, research into these historically overlooked species is increasing. One such taxon is milkweed (Asclepias spp.), a genus primarily native to North America that serves as an oviposition and food source for various pollinators, especially the monarch butterfly (Danaus plexippus L.). Although exhaustive research has been conducted on Asclepias flower morphology, seed production, and pollinator impact, little cytological work has been published. Knowing the genome size of species can predict their ability to hybridize and the potential of genetic variability within a genus. Our study used 15 different Asclepias species and four interspecific Asclepias hybrids, and the total genomic content was calculated using propidium iodide. We found the 2C genome size ranged from 0.65 to 1.24 picograms. To our knowledge, our research presents data on eight species with previously unknown genomic content and is the first to report 2C values for interspecific Asclepias hybrids.

39. Mutation Breeding of Salvia coccinea with Ethyl Methanesulfonate

Abstract: Salvia coccinea is a valuable flowering annual that attracts hummingbirds and bees to the garden, but few cultivars are commercially available. There is a limited range of petal colors and no leaf variegation. This research aimed to improve the ornamental value of S. coccinea by inducing mutations with ethyl methanesulfonate (EMS). The standard, red-flowered species was selected for treatment by exposing seeds to 0%, 0.4%, 0.8%, or 1.2% EMS for 8, 12, or 24 hours. The optimal treatment rate was determined to be 1.2% EMS for 8 hours, which generated desirable mutations near the median lethal dose (LD50). The M1 population had a 53% germination rate and was completely morphologically uniform. By the M2, mutations included differences in leaf shape and flower size in addition to albina, chlorina, virescens, and chimeral chlorophyll changes. A 1% mutation rate was achieved in this breeding program with seven unstable mutations and six stable mutations. The normalized difference vegetation index (NDVI) values were measured to determine differences in chlorophyll content between lethal albina mutations, chartreuse chlorina and virescens mutations, and typical leaf color. Future work will investigate the stability and heritability of chlorophyll variegation by hybridizing these selections with coral-flowered accessions of S. coccinea.

The risk and protective factors associated with high compliance to these preventive measures are critical to enhancing pandemic preparedness.ObjectiveThis survey aims to assess differences in mental health, knowledge, attitudes, and practices (KAP) of preventive measures for COVID-19 amongst healthcare professionals (HCP) and non-healthcare professionals.DesignMulti-national cross-sectional study was carried out using electronic surveys between May-June 2020.SettingMulti-national survey was distributed across 36 countries through social media, word-of-mouth, and electronic mail.ParticipantsParticipants ≥21 years working in healthcare and non-healthcare related professions.Main outcomeRisk factors determining the difference in KAP towards personal hygiene and social distancing measures during COVID-19 amongst HCP and non-HCP.ResultsHCP were significantly more knowledgeable on personal hygiene (AdjOR 1.45, 95% CI -1.14 to 1.83) and social distancing (AdjOR 1.31, 95% CI -1.06 to 1.61) compared to non-HCP. They were more likely to have a positive attitude towards personal hygiene and 1.5 times more willing to participate in the contact tracing app. There was high compliance towards personal hygiene and social distancing measures amongst HCP. HCP with high compliance were 1.8 times more likely to flourish and more likely to have a high sense of emotional (AdjOR 1.94, 95% CI (1.44 to 2.61), social (AdjOR 2.07, 95% CI -1.55 to 2.78), and psychological (AdjOR 2.13, 95% CI (1.59-2.85) well-being.Conclusion and relevanceWhile healthcare professionals were more knowledgeable, had more positive attitudes, their higher sense of total well-being was seen to be more critical to enhance compliance. Therefore, focusing on the well-being of the general population would help to enhance their compliance towards the preventive measures for COVID-19.

41. Atypical Presentation of Severe Bullous Pemphigoid: A Case Report.

Abstract: Bullous pemphigoid (BP) is the most common autoimmune subepidermal blistering disorder. Typically, patients will present with tense bullae and intense generalized pruritus with a skin biopsy demonstrating subepidermal split with eosinophils and a direct immunofluorescence highlighting autoantibodies against the basement membrane zone. Prognosis varies, and treatment involves an assessment of the severity of disease to determine whether to initiate topical or systemic immunosuppressive agents. We present an atypical presentation of BP that presented as a 3-to-4-week duration of pruritic small vesicular lesions in the upper chest, scabbed circular lesions along the upper extremity and pinnas of bilateral ear. Initially thought to be herpes zoster infection initially treated with valacyclovir for a week following a prior concern of a concomitant superficial skin infection with cephalexin and prednisone. With no clinical improvement, tissue biopsy was performed that confirmed bullous pemphigoid and treatment with steroid taper, doxycycline, and triamcinolone acetonide 0.1% cream was started. The aim of this case report is to present an atypical presentation of BP and to highlight maintaining a high index of suspicion of BP in patients presenting with disseminated significantly pruritic lesions. Abstract: Exposure to environmental stress is clinically established to influence male reproductive health, but the impact of normal cellular metabolism on sperm quality is less well-defined. Here we show that impaired mitochondrial proline catabolism, reduces energy-storing flavin adenine dinucleotide (FAD) levels, alters mitochondrial dynamics toward fusion, and leads to age-related loss of sperm quality (size and activity), which diminishes competitive fitness of the animal. Loss of the 1-pyrroline-5-carboxylate dehydrogenase enzyme alh-6 that catalyzes the second step in mitochondrial proline catabolism leads to premature male reproductive senescence. Reducing the expression of the proline catabolism enzyme alh-6 or FAD biosynthesis pathway genes in the germline is sufficient to recapitulate the sperm-related phenotypes observed in alh-6 loss-of-function mutants. These sperm-specific defects are suppressed by feeding diets that restore FAD levels. Our results define a cell autonomous role for mitochondrial proline catabolism and FAD homeostasis on sperm function and specify strategies to pharmacologically reverse these defects.

Abstract: The emergence of craft specialisation is a key area of interest for archaeologists investigating the socio-economic history and development of past societies. In southwest Asia, as elsewhere, the origins of craft specialisation have been associated with the emergence of surplus food production, households and social stratification. We present evidence for nascent skilled production of green stone beads at the Pre-Pottery Neolithic A (PPNA) site Shubayqa 6, northeast Jordan. Thousands of pieces of debitage, roughouts and finished beads exhibit signs of standardised production that was probably geared towards exchange. This hints towards incipient skilled craft production that was likely part-time and seasonal. We therefore argue that the appearance of specialist artisans in this autonomous and non-hierarchical society has no correlation with surplus food production, households, or social stratification.
