Abstract Background Lauraceae is well known for its significant phylogenetic position as well as important economic and ornamental value; however, most evergreen species in Lauraceae are restricted to tropical regions. In contrast, camphor tree (Cinnamomum camphora) is the most dominant evergreen broadleaved tree in subtropical urban landscapes. Results Here, we present a high-quality reference genome of C. camphora and conduct comparative genomics between C. camphora and C. kanehirae. Our findings demonstrated the significance of key genes in circadian rhythms and phenylpropanoid metabolism in enhancing cold response, and terpene synthases (TPSs) improved defence response with tandem duplication and gene cluster formation in C. camphora. Additionally, the first comprehensive catalogue of C. camphora based on whole-genome resequencing of 75 accessions was constructed, which confirmed the crucial roles of the above pathways and revealed candidate genes under selection in more popular C. camphora, and indicated that enhancing environmental adaptation is the primary force driving C. camphora breeding and dominance. Conclusions These results decipher the dominance of C. camphora in subtropical urban landscapes and provide abundant genomic resources for enlarging the application scopes of evergreen broadleaved trees.

Tuina is a treatment method in traditional Chinese medicine which has analgesic effects and effectively alleviates the symptoms of neuropathic pain (NP). Transient receptor potential vanilloid type 1 (TRPV1) and transient receptor potential ankyrin type 1 (TRPA1) play major roles in transmitting nociceptive sensory signals in the nociceptive primary sensory dorsal root ganglion (DRG) nerve. The nitric oxide (NO)/cyclic guanosine 3′,5′-monophosphate(cGMP) pathway exerts both nociceptive and antinociceptive effects in various chronic pain models. TRPV1 and TRPA1 mediate the influx of calcium, which stimulates the generation of NO. Subsequently, NO activates the NO/cGMP/protein kinase G (PKG) signaling pathway, thereby improving hyperalgesia. In the present study, oa rat model of NP with minor chronic constriction injury (CCI) of the right sciatic nerve of NP was established. The results of behavioral testing showed that, after a one-time tuina intervention, the mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were prolonged to varying degrees in the tuina group compared with the model group. Similarly, the expression of TRPV1, TRPA1, NO, soluble guanylate cyclase β (sGCβ), cGMP, and PKG1 was significantly decreased in the DRG of the tuina and tuina + TRPV1/TRPA1 antagonist group was significantly decreased. These findings suggest that the tuina intervention can effectively improve the symptoms of thermal and mechanical allodynia caused by peripheral nerve injuries. Tuina exerts immediate analgesic effects through the TRPV1/TRPA1-NO-cGMP-PKG signaling pathway.

BackgroundIn some observational studies, attention-deficit/hyperactivity disorder has been linked to allergic diseases, but the findings are debatable. This study aimed to determine whether attention-deficit/hyperactivity disorder (ADHD) is causally related to allergic asthma, allergic rhinitis, pollen allergy, allergic urticaria, and allergic conjunctivitis using the two-sample Mendelian Randomization (MR) approach.MethodsWe did a two-sample Mendelian randomization (MR) study, which chose single nucleotide polymorphisms (SNPs) that are highly associated with attention-deficit/hyperactivity disorder (ADHD) levels from the Psychiatric Genomics Consortium (PGC) on 20,183 cases and 35,191 controls as our instruments. Outcomes datasets included genome-wide association study (GWAS) meta-analysis (n = 1,415,804). The summary statistics of outcome data were obtained from the FinnGen datasets including allergic asthma (10,877 cases and 180,942 controls), allergic rhinitis (8,430 cases and 298,829 controls), pollen allergy (4555cases and 301,734 controls), allergic urticaria (1792 cases and 299,491 controls) and allergic conjunctivitis (15,567 cases and 293,587 controls). Inverse variance weighted, MR-Egger, weighted median, were used to estimate the causal association between ADHD and allergic diseases. Cochran’s Q test was used to quantify the heterogeneity of instrumental variables. MR-Egger intercept test, leave-one-out analysis, and the funnel plot were all used in sensitivity analyses.ResultsThe Mendelian randomization (MR) analysis indicated that ADHD in inverse variance weighted [odds ratio (OR) = 1.0612; 95% confidence interval (CI):1.0192–1.1049; p = 0.0039] lightly increased the risk of allergic asthma. In MR sensitivity analyses of the weighted median, a similar association was found. But no evidence for an effect of ADHD on allergic asthma risk was found in additional methods: MR-Egger (OR = 0.9592, 95% CI: 0.8384–1.0974, p = 0.5457), and weighted median (OR: =1.0341, 95% CI: 0.9785–1.0929, p = 0.2330). Also, no strong evidence for an effect of ADHD on other allergic diseases (allergic rhinitis, pollen allergy, allergic urticaria, and allergic conjunctivitis) incidence was found using the inverse variance weighted (IVW) method, weighted median method, and MR-Egger regression.ConclusionAlthough several studies have found a link between ADHD and allergic diseases, our findings do not support that ADHD could increase allergic diseases incidence. Randomized controlled trials or Mendelian randomization studies with larger samples are still needed to draw more precise conclusions.

Japanese iris (Iris japonica) is a popular perennial ornamental that originated in China; it has a long display period and remains green outdoors throughout the year. winter dormancy characteristics contribute greatly to the evergreenness of herbaceous perennials. Thus, it is crucial to explore the mechanism of winter dormancy in this evergreen herbaceous perennial. Here, we used the hybrid RNA-seq strategy including single-molecule real-time (SMRT) and next-generation sequencing (NGS) technologies to generate large-scale Full-length transcripts to examine the shoot apical meristems of Japanese iris. A total of 10.57 Gb clean data for SMRT and over 142 Gb clean data for NGS were generated. Using hybrid error correction, 58,654 full-length transcripts were acquired and comprehensively analysed, and their expression levels were validated by real-time qPCR. This is the first full-length RNA-seq study in the Iris genus; our results provide a valuable resource and improve understanding of RNA processing in this genus, for which little genomic information is available as yet. In addition, our data will facilitate in-depth analyses of winter dormancy mechanisms in herbaceous perennials, especially evergreen monocotyledons.

With the global temperature increase, diverse endogenous factors and environmental cues can lead to severe obstacles to bud endodormancy release for important economic plants, such as herbaceous peony (Paeonia lactiflora Pall.). Knowing the underlying mechanism in bud endodormancy release is vital for widely planting herbaceous peony at low latitudes with warm winter climates. A systematic study was carried out between the southern Chinese cultivar ‘Hang Baishao’ with low-chilling requirement (CR) trait and the northern cultivar ‘Zhuguang’ with high-CR trait. Peony buds were sampled at regular intervals under natural cold during the crucial bud endodormancy release stage. Physiology and morphology of the buds were observed, and the roles of reactive oxygen species (ROS) and relevant genes in the regulation of bud endodormancy release were also highlighted, which has been rather rare in previous bud dormancy studies of both herbaceous and tree peonies. The expression of the starch metabolism- and sucrose synthesis-related genes PlAMY PlSPS and PlSUS was lower in the high-CR ‘Zhuguang’ and corresponded to a lower content of soluble sugars. The expression of polyamine oxidase gene PlPAO2 correlated with a higher level of hydrogen peroxide (H2O2) in high-CR ‘Zhuguang’ than in low CR ‘Hang Baishao’ during bud endodormancy. Expression of PlMAPKKK5, an intermediate gene in the abscisic acid (ABA) response to ROS signaling, correlated with ROS levels and ABA content. We present the hypothesis that accumulation of ROS increases ABA content and decreases GA3 content and signal transduction leading to reduced expression of PlSVP and PlSOC1. Reduced cell division and increased cellular damage which probably blocked bud endodormancy release were also observed in high-CR ‘Zhuguang’ through histological observation and related genes expression. This study provides a comparative analysis on physiological responses and gene expression patterns of bud dormancy of geophytes in an increasingly unsuitable environment.

Cold acclimation (CA) is a strategy which plants have evolved to increase freezing tolerance. Global climate change could obstruct CA and raise the probability of winter injury, especially for evergreens. Hence, understanding the regulatory mechanism of CA is crucial to improve freezing tolerance in evergreen plants. A comparative study on a pair of closely related evergreen and deciduous iris species in response to cold through CA was conducive to uncovering and complementing the knowledge of CA. We investigated morphological, physiological and biochemical changes, as well as the expression of associated genes in the functional leaves of both iris species from natural CA to deacclimation. Briefly, fast and strong CA in the evergreen iris might cause early expressions of BAM1, NCED3, GPX6, etc., which leads to strong enzyme activity of starch degradation, abscisic acid biosynthesis and reactive oxygen species scavenging. Additionally, genes belonging to the antioxidant system were mainly induced during deacclimation. These results suggest that interspecies differences in the leaf freezing tolerance of irises are associated with the rate and degree of CA, which activates multiple signaling networks with complex interactions and induces the transcription of cold-responsive genes. Moreover, the ICE–CBF–COR signaling cascade may integrate and initiate diverse cold-responsive pathways during CA of the evergreen iris. The findings of this study provide valuable insight to further research on CA mechanisms and implicate genes which could support breeding strategies in herbaceous perennials under climate changes.

Objective: To explore the analgesic initiation mechanism of " three-manipulations and three-acupoints" of tuina on minor chronic constriction injury (minor CCI) model rats. Methods: According to the random number table method, 35 SD rats were randomly divided into five groups: normal group, sham group, model group, tuina group, and tuina + MK-801 group. The model group, tuina group, and tuina + MK-801 group were subjected to ligation of the right sciatic nerve trunk to establish a minor CCI rat model. The sham group was only exposed to the right sciatic nerve without ligation, and the normal group was not subjected to any operation. The normal group was not subjected to any intervention measures. On the seventh day after modeling, the model group and the sham group underwent 9 minutes of grasping restraint, while the tuina group underwent one intervention of three-manipulations (point method, dialing method, and kneading method) and three-acupoints (right " Yinmen" (BL37), " Chengshan" (BL57), and " Yanglingquan" (GB34)acupoints) with each manipulation and acupoint intervention for 1 minute for a total of 9 minutes. The tuina+ MK-801 group received intrathecal injection of MK-801 from the fifth to seventh days after modeling, with a dose of 6 μg (10 μL) per day, tuina intervention was performed 30 minutes after the last intrathecal injection, and the specific operation of tuina was the same as that of the tuina group. Before modeling, after modeling, and after intervention, each group of rats was subjected to cold sensitivity threshold (CST) and mechanical withdrawal threshold (MWT) testing. After intervention, immunohistochemistry was used to detect the positive expression of cyclic guanosine monophosphate (cGMP) in the spinal dorsal horn (SDH) at L4-6 segments; protein expressions of N-methyl-D-aspartate receptor 1 (NMDAR1), neurogenic nitric oxide synthase (nNOS), soluble guanylyl cyclase β (sGCβ), and protein kinase G1 (PKG1) in SDH at L4-6 segments were detected by Western blotting; mRNA expressions of NMDAR1, nNOS, sGCβ, cGMP, and PKG1 in SDH at L4-6 segments were detected by real-time PCR. Results: Compared with the normal and sham groups, after modeling, CST increased and MWT decreased in the model group, tuina group and tuina+ MK-801 group (P<0.05); after intervention, the positive protein expression of cGMP was increased, the protein expressions of NMDAR1, nNOS, sGCβ, and PKG1 were increased, and mRNA expressions of NMDAR1, nNOS, sGCβ, cGMP, and PKG1 were increased in SDH at L4-6 segments in the model group (P<0.05). Compared with the model group, after intervention, CST decreased and MWT increased in the tuina group and tuina+ MK-801 group (P<0.05); the positive protein expression of cGMP was decreased, the protein expressions of NMDAR1, nNOS, sGCβ, and PKG1 were decreased, and mRNA expressions of NMDAR1, nNOS, sGCβ, cGMP, and PKG1 were decreased in SDH at L4-6 segments in the tuina group and tuina+ MK-801 group (P<0.05). Conclusion: One-time tuina intervention can effectively improve the symptoms of thermal and mechanical hyperalgesia induced by peripheral nerve injury, which may initiate analgesia through the NMDAR1/cGMP/protein kinase G signaling pathway, thereby exerting immediate analgesic effect.
