The Amur tiger and leopard, once roaming over the Eurasian continent, are now endangered and confined to the Sikhote-Alin Mountains, Russia—a landscape that has been increasingly fragmented due to human activities. The ultimate fate of these big cats depends on whether they can resettle in their previous main historical range in NE China. Recent sightings of these animals along the China–Russia border have aroused new hope, but direct evidence is lacking. The main objectives of our study were (1) to determine the abundance and spatiotemporal patterns of tigers, leopards, and primary prey; (2) to investigate factors influencing the resettlement of the two big cats; and (3) to propose a landscape-scale conservation plan to secure the long-term sustainability of the Amur tiger and leopard. We monitored the two felids, their prey, and human activities, with 380 camera-trap stations, for a total of 175,127 trap days and over an area of 6000 km2 in NE China. We used the constraint line method to characterize cattle grazing and human influences on tigers, leopards, and their prey species. Our results show that, unexpectedly, at least 26 tigers and 42 leopards are present within China, which are confined primarily to a narrow area along the border with Russia. We have further identified that cattle grazing and human disturbances are the key hurdles to the dispersal of the tigers and leopards farther into China where suitable habitat is potentially available. Amur tigers and leopards are returning to China, indeed, but their long-term resettlement is not likely without active and timely conservation efforts on landscape and regional scales. To overcome the hurdles to the resettlement of tigers and leopards in China, we propose a “Tiger and Leopard Resettlement Program” that will engage the government, local communities, and researchers, so that the long-term sustainability of the Amur tigers and leopards can be ensured.

Mitochondrial gene transfer to the nuclear genome could affect the accuracy of results in population genetics and evolutionary studies using mitochondrial gene markers. In a population genetics study of the red turpentine beetle (Dendroctonus valens), an invasive species in China, we found numerous ambiguous sites existing in the Cytochrome Oxidase I (COI) gene sequences obtained directly from polymerase chain reaction (PCR) products amplified from total genomic DNA using universal primers. By comparing the profiles of restriction endonuclease digestions and the sequences of PCR products amplified from mitochondrial DNA and nuclear DNA of the same individuals, we confirmed it was a phenomenon of mitochondrial gene transfer to the nuclear genome. Large numbers of COI pseudogenes were detected in this species. According to different levels of condon position bias and phylogenetic analysis, these should have originated from independent integration events. The impact of nuclear mitochondrial DNA sequences on population genetics analyses was discussed.

SSR markers have become the most popular resource for studying population genetic variation in eukaryotes. However, few studies with SSR markers have been carried out with phytoparasitic nematodes. In this study a primary survey on EST-SSRs was made utilizing bioinformatics methods to derive SSRs from expressed sequence tags (ESTs) of 16 species of PPN, which belong to 7 genera and 5 families. The results showed that trinucleotide repeats were the most abundant SSRs in coding ESTs, while tetranucleotide SSRs were predominant in non-coding ESTs and genome sequences. AG/CT, AAC/GTT and (AAAN)n motifs were predominant, and CG/GC, ACT/AGT motifs were scanty in the ESTs of most genera and species. SSRs were more abundant in non-coding ESTs than in coding ESTs. The distribution frequencies of SSR motifs in coding ESTs, non-coding ESTs and genomes are different. Our results will provide useful information for screening SSRs from each genus and species for further genetic study of phytoparastic nematodes.

Two species of butterflies, Euphydryas aurinia and Melitaea phoebe , coexist as two metapopulations in a 38-patch network in Hebei Province, China. A Markovian model, whose transition matrix is the product of two matrices which represent the local extinction and recolonization process respectively, is used to describe the metapopulation dynamics. The application of this model to the metapopulation, consisting of 12 local populations in the northern subregion, shows that the expected life times of E. aurinia and M. phoebe are 160 and 121 years respectively and usually nearly half of the patches are occupied by E. aurinia , while only 1–3 patches are occupied by M. phoebe . We claim that E. aurinia can persist for a long time while M. phoebe faces relatively big extinction risk. By comparing the population dynamics with and without migration, we find M. phoebe benefits much more from migration than E. aurinia . Most patches are occupied mainly by local populations for E. aurinia , while by immigrants from the 8th patch for M. phoebe , meaning that E. aurinia has a classical metapopulation structure while M. phoebe has a source–sink metapopulation structure.

Sequences of 479 bp region of the mitochondrial COI gene were applied to detect population genetic diversity and structure of Dendroctonus valens populations. By comparing the genetic diversity between native and invasive populations, it was shown that the genetic diversity of Chinese populations was obviously lower than that of native populations with both indices of haplotype diversity and Nei's genetic diversity, suggesting genetic bottleneck occurred in the invasive process of D. valens, and was then followed by a relatively quick population buildup. According to phylogenetic analyses of haplotypes, we suggested that the origin of the Chinese population was from California, USA. Phylogenetic and network analysis of native populations of D. valens revealed strong genetic structure at two distinct spatial and temporal scales in North America. The main cause resulting in current biogeographic pattern was supposedly due to recycled glacial events. Meanwhile, a cryptic species might exist in the Mexican and Guatemalan populations.

The butterfly Euphydryas aurinia occurs as a classical metapopulation in Yanjiaping village, Heibei province, China. In the patch network under study there were 38 habitat patches. Most patches were uncultivated, consisting of meadows and fallows, while others had been cultivated, i.e., used as small croplands. In the cultivated patches, the habitat for the butterfly can be classified into two types: meadow type (MTH) and cropland type (CTH). In contrast, the uncultivated habitat patches consists only of MTH. We examined the habitat utilization of ovipositing females and larvae of E. aurinia to assess the effect of mixed land use on the butterfly’s occurrence. More egg clusters and pre-hibernating larval groups were distributed throughout CTH than MTH. This dependence on CTH may be due to the preference of egg-laying females for large-sized host plants, which were mainly concentrated in CTH. Compared to the MTH, the mortality rate in CTH was lower. Therefore, for both eggs and pre-hibernating larvae, the quality of CTH was higher than that of MTH. The conditions in MTH, on the other hand, were important for the development of post-hibernation larvae. A combination of extensive farming and animal husbandry has created a mosaic of meadows and croplands in the habitats studied here, which is typical of rural areas in some developing countries. This study implies that patchy land use typical of traditional agriculture facilitates the long-term persistence of E. aurinia.

Mark–release–recapture studies were conducted on two species of chequerspot butterfly, Euphydryas aurinia and Melitaea phoebe, in the same habitat patch network in Yanjiaping, a small basin in the Taihang Mountains, north-west of Beijing, China, in 2000. 2. Euphydryas aurinia tended to stay in the habitat patches and to move to neighbouring patches, whereas M. phoebe moved widely among patches in the entire network. 3. The parameters of the virtual migration model showed higher daily emigration propensity in M. phoebe and in E. aurinia males than in E. aurinia females, and significantly greater average daily movement distance in M. phoebe than in E. aurinia. 4. The results are consistent with the previous findings showing genetic structuring among local populations of E. aurinia but not among local populations of M. phoebe. 5. Based on the genetic and ecological results, it was concluded that E. aurinia has a classic metapopulation in the study area, whereas M. phoebe appears to have a source–sink metapopulation. 6. In 2000, when there was an overall increase in the abundance of the two species, the limited mobility of E. aurinia resulted in an increase in the average local population size, whereas the increase in the number of local populations in M. phoebe was due to its high mobility.

According to investigation on two species of melitaeine butterflies in Yanjiaping Village, Chicheng County, Hebei Province, China, between 1998–2002, together with the use of 1–10000 contour map of the local area, some conclusions are shown by the SPSS and GIS analysis of data obained from GPS: (1) The two species of melitaeine butterflies have different metapopulation structures.M. phoebe is a source-sink metapopulation, whileE. aurinia is a classical metapopulation, supporting the analytic result from our former genetic research. (2) The two species of melitaeine butterflies exhibit different trends of population dynamics.M. phoebe source-sink metapopulation is very unsteady, and is always small, thus has a tendency to go extinct gradually. ButE. aurinia classical metapopulation is stable, and has maintained a larger population size. Therefore, it stands a better chance of long-term survival. (3) The two species of melitaeine butterflies are significantly related in both patch occupancy and local population size. (4) The effect of isolation is significant on the metapopulations of these two species of melitaeine butterflies, consistent with the classical theories, whereas the effect of patch area is not significant on the metapopulations of these two species of melitaeine butterflies, which is inconsistent with the classical theories. Therefore, other factors, such as habitat quality, should be considered for their influences on metapopulations.

We analyzed genetic differentiation within metapopulations of two species of checkerspot butterfly, Euphydryas aurinia and Melitaea phoebe, in China. To generate genetic information, we used a new molecular technique, DALP – direct amplified length polymorphism. AMOVA results showed that most of the variation occurred among individuals within local populations of both E. aurinia and M. phoebe. However, while there was differentiation among local population in E. aurinia (P < 0.001), there was no subdivision in metapopulation of M. phoebe (P = 0.210). This is consistent with the behavior of M. phoebe adults being more dispersive than E. aurinia. Within the M. phoebe metapopulation, three neighboring patches were always occupied during the observation period (1998–2000). In addition, the number of individuals in these three populations accounted for the majority of M. phoebe larvae, and hence we conclude that the M. phoebe metapopulation might exist as a source-sink metapopulation. On the other hand, the E. aurinia metapopulation is an example of a classical metapopulation. Therefore, the conservation management of these two species should reflect these differences.

The chemical compounds that exist in plant leaf tissue are of great importance for homopteran insects during the proccess of host-plant acceptance. From the leaf surface to the phloem tube, various groups of chemicals can be detected by insect's receptors which lead to different feeding responses. The effects of sugars and alkaloids in leaf surface; polysaccharides, pH gradients and phenolics in mesophyll; amino acids, proteins, carbohydrates and secondary substances in phloem tube on phloem finding and host-plant acceptance are reviewed respectively in this paper, which also includes the potential effects of mesophyll structures on insect's feeding preference. Furthermore the mechanism by which the insects perceive these chemical and mechanical cues, correlated with the EPG (electrical penetration graph) patterns (a series of visible signals showing the stylet activities in plant tissue) is also discussed.

The preference of greenhouse whiteflies varies with different species of host plants and leaves of different age. They prefer young leaves of tobacco, cucumber, broadbean and cowpea significantly more than the young leaves of paprika and old leaves of broadbean and cowpea. Behavioural studies indicated that the leaf-time and total probing time were significantly longer on preferred leaves than on non-preferred leaves. On non-preferred leaves, probing may continue about 2 minutes but not longer than 5 minutes. On preferred leaves, there were both frequent probing for short intervals and probing (+feeding) which continued for 10 minutes or more. Morphological studies indicated that, in general, it needed 10 to 15 minutes for the stylet to reach the phloem and feed. The probing process takes no more than 10 to 15 minutes. Probing can be divided into test probing which takes less than 2 minutes to reach depths of 20 to 50 pm, and exploratory probing which surpasses 5 minutes. Test probing is the key process for determining host plant selection, and the chemical composition of the leaf surface wax may play an important role.

Social-economic factors are considered as the key to understand processes contributing to biological invasions. However, there has been few quantified, statistical evidence on the relationship between economic development and biological invasion on a worldwide scale. Herein, using principal factor analysis, we investigated the relationship between biological invasion and economic development together with biodiversity for 91 economies throughout the world. Our result indicates that the prevalence of invasive species in the economies can be well predicted by economic factors (R(2) = 0.733). The impact of economic factors on the occurrence of invasive species for low, lower-middle, upper-middle and high income economies are 0%, 34.3%, 46.3% and 80.8% respectively. Greenhouse gas emissions (CO(2), Nitrous oxide, Methane and Other greenhouse gases) and also biodiversity have positive relationships with the global occurrence of invasive species in the economies on the global scale. The major social-economic factors that are correlated to biological invasions are different for various economies, and therefore the strategies for biological invasion prevention and control should be different.

BACKGROUND: MicroRNAs (miRNAs) are considered to be very important in regulating the growth, development, behavior and stress response in animals and plants in post-transcriptional gene regulation. Pinewood nematode, Bursaphelenchus xylophilus, is an important invasive plant parasitic nematode in Asia. To have a comprehensive knowledge about miRNAs of the nematode is necessary for further in-depth study on roles of miRNAs in the ecological adaptation of the invasive species. METHODS AND FINDINGS: Five small RNA libraries were constructed and sequenced by Illumina/Solexa deep-sequencing technology. A total of 810 miRNA candidates (49 conserved and 761 novel) were predicted by a computational pipeline, of which 57 miRNAs (20 conserved and 37 novel) encoded by 53 miRNA precursors were identified by experimental methods. Ten novel miRNAs were considered to be species-specific miRNAs of B. xylophilus. Comparison of expression profiles of miRNAs in the five small RNA libraries showed that many miRNAs exhibited obviously different expression levels in the third-stage dispersal juvenile and at a cold-stressed status. Most of the miRNAs exhibited obviously down-regulated expression in the dispersal stage. But differences among the three geographic libraries were not prominent. A total of 979 genes were predicted to be targets of these authentic miRNAs. Among them, seven heat shock protein genes were targeted by 14 miRNAs, and six FMRFamide-like neuropeptides genes were targeted by 17 miRNAs. A real-time quantitative polymerase chain reaction was used to quantify the mRNA expression levels of target genes. CONCLUSIONS: Basing on the fact that a negative correlation existed between the expression profiles of miRNAs and the mRNA expression profiles of their target genes (hsp, flp) by comparing those of the nematodes at a cold stressed status and a normal status, we suggested that miRNAs might participate in ecological adaptation and behavior regulation of the nematode. This is the first description of miRNAs in plant parasitic nematodes. The results provide a useful resource for further in-depth study on molecular regulation and evolution of miRNAs in plant parasitic nematodes.

Increasing levels of global trade and intercontinental travel have been cited as the major causes of biological invasion. However, indirect factors such as economic development that affect the intensity of invasion have not been quantitatively explored. Herein, using principal factor analysis, we investigated the relationship between biological invasion and economic development together with climatic information for China from the 1970s to present. We demonstrate that the increase in biological invasion is coincident with the rapid economic development that has occurred in China over the past three decades. The results indicate that the geographic prevalence of invasive species varies substantially on the provincial scale, but can be surprisingly well predicted using the combination of economic development (R(2) = 0.378) and climatic factors (R(2) = 0.347). Economic factors are proven to be at least equal to if not more determinant of the occurrence of invasive species than climatic factors. International travel and trade are shown to have played a less significant role in accounting for the intensity of biological invasion in China. Our results demonstrate that more attention should be paid to economic factors to improve the understanding, prediction and management of biological invasions.

In this study, the oviposition sites of Euphydryas aurinia and Meiltaea phoebe adult females in the same habitat network were investigated, aimed to identify the factors affecting the patch quality. The results showed that the egg-laying females of both melitaeine butterfly species preferred to oviposit in the open areas with large-sized host plants and surrounded by short vegetations, especially on warm and sunny slopes. For E. aurinia, most of its egg clusters were concentrated within the area less than 3 m to the edge of cropland, and the average distance from each egg cluster to the edge of nearest cropland was 3.55 +/- 0.33 m (n = 246); but for M. phoebe, a few of its egg clusters distributed in the area less than 3 m to the edge, and the average distance was 7.43 +/- 1.53 m (n = 25). It was suggested that the criteria to evaluate the patch quality and the conservation strategies for the two butterfly species should be different, due to their special habitat requirements of ovipositing females.

The sorghum is a plant, which has been intentionally introduced in China for foods needs. It is a plant of African origin, which is much cultivated in the northern hemisphere. For millions of people in the semiarid tropic temperature of Asia and Africa, sorghum is the most important staple food. Sorghum is becoming one of the most important crops in China. There are about 7000 varieties and hybrids including native and introduced species being cultivated in the country. This paper describes three new species of oribatid mites from Shergyla Mts., southeast Tibet, at elevations between 4100 and 4700 m.a.s.l. They are Oxybrachioppia glabriseta sp. nov. belonging to the family Oppiidae Grandjean and Umbellozetes squamaceus sp. nov. and U. combinatus sp. nov. belonging to the family Tegoribatidae Grandjean. The new species were collected from alpine meadow, mosses and forest litter, lichens on stones and thin soils, respectively. The holotypes are deposited in the Institute of Zoology, Academia Sinica, Beijing, China. The paratypes are deposited in Beijing Normal University, Beijing, China, Institute of Zoology, Academia Sinica, Beijing, China, and University of Bergen, Bergen, Norway.
