Abstract At present, the characteristics of double-hit multiple myeloma (DHMM) are unknown. We retrospectively analyzed the clinical data from 433 new diagnosed MM patients and found that DHMM have a higher β2-MG level and percentage of bone marrow plasma cell. Cox regression analysis showed that the prognosis of DHMM was not limited by clinical indicators. The abnormal proliferation of bone marrow in DHMM is obvious, and the proportion of poorly differentiated plasma cell is high. By collecting specimens from our center and performing flow cytometry to analyze the immunophenotypic and functional characteristics of lymphocyte subpopulations, we found that DHMM had a higher ratio of Tregs cells, and the proportion of iTregs cells was also significantly higher than non-DHMM (P

Abstract Respiratory failure, intracranial hemorrhage and infection were more common in hyperleukocytic acute myeloid leukemia patients than in non-hyperleukocytic leukemia patients. Compared with non-apheresis treatment, the white blood cells decreased significantly and the infection rate decreased after apheresis treatment. However, the treatment time of leukapheresis in patients with hyperleukocytic leukemia is very long, while it is more damaging to cells. In this study, which conducted a retrospective analysis on patients with hyperleukocytic acute myeloid leukemia, the process of centrifugation of normal cells and patients’ cells by apheresis machine was simulated in vitro. Through selecting 5 healthy persons and 11 patients with hyperleukocytic acute myeloid leukemia, extracting their blood samples and performing in vitro centrifugation at different speeds or duration, we observed the changes of the numbers and morphology of peripheral blood cells in healthy people and patients, so as to explore the optimal centrifugation parameters during leukapheresis. The cells obtained by the optimal centrifugation parameters were cryopreserved and two groups of mice (10 mice in each group) were used to establish leukemia animal models. Through the research, it is found that when the centrifugal speed is below 6000 rpm, the damage to blood cells in healthy people and in patients with hyperleukocytic leukemia is not obvious. When the centrifugal speed is above 6000 rpm, the platelets will be damaged significantly. The cells obtained under the optimal centrifugation parameters can be successfully cryopreserved and used to establish leukemia animal models. This study is of great significance for improving the efficiency and reducing the side effects of leukapheresis, and is helpful to improve the treatment of white blood cells reduction.

Real-time and online screening techniques for single load effect signal monitoring are one of the key issues in smart structure monitoring. In this paper, an online signal sifting framework called online recursive empirical mode decomposition (EMD) is proposed. The framework is based on an improved EMD that optimizes the boundary effect by using extreme value recursion and eigensystem realization algorithm (ERA) extension, and combines the intrinsic mode functions (IMFs) correlation coefficient and adaptive filtering to select IMFs for signal reconstruction to achieve the sifting purpose. When applied to simulated signals, the method satisfies the requirements of signal sifting in an online environment with high adaptivity, low parameter sensitivity and good robustness. The method was applied to the dynamic strain data collected by the health monitoring system of Daishan Second Bridge to achieve real-time online sifting of strain signals caused by traffic loads, which provided the basis for subsequent data analysis applications and confirmed the value of the application in a real bridge health monitoring system.

This article presents a new protection device using aluminum foam to enhance the impact resistance of bridge columns. First, the protection device is designed according to the characteristics of aluminum foam material. The geometric configuration and structure of the device are described. Second, the impact performance of bridge column is analyzed, including impact force analysis, damage analysis, and the influence of axial load. Third, three-dimensional solid element models of columns with and without the protection device are developed in order to verify the effect of the protection device. By comparing dynamic responses of vehicle impact on columns with and without the protection device, it is considered that the protection device has certain protection effect: after installing the protective device, the peak value of impact force reduces by 37.5%, the maximum displacement of column top reduces by 23.7%, the maximum stress at column bottom reduces by 51.6%, the maximum stress at column bottom reduces by 51.6%, the maximum acceleration of the vehicle reduces by 40.6%, and 86.84% of the impact energy is absorbed by the protection device. Finally, the devices with different foam thicknesses and porosities are comparatively analyzed to investigate the influence of these design parameters on impact performance. The results show that the increase in the thickness of aluminum foam has positive effects on the protection capability. The protection capability improves with aluminum foam porosity increasing when the porosity is less than 60%.

Abstract Mechanical force contributes to perforin pore formation at immune synapses, thus facilitating the cytotoxic T lymphocytes (CTL)-mediated killing of tumor cells in a unidirectional fashion. How such mechanical cues affect CTL evasion of perforin-mediated autolysis remains unclear. Here we show that activated CTLs use their softness to evade perforin-mediated autolysis, which, however, is shared by T leukemic cells to evade CTL killing. Downregulation of filamin A is identified to induce softness via ZAP70-mediated YAP Y357 phosphorylation and activation. Despite the requirements of YAP in both cell types for softness induction, CTLs are more resistant to YAP inhibitors than malignant T cells, potentially due to the higher expression of the drug-resistant transporter, MDR1, in CTLs. As a result, moderate inhibition of YAP stiffens malignant T cells but spares CTLs, thus allowing CTLs to cytolyze malignant cells without autolysis. Our findings thus hint a mechanical force-based immunotherapeutic strategy against T cell leukemia.
