Abstract Almond hulls can be used as an inexpensive source of nutrients for producing fungi that can be used as nontraditional foods such as alternative protein, probiotics, and food ingredients. Nutrients were extracted from almond hulls using hot water and their utility in cultivating filamentous fungus Aspergillus awamori (A. awamori) in submerged cultivation was investigated using batch fermentation in flasks with passive aeration. The almond hulls extract supplied all the nutrients required for A. awamori growth. The uptake preference of various sugars was investigated and the growth kinetic parameters of A. awamori were determined. Utilization of sugars by A. awamori grown in almond hull extract proceeded sequentially with preference for glucose followed by sucrose, fructose, and then xylose. Compared to potato dextrose broth medium, the fungal biomass produced using almond hull extract as growth medium had higher protein (19.59%) and lower fat (1.50%) contents. The modified Gompertz model described the kinetics of A. awamori well with a lag phase of 0.18 days and a specific growth rate of 1.77 d−1.

Objective To observe the effects of 4 weeks of voluntary wheel running on depressive-like behavior in a rat chronic stress-induced depression model to explore the anti-depressive mechanism of exercise. Methods In this observational study, 36 Sprague-Dawley rats were randomly divided into control, stress model, and stress exercise groups (12 rats/group). The control group received no intervention, and the stress model and stress exercise group rats underwent chronic mild unpredictable stress and isolation. The stress exercise group rats also underwent 4 weeks of voluntary wheel running. Behavioral changes and hippocampal protein and mRNA expression levels were detected. Results Voluntary wheel running significantly increased horizontal and vertical movements, sucrose intake, and the sucrose preference percentage and reduced immobility time in the forced swimming test in depression model rats. The hippocampal tumor necrosis factor-α, interleukin (IL)-6, IL-1β, and quinolinic acid levels were significantly decreased, while the IL-4, IL-10, and kynurenic acid levels were significantly increased. Kynurenine-3-monooxygenase and 3-hydroxyanthranilate-3, 4-dioxygenase mRNA levels were downregulated, and kynurenine aminotransferase mRNA was upregulated. Conclusion Voluntary wheel running improved depressive-like behavior in depression model rats. The mechanism may be related to a kynurenine pathway metabolite level imbalance, which has neurotoxic and neuroprotective effects, caused by long-term voluntary wheel running.

Context: It is very necessary to delay ovarian aging and prevent age-related health problems. The active ingredient in Honghua Xiaoyao tablet (HHXYT) has the effects of anti-oxidation, anti-inflammation, immune regulation and so on.Objective: To explore the effect and mechanism of Honghua Xiaoyao tablet on aging model mice.Materials and methods: The aging model was established by intraperitoneal injection of D-galactose in model mice. The mice in the HHXYT-L,M,H group were given 0.3 g/kg, 0.6 g/kg and 1.2 g/kg Honghua Xiaoyao tablet suspension respectively, and the HHXYT-M + E2 group was given 0.6 g/kg HHXYT +0.13 mg/kg estradiol valerate for 30 days. In this study, ELISA, HE, Western blot, IH and TUNEL were used.Results: HHXYT + E2 can improve the gonadal index, estrous cycle of aging mice. In HHXYT-M + E2 group, the level of FSH and LH decreased, while E2 and AMH increased significantly. The number of growing follicles in HHXYT-M + E2 group increased, which was better than that of HHXYT alone. Western blot results showed that HHXYT-M + E2 group decreased the expression of Bax, cleaved-Parp, cleaved-Casp-3 and CytC molecules and increased the expression of Bcl-2 in ovarian tissue. FSHR expression decreased in model group and increased in HHXYT group. TUNEL staining showed that the number of apoptotic cells in HHXYT group was reduced, and the HHXYT-M + E2 group was the most significantly.Discussion and conclusion: HHXYT can improve the level of sex hormones and increase the number of growing follicles in aging mice. HHXYT-M + E2 group has the best effect, and its mechanism may be related to reducing ovarian granulosa cell apoptosis.

Abstract Lung metastasis is the major cause of death in patients with triple-negative breast cancer (TNBC). Tumor-associated macrophages (TAMs) represent the M2-like phenotype with potent immunosuppressive activity, and play a pro-tumor role in TNBC lung metastasis. Sohlh2 belongs to the basic helix-loop-helix transcription factor family. However, its role in macrophages polarization remains unknown, especially in TNBC progression. Here we demonstrated that Sohlh2 overexpression promoted M2 macrophage polarization. Moreover, high expression of Sohlh2 in M2-like macrophage enhanced TNBC cell growth, migration and lung metastasis in vivo and in vitro. Mechanistically, we revealed that Sohlh2 functioned through up-regulating LXRα, ABCA1, ABCG1 expression and disturbing the lipid homeostasis on the membrane of macrophages. Sohlh2 could directly bind to the promoter of LXRα and promote its transcription activity. E3 ubiquitin ligase TRIM21 promoted Sohlh2 ubiquitination and degradation, and suppressed M2 macrophage polarization and TNBC progression. Collectively, our findings suggested that Sohlh2 in macrophage could be a novel therapeutic target for TNBC metastatic treatment.

To address the lack of specialized machinery for the mechanical weeding of SCSCP in the Huang Huai Hai region, this study designs a mechanized inter-row weeding machine for SCSCP. The machine features a reciprocating weeding shovel and an adaptive contouring mechanism for cultivation and soil loosening. This paper details the machine’s principles by analyzing the geometric relationship and mechanical model between the corresponding profiling quantities, which determine the relevant parameters for adaptive contouring to ensure stable operation on undulating ground. Furthermore, by optimizing the design of the weeding shovel’s reciprocating motion mechanism, combining EDEM simulation with the weeding shovel–soil interaction, it has been determined that, at various PTO shaft speeds, the optimal weeding efficacy is achieved with a blade-type weeding shovel structure when operating at a forward speed of 3.5 km/h. Field experiments were conducted with different PTO shaft speeds and weeding depths, using weeding and seedling injury rates as performance indicators. The results showed that, based on the optimal speed, the PTO shaft speed is 760 r/min, the operating depth is 3–5 cm, and the average row weeding rate is 90.4%. The average soybean and corn seedling injury rate is 3.4% and 4.2%, meeting the technical requirements for mechanical weeding.

Abstract Disturbance in the redox balance of alveolar epithelial cells (AECs) was considered as a causal factor for pulmonary fibrosis. The regulatory mechanisms of redox hemostasis in the development of pulmonary fibrosis remain largely unknown. Using a type II AEC-specific Sohlh2 conditional knock-in (CKI) mouse model, we found that Sohlh2, a basic HLH transcription factor, accelerated age-related pulmonary fibrosis. High-fat diet (HFD) resulted in a tremendous increase in lung inflammation and fibrotic changes in the lung tissues of Sohlh2 CKI mice. Sohlh2 overexpression led to a significant rise of intracellular ROS and apoptosis in the lung, mouse primary AECIIs, and human A549 cells, which was attenuated by ROS inhibitor (NAC). Sohlh2 enhanced oxidative stress via repressing p62/Keap1/Nrf2 mediated anti-oxidative signaling pathway. p62, a direct target of Sohlh2, mediated Sohlh2 effects on ROS generation and apoptosis in A549 cells. Hence, our findings elucidate a pivotal mechanism underlying oxidative stress-induced pulmonary fibrosis, providing a framework for aging-related disorder interventions.

Abstract Enzymatic hydrolysis of sugar beets is an efficient alternative to conventional hot water extraction where overall soluble extraction is valued. The economic analysis of a sugar beet soluble sugar production plant was designed and developed using SuperPro Designer software. The commercial scale operation was sized to process 100 million metric ton (MT)/day of sugar beets. The base case process includes transportation, grinding, thermal pretreatment, enzymatic hydrolysis, solid–liquid separation, and evaporation. The final product from the model was assumed to be a sugar syrup stream at 65% total solids content. Since the breakeven selling price was sensitive to the cost of enzymes, alternative scenario A added an ultrafiltration unit operation to separate the enzymes and reuse them in subsequent batches of hydrolysis. Steam was another major contributor to the operating cost. Alternative scenario B included a natural gas boiler to generate steam from biogas produced from a co‐located dairy manure digester. The breakeven selling price of the sugar syrup from the base case, alternative scenario A, and alternative scenario B were $965, $911, and $955 per metric ton, respectively. Sensitivity analysis showed that the breakeven selling price is sensitive to the raw material cost, especially for the pectic enzymes.

Biogas digestates produced from anaerobically processed food or dairy manure have demonstrated some potential to suppress different pathogens and pests. In this study, digestates from these two substrates processed mesophilically (20 to 40°C) or thermophilically (>40°C) were compared for their nematode-suppressive potential. In bioassays, the suppressive potential of Heterodera schachtii or Meloidogyne incognita was tested at different amendment rates. Testing was expanded to field microplot experiments with M. incognita and greenhouse tests with Pratylenchus vulnus. In the bioassay with H. schachtii, fewer nematodes penetrated radish roots after amendment with thermophilic than with mesophilic digestates. Root penetration was lowest after treatment with digestates of food waste and thermophilic dairy manure at the highest rate. In a watermelon bioassay with M. incognita, nematode-induced galling and egg mass production were reduced after amendment with food waste digestates compared with manure digestates and after thermophilic compared with mesophilic digestates in the case of egg masses. In a greenhouse experiment with peach rootstock ‘Nemaguard’, the numbers of P. vulnus in roots were diminished after amendment with digestates at the highest rate. In microplot experiments with bell peppers, plants initially were more vigorous after amendment with thermophilic digestates than after mesophilic digestate additions. This effect was transient and was no longer measurable at harvest. In summary, food waste digestates, especially those that were thermophilically produced, had nematode suppressive and plant growth-improving properties. Abstract The cross coupling between receiver coils complicates the control and optimization of “one‐to‐may” multi‐load IPT systems. In this paper, the cross‐coupling effect in a two‐load IPT system is thoroughly discussed. It is noted that the input impedance, the total output power and the power distribution ratio among receivers are all affected. After developing a new leakage‐inductance‐based model considering the cross‐coupling effect, it is suggested that the operating frequency is adjusted according to the cross‐coupling coefficient to ensure a constant power distribution ratio. The total output power and the input impedance are regulated by coordinated adjustment of the inverter's duty cycle and the transmitter‐side compensation capacitance with switched control capacitor (SCC) control. Based on the proposed hybrid control strategy (frequency, duty cycle, and SCC), the power distribution among loads and the total output power are kept constant under variable cross‐coupling coefficient, and the input impedance are shaped to be inductive to achieve zero voltage switching (ZVS) realization for all switches in the inverter. The theoretical analysis is finally verified by experimental results on a 35 W two‐load IPT prototype.

Power converters are essential parts of microgrids (MGs), but they cause several problems. One of the most serious concerns with a DC microgrid (DCMG) is the constant power load (CPL), which has an incremental negative impedance (INI). INI has a negative dampening effect on the power efficiency of a power system. Sliding-mode control (SMC) has a hard attitude against external disturbances when compared to traditional control approaches like lead–lag, linear quadratic regulator, and PID. This paper suggests an adaptive sliding-mode control (ASMC) for a buck converter to supply a resistive and a CPL in a DCMG. The proposed controller provides large-signal stability during steady-state operation and maintains output voltage when the source voltage or CPL is changed. Moreover, the suggested controller significantly reduced the chattering effect, which is a major drawback of conventional SMC. An application of DCMG is presented to reveal the efficiency of the suggested ASMC method under both source voltage variation and constant power load variation.

Electricity theft has a considerable negative effect on energy suppliers and power infrastructure, leading to non-technical losses and business losses. Power quality deteriorates and overall profitability falls as a result of energy theft. By fusing information and energy flow, smart grids may assist solve the issue of power theft. The examination of smart grid data aids in the detection of power theft. However, the earlier techniques were not very good in detecting energy theft. In this work, we suggested an electricity theft detection approach using smart meter consumption data in order to handle the aforementioned issues and assist and assess energy supply businesses to lower the obstacles of limited energy, unexpected power usage, and bad power management. In specifically, the Deep CNN model effectively completes two tasks: it differentiates between energy that is not periodic and that is, while keeping the general features of data on power consumption. The trial’s results show that the deep CNN model outperforms prior ones and has the best level of accuracy for detecting energy theft.

For LED color-mixing, the different color LEDs need to be controlled independently, so a fully decoupled current control and current balancing of the modular structure is proposed for LED color-mixing system. The capacitive current-balancing structure is combined with the quasi-boost converter to realize the decoupled current control and current balancing in each module. The capacitive current-balancing structure is based on the relative reactance of the capacitor with respect to the equivalent resistance of the associated LED string. The inductor in quasi-boost converter is used to further stable the output current flowing through each LED string, and the switch in quasi-boost converter for controlling the current of each LED string adopts the parallel connection to protect LEDs from the current-spike caused by the switch. The structure is modularity and scalability so that the different color LEDs can be added and removed easily. Compared with SIMO (Single-Inductor-Multiple-Output) structure, the proposed structure has no cross-coupling issues. The LED string currents working with PWM mode have a good current-balancing performance since the equivalent resistance of LED string is much smaller than the reactance of capacitors. In addition, the resonant capacitors separate the grounds of the power source and LED strings, so all the switches in parallel with LED strings do not need the isolated gate drivers. A prototype with RGB modules has been built and evaluated, and the experimental results have a good agreement with the theoretical analysis.

Abstract Gastric cancer (GC) remains the third leading cause of cancer-related deaths. Chemoresistance is the major determinant of GC treatment failure. To explore the molecular mechanisms of GC chemoresistance, mass spectrometry was performed to detect the genes altered in expression between chemoresistant and chemosensitive GC. PRKA kinase anchor protein 8L (AKAP-8L) was identified as one of the top upregulated genes in chemoresistant GC tissues. Moreover, the higher AKAP-8L expression was associated with the lower survival rate in GC patients. Overexpression of AKAP-8L enhanced the GC cell stemness and chemoresistance of oxaliplatin in vivo and in vitro. AKAP-8L deficiency obtained the opposite results. Mechanistically, AKAP-8L interacted with Stearoyl-CoA desaturase 1 (SCD1) mRNA and IGF2BP1 protein, and regulated SCD1 mRNA stability via IGF2BP1-dependent manner. SCD1 played a critical role in mediating the function of AKAP-8L in GC cell stemness and chemoresistance. Clinically, AKAP-8L and SCD1 protein levels was positively associated with human GC chemoresistance. Taken together, our results demonstrated that AKAP-8L facilitates GC chemoresistance via regulating SCD1-mediated stemness of GC cells. AKAP8L may represent a novel therapeutic target to overcome GC chemoresistance.

Biomass from specialty crops, including almonds, walnuts, and numerous others, serves as an important resource for energy and materials as agricultural systems evolve towards greater sustainability and circularity in management and operations. Biochar was produced from almond shells in a laboratory furnace at temperatures between 300 and 750 °C for residence times of 30 and 90 min with moisture contents of 5% to 15% wet basis. Response surface methodology was used to optimize the biochar yield. Feedstock and product temperatures were continuously monitored throughout the experiments. In addition, larger batches of biochar were also produced in a fixed-bed pilot-scale pyrolyzer. The yield of biochar was determined as a weight fraction of the amount of oven-dry almond shells used in each experiment. Physical and chemical characteristics of biochars were evaluated. Pyrolysis temperature and time were found to be the significant parameters affecting the biochar yield, with second-order regression models derived to fit yield results. As anticipated, highest biochar yields (65%) were obtained at a pyrolysis temperature of 300 °C and a pyrolysis time of 30 min due to the limited volatilization at this short residence at low temperature affecting torrefaction of the feedstock. The average biochar yield from the fixed-bed pilot-scale experiments was 39.5% and more closely aligned with the fixed carbon from standard proximate analyses. Higher pyrolysis temperatures resulted in higher C:N ratio and pH with the highest C:N ratio of 19:1 and pH of 10.0 obtained at a pyrolysis temperature of 750 °C for 90 min. Particle density increased with the increase of pyrolysis temperature. Results of this study can aid in predicting biochar yields from almond shells under different pyrolysis conditions and determining the amount of biochar required for different applications.

Abstract This study is to explore the therapeutic effect and potential mechanisms of exosomal microRNAs (miRNAs) derived from the ovaries with primary ovarian insufficiency (POI). The POI mouse model was established by intraperitoneal injection of cyclophosphamide (CTX) and busulfan. The apoptosis of granulosa cells (GCs) incubated with exosomes extracted from ovarian tissues of control and POI groups was analyzed by flow cytometry. Then, high‐throughput sequencing was performed to detect the difference of miRNAs profile in ovarian tissue‐derived exosomes between the control and POI mice. The effect of differential miRNA on the apoptosis of CTX‐induced ovarian GCs was analyzed by flow cytometry. The results showed that POI mouse model was successfully established. Exosomes extracted from ovarian of normal and POI group have different effects on apoptosis of GCs induced by CTX. miRNA‐seq found that exosomal miR‐122‐5p in POI group increased significantly. miR‐122‐5p as the dominant miRNA targeting BCL9 was significantly upregulated in ovarian tissues of chemotherapy‐induced POI group. Exosomes derived from the ovaries in the control group and miR‐122‐5p inhibitor group attenuated the apoptosis of primary cultured ovarian GCs. In conclusion, exosomal miR‐122‐5p promoted the apoptosis of ovarian GCs by targeting BCL9, suggested that miR‐122‐5p may function as a potential target to restore ovarian function.

Abstract Cells cultivated in bioreactors offer many possibilities for the production of novel and nutritious food products. Scientific and technological advances in cellular agriculture and processing technologies have allowed for the development of new techniques to utilize in vitro animal cells, plant cells, and microorganisms to mimic the organoleptic and nutritional properties of traditional foods as well as to potentially develop entirely new product classes. This review compiles and discusses the state‐of‐the‐art cellular production and processing systems including 3D printing of customizable cell‐cultivated food products. In addition to the technological state‐of‐the art, this article reviews the nutritional characteristics of cell‐cultivated foods, introduces examples of new food products, and compiles economic characteristics and environmental impacts of each production system as assessed through technoeconomic analyses and lifecycle assessments. The factors influencing consumer acceptance of cell‐cultivated foods are articulated and the potential implications of these new technologies on traditional agricultural industries and food chains are discussed. Lastly, future research and development trajectories are introduced with suggestions for continued development.

The symmetrical segmentation of brain tumor images is crucial for both clinical diagnosis and computer-aided prognosis. Traditional manual methods are not only asymmetrical in terms of efficiency but also prone to errors and lengthy processing. A significant barrier to the process is the complex interplay between the deep learning network for MRI brain tumor imaging and the harmonious compound of both local and global feature information, which can throw off the balance in segmentation accuracy. Addressing this asymmetry becomes essential for precise diagnosis. In answer to this challenge, we introduce a balanced, end-to-end solution for brain tumor segmentation, incorporating modifications that mirror the U-Net architecture, ensuring a harmonious flow of information. Beginning with symmetric enhancement of the visual quality of MRI brain images, we then apply a symmetrical residual structure. By replacing the convolutional modules in both the encoder and decoder sections with deep residual modules, we establish a balance that counters the vanishing gradient problem commonly faced when the network depth increases. Following this, a symmetrical threefold attention block is integrated. This addition ensures a balanced fusion of local and global image features, fine-tuning the network to symmetrically discern and learn essential image characteristics. This harmonious integration remarkably amplifies the network’s precision in segmenting MRI brain tumors. We further validate the equilibrium achieved by our proposed model using three brain tumor segmentation datasets and four metrics and by juxtaposing our model against 21 traditional and learning-based counterparts. The results confirm that our balanced approach significantly elevates performance in the segmentation of MRI brain tumor images without an asymmetrical increase in computational time.

The respiratory status of dairy cows can reflect their heat stress and health conditions. It is widely used in the precision farming of dairy cows. To realize intelligent monitoring of cow respiratory status, a system based on infrared thermography was constructed. First, the YOLO v8 model was used to detect and track the nose of cows in thermal images. Three instance segmentation models, Mask2Former, Mask R-CNN and SOLOv2, were used to segment the nostrils from the nose area. Second, the hash algorithm was used to extract the temperature of each pixel in the nostril area of a cow to obtain the temperature change curve. Finally, the sliding window approach was used to detect the peaks of the filtered temperature curve to obtain the respiratory rate of cows. Totally 81 infrared thermography videos were used to test the system, and the results showed that the AP50 of nose detection reached 98.6%, and the AP50 of nostril segmentation reached 75.71%. The accuracy of the respiratory rate was 94.58%, and the correlation coefficient R was 0.95. Combining infrared thermography technology with deep learning models can improve the accuracy and usability of the respiratory monitoring system for dairy cows.

Prior to dispatch from manufacturing facilities, seeders require rigorous performance evaluations for their seeding capabilities. Conventional manual inspection methods are notably less efficient. This study introduces a wheat seeding detection approach anchored in an enhanced YOLOv5s image-processing technique. Building upon the YOLOv5s framework, we integrated four CBAM attention mechanism modules into its model. Furthermore, the traditional upsampling technique in the neck layer was superseded by the CARAFE upsampling method. The augmented model achieved an mAP of 97.14%, illustrating its ability to elevate both the recognition precision and processing speed for wheat seeds while ensuring that the model remains lightweight. Leveraging this advanced model, we can effectively count and locate seed images, enabling the precise calculation and assessment of sowing uniformity, accuracy, and dispersion. We established a sowing test bench and conducted experiments to validate our model. The results showed that after the model was improved, the average accuracy of wheat recognition was above 97.55% under different sowing rates and travel speeds. This indicates that this method has high precision for the total number of seed particles. The sowing rate and sowing travel speed were consistent with manual measurements and did not significantly affect uniformity, accuracy, or dispersion.

This study aimed to assess the feasibility of a novel weeding and fertilization scheme, namely, mechanical weeding plus a one-time deep application of a reduced amount of slow-release fertilizer for rice cultivation. The effects of the weeding and fertilization method on rice yield and quality were investigated using a split plot test as the research method. Two weeding methods, namely, chemical weeding (CW) and mechanical weeding (MW), and four fertilization methods were tested, including the conventional fertilization method (quantitative split broadcast application of fast-release N fertilizer (CK)), the quantitative split broadcast application of 80% fast-release N fertilizer (LCK), the one-time base application of slow-release fertilizer (SR), and the one-time deep application of 80% slow-release fertilizer (LSR). The results showed that the rice yield under MW with LSR treatment can maintain a high level—higher than 9.2 t ha−1 per year. This was attributed to the slow-release fertilizer and deep fertilization, which increased the number of stems and tillers in the pre-fertility and spike rate, respectively, resulting in a high panicle number with a 20% reduction of N fertilizer. Furthermore, mechanical weeding improved the seed-setting rate, resulting in a higher number of grains per panicle, a higher panicle number, and an increased thousand-grain weight, thereby maintaining a high yield. At the same time, the quality of rice under MW with LSR treatment improved, specifically reflected in the significant improvement of the processing and appearance quality of rice, a slight increase in protein content, and a reduction in the amylose content, thereby improving its nutritional quality while maintaining good cooking quality.

Images captured during rainy days present the challenge of maintaining a symmetrical balance between foreground elements (like rain streaks) and the background scenery. The interplay between these rain-obscured images is reminiscent of the principle of symmetry, where one element, the rain streak, overshadows or disrupts the visual quality of the entire image. The challenge lies not just in eradicating the rain streaks but in ensuring the background is symmetrically restored to its original clarity. Recently, numerous deraining algorithms that employ deep learning techniques have been proposed, demonstrating promising results. Yet, achieving a perfect symmetrical balance by effectively removing rain streaks from a diverse set of images, while also symmetrically restoring the background details, is a monumental task. To address this issue, we introduce an image-deraining algorithm that leverages multi-scale dilated residual recurrent networks. The algorithm begins by utilizing convolutional activation layers to symmetrically process both the foreground and background features. Then, to ensure the symmetrical dissemination of the characteristics of rain streaks and the background, it employs long short-term memory networks in conjunction with gated recurrent units across various stages. The algorithm then incorporates dilated residual blocks (DRB), composed of dilated convolutions with three distinct dilation factors. This integration expands the receptive field, facilitating the extraction of deep, multi-scale features of both the rain streaks and background information. Furthermore, considering the complex and diverse nature of rain streaks, a channel attention (CA) mechanism is incorporated to capture richer image features and enhance the model’s performance. Ultimately, convolutional layers are employed to fuse the image features, resulting in a derained image. An evaluation encompassing seven benchmark datasets, assessed using five quality metrics against various conventional and modern algorithms, confirms the robustness and flexibility of our approach.

Based on the photonic topological transition (PTT), the transmission properties of a black phosphorus/aluminum oxide multilayered hyperbolic metamaterial (HMM) are theoretically investigated in the mid-infrared region. The results demonstrate that an angular transparency window appears near the transition point of PTT, which is achieved by tailoring the topology of the HMM’s equi-frequency surface. The angular full width at half maximum of the transparency window is 2.34°, and the transmittance is higher than 99.8% at normal incidence. In addition, the operating wavelength can be flexibly tuned by adjusting the concentration of electrons. Besides, a layered cascade structure with a wide operating wavelength (1 µm) and an enhanced angular selectivity performance is proposed, which resolves the shortcoming of a single working wavelength. In addition, the spectral-selective behavior of absorption is also explained based on the PTT. These attractive properties make the black phosphorus-based HMM hold promise for potential applications in angularly selective systems and energy harvesting.

BackgroundThere are controversies on the pathophysiological alteration in patients with atrial fibrillation (AF) undergoing pulmonary vein isolation using different energy sources.ObjectivesWe evaluated the changes in plasma proteins in acute phase post-ablation in patients receiving cryoballoon ablation, radiofrequency balloon ablation, or radiofrequency ablation.MethodsBlood samples from eight healthy controls and 24 patients with AF were taken on the day of admission, day 1, and day 2 post-ablation and analyzed by the Olink proximity extension assay. Proteins were identified and performed with enrichment analysis. Protein–protein interaction network and module analysis were conducted using Cytoscape software.ResultsOf 181 proteins, 42 proteins in the cryoballoon group, 46 proteins in the radiofrequency balloon group, and 43 proteins in the radiofrequency group significantly changed after ablation. Most of the proteins altered significantly on the first day after ablation. Altered proteins were mainly involved in cytokine–cytokine receptor interaction. Both balloon-based ablations showed a similar shift toward enhancing cell communication and regulation of signaling while inhibiting neutrophil chemotaxis. However, radiofrequency ablation presented a different trend. Seed proteins, including osteopontin, interleukin-6, interleukin-10, C-C motif ligand 8, and matrix metalloproteinase-1, were identified. More significant proteins associated with hemorrhage and coagulation were selected in balloon-based ablations by machine learning.ConclusionPlasma protein response after three different ablations in patients with AF mainly occurred on the first day. Radiofrequency balloon ablation shared similar alteration in protein profile as cryoballoon ablation compared with radiofrequency ablation, suggesting that lesion size rather than energy source is the determinant in pathophysiological responses to the ablation.

Abstract Background The association between exposure to air pollution and sudden sensorineural hearing loss (SSNHL) has not been extensively discussed in the literature. Therefore, we conducted this nationwide study to evaluate the risk of SSNHL in Taiwanese residents with exposure to air pollution. Methods We enrolled subjects aged older than 20 years with no history of SSNHL from 1998 to 2010, and followed up until developing SSNHL, withdrawn from the National Health Insurance program, and the end of the database (2011/12/31). The air quality data are managed by Taiwan Environmental Protection Administration. The annual concentrations of PM2.5, SO2, CO, NO, and NO2 from 1998 to 2010 were classified into the three levels according to tertiles. We calculated the annual average of pollutants from baseline until the end of the study, and classified into tertiles. The adjusted hazard ratio (aHR) was estimated by using the multivariate Cox proportional hazard model. Results When considered continuous air pollutants concentration, subjects who exposed with higher concentration of CO (aHR = 2.16, 95% CI 1.50–3.11), NO (aHR = 1.02, 95% CI 1.01–1.03), and NO2 (aHR = 1.02, 95% CI 1.01–1.04) developing significant higher risk of SSNHL. When classified air pollutants concentration into low, moderate and high level by tertiles, and selected low level as reference, patients exposed with moderate (aHR = 1.56, 95% CI 1.20–2.04) or high level (aHR = 1.33, 95% CI 1.01–1.75) of PM2.5 showed significant higher risk of developing SSNHL. Conclusion This study indicated an increased risk of SSNHL in residents with long-term exposure to air pollution. Nevertheless, further experimental, and clinical studies are needed to validate the study findings.

The rapid growth of human civilizations has led to imminent pressures to develop new food products with increased nutritional characteristics and decreased environmental footprints. Filamentous fungi, a class of microorganisms that have been utilized in a wide variety of foods for thousands of years, have recently garnered widespread attention in research communities and commercial ventures seeking to explore new and innovative applications in a diverse array of food products including, but not limited to, their more established applications as alternative proteins. Technological advances in the cultivation and processing of filamentous fungi have created new frontiers in the control of textures, flavors, and nutritional properties of fungi-based foods. This review highlights technological advances in the production of fungi-based foods from cultivation to product manufacturing, presents the current state of the art in fungi-based food products, and offers thoughts on their future trajectories. Emphasis is given to circular bioprocessing concepts for the sustainable utilization of agricultural and food processing byproducts.

Abstract The pathophysiology of coronavirus disease 19 (COVID-19) involves a multitude of host responses, yet how they unfold during the course of disease progression remains unclear. Here, through integrative analysis of clinical laboratory tests, targeted proteomes, and transcriptomes of 963 patients in Shanghai, we delineate the dynamics of multiple circulatory factors within the first 30 days post-illness onset and during convalescence. We show that hypercortisolemia represents one of the probable causes of acute lymphocytopenia at the onset of severe/critical conditions. Comparison of the transcriptomes of the bronchoalveolar microenvironment and peripheral blood indicates alveolar macrophages, alveolar epithelial cells, and monocytes in lungs as the potential main sources of elevated cytokines mediating systemic immune responses and organ damages. In addition, the transcriptomes of patient blood cells are characterized by distinct gene regulatory networks and alternative splicing events. Our study provides a panorama of the host responses in COVID-19, which may serve as the basis for developing further diagnostics and therapy.

Yield is an important indicator in evaluating rice planting, and it is the collective result of various factors over multiple growth stages. To achieve a large-scale accurate prediction of rice yield, based on yield estimation models using a single growth stage and conventional spectral transformation methods, this study introduced the continuous wavelet transform algorithm and constructed models under the premise of combined multiple growth stages. In this study, canopy reflectance spectra at four important stages of rice elongation, heading, flowering and milky were selected, and then, a rice yield estimation model was constructed by combining vegetation index, first derivative and wavelet transform based on random forest algorithm or multiple stepwise regression. This study found that the combination of multiple growth stages significantly improved the model accuracy. In addition, after two validations, the optimal model combination for rice yield estimation is first derivative-wavelet transform-vegetation index-random forest model based on four growth stages, with the coefficient of determination (R2) of 0.86, the root mean square error (RMSE) of 35.50 g·m−2 and the mean absolute percentage error (MAPE) of 4.6% for the training set, R2 of 0.85, RMSE of 33.40 g.m−2 and MAPE 4.30% for the validation set 1, and R2 of 0.80, RMSE of 37.40 g·m−2 and MAPE of 4.60% for the validation set 2. The research results demonstrated that the established model could accurately predict rice yield, providing technical support and a foundation for large-scale statistical estimating of rice yield.

It has been well recognized that the development and use of artificial materials with high osteogenic ability is one of the most promising means to replace bone grafting that has exhibited various negative effects. The biomimetic features and unique physiochemical properties of nanomaterials play important roles in stimulating cellular functions and guiding tissue regeneration. But efficacy degree of some nanomaterials to promote specific tissue formation is still not clear. We hereby comparatively studied the osteogenic ability of our treated multi-walled carbon nanotubes (MCNTs) and the main inorganic mineral component of natural bone, nano-hydroxyapatite (nHA) in the same system, and tried to tell the related mechanism. In vitro culture of human adipose-derived mesenchymal stem cells (HASCs) on the MCNTs and nHA demonstrated that although there was no significant difference in the cell adhesion amount between on the MCNTs and nHA, the cell attachment strength and proliferation on the MCNTs were better. Most importantly, the MCNTs could induce osteogenic differentiation of the HASCs better than the nHA, the possible mechanism of which was found to be that the MCNTs could activate Notch involved signaling pathways by concentrating more proteins, including specific bone-inducing ones. Moreover, the MCNTs could induce ectopic bone formation in vivo while the nHA could not, which might be because MCNTs could stimulate inducible cells in tissues to form inductive bone better than nHA by concentrating more proteins including specific bone-inducing ones secreted from M2 macrophages. Therefore, MCNTs might be more effective materials for accelerating bone formation even than nHA.

This paper focuses on the steering motion control of an in-wheel motor-drive robot. The influence of the pulse-width modulation (PWM) duty cycle on steering motion and the steering control method have not yet been proved. Thus, this study aimed to design a steering controller for the off-center steer-by-wire system of a robot. The influence of the PWM duty cycle on the steering motion under different conditions is firstly tested on a test bench. Based on the optimal duty cycles of different cases found in the test, a two-stage fuzzy controller of the duty cycle is designed for the steering system. The first stage of the controller is used to dynamically adjust the PWM duty cycle of the electromagnetic friction lock (EFL). The second stage is designed to realize the self-tuning of the fuzzy controller’s quantization factor and the scale factor. Through two-stage control, the motion of the in-wheel motor and the EFL can be coordinated to realize stable and rapid steering. Considering the robots’ primary application in field roads at present, road tests were ultimately conducted to verify the proposed method. The test results show that the angle response rate of the steering arm is elevated with the increase in the steering angle signal. The proposed controller can sensitively track the target angles with smaller overshoot, yaw rate and lateral acceleration, and better steering accuracy than the PID (proportional–integral–differential) controller under different working conditions.

Abstract GNA13, encoding one of the G protein alpha subunits of heterotrimeric G proteins that transduce signals of G protein-coupled receptors (GPCR), is frequently mutated in germinal center B-cell-like diffuse large B-cell lymphoma (GCB-DLBCL) with poor prognostic outcomes. Due to the “undruggable” nature of GNA13, targeted therapy for these patients is not available. In this study, we found that palmitoylation of GNA13 not only regulates its plasma membrane localization, but also regulates GNA13’s stability. It is essential for the tumor suppressor function of GNA13 in GCB-DLBCL cells. Interestingly, GNA13 negatively regulates BCL2 expression in GCB-DLBCL cells in a palmitoylation-dependent manner. Consistently, BCL2 inhibitors were found to be effective in killing GNA13-deficient GCB-DLBCL cells in a cell-based chemical screen. Furthermore, we demonstrate that inactivating GNA13 by targeting its palmitoylation enhanced the sensitivity of GCB-DLBCL to the BCL2 inhibitor. These studies indicate that the loss-of-function mutation of GNA13 is a biomarker for BCL2 inhibitor therapy of GCB-DLBCL and that GNA13 palmitoylation is a potential target for combination therapy with BCL2 inhibitors to treat GCB-DLBCL with wild-type GNA13.

Cervical cancer is the fourth most prevalent disease among women. Prompt diagnosis and its management can significantly improve patients' survival rates. Therefore, routine screening for cervical cancer is of paramount importance. Herein, we explore the potential of a deep learning model to automatically distinguish abnormal cells from normal cells. The ThinPrep cytologic test dataset was collected from the fourth central hospital of Baoding city, China. Based on the dataset, four classification models were developed. The first model was a 10-layer convolutional neural network (CNN). The second model was an advancement of the first model equipped with a spatial pyramid pooling (SPP) layer (CNN + SPP) to treat cell images based on their sizes. Based on the first model, the third model replaced the CNN layers with the inception module (CNN + Inception). However, the fourth model incorporated both the SPP layer and the inception module into the first model (CNN + inception + SPP). The performances of the four models are estimated and compared by using the same testing data and evaluation index. The testing results demonstrated that the fourth model yields the best performance. Moreover, the area under the curve (AUC) for module four was 0.997.

Tomatoes contain active phytochemicals, including phytoene, phytofluene, beta-caroten, flavonoids, carotenoids, and lycopene but theirs effects on Weight, Body Mass Index, Blood Pressure and Inflammatory Factors is controversial. The aim of this systematic review and meta-analyze is reported the effects of Tomato consumption on Weight, Body Mass Index, Blood Pressure and Inflammatory Factors such as CRP and IL-6 in Randomized Controlled Trials. Literature search was conducted by a systematic search in PubMed/MEDLINE, Scopus, and Web of sciences (WOB) up to August 2019. There was no language or release time data restrictions. Two researcher conducted data extraction, independently. Dersimonian-laird random effect models were conducted to combining result of included studies. PRISMA guidelines (Preferred Items for Reporting of Systematic Reviews and Meta-Analyses) used for this meta-analysis. Nine studies met inclusion criteria for this meta-analysis. Combined results show not significant change in weight of participants (WMD: −0.03 kg, 95%CI: −0.59, 0.53, Pheterogeneity = 0.98) and BMI by tomato intervention (WMD: 0.07 kg/m2, 95%CI: −0.14, 0.29, Pheterogeneity = 0.99). Furthermore, blood pressure change was not significant too but pooled results show an significant reduction in IL-6 levels following intervention by tomato compared to control group (WMD: −0.12 pg/ml, 95% CI: −0.23, −0.02, Pheterogeneity = 0.001). Although, the pooled results from effect of tomato on CRP were not significant (WMD: 0.98 mg/l, 95%CI: −0.23, 219, Pheterogeneity = 0.09) but time-response analysis showed significant time dependent relation between tomato intervention and CRP levels. The results demonstrated a significant reduction in IL-6 by tomato consumption. Tomato reduce CRP levels in short period too. Keywords: Tomato, Weight, Blood pressure, IL-6, CRP

BackgroundRenal cell carcinoma is the most common malignant tumor of the kidney. The 5-year survival of renal cell carcinoma with distant metastasis is very low. Sohlh2 is a newly discovered tumor suppressor gene playing inhibitory roles in a variety of tumors, but its role in renal cell carcinoma has not been reported.MethodsTo clarify the role of Sohlh2 in the occurrence and development of renal cell carcinoma, we constructed stably transfected human renal cell carcinoma cell lines with Sohlh2 overexpression and Sohlh2 knockdown, separately. First, we studied the effects of Sohlh2 on proliferation, migration, invasion, and epithelial–mesenchymal transition (EMT) of renal cell carcinoma cells in vitro and in vivo. Then, we detected whether Sohlh2 functions through DNMT3a/Klotho using Western blotting, qPCR, and Cell Counting Kit-8 (CCK-8) assay. Finally, we collected 40 resected renal cell carcinoma samples to study the relevance between Sohlh2, DNMT3a, and Klotho by immunohistochemistry.ResultsOur results showed that Sohlh2 was downregulated in renal cell carcinoma, and its expression level was negatively correlated with tumor staging. Both in vitro and in vivo experiments confirmed that Sohlh2 overexpression inhibited the proliferation, migration, invasion, metastasis, and EMT of renal cell carcinoma. Sohlh2 functions through demethylation of Klotho by downregulating the expression of DNA methyltransferase of DNMT3a. In renal cell carcinoma, Sohlh2 was positively correlated with Klotho and negatively correlated with DNMT3a.ConclusionSohlh2 functions as a tumor suppressor gene in renal cell carcinoma by demethylation of Klotho via DNMT3a. Sohlh2 correlated with Klotho positively and with DNMT3a negatively in renal cell carcinoma. Our study suggests that Sohlh2 and DNMT3a/Klotho can be used as potential targets for the clinical treatment of renal cell carcinoma.

Precision dairy farming technology is widely used to improve the management efficiency and reduce cost in large-scale dairy farms. Machine vision systems are non-contact technologies to obtain individual and behavioral information from animals. However, the accuracy of image-based individual identification of dairy cows is still inadequate, which limits the application of machine vision technologies in large-scale dairy farms. There are three key problems in dairy cattle identification based on images and biometrics: (1) the biometrics of different dairy cattle may be similar; (2) the complex shooting environment leads to the instability of image quality; and (3) for the end-to-end identification method, the identity of each cow corresponds to a pattern, and the increase in the number of cows will lead to a rapid increase in the number of outputs and parameters of the identification model. To solve the above problems, this paper proposes a cascaded dairy individual cow identification method based on DeepOtsu and EfficientNet, which can realize a breakthrough in dairy cow group identification accuracy and speed by binarization and cascaded classification of dairy cow body pattern images. The specific implementation steps of the proposed method are as follows. First, the YOLOX model was used to locate the trunk of the cow in the side-looking walking image to obtain the body pattern image, and then, the DeepOtsu model was used to binarize the body pattern image. After that, primary classification was carried out according to the proportion of black pixels in the binary image; then, for each subcategory obtained by the primary classification, the EfficientNet-B1 model was used for secondary classification to achieve accurate and rapid identification of dairy cows. A total of 11,800 side-looking walking images of 118 cows were used to construct the dataset; and the training set, validation set, and test set were constructed at a ratio of 5:3:2. The test results showed that the binarization segmentation accuracy of the body pattern image is 0.932, and the overall identification accuracy of the individual cow identification method is 0.985. The total processing time of a single image is 0.433 s. The proposed method outperforms the end-to-end dairy individual cow identification method in terms of efficiency and training speed. This study provides a new method for the identification of individual dairy cattle in large-scale dairy farms.

California is the leading dairy state in the United States. The total sale of milk and its products represents about $6.3 billion annually out of the $50 billion generated from all agricultural production in the state. However, methane emissions from dairy manure and enteric fermentation represented nearly half of all annual methane emissions in California, with dairy manure accounting for 25%, and enteric fermentation for 20%. Methane emissions originating from manure are produced primarily from anaerobic settling basins and lagoons, which are the most common manure storage systems in the state. To achieve sustainability on dairy farms and to comply with state regulations for air and climate pollutants, dairy farms have implemented technologies such as anaerobic digestion and alternative manure management technologies. In addition, governmental incentive programs have been deployed to partially fund these technologies for eligible dairies in the state. The present article reviews the design and operations, effectiveness, and economics of the most common technologies employed in Californian dairies in reducing methane emissions. The technologies studied include anaerobic digesters, mechanical separators, compost-bedded pack barns, manure vacuuming followed by drying, and weeping walls. The current status and estimated effectiveness of government incentive programs are reviewed and recommendations for improvements presented. Finally, future trends and research needs for mitigating the emissions in Californian dairies are identified.

Abstract Background DNMT3A R882H, a frequent mutation in acute myeloid leukemia (AML), plays a critical role in malignant hematopoiesis. Recent findings suggest that DNMT3A mutant acts as a founder mutation and requires additional genetic events to induce full-blown AML. Here, we investigated the cooperation of mutant DNMT3A and NRAS in leukemogenesis by generating a double knock-in (DKI) mouse model harboring both Dnmt3a R878H and Nras G12D mutations. Methods DKI mice with both Dnmt3a R878H and Nras G12D mutations were generated by crossing Dnmt3a R878H knock-in (KI) mice and Nras G12D KI mice. Routine blood test, flow cytometry analysis and morphological analysis were performed to determine disease phenotype. RNA-sequencing (RNA-seq), RT-PCR and Western blot were carried out to reveal the molecular mechanism. Results The DKI mice developed a more aggressive AML with a significantly shortened lifespan and higher percentage of blast cells compared with KI mice expressing Dnmt3a or Nras mutation alone. RNA-seq analysis showed that Dnmt3a and Nras mutations collaboratively caused abnormal expression of a series of genes related to differentiation arrest and growth advantage. Myc transcription factor and its target genes related to proliferation and apoptosis were up-regulated, thus contributing to promote the process of leukemogenesis. Conclusion This study showed that cooperation of DNMT3A mutation and NRAS mutation could promote the onset of AML by synergistically disturbing the transcriptional profiling with Myc pathway involvement in DKI mice.

Background Antibiotic resistance genes (ARGs) are considered to be emerging environmental contaminants of concern potentially posing risks to human and animal health, and this research studied the prevalence of antimicrobial resistance in dairy manure. Methods This study is focused on investigating prevalence of ARGs in California dairy farm manure under current common different manure management. A total of 33 manure samples were collected from multiple manure treatment conditions: (1) flushed manure (FM), (2) fresh pile (FP), (3) compost pile (CP), (4) primary lagoon (PL), and (5) secondary lagoon (SL). After DNA extraction, all fecal samples were screened by PCR for the presence of eight ARGs: four sulfonamide ARGs (sulI, sulII, sulIII, sulA), two tetracycline ARGs (tetW, tetO), two macrolide-lincosamide-streptogramin B (MLSB) ARGs (ermB, ermF). Samples were also screened for two mobile genetic elements (MGEs) (intI1, tnpA), which are responsible for dissemination of ARGs. Quantitative PCR was then used to screen all samples for five ARGs (sulII, tetW, ermF, tnpA and intI1). Results Prevalence of genes varied among sample types, but all genes were detectable in different manure types. Results showed that liquid-solid separation, piling, and lagoon conditions had limited effects on reducing ARGs and MGEs, and the effect was only found significant on tetW (p = 0.01). Besides, network analysis indicated that sulII was associated with tnpA (p < 0.05), and Psychrobacter and Pseudomonas as opportunistic human pathogens, were potential ARG/MGE hosts (p < 0.05). This research indicated current different manure management practices in California dairy farms has limited effects on reducing ARGs and MGEs. Improvement of different manure management in dairy farms is thus important to mitigate dissemination of ARGs into the environment.

To solve the problems of high labor costs, a low weeding rate and a high seedling injury rate in the direct seeding of rice fields, this paper presents a reciprocating inter-row weeding machine for strip-seeded rice. The machine uses a combination of weeding wheels and weeding shovels to improve the efficiency of weeding between rice rows. Its reciprocating mechanism was designed and optimized. The simulation model of weeding teeth–paddy soil interaction was established in EDEM. The structural parameters of the weeding teeth were optimized, and the bending angle of the optimized weeding teeth was 55°. A prototype trial production and field tests were carried out. The results showed that the prototype’s inter-row weeding rate was between 80.2% and 85.3% and the seedling injury rate was between 3.5% and 5.1% when the prototype’s working speed was 1~3 km h−1. The faster the speed of the prototype, the lower the inter-row weeding rate and the higher the seedling injury rate.

Polyhydroxyalkanoates (PHA) is an emerging biodegradable plastic family that can replace a broad spectrum of conventional thermoplastics and is promising in the sustainable process development and valorization of organic waste. This study established a novel production system of PHA from food waste through halophilic microbial fermentation with spent medium recycling. The essential processing parameters for batch cultivation of Haloferax mediterranei were optimized for food waste substrate (a 40 g/L loading and 2.5 vvm of aeration), which achieved a yield of 0.3 g PHA/g COD consumed. A batch bioreactor system was developed, which produced 7.0 ± 0.7 g/L cell dry mass and 4.5 ± 0.2 g/L PHA with a 20% dissolved oxygen (DO) level. A DO above 50% saturation resulted in faster cell growth and similar cell mass production but 25% less PHA production. The spent saline medium, treated with H2O2 and rotary evaporation, was successfully reused for four consecutive batches and provided consistent PHA concentrations and product qualities.

Abstract IRF8 is a key regulator of innate immunity receptor signaling and plays diverse functions in the development of hematopoietic cells. The effects of IRF8 on hematopoietic stem cells (HSCs) are still unknown. Here, it is demonstrated that IRF8 deficiency results in a decreased number of long‐term HSCs (LT‐HSCs) in mice. However, the repopulation capacity of individual HSCs is significantly increased. Transcriptomic analysis shows that IFN‐γ and IFN‐α signaling is downregulated in IRF8‐deficient HSCs, while their response to proinflammatory cytokines is unchanged ex vivo. Further tests show that Irf8−/− HSCs can not respond to CpG, an agonist of Toll‐like receptor 9 (TLR9) in mice, while long‐term CpG stimulation increases wild‐type HSC abundance and decreases their bone marrow colony‐forming capacity. Mechanistically, as the primary producer of proinflammatory cytokines in response to CpG stimulation, dendritic cells has a blocked TLR9 signaling due to developmental defect in Irf8−/− mice. Macrophages remain functionally intact but severely reduce in Irf8−/− mice. In NK cells, IRF8 directly regulates the expression of Tlr9 and its deficiency leads to no increased IFNγ production upon CpG stimulation. These results indicate that IRF8 regulates HSCs, at least in part, through controlling TLR9 signaling in diverse innate immune cells.

This study focuses on a wheeled mobile robot used for detection, weeding and information monitoring in agriculture. However, it is difficult to reach satisfactory motion mode switching (MMS) performance. This paper aimed at exploring the optimal control parameters guaranteeing smooth MMS of four-wheel steering. Single factor tests were first conducted using a test-bench. A binary quadratic general rotation combination test was designed to obtain the optimal parameters. An entropy weight method was introduced to construct the four indexes as a comprehensive index. The optimal combination of the parameters was obtained, based on the regression equation. The results showed that the two factors and their interaction had a significant impact on the comprehensive index (p < 0.05). The best combinations of the speed of the stepper motor and locking voltage were 56 r·min−1 and 3.96 V for 15° steering, 72 r·min−1 and 4.35 V for 30°, and 107 r·min−1 and 5.50 V for 45°, respectively. A verification test was performed using the prototype of the robot chassis. The results demonstrated that the MMS process was smooth and stable, and the proposed method was effective. This study is a beneficial exploration of the experimental method concerning wheeled robots.

Vacuum-insulated glazing (VIG) has excellent sound and heat insulation and anti-fogging properties, having been a typical structure–function-integrated glass deep processing product. However, overlapping, vacancy and excessive spacing distance of the supporting pillars will increase the concentrated stress for the glass substrate, raising the potential risk of failure. Therefore, this study, aiming to address the high cost of sample preparation and the multiple factors affecting stress distribution, developed a multi-objective design for supporting stress. In this paper, a multi-objective optimization model was designed based on comprehensive mechanical analysis under square-distributed supporting. The critical supporting pillars radius as well as the critical spacing distance were solved and met the strength requirement. Case simulation demonstrated that a 0.2 mm or more radius and a 63 mm or less spacing distance for the supporting pillars were acceptable placement methods which conformed to the design requirements. This research will act as a theoretical reference for future studies, promoting the in-depth development of VIG and exploration of high-strength safety products.

It is of great fundamental significance and practical application to understand the binding sites and dynamic process of the interaction between doxorubicin (DOX) and DNA molecules. Based on the Confocal Raman spectroscopy, the interaction between DOX and calf thymus DNA has been systemically investigated, and some meaningful findings have been found. DOX molecules can not only interact with all four bases of DNA molecules, i.e., adenine, thymine, cytosine, guanine, and phosphate, but also affect the DNA conformation. Meanwhile, the binding site of DOX and its derivatives such as daunorubicin and epirubicin is certain. Furthermore, the interaction between DOX and DNA molecules is a dynamic process since the intensities of each characteristic peaks of the base, e.g., adenine, cytosine, and phosphate, are all regularly changed with the interaction time. Finally, a dynamic mechanism model of the interaction between DOX and DNA molecules is proposed; that is, there are two kinds of interaction between DOX and DNA molecules: DOX-DNA acts to form a complex, and DOX-DOX acts to form a multimer. The two effects are competitive, as the former compresses DNA molecules, and the latter decompresses these DNA molecules. This work is helpful for accurately understanding and developing new drugs and pathways to improve and treat DOX-induced cytotoxicity and cardiotoxicity.

Leaf area index (LAI) is one of the indicators measuring the growth of rice in the field. LAI monitoring plays an important role in ensuring the stable increase of grain yield. In this study, the canopy reflectance spectrum of rice was obtained by ASD at the elongation, booting, heading and post-flowering stages of rice, and the correlations between the original reflectance (OR), first-derivative transformation (FD), reciprocal transformation (1/R), and logarithmic transformation (LOG) with LAI were analyzed. Characteristic bands of spectral data were then selected based on the successive projections algorithm (SPA) and Pearson correlation. Moreover, ridge regression (RR), partial least squares (PLS), and multivariate stepwise regression (MSR) were conducted to establish estimation models based on characteristic bands and vegetation indices. The research results showed that the correlation between canopy spectrum and LAI was significantly improved after FD transformation. Modeling using SPA to select FD characteristic bands performed better than using Pearson correlation. The optimal modeling combination was FD-SPA-VI-RR, with the coefficient of determination (R2) of 0.807 and the root-mean-square error (RMSE) of 0.794 for the training set, R2 of 0.878 and RMSE of 0.773 for the validation set 1, and R2 of 0.705 and RMSE of 1.026 for the validation set 2. The results indicated that the present model may predict the rice LAI accurately, meeting the requirements of large-scale statistical monitoring of rice growth indicators in the field.

Liquid dairy manure, which is produced in enormous quantities in flush dairy manure management systems, is commonly used as an alternative to chemical fertilizers. It provides nutrient benefits to crops and soils. While dairy waste is a well-accepted and widely used fertilizer, the presence of indicator organisms and human pathogens in manure may lead to pathogen contamination in crops and soils. This study is focused on the examination of ozone gas-based sterilization. In the past, ozone (O3) has been used for sanitizing various foods and solid surfaces, but the potential of O3 for eliminating human pathogens in liquid dairy waste is not studied yet. Pathogens such as Salmonella Typhimurium and Escherichia coli O157:H7 are reported to be present in liquid dairy manure, and this research evaluated the effects of various levels of ozone on the survival of these two pathogens. We designed a continuous type O3 treatment system that has four major components: (1) ozone generator using oxygen; (2) ozone concentration control by mixing with pure air; (3) continuous monitoring of ozone concentrations; and (4) ozone experiment chambers. Various levels of ozone (43.26, 87.40, and 132.46 mg·L−1) were produced in the ozone system, and subsequently, ozone was diffused through liquid manure. Liquid manure was exposed to ozone for multiple durations (30, 60, and 120 min). To determine the effectiveness of O3 in eliminating pathogens, time-series samples were collected and analyzed for determining the levels of S. typhimurium and E. coli O157:H7. Preliminary results showed that ozone concentrations of 132.46 mg/L, and exposure time of 120 min resulted in the reduced levels of E. coli and Salmonella. Low levels of ozone and limited exposure time were found to be less effective in pathogen removal potentially due to high solid contents. Additional studies carrying out experiments to evaluate the impacts of solids in combination with ozone concentrations will provide further insights into developing full-scale ozone-based treatment systems.

Antibiotic resistance genes (ARGs) are emerging contaminants causing serious global health concern. Interventions to address this concern include improving our understanding of methods for treating waste material of human and animal origin that are known to harbor ARGs. Anaerobic digestion is a commonly used process for treating dairy manure, and although effective in reducing ARGs, its mechanism of action is not clear. In this study, we used three ARGs to conducted a longitudinal bench scale anaerobic digestion experiment with various temperatures (28, 36, 44, and 52°C) in triplicate using fresh dairy manure for 30 days to evaluate the reduction of gene abundance. Three ARGs and two mobile genetic elements (MGEs) were studied: sulfonamide resistance gene (sulII), tetracycline resistance genes (tetW), macrolide-lincosamide-streptogramin B (MLSB) superfamily resistance genes (ermF), class 1 integrase gene (intI1), and transposase gene (tnpA). Genes were quantified by real-time quantitative PCR. Results show that the thermophilic anaerobic digestion (52°C) significantly reduced (p < 0.05) the absolute abundance of sulII (95%), intI1 (95%), tnpA (77%) and 16S rRNA gene (76%) after 30 days of digestion. A modified Collins-Selleck model was used to fit the decay curve, and results suggest that the gene reduction during the startup phase of anaerobic digestion (first 5 days) was faster than the later stage, and reductions in the first five days were more than 50% for most genes.

In order to solve the problems of blocking the drainage ditch and reducing the soil flatness caused by soil accumulation when using compound planter with plowshare to ditch, a spiral soil separation mechanism (SSSM) is designed. The SSSM is analyzed. In order to obtain the optimal parameters of the SSSM, based on the discrete element method, the multifactor test is carried out with the embedded depth, pitch, and rotation speed of the spiral blade as the test factors and the soil separation distance and uniformity as the evaluation index. The optimal parameters are the embedded depth 49 mm, pitch 331 mm, and rotation speed of the spiral blade 318 r min−1. The field experiment is carried out with these parameters, with soil separation distance 900 mm and standard deviation of soil height 7.8 mm, which is consistent with the simulation results. No blockage of drainage ditch was found, which shows that this device can effectively solve the problem. This study can provide a reference for the design of soil separation equipment using spiral soil separation device.

microRNAs (miRNAs or miRs) play important roles in modulating the occurrence and progression of atherosclerosis and acute coronary syndrome (ACS). Herein, this study aimed to investigate the possible role of miR-9 in the development of atherosclerosis. Initially, the differentially expressed genes associated with ACS were screened and miRNAs that regulate syndecan-2 (SDC2) were predicted using microarray analysis. Furthermore, the biological functions of miR-9 and SDC2 on aortic plaque area, proliferation of collagen fibers, Mac-3-labeled macrophages, inflammatory response, and levels of the focal adhesion kinase/extracellular signal-regulated kinase (FAK/ERK) signaling pathway-related proteins in atherosclerosis were evaluated after ectopic miR-9 expression or SDC2 depletion in ACS mice using oil red O staining, Masson’s trichrome staining, immunohistochemistry, and Western blot analysis, respectively. SDC2 was highly-expressed, while miR-9 was poorly-expressed in atherosclerosis. Additionally, miR-9 targeted SDC2 and negatively-regulated its expression. Up-regulation of miR-9 reduced aortic plaque area, the proliferation of collagen fibers, Mac-3-labeled macrophages and levels of IL-6, IL-1β, and TNF-α by suppressing SDC2 and the FAK/ERK signaling pathway, thereby ameliorating atherosclerosis in ACS mice. In conclusion, the current study provides evidence that miR-9 retards atherosclerosis by repressing SDC2 and the FAK/ERK signaling pathway, highlighting a new theoretical basis for the treatment of atherosclerosis.

Abstract China produces large amount of cotton stalk (CS) residues as agricultural biomass, which are incinerated on-site, causing air pollution. The high organic content of CS could be utilized for biogas production, but the direct digestion without pretreatment always leads to a low methane yield and biodegradability, due to the complicated structure of lignocellulose. In order to search best fitting pretreatment methods in effective anaerobic digestion (AD) of CS, effects of various pretreatments including KOH, NaOH, Ca(OH)2, alkali hydrogen peroxide (AHP), H2SO4, H3PO4 and steam explosion (SE) were studied. It was seen that all treatments resulted in varying methane yields. Among all the pretreatments, acid pretreatment is not suitable for AD of CS. The results showed that the highest cumulative methane yield (CMY) of 192.4 mL·gVS−1 was obtained after 3% AHP pretreatment of CS, and the methane yield improved by 254.3% than the untreated CS. Therefore, AHP treatment was proven to be an efficient pretreatment technique. XRD and FTIR analyses had shown that pretreated CS had favorable structural changes. This research is beneficial in developing environment friendly and cost-effective pretreatment technologies to utilize CS for methane production in future application.

In the multiple cropping regions of southern China, straw returning has become a widely practiced agronomic measure for rice cultivation. However, excessive straw often leads to a high proportion of stubble in topsoil, which prolongs the leveling time of the paddy field and delays the transplanting date for rice seedlings. In particular, scholars in this region have successively improved multiple paddy field levelers to realize excellent straw returning and subsequent land preparation synchronously, but the economic benefit from land preparation to crop harvest was less reported. Therefore, this study carried out a 2-year rice cultivation experiments to compare the effects of paddy field preparation methods on rice growth and economic benefits within the same growing duration. Three treatments were designed: traditional tillage (TT), double axis rotary tillage (DR) and multiple operations for paddy field preparation (DR + ML), with three repeats. The results showed that DR + ML treatment simplified the operation process while improving the quality of land preparation. Within the same growing duration, DR + ML treatment could reduce the paddy field preparation time and extend the growing time in the field by 5–6 days. Furthermore, in comparison to TT treatment, DR + ML showed advantages in stimulating plant development, increasing dry matter accumulation (DMA), and thereby increasing rice yield by more than 12%. The economic benefits were mainly reflected in saving operation cost of paddy field preparation and improving the output (grain yield), which can generally increase the total profit by 58%. The implementation of this study can provide a reference for a simplified high yield cultivation technique in rice-related multiple cropping systems.
