Los jóvenes de entre 16 y 24 años, popularmente conocidos como Generación Z, posmilénica o centurial, son la primera generación en disponer de un acceso fácil y frecuente a internet desde una edad temprana, por lo que está expuesta, como ninguna otra generación con anterioridad, a una presencia constante de la tecnología y de los recursos digitales tanto en su proceso educativo como en su vida personal. En este artículo se analiza la percepción que de las noticias falsas tiene la sociedad española y, especialmente, los jóvenes integrantes de la denominada Generación Z. Saber qué medios consultan, qué los lleva a compartir determinados contenidos y con qué perspectivas afrontan el problema de la desinformación son algunos de los objetivos de esta investigación. Para ello, se ha empleado una metodología mixta que combina un estudio demoscópico y grupos de discusión que proporcionan una visión más completa y matizada del fenómeno. Los resultados permiten comprobar que estos jóvenes constituyen el grupo demográfico más disruptivo con el proceso comunicativo tradicional y son, a su vez, los más expuestos a las redes sociales y los más indiferentes a los medios tradicionales y al fenómeno de la desinformación.

Saffron (Crocus sativus L.), a historically significant crop valued for its nutraceutical properties, has been poorly explored from a phytosanitary perspective. This study conducted a thorough examination of viruses affecting saffron samples from Spanish cultivars, using high-throughput sequencing alongside a systematic survey of transcriptomic datasets from Crocus sativus at the Sequence Read Archive. Our analysis unveiled a broad diversity and abundance, identifying 17 viruses across the 52 analyzed libraries, some of which were highly prevalent. This includes known saffron-infecting viruses and previously unreported ones. In addition, we discovered 7 novel viruses from the Alphaflexiviridae, Betaflexiviridae, Potyviridae, Solemoviridae, and Geminiviridae families, with some present in libraries from various locations. These findings indicate that the saffron-associated virome is more complex than previously reported, emphasizing the potential of phytosanitary analysis to enhance saffron productivity.

Abstract Background Saffron (Crocus sativus) is considered the world's most expensive spice. Used mainly as a colorant for foodstuffs, it is highly appreciated for its aromatic and flavouring properties. Since no molecular markers for this species have been found in the literature, the objective of this study was to determine whether phenotypical differences found in C. sativus were supported by molecular analyses. Findings Thirty primers from Operon Technologies were used in random amplified polymorphic DNA (RAPD) analysis, forty eight primers were screened using intersimple sequence repeats (ISSR) method and fifteen primers derived from a microsatellites library flanking sequences with repeat motifs were assayed in forty three isolates of C. sativus from eleven different countries and a C. kotschyanus isolate was used as outgroup. No polymorphic bands were detected in any of the accessions combining the different approaches used in this study. Conclusion According to our findings, all accessions appear identical clones, not only because morphological characters but also at a molecular level. These data strongly suggested that C. sativus is a monomorphic species. Thus, genome sequencing is needed to find molecular markers for saffron.

Crocins are high-value water soluble pigments that have long been recognized for their medicinal properties, and whose demand has increased worldwide in recent years. In the present study transgenic tomato fruits engineered for the production of crocins and hp3/Bsh tomato mutants with alterations in the carotenoid metabolism, have been combined to increase the levels of crocins in tomato fruits. Tomato fruits from F4 plants showed high antioxidant capacity and the crocin-rich juice of the produced tomato fruit was protected neuroblastoma cells against oxidative insult, through its ability to activate factor 2 related to nuclear erythroid factor 2 (Nrf2). The bio-accessibility of crocins in the juice showed values similar to the ones observed with saffron. Overall, our results support that crocin-fortified tomatoes could result in higher crocin availability and have additional health-promoting effects and could provide better protection against oxidative stress related chronic diseases in humans.

Roma ciudad abierta no solo es considerada una película que retrata la resistencia italiana y que promueve la propaganda antifascista, sino que, además, representa un ejemplo del estilo rosselliniano, de su forma de captar la realidad en su crudeza. Una realidad marcada por hombres y mujeres que sufren, que luchan contra las adversidades y que anhelan un futuro diferente. Sin caer en el pesimismo ni en la desilusión, Rossellini presenta la tragedia que Italia estaba viviendo en los años finales de la Segunda Guerra Mundial. El objetivo de la presente investigación es reflexionar sobre la importancia de la película, destacando cómo, a través de una obra atemporal, el cineasta crea una obra maestra para que los trágicos acontecimientos de la época no caigan en el olvido. Tras un atento analisis de la película, se pondrá el acento en la forma de representar el miedo y la muerte que impregnaban la sociedad romana, entre realismo y simbolismo. La película creó escuela, siendo pionera en la idea de fomentar una conciencia crítica sobre acontecimientos histórico-sociales cercanos en el tiempo.

Carotenoids are C40 isoprenoids with well-established roles in photosynthesis, pollination, photoprotection, and hormone biosynthesis. The enzymatic or ROS-induced cleavage of carotenoids generates a group of compounds named apocarotenoids, with an increasing interest by virtue of their metabolic, physiological, and ecological activities. Both classes are used industrially in a variety of fields as colorants, supplements, and bio-actives. Crocins and picrocrocin, two saffron apocarotenoids, are examples of high-value pigments utilized in the food, feed, and pharmaceutical industries. In this study, a unique construct was achieved, namely O6, which contains CsCCD2L, UGT74AD1, and UGT709G1 genes responsible for the biosynthesis of saffron apocarotenoids driven by a patatin promoter for the generation of potato tubers producing crocins and picrocrocin. Different tuber potatoes accumulated crocins and picrocrocin ranging from 19.41–360 to 105–800 μg/g DW, respectively, with crocetin, crocin 1 [(crocetin-(β-D-glucosyl)-ester)] and crocin 2 [(crocetin)-(β-D-glucosyl)-(β-D-glucosyl)-ester)] being the main compounds detected. The pattern of carotenoids and apocarotenoids were distinct between wild type and transgenic tubers and were related to changes in the expression of the pathway genes, especially from PSY2, CCD1, and CCD4. In addition, the engineered tubers showed higher antioxidant capacity, up to almost 4-fold more than the wild type, which is a promising sign for the potential health advantages of these lines. In order to better investigate these aspects, different cooking methods were applied, and each process displayed a significant impact on the retention of apocarotenoids. More in detail, the in vitro bioaccessibility of these metabolites was found to be higher in boiled potatoes (97.23%) compared to raw, baked, and fried ones (80.97, 78.96, and 76.18%, respectively). Overall, this work shows that potatoes can be engineered to accumulate saffron apocarotenoids that, when consumed, can potentially offer better health benefits. Moreover, the high bioaccessibility of these compounds revealed that potato is an excellent way to deliver crocins and picrocrocin, while also helping to improve its nutritional value.

Crocins are high-value soluble pigments that are used as colorants and supplements, their presence in nature is extremely limited and, consequently, the high cost of these metabolites hinders their use by other sectors, such as the pharmaceutical and cosmetic industries. The carotenoid cleavage dioxygenase 2L (CsCCD2L) is the key enzyme in the biosynthetic pathway of crocins in Crocus sativus. In this study, CsCCD2L was introduced into Nicotiana tabacum and Nicotiana glauca for the production of crocins. In addition, a chimeric construct containing the Brevundimonas sp. β-carotene hydroxylase (BrCrtZ), the Arabidopsis thaliana ORANGE mutant gene (AtOrMut), and CsCCD2L was also introduced into N. tabacum. Quantitative and qualitative studies on carotenoids and apocarotenoids in the transgenic plants expressing CsCCD2L alone showed higher crocin level accumulation in N. glauca transgenic plants, reaching almost 400 μg/g DW in leaves, while in N. tabacum 36 μg/g DW was obtained. In contrast, N. tabacum plants coexpressing CsCCD2L, BrCrtZ, and AtOrMut accumulated, 3.5-fold compared to N. tabacum plants only expressing CsCCD2L. Crocins with three and four sugar molecules were the main molecular species in both host systems. Our results demonstrate that the production of saffron apocarotenoids is feasible in engineered Nicotiana species and establishes a basis for the development of strategies that may ultimately lead to the commercial exploitation of these valuable pigments for multiple applications.

SUMMARY: Crocins are glucosylated apocarotenoids present in flowers and fruits of a few plant species, including saffron, gardenia, and Buddleja. The biosynthesis of crocins in these plants has been unraveled, and the enzymes engineered for the production of crocins in heterologous systems. Mullein (Verbascum sp.) has been identified as a new source of crocins and picrocrocin. In this work, we have identified eight enzymes involved in the cleavage of carotenoids in two Verbascum species, V. giganteum and V. sinuatum. Four of them were homologous to the previously identified BdCCD4.1 and BdCCD4.3 from Buddleja, involved in the biosynthesis of crocins. These enzymes were analyzed for apocarotenogenic activity in bacteria and Nicotiana benthamiana plants using a virus‐driven system. Metabolic analyses of bacterial extracts and N. benthamiana leaves showed the efficient activity of these enzymes to produce crocins using β‐carotene and zeaxanthin as substrates. Accumulations of 0.17% of crocins in N. benthamiana dry leaves were reached in only 2 weeks using a recombinant virus expressing VgCCD4.1, similar to the amounts previously produced using the canonical saffron CsCCD2L. The identification of these enzymes, which display a particularly broad substrate spectrum, opens new avenues for apocarotenoid biotechnological production. [ABSTRACT FROM AUTHOR]

Abstract Crocins, the red soluble apocarotenoids of saffron, accumulate in the flowers of Crocus species in a developmental and tissue-specific manner. In Crocus sieberi, crocins accumulate in stigmas but also in a distinct yellow tepal sector, which we demonstrate contains chromoplast converted from amyloplasts. Secondary metabolites were analysed by LC-DAD-HRMS, revealing the progressive accumulation of crocetin and crocins in the yellow sector, which were also localized in situ by Raman microspectroscopy. To understand the underlying mechanisms of crocin biosynthesis, we sequenced the C. sieberi tepal transcriptome of two differentially pigmented sectors (yellow and white) at two developmental stages (6 and 8) by Illumina sequencing. A total of 154 million high-quality reads were generated and assembled into 248,099 transcripts. Differentially expressed gene analysis resulted in the identification of several potential candidate genes involved in crocin metabolism and regulation. The results provide a first profile of the molecular events related to the dynamics of crocetin and crocin accumulation during tepal development, and present new information concerning apocarotenoid biosynthesis regulators and their accumulation in Crocus. Further, reveals genes that were previously unknown to affect crocin formation, which could be used to improve crocin accumulation in Crocus plants and the commercial quality of saffron spice.

Biogenic-silver nanoparticles emerge as new nanosilver platforms that allow us to obtain silver nanoparticles via “green chemistry”. In our study, biogenic-silver nanoparticles were obtained from Iris tuberosa leaf extract. Nanoparticles were characterized by a UV-vis spectroscopy, dynamical light scattering technique. The transmission electron microscope revealed spheric and irregular nanoparticles with 5 to 50 nm in diameter. Antimicrobial properties were evaluated against typical microbial contaminants found in cosmetic products, showing high antimicrobial properties. Furthermore, natural moisturizing cream was formulated with biogenic-silver nanoparticles to evaluate the preservative efficiency through a challenge test, indicating its promising use as preservative in cosmetics.

El ingreso de Silvio Berlusconi en la arena política italiana ha provocado un cambio en la comunicación política, inaugurando una etapa marcada por el protagonismo de los medios de comunicación, el poder de la imagen, la política como espectáculo y la irrupción del video-poder. Su modelo resultó exitoso y apetecible de ser emulado. Y así fue: la campaña electoral de Donald Trump mostró evidentes analogías con el modelo berlusconiano. A través del análisis del discurso político de los dos políticos y de la literatura publicada sobre el tema, se pondrá de manifiesto una nueva forma de comunicar, más próxima al mundo del espectáculo y del entretenimiento. Tanto Berlusconi como Trump son deliberadamente transgresores, adoptan eslóganes provocadores y pegadizos, recurren a expresiones ofensivas. Dos modelos análogos y aún así con algunas diferencias que hacen presagiar una nueva forma de comunicar.

En el caso de la crisis armada que vive Siria desde marzo del año 2011, el uso de las redes sociales como instrumento de comunicación está motivado por intereses propagandísticos y humanitarios. Esta tendencia es evidente entre los cristianos de Siria. Este trabajo, pues, se centra en dos páginas de Facebook vinculadas a este colectivo. A partir de un análisis de contenido se describen y analizan las posibilidades de comunicación que ofrecen las redes sociales a grupos específicos en un entorno de conflicto armado multicausal en el que la dimensión confesional es relevante.
